Notizen: Abstract Within the Jet Propulsion Laboratory in Pasadena, California, state of the art computer graphics animation is done in the Computer Graphics Laboratory. The topics of the animations cover many scientific disciplines. Specific features of the system developed there, both hardware and software, are discussed. The prime mover of the effort is Dr. James F. Blinn of Pasadena; his role and experiences are elaborated. Their current largest project is The Mechanical Universe; the system is used for its production.

Notizen: Abstract Alpine lakes in siliceous catchments of Tyrol and Carinthia (Austria) show signs of acidification. About 9% of the studied lakes have no alkalinity, more than 20% are below pH 6. About two thirds of all lakes have acid neutralizing capacities below 100 μeq 1−1. In spite of moderate precipitation acidity, some lakes show considerable concentrations of dissolved reactive aluminum during or shortly after snowmelt. High altitude lakes of the Alps are definitely more acidic than high mountain lakes in remote areas. Large differences in water and soil chemistry of nearby situated lakes were attributed to heterogeneities of bedrock geology. Paleolimnological investigations on former pH values of five lakes, based on diatom assemblages in the sediment, showed different developments: recent and past acidification, stable conditions, and alkalinization.

Notizen: Abstract The role of histamine in brain function is discussed. A brief review is presented on the three types of histamine receptors with regard to their biochemistry and functions, as well as on specific ligands, both agonists and antagonists. It is concluded that several aspects of the pharmacology of histamine in the central nervous system are still a matter of empirism and speculations. The recent publications on the brain-penetrating H2 antagonist zolantidine and the selective H3 agonists and antagonists are expected to contribute to the knowledge of the histaminergic pathways in the brain. Therapeutical areas within the central nervous system are related to behaviour (including wake-sleep), neuroendocrinal processes, the control of muscle activity and cerebral circulation.

Notizen: Abstract From a pharmacochemical point of view the existing anti-leprotics as well as possible innovations in the chemotherapy of leprosy are discussed. Of the main anti-leprotics, which are used nowadays — dapsone, rifampicin, clofazimine, isoniazide, ethionamide and prothionamide — the mechanism of action, the main problems in their application and possibilities to develop improved variants are reviewed. Based on the chemistry ofMycobacterium leprae, the target systems for new anti-leprotics are identified. These systems include the cell wall, the catabolism of reactive oxygen species, the metabolisms of carbon sources, the amino acid metabolism and the uptake of iron. Two possible new lead structures from other fields, 4-quinolones and mycobacterial ribonucleotide reductase inhibitors are presented.

Notizen: Abstract Developments in the receptor concept have greatly influenced our current knowledge of the beta-adrenoceptor. The triad of pharmacology, organic chemistry and studies in structure-activity relationships is discussed along historical lines, as it has been and still is an impetus for progress in the biochemistry of ligand-receptor interactions. With respect to the beta-adrenoceptor complex these advances which have led to a model in which three protein structures are functionally interacting within the frame of the cell wall: the beta-adrenoceptor, the regulatory guanine nucleotide binding protein, and the enzyme adenylate cyclase, are reviewed.

Notizen: Abstract Etoposide (VP 16-213) and teniposide (VM 26) are semisynthetic epipodophyllotoxin derivatives active against a variety of tumours. The clinical efficacy has led to an increasing interest in these compounds. This review presents information on the mechanism of action, biochemical pharmacology, bioanalysis, metabolism and pharmacokinetics of etoposide and teniposide.

Notizen: Abstract It is now known that cardiac atria play an important role in blood pressure and volume regulation. Mechanical distension of the atria results in the release of a potent diuretic and natriuretic agent or agents termed the atrial natriuretic factor (ANF). Several structurally related forms of ANF exist in man and it is thought that these represent precursory forms of a single optimally active molecule and/or the presence of more than one form of active ANF. The chemical structure of ANF between different mammalian species is similar. ANF receptors have been identified in kidney, brain, vascular endothelial and smooth muscle cells, tracheal and bronchial smooth muscle and the adrenal glands of many mammalian species, including man. This would suggest that ANF influences blood pressure and volume homoeostasis by affecting any one of a number of biochemical or physiological mechanisms via different target tissues. ANF is now considered a potentially valuable therapeutic agent for the treatment of hypertension and congestive heart failure. Synthesis of potent receptor antagonists could be extremely useful in the treatment of various clinical situations which are produced or complicated by endogenously produced ANF, such as chronic orthostatic hypotension.

Notizen: A morphological study was made to examine the friction and wear mechanisms of ultrahigh molecular weight polyethylene in air and water by means of transmission electron microscopy of a carbon replica film of a very thin layer of the polymer surface slid against a steel ball. Electron diffractions revealed that a molecular orientation with a typical fiber pattern was formed in the friction in air, while thin ribbon-like crystals with particular crystal orientations were formed in water in a way that the sliding took place on the crystallographic a - c plane, giving rise to a low friction coefficient of the polymer.
