    Description: 〈p〉No definitive evidence of spacetime supersymmetry (SUSY) that transmutes fermions into bosons and vice versa has been revealed in nature so far. Moreover, the question of whether spacetime SUSY in 2 + 1 and higher dimensions can emerge in generic lattice microscopic models remains open. Here, we introduce a lattice realization of a single Dirac fermion in 2 + 1 dimensions with attractive interactions that preserves both time-reversal and chiral symmetries. By performing sign problem–free determinant quantum Monte Carlo simulations, we show that an interacting single Dirac fermion in 2 + 1 dimensions features a superconducting quantum critical point (QCP). We demonstrate that the 〈f〉N=2〈/f〉 spacetime SUSY in 2 + 1 dimensions emerges at the superconducting QCP by showing that the fermions and bosons have identical anomalous dimensions 1/3, a hallmark of the emergent SUSY. We further show some experimental signatures that may be measured to test such emergent SUSY in candidate systems.〈/p〉

    Description: 〈p〉Colloidal dispersions with liquid crystallinity hold great promise for fabricating their superstructures. As an example, when graphene oxide (GO) sheets are assembled in the liquid crystalline state, they can turn into ordered macroscopic forms of GO such as fibers via the wet spinning process. Here, we report that by reinforcing intersheet interactions, GO liquid crystals (LCs) turn into mechanically robust hydrogels that can be readily drawn into highly aligned fibrillar structures. GO hydrogel fibers with highly aligned sheets (orientation factor, 〈i〉f〈/i〉 = 0.71) exhibit more than twice the ionic conductivity compared to those with partially aligned structures (〈i〉f〈/i〉 = 0.01). The hierarchically interconnected two-dimensional nanochannels within these neatly aligned GOLC hydrogel fibers may facilitate controlled transport of charge carriers and could be potentially explored as cables for interconnecting biosystems and/or human-made devices.〈/p〉

    Description: 〈p〉With accelerating trends in miniaturization of semiconductor devices, techniques for energy harvesting become increasingly important, especially in wearable technologies and sensors for the internet of things. Although thermoelectric systems have many attractive attributes in this context, maintaining large temperature differences across the device terminals and achieving low–thermal impedance interfaces to the surrounding environment become increasingly difficult to achieve as the characteristic dimensions decrease. Here, we propose and demonstrate an architectural solution to this problem, where thin-film active materials integrate into compliant, open three-dimensional (3D) forms. This approach not only enables efficient thermal impedance matching but also multiplies the heat flow through the harvester, thereby increasing the efficiencies for power conversion. Interconnected arrays of 3D thermoelectric coils built using microscale ribbons of monocrystalline silicon as the active material demonstrate these concepts. Quantitative measurements and simulations establish the basic operating principles and the key design features. The results suggest a scalable strategy for deploying hard thermoelectric thin-film materials in harvesters that can integrate effectively with soft materials systems, including those of the human body.〈/p〉

    Description: 〈p〉Surface-enhanced Raman scattering (SERS) probes based on a charge transfer (CT) process with high stability and reproducibility are powerful tools under open-air conditions. However, the key problem ahead of practical usage of CT-based SERS technology is how to effectively improve sensitivity. Here, a novel ternary heterostructure SERS substrate, Fe〈sub〉3〈/sub〉O〈sub〉4〈/sub〉@GO@TiO〈sub〉2〈/sub〉, with a significant enhancement factor of 8.08 x 10〈sup〉6〈/sup〉 was first synthesized. We found the remarkable enhanced effect of SERS signal to be attributed to the resonance effect of CuPc, CT between GO and TiO〈sub〉2〈/sub〉, and enrichment from a porous TiO〈sub〉2〈/sub〉 shell. In addition, we developed a robust SERS probe with good recyclability under visible light illumination on Fe〈sub〉3〈/sub〉O〈sub〉4〈/sub〉@GO@TiO〈sub〉2〈/sub〉 nanocomposites toward ultrasensitive detection of cancer cells down to three cells. We have now successfully applied this probe for in situ quantification and imaging of programmed cell death receptor ligand 1 (PD-L1) on triple-negative breast cancer cell surface at the single-cell level and for monitoring the expression variation of PD-L1 during drug treatment.〈/p〉

    Description: 〈p〉Slow slip events have been suggested to trigger subduction earthquakes. However, examples to date have been poorly recorded, occurring offshore, where data are sparse. Better understanding of slow slip events and their influence on subsequent earthquakes is critical for hazard forecasts. We analyze a well-recorded event beginning 6 months before the 2012 〈i〉M〈/i〉〈sub〉w〈/sub〉 (moment magnitude) 7.6 earthquake in Costa Rica. The event migrates to the eventual megathrust rupture. Peak slip rate reached a maximum of 5 mm/day, 43 days before the earthquake, remaining high until the earthquake. However, changes in Mohr-Coulomb failure stress at the hypocenter were small (0.1 bar). Our data contradict models of earthquake nucleation that involve power law acceleration of slip and foreshocks. Slow slip events may prove useful for short-term earthquake forecasts.〈/p〉

    Description: 〈p〉N〈sub〉2〈/sub〉-fixing colonies of cyanobacteria and aggregates of phytoplankton and detritus sinking hundreds of meters per day are instrumental for the ocean’s sequestration of CO〈sub〉2〈/sub〉 from the atmosphere. Understanding of small-scale microbial processes associated with phytoplankton colonies and aggregates is therefore crucial for understanding large-scale biogeochemical processes in the ocean. Phytoplankton colonies and sinking aggregates are characterized by steep concentration gradients of gases and nutrients in their interior. Here, we present a mechanistic mathematical model designed to perform modeling of small-scale fluxes and evaluate the physical, chemical, and biological constraints of processes that co-occur in phytoplankton colonies and sinking porous aggregates. The model accurately reproduced empirical measurements of O〈sub〉2〈/sub〉 concentrations and fluxes measured in sinking aggregates. Common theoretical assumptions of either constant concentration or constant flux over the entire surface did not apply to sinking aggregates. Consequently, previous theoretical models overestimate O〈sub〉2〈/sub〉 flux in these aggregates by as high as 15-fold.〈/p〉

    Description: 〈p〉How appetite is modulated by physiological, contextual, or pharmacological influence is still unclear. Specifically, the discovery of appetite modulators is compromised by the abundance of side effects that usually limit in vivo drug action. We set out to identify neuroactive drugs that trigger only their intended single behavioral change, which would provide great therapeutic advantages. To identify these ideal bioactive small molecules, we quantified the impact of more than 10,000 compounds on an extended series of different larval zebrafish behaviors using an in vivo imaging strategy. Known appetite-modulating drugs altered feeding and a pleiotropy of behaviors. Using this multibehavioral strategy as an active filter for behavioral side effects, we identified previously unidentified compounds that selectively increased or reduced food intake by more than 50%. The general applicability of this strategy is shown by validation in mice. Mechanistically, most candidate compounds were independent of the main neurotransmitter systems. In addition, we identified compounds with multibehavioral impact, and correlational comparison of these profiles with those of known drugs allowed for the prediction of their mechanism of action. Our results illustrate an unbiased and translational drug discovery strategy for ideal psychoactive compounds and identified selective appetite modulators in two vertebrate species.〈/p〉

