    Description: Interfacial solar steam generation is emerging as a promising technique for efficient desalination. Although increasing efforts have been made, challenges exist for achieving a balance among a plethora of performance indicators—for example, rapid evaporation, durability, low-cost deployment, and salt rejection. Here, we demonstrate that carbonized manure can convert 98% of sunlight into heat, and the strong capillarity of porous carbon fibers networks pumps sufficient water to evaporation interfaces. Salt diffusion within microchannels enables quick salt drainage to the bulk seawater to prevent salt accumulation. With these advantages, this biomass-derived evaporator is demonstrated to feature a high evaporation rate of 2.81 kg m−2 h−1 under 1 sun with broad robustness to acidity and alkalinity. These advantages, together with facial deployment, offer an approach for converting farm waste to energy with high efficiency and easy implementation, which is particularly well suited for developing regions.

    Description: Unicellular eukaryotes have been suggested as undergoing self-inflicted destruction. However, molecular details are sparse compared with the mechanisms of programmed/regulated cell death known for human cells and animal models. Here, we report a molecular cell death pathway in Saccharomyces cerevisiae leading to vacuole/lysosome membrane permeabilization. Following a transient cell death stimulus, yeast cells die slowly over several hours, consistent with an ongoing molecular dying process. A genome-wide screen for death-promoting factors identified all subunits of the AP-3 complex, a vesicle trafficking adapter known to transport and install newly synthesized proteins on the vacuole/lysosome membrane. To promote cell death, AP-3 requires its Arf1-GTPase-dependent vesicle trafficking function and the kinase Yck3, which is selectively transported to the vacuole membrane by AP-3. Video microscopy revealed a sequence of events where vacuole permeability precedes the loss of plasma membrane integrity. AP-3-dependent death appears to be conserved in the human pathogenic yeast Cryptococcus neoformans.

    Description: Most tropical corals live in symbiosis with Symbiodiniaceae algae whose photosynthetic production of oxygen (O2) may lead to excess O2 in the diffusive boundary layer (DBL) above the coral surface. When flow is low, cilia-induced mixing of the coral DBL is vital to remove excess O2 and prevent oxidative stress that may lead to coral bleaching and mortality. Here, we combined particle image velocimetry using O2-sensitive nanoparticles (sensPIV) with chlorophyll (Chla)-sensitive hyperspectral imaging to visualize the microscale distribution and dynamics of ciliary flows and O2 in the coral DBL in relation to the distribution of Symbiodiniaceae Chla in the tissue of the reef building coral, Porites lutea. Curiously, we found an inverse relation between O2 in the DBL and Chla in the underlying tissue, with patches of high O2 in the DBL above low Chla in the underlying tissue surrounding the polyp mouth areas and pockets of low O2 concentrations in the DBL above high Chla in the coenosarc tissue connecting neighboring polyps. The spatial segregation of Chla and O2 is related to ciliary-induced flows, causing a lateral redistribution of O2 in the DBL. In a 2D transport-reaction model of the coral DBL, we show that the enhanced O2 transport allocates parts of the O2 surplus to areas containing less chla, which minimizes oxidative stress. Cilary flows thus confer a spatially complex mass transfer in the coral DBL, which may play an important role in mitigating oxidative stress and bleaching in corals.

    Description: Cytoplasmic extracts from unfertilized Xenopus eggs have made important contributions to our understanding of microtubule dynamics, spindle assembly, and scaling. Until recently, these in vitro studies relied on the use of heterologous tubulin. This protocol allows for the purification of physiologically relevant Xenopus tubulins in milligram yield, which are a complex mixture of isoforms with various post-translational modifications. The protocol is applicable to any cell or tissue of interest. For complete details on the use and execution of this protocol, please refer to Hirst et al. (2020).

    Description: Dynamic microtubules are essential for many processes in the lives of eukaryotic cells. To study and understand the mechanisms of microtubule dynamics and regulation, in vitro reconstitution with purified components has proven a vital approach. Imaging microtubule dynamics can be instructive for a given species, isoform composition, or biochemical modification. Here, we describe two methods that visualize microtubule dynamics at high speed and high contrast: (1) total internal reflection fluorescence microscopy and (2) label-free interference reflection microscopy.

    Description: Cell-free extract derived from the eggs of the African clawed frog Xenopus laevis is a well-established model system that has been used historically in bulk aliquots. Here, we describe a microfluidic approach for isolating discrete, biologically relevant volumes of cell-free extract, with more expansive and precise control of extract shape compared with extract-oil emulsions. This approach is useful for investigating the mechanics of intracellular processes affected by cell geometry or cytoplasmic volume, including organelle scaling and positioning mechanisms.     Description: After fertilization, microtubule (MT) sperm asters undergo long-range migration to accurately position pronuclei. Due to the large sizes of zygotes, the forces driving aster migration are considered to be from pulling on the astral MTs by dynein, with no significant contribution from pushing forces. Here, we re-investigate the forces responsible for sperm aster centration in sea urchin zygotes. Our quantifications of aster geometry and MT density preclude a pulling mechanism. Manipulation of aster radial lengths and growth rates, combined with quantitative tracking of aster migration dynamics, indicates that aster migration is equal to the length of rear aster radii, supporting a pushing model for centration. We find that dynein inhibition causes an increase in aster migration rates. Finally, ablation of rear astral MTs halts migration, whereas front and side ablations do not. Collectively, our data indicate that a pushing mechanism can drive the migration of asters in a large cell type.

