    Description: The year 2017 marks the 20th anniversary of the molecular cloning of the long sought-after capsaicin receptor, now known as TRPV1 (Transient Receptor Potential Vanilloid 1). This seminal discovery has opened up a “hot” new field of basic research and launched drug discovery efforts into the large family (by the latest count, 28 mammalian members and 27 in humans) of TRP ion channels. Indeed, it took less than a decade for the first potent, small molecule TRPV1 antagonists to enter phase 1 clinical trials, closely followed by TRPA1 and TRPM8 antagonists. The literature on TRP channels is immense. TRPV1 alone is a keyword in over 5000 publications searchable in PubMed. Clearly, it is not possible to capture the entire literature in a single thematic issue. Consequently, the selection of articles presented in this book represents a sampling of the literature, and is admittedly subjective. We tried to survey the wide range of human diseases in which TRP channels have been implicated, ranging from chronic pain through asthma and diabetes to cancer, and highlight the channels that appear to hold the greatest promise for therapeutic targeting. With this book, we hope to convince readers that TRP channels constitute a formidable family of potential therapeutic targets that will likely continue to demand attention.

    Description: Resilience has become an important topic on the safety research agenda and in organizational practice. Most empirical work on resilience has been descriptive, identifying characteristics of work and organizing activity which allow organizations to cope with unexpected situations. Fewer studies have developed testable models and theories that can be used to support interventions aiming to increase resilience and improve safety. In addition, the absent integration of different system levels from individuals, teams, organizations, regulatory bodies, and policy level in theory and practice imply that mechanisms through which resilience is linked across complex systems are not yet well understood. Scientific efforts have been made to develop constructs and models that present relationships; however, these cannot be characterized as sufficient for theory building. There is a need for taking a broader look at resilience practices as a foundation for developing a theoretical framework that can help improve safety in complex systems. This book does not advocate for one definition or one field of research when talking about resilience; it does not assume that the use of resilience concepts is necessarily positive for safety. We encourage a broad approach, seeking inspiration across different scientific and practical domains for the purpose of further developing resilience at a theoretical and an operational level of relevance for different high-risk industries. The aim of the book is twofold: 1. To explore different approaches for operationalization of resilience across scientific disciplines and system levels. 2. To create a theoretical foundation for a resilience framework across scientific disciplines and system levels.     Description: Drug metabolism comprises the identification, characterization, and quantification of the chemicals or compounds produced in an animal or human upon administration of a drug. Research practices not only require the chemical structure but also aim to determine the pharmacological activities and/or toxicity of these compounds. This is first performed in animals, as studies attempt to identify and quantify metabolites, and later in humans, with care to further characterize metabolites that are either unique to or produced disproportionately in humans compared to animals. Characterization includes the determination of enzyme systems or other biological mechanisms that produce each identified metabolite; this information is used to predict potential drug-drug interactions with other compounds that increase or decrease metabolite formation and sources of biological variability in response or toxicity with varying patient genetics, which affect CYP isoform expression. This book’s purpose is to provide some understanding of the biology and current technology applied in the field of drug metabolism.

    Description: The Cross Domain City of the Future Graduation Lab, situated in the Faculty of Architecture and the Built Environment at TU Delft, has been a pioneer in experimenting with a multidisciplinary approach to education on the built environment. Drawing upon this expertise over the past years, this book reflects on multidisciplinarity in the built environment and its implementation in education on the built environment. How should one approach multidisciplinarity in education and practice? What encompasses its core elements, benefits, and challenges? By addressing these questions, the book aims to inform students and practitioners within the realm of the built environment by sharing insights from experiences in multidisciplinary education. It presents eight conclusions regarding the future of multidisciplinary education and, thereby, seeks to contribute to a more humane and sustainable future for cities: 1.Process is central to multidisciplinary collaboration. Negotiating positions, ensuring an environment of respect, balance and open-mindedness, and setting a common vocabulary. 2.Multidisciplinarity can be a way to foster innovation. It triggers complementarity and confrontation. As with any innovation, there is potential for greater outcomes, but, at the same time, extra risks emerge. These need to be managed. 3.Multidisciplinarity could be better integrated into organisational structures. 4.Disciplinarity and multidisciplinarity are in mutual coexistence. They are inseparable. They can complement and contradict each other. 5.Problem precedes solution, not the opposite. Framing the problem, or ‘problematizing’, is a considerable share of the actual solution. This is particularly applicable to multidisciplinarity. 6.Multidisciplinarity is by nature composed of fluid boundaries. Navigating through an enormous diversity of perspectives requires agility, flexibility, independence, spirit of adventure and embracing uncertainty. 7.Professionals should be trained as ‘T-shape’: grounded in their field while able to dialogue with other fields. 8.Both generalists and specialists are needed. Education should provide opportunities for both.

