    Beschreibung: Syneruptive gas flux time series can, in principle, be retrieved from satellite maps of SO2 collected during and immediately after volcanic eruptions, and used to gain insights into the volcanic processes which drive the volcanic activity. Determination of the age and height of volcanic plumes are key prerequisites for such calculations. However, these parameters are challenging to constrain using satellite-based techniques. Here, we use imagery from OMI and GOME-2 satellite sensors and a novel numerical procedure based on back-trajectory analysis to calculate plume height as a function of position at the satellite measurement time together with plume injection height and time at a volcanic vent location. We applied this new procedure to three Etna eruptions (12 August 2011, 18 March 2012 and 12 April 2013) and compared our results with independent satellite and ground-based estimations. We also compare our injection height time-series with measurements of volcanic tremor, which reflects the eruption intensity, showing a good match between these two datasets. Our results are a milestone in progressing towards reliable determination of gas flux data from satellite-derived SO2 maps during volcanic eruptions, which would be of great value for operational management of explosive eruptions.

    Beschreibung: The performance of a linear depth inversion algorithm, cBathy, applied to coastal video imagery was assessed using observations of water depth from vessel-based hydrographic surveys and in-situ altimeters for a wide range of wave conditions (0.3 〈 significant wave height 〈 4.3 m) on a sandy Atlantic Ocean beach near Duck, North Carolina. Comparisons of video-based cBathy bathymetry with surveyed bathymetry were similar to previous studies (root mean square error (RMSE) = 0.75 m, bias = −0.26 m). However, the cross-shore locations of the surfzone sandbar in video-derived bathymetry were biased onshore 18–40 m relative to the survey when offshore wave heights exceeded 1.2 m or were greater than half of the bar crest depth, and broke over the sandbar. The onshore bias was 3–4 m when wave heights were less than 0.8 m and were not breaking over the sandbar. Comparisons of video-derived seafloor elevations with in-situ altimeter data at three locations onshore of, near, and offshore of the surfzone sandbar over ∼1 year provide the first assessment of the cBathy technique over a wide range of wave conditions. In the outer surf zone, video-derived results were consistent with long-term patterns of bathymetric change (r2 = 0.64, RMSE = 0.26 m, bias = −0.01 m), particularly when wave heights were less than 1.2 m (r2 = 0.83). However, during storms when wave heights exceeded 3 m, video-based cBathy over-estimated the depth by up to 2 m. Near the sandbar, the sign of depth errors depended on the location relative to wave breaking, with video-based depths overestimated (underestimated) offshore (onshore) of wave breaking in the surfzone. Wave speeds estimated by video-based cBathy at the initiation of wave breaking often were twice the speeds predicted by linear theory, and up to three times faster than linear theory during storms. Estimated wave speeds were half as fast as linear theory predictions at the termination of wave breaking shoreward of the sandbar. These results suggest that video-based cBathy should not be used to track the migration of the surfzone sandbar using data when waves are breaking over the bar nor to quantify morphological evolution during storms. However, these results show that during low energy conditions, cBathy estimates could be used to quantify seasonal patterns of seafloor evolution.

    Beschreibung: Completion of the Xenopus laevis genome sequence from inbred J strain animals has facilitated the generation of germline mutant X. laevis using targeted genome editing. In the last few years, numerous reports have demonstrated that TALENs are able to induce mutations in F0 Xenopus embryos, but none has demonstrated germline transmission of such mutations in X. laevis. In this report we used the oocyte host-transfer method to generate mutations in both tyrosinase homeologs and found highly-penetrant germline mutations; in contrast, embryonic injections yielded few germline mutations. We also compared the distribution of mutations in several F0 somatic tissues and germ cells and found that the majority of mutations in each tissue were different. These results establish that X. laevis J strain animals are very useful for generating germline mutations and that the oocyte host-transfer method is an efficient technique for generating mutations in both homeologs.

    Beschreibung: As the sampling frequency and resolution of Earth observation imagery increase, there are growing opportunities for novel applications in population monitoring. New methods are required to apply established analytical approaches to data collected from new observation platforms (e.g., satellites and unmanned aerial vehicles). Here, we present a method that estimates regional seasonal abundances for an understudied and growing population of gray seals (Halichoerus grypus) in southeastern Massachusetts, using opportunistic observations in Google Earth imagery. Abundance estimates are derived from digital aerial survey counts by adapting established correction-based analyses with telemetry behavioral observation to quantify survey biases. The result is a first regional understanding of gray seal abundance in the northeast US through opportunistic Earth observation imagery and repurposed animal telemetry data. As species observation data from Earth observation imagery become more ubiquitous, such methods provide a robust, adaptable, and cost-effective solution to monitoring animal colonies and understanding species abundances.

