    Description: Without some form of regenerative life support system, long duration space habitation or travel will be limited severely by the prohibitive costs of resupplying air, water, and food from Earth. Components under consideration for inclusion in a regenerative life support system are based on either physicochemical or biological processes. Physicochemical systems would use filtration and elemental phase changes to convert waste materials into usable products, while biological systems would use higher plants and bioreactors to supply crew needs. Neither a purely biological nor strictly a physicochemical approach can supply all crew needs, thus, the best each approach can offer will be combined into a hybrid regenerative life support system. Researchers at Kennedy Space Center (KSC) Advanced Life Support Breadboard Project have taken the lead on bioregenerative aspects of space life support. The major focus has been on utilization of higher plants for production of food, oxygen, and clean water. However, a key to any regenerative life support system is recycling and recovery of resources (wastes). In keeping with the emphasis at KSC on bioregenerative systems and with the focus on plants, this paper focuses on research with biologically-based options for resource recovery from inedible crop residues.

    Description: A large amount of inedible plant material, generated as a result of plant growth in a Controlled Ecological Life Support System (CELSS), should be pretreated and converted into forms that can be recycled on earth as well as in space. The main portion of the inedible biomass is lignocellulosic material. Enzymatic hydrolysis of this cellulose would provide sugars for many other uses by recycling carbon, hydrogen, oxygen, and nitrogen through formation of carbon dioxide, heat, and sugars, which are potential foodstuffs. To obtain monosaccharides from cellulose, the protective effect of lignin should be removed. White-rot fungi degrade lignin more extensively and rapidly than other microorganisms. Pleurotus ostreatus degrades lignin effectively, and produces edible and flavorful mushrooms that increase the quality and nutritional value of the diet. This mushroom is also capable of metabolizing hemicellulose, thereby providing a food use of this pentose containing polysaccharide. This study presents the current knowledge of physiology and biochemistry of primary and secondary metabolisms of basidiomycetes, and degradation mechanism of lignin. A better understanding of the ligninolytic activity of white-rot fungi will impact the CELSS Program by providing insights on how edible fungi might be used to recycle the inedible portions of the crops.

    Description: Although terrestrial CO2 concentrations, [CO2] are not expected to reach 1000 micromoles mol-1 for many decades, CO2 levels in closed systems such as growth chambers and glasshouses, can easily exceed this concentration. CO2 levels in life support systems in space can exceed 10000 micromoles mol-1 (1%). Here we studied the effect of six CO2 concentrations, from ambient up to 10000 micromoles mol-1, on seed yield, growth and gas exchange of two wheat cultivars (USU-Apogee and Veery-l0). Elevating [CO2] from 350 to 1000 micromoles mol-1 increased seed yield (by 33%), vegetative biomass (by 25%) and number of heads m-2 (by 34%) of wheat plants. Elevation of [CO2] from 1000 to 10000 micromoles mol-1 decreased seed yield (by 37%), harvest index (by 14%), mass per seed (by 9%) and number of seeds per head (by 29%). This very high [CO2] had a negligible, non-significant effect on vegetative biomass, number of heads m-2 and seed mass per head. A sharp decrease in seed yield, harvest index and seeds per head occurred by elevating [CO2] from 1000 to 2600 micromoles mol-1. Further elevation of [CO2] from 2600 to 10000 micromoles mol-1 caused a further but smaller decrease. The effect of CO2 on both wheat cultivars was similar for all growth parameters. Similarly there were no differences in the response to high [CO2] between wheat grown hydroponically in growth chambers under fluorescent lights and those grown in soilless media in a glasshouse under sunlight and high pressure sodium lamps. There was no correlation between high [CO2] and ethylene production by flag leaves or by wheat heads. Therefore, the reduction in seed set in wheat plants is not mediated by ethylene. The photosynthetic rate of whole wheat plants was 8% lower and dark respiration of the wheat heads 25% lower when exposed to 2600 micromoles mol-1 CO2 compared to ambient [CO2]. It is concluded that the reduction in the seed set can be mainly explained by the reduction in the dark respiration in wheat heads, when most of the respiration is functional and is needed for seed development.

    Description: In a study of lunar and Mars settlement concepts, an analysis was made of fundamental design assumptions in five technical areas against a model list of occupational and environmental health concerns. The technical areas included the proposed science projects to be supported, habitat and construction issues, closed ecosystem issues, the "MMM" issues (mining, material processing, and manufacturing), and the human elements of physiology, behavior, and mission approach. Four major lessons were learned. First it is possible to relate public health concerns to complex technological development in a proactive design mode, which has the potential for long-term cost savings. Second, it became very apparent that prior to committing any nation or international group to spending the billions to start and complete a lunar settlement, over the next century, that a significantly different approach must be taken from those previously proposed, to solve the closed ecosystem and "MMM" problems. Third, it also appears that the health concerns and technology issues to be addressed for human exploration into space are fundamentally those to be solved for human habitation of the Earth (as a closed ecosystem) in the 21st century. Finally, it is proposed that ecosystem design modeling must develop new tools, based on probabilistic models as a step up from closed circuit models.

    Description: The requirements for a human life support system for long-duration space missions are reviewed. The system design of a controlled ecological life support system is briefly described, followed by a more detailed account of the study of the conceptual design of a Lunar Based CELSS. The latter is to provide a safe, reliable, recycling lunar base life support system based on a hybrid physicochemical/biological representative technology. The most important conclusion reached by this study is that implementation of a completely recycling CELSS approach for a lunar base is not only feasible, but eminently practical. On a cumulative launch mass basis, a 4-person Lunar Base CELSS would pay for itself in approximately 2.6 years relative to a physicochemical air/water recycling system with resupply of food from the Earth. For crew sizes of 30 and 100, the breakeven point would come even sooner, after 2.1 and 1.7 years, respectively, due to the increased mass savings that can be realized with the larger plant growth units. Two other conclusions are particularly important with regard to the orientation of future research and technology development. First, the mass estimates of the Lunar Base CELSS indicate that a primary design objective in implementing this kind of system must be to minimized the mass and power requirement of the food production plant growth units, which greatly surpass those of the other air and water recycling systems. Consequently, substantial research must be directed at identifying ways to produce food more efficiently. On the other hand, detailed studies to identify the best technology options for the other subsystems should not be expected to produce dramatic reductions in either mass or power requirement of a Lunar Base CELSS. The most crucial evaluation criterion must, therefore, be the capability for functional integration of these technologies into the ultimate design of the system. Secondly, this study illustrates that existing or near-term technologies are adequate to implement a Lunar Base CELSS. There are no apparent "show-stoppers" which require the development of new technologies. However, there are several areas in which new materials and technologies could be used for a more efficient implementation of the system, e.g., by decreasing mass or power requirement and increasing recycling efficiency. These areas must be further addressed through research and development. Finally, although this study focused on the development of a Lunar Base CELSS, the same technologies and a nearly identical design would be appropriate for a Mars base. Actually, except for the distance of transportation, the implementation of a CELSS on Mars would even be easier than it would be on the Moon. The presence of atmospheric CO2 on Mars, although in low concentration, coupled with the fact that the day/night cycle on Mars is very similar to that on Earth, makes the use of light-weight, greenhouse-like structures for growing food plants even more feasible than on the Moon. There are some environmental problems, which would have to be dealt with, like dust storms and the large amount of the ultraviolet radiation incident on the planet's surface. However, the materials and methods are largely available today to develop such a life support system for a Mars base.

    Description: Pepper plants (Capsicum annuum L. cv., Hungarian Wax) were grown under metal halide (MH) lamps or light-emitting diode (LED) arrays with different spectra to determine the effects of light quality on plant anatomy of leaves and stems. One LED (660) array supplied 90% red light at 660 nm (25nm band-width at half-peak height) and 1% far-red light between 700-800nm. A second LED (660/735) array supplied 83% red light at 660nm and 17% far-red light at 735nm (25nm band-width at half-peak height). A third LED (660/blue) array supplied 98% red light at 660nm, 1% blue light between 350-550nm, and 1% far-red light between 700-800nm. Control plants were grown under broad spectrum metal halide lamps. Plants were gron at a mean photon flux (300-800nm) of 330 micromol m-2 s-1 under a 12 h day-night photoperiod. Significant anatomical changes in stem and leaf morphologies were observed in plants grown under the LED arrays compared to plants grown under the broad-spectrum MH lamp. Cross-sectional areas of pepper stems, thickness of secondary xylem, numbers of intraxylary phloem bundles in the periphery of stem pith tissues, leaf thickness, numbers of choloplasts per palisade mesophyll cell, and thickness of palisade and spongy mesophyll tissues were greatest in peppers grown under MH lamps, intermediate in plants grown under the 660/blue LED array, and lowest in peppers grown under the 660 or 660/735 LED arrays. Most anatomical features of pepper stems and leaves were similar among plants grown under 660 or 660/735 LED arrays. The effects of spectral quality on anatomical changes in stem and leaf tissues of peppers generally correlate to the amount of blue light present in the primary light source.

    Description: Researchers report the results of experiments conducted onboard MIR in 1990, 1995, and 1996 in raising edible crops. In the 1990 experiment, radishes and Chinese cabbage were grown successfully, though the experimental plants were up to four times smaller than controls at harvest. The 1995 experiment in growing wheat through a complete life cycle was not completed. The 1996 experiment was successful in growing wheat through a complete life cycle to the seed stage. No seeds developed on any of the 279 ears harvested in that experiment. Reasons for the seedless development are explored.

    Description: The maintenance of optimal levels of oxygen in the gaseous environment of a plant growth chamber during light and dark periods is an essential criterion for the correct growth of plants. The use of solid oxide electrolyzers to control the oxygen levels by removing the excess gaseous oxygen during periods of illumination and full-scale photosynthesis is described. A part of the oxygen removed can be stored and supplied back to the plants during dark periods. The excess oxygen can be used by the crew. The electrolizer can be additionally used in its open circuit mode, to sense the oxygen concentrations in the plant chamber. The solid oxide electrolysis process is described.

