    Description / Table of Contents: Résumé La largeur de la métaphyse tibiale, la zone indifférenciée, la zone sériée et les cellules en dégénerescence ont été observées chez des rats Sprague-Dawley normaux et hypophysectomisés. La production cellulaire de la métaphyse est déterminée sur la base de la croissance osseuse longitudinale déterminée par l'oxytétracycline et la taille des cellules en dégénérescence. La diminution de la croissance osseuse longitudinale, en fonction de l'augmentation de l'âge et après hypophysectomie, est due partiellement, à la diminution de production cellulaire et partiellement à une décroissance de la taille des cellules en dégénérescence dans la métaphyse. L'influence de la production cellulaire et de l'activité mitotique prédomine.
    Abstract: Zusammenfassung Die Breite der proximalen Wachstumsplatte der Tibia, deren undifferenzierter und säulenförmiger Zone und die Größe der nahe bei der Metaphyse auftretenden degenerativen Zellen wurden in normalen und hypophysektomierten Sprague-Dawley-Ratten bestimmt. Die Zellproduktion in der Wachstumsplatte wurde aus dem longitudinalen Knochenwachstum berechnet, welches mittels Oxytetracyclin und der Größe der degenerativen Zellen bestimmt wurde. Es wurde festgestellt, daß die Abnahme des longitudinalen Knochenwachstums bei zunehmendem Alter und nach Hypophysektomie zum Teil einem Rückgang in der Zellproduktion, zum Teil einer Verminderung der Größe der degenerativen Zellen in der Wachstumsplatte zuzuschreiben ist. Der Einfluß der Zellproduktion, und somit der mitotischen Aktivität, herrscht vor.
    Notes: Abstract The width of the proximal growth plate of the tibia, its undifferentiated and columnar zone and the size of the degenerative cell close to the metaphysis, were determined in normal and hypophysectomized Sprague-Dawley rats. The cell production in the growth plate was calculated from the longitudinal bone growth determined with oxytetracycline and the degenerative cell size. It was found that the decrease in longitudinal bone growth with increasing age and after hypophysectomy, is due partly to a decrease in cell production, and partly to a decrease in degenerative cell size in the growth plate. The influence of cell production and thus the mitotic activity predominates.

    Notes: Summary Rats were injected with45Ca and horseradish peroxidase to determine the patterns of45Ca incorporation into incisor enamel and the morphological types of the overlying maturation ameloblasts.45Ca autoradiography showed no differences in the patterns of incorporation into enamel between routinely embedded and freeze-dried specimens. Enamel overlaid by ruffle-ended ameloblasts was much more heavily labeled while that overlaid by smooth-ended ameloblasts showed only moderate labeling. The observations lend further support to the hypothesis that the ruffle-ended cells are very active in mineralizing enamel and that the smooth-ended cells are in a passive, restorative phase.

    Notes: Summary Preliminary indications of the occurrence of actin and myosin in crude matrix vesicle preparations have been reported previously. In the present study extracellular matrix vesicles from rat alveolar bone were isolated. They were further purified by a sucrose density gradient. SDS-polyacrylamide gel electrophoresis of the purified vesicles revealed the presence of a polypeptide with a molecular weight of 43 K daltons and with electrophoretic mobility identical to that of blood platelet actin. The limited proteolysis of both 43 K dalton vesicular polypeptide and actin byStaphylococcus aureus-V8-protease revealed three fragments with identical electrophoretic mobility. In addition, the vesicular preparations inhibited the activity of DNase I, a property typical of actin monomers. Filamentous material extracted from matrix vesicles showed ultrastructural features of F-actin. Reaction of this material with heavy meromyosin resulted in arrowhead formation, which is characteristic of acto-heavy meromyosin. The occurrence of actin in extracellular matrix vesicles may account for their budding from the osteoblastic plasma membrane, their possible motility in the matrix, and maintenance of the spherical shape.

