    Notes: Abstract Comparison of the effects of temperature on the growth in culture (increase in protein) of Frankia showed that three strains isolated from Casuarina were more tolerant of high temperature (45°C) than a strain from Alnus rubra. Optimal temperatures for growth of the Casuarina strains were in the range 25–30°C. Growth of the Alnus strain was good at 25°C but poor at 37°C. High temperatures (35–40°C) during storage for 7 months of these Frankia strains in sand, inoculated initially with liquid culture or with Frankia incorporated into alginate beads and permitted to dry, resulted in substantial loss of infectivity for the host plant species. Loss in infectivity was greater with an Alnus Frankia strain than strains from Casuarina cunninghamiana, C. equisetifolia and C. junghuniana. Three Frankia strains from C. equisetifolia were incorporated into a sand/perlite mixture with three different moisture regimes (field moisture capacity – wet: watered and maintained at field capacity; watered to field capacity but then allowed to dry – moderately wet; or watered to half field capacity and then permitted to dry – dry) and then stored for 12 weeks at 25°C and 35°C. Assessment by the most probable number (MPN) technique of the infectivity of the sand mixture for nodulation of C. equisetifolia showed significant interactions between Frankia strain, temperature and soil moisture content. The infectivity of Frankia strains ORS020607 and UGL020602q was not affected by incubation in wet sand at 25°C but fell by more than half after 12 weeks in moderate and dry conditions. Changes in infectivity were similar when incubation was at 35°C. By contrast, the infectivity of UGL020603q fell substantially under all moisture conditions and at both temperatures. The data show the importance of screening for tolerance of both temperature and moisture content when selecting strains for preparation of inoculum for use in hot climates.

    Notes: Abstract Emission of N2O from cultivated and fertilised soils may contribute significantly to the total global N2O emission. This study included laboratory and field investigations of the N2O production from a dry stubble field as influenced by addition of water, nitrogen and glucose. N2O fluxes were measured using a closed-chamber technique, and the O2 content in the soil was measured using soil probes. Results from a laboratory soil core technique were correlated to the relative N2O emission observed in the field. When the soil water content in the field increased from 14% to 60% water-filled pore space, the N2O emission increased from non-significant to a constant emission of 30 μg N m–2 h–1. At this soil water content the production of N2O was limited by the availability of nitrogen and carbon. Application of nitrogen at soil temperatures of 13 and 21°C in a pre-wetted soil increased the N2O emission 3.1- and 3.7-fold, respectively, whereas nitrogen plus carbon application increased the N2O emission 13.3- and 7.3-fold, respectively. In both treatments the N2O emission rates were affected by fluctuations in soil temperature and O2 content. The results indicate that even in a soil producing very little N2O under dry conditions, the soil microbial community maintains a potential to produce N2O when favourable conditions occur in terms of availability of water, nitrogen and carbon.

    Notes: Abstract Fluxes of N2O were studied in a Norway spruce forest in the southwest of Sweden. Three differently treated catchments were compared: Limed (6 t dolomite ha–1), Nitrex (additional N-deposition corresponding to 35 kg ha–1 year–1, in small doses) and Control (used as control site). The N-retention was still high (95%) after 2years of N-addition at the Nitrex site when the flux measurements were performed. Each catchment contained both well-drained and poorly drained soils (covered with Sphagnum sp.). The emissions of N2O were in general low with both a high spatial and temporal variation for all three sites. The measured emissions were 25, 71 and 96 (gN2O-N ha–1 year–1) for the well-drained Limed, Control and Nitrex sites, respectively. The average emissions of N2O from the wet areas were significantly higher than the well-drained areas within the catchments. For the wet areas the measured emissions were larger: 90, 118 and 254 (g N2O-N ha–1 year–1) for the Limed, Control and Nitrex sites, respectively. Comparison between treatments showed the wet Nitrex site to have a significantly higher emission than all other sites. The increased N-deposition at the Nitrex site increased the N2O emissions by 0.2% of the added N for the well-drained soils and about 1% for the wet areas, compared with the control site. Since the wet areas represented only a small part of the forest, their larger emissions did not contribute significantly to the overall emission of the forest. Neither temperature nor water content of the soil was well correlated with the N2O emissions. Soil gas samples showed that most of the N2O was produced below a 0.3-m depth in the soil.

    Description: Saturn's giant moon Titan has a thick (1.5 bar) nitrogen atmosphere, which has a temperature structure that is controlled by the absorption of solar and thermal radiation by methane, hydrogen, and organic aerosols into which methane is irreversibly converted by photolysis. Previous studies of Titan's climate evolution have been done with the assumption that the methane abundance was maintained against photolytic depletion throughout Titan's history, either by continuous supply from the interior or by buffering by a surface or near surface reservoir.     Description: Angiogenesis is thought to depend on a precise balance of positive and negative regulation. Angiopoietin-1 (Ang1) is an angiogenic factor that signals through the endothelial cell-specific Tie2 receptor tyrosine kinase. Like vascular endothelial growth factor, Ang1 is essential for normal vascular development in the mouse. An Ang1 relative, termed angiopoietin-2 (Ang2), was identified by homology screening and shown to be a naturally occurring antagonist for Ang1 and Tie2. Transgenic overexpression of Ang2 disrupts blood vessel formation in the mouse embryo. In adult mice and humans, Ang2 is expressed only at sites of vascular remodeling.     Description: The discovery of anticancer drugs is now driven by the numerous molecular alterations identified in tumor cells over the past decade. To exploit these alterations, it is necessary to understand how they define a molecular context that allows increased sensitivity to particular compounds. Traditional genetic approaches together with the new wealth of genomic information for both human and model organisms open up strategies by which drugs can be profiled for their ability to selectively kill cells in a molecular context that matches those found in tumors. Similarly, it may be possible to identify and validate new targets for drugs that would selectively kill tumor cells with a particular molecular context.     Description: Exposure of the yeast Saccharomyces cerevisiae to high extracellular osmolarity induces the Sln1p-Ypd1p-Ssk1p two-component osmosensor to activate a mitogen-activated protein (MAP) kinase cascade composed of the Ssk2p and Ssk22p MAP kinase kinase kinases (MAPKKKs), the Pbs2p MAPKK, and the Hog1p MAPK. A second osmosensor, Sho1p, also activated Pbs2p and Hog1p, but did so through the Ste11p MAPKKK. Although Ste11p also participates in the mating pheromone-responsive MAPK cascade, there was no detectable cross talk between these two pathways. The MAPKK Pbs2p bound to the Sho1p osmosensor, the MAPKKK Ste11p, and the MAPK Hog1p.     Description: The neurofibromatosis type 1 (NF1) tumor suppressor protein is thought to restrict cell proliferation by functioning as a Ras-specific guanosine triphosphatase-activating protein. However, Drosophila homozygous for null mutations of an NF1 homolog showed no obvious signs of perturbed Ras1-mediated signaling. Loss of NF1 resulted in a reduction in size of larvae, pupae, and adults. This size defect was not modified by manipulating Ras1 signaling but was restored by expression of activated adenosine 3', 5'-monophosphate-dependent protein kinase (PKA).     Description: Adenosine 3',5'-monophosphate (cAMP) and cAMP-dependent protein kinase (PKA) are regulators of development in many organisms. Dictyostelium uses cAMP as an extracellular chemoattractant and as an intracellular signal for differentiation. Cells that are mutant in adenylyl cyclase do not develop. Moderate expression of the catalytic subunit of PKA in adenylyl cyclase-null cells led to near-normal development without detectable accumulation of cAMP.
