Beschreibung: We have used 19.9 million papers over 5 decades and 2.1 million patents to demonstrate that teams increasingly dominate solo authors in the production of knowledge. Research is increasingly done in teams across nearly all fields. Teams typically produce more frequently cited research than individuals do, and this advantage has been increasing over time. Teams now also produce the exceptionally high-impact research, even where that distinction was once the domain of solo authors. These results are detailed for sciences and engineering, social sciences, arts and humanities, and patents, suggesting that the process of knowledge creation has fundamentally changed.〈br /〉〈span class="detail_caption"〉Notes: 〈/span〉Wuchty, Stefan -- Jones, Benjamin F -- Uzzi, Brian -- New York, N.Y. -- Science. 2007 May 18;316(5827):1036-9. Notizen: A novel procedure for the numerical modelling of current transport in semiconductor devices is presented. The method is based on high-order trigonometric expansions (Fourier series) of the solution. The expansion coefficients are calculated in a Galerkin-type algorithm. The method offers infinite-order accuracy regardless of the number of spatial dimensions of the model. Well-conditioning and diagonal dominance of the discrete equations render the numerical procedure stable and effective. Significant advantages are expected, particularly for the solution of strongly non-linear multidimensional device models. Properties of the algorithm are demonstrated on standard semiconductor devices.

Notizen: This paper introduces a simple model for solving a system of first-order rate equations using the TLM numerical method. A general algorithm is developed by setting up a matrix of discrete compartments. The rate constants provide the linkage between each compartment and the neighbouring ones. The method is demonstrated by some examples in which saturation conditions are considered. The TLM solution is compared with analytical solutions and the Rung-Kutta method of solving differential equations.

Notizen: A simulation of power transformers following switching operations is presented in which each core section is represented by a non-linear inductance, the differential inductance being determined from the flux density in that section of the core. In analysing the resultant circuit model for the transformer it is shown that the circuit equations have dyadic properties which can be used to give a more efficient numerical solution.

Notizen: An approach, based on the finite element method (FEM), is used for evaluating the electromagnetic field distribution in a class of E-plane metal insert filters, operating in the Ka-band. Artificial boundary conditions are inserted directly in a functional, enabling a solution of the scalar wave equation by finite element method. The scattering parameters of the filter, the electromagnetic field and, therefore, the current distributions on the thin metal inserts are computed. Then, the power dissipated in the metallic septa, which is one of the major limitations for the maximum CW-power that can be handled by these types of filter, is evaluated using a perturbational approach. Finally, the maximum field intensity is determined, allowing an estimate of the maximum peak power that these structures can handle.

Notizen: The multigrid technique, which is mainly used in the finite difference method, is applied to the TLM (transmission-line matrix or modeling) procedure. In the multigrid TLM, the field region is covered with sets of regular transmission-line matrices of different mesh sizes. The finer grid pattern, which overlaps the coarser grid, fills the region with high field gradients. Data obtained in the coarser grid are transferred to the finer grid through an interpolation process and used as boundary conditions for the latter. The finer grid data are then transferred back to the coarser one, thus improving the coarse grid values. The method is applied to several diffusion problems and significant improvements in accuracy and efficiency are shown.

Notizen: Numerical quadratures encountered in solving integral equations and in finite element analysis often involve singular integrands, or integrands with very rapid local variation whose numerical stability resembles that of singularities. It is shown that specialized quadrature formulae of Gauss-Christoffel type can be generated for such integrands, using a recursive procedure based on the properties of the underlying orthogonal polynomials. Quadrature formulae of moderately high degree can be computed rapidly enough to allow them to be constructed as needed. An algorithm for generating quadrature formulae is given in detail. Singular or near -singular functions encountered in finite element analysis typically require three -point or four -point quadratures; such formulae are readily obtained, to seven -figure accuracy, in computing times short enough to regard the formulae as disposable and not worth preserving in tables.

