    Notes: Heparin-coated surfaces exhibit prolonged compatibility with blood in vitro and in vivo. Deposition of platelets occurs on heparinized surfaces and results in thrombocytopenia, if the surface area is sufficient. Interaction of platelets with heparin-coated materials appears to depend on a protein intermediate. Precoating of heparinized surfaces with fibrinogen solutions, platelet-free plasma, serum, or exhausted or bentonite-adsorbed plasma does not protect against platelet interaction but incubation in albumin reduces platelet adsorption. The behavior of platelets in respect to protein-coated heparinized surfaces parallels the selective adsorption of specific proteins by platelets in suspension.

    Notes: This work was motivated by the need for a systematic compatibility screen for materials of interest for heart assist devices. A practical polymer screening and monitoring program for biochemical engineering materials was developed. Initial emphasis was placed on screening to measure degradation to low molecular weight, water miscible moieties over simulated in vivo exposure. Infrared spectrographic analysis of the simulated body fluid showed that the level of organic moiety corresponds to general clinical experience. Further confirmation of the technique came from evaluation of control polymers from The University of Texas Drug Research Laboratory. Correlation of results with cell culture viability in the presence of the pseudo body fluid is being carried out.

    Notes: After utilizing the nylon velour Silastic and polypeptide laminates in this experimental pilot study, we can conclude the following: 1Both nylon velour Silastic and polypeptide laminates are well tolerated in the clean wound and serve as a satisfactory framework into which fibroblastic proliferation can occur resulting in adherence or take.2In contaminated wounds, infection resulted in exudation with an accumulation of fluid at the interface of granulation tissue and velour. The exudate, not resorbed and unable to transcend the laminate barrier completely, serves as a nidus of progressive bacterial growth and causes separation and rejection of this synthetic fabric cover. The fluid accumulation appeared more common utilizing the Silastic laminate, while the polypeptide laminate appeared to permit an escape of this exudate to inhibit or delay the separation.3The Silastic laminated velour became firm and inflexible, predisposing with motion, to a mechanical type of separation at the margin of fabric and normal skin, possibly a contributing factor in exogeneous infection. This was seen also with the polypeptide laminate, but to a much lesser degree.4The nylon velour Silastic laminate as it is presently constructed is not acceptable as a synthetic skin cover for an infected burn wound, while the polypeptide laminated velour appears to possess physical properties which make it suitable for further investigation as an artificial skin substitute.

    Notes: Work on the rates of diffusion of urea through two different types of porous glass is reported and compared with data obtained using a commercial cellophane membrane from an artificial kidney. The results show that the rate of urea diffusion per unit area through the larger pored glass can be 3-4 times higher than through the cellophane membrane.

    Notes: Hydrophilic poly (glycolmethacrylate) HydronU. S. Trade Mark, National Patent Development Corporation, 375 Park Avenue, New York, N. Y. gels were used as vehicles for selected antibiotics. Polymer vehicles with various antibiotics were prepared and their biological activity tested by sensitivity tests on solid and liquid media by means of reference microbe strains. Polymer vehicles with chosen antibiotics were implanted in rats and the rate of diffusion tested in vivo by removing the vehicles at fixed time intervals and ascertaining the content of the remaining antibiotic in the vehicle. In the clinical part, the favorable results of application of antibiotics saturated gel vehicles in tympanoplastic operations and healing of inflammatory states after middle-ear operations are described.

    Notes: We regard and handle intercellular tissue like gels with ordered fiber molecules. By this preparative histolysis and histogenesis the formation of bone and tooth can be looked upon as a chemical reaction of a cell-secretion with the interstitial liquid seen as colloidal. The symplex alginate-collagen was directed by diffusion of Ca and H ions and then mineralized like bone and tooth. Collagen in the symplex with alginate gives big pores similar to the spongiosa of bone. Implantation in animals shows healing without rejection and inflammation. The ionotropic gel for a leading structure was remineralized and calcified with hydroxyapatite. Higher temperature gives better crystals by the increasing rate of diffusion and recrystallization. By alternately changing waves of phosphate and calcium, the content of calcium phosphate of the ionotropic gels could be increased to 50%. The mechanical stability was then increased. Waves of ions cause a shrinking of the gel at the same time. This is the reason for the high stability of bone and tooth, shell and pearl. Citrate favors the transformation of the secondary phosphate Brushit into apatite even at room temperature. X-Ray diffractions of these crystals show the formation of 90% apatite in ionotropic gels by directed intermicellar crystallization, and only 10% Brushit.

    Notes: An elastomeric terpolymer of butyl acrylate, methyl methacrylate and methacrylamide was synthesized by emulsion polymerization using an anionic sufactant. The latex terpolymer particles where shown to have a negative charge and the solid terpolymer was negatively charged with respect to Ringer's solution. Caval rings prepared from the latex and properly post treated and cured remained patent after implantation in the inferior vena cava of dogs through a 2 week period, thus demonstrating significant thromboresistance.

    Notes: Heparin has been ionically bonded to glass surfaces. The heparin coating resists fluid shear stresses as high as 104 dynes/cm2 at 30°C for periods in excess of 300 hr, and 1000 W of ultrasonic cleaning at 62°C for periods of more than 45 min. It also prevents the clotting of human blood.

