    Notizen: Abstract We studied the effect of early tutoring on the subsequent sexual preferences and reproductive activity of female domestic canaries (Serinus canaria). Young female canaries were exposed during the first 4 months of life to songs of either domestic or wild canaries. When adult, these females were again exposed to domestic or wild songs. In the first experiment, the sexual responses of the females to unfamiliar domestic and wild songs were quantified with the copulation solicitation display (CSD) assay. In the second experiment, the same females were tested again with modified tutoring songs. In the third experiment, song stimulation of nest-building and egg-laying was studied. Domestic-strain-tutored females gave more CSDs to domestic than to wild songs. In contrast, wild-strain-tutored females showed no sexual preference. We propose that the sexual preference of adult domestic-strain-tutored female canaries for domestic songs is the consequence of learning and categorisation processes. The discrepancy between the results of the domestic-strain-tutored females and those of the wild-strain-tutored females suggests that female canaries have a predisposition to learn songs of their own strain rather than songs of an alien strain. In the third experiment nest-building and egg-laying activities appeared to be unaffected by early tutoring conditions: there was no significant differential effect of the different tutoring and exposure conditions on nest-building and egg-laying scores. Mate attraction and stimulation of females’ reproductive activity appear to be two separate functions of male song, which may have been shaped by different evolutionary constraints.

    Notizen: Abstract  Serum 25-hydroxyvitamin D3 [25(OH)D3] is produced in the skin in response to exposure to ultraviolet radiation, and is a good indicator of vitamin D nutritional status. The aim of this study was to determine summer/winter differences in serum 25(OH)D3 and parathyroid hormone (PTH) in Japanese women and how the summer and winter values are related. The subjects were 122 healthy Japanese women aged 45–81 years (average age: 65.7 years). They were medically examined twice, in September 1997 and February 1999. Serum 25(OH)D3 and intact PTH were determined by high-performance liquid chromatography and a two-site immunoradiometric assay respectively. Lifestyle information was obtained through an interview. The seasonal differences (winter minus summer) in 25(OH)D3 [Δ25(OH)D3] and intact PTH concentrations were –18.8 nmol/l (SD 19.2, P〈0.0001) and 0.98pmol/l (SD 1.02, P〈0.0001) respectively. The correlation coefficient between summer (x) and winter (y) 25(OH)D3 levels was 0.462 (P〈0.0001), with a linearly fitted line of y=0.42x+26.4. This relationship was interpreted as subjects with higher summer 25(OH)D3 values having greater reductions in winter 25(OH)D3 concentrations. There were inter-individual differences in Δ25(OH)D3, although the summer and winter 25(OH)D3 concentrations were well-correlated. Since Δ25(OH)D3 was not associated with any of the lifestyle factors, seasonal differences in the 25(OH)D3 concentrations of an individual appeared to reflect her ability to produce 25(OH)D3 photochemically in the skin. Sun bathing would be a less effective means of attaining adequate vitamin D nutritional status in a person with a small seasonal difference in 25(OH)D3, i.e., one with a low 25(OH)D3 level.

    Notizen: Abstract All yeast mitochondrial mRNAs terminate at their 3′ ends with a conserved dodecamer sequence, a site for high-affinity binding by DBP (dodecamer binding protein). Using purified DBP, we show that binding requires an intact dodecamer site and is enhanced by the presence in an oligonucleotide of the immediate 4–5 upstream nucleotides. Binding affinity varied from 0.25 to 0.85 nM towards a set of RNA oligonucleotides containing messenger specific upstream sequences in addition to the dodecamer site. Furthermore, we show that phosphatase treatment of DBP abolishes its specific binding, indicating the involvement of reversible phosphorylation in the regulation of its binding activities. This finding will further our understanding of the mechanism of DBP in the regulation of RNA metabolism in yeast mitochondria.

    Notizen: Summary The major driving forces in the eukaryotic cell cycle are the cyclin-dependent kinases (Cdk). Cdks can be activated through dephosphorylation of inhibitory phosphorylations catalyzed by the Cdc25 phosphatase family. In higher-eukaryotic cells, there exist three Cdc25 family members, Cdc25A, Cdc25B, and Cdc25C. While Cdc25A plays a major role at the G1-to-S phase transition, Cdc25B and C are required for entry into mitosis. The regulation of Cdc25C is crucial for the operation of the DNA-damage checkpoint. Two protein kinases, Chk1 and Cds1, can be activated in response to DNA damage or in the presence of unreplicated DNA. Chk1 and Cds1 may phosphorylate Cdc25C to prevent entry into mitosis through inhibition of Cdc2 (Cdk1) dephosphorylation.