    Description: The influences of atmospheric carbon dioxide on the fractionation of carbon isotopes and the magnesium incorporation into biogenic marine calcite were investigated using samples of the calcareous alga Amphiroa and benthic foraminifer Sorites grown in the Biosphere 2 Ocean system under variable atmospheric CO2 concentrations (approximately 500 to 1200 ppm). Carbon isotope fractionation was studied in both the organic matter and the skeletal carbonate. Magnesium analysis was to be performed on the carbonate removed during decalcification. These data have not been collected due to technical problems. Carbon isotope data from Amphiroa yields a linear relation between [CO2] and Delta(sup 13)C(sub Corg)values suggesting that the fractionation of carbon isotopes during photosynthesis is positively correlated with atmospheric [CO2]. [CO2] and Delta(sup 13)C(sub Corg) values for Sorites produce a relation that is best described by a hyperbolic function where Delta(sup 13)C(sub Corg) values increase between 300 and 700 ppm and decrease from 700 to 1200 ppm. Further investigation of this relation and Sorites physiology is needed.

    Description: Extensive use and reliance on laboratory fume hoods exist at LeRC for the control of chemical hazards (nearly 175 fume hoods). Flow-measuring devices are necessary to continually monitor hood performance. The flow-measuring device should he tied into an energy management control system to detect problems at a central location without relying on the users to convey information of a problem. Compatibility concerns and limitations should always be considered when choosing the most effective flow-measuring device for a particular situation. Good practice on initial hood design and placement will provide a system for which a flow-measuring device may be used to its full potential and effectiveness.

    Description: This project presents guidelines for the integration of the Intelligent Crop Scheduler (ICS) developed by Leon (1995) with a version of the Biological Life Support System (BLSS) simulation based on the work by Volk and Rummel (1987). These guidelines will also aid in defining the appropriate model detail of simulation-based schedulers. ICS determines what, when and how much to plant of different crops. These decisions are made such that the critical reservoir levels are maintained close to their nominal or desired settings during the duration of the mission. Initial feasibility of the approach has been demonstrated using simplified implementations of the scheduling approach and CELSS 'world' model developed by the investigator at JSC in the Summer of 1995. The BLSS model incorporates more detail. The increased fidelity is in terms of added mass-regeneration formalism (biochemical stoichiometry), more plants, and more accurate modeling of the mechanical system. This report describes the main features of the Crop scheduler and CELSS simulator, discusses integration issues, and provides with detailed guidelines for the integration of these two applications.

    Description: The Marsh Biome, which was modeled after the mangroves and marshes of southwest Florida, has an area of 441.2 sq m separated into three hydrologically independent sections: the Freshwater, Oligohaline and Salt Marshes. The divisions are made based on their salinity (approximately 0, 4, and 34 ppt. respectively), but they also contain different biological communities. The Freshwater and Oligohaline Marshes are mostly filled with various grasses and several trees, while the Salt Marsh houses regions of red, black, and white mangroves (Rhizophora mangle, Avicennia germinans, and Languncularia racemosa respectively). Overall, there are an estimated 80 species of plants within the biome. Water in the Salt Marsh follows a meandering stream from the algal turf scrubbers (apparatuses that clean the water of its nutrients and heavy metals while increasing dissolved oxygen levels) which have an outlet in the Salt Marsh section near sites 4 and 5 to the Fringing Red Mangrove section. The sections of the Salt Marsh are separated by walls of concrete with openings to allow the stream to flow through. Throughout this study, conducted through the months of June and July, many conditions within the biome remained fairly constant. The temperature was within a degree or two of 25 C, mostly depending on whether the sample site was in direct sunlight or shaded. The pH throughout the Salt Marsh was 8.0 +/- 0.2, and the lower salinity waters only dropped below this soon after rains. The water rdepth and dissolved oxygen varied, however, between sites.

    Description: In recent decades, the concentration of methane in the earth's atmosphere increased 1-2% annually. It's rate of increases, combined with methane's effectiveness as a greenhouse gas, has led to an intensive research effort to determine the sources and sinks of the gas in the environment. Biosphere 2 offers a unique opportunity to contribute to the effort because it lacks a major photochemical sink present in the Earth's atmosphere. Researchers can therefore concentrate on biological processes involved in methane cycles. Wetlands are a large source of atmospheric methane, due to anoxic conditions in the sediments and the abundance of organic materials. In order to determine if these conditions in Biosphere 2 also promote methane production, this study looked for the fluxes of methane and methods of transport of the gas from from the water and sediments to the atmosphere in the Marsh Biome. Fluxes of methane from the sediments and waters were measured using static chambers, peepers, and leaf bags. Fluxes and vertical profiles of methane in the sediments show that substantial amounts of methane are being produced in the marsh and are being transported into the Biosphere 2 environment.

    Description: This paper presents an alternative concept that employs existing technologies and off-the-shelf components to liquefy gaseous oxygen from an in-situ propellant production (ISPP) unit and to store the liquid oxygen without boil-off loss. A primary goal is to minimize active components with a secondary goal of designing the active component in a protected or failure-free environment. The resulting design requires only one active component, a compressor operating in a closed and consequently more protected system. The design avoids pumps as active components by employing cryopumping for gaseous oxygen (GOX) and gravity transfer for liquid oxygen (LOX).

    Description: As the environmental changes occur throughout the world in rapid rate, we need to have further understandings for our planet. Since the ecosystems are so complex, it is almost impossible for us to integrate every factor. However, mathematical models are powerful tools which can be used to simulate those ecosystems with limited data. In this project, I collected light intensity, canopy leaf temperature and Air Handler (AHU) temperature, and nitrogen concentration in the leaves for different profiles in the rainforest mesocosm. These data will later be put into mathematical models such as "big-leaf" and "sun/shade" models to determine how these factors will affect CO2 exchange in the rainforest. As rainforests are diminishing from our planet and their existence is very important for all living things on earth, it is necessary for us to learn more about the unique system of rainforests and how we can co-exist rather than destroy.

    Description: To meet NASA's objective of using the unique aspects of the space environment to expand fundamental knowledge in the biological sciences, the Space Station Biological Research Project at Ames Research Center is developing, or providing oversight, for two major suites of hardware which will be installed on the International Space Station (ISS). The first, the Gravitational Biology Facility, consists of Habitats to support plants, rodents, cells, aquatic specimens, avian and reptilian eggs, and insects and the Habitat Holding Rack in which to house them at microgravity; the second, the Centrifuge Facility, consists of a 2.5 m diameter centrifuge that will provide acceleration levels between 0.01 g and 2.0 g and a Life Sciences Glovebox. These two facilities will support the conduct of experiments to: 1) investigate the effect of microgravity on living systems; 2) what level of gravity is required to maintain normal form and function, and 3) study the use of artificial gravity as a countermeasure to the deleterious effects of microgravity observed in the crew. Upon completion, the ISS will have three complementary laboratory modules provided by NASA, the European Space Agency and the Japanese space agency, NASDA. Use of all facilities in each of the modules will be available to investigators from participating space agencies. With the advent of the ISS, space-based gravitational biology research will transition from 10-16 day short-duration Space Shuttle flights to 90-day-or-longer ISS increments.

    Description: Calcium bioavailability of vegetarian diets containing various proportions of candidate crops for a controlled ecological life-support system (CELSS) was determined by femur 45Ca uptake. Three vegetarian diets and a control diet were labeled extrinsically with 45Ca and fed to 5-wk old male rats. A fifth group of rats fed an unlabeled control diet received an intraperitoneal (IP) injection of 45Ca. There was no significant difference in mean calcium absorption of vegetarian diets (90.80 +/- 5.23%) and control diet (87.85 +/- 5.25%) when calculated as the percent of an IP dose. The amounts of phytate, oxalate, and dietary fiber in the diets did not affect calcium absorption.

    Description: A study evaluating alternative methods for long term operation of biomass production systems was recently completed at the Kennedy Space Center (KSC). The 418-day study evaluated repeated batch versus mixed-aged production of potato grown on either standard 1/2-strength Hoagland's nutrient solution or solutions including nutrients recycled from inedible plant material. The long term effects of closure and recycling on microbial dynamics were evaluated by monitoring the microbial communities associated with various habitats within the plant growth system (i.e., plant roots, nutrient solution, biofilms within the hydroponic systems, atmosphere, and atmospheric condensate). Plate count methods were used to enumerate and characterize microorganisms. Microscopic staining methods were used to estunate total cell densities. The primary finding was that the density and composition of microbial communities associated with controlled environmental plant growth systems are stable during long term operation. Continuous production resulted in slightly greater stability. Nutrient recycling, despite the addition of soluble organic material from the waste processing system, did not significantly increase microbial density in any of the habitats.

    Description: Testing of the International Space Station (ISS) U.S. Segment baseline configuration of the Atmosphere Revitalization Subsystem (ARS) by NASA's Marshall Space Flight Center (MSFC) was conducted as part of the Environmental Control and Life Support System (ECLSS) design and development program. This testing was designed to answer specific questions regarding the control and performance of the baseline ARS subassemblies in the ISS U.S. Segment configuration. These questions resulted from the continued maturation of the ISS ECLSS configuration and design requirement changes since 1992. The test used pressurized oxygen injection, a mass spectrometric major constituent analyzer, a Four-Bed Molecular Sieve Carbon Dioxide Removal Assembly, and a Trace Contaminant Control Subassembly to maintain the atmospheric composition in a sealed chamber at ISS specifications for 30 days. Human metabolic processes for a crew of four were simulated according to projected ISS mission time lines. The performance of a static feed water electrolysis Oxygen Generator Assembly was investigated during the test preparation phases; however, technical difficulties prevented its use during the integrated test. The Integrated ARS Test (IART) program built upon previous closed-door and open-door integrated testing conducted at MSFC between 1987 and 1992. It is the most advanced test of an integrated ARS conducted by NASA to demonstrate its end-to-end control and overall performance. IART test objectives, facility design, pretest analyses, test and control requirements, and test results are presented.

