    Notes: Abstract A new magnetic discharge stabilization technique for coaxial laser systems is described. The approach utilizes crosses electric and magnetic fields to create and maintain a large and rapidly rotating plasma volume which does not experience glow-to-arc transitions. Very high cw specific discharge power loadings have been achieved even without the benefit of external gas cooling or circulation. Performance is insensitive to gas composition and pressure such that high power coaxial discharges have been run in CO2 laser gas mixtures up to several hundred torr. Stable cw discharges have also been obtained in mixtures containing several torr of SF6. The technique appears to be readily scalable to give very large excited volumes in systems with comparatively small overall physical dimensions.

    Notes: Abstract We have designed and tested a pulsed single-mode dye laser for spectroscopy experiments. The dye laser is pumped by a 6 kHz copper vapor laser to produce up to 230 mW average power at over 5% efficiency in a nearly diffraction limited beam. The dye laser pulse closely resembles the 30 ns pump pulse. Using an electronic servo loop, the 60 MHz wide line can be scanned, without mode hopping, through the 16 GHz permitted by the travel on the piezo driven mirror mount.

    Notes: Abstract A far-infrared (FIR) NH3 laser was resonantly pumped with a line-tunable infrared (IR) NH3 laser. The number of the observed FIR laser lines amounted to 33. Most of them belonged toaR(J,K) rotation-inversion transition in (0, 1, 0, 0) vibrational state. The line tunability of sealed-off FIR NH3 laser was almost achieved in 90, 115, 150, and 220 μm wavelength regions by the selective line tuning of the pumping IR NH3 laser.

    Notes: Abstract The cw output power of the uv CuII laser has been optimized with respect to the hollow cathode geometry, the discharge current, the fill gas pressure and the resonator mirror parameters. A maximum laser output power of 900 mW for multiline operation at 248.6, 259.1, 260.0, and 270.3 nm was achieved with 100 A discharge current, 260 V voltage and 16 mbar fillgas pressure, when a hollow cathode of 1.2 m length and 2×6 mm2 cross section was employed. The single-pass pain gl has been estimated to 7%. A hollow cathode cross section of 1.5×4.5 mm2 is suggested as an optimum geometry. In addition, some investigations on the mechanism of the laser power decay in Ne−Cu-discharges are presented.

    Notes: Abstract A theoretical treatment of the influence of a saturable absorber in close contact with a highly reflecting mirror on a light pulse is given. The shaping of a Gaussian pulse during a single passage through such an absorber is calculated for various absorber lengths. The influence of absorber position and length on the pulse parameters and the stability of a continuously pumped dye laser are investigated. The shortest pulses and the most favourable operation conditions of the laser are to be expected in the case of a short contacted saturable absorber.

    Notes: Abstract Single-mode selection with Michelson selectors in cw standing-wave dye lasers is analyzed. The application of single and double Michelson mode selectors for compensation of spatial hole burning effects is described in detail. Upper limits for the pumping are considered. It is shown that with spatial hole burning compensation, even the single Michelson selector provides sufficient selectivity for high power single-mode operation in linear cavities.

    Notes: Abstract A simple method for single-frequency operation and stabilization of a low power He−Ne laser has been described and used. With a proper resonator length very weak sideband modes on each side of the strong mode produce a feedback signal to a piezoelectric pusher which controls the resonator length. The long-term frequency stability was estimated to be about ±10 MHz.

    Notes: Abstract Simultaneous laser operation at the wavelengths of λ1=637 and λ2=717 in Pr0.5La0.5P5O14 single crystals was observed. The crystals were excited by nanosecond publses from a Coumarin 2 dye laser (λ=450 nm). Laser emissions and their interdependence were studied both in crystals without an external resonator and in crystals placed within a plane parallel resonator. The energetic, time and frequency characteristics for both laser actions were examined. The effect of bifrequential emission was also found in crystals of different ratios of Pr and La ions concentration and in pure praseodymium pentaphosphate.

    Notes: Abstract We report infrared laser emission in the region of 3 to 5 μm from sodium vapour optically pumped by a pulsed dye laser with wavelengths ranging from 585 to 610nm. Twophoton excitation processes are believed to be responsible for the primary excitation. Both molecular transitions (4 to 5 μm) between high lying states, and atomic transitions (52 S 1/2−42 P 3/2,1/2 at 3.41 μm) have been identified.

    Notes: Abstract The presented theoretical model for a mode-locked Nd-glass laser simultaneously takes into account dynamics of the mode-locking dye, amplification saturation and radiation background. A systematic variation of laser parameters gives insight into the pulse formation process and allows to improve the laser design. The calculations show that it should be possible to decrease considerably the duration of light pulses of a mode-locked Nd-glass laser. Using a new mode-locking dye with a switching time of τ=2.7×10−12 s we obtained stable laser operation and a pulse duration of 1.7×10−12s.