Notizen: This paper presents an overview of the 3D transmission-line matrix method of analysis, describing the various nodes which are available to date: expanded node, asymmetrical condensed node, symmetrical condensed node. The most general scattering matrices of these nodes are given. The various techniques used to implement irregularly graded meshes are also described. A comparison of all the processes applied to the same structure is made. Conclusions on their use are then discussed.

Notizen: A new TLM-based concept analogous to the Green's function approach in classical electromagnetic theory is presented. It employs the procedure known as time domain diakoptics. The response of a TLM mesh to a unit impulse excitation at selected input points is interpreted as a discrete Green's function in the time domain, and the term ‘Johns matrix’ is proposed for this characteristic response. As in classical theory, the response of the mesh to an arbitrary excitation is found by convolving the excitation with its Johns matrix. This concept extends the generalized scattering parameter concept into the time dimension, opens unprecedented possibilities for partioning time domain problems at the field level, and permits large-scale preprocessing of substructures for computer-aided design. It also represents and elegant way of modelling broadband absorbing boundary conditions, imperfectly conducting walls and general frequency dispersive boundaries in the time domain.

Notizen: The paper outlines the operating principles of a 3 -phase to single -phase cycloinverter, as used in heating and melting applications, and presents a digital computer model on which performance predictions may be based. Ideal power switches, an input filter and a compensated heating load are assumed in formulating an initial set of state -variable equations which are valid when all the switching devices are conducting. Connection matrix techniques are then used to generate the mesh equations relevant to the continually changing pattern of the switching devices, leading to a very versatile simulation program. Experimental results from a laboratory -scale unit are presented for different output power levels, and these are compared with corresponding computed results to illustrate the close similarity that exists.

Notizen: Rigorous full -wave analysis techniques are presently receiving much attention in the design of MICs and, in particular, of MMICs due to increasing circuit packing densities and structural complexity. In this paper, the problems associated with such techniques and previous related work are briefly outlined in the introduction. To obtain self -consistency, the electrodynamic Green's functions and related terms are then formulated for the shielded (M)MIC problem. The final operator equation for the numerical solutions derived and the associated functions space are presented and discussed. The central portion of the paper describes a new numerical solution using a discretized Green's function database technique. The resulting linear system of equations is solved by direct inversion for geometrical complexities involving up to about 1000 unknowns; for a higher number of unknowns, an iterative solution is generated. As an alternative to the database technique developed, a variety of spectral domain iterative solutions has been written and tested as well. This includes application of the conjugate gradient method to the normal operator equation (CGN algorithm), an implicit iterative Galerkin approximation called the modified planar conjugate gradient technique (MPCG) and monotonically convergent iteration procedure being a version of the conjugate residual algorithm (CR). Supplementary to this, the extraction of (M)MIC design data from the numerical 3D solutions obtained and error considerations are presented. The paper concludes with a variety of analysis examples of medium to high geometrical complexity and with verification of some results by comparison with measurements and with numerical data from other sources.CPU times required on typical workstations (Micro VAX, HP 9000, etc.) are moderate, thus rendering the techniques presented as useful in the solution of MIC and MMIC design problems.

Notizen: This paper presents the dispersion characteristics of the FD-TD algorithm by showing the numerical phase and group velocities of the TEM, TE or TM modal solutions. For the TEM wave propagation, there exist three fundamental directions to which phase and group velocities can be expressed by only a single variable (wave number k or frequency ω). Those special directions were chosen to show the dispersive nature of the FD-TD algorithm of which the group and phase velocities depend explicitly on frequencies. In view of similarities between the TLM and FD-TD algorithms, a comparison of phase velocity characteristics of both methods was made. Under the special condition where the stability factor s is larger than 1/2, the FD-TD algorithm is found less dispersive than that of the original TLM. However, newly developed symmetrical TLM method appears less dispersive than the FD-TD algorithm. In the numerical simulation of waveguide modal solutions, it was found that there exists an optimum frequency which provide least numerical error in the FD-TD application. A procedure, which determines the stability factor s and the maximum allowed frequency Fmax for the best numerical results, is proposed in this paper.

