    Notizen: Summary To assess the role of the hypothalamo-hypophysial complex in the control of photoperiodically induced vernal premigratory responses in the White-crowned Sparrow, the effects of hypothalamic lesions and systemic administration of several hormones on these responses were investigated. Lesions that destroyed the posterior median eminence (PME) or the entire median eminence (ME) inhibited photoperiodically induced testicular growth, premigratory fattening and Zugunruhe. Lesions that destroyed the anterior median eminence (AME) alone had no appreciable effects on testicular growth, fattening or Zugunruhe. Lesions in the basal infundibular nucleus (IN) that resulted in complete inhibition of testicular growth abolished Zugunruhe, but allowed varying degrees of fattening. The systemic administration of prolactin, testosterone propionate (TP) or the combination thereof in the PME-lesioned birds induced fattening similar to that observed in photostimulated controls but did not induce Zugunruhe. It is concluded that testosterone and prolactin are the most important hormones involved in the control of vernal premigratory fattening. The role of these hormones in the induction of vernal Zugunruhe is not positively proven. The author is grateful to Professor Donald S.     Notizen: Summary The supraependymal cells (SEC) are a normal component of the wall of the cerebral ventricles. In the hypothalamic area of the third ventricle they are restricted, in healthy animals, to the ependymal projection of the hypophyseotropic area. Here the SEC show great polymorphism. In addition to bipolar, multipolar and stellate or spider-like cells, transitional forms between these types can be seen. Their perikarya and processes can either remain at some distance from the ependyma or be in close contact with it. The processes may protrude between the ependymal cells or show surface differentiations that resemble the thin cytoplasmic folds of the mesenchymal wandering cells. Considering this and the variations in the number of cells, for example during the ovarian cycle, the SEC can be interpreted as mesenchymal cells, probably related to microglial cells of the subependymal layer. It is suggested that the SEC have a phagocytotic function and may be involved in the normal renewal of the ependyma. A definitive explanation for the restriction of the SEC to the hypophyseotropic area as well as the elucidation of their function remain to be found.

    Notizen: Summary Within the suprachiasmatic nucleus (SCN) of the rat the fine structure of the synapses and some features of their topological arrangement were studied. Five types of synapses could be distinguished with certainty: A. Two types of Gray-type-I (GTI) or asymmetrical synapses (∼33%). The presynaptic elements contain strikingly different types of mitochondria. Size of clear vesicles: ∼ 450 Å. Synapses with subjunctional bodies often occur, among these also “crest synapses”. Localization: dendritic shafts and spines, rarely somata. B. Three types of Gray-type-2 (GTII) or symmetrical synapses (∼66%)∶:1) Axo-dendritic and -somatic (=AD) synapses. Size of clear vesicles: ∼500 Å. 2) Invaginated axo-dendritic and -somatic (=IAD) synapses with club-like postsynaptic protrusions within the presynaptic elements (PreEl). Size of clear vesicles is very variable: ∼ 400–1,000 Å. 3) Dendro-dendritic, -somatic and somato-dendritic (=DD) synapses occurring at least partly in reciprocal arrangements. They represent an intrinsic system. Shape of clear vesicles: often oval; sucrose treatment partly produces flattening. Dense core-vesicles (dcv) are found in all GTII- and most of the GTI-synapses after three-dimensional reconstruction. All types of synapses (mostly GTII-synapses) can be enclosed by multilamellar astroglial formations. The synapses often occur in complex synaptic arrangements. Dendrites and somata of females show significantly more multivesiculated bodies than those of males. Further pecularities of presynaptic (PreEls) and postsynaptic elements (PostEls) within the SCN are described and discussed.

    Notizen: Summary The ependyma lining the lateral recess of the third ventricle of the teleost inferior lobe has been studied by light and electron microscopy, including Golgi impregnation methods. As many as five different cell types appear to line the ventricle, but some of these may be similar cells in different stages of activity. One cell type contains small dense-cored vesicles and appears to have processes extending into deeper portions of the lobe. Golgi preparations reveal subependymal cells with apical processes extending to the ventricle and basal extensions which may reach the pial surface. The present observations are discussed in relation to similar studies in other fishes, amphibians and mammals. Possible functions for the various cells observed are suggested.

    Notizen: Summary To investigate whether the hypothalamus is involved in the cytodifferentiation of the anterior pituitary gland, rat foetuses were encephalectomized in utero on day 16 of pregnancy. Pituitary sections from encephalectomized and normal littermate foetuses were studied on day 21 with the immunofluorescence technique using antibodies and α-MSH, anti β-MSH, anti α-(17–39) ACTH and anti β-(1–24) ACTH. On day 16, only the anti β-MSH revealed a few cells in the pars distalis but not in the pars intermedia. On the other hand, on day 21, the pituitary cells reacting with antibodies anti α-MSH, anti β-MSH and anti α-(17–39) ACTH were as numerous in the encephalectomized foetuses as in the normal littermate foetuses. The cells revealed with the antibody anti β-(1–24) ACTH were less numerous and less fluorescent in the pars distalis and intermedia of the hypophysis of the encephalectomized foetuses. On day 21, the adrenals of the encephalectomized foetuses were atrophied in comparison with those of the normal littermate foetuses but they were larger than on day 16. These data suggest that the cytodifferentiation of the corticotroph and melanotroph cells of the hypophysis occurs without the influence of the hypothalamus which is necessary for the normal release of ACTH.