    Notes: Abstract The characteristics of near uv dye lasers pumped with short duration, 308 nm pulses from a XeCl laser are described with particular emphasis on the parameters influencing high average power operation. The solvent is shown to effect the operation of all dye lasers studied, particularly the photochemical stability. Measurements of the efficiency, as a function of the integrated pump energy have been made for a series of dye-solvent combinations. With the best combinations, average output powers in excess of 1W and conversion efficiencies greater than 25% were obtained at a repetition rate of ∼100 Hz.

    Notes: Abstract Near-infrared laser action of atomic carbon was obtained using a hollow cathode discharge tube.     Notes: Abstract This work was an experimental investigation of a potential large-scale excimer laser system, in which a high-pressure KrF gas mix was pumped by the 4-MeV electron beam from an available linear accelerator. As a function of the mix and the pressure, fluorescence from KrF* and/or Kr2F* were produced with good efficiency. Comparing measured data with a kinetic model, it was found that the kinetics followed the “normal” patterns seen in low-energy electron pumping. The data fitting process also resulted in a new value for the three-body quenching rate constant for reactions involving Ar. A critical factor in the development of this laser will be an improvement in beam control over that afforded by a simple solenoidal magnet. The conditions necessary for lasing are predicted; no inherent limitations on scaling of the device were found.

    Notes: Abstract We investigated time-dependent effects related to the bistable operation of a CO2 laser with an intracavity nonlinear absorber (SF6). The response of the system to small perturbations was sensitively dependent on the operating laser line [P(12)→P(30)] of the 10.6 μm band. The narrow bistable region forP(12), due to a very narrow hysteresis loop, can be effectively utilized for optical switching, because of its pronounced sensitivity to variations in operational parameters. The maximum admissable beam blocking time (intracavity) for spontaneous return to lasing was measured forP(16), for a range of absorber pressures. This τm is related to the relaxation time of the upper level in the absorber.

    Notes: Abstract A tunable diode laser and a multipass optical cell are used to investigate the feasibility of performing radiocarbon dating by means of infrared spectroscopy. We show that the detection of14CO2 at concentrations of14C/12C≲10−12 is not limited by interferences from the background of normal CO2 molecules, provided the gas sample is cooled. Using short-time constants, the present sensitivity of the infrared technique is comparable to that of an ideal disintegration counter. The experimental results are extrapolated to long time constants to demonstrate that radiocarbon dating by infrared spectroscopy is feasible, and that carbon samples with a mass of less than 1 gm can be dated by this technique.

    Notes: Abstract Newly found reflection properties of the Fizeau wedge interferometer are shown to be of particular advantage in obtaining efficient multi-wavelength operation of dye lasers. Use is made of the fact that the reflecting Fizeau wedge can ensure at once a selective feedback for a given wavelength and quasi-total reflection in an other direction for all other wavelengths. Fully independent tuning of two 0.02 nm lines is obtained with short pulse (2 ns) N2-laser pumping using a single Fizeau wedge.

    Notes: Abstract A synchronously mode-locked, cavity-dumped picosecond dye laser is described. The structure and intensity of the picosecond pulses measured under different conditions are reported. It was found that the structure of the pulses from the synchronously pumped dye laser depends critically on the length of the Ar+ laser pulses. At the shortest Ar+ laser pulses of about 70 ps the dye pulses are as short as 1.1 ps. With Ar+ laser pulses of 200 ps the dye laser pulses contains a broad satellite pulse which contains a large fraction of the total intensity. When a cavity dumper is added to the system one gets dye laser pulses 15–20 ps long with a substructure, which indicates incomplete mode-locking. Well mode-locked 1.5–2.0 ps pulses were obtained in the red part of the dye laser action spectrum, i.e. 620–650 nm for R6G, 595–608 nm for R 110 and 657–662 nm for RB, respectively. Addition of mode-locking dyes also improved the pulse quality at some wavelengths.

    Notes: Summary Embryonic lethal mutations at the Notch locus are known to produce a conspicuous central nervous system hypertrophy accompanied by a hypotrophy of the epidermal sheath. We have studied several zygotic mutants belonging to four different autosomal complementation groups which produce the same phenotype. The embryonic development of the new mutants, as well as that of Notch, consists of an initial enlargement of the neurogenic region at the expenses of epidermal cell precursors. The possibility is discussed that these five loci are involved in the determination of neural and epidermal cell precursors.