    Description: 〈p〉A methionine substitution at lysine-27 on histone H3 variants (H3K27M) characterizes ~80% of diffuse intrinsic pontine gliomas (DIPG) and inhibits polycomb repressive complex 2 (PRC2) in a dominant-negative fashion. Yet, the mechanisms for this inhibition and abnormal epigenomic landscape have not been resolved. Using quantitative proteomics, we discovered that robust PRC2 inhibition requires levels of H3K27M greatly exceeding those of PRC2, seen in DIPG. While PRC2 inhibition requires interaction with H3K27M, we found that this interaction on chromatin is transient, with PRC2 largely being released from H3K27M. Unexpectedly, inhibition persisted even after PRC2 dissociated from H3K27M-containing chromatin, suggesting a lasting impact on PRC2. Furthermore, allosterically activated PRC2 is particularly sensitive to H3K27M, leading to the failure to spread H3K27me from PRC2 recruitment sites and consequently abrogating PRC2’s ability to establish H3K27me2-3 repressive chromatin domains. In turn, levels of polycomb antagonists such as H3K36me2 are elevated, suggesting a more global, downstream effect on the epigenome. Together, these findings reveal the conditions required for H3K27M-mediated PRC2 inhibition and reconcile seemingly paradoxical effects of H3K27M on PRC2 recruitment and activity.〈/p〉

    Description: 〈p〉Intracellular delivery of mRNA, DNA, and other large macromolecules into cells plays an essential role in an array of biological research and clinical therapies. However, current methods yield a wide variation in the amount of material delivered, as well as limitations on the cell types and cargoes possible. Here, we demonstrate quantitatively controlled delivery into a range of primary cells and cell lines with a tight dosage distribution using a nanostraw-electroporation system (NES). In NES, cells are cultured onto track-etched membranes with protruding nanostraws that connect to the fluidic environment beneath the membrane. The tight cell-nanostraw interface focuses applied electric fields to the cell membrane, enabling low-voltage and nondamaging local poration of the cell membrane. Concurrently, the field electrophoretically injects biomolecular cargoes through the nanostraws and into the cell at the same location. We show that the amount of material delivered is precisely controlled by the applied voltage, delivery duration, and reagent concentration. NES is highly effective even for primary cell types or different cell densities, is largely cargo agnostic, and can simultaneously deliver specific ratios of different molecules. Using a simple cell culture well format, the NES delivers into 〉100,000 cells within 20 s with 〉95% cell viability, enabling facile, dosage-controlled intracellular delivery for a wide variety of biological applications.〈/p〉

    Description: 〈p〉Scholars endeavor to understand the relationship between human evolution and climate change. This is particularly germane for Neanderthals, who survived extreme Eurasian environmental variation and glaciations, mysteriously going extinct during a cool interglacial stage. Here, we integrate weekly records of climate, tooth growth, and metal exposure in two Neanderthals and one modern human from southeastern France. The Neanderthals inhabited cooler and more seasonal periods than the modern human, evincing childhood developmental stress during wintertime. In one instance, this stress may have included skeletal mobilization of elemental stores and weight loss; this individual was born in the spring and appears to have weaned 2.5 years later. Both Neanderthals were exposed to lead at least twice during the deep winter and/or early spring. This multidisciplinary approach elucidates direct relationships between ancient environments and hominin paleobiology.〈/p〉

    Description: 〈p〉Persistent episodes of extreme weather in the Northern Hemisphere summer have been associated with high-amplitude quasi-stationary atmospheric Rossby waves, with zonal wave numbers 6 to 8 resulting from the phenomenon of quasi-resonant amplification (QRA). A fingerprint for the occurrence of QRA can be defined in terms of the zonally averaged surface temperature field. Examining state-of-the-art [Coupled Model Intercomparison Project Phase 5 (CMIP5)] climate model projections, we find that QRA events are likely to increase by ~50% this century under business-as-usual carbon emissions, but there is considerable variation among climate models. Some predict a near tripling of QRA events by the end of the century, while others predict a potential decrease. Models with amplified Arctic warming yield the most pronounced increase in QRA events. The projections are strongly dependent on assumptions regarding the nature of changes in radiative forcing associated with anthropogenic aerosols over the next century. One implication of our findings is that a reduction in midlatitude aerosol loading could actually lead to Arctic de-amplification this century, ameliorating potential increases in persistent extreme weather events.〈/p〉

    Description: 〈p〉The small molecules thalidomide, lenalidomide, and pomalidomide induce the ubiquitination and proteasomal degradation of the transcription factors Ikaros (IKZF1) and Aiolos (IKZF3) by recruiting a Cys〈sub〉2〈/sub〉-His〈sub〉2〈/sub〉 (C2H2) zinc finger domain to Cereblon (CRBN), the substrate receptor of the CRL4〈sup〉CRBN〈/sup〉 E3 ubiquitin ligase. We screened the human C2H2 zinc finger proteome for degradation in the presence of thalidomide analogs, identifying 11 zinc finger degrons. Structural and functional characterization of the C2H2 zinc finger degrons demonstrates how diverse zinc finger domains bind the permissive drug-CRBN interface. Computational zinc finger docking and biochemical analysis predict that more than 150 zinc fingers bind the drug-CRBN complex in vitro, and we show that selective zinc finger degradation can be achieved through compound modifications. Our results provide a rationale for therapeutically targeting transcription factors that were previously considered undruggable.〈/p〉

    Description: 〈p〉Speleothem oxygen isotope records have revolutionized our understanding of the paleo East Asian monsoon, yet there is fundamental disagreement on what they represent in terms of the hydroclimate changes. We report a multiproxy speleothem record of monsoon evolution during the last deglaciation from the middle Yangtze region, which indicates a wetter central eastern China during North Atlantic cooling episodes, despite the oxygen isotopic record suggesting a weaker monsoon. We show that this apparent contradiction can be resolved if the changes are interpreted as a lengthening of the Meiyu rains and shortened post-Meiyu stage, in accordance with a recent hypothesis. Model simulations support this interpretation and further reveal the role of the westerlies in communicating the North Atlantic influence to the East Asian climate.〈/p〉

    Description: 〈p〉The pathologic accumulation and aggregation of α-synuclein (α-syn) underlies Parkinson’s disease (PD). The molecular mechanisms by which pathologic α-syn causes neurodegeneration in PD are not known. Here, we found that pathologic α-syn activates poly(adenosine 5'-diphosphate–ribose) (PAR) polymerase-1 (PARP-1), and PAR generation accelerates the formation of pathologic α-syn, resulting in cell death via parthanatos. PARP inhibitors or genetic deletion of PARP-1 prevented pathologic α-syn toxicity. In a feed-forward loop, PAR converted pathologic α-syn to a more toxic strain. PAR levels were increased in the cerebrospinal fluid and brains of patients with PD, suggesting that PARP activation plays a role in PD pathogenesis. Thus, strategies aimed at inhibiting PARP-1 activation could hold promise as a disease-modifying therapy to prevent the loss of dopamine neurons in PD.〈/p〉

