Description: Bacterial biofilm is a complex structure with diverse bacterial cells in a highly organized and ordered group within a matrix of extracellular polymeric substances they produce. Microbes attach to surfaces to develop biofilms, a sophisticated process regulated by factors such as nutritional status and biotic/abiotic surface features. An established biofilm structure mainly comprises bacterial cells, proteins, nucleic acid, and exo-polysaccharides that are extracellular macromolecules excreted as tightly bound layers in microbes, providing a perfect niche for bacteria to exchange genetic material between cells. In addition, bacterial cells in the matrix also communicate via quorum sensing, which greatly impacts biofilm processes. Under clinical circumstances, bacterial biofilm shows great resistance to antibiotics, disinfectants, and body defense systems, making it difficult for clinicians to eradicate and facilitate many infectious disease processes, leading to chronic infections of patients with long-term hospitalization and high mortality rates. Therefore, it is very important to understand the recent advances in forming, regulating, and eradicating biofilms in human infections to better prevent, control, and treat biofilm infections in humans. Written by an international team of basic and clinical researchers, the chapters of this book provide novel insights and advanced knowledge for life science researchers, clinical researchers, doctors, and other interested readers on some of the latest developments in biofilms.

Description: Modern medicine is impossible without antibiotics, but global antibiotic usage has led to the development of increasing numbers of multi-drug resistant (MDR) bacteria. Thus, we still have problems with infectious disease treatment despite an arsenal of antibiotics. This has forced researchers to develop new drugs that will be effective against resistant bacteria. Some of these prospective molecules are antimicrobial peptides (AMPs), which are an important component of the innate immune system of various organisms in nature. Currently, more than 3,000 AMPs have been reported with different activities against different bacterial species including resistant phenotype bacteria. AMPs display remarkable structural and functional diversity that is not completely understood. As such, this book presents a comprehensive overview of AMPs and their mechanism of action against MDR bacteria.

Description: Oxidative stress is a major contributor to the etiology of chronic disorders like cancer, diabetes, neurodegenerative diseases, and cardiovascular diseases. Long-term exposure to elevated levels of pro-oxidant substances can lead to structural damage in mitochondrial DNA as well as functional changes in a number of enzymes and cellular components, which can lead to abnormalities in gene expression. Modern lifestyles, which include eating processed food, exposure to a variety of chemicals, and not exercising, are significant factors in the development of oxidative stress. However, the ability of medicinal plants with antioxidant capabilities to cure or prevent a number of human illnesses in which oxidative stress appears to be a contributing factor has been demonstrated. A growing body of research links free radicals to the etiology of many diseases, supporting the use of antioxidants as a promising therapeutic strategy for the management of pathologies caused by free radicals. Despite these remarkable advances, there is still much to learn about the relationship between free radicals and antioxidants. Understanding the principles behind pathological and physiological disorders caused by free radicals is crucial. Importance of Oxidative Stress and Antioxidant System in Health and Disease contributes to understanding the fundamental principles of oxidative stress and the effects of antioxidants on disease and health.

Description: The study of staphylococci has a long way to go, due to the great significance both for humans and for dairy or companion animals of their large number of virulence factors. Their resistance to antibiotics makes these microorganisms become highly pathogenic species that can cause infections ranging from mild to fatal. Staphylococci, especially S. aureus and strains resistant to methicillin (MRSA), are among the most studied microorganisms worldwide. This book covers recent advances in the study of staphylococci. The book is divided into four sections that cover four important events for the study and combat of staphylococci: colonization, epidemiology and pathogenesis, diagnosis, and new treatments. Staphylococci can present a broad spectrum of resistance to antibiotics, which is why their elimination has become difficult. The final section of the book is devoted to new compounds for the fight against staphylococci. We hope that the information contained in this book will be useful for the study and investigation of these medically important microorganisms.

Description: More than 2,500 serotypes of Salmonella exist. However, only some of these serotypes have been frequently associated with food-borne illnesses. Salmonella is the second most dominant bacterial cause of food-borne gastroenteritis worldwide. Often, most people who suffer from Salmonella infections have temporary gastroenteritis, which usually does not require treatment. However, when infection becomes invasive, antimicrobial treatment is mandatory. Symptoms generally occur 8 to 72 hours after ingestion of the pathogen and can last 3 to 5 days. Children, the elderly, and immunocompromised individuals are the most susceptible to salmonellosis infections. The annual economic cost due to food-borne Salmonella infections in the United States alone is estimated at $2.4 billion, with an estimated 1.4 million cases of salmonellosis and more than 500 deaths annually. This book contains nineteen chapters which cover a range of different topics, such as the role of foods in Salmonella infections, food-borne outbreaks caused by Salmonella, biofilm formation, antimicrobial drug resistance of Salmonella isolates, methods for controlling Salmonella in food, and Salmonella isolation and identification methods.