    Description: In this report we describe the details of our empirical protocol effort investigating skill in extravehicular mass handling using NASA's principal mass handling simulator, the precision air bearing floor. Contents of this report include a description of the necessary modifications to the mass handling simulator; choice of task, and the description of an operationally relevant protocol. Our independent variables are presented in the context of the specific operational issues they were designed to simulate. The explanation of our dependent variables focuses on the specific data processing procedures used to transform data from common laboratory instruments into measures that are relevant to a special class of nested control systems (discussed in Volume 1): manual interactions between an individual and the substantial environment. The data reduction is explained in the context of the theoretical foundation described in Volume 1. Finally as a preface to the presentation of the empirical data in Volume 3 of this report series, a set of detailed hypotheses is presented.

    Description: Phase 2 of the Lunar-Mars Life Support Test Project was conducted in June and July of 1996 at the NASA Johnson Space Center. The primary objective of Phase 2 was to demonstrate and evaluate an integrated physicochemical air revitalization and regenerative water recovery system capable of sustaining a human crew of four for 30 days inside a closed chamber. The crew (3 males and 1 female) was continuously present inside a chamber throughout the 30-day test. The objective of this paper was to describe crew interactions and human factors for the test. Crew preparations for the test included training and familiarization of chamber systems and accommodations, and medical and psychological evaluations. During the test, crew members provided metabolic loads for the life support systems, performed maintenance on chamber systems, and evaluated human factors inside the chamber. Overall, the four crew members found the chamber to be comfortable for the 30-day test. The crew performed well together and this was attributed in part to team dynamics, skill mix (one commander, two system experts, and one logistics lead), and a complementary mix of personalities. Communication with and support by family, friends, and colleagues were identified as important contributors to the high morale of the crew during the test. Lessons learned and recommendations for future testing are presented by the crew in this paper.

    Description: Collection and analysis of spacecraft cabin air samples are necessary to assess the cabin air quality with respect to crew health. Both toxicology and engineering disciplines work together to achieve an acceptably clean cabin atmosphere. Toxicology is concerned with limiting the risk to crew health from chemical sources, setting exposure limits, and analyzing air samples to determine how well these limits are met. Engineering provides the means for minimizing the contribution of the various contaminant generating sources by providing active contamination control equipment on board spacecraft and adhering to a rigorous material selection and control program during the design and construction of the spacecraft. A review of the rationale and objectives for sampling spacecraft cabin atmospheres is provided. The presently-available sampling equipment and methods are reviewed along with the analytical chemistry methods employed to determine trace contaminant concentrations. These methods are compared and assessed with respect to actual cabin air quality monitoring needs. Recommendations are presented with respect to the basic sampling program necessary to ensure an acceptably clean spacecraft cabin atmosphere. Also, rationale and recommendations for expanding the scope of the basic monitoring program are discussed.

    Description: The space operations incident reporting tool (SOIRT) is an instrument used to record information about an anomaly occurring during flight which may have been due to insufficient and/or inappropriate application of human factors knowledge. We originally developed the SOIRT form after researching other incident reporting systems of this type. We modified the form after performing several in-house reviews and a pilot test to access usability. Finally, crew members from Space Shuttle flights participated in a usability test of the tool after their missions. Since the National Aeronautics and Space Administration (NASA) currently has no system for continuous collection of this type of information, the SOIRT was developed to report issues such as reach envelope constraints, control operation difficulties, and vision impairments. However, if the SOIRT were to become a formal NASA process, information from crew members could be collected in a database and made available to individuals responsible for improving in-flight safety and productivity. Potential benefits include documentation to justify the redesign or development of new equipment/systems, provide the mission planners with a method for identifying past incidents, justify the development of timelines and mission scenarios, and require the creation of more appropriate work/rest cycles.

    Description: It is widely accepted that human error is a major contributing factor in aircraft accidents. There has been a significant amount of research in why these errors occurred, and many reports state that the design of flight deck can actually dispose humans to err. This research has led to the call for changes in design according to human factors and human-centered principles. The National Aeronautics and Space Administration's (NASA) Langley Research Center has initiated an effort to design a human-centered flight deck from a clean slate (i.e., without constraints of existing designs.) The effort will be based on recent research in human-centered design philosophy and mission management categories. This design will match the human's model of the mission and function of the aircraft to reduce unnatural or non-intuitive interfaces. The product of this effort will be a flight deck design description, including training and procedures, and a cross reference or paper trail back to design hypotheses, and an evaluation of the design. The present paper will discuss the philosophy, process, and status of this design effort.

    Description: The Autogenic-Feedback Training Exercise (AFTE) method of the present invention is a combined application of physiologic and perceptual training techniques. such as autogenic therapy and biofeedback. This combined therapy approach produces a methodology that is appreciably more effective than either of the individual techniques used separately. The AFTE method enables sufficient magnitude of control necessary to significantly reduce the behavioral and physiologic reactions to severe environmental stressors. It produces learned effects that are persistent over time and are resistant to extinction and it can be administered in a short period of time. The AFTE method may be used efficiently in several applications, among which are the following: to improve pilot and crew performance during emergency flying conditions; to train people to prevent the occurrence of nausea and vomiting associated with motion and sea sickness, or morning sickness in early pregnancy; as a training method for preventing or counteracting air-sickness symptoms in high-performance military aircraft; for use as a method for cardiovascular training, as well as for multiple other autonomic responses, which may contribute to the alleviation of Space Motion Sickness (SMS) in astronauts and cosmonauts; training people suffering from migraine or tension headaches to control peripheral blood flow and reduce forehead and/or trapezius muscle tension; training elderly people suffering from fecal incontinence to control their sphincter muscles; training cancer patients to reduce the nauseagenic effects of chemotherapy; and training patients with Chronic Intestinal Pseudo-obstruction (CIP).

    Description: Helmet Mounted Displays (HMDs) present flight, navigation, and weapon information in the pilot's line of sight. The HMD was developed to allow the pilot to retain aircraft and weapon information while looking off boresight. This document reviews current state of the art in HMDs and presents a design guide for the HMD engineer in identifying several critical HMD issues: symbol stabilization, inadequate definitions, undefined symbol drive laws, helmet considerations, and Field Of View (FOV) vs. resolution tradeoff requirements. In particular, display latency is a key issue for HMDs. In addition to requiring further experimental studies, it impacts the definition and control law issues. Symbol stabilization is also critical. In the case of the Apache helicopter, the lack of compensation for pilot head motion creates excessive workload during hovering and Nap Of the Earth (NOE) flight. This translates into excessive training requirements. There is no agreed upon set of definitions or descriptions for how HMD symbols are driven to compensate for pilot head motion. A set of definitions is proposed to address this. There are several specific areas where simulation and flight experiments are needed: development of hover and NOE symbologies which compensate for pilot head movement; display latency and sampling, and the tradeoff between FOV, sensor resolution and symbology.

    Description: At the present time, myoelectric prostheses perform only one function of the hand: open and close with the thumb, index and middle finger coming together to grasp various shaped objects. To better understand the limitations of the current single-function prostheses and the needs of the individuals who use them, The Institute for Rehabilitation and Research (TIRR), sponsored by the National Institutes of Health (August 1992 - November 1994), surveyed approximately 2500 individuals with upper limb loss. When asked to identify specific features of their current electric prosthesis that needed improvement, the survey respondents overwhelmingly identified the lack of wrist and finger movement as well as poor control capability. Simply building a mechanism with individual finger and wrist motion is not enough. Individuals with upper limb loss tend to reject prostheses that require continuous visual monitoring and concentration to control. Robotics researchers at NASA's Johnson Space Center (JSC) and Rice University have made substantial progress in myoelectric teleoperation. A myoelectric teleoperation system translates signals generated by an able-bodied robot operator's muscles during hand motions into commands that drive a robot's hand through identical motions. Farry's early work in myoelectric teleoperation used variations over time in the myoelectric spectrum as inputs to neural networks to discriminate grasp types and thumb motions. The resulting schemes yielded up to 93% correct classification on thumb motions. More recently, Fernandez achieved 100% correct non-realtime classification of thumb abduction, extension, and flexion on the same myoelectric data. Fernandez used genetic programming to develop functions that discriminate between thumb motions using myoelectric signal parameters. Genetic programming (GP) is an evolutionary programming method where the computer can modify the discriminating functions' form to improve its performance, not just adjust numerical coefficients or weights. Although the function development may require much computational time and many training cases, the resulting discrimination functions can run in realtime on modest computers. These results suggest that myoelectric signals might be a feasible teleoperation medium, allowing an operator to use his or her own hand and arm as a master to intuitively control an anthropomorphic robot in a remote location such as outer space.

    Description: The National Aeronautics and Space Administration (NASA) has implemented the Advanced Air Transportation Technology (AATT) program to investigate future improvements to the national and international air traffic management systems. This research, as part of the AATT program, developed preliminary design requirements for an advanced Airline Operations Control (AOC) dispatcher's workstation, with emphasis on flight planning. This design will support the implementation of an experimental workstation in NASA laboratories that would emulate AOC dispatch operations. The work developed an airline flight plan data base and specified requirements for: a computer tool for generation and evaluation of free flight, user preferred trajectories (UPT); the kernel of an advanced flight planning system to be incorporated into the UPT-generation tool; and an AOC workstation to house the UPT-generation tool and to provide a real-time testing environment. A prototype for the advanced flight plan optimization kernel was developed and demonstrated. The flight planner uses dynamic programming to search a four-dimensional wind and temperature grid to identify the optimal route, altitude and speed for successive segments of a flight. An iterative process is employed in which a series of trajectories are successively refined until the LTPT is identified. The flight planner is designed to function in the current operational environment as well as in free flight. The free flight environment would enable greater flexibility in UPT selection based on alleviation of current procedural constraints. The prototype also takes advantage of advanced computer processing capabilities to implement more powerful optimization routines than would be possible with older computer systems.

