    Notes: Andalusite porphyroblasts are totally pseudomorphosed by margarite–paragonite aggregates in aluminous pelites containing the peak mineral assemblage andalusite, chlorite, chloritoid, margarite, paragonite, quartz ± garnet, in a NW Iberia contact area. Equilibria at low P–T are investigated using new KFMASH and (mainly) MnCNKFMASH grids constructed with Thermocalc 3.21. P–T and T–X pseudosections with phase modal volume isopleths are constructed for compositions relatively richer and poorer in andalusite to model the assemblages in an andalusite-bearing rock that contains a thin andalusite-rich band (ARB) during retrogression. Their compositions, prior to retrogression, are used in the modelling, and have been retrieved by restoring the pseudomorph-forming elements into the current-depleted matrix, except for Al2O3 which is assumed to be immobile. Compositional differences between the thin band and the rest of the rock have not resulted in differences in andalusite porphyroblast retrogression. The absence of chloritoid resorbtion implies either a pressure increase at constant reacting-system composition, or that its composition changed during retrogression at constant pressure, by becoming enriched in the progressively replaced andalusite porphyroblasts. T–X pseudosections at 1 kbar model this latter process using as end-members in X, first, the restored original rock and ARB compositions, and, then the same process, taking into account the change in composition of both as retrogression proceeded. The MnNCKFMASH pseudosections of rocks with different Al contents facilitate making further deductions on the rock-composition control of the resulting assemblages upon retrogression. Andalusite eventually disappears in relatively Al-poor rocks, resulting, as in this study, in a rock formed by chloritoid–chlorite as the only FM minerals, plus margarite–paragonite pseudomorphs of andalusite. In rocks richer in Al, chlorite would progressively disappear and a kyanite/andalusite–chloritoid assemblage would eventually be stable at retrograde conditions. The Al-silicate, stable during retrogression in Al-rich rocks, indicates pressure conditions and hence the tectonic context under which retrogression took place.

    Notes: In statistically optimised P–T estimation, the contributions to overall uncertainty from different sources are represented by ellipses. One source, for a diffusion-controlled reaction at non-equilibrium, is diffusion modelling of the reaction texture. This modelling is used to estimate ratios, Q, between free-energy differences, ΔG, of reactions among mineral end-members, to replace the equilibrium condition ΔG = 0. The associated uncertainty is compared with those already inherent in the equilibrium case (from end-member data, activity models and mineral compositions). A compact matrix formulation is introduced for activity coefficients, and their partial derivatives governing error propagation. The non-equilibrium example studied is a corona reaction with the assemblage Grt–Opx–Cpx–Pl–Qtz. Two garnet compositions are used, from opposite sides of the corona. In one of them, affected by post-reaction Fe, Mg exchange with pyroxene, the problem of reconstructing the original composition is overcome by direct use of ratios between chemical-potential differences, given by the diffusion modelling. The number of geothermobarometers in the optimisation is limited by near-degeneracies. Their weightings are affected by strong correlations among Q ratios. Uncertainty from diffusion modelling is not large in comparison with other sources. Overall precision is limited mainly by uncertainties in activity models. Hypothetical equilibrium P–T are also estimated for both garnet compositions. By this approach, departure from equilibrium can be measured, with statistical uncertainties. For the example, the result for difference from equilibrium pressure is 1.2 ± 0.7 kbar.

    Notes: This paper characterizes the metamorphic thermal structure of the Higo Metamorphic Complex (HMC) and presents the results of a numerical simulation of a geotherm with melt migration and solidification. Reconstruction of the geological and metamorphic structure shows that the HMC initially had a simple thermal structure where metamorphic temperatures and pressures increased towards apparent lower structural levels. Subsequently, this initial thermal structure has been collapsed by E–W and NNE–SSW trending high-angle faults. Pressure and temperature conditions using the analysis of mineral assemblages and thermobarometry define a metamorphic field P–T array that may be divided into two segments: the array at apparent higher structural levels has a low-dP/dT slope, whereas that at apparent lower structural levels has a high-dP/dT slope. This composite array cannot be explained by heat conduction in subsolidus rocks alone. Migmatite is exposed pervasively at apparent lower structural levels, but large syn-metamorphic plutons are absent at the levels exposed in the HMC. Transport and solidification of melt within migmatite is a potential mechanism to generate the composite array. Thermal modelling of a geotherm with melt migration and solidification shows that the composite thermal structure may be formed by a change of the dominant heat transfer from an advective regime to a conduction regime with decreasing depth. The model also predicts that strata beneath the crossing point will consist of high-grade solid metamorphic rocks and solidified melt products, such as migmatite. This prediction is consistent with the observation that migmatite was associated with the very high-dP/dT slope. The melt migration model is able to generate the very high-dP/dT segment due to the high rate of heat transfer by advection.