    Notes: Summary New alleles of thetrithorax locus have been isolated and analysed. The phenotypes of different allelic combinations confirm that a decrease or loss of function of the locus is responsible for homoeotic transformations of the adult thoracic and abdominal segments. Since neither these homoeotic transformations nor larval lethality are complemented by the previously described mutationRg-bx, it is concluded that the latter is an allele oftrithorax. The effect of near loss of function of thetrithorax locus after the completion of embryogenesis has been investigated by clonal analysis. This has produced two unexpected results: 1) there is a requirement for the activity of the locus after embryogenesis is completed; 2) the locus is required for the normal development of at least some mesothoracic structures. These results are discussed with respect to the selector gene hypothesis and other putative activator gene mutations.

    Notes: Abstract In preparation for the isolation and biochemical characterization of putative RNA polymerase mutants, DNA-dependent RNA polymerases of Drosophila melanogaster adults were isolated and partially characterized. Approximately 70% of the female adult RNA polymerase is located in ovaries. Multiple forms of ovarian RNA polymerases I and II are separable by DEAE-Sephadex chromatography. The two forms of RNA polymerase II differ in ammonium sulfate optima. RNA polymerase IIA is more active with double-stranded DNA as template, whereas RNA polymerase IIB transcribes single-stranded DNA most efficiently. Rechromatography of RNA polymerase IIA on DEAE-Sephadex results in the loss of ability of this form to transcribed double-stranded DNA most efficiently. Ovariectomized carcasses have two forms of RNA polymerase I and one form of RNA polymerase II and each transcribes single-stranded DNA most efficiently. As judged by gel filtration chromatography, female adult extracts have forms of RNA polymerase II that differ in molecular weight and template preference.

    Notes: Abstract Two leucine aminopeptidases from Drosophila melanogaster larvae have been partially purified. The LAP A and D enzymes have similar biochemical characteristics including molecular weights of ≅280,000 daltons, Michaelis-Menten constants of ≅0.05 mM, associations with metal cofactors, and specificities toward natural and chromogenic substrates. They differ in their pH optima and spatial distributions. If the closely linked genes that code for these enzymes have resulted from a tandem gene duplication event, it is suggested that there has been subsequent evolutionary divergence. This would provide Drosophila larvae with two related, but functionally distinct enzymes.

    Notes: Abstract A method for assaying Drosophila melanogaster adult DNA-dependent RNA polymerase II in crude extracts from as few as two females or three males is described. Preparation of the extracts involves incubating homogenates at 25 C for 60 min and subsequent treatment with Macaloid. Eighty-five percent of the activity in the extracts is inhibited by 1 µg/ml α-amanitin and this fraction is attributed to RNA polymerase II. RNA polymerase II activity in the extracts shows a good dose dependence and a partial dependence on added DNA, Mn2+, and all four ribonucleoside triphosphates. The kinetics of heat inactivation of RNA polymerase II in crude extracts could be reproducibly measured. Flies of different genotypes had different initial rates of RNA polymerase II heat inactivation. The isolation of Drosophila melanogaster α-aminitin-resistant mutants is also reported. Using the assay described in this paper, it appears that the basis for the resistance is an altered RNA polymerase II. The mutation has been mapped to the third chromosome by chromosome replacement.

    Notes: Abstract An electrophoretic variant in the LDH (l-lactate:NAD oxidoreductase, E.C.1.1.1.27) of Drosophila melanogaster was observed on starch (or polyacrylamide) gels. This variant was found to exhibit an identical isozymic pattern (three isozymes with a decreasing staining density) on starch gel and map position as the Adh locus. On the other hand, anodal polyacrylamide gel electrophoresis in crude extracts has shown LDH to consist of nine bands and ADH of four bands. We have shown that ADH (Alcohol:NAD oxidoreductase, E.C.1.1.1.1) also oxidizes l(+)-lactate or d(−)-lactate with the NAD, while LDH oxidizes ethanol. By using various genetic and biochemical techniques, we have shown that the observed Ldh electrophoretic variant was not a real one and could be attributed to the presence of ADH. We have called this phenomenon “pseudopolymorphism,” and the problem of enzyme specificity has been examined. The appearance of a band in an assay using lactic acid as a substrate is not sufficient evidence for the presence of LDH. Hence, caution is called for before characterizing an electrophoretic band on a gel as being equivalent to the presence of a genetic locus. Out of the nine electrophoretic zones of activity observed on polyacrylamide gel (or out of the six previously observed) using crude extract, only two (one major and one minor) belong to LDH, as revealed by purified enzyme preparations. Furthermore, purified LDH exhibits activity in two bands on starch gel (out of three observed in crude extracts), which appear in different positions as compared with those of ADH. Finally, one band which responds to the presence of d(−)-lactate but not to l(+)-lactate has been revealed.