The personal computer revolution has changed the way we deal with many familiar tasks. For example, word processing on personal computers has supplanted the typewriter for many applications.     Description: The applicability of plant-based life support systems has traditionally suffered from the limitations imposed by the high energy demand of controlled environment growth chambers. Theme types of systems are typically less than 2% efficient at converting electrical energy into biomass. The remaining 98% of supplied energy is converted to thermal energy. Traditionally this thermal energy is discharged to the ambient environment as waste heat. This paper describes an energy efficient plant-based life support system which has been designed for use at the Amundsen-Scott South Pole Station. At the South Pole energy is not lost to the environment. What is lost is the ability to extract useful work from it. The CELSS Antarctic Analog Program (CAAP) has developed a system which is designed to extract useful work from the waste thermal energy generated from plant growth lighting systems. In the CAAP system this energy is used to purify Station Sewage.

    Description: The Man-Machine Interaction Design and Analysis System (MIDAS) under joint U.S. Army and NASA cooperative is intended to assist designers of complex human/automation systems in successfully incorporating human performance capabilities and limitations into decision and action support systems. MIDAS is a computational representation of multiple human operators, selected perceptual, cognitive, and physical functions of those operators, and the physical/functional representation of the equipment with which they operate. MIDAS has been used as an integrated predictive framework for the investigation of human/machine systems, particularly in situations with high demands on the operators. We have extended the human performance models to include representation of both human operators and intelligent aiding systems in flight management, and air traffic service. The focus of this development is to predict human performance in response to aiding system developed to identify aircraft conflict and to assist in the shared authority for resolution. The demands of this application requires representation of many intelligent agents sharing world-models, coordinating action/intention, and cooperative scheduling of goals and action in an somewhat unpredictable world of operations. In recent applications to airborne systems development, MIDAS has demonstrated an ability to predict flight crew decision-making and procedural behavior when interacting with automated flight management systems and Air Traffic Control. In this paper, we describe two enhancements to MIDAS. The first involves the addition of working memory in the form of an articulatory buffer for verbal communication protocols and a visuo-spatial buffer for communications via digital datalink. The second enhancement is a representation of multiple operators working as a team. This enhanced model was used to predict the performance of human flight crews and their level of compliance with commercial aviation communication procedures. We show how the data produced by MIDAS compares with flight crew performance data from full mission simulations. Finally, we discuss the use of these features to study communication issues connected with aircraft-based separation assurance.

In order to support that cognitive function definition, we have extended the Man Machine Integrated Design and Analysis System (MIDAS) to include representation of multiple cognitive agents (both human operators and intelligent aiding systems) operating aircraft, airline operations centers and air traffic control centers in the evolving airspace. The demands of this application require representation of many intelligent agents sharing world-models, and coordinating action/intention with cooperative scheduling of goals and actions in a potentially unpredictable world of operations. The MIDAS operator models have undergone significant development in order to understand the requirements for operator aiding and the impact of that aiding in the complex nondeterminate system of national airspace operations. The operator model's structure has been modified to include attention functions, action priority, and situation assessment. The cognitive function model has been expanded to include working memory operations including retrieval from long-term store, interference, visual-motor and verbal articulatory loop functions, and time-based losses. The operator's activity structures have been developed to include prioritization and interruption of multiple parallel activities among multiple operators, to provide for anticipation (knowledge of the intention and action of remote operators), and to respond to failures of the system and other operators in the system in situation-specific paradigms. The model's internal representation has been be modified so that multiple, autonomous sets of equipment will function in a scenario as the single equipment sets do now. In order to support the analysis requirements with multiple items of equipment, it is necessary for equipment to access the state of other equipment objects at initialization time (a radar object may need to access the position and speed of aircraft in its area, for example), and as a function of perception and sensor system interaction. The model has been improved to include multiple world-states as a function of equipment am operator interaction. The model has been used -1o predict the impact of warning and alert zones in aircraft operation, and, more critic-ally, the interaction of flight-deck based warning mechanisms and air traffic controller action in response to ground-based conflict prediction and alerting systems. In this operation, two operating systems provide alerting to two autonomous, but linked sets of operators, whose view of the system and whose dynamics in response are radically different. System stability and operator action was predicted using the MIDAS model.

    Description: This report describes the verification and utilization of the AromaScan(TM) (Hollis, NH) instrument for the ground-based evaluation of odor containment by various spaceflight habitats developed at NASA's Ames Research Center (ARC). The AromaScan(TM) instrument is an electronic odor detection system consisting of 32 polymer sensors that respond differentially to 10 different chemical groups present in an air sample. The AromaScan(TM) system also includes neural network software for constructing a database of known odors, against which an unknown odor can be compared. At present, the standard method for characterizing rodent odor containment during the development and testing of spaceflight hardware is the use of a human odor assessment panel. However, this can be a very time consuming and costly process, and the results are inherently subjective. The AromaScan(TM) system should produce more consistent and objective results, as well as a cost savings in the long term. To test and verify the AromaScan(TM) instrument, daily air samples will be collected from the exhaust port of rodent habitats, during experiment development tests, then injected into the instrument and used to create a database of recognizable odors. Human sniff tests will be performed in conjunction with the AromaScan(TM) analysis, and the results will be correlated. We will then teach the neural network to differentiate between an acceptable and an unacceptable odor profile, as defined by the human sniff test, and to be able to accurately identify an odor that would not pass a sniff panel. The results of our efforts will be to verify that the AromaScan(TM) system is a valuable alternative to human sniff panel assessments for the early iterative process of designing and testing rodent waste filters for spaceflight. Acceptance by a human panel will remain one of the final criteria for successful rodent habitat development.

    Description: The Controlled Ecological Life Support System (CELSS) Antarctic Analog Project (CAAP), is a joint endeavor between the National Science Foundation, Office of Polar Programs (NSF-OPP) and the NASA. The fundamental objective is to develop, deploy, and operate a testbed of advanced life support technologies at the Amundsen-Scott South Pole Station that enable the objectives of both the NSF and NASA. The functions of food production, water purification, and waste treatment, recycle and reduction provided by CAAP will improve the quality of life for the South Pole inhabitants, reduce logistics dependence, enhance safety and minimize environmental impacts associated with human presence on the polar plateau. Because of the analogous technical, scientific, and mission features with Planetary missions such as a mission to Mars, CAAP provides NASA with a method for validating technologies and overall approaches to supporting humans. Prototype systems for sewage treatment, water recycle and crop production are being evaluated at Ames Research Center. The product water from sewage treatment using a Wiped-Film Rotating Disk is suitable for input to the crop production system. The crop production system has provided an enhanced level of performance compared with projected performance for plant-based life support: an approximate 50% increase in productivity per unit area, more than a 65% decrease in power for plant lighting, and more than a 75% decrease in the total power requirement to produce an equivalent mass of edible biomass.

    Description: Personal thermoregulatory systems which provide chest cooling are used in the industrial and aerospace environments to alleviate thermal stress. However, little information is available regarding the physiologic and circulatory changes produced by routine operation of these systems. The objectives of this study were to compare the effectiveness of two passive and two active cooling vests, and to measure the body temperature and circulatory changes produced by each cooling vest configuration. The MicroClimate Systems and the Life Enhancement Tech(LET) lightweight liquid cooling vests, the Steele Vest and LET's Zipper Front Garment were used to cool the chest region of 11 male and 10 female subjects (25 to 55 yr.) in this study. Calf, forearm and finger blood flows were measured using a tetrapolar impedance rheograph. The subjects, seated in an upright position at normal room temperature (approx.21 C), were tested for 60 min. with the cooling system operated at its maximum cooling capacity. Blood flows were recorded continuously using a computer data acquisition system with a sampling frequency of 250 Hz. Oral, right and left ear temperatures and cooling system parameters were logged manually every 5 min. Arm, leg, chest and rectal temperatures; heart rate; respiration; and an activity index were recorded continuously on a URI Inc. Biolog ambulatory monitor. In general, the male and female subjects' rectal and ear temperature responses to cooling were similar for all vest configurations tested. Oral temperatures during the recovery period were significantly (P〈0.05) lower than during the control period, approx.0.2 - 0.5 C, for both men and women wearing any of the four different garments. The corresponding car temperatures were significantly (P〈0.05) decreased approx.0.2 - 0.3 C by the end of the recovery period. Compared to the control period, no significant differences were found in rectal temperatures during cooling and recovery periods. These results show that all vest configurations elicit a similar thermal response in both male and female subject groups. However, subject population variance was rather large and may have masked differences between the vests. One vest may prove more effective than another for a given individual, and experience is the only means of determining this.

    Description: The environmental control and life support system on a spacecraft must maintain a safe and comfortable environment in which the crew can live and work. The system's functions include supplying the crew with oxygen and water, as well as removing carbon dioxide, water vapor, and trace contaminants from cabin air. Although open-loop systems have been used in the past, logistics and safety factors of current and future missions in space make near-complete recycling of the cabin's air and water desirable. The recycling process may include separation and reduction of carbon dioxide, removal of trace gas-phase contaminants, recovery and purification of humidity condensate, purification and polishing of wastewater streams, and other processes. Several of these operations can be performed totally or in part by adsorption processes. Adsorption processes are frequently good candidates for separation and purification in space by virtue of such characteristics as gravity independence, high reliability, relatively high energy efficiency, design flexibility, technological maturity, and regenerability. For these reasons, adsorption has historically played a key role in life support on U.S. and Russian piloted spacecraft. This article focuses on three current spacecraft life support applications that often use adsorption technology: carbon dioxide separation from cabin air, gas-phase trace contaminant control, and potable water recovery from waste streams. In each application, adsorption technology has been selected for use on the International Space Station. The requirements, science, and hardware for each application are discussed. Eventually, human space exploration may lead to construction of planetary habitats. These habitats may have additional applications, such as control of greenhouse gas composition and purification of hydroponic solutions, and may have different requirements and resources available to them, such as gases present in the planetary atmosphere. Adsorption separation and purification processes may continue to fulfill environmental control and life support needs well into the future.

