    Notes: Summary To test the importance of 24-hydroxylation of vitamin D3 on bone mineralization, rat pups born to vitamin D-deficient females were given either 25-hydroxyvitamin D3 or 24,24-difluoro-25-hydroxyvitamin D3 for 16 days beginning at the time of weaning. Following such treatment analysis of blood samples revealed no detectable 24R,25-(OH)2D3 and 1,25-(OH)2D3 in the rats given the difluoro compound while revealing the expected 24,24-difluoro-25-hydroxyvitamin D3 and 24,24-difluoro-1,25-dihydroxyvitamin D3. The rats given 25-hydroxyvitamin D3 had the expected levels of 25-hydroxyvitamin D3, 24,25-dihydroxyvitamin D3, and 1,25-dihydroxyvitamin D3. Following sacrifice at day 17, postweaning bone mineralization and modeling were studied in long bones using histological methods. Bones taken from vitamin D-deficient rats at the beginning and end of the experimental period had lesions typical of rickets. These included wide growth plates, excessive amounts of osteoid, and metaphyseal fibrosis. Following treatment with either 25-hydroxyvitamin D3 or 24,24-difluoro-25-hydroxyvitamin D3, bone mineralization returned to normal. Growth plate widths and the amount of osteoid on bone surfaces were both substantially reduced and to a similar degree in both treatment groups. Normal cartilage core formation and trabecularization of the metaphyseal primary spongiosa were also restored to a similar degree in both groups. In effect, no difference was observed in any bone parameter studied between the 25-hydroxyvitamin D3- and the 24,24-difluoro-25-hydroxyvitamin D3-treated animals. These results provide strong evidence that 24-hydroxylation of the vitamin D molecule plays little or no role in the modeling and mineralization of bone.

    Notes: Summary Using the technique of short interval sequential tetracycline labeling, it was documented that the apposition of mineralized bone matrix in adult male Sprague-Dawley rats was inhibited by hydrocortisone. The inhibition occurred as early as six days after the onset of the treatment and was dose dependent over a dose range of 0.62 to 20 mg per kg body weight per day. Vitamin D2 supplements by injection protected bone from this hydrocortisone action. 64 I. U. of vitamin D2 injected daily was able to prevent the inhibition of bone apposition by 20 mg per kg body weight per day of hydrocortisone. The results imply that vitamin D or its metabolites may compete with hydrocortisone in some cellular mechanisms and support the usefulness of vitamin D supplements in the treatment and the prevention of steroid-induced osteoporosis.

    Notes: Summary The therapeutic effects of vitamin D analogs, 1,24(R)-dihydroxycholecalciferol [1,24(R)(OH)2D3], 1,24(S)-dihydroxycholecalciferol [1,24(S)(OH)2D3], and 1,25-dihydroxycholecalciferol [1,25(OH)2D3] on immobilization osteoporosis were studied in rats. The right hind limb was immobilized through application of a plaster cast following the section of the sciatic nerve. The left hind limb was intact. Vitamin D analogs were orally administered for 6 weeks at dose levels of 0.02 and 0.10µg/kg/day, respectively. The mean lengths of the immobilized femurs were not significantly different from those of the intact femurs in all the experimental groups. In the immobilized femur of animals treated with 1,24(R)(OH)2D3, 0.10µg/kg, dry and ash weights were heavier and calcium and phosphorus contents greater than those in the nontreated group. Furthermore, the amount of calcified bone mass and the cortical thickness of the femurs of the immobilized limb in 1,24(R)(OH)2D3-treated animals were greater than those in the nontreated animals. Treatment with 1,25(OH)2D3 at 0.10µg/kg caused an increase of the bone mass in both immobilized and intact femurs when compared with those of the control group. It was concluded that the administration of 1,24(R)(OH)2D3 diminished the effect of immobilization in the development of osteoporosis without any side effects.

    Notes: Summary A simplified method is described for the measurement of calcium absorption in the dog. This method uses45Ca and takes less than 8 h to perform. Calcium absorption was measured in normal and uremic dogs on different intakes of calcium and phosphorus and compared to the results obtained in the same animal maintained on the same diet by another method using47Ca and a double dilution procedure. The correlation coefficient between the two methods was 0.99 (P〈0.001). The advantages of the simplified method are discussed.