Die Gefäßfortsätze von ependymalen und subependymalen Zellen bilden in der subependymalen Zone und in der Zona interna des Ratteninfundibulum mit Nervenfasern kompakte neuro-gliöse Faserbündel, die sich in der Zona externa aufzweigen. 2. In allen Zonen des Infundibulum kommen zwischen den Nervenfasern und den Gefäßfortsätzen zahlreiche neuro-gliöse Synapsen vor. In den Gefäßfortsätzen fällt die hohe Zahl an Mikrotubuli sowie die zahlreichen, vielgestaltigen Einschlüsse auf. 3. In der subependymalen Zone sind die Interzellularspalten an bestimmten Stellen außerordentlich weit. Sie haben eine kanalartige Beschaffenheit und enthalten feine Bündel von Nervenfasern. 4. Von den lateralen Anteilen des Infundibulum her erreichen Dendriten von Ganglienzellen des Nucleus infundibularis die Mitte des Infundibulum. In dieser Region sind axodendritische Synapsen anzutreffen. 5. Morphometrische Analysen der Nervenfaserendigungen der Zona externa von Normaltieren zeigen, daß die prozentuale Verteilung der nach Granulagröße differenzierten Nervenfaserklassen für Mitte und Seite der Zona externa etwa gleich ist. Zwischen der Größe der Elementargranula und der Anschnittfläche der zugehörigen Nervenfasern besteht eine direkte Beziehung. 6. Die Nervenfaserendigungen erreichen die Basalmembran des perikapillären Raumes fast ausschließlich im Bereich von gefäßwärts gerichteten Vorwölbungen der Zona externa. Das Ausmaß, in dem Nervenfasern im Vergleich zu den Gefäßfortsätzen von Ependymund Gliazellen den perivaskulären Raum erreichen, ist medial weitaus größer als lateral. 7. Bei bilateral adrenalektomierten Ratten nimmt in bestimmten, vorwiegend lateral gelegenen Nervenfasern die Zahl und Größe der Elementargranula in Abhängigkeit von der Überlebensdauer zu. Dies dürfte auf eine verstärkte Synthese und Speicherung von Corticotropin-Releasing Factor in diesen Nervenfasern zurückzuführen sein. Gegenüber dem Normalbefund ist die neurohämale Kontaktfläche erheblich vergrößert. Der perivaskuläre Raum enthält zerfallene Nervenfaserteile, die durch Bindegewebszellen phagocytiert werden. Diese Veränderungen dürften durch eine unter Versuchsbedingungen verstärkte Wachstumstendenz der Nervenfasern in Richtung auf die Blutgefäße und durch eine Abschnürung der Nerven-faserendigungen ausgelöst werden.
    Notes: Summary 1. In the subependymal and internal zones of the rat median eminence nerve fibres and vascular processes of ependymal and subependymal cells form neuro-glial bundles. They branch in the external zone. 2. In all these three zones of the infundibulum numerous neuro-glial synapses are found between nerve fibres and vascular processes of glial cells. The vascular processes contain a high number of microtubules as well as polymorphous granular inclusions. 3. In certain regions of the subependymal layer the intercellular spaces are enlarged. They form channel-like spaces containing small bundles of delicate nerve fibres. 4. Nerve cells of the infundibular nucleus located in the lateral parts of the infundibulum send dendrites to the medial parts of the infundibulum. In this area axo-dendritic synapses are found. 5. For morphometric analysis, the nerve fibres of the external zone were classified according to the diameter of their granules. It is shown that in the different regions of the external zone the distribution of the various types of nerve fibre is similar. Moreover it can be seen that a direct correlation exists between the size of the sectional plane of a given nerve fibre and the size of the granules it comprises. 6. Nerve fibre endings abutting on the basement membrane of the pericapillary space are mostly found in bulb-like protrusions of the external zone. The extent to which nerve fibres reach the perivascular space—as compared with the vascular processes of ependymal and glial cells—is higher in the medial than in the lateral parts of the infundibulum. 7. In bilaterally adrenalectomized rats the number and diameter of elementary granules increases in nerve fibres located laterally. This increase is directly related to the survival time and may be due to an enhanced synthesis and storage of Corticotropin-Releasing Factor in these nerve fibres. Compared with the findings in untreated animals the neurohemal contact area is significantly enlarged. The perivascular space contains degenerating nerve fibres which are undergoing phagocytosis by connective tissue cells. It is assumed that these alterations are due to the increased growth of nerve fibres towards the vessels of the “Mantelplexus”, and that, following adrenalectomy, this excessive growth leads to a pinching off of nerve fibres.

    Notes: Summary The adrenergic innervation of the extrarenal blood vessels of the rat left kidney was investigated by fluorescence histochemistry and by electron microscopy. The trunk of the renal artery proximal to the aorta is elastic and appears to be very sparsely innervated. In contrast, near the kidney the renal artery—which divides into 3 to 4 large branches of the muscular type possesses a dense adrenergic innervation. The adrenergic terminal axons are situated in the adventitia close to the external elastic lamella, but only rarely in close contact with smooth muscle cells. In most instances several terminal axons are grouped and enclosed by a Schwann cell, single axons being rare. All terminal axons are able to take up and to store 5-hydroxydopamine which strongly suggests that they are adrenergic. The innervation of the renal vein is more sparse than that of the muscular arteries but somewhat denser than that of the elastic artery. In addition, close to the origin of the renal artery the presence of “small intensively fluorescent” (SIF) cells as well as of some adrenergic ganglion cells is noted. The latter are situated in the adrenergic nonterminal axon bundles, which run parallel to the blood vessels. It is concluded that the uneven adrenergic innervation along the artery as well as individual variations in the branching of the artery are the main causes of the unusually high individual variations of the NA content of this organ such as used in pharmacological experiments.

    Notes: Summary 4-day cyclic adult female Wistar rats were injected subcutaneously with testosterone propionate on diestrus 1 at 16:00 and on diestrus 2 at 10:00 respectively. Non-injected females served as controls. Autopsy was performed on diestrus 2 at 23:00, and on proestrus at 14:00 and 17:00 respectively. The blue Alcian-PAS staining was used to evidence FSH (β) and LH (γ) pituitary cells. In control animals and in diestrus 2 injected females only a small number of FSH cells could be detected on diestrus 2 at 23:00. This number increased markedly on proestrus at 14:00 and decreased on proestrus at 17:00. A similar evolution was observed in diestrus 1 testosterone injected females, but the number of FSH cells appeared higher at any stage of autopsy in these females than in diestrus 2 injected females and in control rats. In control females, numerous LH cells were observed on diestrus 2 at 23:00. The number of these cells was diminished on proestrus at 14:00 and still more at 17:00. On the contrary few LH cells were detected in testosterone injected females on the evening of diestrus 2. An increase of these cells occurred on proestrus at 14:00, followed at 17:00 by only a weak diminution as established by comparison with control animals. An inhibition of FSH release and a suppression of the proestrus surge of LH were therefore supposed to cause, on one hand, the slowing up of follicular growth observed in diestrus 1 injected females and, on the other hand, the blockage of ovulation noted in both diestrus 1 and diestrus 2 treated animals.

    Notes: Summary Neural lobes of adult rats have been explanted and organ-cultured for 5–10 days. While perivascular cells and neurosecretory fibers undergo a progressive degeneration, scattered pituicyte-like cells are observed mostly associated with each other in number of two or three cells for group. Cultured pituicytes are quite similar to in vivo pituicytes except for some particular features as the shape of the nucleus and the number of dense bodies. Furthermore they share with the in vivo pituicytes the phagocytic capacity which appears even increased as far as neurosecretory fibers are concerned. Finally, degenerating pituicytes and free lipid droplets are observed in the intercellular spaces: the significance of these facts is discussed and tentatively interpreted. The organotypic culture of the neural lobe seems to represent an experimental model useful to obtain a relatively pure population of pituicytes.