Notizen: The TLM technique has been enhanced to allow the accurate modelling of EM diffusion through advanced composite material shells. This is a significant development owing to the rapidly increasing use of conducting composite materials in aerospace vehicle design. The accurate EM modelling of this class of materials is important for correct design of vehicles with regard to electromagnetic compatibility (EMC).Existing 3-D time-domain TLM models a non-perfectly conducting shell using a resistive sheet (or sheets) to simulate the surface. The results from this model are shown to be slightly inaccurate and require lengthy computer solution times. A separate 1-D, frequency-domain transmission-line model has been developed elsewhere to better analyse the diffusion properties of composite materials. The incorporation of the results from this frequency domain code into the 3-D time-domain TLM algorithm is outlined.

Notizen: The calculation of the radar cross-section (RCS) of complex bodies using the symmetrical condensed TLM method is presented. The technique is based on a near-to-far field transformation of the TLM calculated near fields. Several two-dimensional examples are presented which validate the method. The main advantage of utilizing techniques such as TLM for RCS computation lies in the ability to model arbitrary bodies with complex material compositions.

Notizen: The nature and significance of time-domain approximations needed for a diakoptic solution using transmission-line modelling (TLM) are examined. Truncation, filtering and repeated convolution of time responses result in severe restrictions in the accuracy of time-domain diakoptics. The frequency-domain diakoptic method has several advantages, including speed and simplicity, and warrants further study.

Notizen: A de model for short-channel MOSFETs is presented in this paper. Several second-order effects associated with small-geometry MOSFETs such as mobility degradation, carrier velocity saturation and channel length modulation are included in the model. The analysis emphasizes the modeling of the output conductance and the transconductance which are important in analogue circuit simulation. The theoretical predictions of the model are in good agreement with the experimental data available in the literature.

Notizen: A new P-N junction device model is presented. It consists of two numerical unidimensional macroscopic models. The first is a drift-diffusion model. The second is an energy model accounting for the carrier energy relaxation effects. Because device optimization requires systematic investigations, the computing time is of paramount importance and, so an accurate comparison between these two models was undertaken under both static steady-state and dynamic 94 GHz conditions in order to point out the relative interest of the energy model. This study is mainly devoted to silicon millimetre-wave IMPATTs. This comparison has highlighted the superiority of the energy model for the description of low field transport phenomena. But as IMPATT RF properties are mainly governed by high field transport, the RF power levels predicted by the two models have been found to be similar. Moreover it has shown that the energy model tends to underestimate both the efficiency and the diode terminal negative resistance level. This parasitic effect is inherent to the description of the carrier generation process by impact ionization which in this model is assumed to be governed by the carrier energy.

Notizen: The response of capacitive array sensors in the presence of flawed solid materials is simulated using finite elements and infinite elements with exponential decay. Conventional finite elements are used to model the critical regions near the probe and the surface of the solid. Infinite elements are used to represent the farfield conditions of the space surrounding the probe and the solid. The method is first applied to problems with analytic solutions to determine the accuracy of the results obtained using the infinite elements. The response of a capacitive array sensor is then simulated using a line integral which measures the relative change in admittance between flawed and unflawed solids. Examples of capacitive probe responses are given for several parametric variations of the flaw size and dielectric constant of the solid.

Notizen: Biological mineralization is a cell-mediated process which is believed to be triggered by a “nucleating agent.” Various matrix structures, such as matrix vesicles, collagen fibrils and macromolecules, have been claimed to be the source of this substance, since these components have been found by transmission electron microscopy (TEM) of thin sections to be associated with early mineral crystals. Systematic image analysis of the relationships revealed in electron micrographs between specific matrix components and early mineral deposits has shown that unequivocal image interpretation is not possible. This is due principally to the problems posed by overprojection and truncation phenomena, since the structures being analyzed lie within the same dimensional range as thin section thickness. Various examples are illustrated and discussed. The site at which mineral crystals are initially laid down thus cannot be identified with any matrix structure using thin section TEM. Possible technical approaches to resolve this problem of image analysis are discussed.

