    Description: Cis and trans dichlorodiaminoplatinum (II) compounds bind to DNA and form DNA cross-links, which are usually considered to be irreversible. Thiourea can reverse these cross-links without any apparent breakdown of the DNA. In addition, cis- and trans-Pt (II) treatment of lambda decreases its transfectivity.     Description: Of the proteins in mechanically disrupted chicken gizzard fibers (no functional sarcolemma) only the 20,000-dalton light chains of myosin underwent large Ca2+-and Sr2+-dependent changes in phosphorylation. Phosphorylation closely corresponded with the Ca2+- and Sr2+-activated tensions.     Description: Norepinephrine reversibly antagonizes three calcium-dependent potentials recorded from rat postganglionic neurons. Norepinephrine inhibits the development of a shoulder on the aciton potential, the magnitude of the hyperpolarizing afterpotential, and the rate of rise and amplitude of the calcium spike. The action of norepinephrine is antagonized by the alpha-adrenergic antagonist phentolamine, but not by MJ 1999, a beta-adrenergic antagonist.     Description: A variant of the MPC 11 cell line, M 311, produces a short immunoglobulin heavy chain. When compared with the parental gamma 2b heavy chain, M 311 was found to have a carboxyl terminal deletion comprising the CH3 domain. The COOH-terminal cyanogen bromide (CNBr) cleavage fragment of M 311 is identical to a corresponding segment ofa parental heavy chain CNBr fragment, with the exception of a substitution of asparagine for lysine at the COOH-terminal residue. This observation enabled prediction of both the parental DNA sequence in this region and the genetic mechanism which generated the variant, a frameshift followed by premature termination.     Description: Tetramethylrhodamine labeled N-formyl-Nle-Leu-Phe-Nle-Tyr-Lys is a potent chemoattractant for human neutrophils. Binding of this peptide to living neutrophils was observed by means of video intensification microscopy. At 37 degrees C, diffuse membrane fluorescence was seen initially, followed by rapid aggregation and internalization of the fluorescent peptide.     Description: A study was made of the function of the intervening sequences in the ovalbumin gene, Radioactively labeled DNA probes for the intervening sequences were prepared and RNA's were isolated from whole cells, nuclei, and polysomes of estrogen-stimulated chick oviducts. The concentrations of messenger RNA (mRNA) transcripts from ovalbumin structural sequences (mRNAov) and transcripts corresponding to intervening sequences were then estimated by hybridization to cloned DNA probes. Oviduct tissue contains approximately 58,000 molecules of mRNAov sequences per tubular gland cell and most of these sequences are present in the cytoplasm. In contrast, there are 200 to 300 molecules of RNA per cell which are transcribed from the intervening sequences of the natural ovalbumin gene and almost all of these are found in the nucleus. The morphology of these intraerythrocytic protozoa and the range of hosts that could be infected experimentally suggested that the parasite was B. gibsoni.     Description: The channels in the junctions of various mammalian cell types--primary cultures and lines--were probed with a series of linear fluorescent amino acid and peptide molecules of different size and charge. Permeability is limited by probe size and electronegativity, these two factors apparently being related reciprocally. In respect to both factors, mammalian junctional channels are more restrictive than insect channels; hence the mammalian channels are narrower, more polar, or both.     Description: By using 4.45-angstrom radiation generated by Cl+15 ions in a laser plasma and nanosecond exposures, low-angle x-ray diffraction patterns were obtained from dried rat spinal nerves and a powder of cholesterol.     Description: The effects of thyrotropin-releasing hormone and 17 beta-estradiol on the electrical membrane properties of a prolactin-secretin pituitary cell line (GH3/B6) were studied with intracellular microelectrode recordings. Of the cells tested, 50 percent were excitable and displayed calcium-dependent action potentials when depolarized. When injected directly on the membrane of an excitable cell, thyrotropin-releasing hormone and 17 beta-estradiol induced action potentials within 1 minute. The spiking activity was preceded by a progressive increase of the input resistance without any detectable change in the resting membrane polarization. The results reveal a rapid effect of both substances on the membrane of GH3/B6 cells. In the case of thyrotropin-releasing hormone, which has both a short-term effect on release of prolactin and a long-term effect on its synthesis, the induced electrical activity may be associated with the stimulation of prolactin production.     