    Notes: Investigations on the Thermal Decomposition of n-Dodecylcyclopentane in the Gas PhaseIn the thermal decomposition of n-dodecylcyclopentane at 670 and 750°C in a laboratory tubular reactor mixtures of hydrogen and 57 and 113 hydrocarbons respectively are formed. The mixtures have been analyzed and identified by capillary gas chromatography and by mass spectrometry. The distribution of the reaction products is used for mechanistic discussions.

    Notes: The introduction of chick erythrocyte nuclei into mammalian cell cytoplasms results in their reactivation as evidenced by the de novo transcription of chick genes and the synthesis of both globin and constitutive proteins. In the present study, chick erythrocytes have been fused to L6 rat myoblasts and to alphaamanitin-resistant variants of L6 to determine whether the chick or the mammalian RNA polymerase II was responsible for transcription of chick genes. Heterokaryons formed by fusing chick erythrocytes with alpha-amanitin-resistant L6 myoblasts synthesize both chick globin and chick constitutive proteins in the continued presence of 5 μg/ml alpha amanitin ten days postfusion. Both the synthesis of globin and other chick polypeptides occurs at levels comparable to those observed for untreated heterokaryons. Synthesis occurs under conditions in which insignificant chick RNA polymerase II activity can be detected irv wild-type heterokaryons by autoradiography. These results demonstrate that RNA polymerase II is one of the mammalian proteins that is selectively taken up by the chick nucleus during reactivation in the presence of alpha amanitin. Furthermore, the mammalian RNA polymerase II alone can account for the transcription of both differentiation specific and constitutive genes in the chick nucleus.

    Notes: The protonation and deprotonation rates of the coordinated amide groups in the Cu2+-complexes of 1,8-diamino-3,6-diaza-2,7-octanedione (DED = L) have been studied by stopped-flow techniques. Starting at low pH from Cu2+ and DED the dimeric Cu2L24+-complex, fully formed within the mixing time of the stopped-flow instrument, reacts in two consecutive steps to yield the final product CuLH-2. The rate constants of the forward and backward reactions have been determined and are given in Table 1. The intermediate was identified as Cu2L2H-22+ by measuring its VIS.-absorption spectrum.The rate constants for the interconversion of the amide groups from the O- to the N-coordinated form in the Cu2+-complexes of DED, 2,10-dioxo-1, 4, 8, 11 tetraazaundecane (DANA) and triglycine (TRIGLY) are compared with each other. It is shown that these rate constants are similar, no matter whether the amide group is terminal or internal as long as the rotation is easily possible as is the case in the dimeric species Cu2L24+ and Cu2L2H-22+. However, for CuLH-2 the inter-conversion only takes place after opening of one of the chelate rings in a rapid protonation preequilibrium.

E-2a isomerizes thermally to give the Z-configuration Z-2a in a very smooth reaction. For systematical investigations the 15-membered ring enynes E-5a and Z-5a were generated. They behave like openchained 1,3-enynes.
    Notes: Die Synthese von (E)-1-Cyclododecen-3-in (E-2a) führt zur Isolierung des ersten trans-Enins mit geometrischer Ringspannung. E-2a wandelt sich thermisch leicht in die Z-Konfiguration Z-2a um. Zur systematischen Untersuchung werden die 15-Ring-Enine E-5a und Z-5a hergestellt. Sie verhalten sich wie offenkettige 1,3-Enine.

    Notes: The characteristics of pure preparations of short-tail fibers of bacteriophage T4 have been studied in the optical and electron microscope. Three main structures were observed: 1) spheres of 8.1 nm diameter; 2) fibers 43 nm long and 3.8 nm thick; and 3) fibers 54 nm long and 3.2 nm thick. Both types of fibers exhibited a regular beaded appearance. The 43-nm fibers were the most abundant structure. During the process of purification of the short-tail fibers, the formation of aggregates was observed each time the material containing the short-tail fibers was dialyzed against saline solutions. These aggregates became increasingly fibrous (as observed in the optical microscope) as the material used was increasingly enriched in short-tail fibers. Finally, most of the aggregates were of the fibrous type when they were formed from a purified preparation of short-tail fibers. In the electron microscope, it was found that the filamentous aggregates were organized in well-defined bundles. The amino acid composition of the highly purified short-tail fibers was also determined. Among the known fibrous proteins, the ones that most resemble the amino acid composition of the short-tail fibers are actin and fibrinogen. These observations are discussed in relation to the T4 short-tail fiber structure and their localization on the hexagonal baseplate of the T4 tail structure.