Notizen: A computer graphics system for reconstruction from serial section micrographs was applied to intracellular details of a yeast target cell (Saccharomyces cerevisiae cell) induced by the α factor mating pheromone and was also applied to a periodontal structure of a dog tooth moved orthodontically. In the former, intracellular organelles and a distribution of vesicles could be clearly observed through the cell membrane using the transparent display method in which the smoothing of the reconstructed outer cell membrane surface by computer processing was applied to the transparent display. In the latter case, by cutting through a reconstructed dog tooth and its periodontal tissues, labiolingual and mesiodistal cut surfaces of the tooth and of adjacent alveolar bone could be observed with fine details (232 sections were used).

Notizen: In order to determine the best conditions to carry out quantitative ultrastructural studies in plant specimens, five different fixation techniques, including some of the most reported electron microscopy fixatives (glutaraldehyde-paraformaldehyde, osmium tetroxide, potassium permanganate), were assayed in onion root meristems to check their ability to induce morphometric changes in Golgi apparatus ultrastructure. Although the parameters evaluated showed in all cases the same tendency, values obtained after permanganate fixation were always higher than those found after aldehyde techniques (especially aldehyde-osmium). Aldehyde followed by osmium fixation appears as the most indicated fixation method when accurate quantitative ultrastructural studies are to be developed.

Notizen: Optimal imaging of complex structures requires proper alignment relative to the optic axis of the electron microscope. This is especially important for high-voltage and intermediatevoltage microscopes, which form an in-focus image throughout the entire thickness of the object. As a result, structures at different specimen heights form overlapping and confused images that severely curtail the usefulness of these instruments. The work described here provides a generalized, flexible method for optimizing specimen orientation and eliminating or limiting image overlap by means of a commonly used double-tilt stage. Analysis of the motion about the two axes provides accurate tilting for any azimuthal direction whether or not it corresponds to a mechanical axis of the stage. An object can be positioned to minimize image overlap, to record stereopairs for any parallax axis, and to record three-dimensional data sets by the conical collection geometry.Images of muscle paracrystals are shown after tilting about an axis perpendicular to a symmetry direction. The tilted image displays higher-order symmetry, which is altered by changes of one degree. Precision double-tilting for optimizing stereopairs is shown for a desmosome recorded using different parallax axes and pretilts. A tomographic conical data-collection scheme is demonstrated by imaging a microtubule axoneme for a specific cone half-angle and arbitrary azimuthal angles.

Notizen: The relative sensitivity of an analytical electron microscope and energy-dispersive x-ray detector to x-rays of various elements is investigated through an extensive kASi factor study. Elemental standards, primarily National Bureau of Standards multielement research glasses, were dry-ground into submicrometer-sized particles and analyzed at 200 kV accelerating potential. The effect of self-absorption of x-rays by the particle has been corrected for, allowing the experimental kASi factors from this study to approximate those that could be obtained from “infinitely thin” specimens.Whenever possible, elemental k-factors were determined by the analysis of many (up to a maximum of nine) different standard materials. Experimental kASi factors were calculated for a wide range of Kα, Lα, and Mα x-ray lines. For comparison, theoretical kASi factors, employing a variety of ionization cross sections, were computed. Good agreement is obtained between several of the theoretical k-factor models and the experimental results. Mass volatilization of Na and K from the small glass particles during analysis is discussed, as are observations that the grinding and/or dispersing of standard materials in a liquid (such as ethanol) may promote leaching of certain elements from the particle matrix.

