    Notes: Summary The concentration of18F was measured in three parts of the femur and in the blood of 36 mice killed at intervals of from 1 min to 2 h after intravenous injection. Blood supply to the bone sections, calculated by dividing the increase in18F concentration in bone by the integral of18F concentration in blood between selected time intervals, was shown to be unaffected by the return of18F from bone to blood at times earlier than 90 min following injection. The supply of blood per unit mass of bone was greatest in distal end of the femur, less in proximal end and least in shaft. In whole femur average blood flow was 230 mm3 per gram fresh bone per min. The deposition of239Pu in the three parts of the mouse femur was shown to be linearly related to blood flow and the nature of the relationship is discussed.

    Notes: Summary Male mice homozygous for the recessive p-locus allele p s are sterile. Spermatogenesis is normal through the completion of meiosis, but is abnormal from the Golgi phase of spermiogenesis on. During acrosome formation various abnormalities appear, ranging from atypical Golgi-acrosome complexes to multiple acrosome “implantation” sites on the spermatid nuclei. In many developing acrosomal caps the distribution of acrosomal material is irregular. During the nuclear elongation phase, bundles of microtubules oriented parallel to the manchette have been seen lying in cylindrical invaginations of some spermatid nuclei. In later spermatids, chromatin condensation appears to proceed normally but may be unduly delayed in reaching completion. These various perturbations give rise to a wide spectrum of abnormal spermatozoa ranging from spermatozoa with heads of near-normal morphology to highly bizarre heads which have lost their chromatin. Sperm flagellar ultrastructure is normal and all sperm tails; even those devoid of recognizable heads, are highly motile. These findings support the view that the development of the sperm head is under the control of a group of genes distinct from those mediating events involved in flagella development.

    Notes: Summary Gametogenesis is normal through meiosis and the earliest phases of spermiogenesis in male-sterile mice homozygous for the recessive, pleiotropic, mutation hpy (hydrocephalic-polydactyl). However, structurally complete sperm flagella were not encountered. Instead, partially assembled axonemal structures and/or poorly organized aggregates of other tail components (mitochondria, outer coarse fibers) were seen at the posterior poles of nuclei in older spermatids. The ultrastructure of centrioles in spermatids was normal, but that of axonemes associated with them was not. These findings suggest that the observed flagella dysgenesis results from defects in assembly rather than from defective intiation centers. Released “gametes” usually consisted of distorted nuclei and associated acrosome enclosed in a relatively close fitting plasma membrane. Perturbations of sperm head development were also encountered; they included extreme nuclear elongation, and distortion of the acrosome and underlying nuclear material by inpushings of finger-like processes of Sertoli cells. It is believed that sperm head anomalies are secondary consequences of the mutant condition. The findings support the view that the hpy locus represents one of a number of genes primarily involved in the mediation of flagella development.

    Notes: Summary After gentle trypsinization, the pseudostratified columnar Müllerian epithelium that lines the uterine cervix of newborn mice could be separated from the enclosing stromal tissue. Pure epithelial tubes explanted in vitro and were allowed to grow in a standard medium for 3–4 days forming a confluent colony of rather closely-fitting cells. The cell sheet was studied by a preparatory technique that allows examination of a large number of cells with preserved intercellular spatial orientation. Attempts were made to identify cultured cells according to the morphology of cell types in the cervicovaginal epithelium in vivo. Electron micrographs revealed that, close to the explant, the cultured cell sheet exhibited several features similar to the Müllerian epithelium in vivo. Outside these central areas of the colony was a broad transitional zone consisting of thin platelike cells distinguished by an abundance of microfilaments. At the periphery of the colonies, bulky cells possessing microvilli and a vacuolated cytoplasm tended to overlap adjoining platelike cells. These bulky cells had a morphology resembling that of the superficial cells seen in the upper vagina and common cervical canal of immature and diestrous animals. The epithelial development in the cultures apparently simulated the transformation in vivo from a pseudostratified Müllerian epithelium in the newborn to a stratified epithelium resembling that of the uppermost vagina and common cervical canal of immature animals. Judged by morphological and cytochemical criteria, the Müllerian cells in the outgrowth obviously had many changed features. It thus seems questionable whether the cells grown in vitro are comparable with the corresponding cells in vivo when used for experiments requiring the controlled conditions of the culture environment.