    Notes: Summary Thaw-mount autoradiographic studies after injection of3H-1,25-D3 were conducted on 18-and 20-day-old rat fetuses. In maxillary bones, ribs, and tibia, nuclear concentration of radioactivity was found in osteoprogenitor cells and osteoblasts. Osteocytes and chondrocytes in epiphyseal plates were either unlabeled or weakly labeled. In competition experiments, nuclear concentration of radioactivity was blocked by the injection of a high dose of nonradioactive 1,25-D3 prior to the administration of the labeled hormone, but not by a similar dose of nonradioactive 25-D3. The results are interpreted as indicating that osteoprogenitor cells and osteoblasts are target cells for the direct action of 1,25-D3 on fetal bone.

    Notes: Summary The role of vitamin D in early skeletal development was studied by measuring serum calcium and phosphorus, osseous tissue quantity and mineralization, and endochondral bone elongation in rat fetuses and pups from vitamin D-replete and vitamin D-deficient mothers. At the 20th day of pregnancy there was a slight, yet significant, increase in the amount of osteoid on trabecular bone surfaces in fetuses from vitamin D-deficient mothers. The fetal bones otherwise appeared normal in spite of severe skeletal changes in the vitamin D-deficient mothers. After parturition, the importance of vitamin D in skeletal development becomes progressively more obvious. Serum calcium levels were slightly, yet significantly, lower in vitamin D-deficient than in vitamin D-replete pups and these levels continued to fall in the vitamin D-deficient pups through lactation and after weaning. At 3 days postpartum, there was a small, yet significant, increase in the amount of osteoid on bone surfaces of the vitamin D-deficient pups. The relative amounts of osteoid in the vitamin D-deficient pups continued to increase through lactation and after weaning when compared with vitamin D-replete pups. By the 14th day of lactation and at later periods, there were significant reductions in metaphyseal mineralized tissues in the vitamin D-deficient pups when compared with the vitamin D-replete pups. At weaning and after weaning, there were substantial increases in growth plate thickness and decreases in longitudinal bone growth in the vitamin D-deficient pups when compared with the vitamin D-replete pups. The results from this study indicate that vitamin D does not appear to play a major role in fetal skeletal development. However, after birth, vitamin D becomes progressively more important with age for normal bone development, mineralization, and endochondral growth.

    Notes: Summary To evaluate the relationships of a 25-hydroxy vitamin D absorption test to intestinal fat absorption and to the absorption of radioactively labeled vitamin D and 25-hydroxy vitamin D, an investigation was undertaken in 19 patients with gastrointestinal disorders. A correlation was noted between the results of the 25-hydroxy vitamin D absorption test and baseline 25-hydroxy vitamin D levels. No correlation was found between results of the 25-hydroxy vitamin D absorption test and quantitative fecal fat excretion. Peak values of the 25-hydroxy vitamin D absorption test correlated with net absorption of14C vitamin D. No correlation existed between the values obtained in the 25-hydroxy vitamin D absorption test and3H-25-OH D absorption. These studies indicate that the 25-hydroxy vitamin D absorption test probably does not serve as an effective screening test for intestinal fat malabsorption. The results of the test probably most accurately reflect the body stores of vitamin D at the time of testing, but there appears to be little advantage to performing a 25-hydroxy vitamin D absorption test in lieu of a single determination of the serum level of 25-hydroxy vitamin D as a method of evaluating vitamin D nutritional status.