Notizen: Experimental conditions for obtaining the optimum signal-to-background (S/B) ratio in reflection electron energy-loss spectroscopy (REELS) are investigated. It is shown that the S/ B ratio can be improved by lowering the incident energy of the electrons. The spectra taken from the GaAs (660) specular reflection spot under the surface resonance condition is demonstrated to have the best S/B ratio and lowest surface plasma excitation, which is capable of providing structural information on the top few atomic layers.

Notizen: C-peptide and insulin antigenic sites in rat pancreatic β cells were both labeled by the protein A and immunogold techniques. Gold bead density in the secretory granules was correlated with granule size distribution. Our results demonstrate that the distribution patterns of antigenic sites for C-peptide (which also represents proinsulin sites) and insulin are similar when granule size distribution is correlated to the density of antigenic sites. It was found that the ratio of C-peptide to insulin in granules is independent of granule size. Examination of frequency histograms by a Kolmogorov-Smirnov analysis excluded the possibility that the antigenic sites of C-peptide and insulin are distributed in the same manner. Mean gold bead density for insulin antigen (97 ± 24) was slightly higher than C-peptide density (64 ± 18). The efficiency of labeling was found to be very low; less than 0.1% of the theoretically estimated antigenic sites were labeled.

Notizen: We designed and built a freeze-drying device that ensures the protection of the specimens against contaminants during mounting on the cold stage of the freeze-fracture machine, transferring into the vacuum chamber and deep etching. The device consists of a copper cap that covers the specimen and a thermal connection that ensures thermal transfer between the microtome arm and the copper cap. This device was used to study the ultrastructural features of the erythrocyte membrane skeleton and the immunocytchemical localization of spectrin in an “in situ” approach, by freeze drying and platinum rotary shadowing. Human erythrocytes adhered to polylysine-coated coverslips and were broken by a stream of buffer that mimics the intracellular ionic environment (“inside buffer”). The samples were prefixed in periodate-lysine-paraformaaldehyde fixative, labeled with antispectrin 5-nm gold particles, fixed in glutaraldehyde, mordanted in tannic acid, postfixed in OsO4, repeatedly washed in water, rinsed quickly in 30% ethanol, freeze-dried, and rotary-shadowed. Electron microscopic examination of the replicas revealed the skeletal network on the inner surface of the erythrocyte membrane. Immunocytochemical labeling proved that spectrin represents a fibrillar component of the network. Our data confirm the speculative model of the molecular organization of the erythrocyte skeleton, based on studies on in vitro association of proteic constituents. Both the technique and the device developed by us may lead to a deeper understanding of the spatial organization of the cytoskeletal network of more complex cell types.

Notizen: The use of osmium-potassium ferrocyanide as the secondary fixative greatly improved chondrocyte preservation and stabilized the cartilage matrix proteoglycan. The proteoglycan was similar in appearance to that seen following fixation in the presence of cationic dyes. Extensive glycogen preservation was noted in these cells, occupying the area prior to and during the formation of the secondary center of ossification. The volume and organization of the glycogen within the cell cytoplasm were greater than that following buffered osmium fixation, and the cellular vacuoles within were greatly reduced. The cells forming the secondary center prior to the onset of mineralization were of greatest interest, because other studies compared them with the primary growth plate and described them as showing signs of hypertrophy as early as 5 days postnatally, as is found in the primary growth plate. Our observations indicate that glycogen is present in these cells, and cellular enlargement was not present. The cells do not resemble the hypertrophic chondrocytes of the primary growth plate, as far as cytoplasmic content is concerned, and we suggest that they may contribute to the development of the secondary center in a different fashion.

Notizen: A cross-sectional sample preparation technique is described that relies on lithographic and dry-etching processing, thus avoiding metallographic polishing and ion milling. The method is capable of producing cross-sectional transmission electron microscopy samples with a large amount of transparent area (1 μm × 2.5 mm) which allows the examination of many patterned test sites on the same sample from the same chip of a silicon wafer. An example of the application of the technique is given for localized oxidation through a mask.