    Notes: Summary Evans blue was injected into the soleus muscle of albino mice in order to mark retrogradely the corresponding motoneurons of the spinal cord. Subsequently, reaction kinetics of acid phosphatase were studied in the marked nerve cells. 41–51 motoneurons per animal were counted. They are located in the dorsolateral portion of the Rexed zone IX where they form motor cell columns approximately 1.5 mm in length. After identification of the motoneurons, the enzyme reaction for acid phosphatase was performed by covering the section with a gel film containing naphthol-AS-BI-phosphate-hexazonium-pararosanilin. The formation of the azo dye was measured cytophotometrically at 520 nm. During the first 10 min of registration, a linear decrease in transmission of 0.4 per cent/min was shown. Because of the unimodal distribution of changes in transmission, the motoneurons of the soleus muscle could not be characterized as fast or slow types on the basis of the reaction kinetics of acid phosphatase.

    Notes: Summary The development of the nerve supply of the pituitary pars intermedia (PI) of C3H mice was studied by electron microscopy. Nerve fibres and terminal structures, most probably adrenergic, first appear in the newborn. The adult innervation pattern is achieved by the end of the first postnatal week. In the adult animal two types of nerve terminals were distinguished; type A (peptidergic or neurosecretory) and type B (adrenergic). The peptidergic fibres were scarce and exhibited no synapse-like contacts. It is suggested that they are of secondary importance in a direct nervous hypothalamic control of PI function. Type B terminals were found throughout the PI. They formed synapse-like contacts with the glandular cells, indicating that the primary innervation is exerted by adrenergic neurons. An autonomous differentiation of the glandular cells and in the adult a combined direct nervous and neurohumoral control of PI function is suggested.

    Notes: Summary Ovary-independent (estrogen-independent) irreversible proliferation and cornification of the vaginal epithelium in ovariectomized mice caused by neonatal injections of 20 μg estradiol-17β (E2) was suppressed by injections of 200 IU vitamin A acetate (VA) given simultaneously with E2. More than 2 months after the combined treatment were required for the suppression. A new type of cell of low electron density (Light cells) appeared at 40–120 days of age on the basal lamina of the vaginal epithelium in neonatally E2 plus VA-treated ovariectomized mice only at 50 days. Light cells contained small amounts of organelles. Columnar basal cells resembling those in normal estrous mice were also found in the E2 plus VA-treated mice at 70–120 days. In one of the 120-day-old, E2 plus VA-treated mice, the epithelium consisted of only 2–3 layers of cells. Neither Light cells nor columnar cells were found in the atrophic epithelium.

    Notes: Summary Recent scanning electron microscopic studies confirm the presence of solitary cilia on most epithelial cells along the mammalian nephron and collecting ducts. By transmission electron microscopy we have found that the axonemata of such cilia consist of a maximal number of 9 doublet and no singlet filaments. 10% of the cross-sectioned cilia contain 9 doublets arranged in a peripheral ring (9+0 pattern). 30 % of the cross-sections contain 8 or 7 doublets in peripheral ring and 1 or 2 doublets in the central region (8+1 and 7+2 patterns). Serial sections and goniometer tilt reveal the central doublets to originate as dislodged peripheral doublets. 60% of the sectioned cilia contain filament numbers between 8 and 4. In patterns of 5 and 4 filaments single microtubules predominate. The functional significance of these atypical cilia is discussed.

    Notes: Summary Freeze-etched preparations of the ventricular surfaces of ependymal cells clearly reveal the presence of pinocytotic vesicles opening into the third ventricle and large vacuoles formed by broad cell projections. The density of the vesicular openings is approximately 20 per μm2. The ependymal cells in the median eminence of the frog are adjoined by tight junctions comprised of five to eight interconnected junctional strands, whereas near the median eminence in the mouse only one to two such strands form the tight junction of the ependymal cells. Gap junctions between the adjacent ependymal cells are detected near the median eminence in the mouse but not in the frog.