    Notes: Sapphirine, coexisting with quartz, is an indicator mineral for ultrahigh-temperature metamorphism in aluminous rock compositions. Here a new activity-composition model for sapphirine is combined with the internally consistent thermodynamic dataset used by THERMOCALC, for calculations primarily in K2O-FeO-MgO-Al2O3-SiO2-H2O (KFMASH). A discrepancy between published experimentally derived FMAS grids and our calculations is understood with reference to H2O. Published FMAS grids effectively represent constant aH2O sections, thereby limiting their detailed use for the interpretation of mineral reaction textures in compositions with differing H2O.For the calculated KFMASH univariant reaction grid, sapphirine + quartz assemblages occur at P–T in excess of 6–7 kbar and 1005 °C. Sapphirine compositions and composition ranges are consistent with natural examples. However, as many univariant equilibria are typically not ‘seen’ by a specific bulk composition, the univariant reaction grid may reveal little about the detailed topology of multi-variant equilibria, and therefore is of limited use for interpreting the P–T evolution of mineral assemblages and reaction sequences. Calculated pseudosections, which quantify bulk composition and multi-variant equilibria, predict experimentally determined KFMASH mineral assemblages with consistent topology, and also indicate that sapphirine stabilizes at increasingly higher pressure and temperature as XMg increases. Although coexisting sapphirine and quartz can occur in relatively iron-rich rocks if the bulk chemistry is sufficiently aluminous, the P–T window of stability shrinks with decreasing XMg. An array of mineral assemblages and mineral reaction sequences from natural sapphirine + quartz and other rocks from Enderby Land, Antarctica, are reproducible with calculated pseudosections. That consistent phase diagram calculations involving sapphirine can be performed allows for a more thorough assessment of the metamorphic evolution of high-temperature granulite facies terranes than was previously possible. The establishment of a a-x model for sapphirine provides the basis for expansion to larger, more geologically realistic chemical systems (e.g. involving Fe3+).

    Notes: Large garnet poikiloblasts hosted by leucosome in metapelitic gneiss from Broken Hill reflect complex mineral–melt relationships. The spatial relationship between the leucosomes and the garnet poikiloblasts implies that the growth of garnet was strongly linked to the production of melt. The apparent difficulty of garnet to nucleate a large number of grains during the prograde breakdown of coexisting biotite and sillimanite led to the spatial focussing of melting reactions around the few garnet nuclei that formed. Continued reaction of biotite and sillimanite required diffusion of elements from where minerals were reacting to sites of garnet growth. This diffusion was driven by chemical potential gradients between garnet-bearing and garnet-absent parts of the rock. As a consequence, melt and peritectic K-feldspar also preferentially formed around the garnet. The diffusion of elements led to the chemical partitioning of the rock within an overall context in which equilibrium may have been approached. Thus, the garnet-bearing leucosomes record in situ melt formation around garnet porphyroblasts rather than centimetre-scale physical melt migration and segregation. The near complete preservation of the high-grade assemblages in the mesosome and leucosome is consistent with substantial melt loss. Interconnected networks between garnet-rich leucosomes provide the most likely pathway for melt migration. Decimetre-scale, coarse-grained, garnet-poor leucosomes may represent areas of melt flux through a large-scale melt transfer network.

    Notes: The paper presents a straightforward method of calculating approximate orientation parameters (six degrees of freedom) of a known targeted object relative to a single calibrated camera. These can then initialise an optimised non-linear calculation of the same parameter values. The method uses the mathematics of a scaled parallel projection and is particularly suited to narrow angle images obtained using a telescope, telephoto or zoom lens. However, the method can also be used with cameras having wider fields of view provided the target group occupies a moderately small part of the image.

    Notes: This paper describes three mathematical modelling methods for high-resolution declassified CORONA KH-4B images. Since CORONA images are collected with a panoramic camera, several types of geometric distortions are involved. Two methods use the modified collinearity equations, and the third involves the terrain-dependent rational function model (RFM) which is considered to be a generic sensor model. Comparative analysis of the three mathematical modelling methods is undertaken. The results show that a ± 1·5 pixels level of horizontal and vertical accuracy can be obtained. A digital elevation model (DEM) of a test site is also created.

    Notes: The fifth biennial symposium of the Photogrammetric Society was held at the University of Birmingham on 6th to 8th April. It has been decided to perpetuate the memory of E. H. Thompson by giving his name to the series of symposia which originated at Birmingham in 1971 Eight papers were delivered to the 1979 symposium. They covered a wide variety of topics which included, in addition to a review of Thompson's own contribution to analytical photogrammetry, a modern approach to the handling of parallax measurements; aspects of digitising; low cost automatic contouring; high energy roentgen photogrammetry; applications in anatomy; the photogrammetric operator's point of view; and cadastral surveys. The majority of the papers are published in this issue of The Photogrammetric Record.〈section xml:id="abs1-2"〉〈title type="main"〉RésuméLe cinquième symposium biennal de la Société de Photogrammétrie s'est dérouléà l'université de Birmingham du 6 au 8 Avril.     Notes: The concept of optimum interpolation is briefly explained. The use of polynomial and non-polynomial stepwise regression for obtaining optimum or near optimum interpolation is illustrated. Least squares collocation is presented as an advanced method for optimising stepwise regression whenever the latter does not achieve desired optimum interpolation for given terrain. A Fortran IV program, written to illustrate an optimum interpolation algorithm using stepwise regression and collocation, is found to produce better results than existing interpolation methods. Other applications of such an algorithm are also discussed.〈section xml:id="abs1-2"〉〈title type="main"〉RésuméOn expose brièvement le concept d'interpolation optimale, ainsi que les méthodes d'approximation polynomials ou autres qui permettent de la réaliser localement au mieux. On présente la collocation comme une méthode permettant de parachever l'interpolation; on a trouvé effectivement, avec un programme écrit en Fortran IV, que les résultats obtenus ainsi étaient meilleurs. On envisage aussi d'autre types d'applications de cet algorithme.〈section xml:id="abs1-3"〉〈title type="main"〉ZusammenfassungKurze Erläuterung des Begriffs der optimalen Interpolation. Skizzierung der Anwendung von schrittweiser Regression mit Hilfe von Polynomen oder anderen Verfahren zur Ermittlung optimaler oder genähert optimaler Inter-polationen. Die Kollokation nach kleinsten Quadraten wird als verbesserte Methode zur Optimierung der schrittweisen Regression dargestellt, wenngleich letztere keine optimale Interpolation für ein gegebenes Gelände liefert. Ein Rechenprogramm in Fortran IV, das zur Veranschaulichung eines optimalen Interpolations-Algorithmus mit Hilfe der schrittweisen Regression und Kollokation geschrieben wurde, lieferte bessere Ergebnisse als vorhandene Interpolationsmethoden. Andere Anwendungen eines solchen Algorithmus werden ebenfalls erörtert.