    Description: 〈p〉Broadly neutralizing antibodies against highly variable pathogens have stimulated the design of vaccines and therapeutics. We report the use of diverse camelid single-domain antibodies to influenza virus hemagglutinin to generate multidomain antibodies with impressive breadth and potency. Multidomain antibody MD3606 protects mice against influenza A and B infection when administered intravenously or expressed locally from a recombinant adeno-associated virus vector. Crystal and single-particle electron microscopy structures of these antibodies with hemagglutinins from influenza A and B viruses reveal binding to highly conserved epitopes. Collectively, our findings demonstrate that multidomain antibodies targeting multiple epitopes exhibit enhanced virus cross-reactivity and potency. In combination with adeno-associated virus–mediated gene delivery, they may provide an effective strategy to prevent infection with influenza virus and other highly variable pathogens.〈/p〉

    Description: 〈p〉Gut microbes live in symbiosis with their hosts, but how mutualistic animal-microbe interactions emerge is not understood. By adaptively evolving the opportunistic fungal pathogen 〈i〉Candida albicans〈/i〉 in the mouse gastrointestinal tract, we selected strains that not only had lost their main virulence program but also protected their new hosts against a variety of systemic infections. This protection was independent of adaptive immunity, arose as early as a single day postpriming, was dependent on increased innate cytokine responses, and was thus reminiscent of "trained immunity." Because both the microbe and its new host gain some advantages from their interaction, this experimental system might allow direct study of the evolutionary forces that govern the emergence of mutualism between a mammal and a fungus.〈/p〉

    Description: 〈p〉The synthesis of ultrasmall supported bimetallic nanoparticles (between 1 and 3 nanometers in diameter) with well-defined stoichiometry and intimacy between constituent metals remains a substantial challenge. We synthesized 10 different supported bimetallic nanoparticles via surface inorganometallic chemistry by decomposing and reducing surface-adsorbed heterometallic double complex salts, which are readily obtained upon sequential adsorption of target cations and anions on a silica substrate. For example, adsorption of tetraamminepalladium(II) [Pd(NH〈sub〉3〈/sub〉)〈sub〉4〈/sub〉〈sup〉2+〈/sup〉] followed by adsorption of tetrachloroplatinate [PtCl〈sub〉4〈/sub〉〈sup〉2–〈/sup〉] was used to form palladium-platinum (Pd-Pt) nanoparticles. These supported bimetallic nanoparticles show enhanced catalytic performance in acetylene selective hydrogenation, which clearly demonstrates a synergistic effect between constituent metals.〈/p〉

    Description: 〈p〉Gradient structures exist ubiquitously in nature and are increasingly being introduced in engineering. However, understanding structural gradient–related mechanical behaviors in all gradient structures, including those in engineering materials, has been challenging. We explored the mechanical performance of a gradient nanotwinned structure with highly tunable structural gradients in pure copper. A large structural gradient allows for superior work hardening and strength that can exceed those of the strongest component of the gradient structure. We found through systematic experiments and atomistic simulations that this unusual behavior is afforded by a unique patterning of ultrahigh densities of dislocations in the grain interiors. These observations not only shed light on gradient structures, but may also indicate a promising route for improving the mechanical properties of materials through gradient design.〈/p〉

    Description: 〈span〉〈div〉Summary〈/div〉The strength of the lithosphere plays a key role in the formation and evolution of tectonic plate boundaries. Localized lithospheric deformation associated with plate tectonics requires a mechanism for weakening across the entire width of the lithosphere, including the strongest cold ductile region. We explore the microphysics of weakening of lithospheric materials, and in particular the coupled evolution of mineral grain size and intragranular defects and their control on lithospheric strength. We propose a model for the interaction between grain-boundaries and dislocation density to reduce the net free energy of grains during dynamic recrystallization (DRX). The driving forces for DRX arise from heterogeneity in dislocation density and grain boundary curvature. Our model shows that grain growth driven by variation in grain boundary curvature can be impeded by variation in dislocation density; this occurs because as the grains grow, to minimize their surface energy, their dislocation density and associated internal energy may increase and offset the driving forces for grain growth. The correlation between grain size and dislocation density can for example arise because the dislocation accumulation in smaller grains is suppressed due to the large stress that is needed to bend and elongate a short dislocation (as dictated by the small grain size), while the larger grains can have long dislocations and reach a steady state dislocation density dictated by the applied stress. In a lithospheric setting, slower grain growth means that it would require less mechanical work to establish weak localized shear zones through grain damage, and retard the healing of previously damaged zones. Furthermore, the competition of two different time-scales - that of grain growth and the dislocation kinetics - can lead to oscillating behavior over 1 to 10 years as the grain size and dislocation density advance towards their steady states. These oscillations are likely to have an effect on the rheology of lithospheric rocks, e.g. their strengthening and weakening through time, and have a potential application to geological processes such as postseismic creep in ductile shear zones.〈/span〉

    Description: 〈p〉The bulk of Earth’s biological materials consist of few base substances—essentially proteins, polysaccharides, and minerals—that assemble into large varieties of structures. Multifunctionality arises naturally from this structural complexity: An example is the combination of rigidity and flexibility in protein-based teeth of the squid sucker ring. Other examples are time-delayed actuation in plant seed pods triggered by environmental signals, such as fire and water, and surface nanostructures that combine light manipulation with mechanical protection or water repellency. Bioinspired engineering transfers some of these structural principles into technically more relevant base materials to obtain new, often unexpected combinations of material properties. Less appreciated is the huge potential of using bioinspired structural complexity to avoid unnecessary chemical diversity, enabling easier recycling and, thus, a more sustainable materials economy.〈/p〉

    Description: 〈p〉Exploration of intermediates that enable chemoselective cycloaddition reactions and expeditious construction of fused- or bridged-ring systems is a continuous challenge for organic synthesis. As an intermediate of interest, the oxyallyl cation has been harnessed to synthesize architectures containing seven-membered rings via (4+3) cycloaddition. However, its potential to access five-membered skeletons is underdeveloped, largely due to the thermally forbidden (3+2) pathway. Here, the combination of a tailored precursor and a Pd(0) catalyst generates a Pd-oxyallyl intermediate that cyclizes with conjugated dienes to produce a diverse array of tetrahydrofuran skeletons. The cycloaddition overrides conventional (4+3) selectivity by proceeding through a stepwise pathway involving a Pd-allyl transfer and ring closure sequence. Subsequent treatment of the (3+2) adducts with a palladium catalyst converts the heterocycles to the carbocyclic cyclopentanones.〈/p〉

