    Notizen: Abstract Barley (Hordeum vulgare L. cv. Golf) was grown in solution culture with controlled nitrogen availability in order to study the influence of nitrogen nutrition on ammonia emission from the leaves. Ammonia emission measured in cuvettes connected to an automatic NH3 monitor was close to zero for nitrate grown plants but increased to 0.9–1.3 nmol NH3 m-2 leaf area s-1 after 3–5 days of ammonium nutrition. Increasing concentrations from 0.5 to 10 mM NH4 + in the root medium increased NH3 emission from the shoots, root glutamine synthetase activity and NH4 + concentrations in apoplast, xylem sap and bulk tissue, while apoplastic pH values decreased. Inhibition of glutamine synthetase in nitrate grown barley plants by addition of 1 mM methionine sulfoximine (MSO) to the root medium caused ammonia emission to increase 5 to 10-fold after 2–3 hours. At the same time shoot tissue ammonium concentrations started to increase. Addition of an inhibitor of photorespiration, 1 mM pyrid-2-yl hydroxymethane sulfonate (HPMS) reduced this increase in ammonia emission showing a relation between NH3 emission and photorespiration. Oil seed rape (Brassica napus L. cv. Global) plants grown at 3 different nitogen levels (2N, 4N and 7N) in a sand/soil mixture showed increasing NH3 compensation points with increasing N level. This increase was highly correlated with increasing NH4 + concentrations in the leaf apoplast and total leaf tissue. The NH3 compensation points could be succesfully predicted on basis of the pH and NH4 + concentration in the leaf apoplast.

    Notizen: Abstract Comparisons were made between the efficiency of barley plant regeneration from anther culture (AC) and isolated microspore culture (IMC) for the European winter cultivar `Igri' and the spring F1 Australian breeder's hybrid Amagi Nijo×WI2585. In both cases, IMC produced a higher number of green regenerant plantlets per anther than AC. For `Igri' there was a 100- to 200-fold improvement and for Amagi Nijo×WI2585 there was a five- to ninefold improvement of IMC over AC. To improve the consistency and reliability of the IMC method, we investigated several parameters, including maltose concentration, subculture protocol, microspore plating density and colony plating density. Subculturing during the liquid culture phase produced no significant improvement in the number of microspores developing into colonies. The optimal concentration of maltose in the liquid induction medium was found to be 90 g l–1. Both microspore plating density and colony plating density were found to influence plant regeneration. Microspores produced the highest numbers of colonies when plated at densities greater than 5×104 ml–1, and colonies produced optimal numbers of green plantlets when plated at 12.5–25 colonies/cm2.

    Notizen: Abstract  The Yd2 gene in barley provides protection against barley yellow dwarf luteovirus (BYDV), the most economically devastating virus of cereals worldwide. Because resistance assays to identify Yd2-containing individuals from breeding populations are often difficult, we have developed a closely linked, codominant PCR-based marker for Yd2 using AFLP marker technology. The marker, designated YLM, can be amplified from barley genomic DNA prepared using a rapid and simple extraction procedure and, in a survey of more than 100 barley genotypes, was found to be polymorphic between most Yd2 and non-Yd2 lines. The YLM therefore shows excellent potential as a tool for selecting Yd2-carrying segregants in barley breeding programmes.

    Notizen: Abstract  The partial resistance to leaf rust in barley is a quantitative resistance that is not based on hypersensitivity. To map the quantitative trait loci (QTLs) for partial resistance to leaf rust, we obtained 103 recombinant inbred lines (RILs) by single-seed descent from a cross between the susceptible parent L94 and the partially resistant parent Vada. These RILs were evaluated at the seedling and adult plant stages in the greenhouse for the latent period (LP) of the rust fungus, and in the field for the level of infection, measured as area under the disease progress curve (AUDPC). A dense genetic map based on 561 AFLP markers had been generated previously for this set of RILs. QTLs for partial resistance to leaf rust were mapped using the “Multiple Interval Mapping” method with the putative QTL markers as cofactors. Six QTLs for partial resistance were identified in this population. Three QTLs, Rphq1, Rphq2 and Rphq3, were effective at the seedling stage and contributed approximately 55% to the phenotypic variance. Five QTLs, Rph2, Rphq3, Rphq4, Rphq5, and/or Rphq6 contributed approximtely. 60% of the phenotypic variance and were effective at the adult plant stage. Therefore, only the QTLs Rphq2 and Rhpq3 were not plant-stage dependent. The identified QTLs showed mainly additive effects and only one significant interaction was detected, i.e. between Rphq1 and Rphq2. The map positions of these QTLs did not coincide with those of the race-specific resistance genes, suggesting that genes for partial resistance and genes for hypersensitive resistance represent entirely different gene families. Also, three QTLs for days to heading, of which two were also involved in plant height, were identified in the present recombinant inbred population. These QTLs had been mapped previously on the same positions in different populations. The perspectives of these results for breeding for durable resistance to leaf rust are discussed.