    Description: The future of manned space exploration will be determined through a process which balances the innate need of humanity to explore its surroundings and the costs associated with accomplishing these goals. For NASA, this balance is derived from economics and budgetary constraints that hold it accountable for the expenditure of public funds. These budgetary realities demand a reduction in cost and expenditures of exploration and research activities. For missions venturing out to the edge of habitability, the development of cost effective life support approaches will have a significant influence on mission viability. Over the past several years, a variety of mission scenarios for potential Lunar and Mars missions have been developed. The most promising of these scenarios attempt to provide basic mission requirements at a minimum cost. As a result, these scenarios are extremely power limited. For Closed Environment Life Support System (CELSS) applications, these realities impose both limitations and direction to future research. This paper presents a summary of these mission scenarios and an evaluation of the impact which these power limitations will have on CELSS system design.

    Description: The goal of this research was to develop a dynamic model which can predict the effect of humidity swings on activated carbon adsorption beds used to remove trace contaminants from the atmosphere in spacecraft. Specifically, the model was to be incorporated into a computer simulation to predict contaminant concentrations exiting the bed as a function of time after a humidity swing occurs. Predicted breakthrough curves were to be compared to experimentally measured results. In all respects the research was successful. The two major aspects of this research were the mathematical model and the experiments. Experiments were conducted by Mr. Appel using a fixed-bed apparatus at NASA-Ames Research Center during the summers of 1994 and 1995 and during the first 8 months of 1996. Mr. Appel conducted most of his mathematical modeling work at the University of Virginia. The simulation code was used to predict breakthrough curves using adsorption equilibrium correlations developed previously by M. D. LeVan's research group at the University of Virginia. These predictions were compared with the experimental measurements, and this led to improvements in both the simulation code and the apparatus.

    Description: The Center of Excellence (COE) was created in 1984 to facilitate active collaboration between the scientists at Ames Research Center and the Stanford Psychology Department. As this document will review, over that period of time, the COE served its function well. Funds from the Center supported a large number of projects over the last ten years. Many of the people who were supported by the Center have gone on to distinguished research careers in government, industry and university. In fact, several of the people currently working at NASA Ames were initially funded by the Center mechanism, which served as a useful vehicle for attracting top quality candidates and supporting their research efforts. We are grateful for NASA's support over the years. As we reviewed in the reports for each year, the COE budget generally provided a portion of the true costs of the individual research projects. Hence, the funds from the COE were leveraged with funds from industry and other government agencies. In this way, we feel that all parties benefitted greatly from the collaborative spirit and interactive aspects of the COE. The portion of the support from NASA was particularly important in helping members of the COE to set aside the time to publish papers and communicate advances in our understanding of human performance in NASA-related missions.

    Description: This research project examined the effects of consonance between cockpit displays and alerting system as a technique to encourage pilots to conform to alerting system commands. An experiment used the task of collision avoidance during closely spaced parallel approaches as a case study, building upon previous experiments which identified instances of non-conformance and conflicts between the alerting criteria preferred by pilots, compared to that used by alerting systems. Using a workstation based, part-task simulator, each of 45 subjects completed 45 experiment runs. In each run, the subjects were told they were flying an approach. Their primary task was to keep their wings level despite turbulence through the use of a sidestick. The sidestick commands did not affect the path of the aircraft, however, so that consistent approach paths were be followed. Their secondary task was to indicate when an aircraft on a parallel approach is blundering towards them, as evidenced by the traffic display. Subjects were asked to press different buttons indicating whether they feel an avoidance maneuver is required by the traffic situation or not. At the completion of each run, subjects were asked to rate their confidence in their decision and, if appropriate, to rate the timeliness of automatic alerts when had been given. Three different automatic alert conditions were tested. The "No Automatic Alerts Given" condition is self-explanatory. In the "Automatic Alerts Based on NTZ Criteria" condition, an automatic alert was given when the NTZ criteria was triggered; this criteria is consistent with subject reactions in other studies, in which subjects were found to react, on average, when the other aircraft was 1350 min to the side of the own aircraft. In the "Automatic Alerts Based on MIT Criteria" condition, an automatic alert was given when the MIT criteria was triggered; this criteria was developed by Carpenter and Kuchar for parallel approaches to have better performance, at the cost of increased complexity and higher sensitivity to pilot non-conformance. Three displays were tested. The "Baseline Display" is based on a moving map display, with a top-down view, track-up orientation, iconic presentation of the other aircraft's positions and a text presentation of the other aircraft's altitude. The "NTZ Alert Criteria Shown" display added to the Baseline Display an explicit presentation on the display of the boundary delineating the NTZ criteria, allowing for quick comparison of the other aircraft's position relative to this alert criteria. Likewise, the "MIT Alert Criteria Shown" display added to the Baseline Display an explicit presentation on the display of the boundary delineating the MIT criteria, allowing for quick comparison of the other aircraft's position relative to this alert criteria The test matrix for this experiment was three dimensional, varying displays, alerts and traffic conflict scenarios.

    Description: Exercise will almost certainly play an integral part in minimizing the bone mineral loss and muscular atrophy that occur during spaceflight. It has been hypothesized that an effective exercise regimen can be developed to elicit loads on the lower extremities and require muscle actions which resemble those encountered on Earth. The Penn State Zero-Gravity Simulator (PSZS) is a device which suspends subjects horizontally from multiple latex cords, with each cord negating the weight of a limb segment. A treadmill mounted on the wall under the PSZS enables subjects to run in simulated 0G. Subjects wear a harness to which a number of springs, which provide a gravity replacement load, are connected. The opposite end of each spring is connected to the side of the treadmill. During exercise, astronauts currently wear a similar harness in which the spring tethering load pulls at both the waist and shoulders. Ground reaction forces, muscular activations, and joint angles of the left leg during overground, treadmill, and fully-loaded zero-gravity simulated (ZLS) locomotion were assessed in order to gain insight into the effectiveness of the exercise regimen used by NASA to prevent the muscular atrophy and bone demineralization which occur in weightlessness. There were three hypotheses to this research. It was hypothesized that there will be no differences in peak ground reaction forces and peak loading rates between overground gait and gait in the full body weight loaded conditions in the ZLS. A second hypothesis was that that there will be no differences in hip, knee, and ankle joint positions between walking or running overground, on a standard treadmill, and in full bodyweight loaded conditions in the ZLS. The third hypothesis was that the muscular activations, as a percentage of maximal voluntary contraction, will be similar between walking or running overground, on a standard treadmill, and in full body-weight loaded conditions in the ZLS.

    Description: Human Factors Engineering (HFE) is a discipline whose goal is to engineer a safer, more efficient interface between humans and machines. HFE makes use of a wide range of tools and techniques to fulfill this goal. One of these tools is known as motion and time study, a technique used to develop time standards for given tasks. During the summer of 1995, a human factors motion and time study was initiated with the goals of developing a database of EVA task times and developing a method of utilizing the database to predict how long an EVA should take. Initial development relied on the EVA activities performed during the STS-61 (Hubble) mission. The first step of the study was to become familiar with EVA's, the previous task-time studies, and documents produced on EVA's. After reviewing these documents, an initial set of task primitives and task-time modifiers was developed. Data was collected from videotaped footage of two entire STS-61 EVA missions and portions of several others, each with two EVA astronauts. Feedback from the analysis of the data was used to further refine the primitives and modifiers used. The project was continued during the summer of 1996, during which data on human errors was also collected and analyzed. Additional data from the STS-71 mission was also collected. Analysis of variance techniques for categorical data was used to determine which factors may affect the primitive times and how much of an effect they have. Probability distributions for the various task were also generated. Further analysis of the modifiers and interactions is planned.

    Description: Research and operational experience have shown that one of the major problems with pilot-automation interaction is a lack of mode awareness (i.e., the current and future status and behavior of the automation). As a result, pilots sometimes experience so-called automation surprises when the automation takes an unexpected action or fails to behave as anticipated. A lack of mode awareness and automation surprises can he viewed as symptoms of a mismatch between human and machine properties and capabilities. Changes in automation design can therefore he expected to affect the likelihood and nature of problems encountered by pilots. Previous studies have focused exclusively on early generation "glass cockpit" aircraft that were designed based on a similar automation philosophy. To find out whether similar difficulties with maintaining mode awareness are encountered on more advanced aircraft, a corpus of automation surprises was gathered from pilots of the Airbus A-320, an aircraft characterized by high levels of autonomy, authority, and complexity. To understand the underlying reasons for reported breakdowns in human-automation coordination, we also asked pilots about their monitoring strategies and their experiences with and attitude toward the unique design of flight controls on this aircraft.

    Description: Because plant reproduction is a complex developmental process there are many possible sites of perturbation by the unusual environments of orbital spacecraft. Previous long-duration experiments on Soviet platforms shared features of slowed development through the vegetative stage of plant growth and aborted reproductive function. Our goal has been to understand how special features of the spaceflight environment impact physiological function and reproductive development. In a series of short-duration experiments in the Shuttle mid-deck we studied early reproductive development in Arabidopsis thaliana. Pollen and ovule development aborted at an early stage in the first experiment on STS-54 which utilized closed plant growth chambers. Post-flight analysis suggested that the plants may have been carbon dioxide limited. Subsequent experiments utilized carbon dioxide enrichment (on STS-51) and cabin air flow-through with an air exchange system (on STS-68). Both modifications allowed pollen and ovule development to occur normally on orbit, and full reproductive development up to the stage of an immature seed occurred on STS-68. However, analysis of plant roots from these experiments demonstrated a limitation in rootzone aeration in the spaceflight material that was not mitigated by these procedures. In the future, additional resources (crew time, upgraded flight hardware, and special platforms) will invite more elaborate, long-duration experimentation. On the ISS, a variable speed centrifuge and upgraded plant habitats will permit detailed experiments on the role of gravity in shaping the plant micro-environment, and what influence this plays during reproduction.