    Description: COVID-19 is a rampant worldwide problem. It is caused by the SARS-CoV-2 virus and is manifest in different variants. The Delta variant compromised existing therapeutic and preventive options for this disease and is beginning to be replaced by the Omicron variant. Through pharmaceutical biotechnology, three different treatment approaches to COVID-19 have been developed: computer-aided drug design (CADD); rational drug design in the wet lab; and the advanced drug delivery system. These approaches are heavily influenced by advances in life sciences, such as the development of structural bioinformatics, the establishment of nanobiotechnology as a standard approach in drug design, and major advances in structural biology such as the development of the CryoEM method.     Description: This open access book contains a structured collection of the complete solutions of all essential axisymmetric contact problems. Based on a systematic distinction regarding the type of contact, the regime of friction and the contact geometry, a multitude of technically relevant contact problems from mechanical engineering, the automotive industry and medical engineering are discussed. In addition to contact problems between isotropic elastic and viscoelastic media, contact problems between transversal-isotropic elastic materials and functionally graded materials are addressed, too. The optimization of the latter is a focus of current research especially in the fields of actuator technology and biomechanics. The book takes into account adhesive effects which allow access to contact-mechanical questions about micro- and nano-electromechanical systems. Solutions of the contact problems include both the relationships between the macroscopic force, displacement and contact length, as well as the stress and displacement fields at the surface and, if appropriate, within the half-space medium. Solutions are always obtained with the simplest available method - usually with the method of dimensionality reduction (MDR) or approaches which use the solution of the non-adhesive normal contact problem to solve the respective contact problem.

La tendencia del mercado genera la necesidad de conocer la dinámica del comercio exterior, por lo tanto se requiere tener unas bases sólidas que permitan a las compañías dirigirse a mercados externos diferentes a su lugar de origen. A pesar que en Colombia las Pymes representan un alto porcentaje de la actividad productiva, indus-trial y manufacturera del país, la gran mayoría no cuenta con las competencias requeridas para incursionar en el mercado internacional. Según ANIF (2016) en la gran encuesta pyme se identificó que en promedio, aproximadamente, el 80% de las pymes no realizaron exportaciones durante el 2016, ni muestran interés por hacerlo, esto en consecuencia a la desarticulación entre la acade-mia, la industria y el gobierno.

    Novel Therapeutic Targets and Emerging Treatments for Fibrosis (2021)

    Description: For decades we have known that the overgrowth, hardening and scarring of tissues (so-called fibrosis) represents the final common pathway and best histological predictor of disease progression in most organs. Fibrosis is the culmination of both excess extracellular matrix deposition due to ongoing or severe injury, and a failure to regenerate. An inadequate wound repair process ultimately results in organ failure through a loss of function, and is therefore a major cause of morbidity and mortality in disease affecting both multiple and individual organs.Whilst the pathology of fibrosis and its significance are well understood, until recently we have known little about its molecular regulation. Current therapies are often indirect and non-specific, and only slow progression by a matter of months. The recent identification of novel therapeutic targets, and the development of new treatment strategies based on them, offers the exciting prospect of more efficacious therapies to treat this debilitating disorder.This Research Topic therefore compromises several up-to-date mini-reviews on currently known and emerging therapeutic targets for fibrosis including: the Transforming Growth Factor (TGF)-family; epigenetic factors; Angiotensin II type 2 (AT2) receptors; mineralocorticoid receptors; adenosine receptors; caveolins; and the sphingosine kinase/sphingosine 1-phosphate and notch signaling pathways. In each case, mechanistic insights into how each of these factors contribute to regulating fibrosis progression are described, along with how they can be targeted (by existing drugs, small molecules or other mimetics) to prevent and/or reverse fibrosis and its contribution to tissue dysfunction and failure. Two additional reviews will discuss various anti-fibrotic therapies that have demonstrated efficacy at the experimental level, but are not yet clinically approved; and the therapeutic potential vs limitations of stem cell-based therapies for reducing fibrosis while facilitating tissue repair. Finally, this Research Topic concludes with a clinical perspective of various anti-fibrotic therapies for cardiovascular disease (CVD), outlining limitations of currently used therapies, the pipeline of anti-fibrotics for CVD and why so many anti-fibrotic drugs have failed at the clinical level.