    Notes: A history and analysis of stereoplotting instrument design and construction in the United Kingdom is given, beginning with the earliest instruments of F. V. Thompson dating from 1907 and 1908 and covering all known designs up till the late 1960s. In the first part of the paper, the instruments built in the 1920s and 1930s are described, much new information having come to light recently in the archives of Ban and Stroud Ltd. and in the papers of the late Professor E. H. Thompson. In the second part of the paper, the instruments which were produced during and after the Second World War are discussed in detail, with an emphasis on those designs which are less well known rather than those which were produced in quantity. The ideas and designs of Deville, F. V. Thompson, Fourcade and E. H. Thompson have been the dominating influences throughout the period and a strong thread of continuity can be established between them.〈section xml:id="abs1-2"〉〈title type="main"〉RésuméOn fait l'histoire du restituteur analogique en Grande Bretagne, depuis ses balbutiements (F.     Notes: An algorithm for the generation of digital ground models is described. The algorithm is based on an approximate solution and uses data obtained with simple measuring equipment.     Notes: Problems are sometimes encountered during the relative orientation of close range photography which has a limited usable format and a large Z range. Computer simulations and practical tests with both a comparator and a plotting instrument are used to investigate the problems and possible inaccuracies. Systematic errors in the photogrammetric model are revealed.〈section xml:id="abs1-2"〉〈title type="main"〉RésuméOn rencontre souvent des difficultés lorsqu'on forme un modèle avec des clichés pris à courte distance, à format réduit et lorsque l'objet présente quelque profondeur. On a approfondi ces problèmes en utilisant conjointement simulations et essais pratiques; on a prouvé ainsi que les déformations du modèle étaient systématiques.〈section xml:id="abs1-3"〉〈title type="main"〉ZusammenfassungMitunter ergeben sich bei der relativen Orientierung von Nahbereischsbildern Probleme, weil das nutzbare Bildformat klein und der z-Bereich gross sind. Mit Hilfe von Computer-Simulationen und praktischer Tests mit einem Komparator bzw. einem Auswertegerät wurden die Probleme und mögliche Ungenauigkeiten untersucht. Dabei wurden systematische Fehler in dem photogrammetrischen Modell aufgedeckt.

    Notes: Water currents in an hydraulic model have been determined by means of a photogrammetric method of detecting the movement of tethered buoys. Single vertical photographs, taken by a non-metric camera, have been analysed to produce the required differential measurements.     Notes: This paper describes the production of stereopairs of shadow photographs to simulate X-ray pictures and demonstrates the possibility of determining analytically the precise position in the object space of foreign bodies using a digitiser as a monocomparator and projective transformation theory to dispense with the necessity of undertaking normal inner and relative orientation analytical procedures.A model is constructed and comparison accuracy tests are made using shadow and X-ray picture pairs. Dazu wurde ein Monokomparator als Digitalisiervorrichtung und die Theorie der projektiven Transformation angewendet, urn auf die sonst übliche analytische Bestimmung der inneren und relativen Orientierung verzichten zu können.Es wurden ein Modell konstruiert und vergleichende Genauigkeitstests für aus Schatten-Fotos bzw. Röntgenbildern bestehende Bildpaare durchgeführt. Im Beitrag wird die Meinung vertreten, dass die Theorie der projektiven Transformation bei der Nahphotogrammetrie günstiger ist, als die traditionelle Theorie der topographischen Stereophotogrammetrie.

Other speakers dealt, in turn, with future instrumentation in photogrammetry; aerial triangulation and film stability; photomaps and tests related to the perception of map users; map revision; and the recording of archaeological information.     Notes: The aim of this paper is to offer regional evidence from Spain on alcohol abuse among adolescents. Specifically, we identify the determinants of the decision to abuse alcohol with respect to the five most important regions of Spain, as well as for the country as a whole. To this end, we estimate Probit specifications using data drawn from the Spanish Surveys on Drug Use in the School Population corresponding to 1994, 1996 and 1998. The results first reveal patterns that are qualitatively similar, but quantitatively different. Similarly, it would appear that economic policies aimed at reducing the access of adolescents to alcohol may have a positive effect on reducing abuse. Finally, the results suggest that encouragement be given to healthy habits among young people, as well as to the fight against education failure and the launching of information campaigns that accurately portray the current lifestyle of adolescents.

    Notes: The aim of this study was to determine the knowledge and acceptability of soymilk (defined as consumption pattern/usage and taste) by adult consumers, residing in different socio-economic areas (low vs. high) in Cape Town. The study population of 214 participants were selected using convenience sampling. A questionnaire was developed in order to obtain demographic data (four items); basic general knowledge of soymilk (four items); and acceptability of the tested products (consumption patterns and taste) (eight items). There was a significant difference between the two groups (P = 0.000) regarding basic knowledge of soymilk, with significantly more in the high socio-economic area (HSEA) having a score of at least 75%. Gender (P = 0.082) and age (P = 0.122) did not have a significant impact on the consumption patterns of soymilk. There was no significant difference between the two groups with respect to current usage (P = 0.228) and frequency of use (P = 0.213) of soymilk. However, a significantly smaller group (P = 0.000) of participants in the low socio-economic area had previously tasted soymilk compared with those in the HSEA (16.7% vs. 41.5%; P = 0.000). In conclusion, socio-economic status appeared to be associated with knowledge of soymilk. No significant differences with regard to gender, age or current usage in the two groups studied were noted.