    Description / Table of Contents: Résumé Les effects d'une dose de 1 mg de vitamine D3 (40 000 Unités,) administrée par voie intra-veineuse, ont été étudiés sur l'histologie osseuse et le métabolisme du phosphore chez 19 sujets contrôles dont l'histologie était normale et 28 malades présentant les caractères histologiques de l'ostéomalacie par carence vitaminique D. L'administration de la vitamine D n'a entraîné aucune modification histologique ou biologique significative chez les sujets contrôles. Mais chez les ostéomalaciques, il est apparu en moins de sept jours une augmentation très significative du front de calcification à l'interface tissue-ostéoïde-tissue-calcifié. Cette modification s'accompagnait d'une ascension progressive de la phosphatémie et de la réabsorption tubulaire du phosphore atteignant dans le même délai des valeurs normales.
    Abstract: Zusammenfassung Bei 19 Patienten mit normaler Knochenhistologie und bei 28 Patienten mit histologisch gesicherter Osteomalacie wurde die Wirkung von 1 mg Vitamin D3 (40000 Iv) i.v. auf die Knochenhistologie und den Phosphatmetabolismus untersucht. Bei den Kontrollpatienten konnten keine signifikanten Änderungen nach Vitamin D festgestellt werden, wogegen die Osteomalacie-patienten innerhalb von 7 Tagen eine deutliche Zunahme der Verkalkungszone an der Grenze zwischen Osteoid- und Knochengewebe zeigten. Diese Änderung war von einer fortschreitenden Zunahme des Serum-Phosphates, verbunden mit einer gesteigerten renalen tubulären Rückabsorption des Phosphates begleitet; beide kehrten anschließend gleichzeitig wieder zur Norm zurück.
    Notes: Abstract The effect of a 1 mg dose of intravenous Vitamin D3 (40,000 i.u.) on bone histology and phosphate metabolism was investigated in 19 patients with normal bone histology and 28 patients with histological evidence of osteomalacia. No significant changes occurred in the control patients after Vitamin D but the patients with osteomalacia showed a marked increase, within seven days, in the proportion of osteoid having a calcification front. This was accompanied by a progressive rise in the serum phosphate, which was associated with an increase in the renal tubular reabsorption of phosphate to normal.

    Description / Table of Contents: Résumé On a fait des recherches sur les conséquences d'un manque de la vitamine D par rapport a la croissance, a la composition ionique du sang et de l'activité encymatique dans l'os chez le rat. La diét contenait 0,6% de calcium et 0,6% de phosphore. Sans apport de vitamine D la croissance était lente, la cendre de la métaphyse diminuait, le serum calcium abaissait le serum phosphore augmentait. L'activité des encymes dans la métaphyse et la diaphyse de la tibia et du femur était la suivante: alkaline phosphatase, acide β-glycerophosphatase, activité protéolytique a pH 8, ta leucin-aminopéptidase et la glutamatdehydrogenase était augmenté dans la métaphyse; la lactatdehydrogenase et l'acide phénylphosphatase était diminué. Des changements comparables étaient decouvertes dans la diaphyse a l'éxception d'une activité de lactatdehydrogenase et de phenylphosphatase non alterée; en outre l'activité proteolytique a pH 5 était augmentée. Le manque de la vitamine D restait sans influence sur l'aspartattrans-aminase et alanintransaminase. L'activité proteolytique a pH 3,7 n'était pas changée. Ces altération encymatiques ne peuvent pas etre provoquées par une malnutrition parce que'elles n'etaient pas observées chez des animales avec une nuriture identique mais a, laquelle on avait ajouté de la vitamin D. Les encymes cataboliques ne manquait pas, la calcémie basse n'est donc pas éxpliquable par une réduction de la résorption osseuse. Les résultats sont plustôt en faveur d'une résorption osseuse très active.
    Abstract: Zusammenfassung Der Effekt von Vitamin D-Mangel auf Wachstum, Serumionen und Enzymaktivität im Knochen wurde bei der Ratte untersucht. Die Diät enthielt 0,6% Calcium und 0,6% Phosphor. Vitamin D-freie Diät hatte verzögertes Wachstum, Abnahme der Knochenasche in der Metaphyse, erniedrigtes Serumcalcium und erhöhtes Serumphosphat zur Folge. Die enzymatische Aktivität in der Metaphyse und Diaphyse von Tibia und Femur veränderte sich: alkalische Phosphatase, saure β-Glycerophosphatase, proteolytische Aktivität bei pH 8, Leucin-Aminopeptidase und Glutamatdehydrogenase waren in Knochenproben der Metaphyse vermehrt, Lactatdehydrogenase und saure Phenylphosphatase vermindert. Ähnliche Veränderungen der Enzymaktivität im Knochen fanden sich in der Diaphyse, mit Ausnahme unveränderter Aktivität von Lactatdehydrogenase und saurer Phenyphosphatase sowie erhöhter proteolytischer Aktivität bei pH 5. Der Vitamin D-Mangel ließ die Aspartat-und Alanin-Transaminasen sowie die proteolytische Aktivität bei pH 3,7 unverändert. Die Enzymveränderungen sind nicht durch geringere Nahrungsaufnahme bedingt, weil sie bei Tieren mit identischer Ernährung, aber ohne Vitamin D-Entzug, nicht beobachtet wurden. Ein Mangel an Abbaufermenten, welcher Hemmung der Knochenresorption bewirken und für den Abfall des Serumcalciums verantwortlich sein könnte, ließ sich nicht finden. Die Resultate sprechen eher für vermehrte Knochenresorption.
    Notes: Abstract The effect of vitamin D deficiency on growth, serum ions and bone enzymatic activity has been studied in rats. The diet contained 0.6% calcium and 0.6% phosphorus. Omission of vitamin D from the diet resulted in decreased growth, decreased metaphyseal bone ash, decreased serum calcium and increased serum phosphorus. Changes in enzymatic activity in metaphyseal and diaphyseal bone from the tibia and femur also occurred. Alkaline phosphatase, acid β-glycerophosphatase, proteolytic activity at pH 8.0, leucine aminopeptidase, and glutamate dehydrogenase were increased in metaphyseal bone samples. Lactate dehydrogenase and acid phenylphosphatase were decreased. Similar changes in bone enzymatic activity occurred in the diaphysis except that the lactate dehydrogenase and acid phenylphosphatase activities were not decreased and proteolytic activity at pH 5.6 was increased. The vitamin D deficiency did not affect aspartate or alanine transaminase or proteolytic activity at pH 3.7. These enzyme changes were not due to decreased food consumption because they did not occur in pair-fed animals. There was no lack of degradative enzymes that would impair bone resorption and lead to a fall in serum calcium. Rather the results were more consistent with an increase in bone resorption.