    Description: Echolocation has evolved in different groups of animals, from bats and cetaceans to birds and humans, and enables localization and tracking of objects in a dynamic environment, where light levels may be very low or absent. Nature has shaped echolocation, an active sense that engages audiomotor feedback systems, which operates in diverse environments and situations. Echolocation production and perception vary across species, and signals are often adapted to the environment and task. In the last several decades, researchers have been studying the echolocation behavior of animals, both in the air and underwater, using different methodologies and perspectives. The result of these studies has led to rich knowledge on sound production mechanisms, directionality of the sound beam, signal design, echo reception and perception. Active control over echolocation signal production and the mechanisms for echo processing ultimately provide animals with an echoic scene or image of their surroundings. Sonar signal features directly influence the information available for the echolocating animal to perceive images of its environment. In many echolocating animals, the information processed through echoes elicits a reaction in motor systems, including adjustments in subsequent echolocation signals. We are interested in understanding how echolocating animals deal with different environments (e.g. clutter, light levels), tasks, distance to targets or objects, different prey types or other food sources, presence of conspecifics or certain predators, ambient and anthropogenic noise. In recent years, some researchers have presented new data on the origins of echolocation, which can provide a hint of its evolution. Theoreticians have addressed several issues that bear on echolocation systems, such as frequency or time resolution, target localization and beam-forming mechanisms. In this Research Topic we compiled recent work that elucidates how echolocation – from sound production, through echolocation signals to perception- has been shaped by nature functioning in different environments and situations. We strongly encouraged comparative approaches that would deepen our understanding of the processes comprising this active sense.

    Description: In recent years, the long term prognosis of patients with rheumatic disease has dramatically improved, due to the enormous advances in the understanding of the pathogenesis of these diseases that have led to the development of drugs targeting the molecules directly involved in the inflammatory response. The development of these biological drugs has radically changed the therapeutic approach of inflammatory rheumatic diseases, mainly in patients resistant to standard treatment. Also in the field of conventional therapy, many new data have confirmed the effectiveness of “old” drugs, such as hydroxychloroquine, methotrexate and azathioprine, demonstrating several additional beneficial properties for the treatment of Systemic Lupus Erythematosus (SLE), Rheumatoid Arthritis (RA) or other autoimmune diseases. Moreover, immunosuppressant developed to prevent graft rejection such as mycophenolate or tacrolimus have been demonstrated to have an important role in the treatment of rheumatic patients.     Description: The invasive character of a primary cancer is greatly dependent on numerous interactions between tumor cells and their extracellular surroundings. Matricellular receptors are defined as (cell-surface) receptors that bind extracellular matrix (ECM) structural proteins and soluble factors dynamically acting on ECM homeostasis. Matricellular receptors mediate numerous signalings from the extracellular environment to cell nucleus and drive main biological functions that are cell growth, survival and migration. Numerous data from the last decade evidence that matricellular receptors are biosensors that allow to a tumor cell answering to microenvironmental variations, and in this sense they are important contributors to tumor cell malignancy. Matricellular receptors represent thus valuable targets for the development of original anti-cancer strategies. Original reports, bibliographic reviews or hypotheses are welcome to improve the basic knowledge of matricellular receptor properties, their spatio-temporal regulation, the dynamic formation of complex receptors and the impact of such interactions on the invasive properties of tumor cells. Biological, biophysical and pharmacological, as well as in silico contributions will be appreciated.

In addition, absence of clinical trials and other traditional safety mechanisms before medicines are introduced to the wider market results in questionable safe dosage ranges which may produce adverse and unexpected outcomes. Therefore, the use of herbal remedies requires sufficient knowledge about the efficacy, safety and proper use of such products.     Description: Adverse drug reactions (ADRs) have a significant impact on the patient’s health. The purpose of this PhD thesis was to analyse the impact of ADRs on the admissions to the emergency room, using the data obtained from the MEREAFaPS study. Elderly patients resulted the most exposed group to ADRs. Gastrointestinal disorders were the most commonly reported adverse reactions. Acetylsalicylic acid and warfarin were the most reported drugs. 17% of serious ADRs and 32% of the resulting deaths were classified as avoidable. Elderly age was found to be the main risk factor associated with ADRs. It is therefore important to underline the need for a more careful use of drugs in this group of patients.