    Description: In large areas of the world's oceans, there is a relationship between the mass flux of particulate matter and the unsupported 231Pa/230Th (xs231Pa/xs230Th) activity ratio of recent sediments. This observation forms the basis for using the xs231Pa/xs230Th ratio as a proxy for past changes in export productivity. However, a simple relationship between xs231Pa/xs 230Th ratio and particle flux requires that the water residence time in an ocean basin is far in excess of the scavenging residence time of 231Pa, and that the composition of sinking particles maintains a strong preference for the adsorption of 230Th over 231Pa with a constant 230Th/231Pa fractionation factor (F). The best correlation between xs231Pa/xs230Th ratio and mass flux is found in the Pacific Ocean. In the Atlantic, the contrast in the xs231Pa/xs230Th ratios between open ocean (low flux regions) and ocean margins (high flux regions) is much less pronounced due to the shorter residence time of deep water, resulting in less effective boundary scavenging of 231Pa. In the Southern Ocean, south of the Polar Front, there is no more a simple relationship between xs231Pa/xs230Th and particle flux. This is a result of a southward decrease in F, probably reflecting the increased opal content of sinking particles. Opal does not fractionate 231Pa and 230Th significantly. This lack of fractionation results in high xs231Pa/xs230Th ratios in opal-dominated regions, even in areas of very low particle fluxes such as the Weddell Sea. The xs231Pa/xs230Th ratio can therefore only be used as a paleoproductivity proxy if, in the time interval of interest, changes in the basin ventilation rate and differential scavenging of both radionuclides due to changes in the chemical composition of particulate matter can be excluded.

    Description: Glacial landforms in northern Russia, from the Timan Ridge in the west to the east of the Urals, have been mapped by aerial photographs and satellite images supported by field observations. An east-west trending belt of fresh hummock-and-lake glaciokarst landscapes has been traced to the north of 67°N. The southern boundary of these landscapes is called the Markhida Line, which is interpreted as a nearly synchronous limit of the last ice sheet that affected this region. The hummocky landscapes are subdivided into three types according to the stage of postglacial modification: Markhida, Harbei and Halmer. The Halmer landscape on the Uralian piedmont in the east is the freshest, whereas the westernmost Markhida landscape is more eroded. The west- east gradient in morphology is considered to be a result of the time-transgressive melting of stagnant glacier ice and of the underlying permafrost. The pattern of ice-pushed ridges and other directional features reflects a dominant ice flow direction from the Kara Sea shelf. Traces of ice movement from the central Barents Sea are only discernible in the Pechora River left bank area west of 50°E. In the Polar Urals the horseshoe-shaped end moraines at altitudes of up to 560 m a.s.l. reflect ice movement up-valley from the Kara Ice Sheet, indicating the absence of a contemporaneous ice dome in the mountains. The Markhida moraines, superimposed onto the Eemian strata, represent the maximum ice sheet extent in the western part of the Pechora Basin during the Weichselian. The Markhida Line truncates the huge arcs of the Laya-Adzva and Rogovaya ice-pushed ridges protruding to the south. The latter moraines therefore reflect an older ice advance, probably also of Weichselian age. Still farther south, fluvially dissected morainic plateaus without lakes are of pre-Eemian age, because they plunge northwards under marine Eemian sediments. Shorelines of the large ice-dammed Lake Komi, identified between 90 and 110 m a.s.l. in the areas south of the Markhida Line, are radiocarbon dated to be older than 45 ka. The shorelines, incised into the Laya-Adzva moraines, morphologically interfinger with the Markhida moraines, indicating that the last ice advance onto the Russian mainland reached the Markhida Line during the Middle or Early Weichselian, before 45 ka ago.

    Description: The marine-lacustrine transition (isolation contact) in sediment cores from eight lake basins situated 13.5–72 m a.s.l., in the Norwegian-Russian border area north of Nikel, northwest Russia, was identified based on lithological and diatom analysis, radiocarbon dated, and used to construct a relative sea-level (RSL) curve for the Holocene. All the lakes except one (interpreted as having an unconformable slumped transition) show a regressive I-II-III (marine-transitional-lacustrine) facies succession, indicating a postglacial history of continuous emergence. The RSL curve shows rapid emergence between 10 000 and 8000 BP, very slow emergence between 7000 and 5000 BP, increased rate of emergence between 4500 and 4000 BP, and a moderate rate of emergence after 3500 BP. The low rate of emergence around 6000 BP correlates with the Tapes transgression of more coastal regions, but corresponding sea level, at 25–26 ma. present s.l., lies 5–10 m lower than the elevation predicted based on existing isobase maps for the region. The discrepancy suggests a need for further work in order to more rigorously define and map the Tapes transgression and associated shoreline complex in the northern Fennoscandian-Kola region.