    Notizen: Abstract A first exploration of applications of ecological niche modeling and geographic distributional prediction to endangered species protection is developed. Foci of richness of endangered bird species are identified in coastal California and along the southern fringe of the United States. Species included on the Endangered Species List on the basis of peripheral populations inflate these concentrations considerably. Species without protection in the US National Park System are focused particularly in peninsular Florida. Application of this methodology to additional taxa and regions holds promise for diverse conservation applications.

    Notizen: Abstract Migration flows from western Europe, the United States and Canada to east central Europe have received little attention so far. But numbers are now considerable and in the context of globalisation it seems appropriate to conceptualise communities of Westerners in terms of transnational social spaces as defined by Glick Schiller et al. Study of Americans in Prague reveals three main groups: enterprise managers, lifestyle migrants and entrepreneurs in small and medium-sized enterprises. All are engaged in a variety of different processes of community-building, often with clear trends towards a new transnationalism. Yet, the individual attempt to live in two societies is limited by the marked boundaries of the communities and by the degree of social exclusion which they experience from the majority population.

    Notizen: Abstract In a large cross-sectional sample, commercial airline pilots in the United States were asked for their perceptions of job safety hazards. Regression techniques are employed to investigate the relationship between these perceptions and both the length of tenure of pilots and their specific employer within the industry. The latter is found to have a far more significant impact on risk perception. No evidence is found for a learning curve of job risk with respect to experience. Pilots' assessments of inadequacies in training and aircraft maintenance are found to be significantly related to the financial health of their employer.

    Notizen: Summary We have determined the nucleotide sequence of a ribosomal protein gene which codes for the ribosomal protein S6 (rps6). The sequence analysis revealed that the gene comprises 239 amino acids, giving rise to a basic protein with a molecular weight of 27,502 Da. The product of this gene is the equivalent of the ribosomal protein S1O from Saccharomyces cerevisiae. Northern analyses and S1 mapping of both the 5′ and the 3′ end of the transcripts of this gene show that it is transcribed into three distinct transcripts with different sizes and heterogeneous termini. In the DNA region flanking the coding sequence, several conserved elements are present that may be involved in the transcription initiation and termination.

    Notizen: Summary In some strains of mice, eggs when X irradiated during the pronuclear stage, undergo a mitotic block in the G2 phase of the first cell cycle and cleave when the second division takes place in controls. The importance of this effect varies considerably with the strain and depends exclusively on the maternal genotype. In previous work, two-dimensional electrophoresis showed that eggs blocked at the one-cell stage after irradiation, undergo the same modifications in polypeptide synthesis as two-cell controls of the same age, except at the time of normal first mitosis, where three polypeptide sets of 30, 35 and 45 kDa appear only in cleaving controls. In the present study, we have found phosphorylations in dividing controls, on polypeptides of 30, 35 and 45 kDa. These phosphorylations are not seen in blocked irradiated eggs.

    Notizen: Summary Phosphorylation of microtubule protein was tested during differentiation in neuroblastoma cells. Two microtubule proteins were modified, β-tubulin and MAP-1 B. In the first case less than one mol of phosphate was incorporated per mol of protein, whereas several residues were phosphorylated in MAP-1 B. The localization of the phosphorylated residue of β-tubulin indicated that it is present in an isoform, at its carboxy-terminal region, and probably correspond to the serine 444. When comparing thein vivo phosphorylation of tubulin with that produced by casein kinase IIin vitro, a similar pattern was obtained. A similar result was found upon the comparison of the phosphorylation pattern of MAP-1 B after phosphorylationin vivo andin vitro using casein kinase II. These results suggest a role for casein kinase II in the phosphorylation of microtubule proteins in neuroblastoma cells. A result similar to that found for neuroblastoma cells was found after injection of [32P]phosphate into the brain of seven-day-old rats; however, a more complex pattern was found for the phosphorylationin vivo in adult rats.