    Description: In the ancient past, cocoa has been appreciated as a high-calorie food to boost energy in soldiers and for its undefined medicinal and mystical properties. During other times, chocolate has been considered as the forbidden “food of God”: a treasure of pleasure for the mind and the soul. The overall perception of the consumer for chocolate was of a “charming” and appealing food with lots of negative aspects related to high sugar content leading to consider chocolate as “junk food” for its “obesigen” calories. Recently, in association with the renewed interest of nutrition science in alternative source of health-promoting foods and ingredients, a large body of research has been conducted to unravel the pro and cons of cocoa in relation to human health. Epidemiological evidences indicate that cocoa consumption helps preventing cardiovascular disease for its high content in bioactive flavonoids. Clinical trials show that chocolate consumption might improve vascular function, decreasing platelet aggregation and display an antioxidant and anti-inflammatory effect. The putative protective action of cocoa seems to be multi-factorial and involving different aspects of vascular, antioxidant and endothelial function. However, the mechanism(s) that account for the benefits of cocoa it is still unclear. The aim of this Research Topic is therefore to provide the reader with an objective picture of the state of art on the association between cocoa and health, mainly through the evidences of human trials; overwhelmingly considered the golden standard for nutritional science. The Research Topic will cover the analysis of the manufacturing processes of the chocolate and the antioxidant effects in humans as well as the majority of the putative health effects of chocolate and cocoa, such as anti-inflammatory properties, effect on immunity, platelet aggregation, blood pressure, endothelial function and cognitive behavior. Unraveling the functional properties of cocoa will help to understand if the 'food of God' is a primordial gift for the health of mankind.

    Description: G protein-coupled receptors (GPCRs) are integral membrane proteins forming the fourth largest superfamily in the human genome. Many of these receptors play key physiological roles and several pathologies have been associated with receptor functional abnormalities. GPCRs therefore represent important goals for drug design in pharmaceutical companies since they constitute the target of about one third of the drugs currently on the market. However, endogenous GPCRs are most often difficult to study because of a lack of tools to target them specifically and single out their response to physiological or drug-elicited stimulations. Hence, studies mostly focused on recombinant receptors expressed in a variety of cellular models that do not always closely reflect the receptor natural environment and often deal with levels of expression exceeding by far physiological ranges. Recent technological developments combining for example genetically modified animals and advanced imaging approaches have improved our ability to visualize endogenous GPCRs. To date, trailing receptor activation, subsequent intracellular redistribution, changes in signaling cascade up to integrated response to a drug-elicited stimulation is at hand though the impact of a physiological challenge on receptor dynamics remains a major issue. Data however suggest that the receptor may embrace a different fate depending on the type of stimulation in particular if sustained or repeated. This suggests that current drugs may only partially mimic the genuine response of the receptor and may explain, at least in part, their secondary effects. Commonalities and specificities between physiological and drug-induced activation can thus represent valuable guidelines for the design of future drugs.

Yet many sub-Saharan African countries struggle to meet demand. Why is this, and what can be done to reduce energy poverty and further Africa's development? Examining the politics and processes surrounding electricity infrastructure, provision and reform, the author provides an overview of historical andcontemporary debates about access in the sub-continent, and explores the shifting role and influence of national governments and of multilateral agencies in energy reform decisions. He describes a challenging political environment for electricity supply, with African governments becoming increasingly frustrated with the rules and the processes of multilateral donors. Civil society also began to question reform choices, and governments in turn looked to new development partners, such as China, to chart a fresh path of energy transformation. Drawing on over fifteen years of research on Uganda, which has one of the lowest levels of access to electricity in Africa and has struggled to construct several, large hydroelectric dams on the Nile, Gore argues that there is a critical need to recognize how the changing political and social context in African countries, and globally, has affected the capacity to fulfil national energy goals, minimize energy poverty and transform economies.