    Description: Trace contaminant control onboard the International Space Station will be accomplished not only by the Trace Contaminant Control Subassembly but also by other Environmental Control and Life Support System subassemblies. These additional removal routes include absorption by humidity condensate in the Temperature and Humidity Control Condensing Heat Exchanger and adsorption by the Carbon Dioxide Removal Assembly. The Trace Contaminant Injection Test, which was performed at NASA's Marshall Space Flight Center, investigated the system-level removal of trace contaminants by the International Space Station Atmosphere Revitalization, and Temperature/Humidity Control Subsystems, (November-December 1997). It is a follow-on to the Integrated Atmosphere Revitalization Test conducted in 1996. An estimate for the magnitude of the assisting role provided by the Carbon Dioxide Removal Assembly and the Temperature and Humidity Control unit was obtained. In addition, data on the purity of Carbon Dioxide Removal Assembly carbon dioxide product were obtained to support Environmental Control and Life Support System Air Revitalization Subsystem loop closure.

    Description: A filter assembly which is incorporated into the Russian Trace Contaminant Control Assembly was tested for removal of airborne trace chemical contaminants in a closed loop 9 m(exp 3) system. Given contaminant loading rates and maximum allowable atmospheric concentrations, the Russian system was able to maintain system air concentrations below maximum allowable limits. This was achieved for both a new filter system and for a system where a part of it was pre-loaded to emulate 3 years of system age.

    Description: The Launch and Entry Suit (LES) and Advanced Crew Escape Suit (ACES) are worn by astronauts for launch and entry. Previous work by Waligora, et al., 1992, Waligora and Gilbert, 1992, and Dalrymple 1996, have found that carbon dioxide (CO2) accumulation in the LES/ACES helmet may be problematic. CO2 accumulation is important because high inspired levels of CO2 reduce physical function and pose a safety hazard (e.g. levels of CO2 accumulation of 3.6% in the Extravehicular Mobility Unit are sufficient to terminate Extra Vehicular Activities). My task was to design a suitable test protocol for determining the important physiological aspects of LES/ACES use. Three basic issues arose. First was the determination of the astronaut's CO2 inspiration during visor-down use at rest and during walking at 3.5 mph. A sub-issue was the impact of a pneumotach on CO2 since it has been previously observed that when the Aerosport pneumotach was used, performance seemed improved, which might be attributable to a lowered respiration rate when using the pneumotach. The second issue was the energy costs of waLking in the LES/ACES with various G-suit inflation levels, since G-suit inflation increases metabolic costs and metabolic costs influence the C02 production in the LES/ACES helmet. Since G-suit inflation improves orthostatic tolerance after space flight, but likely increases the energy costs of walking, the balance between G-suit inflation and C02 accumulation is an important safety consideration. The third issue which arose from pilot work was the substantial reduction in physical function after a 10 min visor-down period prior to walk.

    Description: The difficulty of accomplishing work in extravehicular activity (EVA) is well documented. It arises as a result of motion constraints imposed by a pressurized spacesuit in a near-vacuum and of the frictionless environment induced in microgravity. The appropriate placement of foot restraints is crucial to ensuring that astronauts can remove and drive bolts, mate and demate connectors, and actuate levers. The location on structural members of the foot restraint sockets, to which the portable foot restraint is attached, must provide for an orientation of the restraint that affords the astronaut adequate visual and reach envelopes. Previously, the initial location of these sockets was dependent upon the experienced designer's ability to estimate placement. The design was tested in a simulated zero-gravity environment; spacesuited astronauts performed the tasks with mockups while submerged in water. Crew evaluation of the tasks based on these designs often indicated the bolt or other structure to which force needed to be applied was not within an acceptable work envelope, resulting in redesign. The development of improved methods for location of crew aids prior to testing would result in savings to the design effort for EVA hardware. Such an effort to streamline EVA design is especially relevant to International Space Station construction and maintenance. Assembly operations alone are expected to require in excess of four hundred hours of EVA. Thus, techniques which conserve design resources for assembly missions can have significant impact. We describe an effort to implement a human modelling application in the design effort for an International Space Station Assembly Mission. On Assembly Flight 6A, the Canadian-built Space Station Remote Manipulator System will be delivered to the U.S. Laboratory. It will be released from its launch restraints by astronauts in EVA. The design of the placement of foot restraint sockets was carried out using the human model Jack, and the modelling results were compared with actual underwater test results. The predicted locations of the sockets was found to be acceptable for 94% of the tasks attempted by the astronauts, This effort provides confidence in the capabilities of this package to accurately model tasks. It therefore increases assurance that the tool maybe used early in the design process.

    Description: The Space Station Furnace Facility (SSFF) is a facility located in the International Space Station United States Laboratory (ISS US Lab) for materials research in the microgravity environment. The SSFF will accommodate basic research, commercial applications, and studies of phenomena of metals and alloys, electronic and photonic materials, and glasses and ceramics. To support this broad base of research requirements, the SSFF will operate, regulate, and support a variety of Experiment Modules (EMs). To meet station requirements concerning the microgravity level needed for experiments, station is providing an active vibration isolation system, and SSFF provides the interface. SSFF physically consists of a Core Rack and two instrument racks (IRs) that occupy three adjacent ISS US Lab rack locations within the International Space Station (ISS). All SSFF racks are modified International Standard Payload Racks (ISPR). SSFF racks will have a 50% larger pass through area on the lower sides than ISPRs to accommodate the many rack to rack interconnections. The Instrument Racks are further modified with lowered floors and an additional removable panel (15" x 22") on top of the rack for access if needed. The Core Rack shall contain all centralized Core subsystems and ISS subsystem equipment. The two Instrument Racks shall contain the distributed Core subsystem equipment, ISS subsystem equipment, and the EMs. The Core System, which includes the Core Rack, the IR structures, and subsystem components located in the IRs serves as the central control and management for the IRs and the EMs. The Core System receives the resources provided by the International Space Station (ISS) and modifies, allocates, and distributes these resources to meet the operational requirements of the furnace. The Core System is able to support a total of four EMs and can control, support, and activate/deactivate the operations of two EMs, simultaneously. The IRs can be configured to house two small EMs or one tall vertical EM, and serve as the interface between the Core and the respective EM. The Core Rack and an adjacent Instrument Rack (containing one or more furnaces) will be delivered to the ISS in one launch. This is Integrated Configuration One (ICI). The Core Rack and IRI will be passive during transport in the Mini Pressurized Logistics Module (MPLM): Any subsequent EMs to operate within IRI are installed on-orbit. The second IR (containing one or more furnaces) is delivered to ISS on a subsequent launch which will establish Integrated Configuration Two (IC2). Additional integrated configurations will be established with the replacement of EMs or Instrument Racks.

    Description: Cultivar evaluation for controlled environments is a lengthy and multifaceted activity. The chapters of this thesis cover eight steps preparatory to yield trials, and the final step of cultivar selection after data are collected. The steps are as follows: 1. Examination of the literature on the crop and crop cultivars to assess the state of knowledge. 2. Selection of standard cultivars with which to explore crop response to major growth factors and determine set points for screening and, later, production. 3. Determination of practical growing techniques for the crop in controlled environments. 4. Design of experiments for determination of crop responses to the major growth factors, with particular emphasis on photoperiod, daily light integral and air temperature. 5. Developing a way of measuring yield appropriate to the crop type by sampling through the harvest period and calculating a productivity function. 6. Narrowing down the pool of cultivars and breeding lines according to a set of criteria and breeding history. 7. Determination of environmental set points for cultivar evaluation through calculating production cost as a function of set points and size of target facility. 8. Design of screening and yield trial experiments emphasizing efficient use of space. 9. Final evaluation of cultivars after data collection, in terms of production cost and value to the consumer. For each of the steps, relevant issues are addressed. In selecting standards to determine set points for screening, set points that optimize cost of production for the standards may not be applicable to all cultivars. Production of uniform and equivalent- sized seedlings is considered as a means of countering possible differences in seed vigor. Issues of spacing and re-spacing are also discussed.

    Description: A study has been conducted with the full-spectrum, calligraphic, computer-generated display system to determine the effect of chromatic content of the visual display upon pilot performance during the landing approach maneuver. This study utilizes a new digital chromatic display system, which has previously been shown to improve the perceived fidelity of out-the-window display scenes, and presents the results of an experiment designed to determine the effects of display color content by the measurement of both vertical approach performance and pilot-describing functions. This method was selected to more fully explore the effects of visual color cues used by the pilot. Two types of landing approaches were made: dynamic and frozen range, with either a landing approach scene or a perspective array display. The landing approach scene was presented with either red runway lights and blue taxiway lights or with the colors reversed, and the perspective array with red lights, blue lights, or red and blue lights combined. The vertical performance measures obtained in this experiment indicated that the pilots performed best with the blue and red/blue displays. and worst with the red displays. The describing-function system analysis showed more variation with the red displays. The crossover frequencies were lowest with the red displays and highest with the combined red/blue displays, which provided the best overall tracking, performance. Describing-function performance measures, vertical performance measures, and pilot opinion support the hypothesis that specific colors in displays can influence the pilots' control characteristics during the final approach.

    Description: The aims under this training grant, as under the subsequent fellowship, were to elaborate the theory and technique of cultivar evaluation for specialized controlled environments, then to employ the technique on selected crops, ultimately conducting cultivar trials, and making the knowledge gained available for use in NASA's space program. We undertook a comprehensive search of the Cornell agricultural library (Mann library) and its data-bases for any and all material relating to cultivar evaluation of vegetable crops, and also developed the logic of how to go about narrowing down the field of contending cultivars when undertaking cultivar trials. The results of this work, the principal outcome of the grant, are reflected in his MS thesis, particularly in Chapter 2, "Commercial and Scientific Literature," and even more so in Chapter 8, "Selecting cultivars and lines for screening." David also attended annual conferences of vegetable crop plant breeders, annual yield trials and breeding trials for vegetable crops, as well as relevant professional conferences such as the ASHS annual meetings, and the others.