Here, we report single-cell RNA sequencing of 14,341 and 6754 cells from first-trimester human placental villous and decidual tissues, respectively. Bioinformatic analysis identified major cell types, many known and some subtypes previously unknown in placental villi and decidual context. Further detailed analysis revealed proliferating subpopulations, enrichment of cell type–specific transcription factors, and putative intercellular communication in the fetomaternal microenvironment.     Description: 〈span〉〈div〉Summary〈/div〉Locating and monitoring passive seismic sources provides us important information for studying subsurface rock deformation, injected fluid migration, regional stress conditions as well as fault rupture mechanism. In this paper, we present a novel passive-source monitoring approach using vector-based elastic time-reversal imaging. By solving the elastic wave equation using observed multicomponent records as boundary conditions, we first compute back-propagated elastic wavefields in the subsurface. Then, we separate the extrapolated wavefields into compressional (P-wave) and shear (S-wave) modes using the vector Helmholtz decomposition. A zero-lag cross-correlation imaging condition is applied to the separated pure-mode vector wavefields to produce passive-source images. We compare imaging results using three implementations, i.e., dot-product, energy and power. Numerical experiments demonstrate that the power imaging condition gives us the highest resolution and is less sensitive to the presence of random noises. To capture the propagation of microseismic fracture and earthquake rupture, we modify the traditional zero-lag cross-correlation imaging condition by summing the multiplication of the separated P- and S-wavefields within local time windows, which enables us to capture the temporal and spatial evolution of earthquake rupture. 2D and 3D numerical examples demonstrate that the proposed method is capable of accurately locating point sources, as well as delineating dynamic propagation of hydraulic fracture and earthquake rupture.〈/span〉

Here, we report a new type of log-periodic quantum oscillations in ultraquantum three-dimensional topological materials. Beyond the quantum limit (QL), we observe the log-periodic oscillations involving up to five oscillating cycles (five peaks and five dips) on the magnetoresistance of high-quality single-crystal ZrTe〈sub〉5〈/sub〉, virtually showing the clearest feature of discrete scale invariance (DSI). Further, theoretical analyses show that the two-body quasi-bound states can be responsible for the DSI feature. Our work provides a new perspective on the ground state of topological materials beyond the QL.〈/p〉

    Description: 〈p〉The intravitreal delivery of therapeutic agents promises major benefits in the field of ocular medicine. Traditional delivery methods rely on the random, passive diffusion of molecules, which do not allow for the rapid delivery of a concentrated cargo to a defined region at the posterior pole of the eye. The use of particles promises targeted delivery but faces the challenge that most tissues including the vitreous have a tight macromolecular matrix that acts as a barrier and prevents its penetration. Here, we demonstrate novel intravitreal delivery microvehicles—slippery micropropellers—that can be actively propelled through the vitreous humor to reach the retina. The propulsion is achieved by helical magnetic micropropellers that have a liquid layer coating to minimize adhesion to the surrounding biopolymeric network. The submicrometer diameter of the propellers enables the penetration of the biopolymeric network and the propulsion through the porcine vitreous body of the eye over centimeter distances. Clinical optical coherence tomography is used to monitor the movement of the propellers and confirm their arrival on the retina near the optic disc. Overcoming the adhesion forces and actively navigating a swarm of micropropellers in the dense vitreous humor promise practical applications in ophthalmology.〈/p〉

    Description: 〈p〉Genomic DNA forms chromatin, in which the nucleosome is the repeating unit. The mechanism by which RNA polymerase II (RNAPII) transcribes the nucleosomal DNA remains unclear. Here we report the cryo–electron microscopy structures of RNAPII-nucleosome complexes in which RNAPII pauses at the superhelical locations SHL(–6), SHL(–5), SHL(–2), and SHL(–1) of the nucleosome. RNAPII pauses at the major histone-DNA contact sites, and the nucleosome interactions with the RNAPII subunits stabilize the pause. These structures reveal snapshots of nucleosomal transcription, in which RNAPII gradually tears DNA from the histone surface while preserving the histone octamer. The nucleosomes in the SHL(–1) complexes are bound to a "foreign" DNA segment, which might explain the histone transfer mechanism. These results provide the foundations for understanding chromatin transcription and epigenetic regulation.〈/p〉

Biocomposites have attracted the attention of automakers for the design of lightweight parts. Hybrid biocomposites made of petrochemical-based and bioresourced materials have led to technological advances in manufacturing. Greener biocomposites from plant-derived fiber and crop-derived plastics with higher biobased content are continuously being developed. Biodegradable composites have shown potential for major uses in sustainable packaging. Recycled plastic materials originally destined for landfills can be redirected and repurposed for blending in composite applications, thus leading to reduced dependence on virgin petro-based materials. Studies on compatibility of recycled and waste materials with other components in composite structure for improved interface and better mechanical performance pose major scientific challenges.     Description: 〈p〉Animals constantly update their body posture to meet behavioral demands, but little is known about the neural signals on which this depends. We therefore tracked freely foraging rats in three dimensions while recording from the posterior parietal cortex (PPC) and the frontal motor cortex (M2), areas critical for movement planning and navigation. Both regions showed strong tuning to posture of the head, neck, and back, but signals for movement were much less dominant. Head and back representations were organized topographically across the PPC and M2, and more neurons represented postures that occurred less often. Simultaneous recordings across areas were sufficiently robust to decode ongoing behavior and showed that spiking in the PPC tended to precede that in M2. Both the PPC and M2 strongly represent posture by using a spatially organized, energetically efficient population code.〈/p〉

    Description: 〈span〉〈div〉Summary〈/div〉Magnitudes of differential stress in the lithosphere, especially in the crust, are still disputed. Earthquake-based stress drop estimates indicate median values 〈 10 MPa whereas the lateral variation of gravitational potential energy per unit area (〈span〉GPE〈/span〉) across significant relief indicates stress magnitudes of ca. 100 MPa in average across a 100 km thick lithosphere between the Indian lowland and the Tibetan plateau. These standard 〈span〉GPE〈/span〉-based stress estimates correspond to membrane stresses, because they are associated with a deformation that is uniform with depth. We show here with new analytical results that lateral variations in 〈span〉GPE〈/span〉 can also cause bending moments and related bending stresses of several hundreds of MPa. Furthermore, we perform two-dimensional thermo-mechanical numerical simulations (1) to evaluate estimates for membrane and bending stresses based on 〈span〉GPE〈/span〉 variations, (2) to quantify minimum crustal stress magnitudes that are required to maintain the topographic relief between Indian lowland and Tibetan plateau for ca. 10 Ma and (3) to quantify the corresponding relative contribution of crustal strength to the total lithospheric strength. The numerical model includes viscoelastoplastic deformation, gravity and heat transfer. The model configuration is based on density fields from the CRUST1.0 data set and from a geophysically and petrologically constrained density model based on 〈span〉in situ〈/span〉 field campaigns. The numerical results indicate that values of differential stress in the upper crust must be 〉 ca. 180 MPa, corresponding to a friction angle of ca. 10°, to maintain the topographic relief between lowland and plateau for 〉 10 Ma. The relative contribution of crustal strength to total lithospheric strength varies considerably laterally. In the region between lowland and plateau and inside the plateau the depth-integrated crustal strength is approximately equal to the depth-integrated strength of the mantle lithosphere. Simple analytical formulae predicting the lateral variation of depth-integrated stresses agree with numerically calculated stress fields, which show both the accuracy of the numerical results and the applicability of simple, rheology-independent, analytical predictions to highly variable, rheology-dependent, stress fields. Our results indicate that (1) crustal strength can be locally equal to mantle lithosphere strength and that (2) crustal stresses must be at least one order of magnitude larger than median stress drops in order to support the plateau relief over a duration of ca. 10 Ma.〈/span〉