    Description: In January 2012, the U.S. White House Office of National Drug Control Policy (ONDCP) asked RAND to generate national estimates of the total number of users, total expenditures, and total consumption for four illicit drugs from 2000 to 2010: cocaine (including crack), heroin, marijuana, and methamphetamine. Drug users in the United States spend on the order of $100 billion annually on these drugs (in 2010 dollars). While this total figure has been stable over the decade, there have been important compositional shifts. From 2006 to 2010, the amount of marijuana consumed in the United States likely increased more than 30 percent, while the amount of cocaine consumed in the United States decreased by approximately 50 percent. These figures are consistent with supply-side indicators, such as seizures and production estimates. Methamphetamine consumption rose sharply from 2000 through the middle of the decade, and this was followed by a large decline through 2008. Heroin consumption remained fairly stable throughout the decade, although there is some evidence of an increase in the later years. For all of the drugs, total consumption and expenditures are driven by the minority of users who consume on 21 or more days each month.

    Description: In humans, ascorbic acid is an essential vitamin, anti-oxidant and co-factor of a variety of metal ion-dependent enzymatic reactions. In this review, the transport of L-ascorbic acid is described from food to target cells. Transport of ascorbic acid across the plasma membrane is facilitated by members of the SLC23 family, SLC23A1/SVCT1 and SLC23A2/SVCT2. We present in silico models of these transporters that provide new insights into the structure of the SLC23 family. While SVCT1 is mainly responsible for uptake of ascorbic acid from the intestine into the blood and for reabsorption in the kidney, the more broadly expressed transporter SVCT2 delivers ascorbic acid into tissues that are in high demand of the vitamin. The oxidized form of ascorbic acid, dehydroascorbic acid (DHA), is a substrate of the GLUT transporters belonging to the SLC2 family. They play important roles in ascorbic acid recycling, such as in the brain and in erythrocytes. Ascorbic acid serves as an essential co-factor of metal ion-dependent enzymes, keeping their metal ions in the reduced state. In addition, it serves as an effective antioxidant in cells with high metabolic activity such as neurons. Thus, it is not too surprising that changes in expression and function of the SVCTs have nutritional and pathological consequences such as during ageing, malnutrition and chronic alcohol abuse or in cancer, neurodegenerative diseases and chronic inflammation diseases. In the future, SVCT1 and SVCT2 may furthermore prove useful as drug delivery systems, to enhance transport of novel pharmaceutical agents more efficiently across the intestinal epithelium and the blood-cerebrospinal fluid barrier.

    Description: Chemiebezogene Kompetenzen werden bereits im Sachunterricht der Grundschule entwickelt und sollen im Chemieunterricht der Sekundarstufe I aufgegriffen und weiterentwickelt werden. Ziel dieser Studie war es, die chemiebezogenen Kompetenzen der Schülerinnen und Schüler in der Übergangsphase zwischen Sachunterricht und Chemieunterricht zu analysieren. In einer Ergänzungsstudie wurden zusätzlich sowohl die Auswirkungen der Schulschließungen aufgrund der SARS-CoV-II Pandemie auf die Kompetenzen der Schülerinnen und Schüler als auch der Einfluss einer veränderten Testadministration untersucht. Insgesamt wurden die Kompetenzen von 2262 Lernenden der Jahrgangsstufen 5 bis 9 zu drei Messzeitpunkten analysiert. Die Ergebnisse zeigen, dass die chemiebezogenen Kompetenzen sowohl zum Ende der Grundschulzeit als auch zu Beginn des Chemieunterrichts über alle Kompetenzbereiche hinweg sehr heterogen ausfallen. Dennoch sind sie zu Beginn des Chemieunterrichts in einigen Kompetenzbereichen höher ausgeprägt als zum Ende der Grundschulzeit. Nach den pandemiebedingten Schulschließungen waren die Kompetenzen ähnlich ausgeprägt wie in den gleichen Jahrgangsstufen vor den Schulschließungen. Eine während der Schulschließungen notwendige Bearbeitung der Testhefte von zu Hause aus führte vor allem bei den jüngeren Schülerinnen und Schülern zu besseren Testergebnissen als die Bearbeitung in der Schule. Um die Entwicklung chemiebezogener Kompetenzen in der Übergangsphase zu optimieren, sollte der Erwerb dieser Kompetenzen bereits im Sachunterricht einheitlicher gestaltet werden.