Description: Infection is a common clinical condition that may cause local inflammation but, in some cases, can lead to systemic inflammation, with sepsis and organ dysfunction. Septic shock is a condition of inadequate tissue perfusion and cellular use of oxygen due to the cytotoxic action of bacterial toxins. There is no relationship between the pathological characteristics and the severity of the primary septic outbreak and the development of septic shock, and the time that elapses until the start of the shock is not predictable. Thus, knowledge of the pathophysiology of septic shock is fundamental for treatment. This book presents a comprehensive overview of infectious agents and their therapeutic control, pathological conditions with infective etiology such as diabetic foot osteomyelitis and infections in neurosurgery, and the pathophysiology, diagnosis, and management of sepsis.

Description: This book serves as a comprehensive survey of the impact of vitamin K2 on cellular functions and organ systems, indicating that vitamin K2 plays an important role in the differentiation/preservation of various cell phenotypes and as a stimulator and/or mediator of interorgan cross talk. Vitamin K2 binds to the transcription factor SXR/PXR, thus acting like a hormone (very much in the same manner as vitamin A and vitamin D). Therefore, vitamin K2 affects a multitude of organ systems, and it is reckoned to be one positive factor in bringing about "longevity" to the human body, e.g., supporting the functions/health of different organ systems, as well as correcting the functioning or even "curing" ailments striking several organs in our body. Vitamin K2 - Vital for Health and Wellbeing has been produced and distributed through the support from Kappa Bioscience, Norway.

Description: Protein phosphorylation reactions are carried out in a cell by protein kinases, which predominantly use ATP as a phosphate donor that is transferred and covalently bound to an amino acid on a substrate protein. Protein phosphorylation was discovered in 1954 by Edmond Fischer who shared the Nobel Prize in Medicine or Physiology in 1992 with Edwin Krebs. There are so many kinases that one was called "Just Another Kinase" for JAK kinase. Their counterpart is protein phosphatases that remove phosphates from phosphorylated proteins. Kinases and phosphatases act as switches in the cell that activates or inactivates protein functions. These reactions are reversible; the cell can quickly react to a situation but can then go back to its initial state.

Description: The presence of flavonoids such as chalcones, flavones, flavonols, anthocyanins, and proanthocyanidins is ubiquitous throughout the plant kingdom. It was previously believed that the oldest plants that produced flavonoids were liverworts and mosses. However, recent discoveries have found genes encoding enzymes in the shikimate pathway responsible for flavonoid biosynthesis in a variety of plants including algae, liverworts, mosses, lycophytes, ferns and horsetails, gymnosperms, and angiosperms. These genes include the first two enzymes for flavonoid biosynthesis, chalcone synthase and chalcone isomerase. Furthermore, recent research has found flavones, isoflavones, and flavonols in microalgae from five distinct evolutionary lineages(Cyanobacteria, Rhodophyta, Chlorophyta, Haptophyta, and Ochrophyta) using ultra-high performance liquid chromatography with tandem mass spectrometry. This implies that plants may have developed the capacity to make flavonoids earlier than previously believed. Flavonoids in the earliest plants that produced them were believed to have acted as protection against UV radiation and control of plant hormone activity. As plants evolved, these roles diversified, and the study of flavonoids has revealed how plants developed the capacity to produce a wide array of specialized metabolites and then evolved the metabolic pathways required to manufacture them. This book delves into the structural diversity of flavonoids found throughout the plant kingdom, the evolution of flavonoid biosynthesis genes in plants, and the molecular interaction of flavonoids with growth hormones. The metabolic routes of flavonoids are thoroughly investigated using both biochemical and molecular biology methods, and the book elucidates the links between genes/proteins and metabolites, as well as between metabolites. The book also explores the interplay between flavonoid metabolism and external environmental factors that are subject to continuous change.