    Description: 〈p〉It is crucial but of a great challenge to study in vivo and in situ drug release of nanocarriers when developing a nanomaterial-based drug delivery platform. We developed a new label-free laser desorption/ionization mass spectrometry (MS) imaging strategy that enabled visualization and quantification of the in situ drug release in tissues by monitoring intrinsic MS signal intensity ratio of loaded drug over the nanocarriers. The proof of concept was demonstrated by investigating the doxorubicin (DOX)/polyethylene glycol–MoS〈sub〉2〈/sub〉 nanosheets drug delivery system in tumor mouse models. The results revealed a tissue-dependent release behavior of DOX during circulation with the highest dissociation in tumor and lowest dissociation in liver tissues. The drug-loaded MoS〈sub〉2〈/sub〉 nanocarriers are predominantly distributed in lung, spleen, and liver tissues, whereas the accumulation in the tumor was unexpectedly lower than in normal tissues. This new strategy could also be extended to other drug-carrier systems, such as carbon nanotubes and black phosphorus nanosheets, and opened a new path to evaluate the drug release of nanocarriers in the suborgan level.〈/p〉

    Description: 〈p〉The sources and nature of organic carbon on Mars have been a subject of intense research. Steele 〈i〉et al.〈/i〉 (2012) showed that 10 martian meteorites contain macromolecular carbon phases contained within pyroxene- and olivine-hosted melt inclusions. Here, we show that martian meteorites Tissint, Nakhla, and NWA 1950 have an inventory of organic carbon species associated with fluid-mineral reactions that are remarkably consistent with those detected by the Mars Science Laboratory (MSL) mission. We advance the hypothesis that interactions among spinel-group minerals, sulfides, and a brine enable the electrochemical reduction of aqueous CO〈sub〉2〈/sub〉 to organic molecules. Although documented here in martian samples, a similar process likely occurs wherever igneous rocks containing spinel-group minerals and/or sulfides encounter brines.〈/p〉

    Description: 〈span〉〈div〉Summary〈/div〉Scanning magnetometers are increasingly used to characterize the magnetization of mineral grains in rock samples. Up-scaling this measurement technique to large numbers of individual particles is hampered by the intrinsic non-uniqueness of potential-field inversion. Here it is shown that this problem can be circumvented by adding tomographic information that determines the location of the possible field sources. Standard potential theory is used to prove a uniqueness theorem which completely characterizes the mathematical background of the corresponding source-localized inversion. It exactly resolves under which conditions a potential field measurement on a surface can be uniquely decomposed into signals from the different source regions. The intrinsic non-uniqueness of potential field inversion prevents that the source distribution inside the tomographically outlined regions can be recovered, but the potential field of each region is uniquely defined. For scanning magnetometers in rock magnetism, this result implies that magnetic dipole vectors of large numbers of individual magnetic particles can be reliably reconstructed from surface scans of the magnetic field, if the particle positions are independently determined. This provides an incentive to improve scanning methods for future paleomagnetic applications.〈/span〉

    Description: 〈span〉〈div〉Summary〈/div〉The most common earthquake forecasting models assume that the magnitude of the next earthquake is independent from the past. This feature severely limits the capability to forecast large earthquakes with high probabilities. Here we investigate empirically on the magnitude-independence assumption, exploring if: i) background and triggered earthquakes have the same frequency-magnitude distribution, ii) variations of seismicity in the space-time-magnitude domain encode some information on the future earthquakes size. For this purpose, and to verify the stability of the findings, we consider seismic catalogues covering different space-time-magnitude windows, such as the Alto Tiberina Near Fault Observatory (TABOO), the California and Japanese seismic catalogues. Our approach is inspired by the nearest-neighbour method proposed by Baiesi & Paczuski (2004) and elaborated by Zaliapin et al. (2008) to distinguish between triggered and background earthquakes. Here we implement the same metric-based correlation to identify the precursory seismicity of any triggered earthquake; this allows us to analyse, for each triggered earthquake, the space-time-magnitude distribution of the seismicity that likely contributed to its occurrence. Our results show that the magnitude-independence assumption holds reasonably well in all catalogues, with a remarkable exception that is consistent with a previous independent study; this departure from the magnitude-independence assumption shows that larger events tend to nucleate at a higher distance from the ongoing sequence. We also notice that the reliability of this assumption may depend on the spatial scale considered; it holds for seismic catalogues of large areas, but we identify possible departures in small areas, reflecting different ways to release locally seismic energy. Finally, we come across an important issue that may lead to misleading results in similar studies, i.e., if a seismic catalogue appears overall complete above a fixed magnitude threshold, it may still yield spurious signals into the analysis. Specifically, we show that some significant departures from the magnitude-independence assumption do not survive when considering spatiotemporal variations of the magnitude of completeness.〈/span〉

    Description: 〈span〉〈div〉Summary〈/div〉Vp/Vs models provide important complementary information to Vp and Vs models, relevant to lithology, rock damage, partial melting, water saturation, etc. However, seismic tomography using body-wave traveltime data from local or regional earthquakes does not constrain Vp/Vs well due to the different resolution of Vp and Vs models, with the Vp models usually better constrained than Vs. Since surface-wave data are most sensitive to Vs, which leads to complementary strengths with respect to body-wave data, we jointly invert body- and surface-wave data to better resolve the Vp/Vs models. In order to show the robustness of our joint inversion method, we compare the results to other approaches, including dividing Vp by Vs models and Vp/Vs parameterization with body-wave or both body- and surface-wave data, using synthetic data and real data from the southern California plate boundary region. We confirm that Vp/Vs models from body-wave inversion obtained by division tend to include artifacts, even when both Vp and Vs models seem reasonable. The joint inversion with Vp/Vs parameterization is found to improve the Vp/Vs ratio model significantly and the resultant Vp/Vs model shows more geologically consistent features, such as high Vp/Vs along fault traces at shallow depths likely indicating fault-related damage. The Vp/Vs model also exhibits contrasts at intermediate depths along the Peninsular Range compositional boundary, and high Vp/Vs in the lower crust near the Salton Sea region correlated with high heat flow and may indicate partial melting. The improved Vp/Vs as well as individual Vp and Vs models are useful for earthquake relocation, high-resolution Moho depth imaging, and interpretation of other data and tectonic evolution in the region.〈/span〉

