    Description: The cause of decadal climate variability over the North Pacific Ocean and North America is investigated by the analysis of data from a multidecadal integration with a state-of-the-art coupled ocean-atmosphere model and observations. About one-third of the low-frequency climate variability in the region of interest can be attributed to a cycle involving unstable air-sea interactions between the subtropical gyre circulation in the North Pacific and the Aleutian low-pressure system. The existence of this cycle provides a basis for long-range climate forecasting over the western United States at decadal time scales.

    Description: The T cell antigen receptor (TCR) beta chain regulates early T cell development in the absence of the TCR alpha chain. The developmentally controlled gene described here encodes the pre-TCR alpha (pT alpha) chain, which covalently associates with TCR beta and with the CD3 proteins forms a pre-TCR complex that transduces signals in immature thymocytes. Unlike the lambda 5 pre-B cell receptor protein, the pT alpha chain is a type I transmembrane protein whose cytoplasmic tail contains two potential phosphorylation sites and a Src homology 3 (SH3)-domain binding sequence. Pre-TCR alpha transfection experiments indicated that surface expression of the pre-TCR is controlled by additional developmentally regulated proteins.     Description: The density of impact craters on large volcanoes on Venus is half the average crater density for the planet. The crater density on some classes of coronae is not significantly different from the global average density, but coronae with extensive associated volcanic deposits have lower crater densities.     Description: Some species of bacteria have low-permeability membrane barriers and are thereby "intrinsically" resistant to many antibiotics; they are selected out in the multitude of antibiotics present in the hospital environment and thus cause many hospital-acquired infections. Some strains of originally antibiotic-susceptible species may also acquire resistance through decreases in the permeability of membrane barriers. Another mechanism for preventing access of drugs to targets is the membrane-associated energy-driven efflux, which plays a major role in drug resistance, especially in combination with the permeation barrier. Recent results indicate the existence of bacterial efflux systems of extremely broad substrate specificity, in many ways reminiscent of the multidrug resistance pump of mammalian cells. One such system seems to play a major role in the intrinsic resistance of Pseudomonas aeruginosa, a common opportunistic pathogen.     Description: Frogs must generate a high level of mechanical power when they jump. The muscular system of frogs that jump is presumably designed to deliver these high powers. The length changes and activation pattern that muscles undergo during jumping were measured, and isolated muscle bundles were driven through this in vivo pattern. During jumping, muscles generated maximum power. Specifically, the muscle fibers (i) operated at optimal sarcomere lengths, (ii) operated at optimal shortening velocities, and (iii) were maximally activated during power generation.     Description: A simple technique has been devised that allows the direct synthesis of native backbone proteins of moderate size. Chemoselective reaction of two unprotected peptide segments gives an initial thioester-linked species. Spontaneous rearrangement of this transient intermediate yields a full-length product with a native peptide bond at the ligation site. The utility of native chemical ligation was demonstrated by the one-step preparation of a cytokine containing multiple disulfides. The polypeptide ligation product was folded and oxidized to form the native disulfide-containing protein molecule.     Description: Parallel processing has changed the way much computational physics is done. Areas such as condensed matter physics, fluid dynamics, and other fields are making use of massively parallel computers to solve immense and important problems in new ways. Simulating wave propagation is another area that has benefited through the use of parallel processing. This is graphically illustrated in this article by various numerical simulations of ultrasonic pulses propagating through solids carried out on a massively parallel computer. These computations are accompanied by visualizations of the resulting wavefield. The calculations and visualizations, together, can be completed in only seconds to several minutes and compare well with experimental data.     Description: Nucleotide sequence information derived from DNA segments of the human and other genomes is accumulating rapidly. However, it frequently proves difficult to use such short DNA segments to identify clones in genomic libraries or fragments in blots of the whole genome or for in situ analysis of chromosomes. Oligonucleotide probes, consisting of two target-complementary segments, connected by a linker sequence, were designed. Upon recognition of the specific nucleic acid molecule the ends of the probes were joined through the action of a ligase, creating circular DNA molecules catenated to the target sequence.     Description: Most differentiation events in higher plants occur continuously in the postembryonic adult phase of the life cycle. Embryogenesis in plants, therefore, is concerned primarily with establishing the basic shoot-root body pattern of the plant and accumulating food reserves that will be used by the germinating seedling after a period of embryonic dormancy within the seed.     Description: Modern molecular genetic and genomic approaches are revolutionizing the study of behavior in the mouse. "Reverse genetics" (from gene to phenotype) with targeted gene transfer provides a powerful tool to dissect behavior and has been used successfully to study the effects of null mutations in genes implicated in the regulation of long-term potentiation and spatial learning in mice. In addition, "forward genetics" (from phenotype to gene) with high-efficiency mutagenesis in the mouse can uncover unknown genes and has been used to isolate a behavioral mutant of the circadian system. With the recent availability of high-density genetic maps and physical mapping resources, positional cloning of virtually any mutation is now feasible in the mouse.     Description: Translation of key proteins in the chloroplast is regulated by light. Genetic and biochemical studies in the unicellular alga Chlamydomonas reinhardtii suggest that light may regulate translation by modulating the binding of activator proteins to the 5' untranslated region of chloroplast messenger RNAs.     