Zusammenfassung 1. Kynurenin-3-Hydroxylase wurde in Homogenaten der Schmeißfliege Calliphora erythrocephala Meigen in vitro untersucht und mit dem Enzym aus Säugerlebermitochondrien verglichen. Die enzymatische Hydroxylierung wurde durch eine spezifische Farbreaktion des 3-Hydroxy-kynurenins und durch Isolierung von 3-Hydroxy-kynurenin-3H nach Inkubation mit Kynurenin-3H nachgewiesen. 2. Die Aktivität des Enzyms ist außerordentlich niedrig und übersteigt in Rohhomogenaten nicht 10−5–10−4 Einheiten/mg Protein. Sie fällt im Homogenat bei 4° C mit einer Halbwertzeit von etwa 1 Tag ab. 3. Die meßbare Reaktion resultiert vor allem aus der Bildung und der Weiterreaktion von 3-Hydroxy-kynurenin (3-HOK), wird aber durch die Konkurrenz um das Cosubstrat ebenfalls beeinflußt. Das Enzym arbeitet mit NADPH als Cosubstrat, während NADH eine geringere Wirkung zeigt. Azid-Ionen wirken stimulierend, eine Erhöhung der Kynurenin-Konzentration von 0,33 auf 3,3 mM stark hemmend. Anthranilsäure und „Desoxokynurenin“ hemmten das Enzym nicht. 4. Das pH-Optimum des calliphora-Enzyms dürfte bei pH 7,2 liegen; in Calliphora-Rohhomogenaten zeigte der pH-abhängige Aktivitätsverlauf jedoch 2 Gipfel bei pH 6,8 und pH 7,6. 5. Im Gegensatz zur Hydroxylase aus Säugerleber liegt bei Calliphora mindestens ein hoher Anteil des Enzyms im gelösten Protein der Zelle (105 00 g-Überstand) vor. Daneben wurde Enzymaktivität in der 1200 g-Partikelfraktion nachgewiesen. Ob dies auf Adsorption beruht, oder ob das Enzym in zwei Kompartimenten vorliegt, ist nicht klar. Die Aktivität im 105000 g-Überstand wird durch die Intensität des Zellaufschlusses nicht verändert. 6. Die höchste spezifische Aktivität wurde in den Malpighischen Schläuchen gefunden; die sehr niedrige Aktivität im übrigen Körper kann in Frage gestellt werden. — Im 105000 g-Überstand von Totalhomogenaten zeigt die Hydroxylaseaktivität einen entwicklungsspezifischen Verlauf, mit einem 1. Gipfel kurz vor der Verpuppung, einem 2., hohen Gipfel kurz danach und einem kontinuierlichen Absinken bis zum Imaginalstadium. In der gleichen Versuchsreihe wurde auch der Konzentrationsverlauf des 3-Hydroxy-kynurenins in der Larvenentwicklung von Calliphora bestimmt.

Summary 1. Kynurenine-3-hydroxylase has been studied in homogenates of the blowfly, Calliphora erythrocephala Meigen, and compared to the respective mammalian enzyme. Enzymatic hydroxylation of the substrate was demonstrated by a specific colour reaction of 3-hydroxy-kynurenine and by isolation of 3-hydroxykynurenine-3H following incubation of tritiated kynurenine. 2. In crude homogenates the hydroxylase activity does not exceed 10−5 to 10−4 units/mg protein. The enzyme is inactivated by about 50% in 24 hours at 4°C. Its apparent activity is determined both by formation and disappearance of product; it may be lowered by other processes competing for the same cofactor. NADPH is required for maximal activity, while some activity is also observed with NADH. The reaction is stimulated by azide ions. While there was no effect by addition of anthranilic acid or “deoxokynurenine”, increasing substrate concentration from 0.33 to 3.3 mM resulted in strong inhibition of the enzyme. Maximal activity should occur at pH 7.2. In crude homogenates however two maxima are observed at pH 6.8 and 7.6; the lower activity at pH 7.2 may be related to a competing reaction. 3. In contrast to mammalian liver, in Calliphora most of the enzyme is found in the soluble fraction (105000 xg supernatant). Some activity was found also in the 1200 xg sediment. No decision could be made as to whether this was due to adsorption or to distribution of the enzyme in two distinct compartments. Apparently the intensity of homogenization did not effect the specific activity of the enzyme in the 105000 xg supernatant. 4. The hydroxylase activity is highest in the Malpighian tubules; it is doubtful, whether there is any activity in other tissues. Specific activity was measured in supernatant fractions during development of the blowfly. It increases from zero in the egg stage through the larval stages to reach a maximum prior to pupation; shortly after pupation a second peak is observed from which activity declines steadily. There is little if any activity in the imagoes. In the same series of experiments the concentration of 3-hydroxy-kynurenine in Calliphora larvae was determined.
