    Notes: Summary In vitamin A-deficient rats, the glutathione level in lung was diminished and microsomal lipid peroxidation much increased. In vitamin A-loaded animals, however, both were depressed below control. Thus vitamin A protection against lipid peroxidation is independent of glutathione.

    Description / Table of Contents: Zusammenfassung Bei 70 fettsüchtigen Personen (26 Männer, 44 Frauen) wurden im Blutserum einige Lipidkomponenten und der Gehalt an Malondialdehyd (MDA) vor ihrer Abmagerungskur bestimmt. Aufgrund der klinischen Laborbefunde waren diese Patienten bei der Gruppe der Personen mit Hyperlipo-proteinämie einzuordnen (Einteilung nach Fredricksson und Lees). Bei 32 Personen war der Blutcholesteringehalt hoch (über 5,7 mmol/1), es bestand eine negative Korrelation zwischen MDA und hochdichten Lipoproteinfraktionen (HDL). Der größte Korrelationskoeffizient wurde in der Gruppe II/a bei Hyper-lipoproteinämie festgestellt (HDL-C, r=−0,74; HDL-2-C, r=−0,54; HDL-3-C, r=−0,78). Bei Personen mit normalem Serumcholesteringehalt konnte keine Korrelation nachgewiesen werden. Die Ergebnisse werden mit den durch Lipidperoxidation oxidierten Cholesterin-derivaten, welche die HDL-Synthese mindern, erklärt. Es scheint, daß bei Hyper-cholesterinämie der Fettsüchtigen die Lipidperoxidation beim Rückgang des HDL-Wertes eine bedeutende Rolle spielt und damit also einen wichtigen Schutz gegen kardiovaskuläre Erkrankungen bildet.
    Notes: Summary In the blood sera of 70 obese persons (26 men, 44 women) some lipid components and malondialdehyde (MDA) level were determined before slimming. The people were classified into groups of hyperlipoproteinaemia on the basis of laboratory results, according to Fredrickson and Lees. In 32 people with high blood cholesterol level (above 5.7 mmol) there were negative correlations between MDA and high-density lipoprotein (HDL) fractions. Correlation coefficients were the greatest in the group IIa of hyperlipoproteinaemia (HDL-C. r=−0.74; HDL-2-C. r=−0.54; HDL-3-C. r=−0.78). Correlations were not found in subjects with a normal cholesterol level. The results were attributed to the formation of oxidized cholesterol products, caused by lipid peroxidation, which may decrease the HDL synthesis. It seems that in hypercholesterolaemia coupled with obesity, lipid peroxidation can contribute to the reduction in HDL levels, which is an important protective factor against cardiovascular diseases.

    Description / Table of Contents: Zusammenfassung Schweine von 50 kg Körpergewicht wurden vor dem Abschlachten 2 Monate lang ohne Premix (Vitaminzusatz zum Grundfutter) gefüttert. Die Lendenstücke der Schweine zeigten erhöhte antioxidative Fähigkeit, d. h. niedrige Peroxidationsgrade, im Gegensatz zu anderen Schweinegruppen, denen verschiedene Mengen an Premix gefüttert wurden. Der Gehalt an Malonaldehyd in Blutserum und Leber von 7wöchigen Hühnchen, denen hohe Dosen an Salinomycin verabreicht wurden, ist gestiegen. Als Resultat von Premix-Entzug zeigte sich eine abnehmende Tendenz in Lipidperoxidation. Es werden die möglichen Einflüsse der Lipidperoxidation und der Zersetzungsprodukte auf die physikalisch-chemischen Eigenschaften von Fleisch und auf die menschliche Gesundheit diskutiert. Diese Ergebnisse drängen uns, Möglichkeiten einer optimalen Ernährung mit optimaler Premix-Versorgung zu finden, die dem aktuellen Bedarf entspricht.
    Notes: Summary Pigs of 50 kg bodyweight were fed without premix for two months before slaughtering. Their loin samples showed a lower degree of lipid peroxidation (indicated by a higher antioxidative capacity) than other pig groups which had consumed various premix levels. Malonaldehyde concentration increased in the blood sera and livers of 7-week-old chickens treated with a high dose of an ionophoric coccidiostatic agent, Salinomycin. Premix deprivation resulted only in a trend to decrease lipid peroxidation. The possible effects of lipid peroxidation and its degradation products on the physico-chemical characteristics of meat and human health is discussed. The results urge that improper feeding practices should be avoided and optimal premix applications should be found which correspond to the actual requirements.