Description: A bioactive, fluorescent derivative of enkephalin, Tyr-D-Ala-Gly-Phe-Leu-Lys-rhodamine, was used to determine the distribution of opiate receptors in living neuroblastoma cells. The receptors appeared in clusters on the cell surface, and no internalization was detected.     Description: To explore possible mechanisms for the metastasis of malignant cells to bone, a model of tumor cell migration was developed, using Walker carcinosarcoma or malignant lymphoma cells. It was found that bone contains a factor that is strongly chemotactic for tumor cells.     Description: When incubated in a calcium-free medium, isolated rat fasciculata cells showed neither an increase in the concentration of guanocine 3',5'-monophosphate (cyclic GMP) nor an increase in corticosterone production in response to adrenocorticotropic hormone (ACTH). In response to submaximum and maximum steroidogenic concentrations of ACTH, corticosterone formation was directly proportional to increases in calcium concentration ranging from 0 to 2.5 mM. Higher concentration of calcium, however, inhibited maximal ACTH-induced steroidogenesis. In the absence of ACTH, calcium did not stimulate cyclic GMP accumulation and corticosterone formation. ACTH-induced corticosterone synthesis, preceded by an increase in cyclic GMP, was restored when ACTH and calcium were both present in the medium. Cyclic GMP or dibutryl cyclic GMP-induced steroidogenesis was substantially reduced in the absence of calcium, but in contrast to the ACTH effect a significant amount of corticosterone formation occurred without calcium.     Description: Macrophage migration inhibition factor (MIF) derived from human lymphoid cell lines was found to lose biologic activity on dialysis. Although activity was not recovered in the dialyzate, mixing experiments demonstrated that the components in the retentate and dialyzate could reassociate to restore activity. The fragment of larger molecular weight (less than 10,000) could inhibit the activity of intact MIF, whereas the smaller molecular weight fragment (5,000 to 10,000) could not. These findings suggest that human MIF is composed of at least two noncovalently linked subunits.     Description: A "recptor unit" for gamma-aminobutyric acid (GABA), which includes brainlike receptor binding sites for tritium-labeled GABA and benzodiazepines (diazepam, clonazepam, and flunitrazepam) and a thermostable endogenous protein (GABA modulin) that inhibits both GABA and benzodiazepine binding, has been demonstrated in membranes prepared from NB2a neuroblastoma and C6 glioma clonal cell lines.     Description: Repetitive stimulation (100 pulses per second for 1 second) of the Schafer collateral-commissural system of the rat hippocampus induces long-term potentiation of synaptic strength and produces significant changes in the subsequent endogenous phosphorylation of a 40,000-dalton protein from synaptic plasma membranes. This effect is not observed after stimulation in calcium-deficient media or after simulation at the rate of one pulse per second for 100 seconds.     Description: Substitution of fluorine for hydrogen in position 2, 5, or 6 of the aromatic ring of norepinephrine markedly alters the alpha- and beta-adrenergic agonist properties of norephinephrine. The 6-fluoro isomer is an beta-adrenergic agonist with virtually no beta agonist activity, while the 2-fluoro isomer is a beta-adrenergic agonist with little alpha activity. The 5-fluoro isomer is equipotent with norepinephrine as an alpha agonist and significantly more potent as a beta agonist.     Description: Vital statistics data for the United States from 1922 to 1936 and from 1950 to 1972 were used to analyze fetal and early neonatal mortality. This analysis corroborates the previously established pattern of the sex ratio of fetal deaths--highest from months 3 to 5, lower from months 6 to 7 or 8, and increasing at term. It also indicates a postponement of late fetal deaths into the early infant period. Whereas earlier research reports have described the pattern of the sex ratio of fetal deaths, this report repeats this analysis for a recent national data base. This line of analysis is extended by using the patterns observed in the data to produce an empirical estimate of the primary sex ratio.     