    Notizen: Abstract The effect of temperature on the ability of Colorado potato beetles (Leptinotarsa decemlineata, Say) to use horse-nettle (Solanum carolinense L.) as a host plant was determined for larvae from colonies originating from two geographically separated populations, one adapted to horse-nettle (NC) and the other unadapted to horse-nettle (MA). Survival and developmental rate on horse-nettle and potato were measured for larvae from both colonies over a range of constant temperatures (12–30 °C) and one fluctuating temperature regime (22 °C to 30 °C). The ability of Colorado potato beetles to use horse-nettle as a larval host was strongly influenced by temperature, but the effects of temperature differed greatly between beetles from the two colonies. Survival of adapted larvae on horse-nettle was highest and comparable to that on potato at the constant 30 °C and the fluctuating temperature regime. Below 30 °C, survival of adapted larvae decreased drastically but some larvae survived at all temperatures except the lowest (12 °C). In contrast, survival of unadapted larvae to adult occurred only at 30 °C, and was low (10%). At lower temperatures, all larvae died. On potato, the effect of temperature was less dramatic, and consistent across colonies. At 12 °C, survival to adult was poor (ca. 10%), but at higher temperatures, survival increased sharply and larvae from both colonies survived equally well. On potato, small but statistically significant differences in developmental rates between beetle colonies were detected at the constant but not at the fluctuating temperature regimes. Also, the developmental day degree requirements (DD) and the low temperature development threshold (T0) values for the various developmental stages did not differ between colonies on potato. On horse-nettle, development times for both colonies were always significantly longer and DD requirements were greater than on potato. At 30 °C, the only constant temperature at which larvae from the unadapted colony completed development, the development rate to adult emergence was similar to that of beetles from the adapted colony. Differences between colonies in performance on horse-nettle were not a result of host-independent, genetically based differences in the thermal requirements of the two populations. Our findings are consistent with the hypothesis that adaptation to horse-nettle by Colorado potato beetle may be facilitated by a genotype × environment interaction involving temperature. These findings have important implications for host plant utilization, host range expansion and selection of pest biotypes adapted to plant resistance traits used in crop protection.

    Notizen: Abstract The wild Bolivian potato, Solanum berthaultii Hawkes, has been used as a source of resistance to the Colorado potato beetle (CPB), Leptinotarsa decemlineata Say, one of the most significant pests of potato. In this study, two reciprocal backcross S. tuberosum x S. berthaultii potato progenies, BCB and BCT, were mapped with RFLP markers and screened for resistance to CPB consumption, oviposition and defoliation. The genotypic and phenotypic data were combined and analysed to locate quantitative trait loci (QTLs) for resistance to CPB. Three QTLs on three chromosomes in BCB, and two QTLs on two chromosomes in BCT influenced resistance. The QTLs were generally additive but one instance of epistasis was noted. Each QTL accounted for 4–12% of the phenotypic variation observed in resistance. In the more resistant BCB population, a three QTL model explained ca. 20% of the variation in CPB oviposition. When alleles at the three QTLs were homozygous S. berthaultii, oviposition was reduced ca. 60% compared to the heterozygotes. The QTLs for resistance to CPB were compared to those previously identified for the type A and B glandular trichomes, which have been implicated in resistance in the same progenies. Generally, the QTLs for resistance to CPB coincided with loci associated with the glandular trichomes confirming the importance of the glandular trichomes in mediating resistance. However, a relatively strong and consistent QTL for insect resistance in both BCB and BCT on chromosome 1 was observed that was not associated with any trichome traits, suggesting the trichomes may not account for all of the resistance observed in these progenies.

    Notizen: Abstract The relationship between Leptinotarsa decemlineata Say egg density and Coleomegilla maculata DeGeer predatory behavior was investigated at different spatial scales (plant-to-plant and plot-to-plot). Both adult C. maculata location and daily egg consumption rates were monitored over time in greenhouse and field tests. Despite aggregation in areas of highest prey density by C. maculata, egg consumption was inversely related to egg mass density at the smallest and the largest spatial scales tested. The experimental data on predation rates in high and low density field treatments were included in a mathematical model to simulate impact of natural enemies on the rate of L. decemlineata adaptation to Bt-toxin-expressing transgenic potato plants when Bt-expressing plants are mixed at the plot-to-plot level with normal potato plants. Results showed that C. maculata predatory behavior could decrease the rate at which L. decemlineata adapted to Bt-toxins if plot-to-plot mixed-plantings were used.

    Notizen: Abstract Peach potato aphids Myzus persicae (Sulzer) (Homoptera: Aphididae) were reared on artificial diets containing the steroidal glycoalkaloids (GAs) α-solanine and α-chaconine in concentrations lower or similar to those observed in potato leaves. The adults proved to be susceptible to high concentrations (80–160 mg GA /100 ml of diet) showing reduced fecundity, diet uptake and increased mortality in comparison to controls. Ingestion of these artificial diets by nymphs delayed maturity and decreased the intrinsic rate of natural increase. GAs in low concentrations marginally stimulated the reproductive performance and diet acceptability of this aphid. The possibility of GAs exerting a defensive role in potato plants against aphids is discussed.