    Notes: Intact human platelets bind cytochalasin B (CB) with a capacity of 100- 120 p mols CB/mg protein or approximately 7 × 104 molecules/cell and dissociation constants (KD) ranging from 2 × 10-8 to 10-6 M. Up to 85% of this saturable binding is displaced by 10-5 M cytochalasin E (CE). This CE-sensitive binding also appears heterogeneous with KD similar to those of the overall binding. The CE-insensitive binding, however, appears as a single component with KD ≌ 4 × 10-7 M. The sedimentable constituents from frozen, thawed, and washed cells also bind CB with KD ranging from 2.4 × 10-8 to 1.5 × 10-6 M and a total capacity of approximately 39 p mols/mg protein which accounts for only 4% of the ligand binding to the intact cell. The major portion (60-80%) of this CB binding is displaceable by 500 mM D-glucose and has a KD of 1.5 × 10-6M, while only 10-15% is CE-sensitive with a KD of 2.4 ± 10-8 M. It is concluded that 95% of the saturable CB binding in platelets is associated with the cytosol of which 80-85% is sensitive to CE and that only 3% of the cellular binding is glucose sensitive, membrane-associated binding. If the CE-sensitive binding associated with the cytosol is entirely to actin, the stoichiometry of this binding is approximately one CB to 30 actin monomers, which is greater by an order of magnitude than that for CB binding to muscle actin.

    Description / Table of Contents: Bromination and Dehydrobromination in the Side Chains of 1,2,3-ThiadiazolesThe bromination of 1,2,3-thiadiazoles with N-bromosuccinimide (NBS) is investigated. The monobromides 3 and 4 and the dibromides 5 and 6 can be isolated. Several methods have been used for the introduction of double bonds by the elimination of HBr. Besides the elimination products 9, 11, 12, and 14 the substitution products 10, 13, 15, 16, and 17 are obtained. A lot of regio- and stereoselectivities can be observed.
    Notes: Bei den 1,2,3-Thiadiazolen 2a-h wird die Bromierung mit N-Bromsuccinimid (NBS) untersucht. Es können die Monobromide 3 und 4 sowie die Dibromide 5 und 6 isoliert werden. Zur Einführung von Doppelbindungen unter HBr-Abspaltung werden mehrere Verfahren geprüft. Neben den Eliminierungsprodukten 9, 11, 12 und 14 erhält man die Substitutionsprodukte 10, 13, 15, 16 und 17. Man beobachtet dabei eine Reihe von Regio- und Stereoselektivitäten.

    Notes: The in vitro incubation of cells from turpentine-induced rat myeloid hyperplastic marrow and peritoneal monocyte/macrophage with 14C-arachidonic acid resulted in the incorporation of the radiolabel into the particulate phospholipids. Challenge of the radiolabeled cells with a highly purified type I CSF (CSF I) from human pancreatic carcinoma cells in continuous culture resulted in the hydrolysis and release of the 14C-arachidonic acid from the cellular phospholipids. The simultaneous challenge of the prelabeled cells with CSF-I and its specific antibody (anti-CSF-I antibody) inhibited the CSF-I induced hydrolysis of 14C-arachidonic acid from the cells. These results confer a specificity on the CSF-I induced release of arachidonic acid from the cellular phospholipids. Our data also demonstrated that the 14C-arachidonic acid released from the cellular phospholipids was further transformed into products of the cyclooxygenation and lipoxygenation pathways by cellular enzyme systems in both populations of cells. Interestingly, our data also indicate that the challenge of the granulocytic hyperplastic marrow cells and the monocyte/macrophage cells with purified CSF-I resulted in a higher generation of lipoxygenase products in the predominantly granulocytic cell population than in the population rich in monocyte/macrophage cells. The biological significance of this observation remains to be further explored. Thus, the CSF-I-induced release of cellular arachidonic acid explains, at least in part, the presence of prostaglandins and other metabolites of arachidonic acid that are found in the media of hemopoietic cells incubated with a variety of CSF preparations.

    Notes: Naturally occurring reticulocytes of week old piglets were used to characterize the maturation process under in vitro conditions. When the reticulocytes were suspended in tissue culture medium fortified with metabolic substrates, nearly all cells were viable after 24 hours incubation and usually more than 85% of the initial cell population survived after an 80 hour period. In cells maintained as long as a week in incubation, an adequate level of total adenine nucleotide with a large accumulation of IMP was found. In most cases, reticulocytes lose their reticular materials within two days and assume normal erythrocyte configuration. Concomitant with the morphological change, the cell volume decreases toward normal erythrocyte size, the extent of which can be accounted for by the intracellular loss of salt and accompanying water. As in the in vivo reticulocyte maturation process, reticulocytes undergoing in vitro maturation lose their membrane permeability to glucose. These findings suggest that the process of reticulocyte maturation occurring in cell culture approaches that which naturally occurs in vivo. Thus, these cells may be used to delineate the mechanism of the loss of membrane transport of glucose which normally occurs in the adult pig cells.