Notizen: A magnetic-field-superimposed field emission gun with low aberrations and equipped with a zirconiated tungsten emitter has been developed for applications where very stable high probe currents are required. It has been tested on a conventional electron microscope at 10 kV and on an electron beam testing system at 1 kV. Probe current i = 250 nA in a probe size d = 0.4 μm is obtained at 10 kV; at 1 kV the resolution is 0.1 μm with i = 5 nA, and 0.4 μm with i = 30 nA. For these probe currents, the spatial broadening effect due to electron-electron interactions in the beam is the preponderant factor limiting the probe size.

Notizen: Experiments on automating the transmission electron microscope rely on the search for minimum variance. This image parameter gives satisfactory results for automatic focusing, astigmatism correction, and beam alignment. We investigate here the different image descriptors that might also be used; we conclude that texture parameters, which are directional, would be better candidates correcting astigmatism and beam tilt.

Notizen: The properties of oxides may be drastically influenced by their microstructures. Thus the knowledge of nonstoichiometric mechanisms is an essential tool for the conception of new inorganic compounds by solid-state chemist. Through three chosen examples of complex oxides with perovskite-related structure, these points are illustrated. The new families of phosphate tungsten bronzes, the antiferromagnetic AMn1 - x FexO3 - y, and the copper oxides LCuO3 - x (La = Ba, Y, La) were studied by high-resolution electron microscopy (HREM) in order to analyze the mechanisms of nonstoichiometry, the defects of which can modify their physical properties (magnetism or electronic conductivity) and the factors that govern their stabilization.

Notizen: The applications of electron diffraction and diffraction contrast electron microscopy with which to study the structure and dynamics of organic thin films are discussed. The techniques of making thin film deposits on substrates and of forming free-standing thin films over holes on the substrate are described. Selected area electron diffraction and diffraction contrast imaging techniques for thin film studies are elaborated, and examples are given. Methods to reduce radiation damage and environmental protection of the thin film specimen are outlined. The interpretation of electron diffraction and imaging data is given for three cases: (1) The effects of film tilting and molecular tilting (with respect to the film plane) are examined. (2) The detection of phase transition is illustrated. (3) The use of labels to mark film domains is shown together with the measurement of dynamic movement.

Notizen: Cytoskeletons underneath the postsynaptic membrane of neuromuscular junctions were studied by using a quick-freeze deep-etched method and immunoelectron microscopy of ultrathin frozen sections. In a quick-freeze deep-etched replica of fresh, unfixed muscles, 8.9 ± 1.5-nm particles were present on the true postsynaptic membrane surface. Underneath this receptor-rich postsynaptic membrane, networks of fine filaments were observed. These cytoskeletal networks were more clearly observed in extracted samples. In these samples, diameters of the filaments which formed networks were measured. In the platinum replica, three kinds of filament were recognized - 12 nm, 9 nm, and 7 nm in diameter. The 12-nm filament seemed to correspond to the intermediate filament. The other two filaments formed meshworks between intermediate filaments and plasma membrane. In ultrathin frozen sections vinculin label was localized just beneath the plasma membrane. Thirty-six percent of the label was within 18 nm from the cytoplasmic side of the plasma membrane and 50% was within 30 nm. Taking the size of the vinculin molecule into account, it was concluded that vinculin is localized just beneath the plasma membrane and might play some role in anchoring filaments which formed meshworks underneath the plasma membrane.

Notizen: A scanning electron microscope of ultra-high-vacuum (UHV-SEM) with a field emission gun (FEG) is operated at the primary electron energies of from 100 eV to 3 keV. The instrument can form the images that contain information on surface chemical composition, chemical bonding state (electronic structure), and surface crystal structure in a microscopic resolution of several hundred angstroms (Å) using the techniques of scanning Auger electron microscope, scanning electron energy loss microscope, and scanning low-energy electron diffraction (LEED) microscope. A scanning tunneling microscope (STM) also has been combined with the SEM in order to obtain the atomic resolution for the solid surface. The instrumentation and examples of their applications are presented both for scanning LEED microscopy and STM.