    Notes: SYNOPSIS Examination of stained specimens (protargol and nigrosinbutanol) of hypotrich ciliates during division allows understanding of morphogenesis of buccal structures and cirri. Midventral, frontal, and transverse cirri in both proter and opisthe of Urostyla marina differentiate from a longitudinal series of numerous oblique ciliary streaks in a manner similar to that of Bakuella, Holosticha, Keronopsii. Pseudourostyla, and Uroleptus. This pattern differs markedly from that in Amphisiella, Kahliella, Paraholosticha, and Paraurostyla, in which the fronto-ventral and transverse cirri arise from a series of fewer (2–5) ciliary streaks in a longitudinal or fan-like array. Rows of marginal cirri in U. marina arise independently, as in Urostyla grandis. On the basis of comparisons of both structural and morphogenetic features of this and other hypotrichs, the family Urostylidae is redefined to contain Urostyla (type), Bakuella, Holosticha, Keronopsis, Pseudourostyla, and Uroleptus. Other genera formerly contained in the families Holostichidae and Urostylidae are placed in synonymy, assigned to other families, or held in uncertain familial relationship pending their rediscovery and further investigation.

    Notes: SYNOPSISUltrastructure of the plasmodium wall and of sporogenesis were studied in Myxosoma funduli Kudo infecting the gills of Fundulus kansae (Garman). Plasmodia were located within the lamellar tissues adjacent to sinuses and capillaries. The plasmodium wall consisted of a single unit membrane which was continuous with numerous pinocytic canals extending into the parasite ectoplasm. The plasmodium membrane was covered by a surface coat of almost uniform thickness which prevented direct parasite-host cell contact. Numerous generative cells and cell aggregates, representing early stages of spore development, were seen in immature plasmodia. Later stages of spore development, including mature spores, were observed in older plasmodia. Sporogenesis was initiated by envelopment of one generative cell, the sporont, by a 2nd, nondividing cell, the envelope cell. The sporont and its progeny proceeded through a series of divisions until there were 10 cells, all compartmentalized within the envelope cell. Subsequently, the 10 cells became structurally differentiated and arranged into two 5-celled spore-producing units, each consisting of 1 binucleate sporoplasm and 2 capsulogenic cells, all surrounded by 2 valvogenic cells. Observations of later developmental stages revealed the major events of capsulogenesis, valvogenesis, and sporoplasm maturation, which occurred concomitantly during spore construction.

    Notes: SYNOPSIS Blastocrithidia culicis, Crithidia deanei, Crithidia fasciculata, Herpetomonas samuelpessoai, Leptomonas seymouri and Leishmania tarentolae grown in cultures were compared by electrophoretic mobility for isoenzymes in 6 enzymes. All species were found distinct in these characteristics. Endosymbiotic C. deanei, which was identical to the aposymbiotic C. deanei in 5 enzymes, had an extra band in aspartate aminotransferase. No differences in isoenzymes were found between members of one species maintained in 2 different culture media.

Lorsque les cellules sont cultivées sur milieu contenant du lactate comme seule source de carbone, des divisions synchrones sont observables, indépen-damment des conditions d'éclairement. Toutefois, il existe une relation entre la phase des divisions cellulaires et les périodes lumière-obscurité appliquée à la culture.Pendant le cycle cellulaire, nous montrons que les synthèses des macromolécules sont discontinues: c'est le cas pour les ADN nucléaire et mitochondrial, les ARN ribosomaux et non-ribosomaux, ainsi que pour certaines protéines (cytochrome c 558). Des variations cycliques touchant la morphologie des mitochondries et des chloroplastes sont observées. Au cours du cycle cellulaire, les processus métaboliques séquentiels accompagnent les modifications de structure des organites. C'est ainsi qu'en début du cycle, au commencement de la phase G1, sont synthétisés les ribosomes cytoplasmiques et qu'ensuite, chez les euglènes vertes, les ARN non-ribosomaux sont formés.Ces synthèses d'ARN précèdent l'accroissement du chondriome et du plastidome dans la cellule. En milieu de phase G1, une nouvelle synthèse d'ARN non-ribosomal commence et est observée avant la synthèse des ADN nucléaire et mitochondrial. En fin de phase G1, démarre la division des organites à partir du chondriome et du plastidome en réseau.SYNOPSIS.Synchronous divisions of Euglena gracilis strain Z can be obtained by various methods. When the cells are cultivated in a medium containing lactate as the sole carbon source, synchronous divisions are observed, independent of the conditions of illumination. Nevertheless, there exists a relationship between the phase of cell division and the periods of light and darkness applied to the culture.During the cell cycle, the synthesis of macromolecules is discontinuous—this is true of nuclear and mitochondrial DNA, ribosomal and nonribosomal RNA, and certain proteins (cytochrome c 558). Cyclic variations in the structure of mitochondria and chloroplasts are also observed. In the course of the cell cycle, sequential metabolic processes accompany structural modifications of the organelles. Also, at the beginning of the cycle, at the start of phase G1, the cytoplasmic ribosomes are synthesized, and then, in green euglenids, nonribosomal RNAs are formed. These syntheses of RNA precede enlargement of the chondriome and plastids. In mid-G1 phase, a new synthesis of RNA begins, which precedes synthesis of nuclear and mitochondrial DNA. At the end of G1 phase, division of organelles starts, beginning with the chondriome and plastids, arranged in a network.