    Notes: The Lippman electrocapillary method was used to survey the adsorption behavior of 25 amino acids at the mercury-solution interface. The amount of surface excess of adsorbed amino acid (Γ;), as evidenced by lowering of interfacial tension, was found to increase with the number of double bonds and/or additional nitrogen atoms which provide electron pairs. No influence of dipole moment on adsorption was observed. Adsorption was more extensive at potentials positive to the potential of zero charge, indicating a possible electrontransfer adsorption mechanism, i.e., a sharing π-electrons rather than dipole interaction with the metal. These results are consistant with the enhanced adsorption of proteins at anodic potentials since the side groups extending from peptide linkages on a given protein are the R groups of various amino acids.

    Notes: Electrochemical cells consisting of stainless steel anodes and platinum black cathodes are attractive as power producers for cardiac pacemakers. The maximum power of these systems is of the right order. The electrocatalytic activity of various oxygen electrodes follows the same order in blood, as ascertained from in vivo studies, and in simple aqueous electrolytes. Subcutaneous implantation of the stainless steel anodes is preferable to intraperitonial insertion. The activity of a stainless steel anode is entirely due to its dissolution.

    Notes: A number of inhibitors of intrinsic blood coagulation have been examined. Particular emphasis was placed on heparin, heparin analogues, and hirudin. These inhibitors interfere primarily with two clotting factors in the intrinsic clotting scheme. These include factor IXa and thrombin. Our data suggest that factor IXa is the heparin-and hirudin-sensitive factor in plasma when coagulation is initiated via factors XII or XI, and that subsequent clotting factors remain in inactive forms. Studies with heparin and hirudin also clarified the role of thrombin in the interaction of factors IXa and VIII. It was concluded from these experiments that factor VIII readily interacts with factor IXa in the presence of calcium ions and phospholipid independent of thrombin.

    Notes: Exposure of heparin-bearing anticoagulant materials to whole blood, plasma, or serum has been shown to lead to partial elution of heparin. Similar effects are obtained when graphite-benzalkonium-heparin (GBH) or poly-4-vinyl-pyridine-heparin (PVPyrH) surfaces are exposed to bovine plasma protein fractions. Loss of heparin from these anticoagulant materials appears to be dependent on the nature of the cationic surface and on the particular protein fraction used. With whole blood and plasma, and with most of the protein fractions studied, it was found that a larger fraction of the original heparin was eluted from GBH than from PVPyrH. Surfaces exposed to bovine Cohn fraction IV lost considerably more heparin than similar surfaces exposed to Cohn fractions I, II, III, or V, and fraction V removed less than the others, at the same or higher concentrations, and less from PVPyrH than from GBH.

    Notes: The compatibility of candidate implant materials with living cells was evaluated in vitro. The materials and their interaction with living human blood and tissue cells were observed, recorded, and analyzed by means of reflected light interference microscopy. Blood cells-materials interfaces were prepared by incubating the materials in contanct with whole blood or platelet-rich plasma. The interaction between the materials and the cells was examined either in a static or a dynamic flow system, and recorded by photography or cinemicrography. The tissue cell-materials interfaces were prepared by cultivating animal and human cells as a monolayer directly on the surface of the material to be evaluated. The resulting implant-cell specimens are suitable for histological fixation and subsequent analysis by modern evaluation techniques such as scanning electron microscopy and elemental analysis by electron microprobe. These studies can be performed on the same implant specimen before, and following its exposure to the biological material. Furthermore, separate analyses of the implant and biochemical analyses of the cells and culture medium can be carried out before and after the interaction. These techniques have been used to investigate the interaction of blood and tissue cells with a variety of promising implant candidate materials such as polymers, metals, glass, and carbon surfaces. As a result of these studies, a number of observations have been made which may be of practical significance to the design engineer in developing and evaluating the safety of prosthetic materials for use in the human body.

    Notes: Factor assays of normal citrated plasma following incubation with nonleaching heparinized surfaces in high surface area-plasma volume ratios showed a marked pattern of reduction in factor IX activity, with lesser depression of II and X. No or modest effect on other factors was observed. In particular, no significant adsorption of factor XII (Hageman) by either cationic or anionic surfaces, was found. No consistent pattern of effects was seen with a group of strongly anionic surfaces, analogous to the sulfate and sulfamate groupings of the heparin molecule. However, certain members of this class showed profound effects on factor XI.

    Notes: Infraref internal reflection spectroscopy has been used to study the adsorption of certain plasma proteins on a variety of hydrophobic polymer surfaces. The behavior of the systems studied was almost identical. Under Static conditions the proteins appear to be rapidly adsorbed as monomolecular layers from solutions varying in concentration between a few mg-% and normal plasma levels. These monolayers are deduced to be closely packed arrays in which the protein molecules appear to retain their native globular form. The bearing of these results on the mechanism of surface-induced coagulation is significant.