Notizen: Four kinds of works on the detection of displacement of atoms in crystals are shown. The irregular small displacement of atoms has been detected, with an accuracy of about 0.1 Å around dislocations and stacking faults in Au crystals as shown by their electron microscope images. The displacement of the atomic images is recorded by aberration-free focus (AFF). Even when the periodic displacement of atoms in SiC and TaS2 crystals is around 0.1% of the lattice constant, this displacement has been revealed as the weak-contrast anomaly in the images. Using in situ observations by a TV system attached to an electron microscope, the rapid movements of atoms that have taken place within 1/30 sec have been recorded. Using the technique of successive subtraction of the images by TV system and image sigma, only the images of moving atoms in Au crystal have been recorded each 1/10 sec.

Notizen: The distribution of neuropeptides exhibits pronounced interspecies heterogeneity. Neuropeptides may function as hormones secreted from chromaffin cells or as neurotransmitters/neuromodulators released from nerve terminals. However, other possible functions such as trophic or intracellular effects should also be considered. Thus, to understand the role of neuropeptides, it is important to explore their localization in different species. The distribution of enkephalins, neurotensin, neuropeptide Y, calcitonin gene-related peptide, and galanin in the adrenal medulla of rat, cat, hamster, and mouse is presented in detail.

Notizen: Manufacture of food products with desired texture and stability increasingly benefits from an understanding of the food microstructure. Freeze-fracture is advantageous in gaining this understanding because it avoids alterations of hydration, retains fat, and maximally preserves air cells. This paper reviews insightful applications of freeze-fracture to various food crystals, colloids, gels, emulsions, and foams.

Notizen: Understanding the relationship between the molecular structure and the macroscopic properties of polymer solutions and gels, oil-water-surfactant emulsions, lyotropic and thermotropic liquid crystals, colloidal dispersions, detergents, and other such “microstructured fluids” is essential to the optimal use of these commercially important materials. Modern rapid-freezing methods followed by freeze-fracture replication techniques are ideally suited to allow the direct visualization of the three-dimensional structure of the particles or units that make up the dispersion, while simultaneously revealing their orientation and distribution with molecular resolution. This paper reviews the necessary experimental conditions required to successfully exploit the freeze-fracture technique as it applies to microstructured fluid systems. The benefits and limitations of structural studies by freeze-fracture techniques as opposed to the more commonly used light, X-ray, and neutron-scattering methods are discussed. Freeze-fracture replicas can also be imaged by scanning tunneling microscopy to reveal directly three-dimensional fracture contours with improved resolution.

Notizen: In this paper, we present a technique for analysis of composition gradients, using an analytical electron microscope, within the primary phase of a two-phase alloy for the case where the second-phase particle size is similar to the size of the irradiated volume. If the composition difference between the two phases is large, the detected compositional fluctuations associated with varying phase fractions may mask any underlying composition gradient of the primary phase. The analysis technique was used to determine grain boundary chromium concentration gradients in a nickel-base superalloy, alloy X-750. The technique may also be of use in other alloy systems.

Notizen: An application of high-resolution transmission electron microscopy (HRTEM) to the study of two types of dislocations in potassium feldspars (K-feldspars) is shown here. HRTEM images were submitted to a filtering in order to improve their interpretation. One type of dislocation - (010)[001]/2 - appears to be dissociated, with (010)[001]/2 planar defect, whereas the other one - (010)[110]/2 - is perfect. This result helps provide an understanding of dislocation activation in this material.

Notizen: The history of high-resolution imaging of organic crystals is summarized, followed by a discussion of radiation damage which reflects current thinking. Methods of specimen preparation and the experimental techniques for high-resolution imaging are described. The results show the application of these techniques to aromatic hydrocarbons, phthalocyanines, and linear-chain aliphatic molecules. Artefacts and image interpretation are also discussed.