    Notes: SYNOPSIS Cell-free synthesis of high molecular weight polypeptides, programmed by RNA from Tetrahymena pyriformis strain W is reported, and methods for preparation of the RNA are described. The RNA was extracted by the SDS-phenol-chloroform-isoamyl alcohol technic. The bulk of extracted RNA was ribosomal and on sucrose gradients peaked at -17S and 25S. After heat denaturation all the 25S RNA was converted to 17S. indicating the presence of hidden breaks, possibly the result of nuclease activity during extraction. Nevertheless, when poly(A)–RNA was collected using oligo-(dT)-cellulose column chromatography, it promoted a 15–fold increase in incorporation of [35S] methionine into TCA-precipitable material. Slab-gel electrophoresis and autoradiography of the product revealed 12 different major polypeptides, varying in weight from 28.000 to 65,000 Daltons. A method for preparation of translatable RNA from Tetrahymena will make possible the comparison of messenger RNAs associated with specific cell structures and with different developmental events.

    Notes: SYNOPSIS. Lorica building was studied experimentally for Tintinnopsis parva Merkle, an agglutinated tintinnid. This species incorporated either siliceous or Ca-rich particles. Evidence of agglutination was seen within 30 min after the addition of particles to cultures of the protozoa. Fully agglutinated loricae were produced by the proter; partially incrusted loricae attributed to an opisthe were also recovered. The cytoplasmic evaginations of unknown function called striae were seen in the scanning electron microscope in better relief than ever before. They may provide the mechanism whereby the enclosed capsules are brought into contact with prey organisms.

A very high percentage of the flies so fed produced established midgut infections, a proportion of which went on to develop into mature metacyclic trypanosomes capable of infecting mammalian hosts. The method offers a safe, clean way of infecting tsetse flies with African trypanosomes which reduces the need for trypanosome-infected animals in the laboratory.

The helical coiling state of a glycerinated stalk in the family Vorticellidae had been regarded, without any evidence, as a reversible and completely equilibrated mechano-chemical system regulated by the external free Ca2+ concentration. Our present detailed observations of many individual stalks of glycerinated Vorticella convallaria revealed that the contraction-extension cycle of the helical stalks caused by increasing and decreasing free Ca2+ concentrations in the medium represents a kind of hysteresis. This phenomenon was observed in extensively glycerinated Vorticella in a medium of very low or physiological ionic strength, as well as in briefly glycerinated ciliates. With regard to the configurational change in various parts of a glycerinated stalk during the contraction-extension cycle, it was found that the initial bending of the stalk caused by increasing free Ca2+ concentration begins to take place near the zooid (head) while the initial reextension of the stalk with decreasing free Ca2+ concentration begins to take place near the zooid (head) while the initial reextension of the stalk with decreasing free Ca2+ concentration takes place simultaneously throughout the entire stalk. The Ca2+ threshold for complete reextension of the part of the stalk near the rootlet was found to be much higher than that near the zooid.

    Notes: SYNOPSIS. Amblyospora sp. in Culex salinarius (Coquillett) is transovarially transmitted and has 2 developmental sequences, one in each host sex. In females, the entire life cycle is restricted to oenocytes which become greatly hypertrophied due to the multiplication of diplokaryotic cells during merogony and come to lie next to ovaries. Sporulation occurs only after a blood meal is taken and is shortly followed by infection of the oocytes and subsequent transmission to the next host generation. In the male host, infections spread from oenocytes to adipose tissue where diplokaryotic cells undergo a 2nd merogony. During this merognic cylce, the number of diplokaryotic cells greatly increases and the infection is spread throughout the body of the larval host. Sporulation is initiated with the physical separation of the 2 members of the diplokaryon and the simulatneous secretion of a pansporoblastic membrane. Subsequent meiotic division and morphogenesis result in the formation of 8 haploid spores enclosed with a pansporoblastic membrane. Buildup of spores and subsequent destruction of host adipose tissue prove fatal to the male host during the 4th larval stage.

Glutamate decarboxylase, γ-aminobutyrate-α-ketoglutarate aminotransferase and NAD-linked and NADP-linked succinic semialdehyde dehydrogenase, all constituting the GABA (γ-aminobutyrate)-shunt pathway of glutamate metabolism are localized in the mitochondrial matrix in a streptomycin-bleached mutant of Euglena gracilis strain Z. Glutamate dehydrogenase, requiring NADP as the cofactor, was distributed in the cytoplasm. An improved version of the controlled digestion method for preparing Euglena mitochondria, which involves use of trypsin and a trypsin inhibitor and removal of broken cells before mechanical disruption of cells, is also described.

    Notes: SYNOPSIS. By phenol extraction and DEAE cellulose column chromatography, tRNA was isolated from Tetrahymena pyriformis strain GL. Following acid hydrolysis of the tRNA, the methylated purine content was determined by Dowex 50 column chromatography and paper chromatography.The most abundant methylated guanine derivative was found to be N2-DMG. Also present were 1-MG, N2-MG, and 7-MG. The most abundant methylated adenine was found to be 1-MA; no 2-MA was detected. Small amounts of the N6-methyladenines were detected.

    Notes: New fish species and geographic records for Rhabdospora thelohani Laguessé, 1895 (rodlet cells) are presented. Additionally, the ultrastructure of R. thelohani in Alburnoides bipunctatus ohridanus Karaman, Borostomias antarcticus (Louml;nnberg), Leuciscus cephalus albus Bonaparte and Rutilus rubilio (Bonaparte) is compared with that reported by other authors and with members of Subphylum Apicomplexa. The ultrastructure of R. thelohani was similar in all the fish species examined: however, the organism was not present in all members of any single species and had intertissue density variations. Rhabdospora thelohani is pyriform. averaging in size 7 × 12 m̈, with a basal nucleus. The surface complex is composed of a layer (0.5 m̈m diameter) formed by microfilaments (9.3 nm) and an outer trilaminar membrane (9.3 nm). The cytoplasm contains structures identical to rhoptries, micronemes and subpellicular microtubules. Mitochondria. Golgi apparatus, and rough endoplasmic reticulum were not observed, although free ribosomes were present and arranged in a vesicular pattern. The observations suggest that the organism moves between cells of epithelial layers and is either released into a lumen intact or passively or actively discharges its contents into a lumen. Results from this study indicate that R. thelohani should be considered a member of Apicomplexa unless definitive evidence is presented to the contrary.