    Notes: The nature of the events occurring immediately after exposing foreign surfaces to fresh flowing blood was assessed using a combination of MAIR infrared spectroscopy, ellipsometry, and contact angle measurements. Within the first 5 sec after contact with blood issuing from the jugular veins of lightly anesthesized dogs, germanium prisms were uniformly coated with strongly adherent proteinaceous filsm having an average optical thickness equivalent to 2 layers of stearic acid (approx. 50 Å) and having critical surface tensions of about 36 dyne/cm. A contact time of 60 sec led to a less uniform coating of average optical thickness equivalent to 5 layers of stearic acid (approx. 125 A), but with similar MAIR spectrum and wettability. The internal refelection spectra were consistent with the presence of either alpha-helical or random-chain configurations, but not with the extended chain beta-structure for proteins. Films cast from purified fibrinogen on platinum foils gave similar spectra and exhibited similar wetting properties; these observations are consistent with the hypotehsis that initial adsorptive events are dominated by fibrinogen deposition. The involvement of small amounts of lipid or other protein remains a distinct possibility, however.

    Notes: In the circulation, adherent thrombi are most likely formed by nucleation and growth processes. The adsorption of an anchoring layer must be followed by further constituent interactions; these by crosslinking introduce cooperative effects which affect nucleation and growth rates. Adsorption of thrombin to nonpolar surfaces has been examined using a clotting time assay. Assay discrimination was determined by coefficients of variation: of the assay itself (0.6%), of reproducing diluted thrombin sources (0.37%) and of reproducing sets of dotting tubes containing fibrinogen (1.5%). Adsorptions are 0.54 mg/m2 at the solution-air interface, 0.77 mg/m2 at the solution-clean wall interface, and 0.04 mg/m2 to walls previously treated with thrombin. Average distance between thrombin molecules is 89 Å, the minimum adsorption energy is -11.7 kcal/mole and estimated thrombus adhesive strength 10kg/cm2. Thrombin can be the anchoring layer for a thrombus which contains thrombin, fibrin and platelets since all constituents interact with each other. The thrombin-fibrin intersection energy is about -7 kcal/mole. The platelet can, at least on interaction with thrombin, undergo structural changes which increase interaction. A variety of interactions and cooperative effects are possible. A problem is to develop protocols and assays which will permit specification of the anchoring layer and characteristics of subsequent interactions.

    Notes: Orthopedic appliances of type 316 (SMO) stainless steel were examined in detail after being removed from human subjects. Techniques included conventional microscopy with various lighting methods, together with electron microscope fractography. Corrosion, fatigue failure, and manufacturing defects were identified and statistically tabulated. Corrosion damage occurred on 91% of all multicomponent devices and the overall incidence of fracture failure was 9%. These results indicate an urgent ueed for stronger, more corrosion-resistant implant alloys.

    Notes: Surgical suture materials were coated with a layer of hydrophilic Hydron gel. By tests carried out on tissue cultures, it has been verified that this layer itself improves the tolerance of the fiber by the organism. In addition, appropriate biologically active substances (e.g., antibiotics), preventing either the development or spreading of an infection along the fiber, may be incorporated into the gel layer. Neither the knotting ability nor other mechanical properties of the fiber are influenced by the gel coating. Preliminary clinical experiments showed good results.

    Notes: A new compound material is described, consisting of collagen sponge and glycol methacrylate, with a density varying from a compact to a highly porous spongelike material. Mechanical properties, swelling capacity, trypsin digestion rate, structural stability, and dynamics of tissue reactivity after subcutaneous and subperiostal implantation of this material were compared with collagen sponge, and the glycol methacrylate polymer alone. It has been shown that combined polymer collagen sponge with high porosity is quickly penetrated by connective tissue which maintains the character of highly vascularized loose connective tissue. The porosity of the material is a decisive factor for tissue permeation and the size of the pores should exceed 100 μ. The combination of the glycol methacrylate polymer gel with a collagen sponge increases the mechanical strength of this hydrophilic material. Optimal conditions for construction of the gel-collagen sponge were found.

    Notes: The most commonly used synthetic polymers in corneal implants are reviewed from the viewpoint of their properties and tissue tolerance. Special attention is given to the properties of these materials in in regard to their application as artificial corneal epithelium and endothelium. It is postulated that an ideal artificial corneal membrane must exhibit, as closely as possible, the physical properties and the functional characteristics of the natural membrane. Of all the known artificial materials, homogeneous hydrogels seem to offer the best possibilities as membranes for corneal surgery. Three types of hydrogels were investigated. Poly(2-hydroxyethyl methacrylate) (PHEMA) and poly(glyceryl methacrylate) (PGMA) are covalently linked tridimensional nonionic networks. The third type of hydrogel has multiple ionic crosslinks and belongs to the family of the polyelectrolyte complex hydrogels, made by reacting poly(vinylbenzyl-trimethylammonium chloride) with sodium poly(styrenesulfonate). The acrylic hydrogels are very well tolerated by living tissues, but the polyelectrolyte complexes gave inconsistent results. Membranes of either type were made having permeability to water similar to that of the corneal layers. Most hydrogels are friable and do not hold sutures; this difficulty was solved by attaching the hydrogels are friable and do not hold sutures; this difficulty was solved by attaching the hydrogel membrane to the cornea by means of alkyl cyanoacrylate adhesives. n-Heptyl and n-butyl 2-cyanoacrylate gave very good results in this application.