    Description: Through the support of NASA's Mission to Planet Earth, Biosphere 2 Center hosted 11 research interns for 6 to 8 weeks each during the summer of 1997. In addition, we were able to offer scholarships to 14 students for Columbia University summer field courses. These two types of programs engaged students in much of the range of activity of practicing Earth Scientists, with an emphasis on the collection and analysis of data in both the field and the laboratory. Research interns and students in the field courses also played an important part in the design and evolution of their research projects. In addition to laboratory and field research, students participated in weekly research seminars by resident and visiting scientists. Research interns were exposed to the geology and ecology of the region via short field trips to the Arizona Sonora Desert Museum, Mount Lemmon, Aravaipa Canyon and the Gulf of California, while field course students were exposed to laboratory-based research via intern-led hands-on demonstrations of their work. All students made oral and written presentations of their work during the summer, and two of the research interns have applied to present their results at the National Conference on Undergraduate Research in Maryland in April, 1998.

    Description: Post space flight orthostatic intolerance among Space Shuttle crew members following exposure to extended periods of microgravity has been of significant concern to the safety of the shuttle program. Following the Challenger accident, flight crews were required to wear launch and entry suits (LES). It was noted that overall, there appeared to be a higher degree of orthostatic intolerance among the post-Challenger crews (approaching 30%). It was hypothesized that the increased heat load incurred when wearing the LES, contributed to an increased degree of orthostatic intolerance, possibly mediated through increased peripheral vasodilatation triggered by the heat load. The use of liquid cooling garments (LCG) beneath the launch and entry suits was gradually implemented among flight crews in an attempt to decrease heat load, increase crew comfort, and hopefully improve orthostatic tolerance during reentry and landing. The hypothesis that the use of the LCG during reentry and landing would decrease the degree of orthostasis has not been previously tested. Operational stand-tests were performed pre and post flight to assess crewmember's cardiovascular system's ability to respond to gravitational stress. Stand test and debrief information were collected and databased for 27 space shuttle missions. 63 crewpersons wearing the LCG, and 70 crewpersons not wearing the LCG were entered into the database for analysis. Of 17 crewmembers who exhibited pre-syncopal symptoms at the R+O analysis, 15 were not wearing the LCG. This corresponds to a 21% rate of postflight orthostatic intolerance among those without the LCG, and a 3% rate for those wearing LCG. There were differences in these individual's average post-flight maximal systolic blood pressure, and lower minimal Systolic Blood pressures in those without LCG. Though other factors, such as type of fluid loading, and exercise have improved concurrently with LCG introduction, from this data analysis, it appears that LCG usage provided a significant degree of protection from post-flight orthostatic intolerance.

    Description: Air purification systems are necessary to provide clean air in the closed environments aboard spacecraft. Trace contaminants are removed using adsorption. One major factor concerning the removal of trace contaminants is relative humidity. Water can reduce adsorption capacity and, due to constant fluctuations, its presence is difficult to incorporate into adsorption column designs. The purpose of the research was to allow for better design techniques in trace contaminant adsorption systems, especially for feeds with water present. Experiments and mathematical modeling research on effects of humidity swings on adsorption columns for air revitalization were carried out.

    Description: Locomotion on a treadmill in 0 G will probably remain a centerpiece of NASA's exercise countermeasures programme. This form of physical activity has the potential to cause large bone and muscle forces as well as loading during a period of continuous treadmill exercise. A critical concern is the provision of a treadmill which can approximate 1 G performance in space. At this point, no adequate objective measurements of in-flight treadmill kinetics or of the human response to this activity have been made. Interpretation of the results obtained in the present study is limited by the following: (1) bungee tensions were not measured; (2) ground reaction forces were not measured in parallel with the kinematic measurements; and (3) the instrumentation used to film the astronauts could itself have been affected by microgravity. Despite these shortcomings, what is apparent is that exercise during NASA missions STS 7 and STS 8 resulted in leg motions that were similar to those found during 1 G locomotion on an inclined passive treadmill and on an active treadmill at an even steeper grade. In addition, it was apparent that the majority of the loads were transmitted through the forefoot, and one can surmise that this style of running would result in physiologically significant tensions in the calf musculature and resultant ankle compressive loading. Further speculation regarding limb loading is complicated by the fact that varying amounts of force are transmitted through (1) the treadmill handle and (2) bungee cords that act as a tether. New generations of treadmills are being manufactured that could provide I important information for planners of long-duration space missions. If these types of treadmill are flown on future missions, it will be possible to control bungee tensions more precisely, control for grade and speed, and, most importantly, provide data on the rates and magnitudes of limb loading. These data could then be incorporated into biomechanical models of the lower limb to more fully understand mechanisms of load transmission from distal to proximal structures and to optimize in-flight exercise protocols in such a way that muscle and bone loss could be reduced.

    Description: Life Systems completed a Ground-based Space Station Experiment Development Study Program which verifies through testing the performance and applicability of the electrochemical Air-Polarized Carbon Dioxide Concentrator (APC) process technology for space missions requiring low (i.e., less than 3 mm Hg) CO2 partial pressure (pCO2) in the cabin atmosphere. Required test hardware was developed and testing was accomplished at an approximate one-person capacity CO2 removal level. Initially, two five-cell electrochemical modules using flight-like 0.5 sq ft cell hardware were tested individually, following by their testing at the integrated APC system level. Testing verified previously projected performance and established a database for sizing of APC systems. A four person capacity APC system was sized and compared with four candidate CO2 removal systems. At its weight of 252 lb, a volume of 7 cu ft and a power consumption of 566 W while operating at 2.2 mm Hg pCO2, the APC was surpassed only by an Electrochemical Depolarized CO2 Concentrator (EDC) (operating with H2), when compared on a total equivalent basis.

    Description: The main goal of this program was to develop an unobtrusive power-assisted EVA glove metacarpalphalangeal (MCP) joint that could provide the crew member with as close to nude body performance as possible, and to demonstrate the technology feasibility of power assisted space suit components in general. The MCP joint was selected due to its being representative of other space suit joints, such as the shoulder, hip and carpometacarpal joint, that would also greatly benefit from this technology. In order to meet this objective, a development team of highly skilled and experienced personnel was assembled. The team consisted of two main entities. The first was comprised of ILC's experienced EVA space suit glove designers, who had the responsibility of designing and fabricating a low torque MCP joint which would be compatible with power assisted technology. The second part of the team consisted of space robotics experts from the University of Maryland's Space Systems Laboratory. This team took on the responsibility of designing and building the robotics aspects of the power-assist system. Both parties addressed final system integration responsibilities.

    Description: This grant was entitled 'Planetary Habitability' and the work performed under it related to elucidating the conditions that lead to habitable, i.e. Earth-like, planets. Below are listed publications for the past two and a half years that came out of this work. The main thrusts of the research involved: (1) showing under what conditions atmospheric O2 and O3 can be considered as evidence for life on a planet's surface; (2) determining whether CH4 may have played a role in warming early Mars; (3) studying the effect of varying UV levels on Earth-like planets around different types of stars to see whether this would pose a threat to habitability; and (4) studying the effect of chaotic obliquity variations on planetary climates and determining whether planets that experienced such variations might still be habitable. Several of these topics involve ongoing research that has been carried out under a new grant number, but which continues to be funded by NASA's Exobiology program.

    Description: This report describes the theoretical and operational foundations for our analysis of skill in extravehicular mass handling. A review of our research on postural control, human-environment interactions, and exploratory behavior in skill acquisition is used to motivate our analysis. This scientific material is presented within the context of operationally valid issues concerning extravehicular mass handling. We describe the development of meaningful empirical measures that are relevant to a special class of nested control systems: manual interactions between an individual and the substantial environment. These measures are incorporated into a unique empirical protocol implemented on NASA's principal mass handling simulator, the precision air-bearing floor, in order to evaluate skill in extravehicular mass handling. We discuss the components of such skill with reference to the relationship between postural configuration and controllability of an orbital replacement unit, the relationship between orbital replacement unit control and postural stability, the relationship between antecedent and consequent movements of an orbital replacement unit, and the relationship between antecedent and consequent postural movements. Finally, we describe our expectations regarding the operational relevance of the empirical results as it pertains to extravehicular activity tools, training, monitoring, and planning.