Notizen: With the aid of epitaxial orientation techniques originally designed for linear polymer crystallization, it is found that a large assortment of linear chain molecules can be prepared for electron diffraction study in a projection onto the molecular axes. This not only facilitates a study of ordered monodisperse molecular crystal structures but also of the disordered state as well, including thermotropic phase transitions and the structure of binary solids. Representative studies of monodisperse and polydisperse phase behavior based on electron diffraction and differential scanning calorimetry measurements are reviewed for n-paraffins, glycerolipids, and cholesteryl esters. The importance of observing the microcrystalline state is readily apparent from these studies - not only because the symmetry of individual small crystals can be determined, but also because local structural variations not detectable in bulk measurements are readily observed.

Notizen: The crystal imperfections in thin films of lanthanide phthalocyanines (LnPc2H, Ln = Nd, Tb, Er, Tm, Yb, and Lu) grown epitaxially on KCl have been ovserved by molecular imaging. Grain and twin boundaries, stacking faults, point defects, vacancies, mosaic structures, and sometimes even some amorphous islands exist in the well-crystallized specimens. Combined with the results reported earlier, the packing characteristics of planar LnPc2H molecules can be well understood.

Notizen: Atomic-level details are easily resolved in the latest generation of intermediate voltage electron microscopes, but structural information on the same scale can only be extracted under certain specific conditions. Some understanding of imaging theory, as well as an awareness of correct operating conditions, is required for reliable image interpretation. Several representative examples are chosen to illustrate the possibilities for atomic-resolution imaging of materials, and perspectives and outlook for the technique are briefly discussed.

Notizen: Lactoferrin, a DNA-binding protein, was complexed to colloidal gold and applied on ultrathin sections of resin-embedded plant tissues and bacterial cultures. Optimal results were obtained when lactoferrin was tagged to colloidal gold particles at pH 9.2. Postfixation with osmium tetroxide and embedding with Epon did not prevent the accessibility of the protein towards its corresponding binding sites. In plant nuclei, labeling was observed over the dense chromatin and to a lesser extent over the dispersed chromatin. Nucleolar labeling was preferentially located over the dense fibrillar component. Gold particles were also found to be associated with chloroplasts and mitochondria. In prokaryotic cells, a dispersed labeling was noted over the cytoplasm and, in some cases, the aggregation of few gold particles suggested the presence of packed DNA fibrils. Various control experiments confirmed the specificity of the labeling pattern obtained. Lactoferrin-gold complex appears to be a valuable probe for the intracellular demonstration of DNA molecules in double-fixed and Epon-embedded tissues.

Notizen: A technique for performing light, scanning, and transverse transmission electron microscopy on cultured cells grown within a single tissue culture flask is described. Permanent light microscopy slides are obtained by removing selected portions of the plastic tissue culture vessel and mounting them on glass slides with an aqueous mounting solution. The images obtained from these slides are superior to viewing through the bottom of the flask with an inverted stage microscope. For scanning electron microscopy, selected areas are also cut from the remainder of the vessel and prepared for viewing. The final portion of the culture container is transferred and attached to a new tissue culture vessel and prepared for transmission electron microscopy using alcohol instead of acetone and propylene oxide during dehydration, infiltration, and embedding.

Notizen: The FORTRAN source code is given for a computer program that calculates the two-dimensional intensity distribution in convergent-beam transmission electron microdiffraction (CBED) patterns from perfect crystals. The program uses the eigenvalue or Bloch-ware method. It allows three-dimensional dynamical diffraction, and so includes all higher-order Laue zone effects without approximation. No symmetry reduction is included. The program accepts noncentrosymmetric or centrosymmetric crystal structures and allows absorption corrections to be included. It uses the “EISPACK” subroutines for the diagonalisation of a general complex matrix. Up to 100 CBED disks may be included. The code is also available via “Bitnet”.