Description: High-resolution electron microscopy and lithium-7 nuclear magnetic resonance measurements were carried out for a disordered carbon material, prepared by heat treatment of polyphenylene, in which lithium was stored electrochemically. The nuclear magnetic resonance spectrum suggests the existence of Li(2) covalent molecules in the carbon material.     Description: The time-dependent velocity field in a rotating Couette system has been decomposed. Successive instantaneous velocity profiles were obtained by an ultrasonic Doppler velocity profile method, and the spatiotemporal velocity field had been analyzed by two-dimensional Fourier transform. The Fourier spectrum is depicted by intrinsic peaks, and the velocity field cpan be reconstructed with these peak components only, indicating a successful decomposition of the velocity field.     Description: Two molecular mechanisms of T cell-mediated cytotoxicity, one perforin-based, the other Fas-based, have been demonstrated. To determine the extent of their contribution to T cell-mediated cytotoxicity, a range of effector cells from normal control or perforin-deficient mice were tested against a panel of target cells with various levels of Fas expression. All cytotoxicity observed was due to either of these mechanisms, and no third mechanism was detected.     Description: Mice rendered deficient in lymphotoxin (LT) by gene targeting in embryonic stem cells have no morphologically detectable lymph nodes or Peyer's patches, although development of the thymus appears normal. Within the white pulp of the spleen, there is failure of normal segregation of B and T cells. Spleen and peripheral blood contain CD4+CD8- and CD4-CD8+ T cells in a normal ratio, and both T cells subsets have an apparently normal lytic function. Lymphocytes positive for immunoglobulin M are present in increased numbers in both the spleen and peripheral blood.     Description: Interspecific competition driving divergence in adaptive radiation has not previously been tested experimentally. Natural selection on a morphologically variable species of stickleback fish was contrasted in the presence and absence of a close relative. Selection was nondirectional when the target species was alone, whereas addition of the second species favored individuals most different from it morphologically and ecologically. Disproportionately severe competition between similar phenotypes indicates frequency-dependent selection, verifying a crucial element of theory of competition and character divergence.     Description: Ordered arrays of charged uniform colloidal polystyrene particles in solution form a single-wavelength laser filter that is transparent to other wavelengths. A usable filter was prepared by solidification within an acrylamide-methylene-bisacrylamide gel. The rejection wavelength of the colloidal array filter changes with gel composition. Cryogenic transmission electron microscopy of two gel formulations shows that the colloidal array filter is composed of AAA stacking of close-packed planes.     Description: The production of strings (disclination lines and loops) has been observed by means of the Kibble mechanism of domain (bubble) formation in the isotropic-nematic phase transition of the uniaxial nematic liquid crystal 4-cyano-4'-n-pentylbiphenyl. The number of strings formed per bubble is about 0.6.     Description: Projected changes in the Earth's climate can be driven from a combined set of forcing factors consisting of regionally heterogeneous anthropogenic and natural aerosols and land use changes, as well as global-scale influences from solar variability and transient increases in human-produced greenhouse gases. Thus, validation of climate model projections that are driven only by increases in greenhouse gases can be inconsistent when one attempts the validation by looking for a regional or time-evolving "fingerprint" of such projected changes in real climatic data. Until climate models are driven by time-evolving, combined, multiple, and heterogeneous forcing factors, the best global climatic change "fingerprint" will probably remain a many-decades average of hemi-spheric- to global-scale trends in surface air temperatures. Century-long global warming (or cooling) trends of 0.5 degrees C appear to have occurred infrequently over the past several thousand years-perhaps only once or twice a millennium, as proxy records suggest.     Description: Bromine is, per atom, far more efficient than chlorine in destroying stratospheric ozone, and methyl bromide is the single largest source of stratospheric bromine. The two main previously known sources of this compound are emissions from the ocean and from the compound's use as an agricultural pesticide. Laboratory biomass combustion experiments showed that methyl bromide was emitted in the smoke from various fuels tested. Methyl bromide was also found in smoke plumes from wildfires in savannas, chaparral, and boreal forest.     Description: Large-scale, westward-extending tongues of warm (Pacific) and cold (Atlantic) water are found between 2000 and 3000 meters both north and south of the equator in the Pacific and Atlantic oceans. They are centered at 5 degrees to 8 degrees north and 10 degrees to 15 degrees south (Pacific) and 5 degrees to 8 degrees north and 15 degrees to 20 degrees south (Atlantic). They are separated in both oceans by a contrasting eastward-extending tongue, centered at about 1 degrees to 2 degrees south, in agreement with previous helium isotope observations (Pacific).     Description: A putative tumor suppressor locus on the short arm of human chromosome 9 has been localized to a region of less than 40 kilobases by means of homozygous deletions in melanoma cell lines. This region contained a gene, Multiple Tumor Suppressor 1 (MTS1), that encodes a previously identified inhibitor (p16) of cyclin-dependent kinase 4. MTS1 was homozygously deleted at high frequency in cell lines derived from tumors of lung, breast, brain, bone, skin, bladder, kidney, ovary, and lymphocyte. Melanoma cell lines that carried at least one copy of MTS1 frequently carried nonsense, missense, or frameshift mutations in the gene.     Description: Specific protein-ligand interactions are critical for cellular function, and most proteins select their partners with sharp discrimination. However, the oligopeptide-binding protein of Salmonella typhimurium (OppA) binds peptides of two to five amino acid residues without regard to sequence. The crystal structure of OppA reveals a three-domain organization, unlike other periplasmic binding proteins. In OppA-peptide complexes, the ligands are completely enclosed in the protein interior, a mode of binding that normally imposes tight specificity.