    Description: 〈span〉〈div〉Summary〈/div〉Interpretation of surface fault scarps and palaeoseismic trenches is a key component of estimating fault slip rates, earthquake recurrence rates and maximum magnitudes for hazard assessments. Often these analyses rely on the assumption that successive earthquakes all breached the surface and that the ruptures are recorded topographically, or by the deposits exposed in a trench. The M〈sub〉〈span〉w〈/span〉〈/sub〉7.2 1992 Suusamyr earthquake, Kyrgyzstan, is an apparently problematic case for such analyses because its ruptures show significant displacement but are only mapped as having broken the surface along small, disparate portions of the fault. Here we present the results of surveys conducted along the Suusamyr Fault to establish whether that is the case. Two sets of ruptures were identified following the earthquake. They are unusually short for their displacement and are separated by a 25 km gap. Using satellite imagery, high-resolution digital elevation models and palaeoseismic trenching we first reassess the distribution of the 1992 ruptures and then reconstruct the Holocene earthquake record to establish the extent to which the 1992 earthquake is representative of the rupture behaviour of this fault. We find evidence for at least two prehistoric surface rupturing earthquakes in the Holocene: one ∼3 ka and one 〉8 ka that, along with the modern event, gives recurrence intervals of ∼3 kyr and ∼5 kyr. Within spatial gaps between segments of the 1992 ruptures there are clear prehistoric surface ruptures and the ruptures in each prehistoric earthquake were discontinuous. We conclude that there is significant variability in the surface rupture pattern of successive earthquakes on the Suusamyr Fault, with implications for the completeness of palaeoseismic records obtained from thrust scarps.〈/span〉

    Description: 〈span〉〈div〉Summary〈/div〉We provide a six-component (6-C) polarization model for 〈span〉P〈/span〉-, 〈span〉SV〈/span〉-, 〈span〉SH〈/span〉-, Rayleigh-, and Love-waves both inside an elastic medium as well as at the free surface. It is shown that single-station 6-C data comprised of three components of rotational motion and three components of translational motion provide the opportunity to unambiguously identify the wave type, propagation direction, and local 〈span〉P〈/span〉- and 〈span〉S〈/span〉-wave velocities at the receiver location by use of polarization analysis. To extract such information by conventional processing of three-component (3-C) translational data would require large and dense receiver arrays. The additional rotational components allow the extension of the rank of the coherency matrix used for polarization analysis. This enables us to accurately determine the wave type and wave parameters (propagation direction and velocity) of seismic phases, even if more than one wave is present in the analysis time window. This is not possible with standard, pure-translational 3-C recordings. In order to identify modes of vibration and to extract the accompanying wave parameters, we adapt the multiple signal classification algorithm (MUSIC). Due to the strong nonlinearity of the MUSIC estimator function, it can be used to detect the presence of specific wave types within the analysis time window at very high resolution. We show how the extracted wavefield properties can be used, in a fully automated way, to separate the wavefield into its different wave modes using only a single 6-C recording station. As an example, we apply the method to remove surface wave energy while preserving the underlying reflection signal and to suppress energy originating from undesired directions, such as side-scattered waves.〈/span〉

    Description: 〈span〉〈div〉Summary〈/div〉The Sentinel-1 mission comprises two synthetic aperture radar satellites, each with a 12 day orbital repeat, orbiting 6 days apart within a narrow tube. The mission design promises the ability to respond quickly to earthquakes with InSAR, and to facilitate production of interferograms with good interferometric correlation globally. We report on our efforts to study global seismicity using Sentinel-1 Interferometric Wide-Swath data between April 2015 and December 2016. We select 35 potentially detectable terrestrial earthquakes in the range 5.5 ≤ 〈span〉Mw〈/span〉 ≤ 7.8 on the basis of their locations, depths and magnitudes, and process the first post-event interferogram with the shortest possible time-span for each using the ISCE software. We evaluate each interferogram for earthquake deformation signals by visual inspection. We can identify deformation signals attributable to earthquakes in 18 of these interferograms (51%); a further six interferograms (17%) have ambiguous interferometric phase affected by tropospheric noise. 11 events (31%) could not be identified from their interferograms. The majority of these failed detections were due to interferogram decorrelation, particularly apparent for earthquakes that occurred between 15°N and 15°S, where climate conditions promote dense vegetation. The majority of the ambiguous interferograms are affected by tropospheric noise, suggesting that techniques to mitigate such noise could improve detection performance. The largest event we do not detect with Sentinel-1 data is a 〈span〉Mw〈/span〉7.0 earthquake that occurred in Vanuatu in April 2016; we also fail to detect the 2016 〈span〉Mw〈/span〉6.2 Kurayoshi earthquake in one out of two possible 24-day interferograms. We propose these as upper and lower estimates on the magnitude of completeness for earthquakes studied with Sentinel-1 data; to raise the magnitude of completeness we suggest that more frequent (e.g. six day) recurrence may be necessary in low latitude areas.〈/span〉

    Description: 〈span〉〈div〉Summary〈/div〉The correct estimation of site-specific attenuation is crucial for the assessment of seismic hazard. Downhole instruments provide in this context valuable information to constrain attenuation directly from data. In this study, we apply an interferometric approach to this problem by deconvolving seismic motions recorded at depth with those recorded at the surface. In doing so, incident and surface-reflected waves can be separated. We apply this technique not only to earthquake data but also to recordings of ambient vibrations. We compute the transfer function between incident and surface-reflected waves in order to infer frequency dependent quality factors for S-waves. The method is applied to a 87 m deep borehole sensor and a co-located surface instrument situated at a hard-rock site in West Bohemia/Vogtland, Germany. We show that the described method provides comparable attenuation estimates using either earthquake data or ambient noise for frequencies between 5-15 Hz. Moreover, a single hour of noise recordings seems to be sufficient to yield stable deconvolution traces and quality factors, thus, offering a fast and easy way to derive attenuation estimates from borehole recordings even in low to mid seismicity regions.〈/span〉

    Description: 〈span〉〈div〉Summary〈/div〉Injection of CO〈sub〉2〈/sub〉 into tight reservoirs produces both gravity change and ground deformation, which provide great opportunities for more accurate coupled inverse modelling. In this study, we incorporate signals generated from several synthetic models to estimate the CO〈sub〉2〈/sub〉 distribution in the reservoir. A relationship is found that connects density variations to volumetric changes associated with injected CO〈sub〉2〈/sub〉, taking advantage of a common set of model parameters for both gravitational and geo-mechanical inverse modelling. This is achieved by assuming that the injected CO〈sub〉2〈/sub〉 increases pressure in the reservoir, which in turn generates extra porosity that is then filled in by the CO〈sub〉2〈/sub〉 mass in the generated space. Tikhonov regularization, supported by the Generalized Cross Validation (GCV) technique for finding the optimized model, is used to solve the ill-posed inverse problems. The results indicate that with a combination of gravity and ground deformation monitoring, the uncertainty and ambiguity in gravimetric modelling due to high levels of noise is mitigated by implementing highly accurate ground deformation measurements, which normally have a higher signal to noise ratio.〈/span〉