    Notes: Summary Two new types of structure of the cytoplasm of an autonomic neuron were described: (1) Inclusions of the rough ergastoplasmic reticulum, which were named “ergastosomes”, contain finely granular material with moderately electron opaque staining with both glutaraldehyde-OsO4 and KMnO4 fixations. A short comment was made on the nature of the contents of the ergastosomes. (2) Tubular formations of endoplasmic reticulum were found in the axons and/or dendrites of the paracervical ganglion.

    Notes: Summary To see whether adenohypophysial tissue has the capacity of self-differentiation, Rathke's pouch together with part of the neurohypophysial primordium was isolated from 12-(Group A), 14- (Group B) and 15-day-old (Group C) rat fetuses and cultivated for 9, 6 and 5 days, respectively. Electron microscopic examination of these explants showed many adenohypophysial cells containing electron dense granules, indicative of cytodifferentiation in all groups. Three cell types were distinguishable on the basis of the size of their granules. Variable amounts of neurohypophysial tissue were seen in some explants of Groups B and C, but not A. This “pars nervosa” contained pituicytes with lipid-like inclusions in their cytoplasm but no neurosecretory axons. The adenohypophysial cells abutting on the pars nervosa were arranged rather regularly like cells of the pars intermedia which contained a few granules of about 200 mμ diameter. From these data it is concluded that in rats granule formation in the adenohypophysial primordial cells starts in the absence of hypothalamic neurosecretory substances. It is uncertain from this study whether these cells actually contain hypophysial hormones.

    Notes: Summary The duct system of the rat exorbital lacrimal gland consists of intercalated ducts, interlobular ducts and excretory ducts. The morphological changes from one type of duct to the next are gradual. At the light microscopical level this consists of a change from a bilaminar epithelium in the intercalated ducts to an epithelium, consisting of approximately three layers — which may be pseudostratified — in the excretory ducts. The basal layer of the intercalated ducts consists of myoepithelial cells, whereas the inner epithelial cells may have both a secretory and an electrolyte transporting function. The interlobular duct epithelium contains many cells with deep infoldings of the basolateral plasma membranes and associated mitochondria, suggesting a similar function to the striated duct epithelium in salivary glands. Numerous basal cells in this epithelium have tentatively been interpreted as unusual myoepithelial cells. Nerve terminals have been observed in the ductal epithelium.

    Notes: Summary In order to characterize different sub-zones in the palisade zone of the rat median eminence, quantitative ultrastructural parameters were applied to brains fixed in aldehyde-osmium tetroxide. The palisade zone has been subdivided in 4 successive sub-zones. Increasing numbers of granular vesicles (GV), especially those smaller than 110 nm, are observed from dorsal to ventral sub-zones. There also are more GV per unit area of nervous tissue in the perivascular than in the more dorsal sub-zones. The individual nerve profiles exhibit a larger size in the perivascular layer than in the more dorsal areas, whereas the number and size of nerve profiles devoid of vesicles diminish from dorsal to ventral. As a consequence more GV occur in the perivascular nerve profiles. In the GV containing nerve profiles the concentration of GV is, however, constant in the different sub-zones. A fluctuating size of the preterminal and terminal parts of the nerve fibres is suggested.

    Notes: Summary Epididymal biopsies from rats that had undergone unilateral or bilateral vasectomies from one to eight months previously were compared with biopsies from their contralateral side or from normal controls to ascertain what ultrastructural changes had occurred. After vasectomy, spermatozoa appeared to dissolve in the lumen of the caput epididymidis and to be absorbed by the principal cells. About 5 weeks after vasectomy, numerous lamellar accumulations became apparent in the supernuclear region. Their resemblance to lysosomes or residual bodies was confirmed by an acid phosphatase reaction. After 10 weeks, similar lamellar and polymorphic accumulations on the contralateral side of animals with unilateral vasectomies indicated that resorption had also increased on the unligated side.

    Notes: Summary The transport of 125I-labeled serum lipoproteins through the aortic endothelium was studied by radioautography. Rat aorta and heart was perfused in vitro with a medium containing human very low density (VLDL), low density (LDL), high density lipoprotein (HDL), delipidated HDL apolipoprotein or rat HDL. In all lipoproteins more than 95% of the radioactivity was TCA precipitable and lipid radioactivity was from 2–4% in HDL, 4–6% in LDL, 7–15% in VLDL. After 18–60 min of perfusion and wash with unlabeled medium, most of the aortic radioactivity was TCA precipitable and the percent of lipid counts was similar to that in the original lipoprotein. Following perfusion with VLDL, LDL, or HDL the radioautographic reaction was seen over the endothelium, the subendothelial space and the inner media, and was separated by an unlabeled zone from the reaction present over the adventitia. Uniform labeling of the entire wall was found after perfusion with HDL apolipoprotein. The presence of silver grains over endothelial cells in regions rich in plasmalemmal vesicles suggested that these organelles participate in the transport of the labeled lipoprotein, as was shown for lactoperoxidase (Stein and Stein, 1972). The present data indicate that cholesterol may enter the aortic wall as a constituent of lipoprotein particles. Since an HDL particle carries less than 1/20 of the cholesterol present in a LDL particle, it seems that the lower susceptibility of the female to atheromatosis might be related to the higher ratio of HDL to LDL particles in the female serum.