    Notizen: Abstract When 100 mM KCl replaced sucrose in a chloroplast thylakoid stock suspension buffer, the membranes were converted from a localized proton gradient to a delocalized proton gradient energy coupling mode. The KCl-suspended but not the sucrose-suspended thylakoids showed pyridine-dependent extensions of the ATP onset lag and pyridine effects on post-illumination phosphorylation. The ATP formation assays were performed in a medium of identical composition, using about a 200-fold dilution of the stock thylakoid suspension; hence the different responses were due to the pretreatment, and not the conditions present in the phosphorylation assay. Such permeable buffer effects on ATP formation provide a clear indicator of delocalized proton gradients as the driving force for phosphorylation. The pyridine-dependent increases in the onset lags (and effects on post-illumination phosphorylation) were not due to different ionic conductivities of the membranes (measured by the 515 nm electrochromic absorption change), H+/e − ratios, or electron transport capacities for the two thylakoid preparations. Thylakoid volumes and [ 14C]pyridine equilibration were similar with both preparations. The KCl-induced shift toward a bulk-phase delocalized energy coupling mode was reversed when the thylakoids were placed back in a low-salt medium. Proton uptake, at the ATP-formation energization threshold flash number, was much larger in the KCl-treated thylakoids and they also had a longer ATP formation onset lag, when no pyridine was present. These results are consistent with the salt treatment exposing additional endogenous buffering groups for interaction with the proton gradient. The concomitant appearance of the pyridine buffer effects implies that the additional endogenous buffering groups must be located on proteins directly exposed in the aqueous lumen phase. Kinetic analysis of the decay of the post-illumination phosphorylation in the two thylakoid preparations showed different apparent first-order rate constants, consistent with there being two different compartments contributing to the proton reservoirs that energize ATP formation. We suggest that the two compartments are a membrane-phase localized compartment operative in the sucrose-treated thylakoids and the bulk lumen phase into which protons readily equilibrate in the KCl-treated thylakoids.

    Notizen: Abstract The rapid phase of fructose-1,6-bisphosphatase (FBPase) inactivation following glucose addition to starved yeast cells [reported previously] is inhibited on addition of 10 mM chloroquine (CQ) at about pH 8. This inhibition of inactivation was shown to be due to the prevention of phosphorylation of the enzyme. CQ was also found to inhibit general protein phosphorylation in the yeast cells. Glycolysis, as observed by changes in intracellular glucose-6-phosphate and extracellular glucose and ethanol concentrations, was shown to be significantly inhibited in cells treated with CQ. Similarly, a decrease in ATP concentrations was observed. However, during the early stages of phosphorylation of FBPase, levels of ATP were similar in cells containing CQ as in those without CQ. Thus, decrease in ATP levels is not thought to be significantly responsible for the inhibition of protein phosphorylation. However, the phosphorylating activity of cyclic AMP-dependent protein kinases is inhibited in vitro by relatively low concentrations of CQ. Thus, prevention of protein phosphorylation by CQ is believed to be due to inhibition of protein kinases in yeast cells.

    Notizen: Abstract A survey of dermatophytes isolated from patients seeking medical advice was made from 1982 to 1984 in the United States. The survey included 59 locations with data from 49 cities and one state. Listing of the isolated dermatophytes and the frequency given by percentage of total follows: Trichophyton rubrum 46.8%, T. tonsurans 33.3%, T. mentagrophytes 10.1%, Microsporum canis 4.5%, Epidermophyton floccosum 3,5%, M. gypseum and T. verrucosum both 0.7%, M. audouinii and T. terrestre both 0.1%, and T. violaceum 0.06%. No isolations of M. ferrugineum or T. schoenleinii were reported. Temporal increases were observed for frequencies of T. mentagrophytes, T. tonsurans and M. canis, and decreases occurred for frequencies of E. floccosum and T. rubrum when the data from this survey were compared by the goodness of fit test to data of the 1979 to 1981 survey. The percent of dermatophyte isolations identified as T. tonsurans correlated significantly with the percentage of blacks in cities of 100000 or more people.

    Notizen: Abstract The beliefs of Americans about the proper relationship between humans and their environment were profoundly affected by waves of immigration from Europe. Immigrants valued ownership of land, individuality, freedom, domination of nature, and technological development. These themes remain strong today as centerpieces of the American dominant social paradigm (DSP). That DSP has been reexamined and found wanting by an increasing proportion of Americans. This departure from the old DSP has progressed further among the public than among the elite who have a greater stake in preserving the status quo. Environmentalists constitute a vanguard trying to lead the people to a new, more environmentally oriented social paradigm. The beliefs of the old DSP and the new environmental paradigm (NEP) are contrasted in Table 2. Briefly, the NEP advocates stress love of nature rather than domination of it; compassion for other peoples, future generations, and other species; planning to avoid risk; limits to growth; fundamental social change; and a new structuring of politics. These two worldviews are likely to be in vigorous conflict for several decades in the USA. Social learning, spurred by deterioration of the old ways, is likely to lead Americans to a new perspective on their relationship to nature.

    Notizen: Abstract The response of the parasitoidCampoletis sonorensis (Cameron) as a natural control agent of the fall armyworm,Spodoptera frugiperda (J. E. Smith), was evaluated at 6 host densities and 2 temperature regime. A type-II functional response was exhibited by femaleC. sonorensis in response to varying densities of all armyworn larvae at both temperatures. Significantly more larvae were parasitized at 25°C for the host densities of 60 and 75, than for the same densities at 30°C. Third instar fall armyworm larvae were found to be the preferred stage for parasitization byC. sonorensis.