    Description: 〈span〉〈div〉Summary〈/div〉As the number of seismic sensors grows, it is becoming increasingly difficult for analysts to pick seismic phases manually and comprehensively, yet such efforts are fundamental to earthquake monitoring. Despite years of improvements in automatic phase picking, it is difficult to match the performance of experienced analysts. A more subtle issue is that different seismic analysts may pick phases differently, which can introduce bias into earthquake locations. We present a deep-neural-network-based arrival-time picking method called ”PhaseNet” that picks the arrival times of both P and S waves. Deep neural networks have recently made rapid progress in feature learning, and with sufficient training, have achieved super-human performance in many applications. PhaseNet uses three-component seismic waveforms as input and generates probability distributions of P arrivals, S arrivals, and noise as output. We engineer PhaseNet such that peaks in the probability distributions provide accurate arrival times for both P and S waves. PhaseNet is trained on the prodigious available data set provided by analyst-labeled P and S arrival times from the Northern California Earthquake Data Center. The dataset we use contains more than seven hundred thousand waveform samples extracted from over thirty years of earthquake recordings. We demonstrate that PhaseNet achieves much higher picking accuracy and recall rate than existing methods when applied to the waveforms of known earthquakes, which has the potential to increase the number of S-wave observations dramatically over what is currently available. This will enable both improved locations and improved shear wave velocity models.〈/span〉

Here we examine LFEs near Parkfield, CA in order to assess several proposed explanations for LFEs’ long durations. We determine LFE rupture areas and location distributions using a new approach, similar to directivity analysis, where we examine how signals coming from various locations within LFEs’ finite rupture extents create differences in the apparent source time functions recorded at various stations. We use synthetic ruptures to determine how much the LFE signals recorded at each station would be modified by spatial variations of the source-station travel time within the rupture area given various possible rupture diameters, and then compare those synthetics with the data. Our synthetics show that the methodology can identify inter-station variations created by heterogeneous slip distributions or complex rupture edges, and thus lets us estimate LFE rupture extents for unilateral or bilateral ruptures. To obtain robust estimates of the sources’ similarity across stations, we stack signals from thousands of LFEs, using an empirical Green’s function approach to isolate the LFEs’ apparent source time functions from the path effects. Our analysis of LFEs in Parkfield implies that LFEs’ apparent source time functions are similar across stations at frequencies up to 8 to 16 Hz, depending on the family. The inter-station coherence observed at these relatively high frequencies, or short wavelengths (down to 0.2 to 0.5 km), suggest that LFEs in each of the 7 families examined occur on asperities. They are clustered in patches with sub-1-km diameters. The individual LFEs’ rupture diameters are estimated to be smaller than 1.1 km for all families, and smaller than 0.5 km and 1 km for the two shallowest families, which were previously found to have 0.2-s durations. Coupling the diameters with the durations suggests that it is possible to model these 〈span〉MW〈/span〉 1-2 LFEs with earthquake-like rupture speeds: around 70% of the shear wave speed. However, that rupture speed matches the data only at the edge of our uncertainty estimates for the family with highest coherence. The data for that family are better matched if LFEs have rupture velocities smaller than 40% of the shear wave speed, or if LFEs have different rupture dynamics. They could have long rise times, contain composite sub-ruptures, or have slip distributions that persist from event to event.〈/span〉

    Description: 〈span〉〈div〉Summary〈/div〉On June 24, 2015, a 230,000 cubic metre landslide slid into the triangle bayou at the intersection of the Yangtze and Daning Rivers and generated a river tsunami that ran up 6.2 metres on the opposite shoreline at Wushan town. The slope failure and resulting waves killed two people and damaged many shipping facilities. Based on field surveys and eyewitness observations, we apply the ‘Tsunami Squares’ method to model the Hongyanzi landslide and its generated waves. Landslide simulations indicate a maximum impact velocity of ∼16 m/s that matches well with an eyewitness video. The computed post-slide mass stopped on the near riverbed with a shape fitting the observed geological profile. Tsunami simulations reveal a large region of wave impacts that coincide with the observed runup heights. The successful reproduction of the dynamics of this landslide-generated river tsunami emphasizes the capacity and efficiency of Tsunami Squares modeling in emergency reaction and risk assessment.〈/span〉

    Description: 〈p〉The LZTR1 protein, an adaptor for cullin 3 (CUL3) ubiquitin ligase complex, is implicated in human disease, yet its mechanism of action remains unknown. We found that Lztr1 haploinsufficiency in mice recapitulates Noonan Syndrome phenotypes; whereas LZTR1 loss in Schwann cells drives dedifferentiation and proliferation. By trapping LZTR1 complexes from intact mammalian cells, we identified the guanosine triphosphatase RAS as a substrate for the LZTR1-CUL3 complex. Ubiquitome analysis showed that loss of Lztr1 abrogated Ras ubiquitination at lysine 170. LZTR1-mediated ubiquitination inhibited RAS signaling by attenuating its association with the membrane. Disease-associated 〈i〉LZTR1〈/i〉 mutations disrupted either LZTR1-CUL3 complex formation, or its interaction with RAS proteins. RAS regulation by LZTR1-mediated ubiquitination provides an explanation for the role of LZTR1 in human disease.〈/p〉

    Description: 〈p〉In genetic screens aimed at understanding drug resistance mechanisms in chronic myeloid leukemia cells, inactivation of the cullin 3 adapter protein-encoding leucine zipper like transcription regulator 1 (〈i〉LZTR1〈/i〉) gene led to enhanced mitogen-activated protein kinase pathway activity and reduced sensitivity to tyrosine kinase inhibitors. Knockdown of the 〈i〉Drosophila LZTR1〈/i〉 ortholog 〈i〉CG3711〈/i〉 resulted in a RAS-dependent gain of function phenotype. Endogenous human LZTR1 associates with the main RAS isoforms. Inactivation of 〈i〉LZTR1〈/i〉 led to decreased ubiquitination and enhanced plasma membrane localization of endogenous KRAS. We propose that LZTR1 acts as a conserved regulator of RAS ubiquitination and MAPK pathway activation. As 〈i〉LZTR1〈/i〉 disease mutations failed to revert loss-of-function phenotypes, our findings provide a molecular rationale for 〈i〉LZTR1〈/i〉 involvement in a variety of inherited and acquired human disorders.〈/p〉