    Notes: The effect of exposure period and concentration of algae on the frequency of infection of aposymbiotic ciliates by algae obtained from the same clone of Paramecium bursaria syngen 2, was studied. The frequency of infection was roughly proportional to the algal concentration and to the exposure time of ciliates to algae. The relationship of algal concentration to infection frequency closely fitted the Poisson distribution curve for N = 1, suggesting that the minimum number of algae required to infect a single ciliate is 1. However, the data also strongly suggested that the average number of algae required to initiate infection of an average ciliate was ˜ 1,000. Three possible resolutions of this situation are: (a) the selection by the ciliate of a rare infective variant from a heterogeneous population: (b) the rare escape of an alga from digestion by the ciliate; and (c) the requirement for a large number of algae-ciliate contacts to induce susceptibility in the ciliate. Splitting the exposure of ciliates to algae into 2 periods of 0.5 h, separated by 5 h in the absence of algae, produced a much higher frequency of infection than a single l-h exposure, supporting the suggestion that the large number of algae is required to induce susceptibility in the ciliate which can then be infected by as few as a single algal cell.

    Notes: A plaque technic was evaluated and used for the isolation and enumeration of small, free-living amebae in lake-bottom samples which were collected each month for one year from a Nebraska lake. Several culture media were evaluated, and a simple glucose-salts medium was chosen. The most frequent ameba in the lake-bottom samples was Acanthamoeba polyhaga which underwent marked increases and decreases in population densities during the collection period. This pattern was not correlated with water temperature, bacterial counts, and nitrate or phosphate levels. Other species of amebae of the genera Hartmannella, Vahlkampfia, Naegleria, Paratetramitus and Echinamoeba were isolated. Most of these were either found infrequently or remained at relatively low, constant densities throughout the year. In addition, 4 species of Acrasieae of the genera Dictyostelium and Polysphondylium were isolated from the lake-bottom samples.

    Notes: SYNOPSIS Hydroxyurea (HU) inhibits increase in cell number in cultures of Crithidia fasciculata. Complete inhibition is produced by 8 mM and higher concentrations. If HU is not removed, population growth resumes in 45–50 h: if HU is removed, partially synchronous growth occurs through 2 cycles. During HU inhibition, the rate of DNA synthesis is reduced to 1% of that in exponentially growing cultures; protein and RNA syntheses continue at slightly reduced rates. Mean cell size and protein and RNA contents per cell increase; rate of oxygen consumption per mg cell protein remains constant. The behavior of a culture upon addition of HU and upon its removal agrees with predictions based on the hypothesis that the only direct effect of HU is to block DNA synthesis. The synchrony produced by HU is judged satisfactory for investigations of kinetoplast and nuclear replication but not for biochemical characterization of other aspects of the cell cycle.

    Notes: SYNOPSIS Dihydrofolate reductase (E.C. 1.5.1.3) from Plasmodium falciparum and from its host, the owl monkey (Aotus trivirgatus). were partially purified and characterized. The molecular weight of the parasite enzyme was estimated to be over 10 times as high as that of the host enzyme. The host enzyme had 2 pH optima whereas the parasite enzyme only one. The activity of the host enzyme was greatly stimulated by KCI and urea, while that of the parasite enzyme was inhibited at high concentrations of such chaotropic agents. Km of the parasite enzyme was significantly higher than that of the host enzyme. The parasite enzyme had much lower Ki for pyrimethamine than the host enzyme. Dihydrofolate reductases isolated from pyrimethamine-resistant and pyrimethaminesensitive strains of P. falciparum were found to be similar.

The main kinetic characteristics of these enzymes and their quantitative assay methods were studied.No activity was detected for α-fucosidase, β-xylosidase, β-glucuronidase, elaidate esterase. acid lipase, and alkaline phospho-diesterase.

In the buccal cortex, the paroral kinety-ribbed wall complex, the 3 polykineties, and the cytostome-cytopharynx were investigated.Comparative survey of ciliate ultrastructure indicates 2 principal orientation patterns for kinetodesmal and postciliary fibers, recognition of which leads to reevaluation of the theory of paroral kinety formation and the ideas of homology based on this theory. Ultrastructurally, the scuticociliates are not distinct from tetrahymenines and peniculines; the 3 groups appear to be 1 assemblage.

    Notes: SYNOPSIS. In this study the characteristics of the synthesis of DNA and RNA in the nuclei of Loxodes were investigated. Loxodes striatus is a primitive ciliate with 2 pairs of structurally differentiated diploid nuclei, the macro- and micronuclei. The macronuclei are differentiated morphologically into a clearly recognizable central core and an outer zone.To determine DNA and RNA synthesis, individual organisms were analyzed by autoradiography after incubating groups of cells with a 3H-labeled precursor ([3H]thymidine for DNA and [3H]uridine for RNA).The following observations were made: (A) All portions of macro- and micronuclei appeared to contain DNA as judged by the localizations of incorporated [3H]thymidine. (B) The macro- and micronuclei did not synthesize DNA at the same time; moreover, the duration of DNA synthesis in the former was much longer than of the latter nucleus. (C) Replication of DNA in the inner core and outer zone of the macronucleus occurred at separate times with little if any overlap. (D) All of the detectable [3H]uridine incorporation was found in the macronucleus and none in the micronucleus. Within the macro-nucleus the central core was more heavily labeled. (E) The quantitative differences in the label of the different components of the nuclear complex were investigated. (F) Contrary to the previously reported information our results suggest that DNA synthesis can occur in adult macronuclei.The possible explanation of these results is discussed in the context of the nuclear evolution of ciliates and of recent information on nuclear differentiation.