    Notes: A method of testing the tensile strength of the porcelain-gold bond has been developed. Tensile strengths of the bond are improved primarily by the formation of an oxide layer on the gold before baking the porcelain and not by the degree of gross surface roughness. The oxide layer may act either as a transition ayer or it may create microsurface roughness. The gross surface roughness probably complicates the wetting action of the porcelain on the gold. Whether the actual bond is of a chemical nature, physical nature, or both, remains uncertain.

    Notes: In this study tissue-suture interaction was evaluated in terms of instantaneous stress and total work at withdrawal, histological response, and photomicrographic suture appearance. Porcine subjects and subcuticular incision closures identical to those of clinical plastic surgery were used; implant periods were 1, 2, and 3 weeks. Monofilamentary sutures (4-0) of nylon, linear polyethylene, polypropylene, and Teflon FEP were used; stainless steel and braided siliconized silk were included for comparison. Perisutural sleeves, formed by ingrowth of epithelium up to 1 cm from the loci of emergence of the suture through epidermis, were found to account for 70-85% of withdrawal work. The subcutaneous closure is superior to interrupted or running stitches in minimizing these “end effects” and attendant exacerbation of tissue-suture interaction. The degree of the latter combined with consideration of suture strength, suppleness, and knot durability indicate that nylon and Teflon FEP monofilaments are sutures of choice for cutaneous closure.

The procedure was varied by using three spruing methods, two different waxes, two melt temperatures, and two different directions of filling. Statistical analysis techniques were used to determine the significance of the results. Spruing method was the most highly influencial of the variables and the flat method gave the best results. This method also is most relevant to clinical procedure. The effect of temperature, in fewer instances, was also in fluential. At a lesser level of signifiacnce direction of filling, and in one instance wax, had an effect.

    Notes: Fifty-eight polymers and other materials were evaluated for compatibility with human blood in an in vitro test system. Assays for platelet factor 3 activation, activation of the coagulation sequences, and release of hemoglobin and adenine nucleotides were performed on blood exposed to each of the 58 test materials. Of the materials tested, collagen, heparinized hydrin rubber, poly (methyl methacrylate), graphite-benzalkonium-heparin, poly (methyl acrylate), isotactic poly(methyl methacrylate), grafted poly (ethylene oxide) on poly (acrylic acid), balata rubber, L-1624 from 3M Co., and chlorinated poly (vinyl chloride) appeared most compatible with blood. Chemical affinities appeared to have little to do with blood compatibility of the materials tested. Electron microscopic examination of blood components adherent to selected test materials following exposure to blood was carried out. Each of the three polymers with “good” and the two polymers with “poor” blood compatibility characteristics which were examined by electron microscopy was coated with a thin unilaminar adsorbate from blood. Platelets and fibrin were focally deposited on this adsorbate. Of the test surfaces examined, only glass was covered by a trilaminar adsorbate. Mechanisms of formation of the adsorbates are discussed.

    Notes: At least ten proteins play a role in normal blood coagulation. Nine of these are important in coagulation promoted by contact with glass, and are probably involved in coagulation triggered by other surfaces. These proteins have been established as separate entities by the existence of human mutants who lack, on an inherited basis, the ability to synthesize one of them. They have been assigned Roman numerals until their chemistry and mode of catalysis are better understood. Several of them have been substantially purified. Experiments with purified preparations indicate that they interact in pairs. Thrombin (activated factor II) converts fibrinogen (factor I) to fibrin by controlled partial proteolysis; the same mechanism probably operates in the conversion of factor IX to activated factor IX by activated factor XI, and gel filtration experiments are presented here which support this view. Similarly, partial proteolysis is probably the mechanism whereby thrombin converts factor XIII to activated factor XIII. Activated factor XIII converts fibrin from a urea-soluble form to a urea-insoluble form by transpeptidation between a glutamine residue on one fibrin strand and a lysine e-amino residue on an adjacent fibrin strand. In general, when a pair of coagulation proteins interacts, one of them, the enzyme, converts the second from enzyme, which then catalyzes yet another similar reaction. The probable exceptions to this rule are factors VIII and V, which interact with phospholipid and another protein in a more complex manner.

    Notes: Recording ellipsometers and surface film balance, breath patterns, and ferric oxide powder “staining” were used to observe adsorption of normal plasma constituents onto metal oxide, silicon oxide, and polymer surfaces. Adsorbed proteins could be identified by their ability to adsorb matter from corresponding antihuman sera. Data indicate that oxidized silicon crystal surfaces adsorbed fibrinogen out of plasmas within 2 sec; within 20 sec, these films lost their ability to attract antifibrinogen and were later partially removed if intact factor XII was present in the solution. One aminated nonheparinized polymer formed films that adsorbed large amounts of fibrinogen and some gamma globulins out of plasma in the ellipsometer, and platelets out of platelet-rich plasma, as well as ferric oxide out of aqueous suspension. After taking up heparin, these polymer films appeared able to adsorb only small amounts of protein or oxide, and no detectable amounts of fibrinogen or platelets. On another aminated polymer, heparin seemed without effect. Interaction of metal surfaces with plasma may be complicated by oxidation.