    Notizen: Abstract Transcription of plastid genes in higher plants is driven by two RNA polymerases. One is encoded in the chloroplast, the other is encoded in the nucleus. RNA synthesis in ribosome-deficient plastids is performed exclusively by the nuclear-encoded enzyme. In vitro capping was used to identify the transcriptional start sites of the genes clpP and rpl23 in ribosome-free plastids of the barley mutant albostrians. No transcript initiation was found at sequences similar to eubacterial promoters. Instead, transcription started near the motif 5′-YRTA-3′, which is also conserved in mitochondrial promoters of higher plants. Our data suggest that the nuclear encoded RNA polymerase is active in mature chloroplasts and is the sole polymerase involved in transcription of rpl123.

    Notizen: Abstract To determine the optimum selection environment for barley (Hordeum vulgare L.) targeted at low-input, stress environment, barley lines were selected for high yield under stress (YS), high yield under non-stress (YNS), or average yield in stress and non-stress conditions (YA) during three breeding cycles (cohorts) of three years each. The lines were then tested in a total of 21 year-location combinations with average grain yields ranging from 0.35 to 4.86 t ha-1. Yield under stress of the YS lines was between 27% and 54% higher than that of the YNS lines, with the top YS lines yielding under stress between 16% and 30% more than the top YNS lines. Realized heritability was between 0.35 and 0.67 when selection was conducted under stress and was significant in all three cohorts. By contrast, selection under non-stress gave a significant response in only one cohort, and its efficiency in improving yield under stress was significantly lower than selection under stress. The best YNS line ranked only 19th for yield under stress. The highest-yielding lines under stress were not only selected under stress, but were also landraces collected in very dry areas (〈 250 mm total annual rainfall). This confirms earlier findings and supports the idea that the most effective way to improve productivity of crops grown in less-favored areas is to use locally adapted germplasm and select in the target environment(s).

    Notizen: Abstract The effects of an amino acid mixture and of plant growth regulators added to the FHG barley anther culture medium were examined using three barley cultivars (Cadette, Léger, and Igri) grown in two environments (growth cabinet and glasshouse). ‘Léger’ and ‘Igri’ were known as responsive, and ‘Cadette’ as recalcitrant to androgenesis. Our first experiment showed that the amino acid-supplemented medium was best for embryogenesis and regeneration of ‘Cadette’ and ‘Igri’ in both environments, and if ‘Léger’ in the growth cabinet. The addition of ABA and TDZ did not improve embryogenesis and plant regeneration, and PAA decreased them in the growth cabinet. The addition of the amino acid mixture in the FHG medium also reduced the percentage of albino plants in the growth cabinet, but growth regulators did not improve the percentage of albino plants, and in some cases increased it. In the growth cabinet, disregarding media, ‘Léger’ produced more embryos than ‘Cadette’ and ‘Igri’, and Léger' and ‘Igri’ produced more green plants than ‘Cadette‘. Percentages of albino plants were higher or ‘Cadette’ than for ‘Igri’ or ‘Léger’. In a second experiment, we compared seven hybrids with their parents for androgenic responsiveness. Hybrids had a higher ability to generate green plants than expected based upon the weighted average reflecting the contribution of each parent.

Igri, has been developed which is amenable to transformation studies using particle bombardment.     Notizen: Abstract Partial sequences (appr. 900bp) spanning from exon 2 to 4 of the gene encoding alcohol dehydrogenase 1 were obtained from four varieties of cultivated barley, Hordeum vulgare ssp. vulgare, and from 21 accessions of its wild progenitor, H. vulgare ssp. spontaneum. Sequence diversity was low and restricted to the non-coding intron sequences. In the 648–660bp long intron 3 both a microsatellite locus and a major duplication (153/156bp) were found. Sequence diversity in H. vulgare ssp. spontaneum was slightly higher than in cultivated barley. In general the Adh1 gene displays only limited polymorphism and is not appropriate for seeking correlations between molecular and eco-geographical data.