    Notes: Summary The response of somatotrophs to large doses of purified bovine growth hormone was examined in uninephrectomized rats drinking 1% sodium chloride in an attempt to prolong the action of the hormone. Growth hormone produced a triphasic response in the ultrastructure of somatotrophs. Uninephrectomy and salt alone induced no changes in somatotrophs as compared to unoperated animals consuming tap water. In the first phase (3 days) the cross sectional area of the cell increased, the size of the largest granule per cell as well as the number of granules per cell increased significantly. All these parameters then reached control level after 7 days of injections during a second phase. Suppression of somatotrophs by exogenous growth hormone was evident by 16 days (third phase) when the area of the cell, the size of the largest secretory granule as well as the number of granules per cell all decreased significantly. Somatotrophs apparently did not react to suppression by a lysosomal mechanism inasmuch as no increase in the number of lysosome-derived structures was seen even at 16 days.

Karyometrische Studien am Nucleus suprachiasmaticus männlicher Wistar-Ratten zeigen 6 Tage nach beidseitiger Blendung eine signifikante Abnahme der Zellkerndurchmesser. Dieses Ergebnis spricht für eine Aktivitätsänderung der Neurone im Nucleus suprachiasmaticus geblendeter Ratten.
    Notes: Summary Intraocular injection of labeled amino acids results, in different mammals, in an accumulation of radioactive material not only in the primary optic centers but also within the hypothalamic suprachiasmatic nucleus (Moore et al., 1972; Hendrickson et al., 1972; Moore, 1973). This finding argues in favour of a direct connection between the retina and the secretory suprachiasmatic nucleus although silver techniques do not show such a pathway. In male Wistar rats the nuclear diameter of the suprachiasmatic neurons decreases significantly 6 days after bilateral experimental ablation of the retina. These results indicate an alteration in activity of the suprachiasmatic neurons in blinded rats.

    Notes: Summary The mitoses of the adenohypophysis in male rats have been observed comparatively after fixation by perfusion and fixation by immersion. After fixation by perfusion, the topographic distribution of mitoses throughout the parenchyma is homogenous and the mitotic index is 1.85 for 1 000 cells. After fixation by immersion, the density of the mitoses is decreasing from the periphery towards the center and the mitotic index is only 0.80 for 1 000 cells. The slow penetration of the fixative during fixation by immersion is considered to be the main cause in the differences observed. This slow action causes a modification in the aspect of the mitoses, and more than fifty percent of them cannot be identified and escape to the counts.

    Notes: Summary The ventral, lateral and dorsal lobes of the rat prostate were studied 2, 3, 5, 7 and 21 days after castration. The major changes were a general reduction of the Golgi area and the RER. The cells became pale and contained less organelles compared with the normals. One of the most conspicuous changes were enormous autophagic vacuoles which were present already after 2 days, seemed to reach a numerical peak 3 days after castration and were seldom encountered after 7 days. Increased number of dense bodies were found within the same period. Lipid droplets were observed in the basal cytoplasm of the epithelial cells, especially in the ventral lobe. The basal cells showed the same involution as in the ordinary epithelium. In addition, macrophages were relatively numerous, and between the epithelial cells also lymphocytes were found. The results are discussed in relation to biochemical data on the different lobes.

    Notes: Summary The seminal vesicles and the coagulating gland of the rat were studied 2, 3, 5, 7 and 21 days after castration. The major changes within the seminal vesicles were primarily formation of whorls of the rough endoplasmic reticulum (RER), followed by a general atrophy with a numerical reduction of the RER-profiles, and with general simplification of the cytoplasm due to loss of the organelles. It was a gradually reduction of secretion granules, diminution of the Golgi apparatus, formation of pigment bodies and autophagic vacuoles. Lipid droplets were observed in the basal cytoplasm of the epithelial cells. In the coagulating gland, similar changes occurred within the Golgi area and the lysosome complex. On the other hand, cisternae of the basal endoplasmic reticulum tended to persist in many cells. The similarity in response strongly suggests that the pathogenetic mechanisms are similar in both organs, i.e. atrophy due to deprivation of the androgenic stimulus. The deprivation of androgen gave rise to an inflammatory-like process with infiltration of lymphocytes and macrophages. The increased number of macrophages may indicate that they contribute in some way to the involution of the prostate by removing the material in the autophagic vacuoles.

Small seminiferous tubules lack advanced germinal elements. Additional connective tissue and myoepithelial layers produce a thickening of the limiting membrane. Some myoepithelial cells are atypical with an electron translucent cytoplasm and nuclei with dense peripheral chromatin. No spermatogenic cells beyond the cap phase of the spermatid are observed. The cytoplasm of Sertoli cells contains large lipid droplets and degenerating germ cells.

    Notes: Summary Neural lobes from rats which had been allowed to drink hypertonic saline for five days were examined electron microscopically and by bioassay of oxytocin levels. The profound changes in the ultrastructure were examined and the reversal of these changes in morphology was followed until the hormone levels returned to normal. The recovery of the gland as measured by the two parameters of structure and hormone content indicated that the morphological recovery apparently preceded the hormone level recovery, a factor which might be explained by continued release of hormone during the period of recovery.

    Notes: Summary A fine structural study of sternothyroid muscle spindles of young adult rat demonstrates the rare occurrence of unique nerve endings. These endings are situated in the juxta-equatorial region of nuclear-bag fibers, adjacent to the annulo-spiral sensory endings. They consist of a bundle of terminal axons less than 0.3 μ in diameter and appear to be disposed nearly longitudinally to the axis of the intrafusal fibers. Whereas, the annulo-spiral endings consist of a single axon, coiling around the intrafusal fibers. Transverse sections of these muscle spindles reveal these unique endings scattered around the muscle fibers, fitting into depressions on their surface. The innermost axons directly face the muscle surface and are separated by a narrow gap less than 200 Å in width. No Schwann cell process appears to be associated with these endings. From their unique multi-axonal composition, these endings are termed “bundled endings”. Investigations of developing muscle spindles show the occasional presence of a similar multi-axonal composition of sensory endings in perinatal rats. It is suggested that “bundled endings” are sensory in nature, carrying an immature feature over to adult life.