    Notes: Corrosion potentials of materials composing dental amalgam have been measured in artificial saliva as the specimens were subjected to an increasing tensile load. Further, tensile tests have been carried out on specimens which had been previously subjected to controlled corrosion.The corrosion potentials of Cu, Ag, Zn, Ag2Hg3, Ag3Sn and finished dental amalgam showed only a slight increase in the anodic direction as the load increased. The potentials of Sn and Sn amalgams, on the other hand, showed a considerable increase.Most of the above results were explained as being a consequence of the rupture and repair of protective films as stress was applied or released. For Sn, however, it was necessary to postulate a complex attack by the corrodant to explain the large potential change.Of the phases of dental amalgam whose tensile strengths were measured after subjection to controlled corrosion, only the γ2 or Sn-Hg phase was affected. Its strength was lowered by corrosion, whether or not stress was applied.It was concluded that stress, as such, probably is not a direct factor in the corrosion of dental amalgam under oral conditions.

    Notes: Observations of microcirculatory blood flow in the wing membrane of the unanesthetized bat, Myotis leucifugus, show that many responses associated with the exposure of blood to foreign surfaces mimic normal behavior in these sites.     Notes: By reflected light microscopy, the thrombi formed on a variety of foreign surfaces have been shown to be composed of isolated cellular aggregates and an interaggregate red cell fibrin mesh. The thrombi viewed by this method were seen in two dimensions as they formed at the foreign surface blood interface. Standard histological evaluation of the thrombi in the third dimension, that is in the depth dimension, revealed columnar shaped cellular aggregates extending from the foreign surface into the blood and a red cell fibrin mesh surrounding the aggregates. The thrombi formed at Epon plate-blood interfaces were viewed with an electron microscope. Plates made of Epon embedding epoxy were used as the foreign surface exposed to blood, and the plates and fixed initial thrombi formed on them were embedded in more Epon. A layer of heavy metal staining, noncellular material was found covering the surface of the plates exposed to blood, with platelets deposited on this layer. Since platelets were not found adherent to the foreign surfaces directly, it is our postulate that the intervening film “conditions” the surface preliminary to platelet adhesion.

    Notes: It has been found that when n-butyl α-cyanoacrylate is sprayed on a blood surface from an aerosol containing Freon as the propellant, the polymeric cyanoacrylate film which forms contains 1.4% propellant under the spray conditions used. Studies of the rate of loss of propellant from the film indicate that ⅓ of the Freon initially present in the film still remains after 90 min of exposure of the film to air. Possible implications of these results are discussed.

    Notes: The use of cyanoacrylate tissue adhesives provides a quick method of closing wounds by nonsuture techniques. This study was undertaken to determine the relative effects on healing of a number of alkyl α-cyanoacrylate tissue adhesives compared to sutured incisions. Tissue bonds were prepared with the alkyl α-cyanoacrylate adhesives and sutures and tested for bond strength in vivo in rats over periods of 1, 3, 7, 10, and 14 days. A trend of increasing bond strength with the higher homologs was immediately apparent. However, this trend was reversed at the time of the 14 day measurements with the higher homologs of the cyanoacrylate adhesives having the lower bond strength. Microscopic examination of pulled wounds closed with methyl α-cyanoacrylate showed the cause to be substrate failure with the exception of the one hour postoperative samples which demonstrated adhesive failure. Wound failure in the remaining three monomers was cohesive, gradually decreasing to substrate failure at 10 days postoperative. The tensile strength of all wounds closed with tissue adhesive dropped considerably on the third day postoperative.

    Notes: A tissue culture test and a 7 day implantation method in rabbits were used to screen the toxic potential of 50 tubings used in one or more medical or paramedical applications. Results of the tests revealed that approximately 32% of the tubings contained one or more ingredients which, when released to cells or tissue, caused a toxic response. A similar study six years previously showed that 52% of the tubings tested had toxic potential. It is apparent that improvements have been made in the medical tubings (from point of view of safety), but steps must still be taken to reduce the potential risk to levels approaching zero.

    Notes: The influences of conditioning treatments and surface topography on the heparin adsorptivity and the thromboresistance of a high-strength, impermeable, isotropic, pyrolytic carbon were investigated. The results of the adsorption studies indicate that the adsorption of heparin on such carbon surfaces is near the amount required for monolayer formation. The adsorbed heparin is rapidly elutriated in plasma. The heparin adsorption is not enhanced by a pretreatment with benzalkonium chloride. There was no relationship between the amount of heparin adsorbed on these materials and their compatibility with blood. Polishing, for example, which reduced heparin adsorption, enhanced the tromboresistance of these carbons, and while chemisorption of oxygen markedly reduced their thromboresistance, it did not influence the amount of heparin that could be adsorbed. In vivo tests showed that polished and outgassed, impermeable isotropic carbons deposited at low temperatures were significantly thromboresistant without the exogenous application of heparin. Examples of applications of these new materials in experimental valves are presented.