Description: Protein aggregation causes malfunction in several biochemical processes. Genetic and spontaneous formations of these transmissible spongiform encephalopathies are fatal to humans and animals. Conformational change of normal form of the protein to misfolded form causes its accumulation. The misfolded infectious protein agent forms the pathogenesis of the disease. This book presents pathology of the disease along with current knowledge of the structure-activity mechanism in the first two sections. Dyshomeostasis of metals is implicated in the pathogenesis of prions, and this influence is discussed further to understand the prion mechanism. Genetic resistance and immunobiology of the disease are elaborated in the following section. Finally, a computational study on the dynamics of the prion propagation provides a structural basis of the mechanism.

Description: Proteins are indispensable players in virtually all biological events. The functions of proteins are coordinated through intricate regulatory networks of transient protein-protein interactions (PPIs). To predict and/or study PPIs, a wide variety of techniques have been developed over the last several decades. Many in vitro and in vivo assays have been implemented to explore the mechanism of these ubiquitous interactions. However, despite significant advances in these experimental approaches, many limitations exist such as false-positives/false-negatives, difficulty in obtaining crystal structures of proteins, challenges in the detection of transient PPI, among others. To overcome these limitations, many computational approaches have been developed which are becoming increasingly widely used to facilitate the investigation of PPIs. This book has gathered an ensemble of experts in the field, in 22 chapters, which have been broadly categorized into Computational Approaches, Experimental Approaches, and Others.

Description: Toxoplasma gondii was first identified more than 100 years ago in the tissues of birds and mammals. Although toxoplasmosis is important all over the world, its approaches to diagnostic strategies considerably differ among countries. Its wide distribution may be attributed to complex transmission patterns and parasite coevolution with multiple hosts. Although T. gondii infections of immunocompetent people are generally considered asymptomatic, infections in immunocompromised individuals, such as those with AIDS or organ transplant recipients, can result in severe consequences. This book, composed of a series of articles, including effective diagnosis of laboratory in toxoplasma infections, congenital toxoplasmosis, relationship between toxoplasmosis and public health genomics, prevalence, genetic diversity of toxoplasmosis, and microparticle vaccines against Toxoplasma gondii by authors from all over the world, presents a wide open point of view for toxoplasmosis.

Description: Even 110 years after its discovery, Chagas disease is still an important public health problem, especially in Latin America where it is endemic. The limited effect of current treatments in the chronic phase of the disease, as well as the lack of information about disease progression, are crucial challenges that justify the intense research on this illness. The interaction between the etiological agent Trypanosoma cruzi and its invertebrate and vertebrate hosts involves the regulation of the parasite virulence and host immune responses. T. cruzi presents mechanisms to evade host defenses, including resistance to oxidative species and vesicles secretion, among many others. In this context, a better understanding of the biochemical and molecular features of the parasite’s success inside its hosts could contribute to the development of novel anti-T. cruzi strategies such as prototypes of drugs and vaccines. This book discusses Chagas disease and its etiological agent, including information on relevant clinical aspects such as diagnosis, treatment, biomarkers, and so on. It also presents information about cellular, molecular, and biochemical characteristics of the parasite-host interactions.

Description: Although there are thousands of parasites all over the world, Schistosoma spp. are unique in terms of their morphology. Although they have separate bodies as “male” and “female,” they live in such a way that the female resides within the male. Schistosoma spp., classified in the family of trematodes, has about twenty members, five of which cause a disease called schistosomiasis (or bilharzia) in humans. This disease causes a variety of symptoms, ranging from simple skin lesions to a dramatic clinical scenario of bladder carcinoma. Although there are many different diagnostic and therapeutic methods, schistosomiasis is still an important threat to humans and a significant challenge for healthcare professionals. This book discusses the morphology, geographic distribution, and evolution of Schistosoma spp., as well as the diagnosis, treatment, and prevention of schistosomiasis.

Description: The first roundworm identified in humans was Ascaris, described by Linnaeus in 1758. Rudolphi’s 1808 classification was the first to describe the Nematoidea. With the development of new molecular techniques, classification and phylogenetic research have also frequently changed; in 2019 a new classification, the phylum Nematoda, was created based on evolutionary relationships, developmental and morphological features and recent molecular evidence. Nematodes, which include hookworms, whipworms, threadworms, and soil-borne worms, are one of the oldest disease-causing creatures, dating back to Ancient Greek, Roman and Mesopotamian civilizations. Most commonly affecting children and women, the disease causes developmental disorders, cognitive impairment and death, with symptoms primarily affecting the organs they inhabit. In this book, the historical background, biology, transmission routes and geographical distribution of some roundworms affecting human health are discussed.