    Notizen: Abstract We have shown previously that transcripts corresponding to the cDNA clone pBH72-F1, with similarities to O-methyltransferases (OMT), accumulated in barley leaves in response to attack by the pathogenic fungus Blumeria graminis (Plant Mol Biol 26 (1994) 1797). To investigate the accumulation pattern in the defence response and the organ localization of the pBH72-F1-encoded polypeptide (F1-OMT), an antiserum was raised against Escherichia coli expressed F1-OMT. The 43 kDa protein was absent in normal leaves but accumulated strongly in response to pathogen attack. The F1-OMT protein accumulated faster in barley lines inoculated with an avirulent B. graminis isolates compared to a virulent isolate. Additionally, F1-OMT related proteins were detected in developing kernels. F1-OMT was expressed as a functional enzyme in E. coli and the substrate specificity was investigated. The enzyme exhibited OMT activity towards flavonoid aglycones with the highest activity against apigenin (4′,5,7-trihydroxyflavone). In contrast, caffeic acid did not serve as substrate for F1-OMT. The product of F1-OMT was analyzed by HPLC and GC-MS and found to be genkwanin (4′,5-dihydroxy-7-methoxyflavone). Initial velocity data were best represented by a sequential bi-bi mechanism, and kinetic parameters of KSAM=10.9µM,Kapigenin=4.6µM and a specific activity of 0.45 µkat/g were obtained. Barley F1-OMT, apigenin 7-O-methyltransferase, is suggested to be involved in the production of a methylated flavonoid phytoalexin.

    Notizen: Abstract A cDNA clone was isolated by differential hybridization from a library prepared from barley leaves inoculated with the fungus Blumeria graminis f.sp. hordei (Bgh). The open reading frame of the insert (designated HvCHS2) encoded a polypeptide with 72–79% identity to chalcone synthases (CHS) and 65–68% identity to stilbene synthases. Alignments of the amino acid sequence of HvCHS2 with the consensus sequence of naringenin-CHS (EC 2.3.1.74) reveals significant differences between HvCHS2 and naringenin-CHS. HvCHS2 transcripts accumulate strongly in barley leaves in response to inoculation with Bgh, whereas only insignificant accumulation of barley naringenin-CHS (CHS1) transcripts is seen upon the inoculation. The accumulation of HvCHS2 transcripts is also elicited by UV light. To compare the activity of HvCHS2 with the activity of CHS1, the two enzymes were expressed in Escherichia coli. Both HvCHS2 and CHS1 catalyse the formation of chalcones. However, HvCHS2 and CHS1 differ in their substrate requirements. CHS1 uses cinnamoyl-CoA and 4-coumaroyl-CoA at comparable rates whereas feruloyl-CoA is a poor substrate for this enzyme. In contrast, HvCHS2 converts feruloyl-CoA and caffeoyl-CoA at the highest rate whereas cinnamoyl-CoA is a poor substrate. Thus, HvCHS2 is a novel pathogen and UV light induces homoeriodictyol/eriodictyol CHS involved in the direct production of flavonoids possessing multi-substituted B-rings.

    Notizen: Abstract The iridoid glucosides lateroside 1, harpagoside 2, ajugol 3, and aucubin 4 were isolated from an ethanolic extract of the roots of the weed Verbascum thapsus that exhibits antigermination activity on seeds of barley (Hordeum vulgare). Bioassays indicated that at 3 mM concentration, compounds 1, 2, and 4 showed moderate inhibition of seed germination. These compounds also reduced root length when they were assayed on pregerminated seeds at 1 mM to 0.001 mM concentration range. Of all compounds tested, aucubin 4 was the most active against root elongation. Compound 3 showed no activity in the bioassays.

    Notizen: Abstract It is commonly agreed that cultivation of wild barley preceded the selection of the domesticated, non-brittle spike type. However, how common was wild barley cultivation before domestication and how many domesticated mutants gave rise to the barley crop could not be inferred from botanical and archaeological evidence. Some clues, nevertheless, can be obtained from the pattern of allozyme diversity in wild and cultivated barley obtained by Kahler and Allard (1981). Parallel variation, in terms of number of alleles per locus and frequency of the various alleles, was found in wild and domesticated barley. This similarity has been taken as an indication of multiple domestications and the frequency of the rarest alleles has been used to estimate that about 100 tough-rachis different mutants were necessary for the inclusion of the allozyme diversity of the wild barley in the domesticated crop. Assuming mutation rate of 10−6 in the locus governing tough rachis, the plant population required to generate these 100 mutants in one year would extend over about 200 hectares, or 10 hectares if the 100 mutants have been formed over a period of 20 years. The simplified calculations suggest that prior to domestication cultivation of wild barley was not a common practice.