    Beschreibung: Injection of human Chorionic Gonadotropin (hCG) directly into the dorsal lymph sac of Xenopus is a commonly used protocol for induction of ovulation, but recent shortages in the stocks of commercially available hCG as well as lack of a well tested alternative have resulted in frustrating experimental delays in laboratories that predominantly use Xenopus in their research. Mammalian Luteinizing Hormones (LH) share structural similarity, functional equivalency, and bind the same receptor as hCG; this suggests that LH may serve as a good alternative to hCG for promoting ovulation in Xenopus. LH has been found to induce maturation of Xenopus oocytes in vitro, but whether it can be used to induce ovulation in vivo has not been examined. Here we compared the ability of four mammalian LH proteins, bovine (bLH), human (hLH), ovine (oLH), porcine (pLH), to induce ovulation in Xenopus when injected into the dorsal lymph sac of sexually mature females. We find that both ovine and human LH, but not bovine or porcine, are good substitutes for hCG for induction of ovulation in WT and J strain Xenopus laevis and Xenopus tropicalis.

    Beschreibung: The amphibian model Xenopus, has been used extensively over the past century to study multiple aspects of cell and developmental biology. Xenopus offers advantages of a non-mammalian system, including high fecundity, external development, and simple housing requirements, with additional advantages of large embryos, highly conserved developmental processes, and close evolutionary relationship to higher vertebrates. There are two main species of Xenopus used in biomedical research, Xenopus laevis and Xenopus tropicalis; the common perception is that both species are excellent models for embryological and cell biological studies, but only Xenopus tropicalis is useful as a genetic model. The recent completion of the Xenopus laevis genome sequence combined with implementation of genome editing tools, such as TALENs (transcription activator-like effector nucleases) and CRISPR-Cas (clustered regularly interspaced short palindromic repeats-CRISPR associated nucleases), greatly facilitates the use of both Xenopus laevis and Xenopus tropicalis for understanding gene function in development and disease. In this paper, we review recent advances made in Xenopus laevis and Xenopus tropicalis with TALENs and CRISPR-Cas and discuss the various approaches that have been used to generate knockout and knock-in animals in both species. These advances show that both Xenopus species are useful for genetic approaches and in particular counters the notion that Xenopus laevis is not amenable to genetic manipulations.

    Beschreibung: Stated preference scenarios often describe outcomes to be valued in terms of intermediate biophysical processes or ecosystem services with indirect utility effects, rather than in terms of final, directly welfare-relevant consequences. This article evaluates whether valid welfare estimates can emerge from this practice. We begin with a theoretical model demonstrating conditions under which stated preference scenarios that include intermediate outcomes will elicit welfare estimates identical to those from parallel scenarios that include associated final outcomes (i.e., convergent validity will hold). The model demonstrates that a necessary condition for convergent validity is the ability of respondents to correctly predict biophysical production functions linking intermediate to final outcomes. Hypotheses from the theoretical model are then evaluated empirically using an application of choice experiments to migratory fish restoration in the U.S. state of Rhode Island. Empirical results are mixed but generally reject convergent validity; welfare estimates are not robust to the use of an intermediate outcome in lieu of a related final outcome in stated preference scenarios, as predicted by theory. Results of the analysis suggest that greater attention should be given to the reliability of welfare estimation when final outcomes cannot be quantified.

    Beschreibung: Resource managers must often make difficult choices in the face of imperfectly observed and dynamically changing systems (e.g., livestock, fisheries, water, and invasive species). A rich set of techniques exists for identifying optimal choices when that uncertainty is assumed to be understood and irreducible. Standard optimization approaches, however, cannot address situations in which reducible uncertainty applies to either system behavior or environmental states. The adaptive management literature overcomes this limitation with tools for optimal learning, but has been limited to highly simplified models with state and action spaces that are discrete and small. We overcome this problem by using a recently developed extension of the Partially Observable Markov Decision Process (POMDP) framework to allow for learning about a continuous state. We illustrate this methodology by exploring optimal control of bovine tuberculosis in New Zealand cattle. Disease testing—the control variable—serves to identify herds for treatment and provides information on prevalence, which is both imperfectly observed and subject to change due to controllable and uncontrollable factors. We find substantial efficiency losses from both ignoring learning (standard stochastic optimization) and from simplifying system dynamics (to facilitate a typical, simple learning model), though the latter effect dominates in our setting. We also find that under an adaptive management approach, simplifying dynamics can lead to a belief trap in which information gathering ceases, beliefs become increasingly inaccurate, and losses abound.