    Notes: Summary The development of the pars tuberalis was studied in the rat fetus from 13 days of gestation to 6 weeks after birth. After the closure of Rathke's pouch, the pars tuberalis anlage is clearly distinguishable from the anlagen of the partes intermedia and distalis. It comprises the entire basal portion of the adenohypophysial anlage; the limit between the anlagen of the pars tuberalis and the pars distalis is defined by Atwell's recess, i.e. the pathway taken by the hypophysial vessels coming from the vascular plexus of the median eminence. At 14 days the pars tuberalis cells are characterized by the presence of glycogen which persists in the adult. Their secretory differentiation (elaboration of granules with a diameter of 100–120 nm) is obvious at 15 days of gestation. It therefore, clearly precedes that of the other hypophysial cell types. Its functional differentiation takes place well before its adhesion to the primary vascular plexus of the portal system. Cystic formations appear just before birth in the pars tuberalis, much later than those of the pars distalis. These observations on the development of the pars tuberalis, together with previous observations on the adult PT in various species, showing that the specific glandular cells of the pars tuberalis are cytologically different from all known adenohypophysial cell types, seem to indicate a specific endocrine function of this lobe.

Special attention was paid to the different types of boutons, and the mode of termination of the striatonigral connection was examined. Two different types of nigra neurons with deep nuclear membrane indentations were observed. The synaptic organization of rat substantia nigra is very complex. Six types of boutons, with many characteristics, could be differentiated: pleomorphicvesicle boutons (40%), elongated-vesicle boutons (10%), small-round-vesicle boutons (10%), large-round-vesicle boutons (20%), “clear-terminal” (15%) and dense-core-vesicle boutons (about 5%). Vesicle-containing dendrites were observed entering dendro-dendritic synapses. One to four days after large unilateral striatal lesions were made, a great number of degenerating boutons were seen in the ipsilateral substantia nigra, while the contra-lateral substantia nigra was entirely free of degeneration. The newly defined pleomorphic-vesicle boutons were found almost exclusively in the process of degeneration. The neuronal origin of the other bouton types is discussed.

    Notes: Summary The pattern of follicular development during the estrous cycles of aged rats was examined and compared with that of mature rats. In both, preovulatory follicles are derived from a select group of small pre-Graafian follicles which begin to develop at estrus and reach the preovulatory size by the morning of proestrus, but the rate of growth, as judged by an increase in the percentage of granulosa cells incorporating 3H-thymidine, is accelerated in the follicles of aged rats. A second mechanism, which accounts for preovulatory follicles in aged rats, involves the rescue from atresia of pre-Graafian and preovulatory follicles. The existence of this mechanism is supported by the observation that at metestrus in aged rats virtually all follicles, regardless of their state of atresia, possess a high percentage of granulosa cells incorporating 3H-thymidine, indicating that the follicles are growing rapidly. However, some of these rapidly growing follicles show signs of atresia such as pyknotic nuclei within their granulosa cell layers. Since follicles in the initial stage of atresia contain defective oocytes (Peluso et al. 1979b), their rescue and development into preovulatory follicles would result in the ovulation of defective oocytes, a fact which accounts in part of the lower fertility in these older animals.

    Notes: Summary The postnatal development of microglial cells was investigated in the neonatal rat brain by use of light- and electron microscopy, including enzyme-histochemical techniques. Microglial cells were selectively stained by demonstration of their nucleoside diphosphatase (NDPase) activity and classified into three types: 1) In the early postnatal period, “primitive microglial cells” showing scantily ramified processes were found in the cerebral cortex, the hippocampal formation, and the hypothalamus. During the course of the first postnatal week the processes of this cell type developed gradually and the cells were transformed into typical ramified microglial cells, called “resting microglial cells”. 2) “Amoeboid microglial cells” showing typical features of macrophages were characteristic of the cerebral white matter. 3) “Round microglial cells” possessing a round soma and few pseudopodia but no characteristic processes occurred in large numbers in the sub ventricular zone of the lateral ventricle and as single elements in the vicinity of blood vessels. Histochemically, thiamine pyrophosphatase (TPPase) was demonstrated only in the fully developed, ramified microglial cells (“resting microglial cells”), which could be readily observed in the central nervous tissue from the age of 14 day. “Round and amoeboid microglial cells” did not show TPPase activity and disappeared after 14 days of postnatal life. By use of electron microscopy, in neonatal rats NDPase activity was apparent in the plasma membrane of the three types of microglial cells (“primitive, round, and amoeboid” types). They showed basically similar submicroscopic characteristics, i.e., well-developed Golgi apparatus, long strands of roughsurfaced endoplasmic reticulum, single dense bodies and vacuoles, and numerous ribosomes. “Amoeboid microglial cells” were characterized by their well-developed cytoplasmic vacuoles and phagocytic inclusion bodies. The present study strongly suggests a mesodermal origin for these microglial elements.

    Notes: Summary The noradrenergic terminals in the substantia gelatinosa of the dorsal horn of the cervical spinal cord of the rat were investigated by means of the histofluorescence technique and electron-microscopic cytochemistry using the glyoxylic acid-KMnO4 fixation technique. In accordance with the topographical distribution of fluorescent catecholaminergic fibers, noradrenergic terminals containing small granular vesicles were frequently observed electron microscopically in the outer layer of the substantia gelatinosa. These terminals were most frequently found to appose without showing typical synaptic features, small-caliber dendrites, spine apparatus, and rarely, large caliber dendrites. Only in a few cases, the noradrenergic terminals exhibited typical synaptic contacts with dendritic elements of small size. In addition, noradrenergic terminals apposed non-noradrenergic terminals containing small agranular vesicles. In rats bearing surgical lesions of the dorsal roots, no noradrenergic terminal were found in contact with the degenerated axon terminals in the substantia gelatinosa. These findings suggest that the noradrenergic afferents to the substantia gelatinosa may exert their influence on sensory transmission via dorsal horn cells.