    Notes: SYNOPSIS. Eimeria dispersa (turkey strain) and Eimeria gallopavonis sporozoites were inoculated into primary cultures of chicken kidney (CK) and turkey kidney (TK) cells. Eimeria dispersa sporozoites were more infective in either cell type than those of E. gallopavonis: at 4 hr, the percentage of infection was 67-98 for E. dispersa but only 23-56 for E. gallopavonis. E. dispersa also survived better in culture: at 2 days, losses of E. dispersa in both cell types were only 4-19%, whereas losses of E. gallopavonis were 35-47% in TK cells and 60–95% in CK cells. However, E. gallopavonis developed further than E. dispersa. Location and increase in numbers of intracellular stages at 4 days indicated that E. dispersa proceeded through 2 schizogonic generations before development stopped.

A mating type analysis was performed on 231 isolates of the cellular slime mold, Polysphondylium pallidum found in 61 samples collected in eastern North America between northern Florida and southern Canada. Seventy-eight percent of the isolates belonged to one of 2 mating types; 18% were incapable of mating with any partner; 3% were homothallic; and 1%, consisting of 2 isolates from a Florida sample, belonged to a separate breeding group. It is suggested that the majority of isolates represent a species capable of local genetic adaptation to a niche, the parameters of which undergo considerable variation over space and time.

Cell size, macromolecular composition, carbohydrate utilization patterns, and O2 concentrations were measured throughout the growth stages of Naegleria gruberi in agitated cultures in a complex medium. Biphasic logarithmic growth occurred during the initial 83 hr of growth and the mean generation time was 7.0 hr and 19 hr during initial and secondary log growth stages, respectively. The maximum yield was 5 × 10* amebaeJml. The pH rose rapidly (1 pH unit) during the secondary log growth phase (52-83 hr) and continued into the stationary growth phase (83-120 hr). Dry weight, total protein, carbohydrate, and RNA per ameba increased just before the secondary log growth phase. RNA increased 31% to 35% per ameba at the end of each phase of log growth. DNA increased ∼ 2-fold throughout the different growth phases. Average cell size increased 90% during biphasic log growth then decreased during stationary phase. O2 tension decreased from 100% to 18% of saturation during the biphasic growth phase, then increased during stationary growth to near 100% saturation. Glucose and total carbohydrate assays showed little utilization of those substrates throughout the growth stages. Naegleria gruberi presumably has a predominantly aerobic metabolism, also its metabolism may change during the different growth phases.

Culture conditions were established to optimize replenishment synchrony. The DNA content of E. gracilis in steady state culture and vitamin B12 deficiency culture was measured by flow cytofluorometry and was consistent with colorimetric determinations. The cell volume and DNA distributions of E. gracilis in synchronous culture were analyzed and the sequential changes during the division cycle were computed. Synchronous culture permits more definitive studies of shifts in cell volume and DNA distributions, in which the biochemical events required for cell division are presumably synchronized.

    Notes: Atypical eukaryotic RNA polymerase activity was demonstrated in nuclei of Crypthecodinium cohnii, a eukaryote devoid of histones. Nuclei were isolated from growing cultures of this dinoflagellate and assayed for endogenous RNA polymerase (EC 2.7.7.6) activity. There was a biphasic response to Mg2+ with optima at ˜ 0.01 and 0.02 M MgCl2, but in contrast to other eukaryotic RNA polymerases, this enzyme activity was inhibited by low MnCl2 concentrations. In the presence of 0.01 M MgCl2 the optimum (NH4)2SO4 concentration was 0.025 M, a concentration at which the nuclei were lysed. Incorporation of [3H]UMP into RNA was inhibited by actinomycin D and dependent on the presence of undegraded DNA, and the reaction product was sensitive to ribonuclease and KOH digestion. Omission of one or more ribonucleoside triphosphates greatly reduced the incorporation. Only a slight enhancement of RNA polymerase activity resulted from the addition of various amounts of native and denatured calf thymus DNA. Spermine caused a marked inhibition while spermidine had little effect on RNA synthesis in the nuclei. Under the optimum conditions described in the present paper the nuclei incorporated ˜ 3 pmoles of [3H]UMP/muml; DNA at 25 C for 15 min, and ˜ 80% of this activity was inhibited by the eukaryotic RNA polymerase II inhibitor, α-amanitin (20 m̈/ml). A unique situation therefore exists in C. cohnii nuclei, in which absence of histones (a prokaryotic trait) is combined with α-amanitin-sensitive RNA polymerase activity (a eukaryotic trait).

    Notes: After demonstration that emetine is amebicidal by inhibiting protein synthesis, the question arose whether active protein synthesis is required for emetine's amebicidal effect. The answer appears to be “no,” as derived from experiments on intact amebae. Responses were compared for log- and stationary-growth phase amebae. In the latter, protein synthesis is significantly slower, and sensitivity to emetine, i.e. degree of inhibition of protein synthesis, was maintained independently of rate of protein synthesis. Both stages equally bound tritiated emetine to their ribcsomes. Binding of [3H]emetine was not affected by certain drugs that interfere with energy metabolism, protein synthesis, and/or ribosomal function, e.g. dinitrophenol, puromycin, chloroquine, and acriflavin. High concentrations of EDTA combined with puromycin (which disaggregates ribosomes into their subunits) lowered binding by 50%.In chase experiments the ribosomes of intact amebae were prelabeled with [3H]emetine or [3H]isoemetine, then exposed to relatively high concentrations of unlabeled emetine. Labeled isoemetine was displaced almost completely, whereas no displacement of [3H]emetine occurred; evidently, the high stability of the emetine-ribosome binding is due in part to a hydrogen-bonding reaction of the C-1' atom of the emetine molecule with the chain-elongation site. Finally, evidence was obtained that capacity to bind emetine is an index of drug resistance.