    Notizen: Summary Transgenic barley plants (Hordeum vulgare L. cv. Kymppi) were obtained by particle bombardment of various tissues. Immature embryos and microspore-derived cultures were bombarded with gold particles coated with plasmid DNA carrying the gene coding for neomycin phosphotransferase II (NPTII), together with plasmid DNA containing the gene for β-glucuronidase (GUS). Bombarded immature embryos were grown to plants without selection and NPTII activity was screened in small plantlets. One plant proved to be transgenic (T0). This chimeric plant passed the transferred nptII gene to its T1 progeny. The presence of the nptII gene was demonstrated by the PCR technique and enzyme activity was analyzed by an NPTII gel assay. Four T0 spikes and 15 T1 offspring were transgenic. The integration and inheritance was confirmed by Southern blot hybridization. Transgenic T2 and T3 plants were produced by isolating embryos from green grains of transgenic T1 and T2 plants, respectively and growing them to plants. After selfing, the ratio of transgenic to non-transgenic T2 offspring was shown to follow the rule of Mendelian inheritance. The general performance of transgenic plants was normal and no reduction in fertility was observed. Microspore-derived cultures were bombarded one and four weeks after microspore isolation. After bombardment, cultures were grown either with or without antibiotic selection (geneticin R or kanamycin). When cultures were grown without selection and regenerated plants were transferred to kanamycin selection in rooting phase, one out of a total of about 1500 plants survived. This plant both carried and expressed the transferred nptII gene. The integration was confirmed by Southern blot hybridization. This plant was not fertile.

    Notizen: Abstract Genetic analyses of heading date, tiller number, plant height, grain yield, kernel weight, and plump and thin kernels were made in three six-rowed barley crosses (Hordeum vulgare L.) involving four cultivars. Six populations, P1 , P2 , F1 , F2 , BC1 , and BC2 , from each cross were grown and evaluated at Fargo and Prosper, North Dakota, 1982. Parental means within crosses generally were different except for tiller number. Comparison of generation means suggested that late heading was dominant to early, high kernel weight was dominant to low, and kernel plumpness was influenced by additive gene action. The relationship between yield and heading date was not consistent among crosses and positive r values were quite low. It should be possible to select early maturing, high yielding segregates with plump kernels. Heterosis over the mid-parent was quite similar among crosses for heading date, but there was no heterosis over the high parent. Inbreeding depression was fairly constant for heading date, but was less consistent for yield. The lack of uniformity for estimates of inbreeding depression can be related to environmental variation and to its influence on type of gene action. The ratio of additive to dominance variance was inconsistent among crosses for heading date and yield. These data suggest selection for these characters should be delayed past the F 2 generation. Broad sense heritabilities for heading date ranged from 42 to 86%. Values obtained for grain yield were more consistent among broad sense than narrow sense estimates. Genetic advance estimates were low due to lack of additive variance.

    Notizen: Abstract The Russian barley cultivar Nevsky lacks γ3 hordein and accumulates most of its hordein in the lumen of the endoplasmic reticulum and only a minor portion in the vacuole. In wild type barley and all other temperate cereals, storage proteins are deposited in the vacuole. F1 crosses revealed that the Nevsky phenotype is recessive; but the extent of hordein accumulation in the endoplasmic reticulum in F2 endosperm lacking γ3 hordein was very much less than in the Nevsky parent. In order to study the Nevsky endosperm phenotype we have measured the levels of seven proteins and two mRNAs involved in protein folding in the ER lumen or ER to Golgi transport during endosperm development. The protein levels were unaltered in Nevsky as compared to the wild-type variety Bomi. When the levels of these seven proteins were correlated with the rate of hordein accumulation, four of these (HSP70, PDI, Sar1p and Sec18p) were consistently up-regulated with hordein synthesis. Accumulation of hordein in the endoplasmic reticulum appears to be determined by the absence of γ3 hordein, or the product of a gene closely linked to it, plus one or more other recessive genes.

    Notizen: Abstract Accumulation of the pathogenesis-related (PR) proteins localised in intercellular spaces of barley primary leaves, chlorophyll content, structure of chloroplasts, and photosynthesis were examined during natural and in vitro induced leaf senescence (cultivation of whole plants in the dark or detached leaves under nutrient deficiency). Some of PR proteins accumulated during natural senescence, but their accumulation pattern was different from those of pathogen-induced as well as during in vitro-induced senescence, which indicate different molecular bases of these processes. Photosynthetic rate and chlorophyll content indicate that natural senescence of barley primary leaves began from 15th day after sowing. In 35-d-old first leaves, the chloroplasts showed typical characteristics of senescence as significant decrease of size, greater grana, and prominent plastoglobuli. The chloroplasts contained more grana under in vitro induced senescence and they had reduced length in the dark. Correspondingly, accumulation of PR proteins was detectable on about the 15th day but the content of some PR proteins increased in later stages of senescence.

    Notizen: Abstract Barley (Hordeum vulgare L., cv. Hemus) plants were grown in nutrient solution with or without 54 µM Cd2+ for 12 d. A treatment with Cd2+ inhibited the growth of young barley plants. The main factor limiting plant growth was net assimilation rate, due to decreased photosynthetic rate and accelerated dark respiration rate. One of the reasons for the reduced photosynthetic rate was the lower chlorophyll and carotenoid content. Cd2+ decreased water potential and transpiration rate, but relative water content in leaves of the treated plants was not significantly changed.