    Notes: Summary In a study of the effect of pinealectomy on thyroid C-cell number, 8 animals out of 66 were found to have thymic tissue in close association with the thyroid. Cells containing immunoreactive calcitonin were found in all of the thyroids but in only one of the 8 pieces of thymus. These cells found in a piece of thymic tissue associated with the right thyroid lobe were located immediately under the capsule and did not form or associate with follicles. Unlike the other animals the rat with thymic calcitonin cells had an unequal distribution of C-cells between the left and right thyroid lobes, but the total number of thyroidal C-cells was the same as that of the other rats. Since the thymus proper was not examined in these 66 animals, ten additional rats were taken for such a study. Thyroid-associated thymic tissue was found in three of these, but none of these thymi showed any immunoreactive cells.

The rat jugular vein is provided with both adrenergic and cholinergic nerve fibers organized in plexuses located at the adventitial-medial border.     Notes: Summary Electron-microscopic survey of selectively stained microglial cells in the cerebral cortex of the rat reveals that the processes of this cell type often encircle axo-dendritic synapses. Enzyme-histochemical methods for thiamine pyrophosphatase (TPPase) or nucleoside diphosphatase (NDPase) were used for the selective marking of the microglial cells; TPPase and NDPase activities were observed in the plasma membrane of microglial cells. The synapses encircled by microglial processes displayed presynaptic structures containing round clear vesicles (50 nm in diameter) and a prominent thickening of the postsynaptic membrane. In vitro, the above-mentioned enzymatic activities were completely suppressed by neuroactive agents such as catecholamines and phenothiazine derivatives. Examination using enzyme-histochemical techniques suggests that a single enzyme may be responsible for both above-mentioned enzymatic reactions. The functional significance of microglial cells in the normal central nervous tissue is discussed.

The most elaborate tight junctions were found in the inner stripe of the outer medulla; rat: 10.6±0.8 strands and 205±24nm; rabbit: 11.6±2.4 strands and 291±55 nm. The elaboration of the tight junctions decreased continuously towards the papillary tip. Inner zone I; rat: 9.3±2.6 strands and 186±38nm, rabbit: 9.5±2.3 strands and 247±59nm. Inner zone II; rat: 7.1±2.2 strands and 129±32nm, rabbit: 8.5±1.4 strands and 199±26nm. Inner zone III; rat: 6.0±1.6 strands and 111 + 19 nm, rabbit: 7.0±1.5 strands and 183±43 nm. In the inner zone III comprising the papillary tip tight junctions with only 1–3 strands were not infrequently seen. Preliminary findings in the kidney of the golden hamster indicate a similar decline of junctional tightness along the collecting duct. These morphological observations suggest that the permeability of the paracellular pathway of the medullary collecting duct increases towards the tip of the papilla, especially in the rat. The functional implications for the medullary recycling of urea and electrolytes, and for the urinary concentrating mechanism are discussed. In addition, the tight junctions of the papillary epithelium are described.

    Notes: Summary Immunoreactive luteinizing hormone-releasing hormone (LHRH)-like material has been demonstrated in the pineal gland of the adult rat. The objective of the present study was to examine the ontogenetic development of this LHRH-like substance in the rat pineal with the peroxidase-antiperoxidase (PAP) method of Sternberger. LHRH-like immunoreactive material was not observed in pineal glands of newborn rats. The amount of material increased progressively from the 6th–12th day of postnatal development. On day 12, the amount of LHRH-like immunoreactivity was consistent and comparable in all pineal glands of male and female animals examined.

    Notes: Summary An ultrastructural and morphometric analysis was made of the nucleolar components in pinealocytes of 40 male Fischer rats sampled at eight times in an LD 12∶12 photoperiod cycle. Comparisons of results from the eight times showed variation in estimated mean volume of the granular component of ±29%, and of the fibrillar component ±11%, in relation to daily means. Peaks in mean volume of total nucleolus and its granular component occurred at 1 h of light. Near maximal and minimal mean volumes of the fibrillar component both occurred during both light and dark. Fibrillar centers (nucleolar organizer regions) of different sizes were found at all sampling times. It is concluded that temporal patterns in 24-h changes in the nucleolar components are most prominent in the granular component, and are more complex than suggested by changes in total nucleolar size or mean dimensions, and than represented by a simple biphasic circadian rhythm. Examples of different stages in the migration of the granular component, and of possible sites of nucleo-cytoplasmic transfer of nucleolar material, are described.

    Notes: Summary Localization of hCG binding sites and the process of endocytosis in pseudopregnant rat ovaries were investigated by indirect electron-microscopic immunocytochemistry. Immature female rats were treated with pregnant-mare serum gonadotropin (PMSG) and human chorionic gonadotropin (hCG) to induce ovarian luteinization. Eight days after priming with PMSG-hCG and 1–6 h before sacrifice the animals were given another injection of hCG to bind the receptors. Receptor sites to hCG localized by reaction product were present in most luteal cells, but not in primary follicular cells. The receptor sites were distributed on luteal cell surfaces facing interstitial spaces. Endocytotic pits containing hCG binding sites were rarely seen 1 h after hCG injection. At 2 h, hCG and presumably its receptor were taken up within endocytotic vesicles with the evidence of reaction product coated on the vesicle wall. With time, fusion of endocytotic vesicles with lysosome occurred and the reaction product appeared in phagolysosomes. The reaction product was localized on phagolysosomal inner surface or in free granular form. These findings suggest that hCG and its receptors were internalized through endocytotic pits and endocytotic vesicles and delivered to lysosomes probably for degradation. An additional experiment for localization of acid phosphatase was also performed to delineate the lysosomes and phagolysosomes.