    Notizen: Summary A continuous cell line was previously obtained by Simian Virus (SV) 40 transformation of primary cultures of dissociated mouse fetal hypothalami. One clone from this cell line has been previously shown to possess some of the ultrastructural features, immunological properties and synthesizing capacities of magnocellular hypothalamic neurons which secrete vasopressin and neurophysins. The present paper reports on the morphological characterization of 14 other clones or subclones of the original cell line, using the following criteria: phase contrast microscopy, electron microscopy, Gomori's aldehyde fuchsin staining, cytochemical detection of β-glucuronidase, immunochemical staining with antisera against bovine neurophysin I, bovine neurophysin II, lys-vasopressin, oxytocin, LH-RH and TRH. The results allowed the conclusion that the clones as well as the subclones can be distributed into two groups: 1) neurosecretory neurons which all possess several of the ultrastructural and cytochemical features of the neurophysin-vasopressin synthesizing clone, and 2) primitive nerve cells which are devoid of such features but display numerous bundles of filaments. In addition some clones were found to display intermediate features between groups 1 and 2. A similar diversity was observed within clones of the original strain and subclones of a neurosecretory clone. It is suggested that the primitive clones could represent precursors of the neurosecretory clones.

    Notizen: Summary Immuno-enzyme cytochemical investigations have shown that, (1) the hypothalamic supraoptic and paraventricular nuclei of the Brattleboro rat, as in the normal rat, contain separate neurons which produce oxytocin + neurophysin; (2) the hereditary inability of the Brattleboro rat to synthesize vasopressin and its associated neurophysin is due to a biochemical defect of separate “neurophysin-vasopressin” neurons in the supraoptic and the paraventricular nuclei. These observations strongly support the hypotheses that (1) vasopressin and its associated neurophysin are formed via a common precursor, and (2) the initial point of intracellular appearance of the hereditary defect in the Brattleboro rat lies in the synthesis of this precursor, which occurs on ribosomes. Moreover, observations have demonstrated that, in the Brattleboro rat, in addition to the hereditary inability of the hypothalamic magnocellular neurosecretory system to synthesize vasopressin, there also exists a similar hereditary defect in the hypothetical parvicellular suprachiasmatic-median eminence neurosecretory system.

    Notizen: Summary The distribution of radioactivity in the rat hypothalamus after intraventricular injection of tritiated luteinizing hormone-releasing hormone (LH-RH) was studied by autoradiography and compared with that expected from Fick's second law which defines the character of ordinary diffusion. The results suggest that LH-RH penetrates through the ependyma and thin subependymal layer in the median eminence and into the suprachiasmatic area by ordinary diffusion. By contrast, concentration of radioactivity in outer layers of the median eminence is increased and, therefore, can not be explained by the diffusion law. In this region incorporation of radioactivity into cell bodies was demonstrated. In the subependymal layer of the median eminence a few rows of silver grains seemingly located in cellular processes were also observed. The diffusion coefficient in the periventricular zone was lower than that in the deeper tissue. It is suggested that the ependymal-subependymal complex of the third and lateral ventricles acts as a passive membrane towards LH-RH contained in the cerebrospinal fluid. The significance of this mechanism for the intrahypothalamic transport of RHs is discussed.

    Notizen: Summary The hypothalamic arcuate nucleus (ARCN) of female rats at 5, 20, 45 and 90 days of age was examined ultrastructurally. Axodendritic and axosomatic synapses were counted in 18,000 μm2 area of the ARCN in each brain. Axodendritic and axosomatic synapses in the ARCN of day 5 rats were very small in number. Axon terminals contained small spherical vesicles (SSVs, 40–60 nm in diameter). Occasionally large granular vesicles (LGVs, 75–130 nm in diameter) were found to coexist with SSVs in the endings. Pre- and postsynaptic membranes were thin. The ARCN at this age exhibited a large extracellular space which decreased with advancing age. In day 20 rats, axodendritic and axosomatic synapses increased in number up to about one-half of those of day 45 or day 90 animals. Synaptic vesicles increased in number and mitochondria were frequently encountered in the axon terminals. Pre- and postsynaptic membranes became thicker than those of day 5 rats. Further increase in the number of axodendritic and axosomatic synapses in the ARCN of day 45 rats was observed, and there were no significant difference in the morphology and incidence of synapses between day 45 and day 90 rats. Synaptic vesicles were numerous and pre- and postsynaptic membranes were thick. In tissue incubated with 5-hydroxydopamine (5-OH-DA) before fixation, small granular vesicles (SGVs, about 50 nm in diameter) which were labeled with 5-OH-DA were detected in a certain number of endings in all material taken from each age group, but the incidence of synapses containing SGVs was usually low. From these results, it can be proposed that an increase in the number of synapses in the ARCN is correlated with functional maturation of the ARC neurons.

    Notizen: Summary Vasopressin was immunohistochemically localized at the electron microscopic (EM) level in the hypothalamic-neurohypophysial system (HNS) of three murids. Antiserum to vasopressin was produced in rabbits injected with lysine vasopressin (LVP) conjugated to egg albumin (EA), anti-EA being precipitated prior to staining. Sternberger's unlabeled antibody peroxidase technique was employed, immunoreactivity being designated by peroxidase anti-peroxidase (PAP) molecules and electron opacity. Immunoreactive neurosecretory granules (NSG) were found in the perikarya of the supraoptic nucleus (SON) in all three murids investigated, although far more profusely in the two wild strains. Immunoreactive axonal NSG were observed in the inner and outer zones of the median eminence (ME), and within most of the axons and terminals in the neurohypophysis. The concentration of primary serum effective for staining the SON (1∶10–1∶50) was far higher than that required for the ME and the neurohypophysis (1:500–1:1,200). AntiLVP also induced electron opacity of granules in cells of the pars intermedia (PI). Discussion centers on the significance of immunoreactive NSG in the neurosecretory (NS) perikarya, on the possibility of an extragranular pool of hormone, and on speculation about the electron opacity of the PI granules.