Description: DNA damage response (DDR) and lesion repair are vital processes ensuring genome integrity through various pathways depending mainly on the nature of DNA injury and cell cycle stage. DDR is finely regulated at many levels in co-ordination with other ongoing processes as is genome replication and cell cycle progression. Posttranslational modifications (PTMs), affecting both protein-protein and protein-DNA interactions, play a crucial role in finely tuning all processes involved in the restoration of genome lesions. Regarding damaged chromatin, PTMs serve in many cases as recruitment platforms for DNA repair mechanisms by facilitating binding sites or regulating interactions between involved proteins. Ubiquitination, the addition of ubiquitin moieties on a target protein, apart from controlling protein availability through degradation, is also involved, together with partner small ubiquitin-like modifier (SUMO), in controlling many pathways involved in DDR by modifying the structure-function relationship and thus interacting with partner molecules. The aim of this book is to cover a broad spectrum of current topics in ubiquitination and to a lesser extent SUMOylation involvement in regulation of DDR and repair in health and disease. This book is intended for pre- and postgraduate students and young scientists in this field. Members of both academic and research institutions, actively involved in the field, have described their current understanding of major mechanisms involved, highlighted key events, described ongoing applications in both developmental diseases and cancer and provided hints for future potential applications.

Description: Proteomics was thought to be a natural extension after the field of genomics has deposited significant amount of data. However, simply taking a straight verbatim approach to catalog all proteins in all tissues of different organisms is not viable. Researchers may need to focus on the perspectives of proteomics that are essential to the functional outcome of the cells. In Integrative Proteomics, expert researchers contribute both historical perspectives, new developments in sample preparation, gel-based and non-gel-based protein separation and identification using mass spectrometry. Substantial chapters are describing studies of the sub-proteomes such as phosphoproteome or glycoproteomes which are directly related to functional outcomes of the cells. Structural proteomics related to pharmaceutics development is also a perspective of the essence. Bioinformatics tools that can mine proteomics data and lead to pathway analyses become an integral part of proteomics. Integrative proteomics covers both look-backs and look-outs of proteomics. It is an ideal reference for students, new researchers, and experienced scientists who want to get an overview or insights into new development of the proteomics field.

Description: Mycobacterium tuberculosis, as recent investigations demonstrate, has a complex signaling expression, which allows its close interaction with the environment and one of its most renowned properties: the ability to persist for long periods of time under a non-replicative status. Although this skill is well characterized in other bacteria, the intrinsically very slow growth rate of Mycobium tuberculosis, together with a very thick and complex cell wall, makes this pathogen specially adapted to the stress that could be generated by the host against them. In this book, different aspects of these properties are displayed by specialists in the field.

Description: Herpesviruses are unique among viruses as they encode for a complex self-regulatory system, aggressively invade and persist in the host, evade immune defense, alter all regulatory mechanisms of the macroorganism and modify the replication of heterologous viruses. Environmental factors influence these unconventional relationships. Consequently, a single herpesvirus species can be attributed to a wide range of diseases as etiological agents or cofactors. This book is intended to give an overview on selected clinical hot topics: herpes simplex virus encephalitis, persistent infection in the gingiva, thymidine kinase gene expression causing male infertility, and pharmaceutical reactivation of Epstein-Barr virus for oncolysis. Immune evasion mechanisms and new ways to formulate vaccines are exhaustively reviewed. Finally, a surprise: bovine herpesviruses could serve as models to study the pathomechanism of herpesviruses.

Description: Epstein-Barr virus (EBV) is a fascinating microorganism, as a “double-facetted” viral agent. After primary infection, it can persist throughout a person’s lifetime in a latent form, from which it can reactivate following specific stimuli (i.e., immunodepression). Unlike other herpesviruses, EBV reactivates a countless number of times with such a high replication rate that it is unable to be controlled by conventional anti-herpesvirus drugs. Moreover, for various reasons, no vaccine is currently available in the market. This book presents a comprehensive overview of EBV, including information on its potential for oncogenic activity, its various isolates, and possible vaccine candidates.