    Notes: Summary The morphology and distribution of the sensory neurones of the pudendal nerve within the spinal ganglia of rats were investigated by use of horseradish peroxidase (HRP). The labelling was visualized in diaminobenzidine (DAB) or tetramethyl-benzidine (TMB)-stained sections. Injection of HRP directly into the pudendal nerve labelled perikarya predominantly in the sixth lumbar DRG (L6). Following injection of HRP into the scrotal skin, however, additional cells were labelled in L5 and SI. Labelling was invariably unilateral. Approximately equal numbers of small (〈30 μm) and large neurones (〉40 μm) were labelled following subcutaneous injections although injections into the nerve marked twice as many small cells as large cells. This suggests that, in the rat, most of the small-diameter fibres within the pudendal nerve ascend through L6. Although a cluster of neurones was observed in one experiment, the remaining 25 experiments did not reveal any somatotopic arrangement since the labelled perikarya were distributed evenly throughout the ganglion. Similar numbers of retrogradely labelled neurones (somatopetal transport of the tracer) were observed in both DAB- and TMB-stained sections, although TMB allowed the demonstration of anterograde (somatofugal) HRP transport by terminal labelling in the superficial laminae of the lumbar spinal cord, extending into laminae II–IV.

    Notes: Summary The hypothalamo-extrahypophyseal neurophysin pathways (HEH) and the three hypothalamic nuclei secreting neurophysins, the supraoptic (SON), paraventricular (PVN) and suprachiasmatic (SCN) nuclei, of normal and hypophysectomized rats were studied by application of the immunoperoxidase procedure. Eight well-defined HEH pathways were recognized. Their main sites of projection were: lateral septum and subfornical organ (1 and 2); tractus diagonalis (3); medial nucleus of the amygdala and lateral ventricle (4); nucleus periventricularis thalami, nucleus habenulae lateralis and periaqueductal gray (5); periaqueductal gray, pineal organ, collicular recess and subependymal region of the fourth ventricle (6); dorsomedial nucleus and premammillary area (7); perimammillary region, corpus trapezoideum, ventral surface of medulla oblongata, nucleus tractus solitarii, nucleus commissuralis, substantia gelatinosa and formatio reticularis lateralis of the medulla oblongata and spinal cord (8). Neurophysin fibers of unknown origin were found in the frontal cerebral cortex. It was noted that in pathway 5 the amount of immunostainable material undergoes changes with age. The three neurophysin-secreting nuclei reacted differently following hypophysectomy. Among the HEH pathways the only one that seemed to be affected by hypophysectomy was that innervating the lateral septum. It is suggested that the neurons that survive hypophysectomy either do not project to the neural lobe or, alternatively, display axon collaterals projecting outside the neural lobe. Such a neuronal population could be the origin of the HEH pathways.

    Notes: Summary The distribution of luteinizing hormone-releasing hormone (LHRH)-immunoreactive perikarya and processes was examined, in the untreated rat, with the unlabeled antibody enzyme method of immunocytochemistry on thick 50 μm vibratome sections. LHRH neurons were primarily observed in the preoptico-anterior hypothalamic and septal areas. Projections from these cell bodies to the median eminence form three distinct pathways, one laterally along the course of the optic tracts, one medially through the periventricular stratum of the third ventricle, and one through the tractus infundibularis. In addition, some of these cell bodies project to the organum vasculosum of the lamina terminalis (OVLT) and the subfornical organ (SFO). LHRH immunoreactive neurons were also noted in the anterior olfactory regions; they project along the medial olfactory tract to the olfactory bulb.

    Notes: Summary Regeneration of splenic tissue after autologous subcutaneous implantation provides a useful model for studying the development of splenic tissue. The development of the various non-lymphoid cells of the white pulp in the rat is described. It appears that regeneration of the implants is initiated by ingrowing vessels and a newly formed reticulum, which forms the microenvironment for the homing lymphocytes. Marginal metallophils are found at their characteristic location at the inner border of the marginal sinus five weeks after implantation. Trapping of antigen-antibody complexes reappears when the first primary follicles can be recognized.

    Notes: Summary Autoradiographic studies showed that in the rat hippocampus “microglia-like reactive cells” (MRC) and astrocytes are capable of proliferation in response to kainic acid (KA)-induced lesions. A marked increase in the number of labeled MRC was observed during the first four days after the induction of the KA-lesion. A proliferative response of astrocytes occurred at two days after the KA-lesion. After the induction of a KA-lesion brain macrophages and oligodendrocytes were only slightly labeled with 3H-thymidine. It appears likely that MRC is the main cellular element responding to this type of lesion.

    Notes: Summary The ultrastructure of tight junctions between uterine luminal epithelial cells of the pregnant rat was studied by the freeze-fracture technique. On day 5 of pregnancy, the day of implantation, the region of tight junctions extended three times as far down the lateral cell membrane as on day 1 of pregnancy, and the strands of the complex interlinked more frequently. These observations suggest that tight junctions on day 5 may be more efficient in preserving the contents of the uterine lumen from dilution or escape than at earlier times of pregnancy.

    Notes: Summary Cholecystokinin-like immunoreactivity has been demonstrated by radioimmunoassay and immunocytochemistry in the spinal cord of various mammals, in particular in nerve fibers of the superficial layers of the posterior column, but had not been detected in neuronal cell bodies. We report immunohistochemical evidence for the presence of a group of cholecystokinincontaining neuronal cell bodies in the lumbar spinal cord of the rat. This group of cells is only visualized after direct injection of colchicine into the spinal cord and is located near the central canal in the intermedio-medial nucleus of area X of Rexed.

    Notes: Summary The morphology and localisation of IgM- and IgG-containing cells in the spleen of rats immunized with sheep red-blood cells (SRBC) were studied by combining immunohistochemical reactions with routine histological and histochemical methods. It was shown that IgM cells occur only in the outer periarteriolar lymphocyte sheath (PALS), whereas IgG cells are present throughout the whole PALS. It has been concluded that these cell types have different routes of migration. Both IgM- and IgG-containing cells were found in the germinal center in close relation with the extracellular immune complexes. The significance of this localisation is discussed.