    Description / Table of Contents: Zusammenfassung Es wurden vor und nach einer 7tägigen Entfettungskur (2,1 MJ/Tag) von 54 Fettsüchtigen stammende Blutproben auf einige Lipidkomponenten (Malondialdehyd [MDA] und die Aktivität der vor freien Radikalen schützenden Enzyme Superoxid-Dismutase [SOD] und Glutathionperoxidase [GSHpx]) untersucht. Nach 7tägiger Krankenhausbehandlung wurde eine Erniedrigung des Körpergeichts, der Triglyceride, des HDL-Cholesterin- und des HDL-3-Cholesterins sowie eine Zunahme von Gesamtcholesterin festgestellt. Nach einer zu Hause fortgesetzten drei Monate dauernden Kur (5,0–6,3 MJ) wurde eine Zunahme von HDL-2-Cholesterin und ein Rückgang von Gesamtcholesterin auf den Ausgangswert beobachtet. Bei an Hyperlipoproteinämie leidenden Vpn. und auch bei denen, die einen erhöhten Cholesterin- und Triglyceridgehalt aufwiesen, wurde eine höhere MDA-Konzentration festgestellt. Nach der Entfettungskur, unabhängig von den Lipiden, fällt die SOD-Aktivität. Die GSHpx-Aktivität ist unverändert, ausgenommen bei Männern mit hohem Cholesterinniveau. Weitere Untersuchungen über die vor freien Radikalen schützenden Enzyme sind erforderlich.
    Notes: Summary In blood samples of 54 obese persons (13 men, 41 women) some lipid components, malondialdehyde (MDA), and the activities of free radical protecting enzymes superoxide-dismutase (SOD) and glutathione-peroxidase (GSHpx), were determined before and after a seven-day slimming diet of 2.1 MJ/day. Body weight, triglycerides, HDL-cholesterol, and HDL-3-cholesterol all decreased, total cholesterol increased after the seven days hospitalization. After continuing the slimming diet (5.0–6.3 MJ/day) at home, HDL-2-cholesterol increased and total cholesterol returned to the initial value after three months. MDA decreased in the groups of hyperlipoproteinemia II/a and II/b and also in the group with high cholesterol and triglyceride levels (above 5.7 and 2.7 mmol/l). Correlation coefficients refer to the dominant role of high TG in the intensity of lipid peroxidation. The activity of SOD fell after the slimming, independent of lipid parameters. GSHpx activity remained unchanged except a drop in the male participants having high cholesterol levels. The behavior of the free radical protecting enzymes needs further examination.

    Notes: Summary Chronic ethanol administration was shown to increase catalase and acyl-CoA oxidase activities in rat myocardium but did not alter the activity of liver peroxisomal enzymes. As a result of alcohol consumption a 2–3-fold increase in the level of lipid peroxidation was observed in the heart tissue while in the liver the induction was much less pronounced.

    Notes: Summary Immunomodulating lipopeptides lauroyl-L-Ala-γ-D-Glu-LL-A2pmNH2-Gly (RP 44.102) and lauroyl-L-Ala-γ-D-Glu-LL-A2pmNH2 (RP 56.142) were found to protect mice against the hepatotoxicity of paracetamol, which is due to cytochrome P-450 dependent formation of toxic metabolites and radicals. In fact they decreased the amount of hepatic microsomal cytochrome P-450, and the level of CCl4-induced lipid peroxidation. In contrast lauroyl-L-Ala-γ-D-Glu-DD-A2pmNH2 (RP 53.204), which only differs by the configuration of the two chiral carbons of A2pm (diaminopimelic acid) and is not an immunomodulating agent, failed to protect against poisoning by paracetamol and had no effect on the level of hepatic cytochrome P-450 or the microsomal CCl4-induced lipid peroxidation. This provides a clear connection between the immunostimulating properties of a compound and its effects on xenobiotic biotransformations.

    Notes: Summary In the course of spontaneous cell-mediated cytotoxicity (SCMC) and antibody-dependent cell-mediated cytotoxicity (ADCC) with human peripheral lymphocytes as effector cells, no lipid peroxidation occurred as measured by the production of ethane and thiobarbituric acid-reactive material. Furthermore, impairment of major cellular defense systems of target cells (K 562 cells for SCMC, Chang liver cels for ADCC), by decreasing their glutathione content, had no effect on either lipid peroxidation or the cytotoxic response.     Notes: Summary We have observed a differential effect of silybin dihemisuccinate on rat liver microsonal oxygen consumption and on lipid peroxidation induced by NADPH-Fe2+-ADP and t-butyl hydroperoxide. These results are ascribed to the antioxidant properties of the flavonoid. The differences observed in the effect of the catalysts may be a consequence of the different capacity of silybin to act as a scavenger of free radicals formed by NADPH-Fe2+-ADP or t-butyl hydroperoxide.

    Notes: Abstract A diterpenoid, totarol (1), fromPodocarpus nagi was evaluated as an antioxidant. This diterpenoid inhibited autoxidation of linoleic acid. Mitochondrial and microsomal lipid peroxidation induced by Fe(III)-ADP/NADH or Fe(III)-ADP/NADPH were also inhibited. Nagilactone E (2), a norditerpene lactone isolated from the same source, had no antioxidative activity. Furthermore, totarol protected red cells against oxidative hemolysis. This diterpene was shown to be effective in protecting biological systems against oxidative stresses.

    Notes: Abstract It has recently been indicated that in the absence of free iron, NADPH initiates oxidative damage of proteins in guinea pig liver microsomes and also lipid peroxidation and protein damage in cardiac microsomes and that ascorbic acid specifically inhibits both the lipid peroxidation and protein damage [Mukhopadhyay CK, Chatterjee IB: J Biol Chem 269: 13390–13397, 1994; Mukhopadhyay Met al.: Mol Cell Biochem 126: 69–75, 1993]. In this paper we demonstrate that Fe(III)-independent NADPH-initiated lipid peroxidation and oxidative damage of proteins occur in the microsomes of all the extrahepatic tissues including lung, kidney, adrenal gland and brain and that both the lipid peroxidation and protein damage are specifically prevented by ascorbic acid. We further demonstrate that when NADPH is replaced by $$O_2^{\bar \cdot } $$ as the electron donor, the $$O_2^{\bar \cdot } $$ lipid peroxidation and protein damage are also inhibited by ascorbic acid.