    Description / Table of Contents: Résumé On a étudié l'effet de la suppression des capsules surrénales au point de vue de la distribution du radiocalcium chez des rats rachitiques et chez des rats traités par la vitamine D. Cette opération a produit une augmentation du dépôt de radiocalcium dans les fémurs des rats rachitiques et également des rats traités par la vitamine D comparés à leurs contrôles respectifs ayant eu une opération simulée. Cependant, une analyse kinétique des données sur l'assimilation squelettique du radiocalcium a démontré que les degrés de déposition du minéral de l'os étaient amoindris par la suppression des capsules surrénales du moins chez les animaux traités par la vitamine D. D'autre part, la vitamine D a augmenté le degré de déposition plus que celui des contrôles rachitiques à la fois chez les animaux ayant eu ou non leurs capsules surrénales supprimées. De plus on a démontré que la vitamine D a rehaussé l'absorption intestinale autant que la déposition squelettique du radiocalcium chez le rat rachitique à qui on a enlevé les capsules surrénales.
    Abstract: Zusammenfassung In dieser Arbeit wurde die Auswirkung der Nebennierenentfernjng auf die Verteilung radioaktivem Calciums in rachitischen und Vitamin D behandelten Ratten untersucht. Entfernung der Nebennieren hatte eine erhöhte Ablagerung von radioaktivem Calcium in den Obserschenkelknochen der rachitischen und mit Vitamin D behandelten Ratten gegenüber den jeweiligen scheinoperierten Kontrollen zur Folge. Jedoch konnte durch kinetische Analyse der Ablagerungswerte von radioaktivem Calcium im Skelet festgestellt werden, daß die Menge der abgelagerten Knochenmineralien durch Entfernung der Nebennieren zumindest bei den mit Vitamin D behandelten Ratten herabgesetzt wurde. Auf der anderen Seite hat die Verabreichung von Titamin D die Ablagerungsmenge bei Ratten, denen die Nebennieren entfernt wurden und in solchen, die nicht operiert wurden, den rachitischen Kontrollen gegenüber erhöht. Zusätzlich konnte gezeigt werden, daß bei rachitischen Ratten nach Nebennierenentfernung und nach Behandlung mit Vitamin D erhöhte Darmabsorption und Skeletablagerung radioaktiven Calciums stattfand.
    Notes: Abstract The effect of adrenalectomy on radiocalcium distribution in rachitic and vitamin D-treated rats was studied. Adrenalectomy resulted in an increased deposition of radiocalcium in the femurs of both rachitic and vitamin D-treated rats when compared with their respective shamoperated controls. However, kinetic analysis of the data on the skeletal uptake of radiocalcium demonstrated that accretion rates of bone mineral were decreased by adrenalectomy at least in vitamin D-treated animals. Vitamin D administration, on the other, hand increased the accretion rate over that of rachitic controls in both adrenalectomized and non-adrenalectomized animals. In addition vitamin D was shown to enhance intestinal absorption as well as skeletal deposition of radiocalcium in the adrenalectomized rachitic rat.