Description: In studies with toad bladders, the uncoupling agent 2,4-dinitrophenol (DNP) reversed the inhibition of CO2 production produced by direct inhibition of transport.     Description: C6 glioma cells and B104 neuroblastoma cells both possess adenylate cyclase activity, but only C6 cells have beta-adrenergic receptors. However, when cocultured with B104 cells, C6 cells show a marked decrease in their ability to accumulate adenosine 3', 5'-monophosphate upon stimulation with beta receptor agonists.     Description: The cytosol extracted from a vascular endothelial cell line binds [3H]estradiol with high affinity and a high degree of specificity. In contrast, in experiments performed with cytosol labeled in the intact cell, progesterone and, to a smaller extent, testosterone gave an apparent inhibition of estradiol binding.     Description: Between 1973 and 1977 the total number of Ph.D.'s holding postdoctoral appointments in the biomedical sciences increased at a rate of more than 550 individuals (12.5 percent) per year. During this same period the total number of doctorates awarded each year in these disciplines showed very little change. The postdoctoral growth can be attributed to substantial increases in both the numbers of recent graduates taking postdoctorals and the length of stay on these appointments. The lack of alternative employment opportunities has contributed heavily to the postdoctoral buildup.     Description: This article classifies the major approaches to the assessment of the process and outcomes of medical care. The apparent need to safeguard and enhance the quality of care has led to the institution of mechanisms that subject care to constant review so that deficiencies may be found and corrected. The article reviews the developments that led to the involvement of the federal government in this activity through its sponsorship of professional standards review organizations (PSRO's). The major features of the PSRO's are described and their possible effects discussed.     Description: This article reports a study of the activities and performance of institutional review boards to protect human research subjects. Researchers and institutional review board members were generally supportive of the review system, although substantial criticisms were also heard. Institutional review boards had some direct impact on half of the proposals reviewed by requiring either modification of or additional information about proposed research. The data, however, raise questions about the effectiveness of some review board actions, for example, with regard to informed consent.     Description: High concentrations (80 to 90 millimolar) of the calcium buffers EGTA and citrate (less than 10(-7) molar free calcium ion) reversibly block excitation-contraction coupling in intact frog skeletal fibers, but do not block caffeine-induced contractures. Solutions containing the same free calcium concentration but lower concentrations of calcium buffer (1 millimolar) do not block excitation-contraction coupling.     Description: A group of oxygenated sterols has been identified as potent and specific inhibitors of sterol biosynthesis. The ability of these compounds to inhibit sterol synthesis in cultured cells and the ineffectiveness of cholesterol under the same conditions suggest that feedback regulation of sterol biosynthesis may be brought about by an oxygenated sterol rather than by cholesterol. The nature of the regulatory sterol may vary in different cells with their specific requirements for cholesterol as a structural component or as a precursor of other steroid products. The use of oxygenated sterols to block sterol synthesis in cultured cells provides new information regarding the role of sterol in cell membrane structure and function. For example, de novo sterol synthesis is required for DNA synthesis and cell division by some cultured cells.     Description: Long-term care for the aged in the United States is overly dependent upon the nursing home. This hospital-like model for long-term care is particularly inappropriate since it imposes a medical solution on a variety of social problems. An adequate long-term care program requires a range of resources in the community as well as in institutions; sheltered housing options seem a desirable alternative to the nursing home. To ensure the quality of nursing home care, the present focus on setting standards for care activities should be abandoned in favor of a focus on care outcomes (physical, mental, and social).