    Notes: Semipurified lipase of Candida rugosa (CRSL) was subjected to chemical modification, and the activities of the modified lipase, in hydrolysis and esterification reactions, were examined. The esterification reactions were carried out in the absence and presence of isooctane. When the enzyme was modified with polyethylene glycol (PEG), two methodologies were studied. The activation of PEG with p-NO2-phenylchloroformate gives better biocatalysts than those obtained with cyanuric chloride-PEG. The chemical modification with PEG increases the stability of pure lipases in isooctane at 50°C (extreme conditions). The chemically modified enzymes are useful for biotransformations in organic solvents. In addition the nitration of tyrosines with tetranitromethane was also studied. © 1997 John Wiley & Sons, Inc.     Notes: To investigate the response of hybridoma cells to hypoosmotic stress, S3H5/γ2bA2 and DB9G8 hybridomas were cultivated in the hypoosmolar medium [Dulbecco's modified Eagle's medium (DMEM) supplemented with 10% serum] resulting from sodium chloride subtraction. Both hybridomas showed similar responses to hypoosmotic stress in regard to cell growth and antibody production. The cell growth and antibody production at 276 mOsm/kg were comparable to those at 329 mOsm/kg (standard DMEM). Both cells grew well at 219 mOsm/kg, though their growth and antibody production were slightly decreased. When the osmolality was further decreased to 168 mOsm/kg, the cell growth did not occur. When subjected to hyperosmotic stress, both cells displayed significantly enhanced specific antibody productivity (qAb). However, the cells subjected to hypoosmotic stress did not display enhanced qAb. Taken together, both hyperosmotic and hypoosmotic stresses depressed the growth of S3H5/γ2bA2 and DB9G8 hybridomas.     Notes: This study demonstrates a new approach to reduce the amount of fungal cellulase required for the conversion of cellulose into ethanol. Escherichia coli KO11, a biocatalyst developed for the fermentation of hemicellulose syrups, was used to produce recombinant endoglucanase as a co-product with ethanol. Seven different bacterial genes were expressed from plasmids in KO11. All produced cell-associated endoglucanase activity. KO11(pLOI1620) containing Erwinia chrysanthemi celZ (EGZ) produced the highest activity, 3,200 IU endoglucanase/L fermentation broth (assayed at pH 5.2 and 35°C). Recombinant EGZ was solubilized from harvested cells by treatment with dilute sodium dodecyl sulfate (12.5 mg/ml, 10 min, 50°C) and tested in fermentation experiments with commercial fungal cellulase (5 filter paper units/g cellulose) and purified cellulose (100 g/L). Using Klebsiella oxytoca P2 as the biocatalyst, fermentations supplemented with EGZ as a detergent-lysate of KO11(pLOI1620) produced 14%-24% more ethanol than control fermentations supplemented with a detergent-lysate of KO11(pUC18). These results demonstrate that recombinant bacterial endoglucanase can function with fungal cellulase to increase ethanol yield during the simultaneous saccharification and fermentation of cellulose. © 1997 Wiley & Sons, Inc.     Notes: An expression system has been developed for the introduction of DNA cassettes into the region between the transcription and translation start sites of a gene of interest. This cassette system was used to engineer mRNA stability through the introduction of hairpins at the 5′ end. A synthetic DNA cassette was designed so that the resulting mRNA hairpin would be positioned one nucleotide from the 5′ mRNA end. The hairpin-containing mRNA exhibited a half-life 3 times that of the mRNA with no hairpin, resulting in increases in both mRNA and protein levels.     Notes: Preferential transport in adsorptive membranes can be used to selectively remove biochemicals directly from fermentation broths. During preferential transport, an adsorbing solute is selectively transported across the membrane while nonadsorbing solutes and cells are retained by the membrane. This technique was used to separate lysozyme directly from a feed containing lysozyme, myoglobin, and yeast cells. We found that because the oscillatory flows used in preferential transport involve strokes that are close to symmetric, they are very efficient in alleviating cake formation due to cell deposition on the membrane surface. Theoretical results suggest that, by optimizing process variables, preferential transport can lead to a continuous concentrated stream of the adsorbing protein. © 1997 John Wiley & Sons, Inc.     Notes: A mathematical model of glycolysis in Saccharomyces cerevisiae is presented. The model is based on rate equations for the individual reactions and aims to predict changes in the levels of intra- and extracellular metabolites after a glucose pulse, as described in part I of this study. Kinetic analysis focuses on a time scale of seconds, thereby neglecting biosynthesis of new enzymes. The model structure and experimental observations are related to the aerobic growth of the yeast. The model is based on material balance equations of the key metabolites in the extracellular environment, the cytoplasm and the mitochondria, and includes mechanistically based, experimentally matched rate equations for the individual enzymes. The model includes removal of metabolites from glycolysis and TCC for biosynthesis, and also compartmentation and translocation of adenine nucleotides. The model was verified by in vivo diagnosis of intracellular enzymes, which includes the decomposition of the network of reactions to reduce the number of parameters to be estimated simultaneously. Additionally, sensitivity analysis guarantees that only those parameters are estimated that contribute to systems trajectory with reasonable sensitivity. The model predictions and experimental observations agree reasonably well for most of the metabolites, except for pyruvate and adenine nucleotides. © 1997 John Wiley & Sons, Inc.     Notes: The design of a reactor in which two interacting cell populations (microorganisms and plants) could grow under controlled conditions was considered. In this reactor, the cell populations are separated by a membrane which permits semi-in vivo study of induced interaction-specific changes in metabolism. In this paper, the interaction of suspension culture of Nicotiana tabacum (tobacco) and the Oomycete, Phytophthora nicotiana was simulated. The results of the computer simulation show the induced metabolic changes as a consequence of the biological interaction. The paper introduces a novel approach in the strategy for the study of interacting population in suspension cultures. This type of system has potential applications in studies of the regulation of secondary metabolism and for the production of high values pharmaceuticals. © 1997 John Wiley & Sons, Inc.     Notes: The strength of adhesion and dynamics of detachment of murine 3T3 fibroblasts from self-assembled monolayers were measured in a radial-flow chamber (RFC) by applying models for fluid mechanics, adhesion strength probability distributions, and detachment kinetics. Four models for predicting fluid mechanics in a RFC were compared to evaluate the accuracy of each model and the significance of inlet effects. Analysis of these models indicated an outer region at large radial positions consistent with creeping flow, an intermediate region influenced by inertial dampening, and an inner region dominated by entrance effects from the axially-oriented inlet. In accompanying experiments patterns of the fraction of cells resisting detachment were constructed for individual surfaces as a function of the applied shear stress and evaluated by comparison with integrals of both a normal and a log-normal distribution function. The two functions were equally appropriate, yielding similar estimates of the mean strength of adhesion. Further, varying the Reynolds number in the inlet, Red, between 630 and 1480 (corresponding to volumetric flow rates between 0.9 and 2.1 mL/s) did not affect the mean strength of adhesion. For these same experiments, analysis of the dynamics of detachment revealed three temporal phases: 1) rapid detachment of cells at the onset of flow, consistent with a first-order homogeneous kinetic model; 2) time-dependent rate of detachment during the first 30 sec. of exposure to hydrodynamic shear, consistent with the first-order heterogeneous kinetic model proposed by Dickinson and Cooper (1995); and 3) negligible detachment, indicative of pseudo-steady state after 60 sec. of flow. Our results provide rigorous guidelines for the measurement of adhesive interactions between mammalian cells and prospective biomaterial surfaces using a RFC. © 1997 John Wiley & Sons, Inc.     Notes: Glucose and lactate metabolic rates were evaluated for cultures of cord blood (CB) mononuclear cell (MNC), peripheral blood (PB) MNC, and PB CD34+ cell cultures carried out in spinner flasks and in T-flasks in both serum-containing and serum-free media. Specific glucose uptake rates (qgluc, in micromoles per cell per hour) and lactate generation rates (qlac) correlated with the percentage of colony-forming cells (CFC) present in the culture for a broad range of culture conditions. Specifically, the time of maximum CFC percentage in each culture coincided with the time of maximum qgluc and qlac in cultures with different seeding densities and cytokine combinations. A two-population model (Qlac = α[CFC] + β([TC] - [CFC]), where [TC] is total cell concentration; Qlac is volumetric lactate production rate in micromoles per milliliter per hour; α is qlac for an average CFC; and β is qlac for an average non-CFC) was developed to describe lactate production. The model described lactate production well for cultures carried out in both T-flasks and spinner flasks and inoculated with either PB or CB MNC or PB CD34+ cells. The values for α and β that were derived from the model varied with both the inoculum density and the cytokine combination. However, preliminary results indicate that cultures carried out under the same conditions from different samples with similar initial CD34+ cell content have similar values for β and β. These findings suggest that it should be possible to use lactate production data to predict the harvest time that corresponds to the maximum number of CFC in culture. The ability to harvest ex vivo hematopoietic cultures for transplantation when CFC are at a maximum has the potential to speed the rate at which immunocompromised patients recover. © 1997 John Wiley & Sons, Inc.     Notes: A mathematical model to estimate the solar irradiance profile and average light intensity inside a tubular photobioreactor under outdoor conditions is proposed, requiring only geographic, geometric, and solar position parameters. First, the length of the path into the culture traveled by any direct or disperse ray of light was calculated as the function of three variables: day of year, solar hour, and geographic latitude. Then, the phenomenon of light attenuation by biomass was studied considering Lambert-Beer's law (only considering absorption) and the monodimensional model of Cornet et al. (1900) (considering absorption and scattering phenomena). Due to the existence of differential wavelength absorption, none of the literature models are useful for explaining light attenuation by the biomass. Therefore, an empirical hyperbolic expression is proposed. The equations to calculate light path length were substituted in the proposed hyperbolic expression, reproducing light intensity data obtained in the center of the loop tubes. The proposed model was also likely to estimate the irradiance accurately at any point inside the culture. Calculation of the local intensity was thus extended to the full culture volume in order to obtain the average irradiance, showing how the higher biomass productivities in a Phaeodactylum tricornutum UTEX 640 outdoor chemostat culture could be maintained by delaying light limitation. © 1997 John Wiley & Sons, Inc.     Notes: Both parallel fermentations with Aspergillus awamori (CBS 115.52) and a literature study on several fungi have been carried out to determine a relation between fungal morphology and agitation intensity. The studied parameters include hyphal length, pellet size, surface structure or so-called hairy length of pellets, and dry mass per-wet-pellet volume at different specific energy dissipation rates. The literature data from different strains, different fermenters, and different cultivation conditions can be summarized to say that the main mean hyphal length is proportional to the specific energy dissipation rate according to a power function with an exponent of -0.25 ± 0.08. Fermentations with identical inocula showed that pellet size was also a function of the specific energy dissipation rate and proportional to the specific energy dissipation rate to an exponent of -0.16 ± 0.03. Based on the experimental observations, we propose the following mechanism of pellet damage during submerged cultivation in stirred fermenters. Interaction between mechanical forces and pellets results in the hyphal chip-off from the pellet outer zone instead of the breakup of pellets. By this mechanism, the extension of the hyphae or hair from pellets is restricted so that the size of pellets is related to the specific energy dissipation rate. Hyphae chipped off from pellets contribute free filamentous mycelia and reseed their growth. So the fraction of filamentous mycelial mass in the total biomass is related to the specific energy dissipation rate as well.To describe the surface morphology of pellets, the hyphal length in the outer zone of pellets or the so-called hairy length was measured in this study. A theoretical relation of the hairy length with the specific energy dissipation rate was derived. This relation matched the measured data well. It was found that the porosity of pellets showed an inverse relationship with the specific energy dissipation rate and that the dry biomass per-wet-pellet volume increased with the specific energy dissipation rates. This means that the tensile strength of pellets increased with the increase of specific energy dissipation rate. The assumption of a constant tensile strength, which is often used in literature, is then not valid for the derivation of the relation between pellet size and specific energy dissipation rate. The fraction of free filamentous mycelia in the total biomass appeared to be a function of the specific energy dissipation in stirred bioreactors. © 1997 John Wiley & Sons, Inc.