    Notes: A large number of activated carbons, carbon blacks, ion exchange resins, and inorganic adsorbents were screened for their capacity to remove urea from dilute neutral solution. An adsorbent capable of performing this task would be of assistance in decreasing the volume of dialyzing fluid required for hemodialysis. All activated carbons and carbon blacks had some adsorption capacity for urea. The most efficient was a carbonized poly(vinylidene chloride). The inorganic adsorbents exhibited negligible urea adsorption capacity, as did ion exchange resins in the ionic form at equilibrium with simulated dialyzing fluid. Several chemical reactions for urea were found not to procede in dilute neutral solution, even with carbon present to adsorb reaction products. Attenuated total reflectance infrared spectroscopy showed no band shifts in urea adsorbed on strong acid ion exchange resin. Measurement of the spectrum of urea adsorbed on carbon was prevented by adsorption of the incident energy by the carbon.

    Notes: Certain thermoplastic polymers possess numerous desirable physical and mechanical properties important for total joint replacement. Their modulus of clasticity and density are close to the cartilage bone composite. Their elasticity would provide valuable damping for the transmission of shock forces. As a class of materials, polymers do not have the compression and tensile strength of metals, but the high strength-to-weight ratio of some polymers makes them especially attractive as a material for the socket half of the joint. The socket shape could be easily molded or machined and at a low cost. As a prime requisite they must have sufficient wear and creep resistance for a lifetime of cyclic loading and sliding. In addition they must be biologically tolerated, and the joint environment must not degrade or otherwise adversely affect the physical and mechanical properties of the polymer.The purpose of this paper is to present a friction and wear analysis of ten thermoplastic polymers selected because their individual physical and mechanical properties suggested good wear and creep resistance. Polyimides with and without graphite have particular potential as a bearing material. The toxicological aspects of these materials are under study and will be the subject of a subsequent report.

    Notes: Finite difference treatment of two-dimensional problems in elastostatics is usually based on the differential equations for the displacement vector or the Airy stress function, depending on whether boundary conditions are on displacement or stress. In either case, determination of stresses requires numerical differentiation and therefore use of a rather fine grid. Moreover, neither method is suited to the treatment of mixed boundary conditions. The alternative method developed in this paper uses the first derivatives of the displacement components at the grid points as basic variables and hence does not require numerical differentiation in the evaluation of stresses. Appropriate finite difference equations are established, and their use is discussed in connection with a specific example with known explicit solution.

At corner nodes the variables are the displacement and the two slopes. At midside nodes the variables are the displacement, the slope normal to the side, and the rate of change of this along the side - i.e., the rate of torsion. Of two options, one divides the triangle into three and takes different quartic displacements in the three regions, and the other uses displacement functions with singularities at the vertices. According to a test which compares the energies of the two formulations by eigenvalues, the second is the better if the integration is accurate. The same test, together with others based on finite differences and on nodal values, etc. checks the correctness of the routine decisively.

    Notes: The direct Finite Element Analysis which was successfully employed in the solution of dynamic flexural traveling wave problems is extended herein to provide the transient behaviour of finite beams and plates in which shear deformation and rotatory inertia are considered. The particle and angular velocities are exponentially damped so that the static solutions for these problems are obtained with the same analysis which provided the dynamic and transient cases. Three special cases are chosen as examples. In the first, a sinusoidally varying shear force is applied at the tip of a cantilever beam. The resonant characteristics of this beam for both the undamped and damped cases are studied. In the second, a step shear loading is applied to a cantilever beam and its damped dynamic history is studied. Finally, a circular plate whose outer edge is simply supported is impacted at its inner edge by a step moment and its damped transient behaviour is determined. The idea of the methods is potentially applicable to dynamic problems in general.

    Notes: The solution of the large numbers of non-linear algebraic equations occurring in analyses of non-linear structural systems using finite differences or finite elements is time consuming even for the fastest computers. This paper presents a method of solution based on a Taylor's expansion technique which is an aid in the solution of such systems of equations.Examples presented show that in the finite element analysis of moderately non-linear structural systems the technique provides sufficiently accurate solutions. For more non-linear systems the technique provides excellent starting values for the Newton-Raphson method.

    Notes: This paper presents a general theory of finite elements. The concept of finite elements is cast in a general topological framework valid for spaces of finite dimension. It is shown that the idea of finite element models can be developed in higher-dimensional spaces, independent of specific co-ordinate systems, for any type of continuous abstract function defined on the space.     Notes: Surface displacements at the end of a semi-infinite, circular cylinder due to an axisymmetric ring of forces on the end are examined. The solution which has been found may then be used to find surface displacements for general axisymmetric loadings by convolution. The solution, in tabular form, is given as corrections to the counter-part half-space solution.The method of solution involves a three step superposition process. First, the displacement due to a ring of forces on a half-space is found by using the Boussinesq solution. Then, the excess tractions on the half-space, over that of the cylinder, are removed. This is done in two parts. The problem of an infinite cylinder with linearly varying pressure and shear over a short length of the lateral surface is solved by using Fourier integrals. This is used for the removal of the pressure and shear on the lateral surface of the cylinder by convolution. Next, the stresses at the mid-section of the infinite cylinder are removed. This is done by finding a set of boundary conditions for the end which yields zero tractions on the lateral surface. Then a series of these boundary conditions is used to approximate the tractions which must be removed.With the solution thus obtained, two sample problems are shown: 1. an elastic cylinder in contact with a half-space; 2. a rigid punch in contact with an elastic cylinder.