    Notizen: Abstract  We examined the utility of otolith minor and trace element chemistry, assayed with inductively coupled plasma mass spectrometry (ICP-MS), as a means of delineating population structure in the Nassau grouper (Epinephelus striatus). We characterized the elemental composition of otoliths collected in 1993 from three locations in Exuma Sound, Bahamas and from Glover Reef, Belize in 1995. A single location in Exuma Sound was sampled in 1994 to test temporal variability in otolith composition. Five elements (Ca, Zn, Sr, Ba and Pb) were routinely detected, at levels significantly above background, by solution-based ICP-MS. Results from analysis of variance of elemental data, expressed as a ratio to Ca, indicated that there were no significant differences among the Exuma locations for any element, but significant variability was found between Glover Reef and the pooled Exuma localities for Zn/Ca, Sr/Ca and Ba/Ca ratios. Significant inter-annual differences at one Exuma Sound location was restricted to Ba/Ca ratios. Discriminant function analysis correctly classified 86% and 95% of the Belize and pooled Exuma sites, respectively. Otoliths from Belize were characterized by low Zn/Ca and high Ba/Ca and Pb/Ca ratios compared to otoliths from fish collected in Exuma Sound. Although differences in Ba levels may be related to upwelling at Glover Reef, more data are needed to definitely link otolith composition with regional differences in water chemistry.

    Notizen: Abstract The phylogenetic position of Dictyostelium inferred from 18S rRNA data contradicts that from protein data. Protein trees always show the close affinity of Dictyostelium with animals, fungi, and plants, whereas in 18S rRNA trees the branching of Dictyostelium is placed at a position before the massive radiation of protist groups including the divergence of the three kingdoms. To settle this controversial issue and to determine the correct position of Dictyostelium, we inferred the phylogenetic relationship among Dictyostelium and the three kingdoms Animalia, Fungi, and Plantae by a maximum-likelihood method using 19 different protein data sets. It was shown at the significance level of 1 SE that the branching of Dictyostelium antedates the divergence of Animalia and Fungi, and Plantae is an outgroup of the Animalia-Fungi-Dictyostelium clade.

    Notizen: Abstract The evolutionary relationships of four eukaryotic kingdoms—Animalia, Plantae, Fungi, and Protista—remain unclear. In particular, statistical support for the closeness of animals to fungi rather than to plants is lacking, and a preferred branching order of these and other eukaryotic lineages is still controversial even though molecular sequences from diverse eukaryotic taxa have been analyzed. We report a statistical analysis of 214 sequences of nuclear small-subunit ribosomal RNA (srRNA) gene undertaken to clarify these evolutionary relationships. We have considered the variability of substitution rates and the nonindependence of nucleotide substitution across sites in the srRNA gene in testing alternative hypotheses regarding the branching patterns of eukaryote phylogeny. We find that the rates of evolution among sites in the srRNA sequences vary substantially and are approximately gamma distributed with size and shape parameter equal to 0.76. Our results suggest that (1) the animals and true fungi are indeed closer to each other than to any other “crown” group in the eukaryote tree, (2) red algae are the closest relatives of animals, true fungi, and green plants, and (3) the heterokonts and alveolates probably evolved prior to the divergence of red algae and animal-fungus-green-plant lineages. Furthermore, our analyses indicate that the branching order of the eukaryotic lineages that diverged prior to the evolution of alveolates may be generally difficult to resolve with the srRNA sequence data.

    Notizen: Abstract Two field experiments were conducted in the Jordan Valley to evaluate potato response to N fertigation. Nitrogen as ammonium sulphate was supplied through irrigation water (fertigation) at rates of 0, 35, 70 and 105 mg N l-1. Soil N application treatment equivalent to the fertigation treatment of 70 mg N l-1 was included. 15N labelled ammonium sulphate was used to evaluate the N recovery and utilization efficiency. Yield increased by the N rate. The soil N application gave higher yield than the zero N and lower than the fertigated treatments. The increase in yield was due to the increase in the size of the tubers. The specific gravity was the highest with the zero N. The index ratios of potato tubers were similar with all treatments. The N derived from fertilizers by both tubers and shoots, increased with the N rate regardless of the method of application. The soil application treatments had fertilizer utilization as high as the fertigation treatments and produced total tuber yield not significantly different from that obtained by the fertigation treatment with similar rate. This might be attributed to the poor fertilizer distribution in the root zone in the fine textured soil. The low value of the fertilizer utilization of the plant receiving the 15N in the preceding season suggested possibilities of rapid transformation and immobilization by the soil microorganisms.

    Notizen: Abstract The potato (Solanum tuberosum) genome contains a highly heterogeneous population of Ty1-copia group retrotransposons. Here we identify the first such transposable element known to be transcribed in this species. The elements are transcriptionally activated during protoplast isolation. The majority of the activated Ty1-copia sequences are similar to elements which are transcriptionally induced under the same conditions in tobacco (Nicotiana tabacum). We also show that a previously identified potato element M166, which has no known equivalent in tobacco is also transcribed under these conditions. It appears that the control of transcription of this particular Ty1-copia group retrotransposons has been broadly conserved between these two species.

    Notizen: Abstract An algorithm was developed for the segmentation and tracking of piglets and tested on a 200-image sequence of 10 piglets moving on a straw background. The image-capture rate was 1 image/140 ms. The segmentation method was a combination of image differencing with respect to a median background and a Laplacian operator. The features tracked were blob edges in the segmented image. During tracking, the piglets were modelled as ellipses initialised on the blobs. Each piglet was tracked by searching for blob edges in an elliptical window about the piglet's position, which was predicted from its previous two positions.