L' étude ultrastructurale de la grégarine Gregarina blaberae a été entreprise après mise au point d'un protocole d'infestation expérimentale de larves vierges de Blattes, Blaberus craniifer. Les observations en microscopie électronique, effectuées en fonction du temps, ont permis de suivre le développement du sporozoïte, ainsi que la croissance et l' évolution du trophozoïte.La croissance est spectaculaire puisqu'en 18 jours elle entraîne la transformation du germe infectieux ou sporozoïte (L = 15 μm, φ=1 μm) en un céphalin—trophozoïte fixéà l' épithélium intestinal—de 250 μm de longueur pour un diamètre de 65 μm. L'organisation ultrastructurale du sporozoïte ne diffère pas de celle de la plupart des sporozoïtes des autres sporozoaires étudiés jusqu' à présent, le conoïde et les corps denses sont présents. La paroi est trimembranaire. toutefois le complexe membranaire interne présente quelques interruptions. Les premiers dictyosomes s' élaborent à partir de l'enveloppe nucléaire. La migration du noyau et des corps denses, puis la régression de toutes les formations caractéristiques du sporozoïte et l' établissement d'une zone corticale venant coiffer l' épimérite, ont lieu dans les 48 heures suivant l'infestation et marquent la transformation sporozoïte-céphalin.L' évolution du céphalin, se poursuit par la formation des ébauches des plis épicytaires qui s'effectue à partir du 3éme jour du développement à la base du deutomérite. Un système régulier de plis longitudinaux ou épicyte s' établit ainsi sur toute la partie extra-cellulaire de la grégarine. Dès les 4éme-5éme jours de développement, un réseau vacuolaire et un chondriome important se différencient dans l' épimérite, tandis qu'un septum de nature fibrillaire isole le protomérite du deutomérite. Le stade suivant—à partir du 6éme jour—est marqué par une ségrégation du matériel glucidique au niveau de ces 2 segments.Le modèle étudié permet de préciser le rôle de l' épimérite dansla nutrition du parasite, l' évolution du chondriome et des membranes corticales au cours de la phase de croissance végétative des grégarines polycystidées.

    Notes: SYNOPSIS. Stomatogenesis was studied in the heterotrich ciliate Blepharisma japonicum stained with protargol. During binary fission not only is a new oral apparatus made for the posterior daughter, but the already existing oral apparatus of the parent cell is reorganized, i.e. partially disassembled and then subsequently reassembled to provide a functional feeding apparatus for the anterior daughter cell. These morphogenetic events, requiring 21/2 to 3 hr, are complete by the time the anterior and posterior daughters separate.In preparation for division, an oral anlage is formed by the rapid proliferation of kinetosomes along 4–5 stomatogenic kinetics directly subtending the cytostome. This field of randomly oriented kinetosomes ultimately gives rise to the feeding apparatus of the posterior daughter cell. Early in division, the oral anlage separates into 2 longitudinal fields of kinetosomes: one is destined to give rise to the undulating membrane and the other forms the adoral zone of membranelles. Shortly after the anlage is established posterior to the cytostome, reorganization of the existing functional mouth is initiated. The morphologic changes associated with this dedifferentiation-redifferentiation sequence lead to the formation of an oral apparatus for the anterior daughter and cannot be distinguished from those characteristically seen during physiologic reorganization.

    Notes: SYNOPSIS. The effects of age and sex of the cat on oocyst shedding, multiplication of Toxoplasma gondii in tissues of cats, and acquisition of immunity were investigated after oral inoculation of cats with Toxoplasma cysts. Twenty-five cats varying in age from 1 week to 39 months were killed 7-97 days after inoculation with T. gondii. Homogenates of brain, heart, mesenteric lymph nodes, retina, and blood from these cats were inoculated into mice to test for Toxoplasma infectivity. Toxoplasma was isolated more frequently and in higher titers in mice receiving inocula from cats of the youngest age group (1 week old). Toxoplasma gondii was isolated from tissues of only 2 of 21 cats older than 2 months (at the time of inoculation), although all of the animals shed oocysts within 1 week after ingesting the parasites.The number of oocysts shed varied among littermates of the same sex and between sexes. Generally, cats younger than 12 months shed more oocysts than older cats. The number of oocysts shed by older cats varied considerably; males generally shed more oocysts than the females. However, the numbers of cats examined were too small for statistical comparison. Nevertheless, the observations suggest that cats older than 12 months should not be used in experiments where numbers of oocysts shed is critical.

    Notes: SYNOPSIS. Changes in the metabolism of Crithidia fasciculata ATCC 11745 when grown in the presence of ethidium bromide were studied. Ethidium bromide-grown cells had decreased respiratory activity as measured by oxygen consumption. More than 50% of the organisms cultivated in a defined medium containing 1.0 mg/liter of ethidium bromide became dyskine-toplastic and had decreased activities of particulate succinate and NADH-linked dehydrogenases as well as of soluble isocitrate dehydrogenase. These cells also had increased activities of particulate α-glycerophosphate dehydrogenase, soluble α-glycerophosphate dehydrogenase, malic enzyme, hexokinase, and malate dehydrogenase. Ethidium bromide-grown cells had a lower level of ATP and contained less DNA than cells grown in its absence.