    Description: 〈p〉Understanding of the processes governing soil organic carbon turnover is confounded by the fact that C feedbacks driven by soil erosion have not yet been fully explored at large scale. However, in a changing climate, variation in rainfall erosivity (and hence soil erosion) may change the amount of C displacement, hence inducing feedbacks onto the land C cycle. Using a consistent biogeochemistry-erosion model framework to quantify the impact of future climate on the C cycle, we show that C input increases were offset by higher heterotrophic respiration under climate change. Taking into account all the additional feedbacks and C fluxes due to displacement by erosion, we estimated a net source of 0.92 to 10.1 Tg C year〈sup〉–1〈/sup〉 from agricultural soils in the European Union to the atmosphere over the period 2016–2100. These ranges represented a weaker and stronger C source compared to a simulation without erosion (1.8 Tg C year〈sup〉–1〈/sup〉), respectively, and were dependent on the erosion-driven C loss parameterization, which is still very uncertain. However, when setting a baseline with current erosion rates, the accelerated erosion scenario resulted in 35% more eroded C, but its feedback on the C cycle was marginal. Our results challenge the idea that higher erosion driven by climate will lead to a C sink in the near future.〈/p〉

    Description: 〈p〉〈i〉Artemisia annua〈/i〉 produces the valuable medicinal component, artemisinin, which is a sesquiterpene lactone widely used in malaria treatment. AaORA, a homolog of CrORCA3, which is involved in activating terpenoid indole alkaloid biosynthesis in 〈i〉Catharanthus roseus〈/i〉, is a jasmonate (JA)–responsive and trichome-specific APETALA2/ETHYLENE-RESPONSE FACTOR that plays a pivotal role in artemisinin biosynthesis. However, the JA signaling mechanism underlying AaORA-mediated artemisinin biosynthesis remains enigmatic. Here, we report that AaORA forms a transcriptional activator complex with AaTCP14 (TEOSINTE BRANCHED 1/CYCLOIDEA/PROLIFERATING CELL FACTOR 14), which is also predominantly expressed in trichomes. AaORA and AaTCP14 synergistically bind to and activate the promoters of two genes, 〈i〉double bond reductase 2〈/i〉 (〈i〉DBR2〈/i〉) and 〈i〉aldehyde dehydrogenase 1〈/i〉 (〈i〉ALDH1〈/i〉), both of which encode enzymes vital for artemisinin biosynthesis. AaJAZ8, a repressor of the JA signaling pathway, interacts with both AaTCP14 and AaORA and represses the ability of the AaTCP14-AaORA complex to activate the 〈i〉DBR2〈/i〉 promoter. JA treatment induces AaJAZ8 degradation, allowing the AaTCP14-AaORA complex to subsequently activate the expression of 〈i〉DBR2〈/i〉, which is essential for artemisinin biosynthesis. These data suggest that JA activation of the AaTCP14-AaORA complex regulates artemisinin biosynthesis. Together, our findings reveal a novel artemisinin biosynthetic pathway regulatory network and provide new insight into how specialized metabolism is modulated by the JA signaling pathway in plants.〈/p〉

    Description: 〈p〉We report the development of a novel platform to enhance the efficacy and safety of follicular lymphoma (FL) treatment. Since lymphoma is a clonal malignancy of a diversity system, every tumor has a different antibody on its cell surface. Combinatorial autocrine-based selection is used to rapidly identify specific ligands for these B cell receptors on the surface of FL tumor cells. The selected ligands are used in a chimeric antigen receptor T cell (CAR-T) format for redirection of human cytotoxic T lymphocytes. Essentially, the format is the inverse of the usual CAR-T protocol. Instead of being a guide molecule, the antibody itself is the target. Thus, these studies raise the possibility of personalized treatment of lymphomas using a private antibody binding ligand that can be obtained in a few weeks.〈/p〉

    Description: 〈p〉With the increasing resistance of many Gram-negative bacteria to existing classes of antibiotics, identifying new paradigms in antimicrobial discovery is an important research priority. Of special interest are the proteins required for the biogenesis of the asymmetric Gram-negative bacterial outer membrane (OM). Seven Lpt proteins (LptA to LptG) associate in most Gram-negative bacteria to form a macromolecular complex spanning the entire envelope, which transports lipopolysaccharide (LPS) molecules from their site of assembly at the inner membrane to the cell surface, powered by adenosine 5'-triphosphate hydrolysis in the cytoplasm. The periplasmic protein LptA comprises the protein bridge across the periplasm, which connects LptB〈sub〉2〈/sub〉FGC at the inner membrane to LptD/E anchored in the OM. We show here that the naturally occurring, insect-derived antimicrobial peptide thanatin targets LptA and LptD in the network of periplasmic protein-protein interactions required to assemble the Lpt complex, leading to the inhibition of LPS transport and OM biogenesis in 〈i〉Escherichia coli〈/i〉.〈/p〉

    Description: 〈p〉Several neurological disorders may benefit from gene therapy. However, even when using the lead vector candidate for intrathecal administration, adeno-associated virus serotype 9 (AAV9), the strength and distribution of gene transfer to the brain are inconsistent. On the basis of preliminary observations that standard intrathecal AAV9 infusions predominantly drive reporter gene expression in brain regions where gravity might cause cerebrospinal fluid to settle, we tested the hypothesis that counteracting vector "settling" through animal positioning would enhance vector delivery to the brain. When rats are either inverted in the Trendelenburg position or continuously rotated after intrathecal AAV9 infusion, we find (i) a significant 15-fold increase in the number of transduced neurons, (ii) a marked increase in gene delivery to cortical regions, and (iii) superior animal-to-animal consistency of gene expression. Entorhinal, prefrontal, frontal, parietal, hippocampal, limbic, and basal forebrain neurons are extensively transduced: 95% of transduced cells are neurons, and greater than 70% are excitatory. These findings provide a novel and simple method for broad gene delivery to the cortex and are of substantial relevance to translational programs for neurological disorders, including Alzheimer’s disease and related dementias, stroke, and traumatic brain injury.〈/p〉

    Description: 〈p〉We report the discovery of a large impact crater beneath Hiawatha Glacier in northwest Greenland. From airborne radar surveys, we identify a 31-kilometer-wide, circular bedrock depression beneath up to a kilometer of ice. This depression has an elevated rim that cross-cuts tributary subglacial channels and a subdued central uplift that appears to be actively eroding. From ground investigations of the deglaciated foreland, we identify overprinted structures within Precambrian bedrock along the ice margin that strike tangent to the subglacial rim. Glaciofluvial sediment from the largest river draining the crater contains shocked quartz and other impact-related grains. Geochemical analysis of this sediment indicates that the impactor was a fractionated iron asteroid, which must have been more than a kilometer wide to produce the identified crater. Radiostratigraphy of the ice in the crater shows that the Holocene ice is continuous and conformable, but all deeper and older ice appears to be debris rich or heavily disturbed. The age of this impact crater is presently unknown, but from our geological and geophysical evidence, we conclude that it is unlikely to predate the Pleistocene inception of the Greenland Ice Sheet.〈/p〉
