    Beschreibung: We propose an equivalence class of nonstationary Gaussian stochastic processes defined in the wavelet domain. These processes are characterized by means of wavelet multipliers and exhibit well-defined time-dependent spectral properties. They allow one to generate realizations of any wavelet spectrum. Based on this framework, we study the estimation of continuous wavelet spectra, i.e., we calculate variance and bias of arbitrary estimated continuous wavelet spectra. Finally, we develop an areawise significance test for continuous wavelet spectra to overcome the difficulties of multiple testing; it uses basic properties of continuous wavelet transform to decide whether a pointwise significant result is a real feature of the process or indistinguishable from typical stochastic fluctuations. This test is compared to the conventional one in terms of sensitivity and specificity. A software package for continuous wavelet spectral analysis and synthesis is presented.

    Beschreibung: Presented here is the complete genome sequence of Thiomicrospira crunogena XCL-2, representative of ubiquitous chemolithoautotrophic sulfur-oxidizing bacteria isolated from deep-sea hydrothermal vents. This gammaproteobacterium has a single chromosome (2,427,734 base pairs), and its genome illustrates many of the adaptations that have enabled it to thrive at vents globally. It has 14 methyl-accepting chemotaxis protein genes, including four that may assist in positioning it in the redoxcline. A relative abundance of coding sequences (CDSs) encoding regulatory proteins likely control the expression of genes encoding carboxysomes, multiple dissolved inorganic nitrogen and phosphate transporters, as well as a phosphonate operon, which provide this species with a variety of options for acquiring these substrates from the environment. Thiom. crunogena XCL-2 is unusual among obligate sulfur-oxidizing bacteria in relying on the Sox system for the oxidation of reduced sulfur compounds. The genome has characteristics consistent with an obligately chemolithoautotrophic lifestyle, including few transporters predicted to have organic allocrits, and Calvin-Benson-Bassham cycle CDSs scattered throughout the genome.

    Beschreibung: The first detailed quantitative observations of the two-dimensional collapse of a granular column along a horizontal channel are presented for a variety of materials. Together with the complementary study for the axisymmetric situation, we conclude that for granular collapses the generally accepted approaches, that are highly dependent on frictional parameters, do not describe the main flow phenomena. The motion divides in two main flow regimes at a∼1.8, where the aspect ratio a=hi∕di and hi and di are the initial height and width of the column. We describe the details of collapse by emphasizing the sequential occurrence of a main spreading followed by a final avalanching phase. For the low a regime, a〈1.8, we derive descriptions of the final geometry by direct physical arguments. For the large a regime, a〉1.8, we determine that nearly all details of the collapse, including the position of the flow front as a function of time, the emplacement time, the self-similar final profiles, and especially their maximum vertical and horizontal extension, are established during the spreading phase and can be expressed in terms of the initial geometrical parameters but are independent of basal and internal friction parameters.

    Beschreibung: Proteins containing the PB1 domain, a protein interaction module conserved in animals, fungi, amoebas, and plants, participate in diverse biological processes. The PB1 domains adopt a ubiquitin-like β-grasp fold, containing two α helices and a mixed five-stranded β sheet, and are classified into groups harboring an acidic OPCA motif (type I), the invariant lysine residue on the first β strand (type II), or both (type I/II). The OPCA motif of a type I PB1 domain forms salt bridges with basic residues, especially the conserved lysine, of a type II PB1 domain, thereby mediating a specific PB1-PB1 heterodimerization, whereas additional contacts contribute to high affinity and specificity of the modular interaction. The canonical PB1 dimerization is required for the formation of complexes between p40phox and p67phox (for activation of the NADPH oxidase crucial for mammalian host defense), between the scaffold Bem1 and the guanine nucleotide exchange factor Cdc24 (for polarity establishment in yeasts), and between the polarity protein Par6 and atypical protein kinase C (for cell polarization in animal cells), as well as for the interaction between the mitogen-activated protein kinase kinase kinases MEKK2 or MEKK3 and the downstream target mitogen-activated protein kinase kinase MEK5 (for early cardiovascular development in mammals). PB1 domains can also mediate interactions with other protein domains. For example, an intramolecular interaction between the PB1 and PX domains of p40phox regulates phagosomal targeting of the microbicidal NADPH oxidase; the PB1 domain of MEK5 is likely responsible for binding to the downstream kinase ERK5, which lacks a PB1 domain; and the scaffold protein Nbr1 associates through a PB1-containing region with titin, a sarcomere protein without a PB1 domain. This Review describes various aspects of PB1 domains at the molecular and cellular levels.

    Beschreibung: Mass budget calculations, validated with satellite gravity observations [from the Gravity Recovery and Climate Experiment (GRACE) satellites], enable us to quantify the individual components of recent Greenland mass loss. The total 2000–2008 mass loss of ~1500 gigatons, equivalent to 0.46 millimeters per year of global sea level rise, is equally split between surface processes (runoff and precipitation) and ice dynamics. Without the moderating effects of increased snowfall and refreezing, post-1996 Greenland ice sheet mass losses would have been 100% higher. Since 2006, high summer melt rates have increased Greenland ice sheet mass loss to 273 gigatons per year (0.75 millimeters per year of equivalent sea level rise). The seasonal cycle in surface mass balance fully accounts for detrended GRACE mass variations, confirming insignificant subannual variation in ice sheet discharge.

    Beschreibung: Background During the Ordovician the global diversity increased dramatically at family, genus and species levels. Partially the diversification is explained by an increased nutrient, and phytoplankton availability in the open water. Cephalopods are among the top predators of todays open oceans. Their Ordovician occurrences, diversity evolution and abundance pattern potentially provides information on the evolution of the pelagic food chain. Methodology/Principal Findings We reconstructed the cephalopod departure from originally exclusively neritic habitats into the pelagic zone by the compilation of occurrence data in offshore paleoenvironments from the Paleobiology Database, and from own data, by evidence of the functional morphology, and the taphonomy of selected cephalopod faunas. The occurrence data show, that cephalopod associations in offshore depositional settings and black shales are characterized by a specific composition, often dominated by orthocerids and lituitids. The siphuncle and conch form of these cephalopods indicate a dominant lifestyle as pelagic, vertical migrants. The frequency distribution of conch sizes and the pattern of epibionts indicate an autochthonous origin of the majority of orthocerid and lituitid shells. The consistent concentration of these cephalopods in deep subtidal sediments, starting from the middle Tremadocian indicates the occupation of the pelagic zone early in the Early Ordovician and a subsequent diversification which peaked during the Darriwilian. Conclusions/Significance The exploitation of the pelagic realm started synchronously in several independent invertebrate clades during the latest Cambrian to Middle Ordovician. The initial rise and diversification of pelagic cephalopods during the Early and Middle Ordovician indicates the establishment of a pelagic food chain sustainable enough for the development of a diverse fauna of large predators. The earliest pelagic cephalopods were slowly swimming vertical migrants. The appearance and early diversification of pelagic cephalopods is interpreted as a consequence of the increased food availability in the open water since the latest Cambrian.