    Notes: A presentation is made of a rectangular plate bending element, the use of which corresponds to the finite difference method. The element stiffness matrix is arrived at by approximation of the strain energy of the element by suitably selected finite difference expressions which contain not only values in respect of the deflection, but also values of its first derivatives. On the basis of the connection found between the two methods, some changes are suggested in the finite difference method, which involve a more realistic way of considering the detailed distribution of the loading. The effect of these modifications is illustrated in two applications.

    Notes: Solution of plane elastic problems by piecewise linear approximation is outlined. This method is based upon Galerkin error distribution technique, which leads to simultaneous algebraic equations identical to those associated with the Finite Element Method.     Notes: A review is given of some mathematical programming methods suitable for optimization of structures needing matrix methods for analysis. An application is presented for elastic flat grillages made of straight orthogonal beams normally loaded. Empirical relationships are used to relate beam section properties so that each beam element has only one design variable. Optimization results are obtained by methods of stress-ratio, linear programming-cutting plane and usable-feasible gradient directions. A comparison of the efficiency of these methods is given for the grillage designs which are shown to have non-convex stress constraints and numerous relative optima. Instances are given of non-fully stressed global optima and fully stressed designs which are not local optima for cases with only stress constraints. Discussions include locating the global optima for grillage designs and extensions of the methods presented to other structural design problems.

    Notes: The paper presents a method for reducing the size of a vibration analysis by generating an inertia matrix referred to a limited set of ‘master’ vibrational freedoms. In designing a computer program to perform this mass condensation process, the emphasis has been on providing a two way interface between structural analysis and vibration analysis programs. The transformation from the full set of degrees of freedom to the master set makes extensive vibration calculations economically feasible giving solutions in terms of the amplitudes of the master freedoms. However, by reversing the transformation after completion of a vibration analysis, it is possible to restore the full set of freedoms, allowing automatic plotting of mode shapes in full detail, and re-entry to the structural analysis program for the purpose of computing dynamic stresses.A worked example is given. This is a realistic machine tool structure rather than the simple plates or beams which are usually used for illustration purposes. Natural frequencies and mode shapes, calculated with various degrees of condensation, are compared with those obtained by using all possible freedoms. A comparison is also made with an intuitive selection of ‘lumped masses’.

    Notes: The application of non-linear programming methods for the optimum design of statically indeterminate structures is discussed, with special emphasis on the design of elastic grillages loaded laterally and in plane. Some features of SUMT (sequential unconstrained minimization technique) are demonstrated by means of numerous examples of varying complexity. The Variable Metric method of search is discussed and compared to Powell's Direct Method. It is shown that non-convex sets of design variables are often encountered in structural problems of the grillage type. SUMT may still be used, but the choice of starting value and initial response factor decisively influences the chance of finding the global optimum.It is demonstrated that a fully stressed design may not necessarily correspond to the minimum weight design. Optimum design of grillages which are simultaneously subjected to lateral and in-plane loads may be performed efficiently by means of non-linear programming.

    Notes: A completely numerical method for steady state linear viscoelastic stress analysis is presented by means of the finite element approach. Numerical representations of the measured viscoelastic constitutive relations are used. This method is developed to obtain steady state solutions to mixed boundary value problems in which the character of the boundary conditions at a point changes with time. Such problems cannot be handled by direct application of the correspondence theorem. A numerical example of viscoelastic sheet rolling is presented along with an experimental verification of the solution by photo-viscoelastic observations.

    Notes: Several metallic orthopedic implants which failed in service and for which the medical histories are known are comprehensively examined using metallographic techniques and electron microprobe analyses to determine the causes of failure.     Notes: Tubing made of a polar plastic, polychlorotrifluoroethylene, was electrically polarized while heated. The zeta potential of tubes polarized negatively inside was consistently higher than that of unpolarized tubes. Tubes polarized positively had a much smaller and variable change in zeta potential. The change is apparently not related to the formation of electrets, but may be due to the orientation of pre-existing dipoles.

    Notes: The effect of phenol and certain phenyl compounds on the different stages of the blood coagulation and fibrinolytic mechanism is investigated. Phenol has a complex action on blood coagulation. In an optimal concentration of 0.3% it produced a definite, though suboptimal platelet factor 3 activity, evolved factor XII, accelerated thrombin-fibrinogen interaction, retarded clot retraction, enhanced the action of streptokinase on plasminogen, and inhibited plasmin. In addition, in the absence of factor XIII and calcium, it increased the solubility of fibrin in urea. Phenyl acetaldehyde, benzyl benzoate, and styrene (phenyl ethylene) accelerated stypven clotting time and thrombin-fibrinogen reaction. Unpolymerized styrene and phenyl ethanol specifically inhibited the action of plasma thromboplastin, formed in a system lacking in factor VIII or IX. A study is also reported on the changes responsible for the stabilization with phenol of brain thromboplastic activity. The action is not purely bactericidal, but is dependent on the presence of brain tissue particles in suspension. The contribution of platelets to plasma thromboplastin formation is examined in the light of these investigations, showing the ability of phosphorus-free compounds to supply a platelet-like activity.