    Notes: Summary The morphological characteristics and distribution of the somata of serotonin-containing neurons in the brainstem of rats and cats were studied by use of the peroxidase-anti peroxidase (PAP) immunohistochemical method employing highly specific antibodies to serotonin. Antibodies were raised in rabbits against an antigen prepared by coupling serotonin to bovine thyroglobulin and using formaldehyde as the coupling reagent. The distribution pattern of serotonin neurons observed in the present material is essentially in agreement with that described by other investigators who used the Falck-Hillarp method. In addition, this immunohistochemical technique revealed serotonin-containing perikarya in the following regions: 1) the periaqueductal gray, especially lateral to the nucleus raphe dorsalis, 2) the nucleus interpeduncularis, 3) the nucleus parabrachialis ventralis and dorsalis, 4) the field of the lemniscus lateralis, and 5) the reticular formation of the pons and medulla oblongata. The described immunohistochemical procedure makes it possible to study central serotonin neurons in detail without pharmacological pretreatment. The wide distribution of serotonin neurons demonstrated in this study should be considered when interpreting experiments dealing with the serotonin system.

    Notes: Summary In the preinfundibular portion of male rat hypothalamus, the arcuate-median eminence region was examined after staining with anti-porcine ACTH 1-39 serum. In several cases, anti-β-endorphin serum was also employed. Both sera stain the same cell bodies. The cell bodies of the immunoreactive neurons are scattered in the subependymal layer, arcuate nucleus and lateral tuberal region. Fibers originating from these cell bodies are distributed extensively throughout these regions. They make synaptic contacts on immunonegative fibers in the arcuate nuclei, or terminate directly in the perivascular space in the internal layer of the median eminence. The fibers, however, are very scarce in the external layer of the median eminence, and do not directly terminate on the neurovascular contact surface. These findings suggest that the ACTH/β-endorphin neuronal system may serve neuronally and humorally as an intrahypothalamic constituent of the hypothalamic pituitary regulation system.

Due to its location the name “paraganglion supracardiale vagi” is suggested. Fluorescence microscopy of the paraganglionic cells displays an intense yellow-green fluorescence indicating the presence of biogenic amines. Ultrastructurally, chief cells containing dense-core vesicles form three kinds of synaptic contacts (afferent, efferent and reciprocal) with enlarged, mitochondria-rich nerve endings.

    Notes: Summary Growing rats (4 weeks old) were kept for 3 weeks at 11° C and 24° C respectively. The cold-adapted animals showed a significantly higher oxygen consumption (64%). Volume density of subsarcolemmal and interfibrillar mitochondria as well as volume density of fat droplets were estimated in M. soleus and the diaphragm of both groups. In cold-adapted animals, the total volume of mitochondria was significantly increased by 24% in diaphragm and 37% in M. soleus. The volume of subsarcolemmal mitochondria was almost doubled in each muscle, but the volume of interfibrillar mitochondria did not change significantly. The surface of the inner mitochondrial membranes per unit volume of mitochondrion in M. soleus was significantly increased both in interfibrillar and subsarcolemmal mitochondria, whereas the surface of the outer mitochondrial membranes per unit volume of mitochondrion was increased only in the subsarcolemmal mitochondria. The volume of fat droplets in the diaphragm and M. soleus of cold adapted animals increased significantly by 62% and 150% respectively.

It was found that these subcellular changes display a different pattern from those underlying the ACTH-induced adrenocortical cell growth in the intact rats. This result supports the view that compensatory adrenal hypertrophy does not involve activation of the hypothalamo-hypophyseal-adrenal axis.

Serotonin-containing nerve fibers were distributed throughout the entire cerebellum including the deep cerebellar nuclei, while 5-HT-positive neuronal somata were not detected in the cerebellum of either species. A different pattern of 5-HT innervation was found among the three layers of the cerebellar cortex. There were also interspecific differences in the pattern of distribution of 5-HT. In the rat, the pool of 5-HT nerve fibers mainly consisted of tangential elements, which were predominant in the molecular layer, while in the cat only a few 5-HT fibers were found in the molecular layer of the cerebellar cortex; dense networks of 5-HT nerve fibers were present in the granular layer. Some differences are evident in the pattern of distribution of 5-HT fibers in cerebellar regions classified on an anatomical and functional basis.

    Notes: Summary Glomus cells from carotid bodies of adult rats dissociated by means of collagenase or collagenase + trypsin were used to study by electron microscopy the endocytotic uptake of cationized ferritin (CF) tracer into subcellular compartments. The glomus cells were incubated with the tracer (1) in a basic salt medium (BM), or (2) in the BM into which calcium ionophore A23187 had been added, or (3) in a potassium-rich medium. Incubation of the cells in BM containing CF for 30 min resulted in attachment of the tracer to the cell membrane and uptake of a few solitary tracer particles into small vesicles and multivesicular bodies. No uptake into the cisternae of the Golgi apparatus was observed. Further incubation in BM containing CF for another 30 min resulted in increased uptake of the tracer into small vesicles and multivesicular bodies. A similar pattern of uptake was observed when the dissociated glomus cells were first preincubated in BM with CF for 30 min and then incubated for 1 min or 30 min in the BM solution containing both the ionophore and CF. Upon such incubation, CF particles were seen to penetrate into coated pits and sites of exocytosis at the cell surface. When the 30-min preincubation in BM was followed by incubation in a CF-containing potassium-rich medium for 15–30 min, uptake into vesicles, small lysosomes and occasionally also into profiles of the smooth endoplasmic reticulum was seen. Endocytotic mechanisms of the glomus cells are outlined.
