Notes: Abstract Peroxidase, esterase, and acid phosphatase isozymes of environmentally induced L and S genotrophs, nuclear DNA reversion types, and the original plastic (Pl) type of the flax variety Stormont Cirrus have been compared by polyacrylamide gel electrophoresis. Differences were observed in particular genotrophs in terms of peroxidase and acid phosphatase isozymes but not in the esterase isozymes. The isozyme pattern of a particular line was not correlated with the nuclear DNA amount. The relationship between the isozyme pattern and the phenotypes of the lines in which they are expressed is discussed.

Notes: Abstract The ontogeny of lactate dehydrogenase (LDH) isozymes was examined in avian hybrids and compared with the isozyme patterns of the parental species. Hybrids were obtained by crossing female Japanese quail (Coturnix coturnix japonica) with male domestic chickens (Gallus gallus domesticus). By use of starch gel electrophoresis and an enzyme-specific stain, traces of embryonic paternally derived LDH were detected in unincubated hybrid eggs. It was concluded that the embryonic genes coding for the B subunits of LDH are activated during the hours between fertilization and oviposition. In early blastoderms, a great excess of maternally stored LDH is present. In the hybrid, the predominantly maternal pattern of isozymes shifts during embryogenesis to a predominantly paternal pattern. This was considered evidence for differential allelic regulation of LDH inactivation. A progressive trend toward the establishment of the adult distribution of isozymes in various tissues was also observed in the hybrid and quail, and found to be similar to chicken LDH isozyme ontogeny.

Notes: Abstract General criteria are described for selecting internal standards for electrophoretic gels. Gel sieving behavior is used to characterize the size and charge of enzymes of interest and of putative standard proteins. For a variety of enzymes of the butterfly Colias meadii, hemoglobin and ferritin are shown to be effective standards.

Notes: Abstract Isozyme patterns may be analyzed quantitatively, without the use of a densitometer, by performing serial twofold dilutions of a sample to a visual end point. The specific activity (S) of a given dehydrogenase isozyme can be assessed in the presence of other isozymes catalyzing the same reaction, by (1) determining the isozyme titer (T) (defined as mg protein/ml in the last visible band) and (2) applying the formula S=K/T, where K is 1.6×10−3 units/ml in the last visible band. The units/ml (U) in the starting material can be calculated from the equation U=K (2) n−1, where n is the number of the slot producing the last visible band.

Notes: Abstract Allelic variation at the Est-II locus was investigated in 22 natural populations of Heliothis zea from maize, separated spatially and temporally. Of the four alleles observed, Est-II b was the predominant one in every population. The relative frequency of Est-II b ranged from 0.57 to 0.85, with a mean of 0.72±0.06, and proved to be remarkably stable in space and time. Temporal changes in Est-II allele frequencies were observed at two of the sampling localities; however, principal component analysis and multiple correlation of genetic and environmental components have provided persuasive evidence that abiotic environmental or geographic variation is not related to differences in Est-II allele frequencies. Possible factors responsible for the maintenance of this polymorphism are discussed and include possible effects of the chemical composition of host plants and pesticides.

Notes: Abstract Effects of thyroxine on incorporation of l-serine-C14 into four carbonic anhydrase isozymes (CA II, CA Ia, CA Ib, CA Ic) and hemoglobin were quantified in reticulocytes of Macaca nemestrina in vitro. Response to thyroxine differed significantly between CA Ia and two allelic variants (CA Ib and CA Ic) and the nonallelic isozyme (CA II). The effects of thyroxine on serine incorporation into hemoglobin and three of the carbonic anhydrase isozymes were shown to be nonlinear with thyroxine concentration.

Notes: Abstract The chromosome numbers of several Mediterranean and Central European species from the genusCentaurea L. have been studied. The material was collected in natural habitats, exceptC. thracica andC. maxima. The numbers forC. cineraria (2n = 36; tetraploid!),C. subtilis (2n = 22),C. sonchifolia (2n = 44),C. sphaerocephala (2n = 44),C. napifolia (2n = 22),C. achaia (2n = 22),C. thracica (2n = 18), andC. maxima (2n = 28) are reported for the first time. For the following species earlier counts are confirmed:C. cineraria (diploid, 2n = 18),C. maculosa (2n = 18),C. rhenana (2n = 18),C. triumfettii (2n = 22 + 0−1 B; first report of accessory chromosome for this species),C. scabiosa (2n = 20 + 0−3 B),C. crassifolia (2n = 30). B-chromosomes are reported for 3 populations ofCentaurea scabiosa from Berlin; they lack in 2 populations from Bayern and Hessen.

Notes: Abstract Optimal values for the quantity of proteins in samples for ultramicroelectrophoretic fractionation in polyacrylamide gel and capillary tubes of different diameters enabling clear and highly reproducible results to be obtained by electrophoresis were established experimentally. A simple method of calculating the necessary sample volumes for protein solutions of different concentrations and capillary tubes of different diameters by means of a table and formula is suggested.

Notes: Abstract The lactate dehydrogenase isozyme spectrum was investigated in the mammary glands, lungs, spontaneous mammary gland tumors, and metastases of tumors in the lungs of female mice of the high-cancer strain C3H previously having given birth to young. Differences in the isozyme spectrum of the metastases and tumors were reflected in the larger fractions migrating to the anode in the metastatic nodes. The presence of a metastasis in the lung was accompanied by a cathodal shift in the isozyme spectrum of the lung tissues.

Notes: Abstract Changes in the activity of malate dehydrogenase (MD) and its isozymes in the blood serum and heart muscle of rats were investigated during hypoxia and under the influence of ATP and cocarboxylase. An additional cathodic isozyme (MD4) appeared in the blood serum during hypoxia. Under the influence of ATP and cocarboxylase the metabolic changes in the blood and myocardium induced by hypoxia did not correlate fully.