    Notes: Summary An initially rare allele with a large effect on a quantitative character is expected to exhibit the following behaviour in artificial selection lines: 1. It should change in frequency, or be lost by chance, at rates predictable from the effects of the allele on the quantitative character and the selection regime imposed. 2. At the phenotypic level the behaviour of the allele should cause (a) asymmetrical responses to bidirectional selection, (b) variation among replicate lines in response to selection corresponding to the behaviour of the allele in individual lines, (c) changes in heritability in lines in which the allele increases in frequency and (d) selection response compatible with the effects of the allele. This paper reports an experimental evaluation of these predictions utilizing a rare allele of large effect (sm lab ) detected in a sample of the Canberra outbred population of Drosophila melanogaster at a frequency of 1/120. Homosygosity for this allele reduced abdominal bristle number by more than 50%, altered the abdominal bristle pattern and reversed the sexual dimorphism for abdominal bristle number. Experiments were done to characterise sm lab and all evidence indicates that it is a single allele with a very large effect. Bidirectional selection for abdominal bristle number was carried out in three high and three low lines from this sample of the Canberra population. The sm lab allele rose in frequency and went to fixation in two of the low lines (in 10 generations) but was lost from the third. These times to fixation were slower than the expectations derived from computer simulations of the behaviour of such an allele but this can be attributed to the lower fitness of sm lab homozygotes. The proportions of lines with the allele fixed or lost were compatible with expectations. At the phenotypic level, the behavior of sm lab had the expected consequences, namely, (i) asymmetrical responses to bidirectional selection, (ii) variation in response among replicate low lines corresponding to the behaviour of sm lab , (iii) changes in heritabilities in the lines in which sm lab went to fixation, and (iv) selection responses compatible with the effects of the allele. A test for rare alleles of large effect was proposed, based on the expected pattern of change in heritability under artificial selection. This test was applied to the high selection lines but no evidence was found for important effects due to rare alleles of large effect increasing abdominal bristle number, a conclusion consistent with other independent evidence. This work provides experimental corroboration of the links between population genetics and quantitative genetics.

    Notes: Summary Significant changes of genotypic structure in 20 lines selected for wing length are detected by analysis of the allelic frequencies of several enzyme loci (XDH, LAP-D, EST-6, 1-APH, ADH, α-GPDH). These changes are not haphazard but a consequence of the effects of selection on the genetic structure of the population, since replicate lines always behave in a parallel way. The changes are larger in the lines selected for short wings, in which the genetic variability decreases considerably. This decrease is the result of selection for homozygosity, detected at the allozyme loci, but most probably reflects homozygosity of more or less extended chromosomal segments. Selection for wing length, especially for short wings, favoured recombinants of the initial founder chromosomes. Only in the 1-APH and the EST-6 loci, separated by 11.7 centimorgans on the genetic map, do the alleles linked in the founder lines change in parallel in the control and long wing lines. The correlated response in the allozyme allele frequencies cannot be accounted for by a direct influence of the allozymes on the variability in wing length. The changes in the EST-6, 1-APH and perhaps in the LAP-D, can be explained by a direct effect of natural selection on the allozyme loci, probably in interaction with the effect of selection for wing length on linked loci. This last effect seems to be the main factor contributing to the change detected in the XDH locus.

    Notes: Summary Phase-contrast microscopy of 2.5 μm plastic sections reveals five different kinds of sense organs on the internal mouthparts of Drosophila melanogaster: a ventral group of sensilla on the epipharynx, three kinds of sense organs in the middle part of the sucking apparatus (the cibarium), one of them being described here for the first time, and an additional type of sensillum in the dorsal pharynx (Fig. 1). Three of these internal sense organs resembling each other structurally form similar projection patterns in the tritocerebrum, whereas external taste receptors on the labial palps show a different pattern in the suboesophageal ganglion (Fig. 12). The central projections of these internal and external sensilla were traced by applying an ethanolic solution of cobalt on the intact cuticle.

    Notes: Abstract Esterase 6 (EST 6) inDrosophila melanogaster is a male reproductive enzyme transferred to females as a component of the seminal fluid [Richmond, R. C., Gilbert, D. G., Sheehan, K. B., Gromko, M. H., and Butterworth, F. W. (1980).Science 207:1483–1485]. Here we report investigations into the relation between EST 6 and remating by females. EST 6 activity in a strain selected for decreased time to remating is increased over control levels. Inseminated females remated to males carrying null or active alleles show no differences in the timing of remating. However, females inseminated by EST 6-active males remate significantly sooner than females inseminated by null males. Interrupted copulation experiments demonstrate that the remating effect is not due to EST 6 alone but requires other components of the ejaculate. Other evidence suggests that sperm stored in the ventral receptacle respond to EST 6 levels and control remating time. As the first mate of a female who will remate, null-EST 6 males have, under laboratory conditions, a significantly higher fitness than males carrying active alleles. Thus the absence of null alleles of EST 6 in natural populations presents a dilemma suggesting that the remating effect of EST 6 may be balanced by other effects on reproduction.