    Notes: Adult albino rabbits were implanted with disks of 316 and 304 stainless steel in the subperiosteum of the anterior calvarium and the left mandible and in the left paravertebral and thigh muscles. The corresponding opposite sites were sham operated. The histopathological slides revealed that there was variation in thickness of the membrane in sections of both the paravertebral and thigh muscles; one side showing greater reaction than the other, which corresponded with the appearance of the implants. Sections of the calvarium revealed minimal bone and soft tissue reaction, although the implants showed corrosion compatible with soft tissue sites. The mandibular sections revealed extensive bone tissue reactions with minimum masseter muscle reactions, but the appearance of the implants due to corrosion was different than those of all other sites. Moderate eosinophilic infiltration was observed in sections containing deposits of corrosion products.

    Notes: Bioelectric charge transfer at the blood-wall interface may be a crucial factor affecting thrombosis on implant materials. A program of studies was conducted to determine the electrokinetic and other physical properties of a wide spectrum of materials including organic polymers, metals, inorganics, heterogeneous compounds, and animal tissue. From these tests, materials were selected for in-vivo cannulation experiments. In this manner, a search was made to find a correlation between surface charge characteristics and thrombosis on cardiovascular implant materials. It was found that materials exhibiting a substantial positive charge were prone to rapid thrombosis. No clear correlation can be stated, as yet, regarding the thrombogenecity of highly charged negative surfaces or slightly charged surfaces. However, many heterogeneous materials have exhibited prolonged in-vivo patency. It appears necessary to isolate surface effects from volumetric effects to seek the role of bioelectric charge transfer in blood thrombus formation.

Hydrolytic stability of this polymer is compared to that of a polyester urethan.     Notes: During the period covered by this investigation, toxicological studies were carried out in hamsters and rabbits. The results of injections, subcutaneously and in the buccal pouch of hamsters, indicated that the silicones used were nontoxic, non-absorbable, and well encapsulated by connective tissue elements with moderate tissue reaction. Further studies of various combinations of roomtemperature vulcanizing silastics indicated similar reactions when injected submucosally in the alveolar ridges and palate of rabbits. The injected materials remained in situ in the injected submucosal area and became well encapsulated by the proliferation of connective tissue and collagenous fibers. There was no evidence of macrophages or foreign body giant-cells in and around the proliferating connective tissue capsule. There were no macroscopic or microscopic signs of tumor formation. The encapsulated masses did not elicit bone resorption when localized adjacent to bone tissue.

    Notes: The effects of variations in composition and processing on the open-circuit electrode potentials measured as a function of time of amalgams made from silver-tin, silver-tin-copper, silver-tin-zinc, and silver-tin-copper-zinc alloys have been determined at 37°C in distilled water, in saline solution, as well as in acetate and citrate solutions buffered to pH values of 3, 7, and 10. The electrochemical potentials of the alloys measured as a function of time in neutral acetate solutions show anodic polarization from an initial cathodic potential value of approximately 0.1 v with respect to the standard hydrogen electrode to a constant potential level depending upon alloy composition. In neutral acetate solution the potential-time curves of silver-tin amalgams with 7:5 mercury to alloy ratio show a rapid anodic polarization from an initial anodic potential value of 0.28 v to a minimum value of 0.05 v at approximately 1 hr followed by a potential reversal to a plateau value of 0.40 v. For 7:5 amalgams in neutral acetate solutions increases in both the trituration time and in the severity of processing deformation cause a small increase in initial potential, with a decrease in anodic polarization, while an increase in amalgam age lowers both the initial potential value and the longer-time potential plateau. Increasing the mercury content of the amalgam to a 2:1 ratio decreases the rate of potential reversal from the minimum, whereas decreasing the ratio to 4:5 decreases the amount of depolarization from the minimum value. The effects of copper and zinc additions to silver-tin alloys are mixed. In neutral acetate solution the 7:5 ratio amalgams from silver-tin-copper alloys show anodic polarization from the same initial value as the silver-tin amalgams, followed by a region of decreased polarization. Both silver-tin-zinc and silver-tin-copper-zinc alloys exhibit a much higher initial anodic potential (0.78 v), and following the achievement of the potential plateau have a slow anodic polarization to the long-time potential value shown by silver-tin amalgam. Three commercial alloy amalgams were studied in neutral acetate solution. The two amalgams containing zinc have a potential behavior similar to that of the experimental zinc-containing amalgam; the non-zinc amalgam shows a rapid anodic polarization from an initial anodic potential value of 0.24 v, a minimum at 1 hr, and a reversal to the long-time potential values of 0.40 v shown by the zinc-containing amalgam.
