    Beschreibung: The most common quorum sensing (QS) system in Gram-negative bacteria occurs via N-acyl homoserine lactone (AHLs) signals. An archetypical system consists of a LuxI-family protein synthesizing the AHL signal which binds at quorum concentrations to the cognate LuxR-family transcription factors which then control gene expression by binding to specific sequences in target gene promoters. QS LuxR-family proteins are approximately 250 amino acids long and made up of two domains; at the N-terminus there is an autoinducer-binding domain whereas the C-terminus contains a DNA-binding helix-turn-helix (HTH) domain. QS LuxRs display surprisingly low similarities (18-25%) even if they respond to structurally similar AHLs. 95% of LuxRs share 9 highly conserved amino acid residues; six of these are hydrophobic or aromatic and form the cavity of the AHL-binding domain and the remaining three are in the HTH domain. With only very few exceptions, the luxI/R cognate genes of AHL QS systems are located adjacent to each other. The sequencing of many bacterial genomes has revealed that many proteobacteria also possess LuxRs that do not have a cognate LuxI protein associated with them. These LuxRs have been called orphans and more recently solos. LuxR solos are widespread in proteobacterial species that possess a canonical complete AHL QS system as well as in species that do not. In many cases more than one LuxR solo is present in a bacterial genome. Scientists are beginning to investigate these solos. Are solos responding to AHL signals? If present in a bacterium which possesses a canonical AHL QS system are solos an integral part of the regulatory circuit? Are LuxR solos eavesdropping on AHLs produced by neighboring bacteria? Have they evolved to respond to different signals instead of AHLs, and are these signals endogenously produced or exogenously provided? Are they involved in interkingdom signaling by responding to eukaryotic signals? Recent studies have revealed that LuxR solos are involved in several mechanisms of cell-cell communication in bacteria implicating them in bacterial intraspecies and interspecies communication as well as in interkingdom signaling by responding to molecules produced by eukaryotes. LuxR solos are likely to become major players in signaling since they are widespread among proteobacterial genomes and because initial studies highlight their different roles in bacterial communication. This Research Topic allows scientists studying or interested in LuxR solos to report their data and/or express their hypotheses and thoughts on this important and currently understudied family of signaling proteins.

    Beschreibung: The cellular prion protein PrPC is a ubiquitous GPI-anchored protein. While PrPC has been the focus of intense research for its involvement in a group of neurodegenerative disorders known as transmissible spongiform encephalopathies (TSE), much less attention has been devoted to its physiological function. This notably relates to the lack of obvious abnormalities of mice, goat or cattle lacking PrPC. This apparently normal phenotype in these PrPC-deficient animals however contrasts with the very high degree of conservation of the prion protein gene (Prnp) in mammalian species (over 80%), and the presence of genes with similarities to Prnp in birds, reptiles, amphibians and fish. This high conservation together with its ubiquitous expression, - albeit at highest levels in the brain-, suggest that PrPC has major physiological functions. Dissecting PrPC function is further complicated by the occurrence, in mammals, of two potentially partially redundant homologues, Doppel, and Shadoo. The biological overlaps between members of the prion protein family are still under investigation and much debated. Similarly, although in vitro analyses have suggested various functions for PrPC, notably in cell death and survival processes, some have yielded conflicting results and/or discrepancies with in vivo studies. This Research Topic brings together the accumulated knowledge regarding the biological roles of the prion protein family, from the animal to the molecular scale.

    Beschreibung: Diacylglycerol kinases (DGKs) phosphorylate diacylglycerol (DG), catalyzing its conversion into phosphatidic acid (PA). This reaction attenuates membrane DG levels, limiting the localization/activation of signaling proteins that bind this lipid. Initially recognized as modulators of classical and novel PKC family members, the function of the DGK has further expanded with the identification of novel DG effectors including Ras Guanyl nucleotide-releasing proteins (RasGRP) and chimaerin Rac GTPases. The product of the DGK reaction, PA, is also a signaling lipid that mediates activation of multiple proteins including the mammalian target of rapamycin (mTOR). The DGK pathway thus modulates two lipids with important signaling properties that are also key intermediates in lipid metabolism and membrane trafficking. The DGK family in eukaryotes comprises 10 different members grouped into five different subfamilies characterized by the presence of particular regulatory motifs. These regions allow the different DGK isoforms to establish specific complexes and/or to be recruited to specific subcellular compartments. The subtle regulation of DG and PA catalyzed byspecific DGKs is sensed by a restricted set of molecules, providing the means for spatio-temporal regulation of signals in highly specialized cell systems. In the recent years, multiple studies have unveiled the functions of specific isoforms, their mechanisms of regulation and their participation in different pathways leading to and from DG and PA. Animal models have greatly helped to understand the specialized contribution of DGK mediated signals, particularly in the immune and central nervous systems. Mice deficient for individual DGK isoforms show defects in T and B cell functions, dendritic spine maintenance, osteoclast and mechanical-induced skeletal muscle formation. Studies in flies and worms link DGK mediated DAG metabolism with mTOR- mediated regulation of lifespan and stress responses. In plants DGK mediated PA formation contributes to plant responses to environmental signals. Aberrant DGK function has been recently associated with pathological states, an expected consequence of the essential role of these enzymes in the regulation of multiple tissue and systemic functions. DGK mutations/deletions have been related to human diseases including diabetes, atypical hemolytic-uremic syndrome, Parkinson disease and bipolar disorders. On the contrary DGK upregulation emerges as a non-oncogenic addition of certain tumors and represents one of the main mechanism by which cancer evades the immune attack. As a result, the DGK field emerges an exciting new area of research with important therapeutic potential.

    Beschreibung: The formation of a complex multicellular organism from a single cell is one of the most amazing processes of biology. Embryonic development is characterised by the careful regulation of cellular behaviours such that cells proliferate, migrate, differentiate and form tissues at the correct place and time. These processes are genetically controlled and depend both on the history of cells, their lineage, and on the activities of signalling pathways, which coordinate the cell interactions leading to organogenesis. The aim of the Frontiers research topic “Signalling pathways in embryonic development” has been to provide a forum for experts in cell and developmental biology to share recent advances in the field of signalling during embryonic development. Sixteen articles in a variety of formats are united in this Topic, offering a valuable collection for researchers looking for an update in the knowledge of signalling pathways operating during embryogenesis. The works, focused mainly on vertebrates, explore different aspects of this theme from cell communication to organ formation and have implications for areas as distant as evolution or pathology. Understanding developmental signalling pathways is important for several reasons. It gives us information about basic mechanisms of cell function and interactions needed for morphogenesis and organogenesis. It uncovers the basis of congenital malformations, since errors at any step of cell signalling during development are a major cause of defects. This fundamental insight gives us clues to understand the mechanisms operating in evolution that explain diversity in form and function.     Beschreibung: Together with the microfilament, microtubule and intermediate-filament networks, septins constitute an integral part of the eukaryotic cytoskeleton. Historically identified as proteins critical for septum formation in the budding yeast Saccharomyces cerevisiae, septin family GTPases are expressed and participate in the process of cytokinesis in most eukaryotes except higher plants. More than a dozen septin genes in mammals, together with various splice variants displaying tissue-specific expression patterns and flexible hetero-polymeric higher-order assembly achieve an unfathomable complexity superior to the other cytoskeletal components. Even though the initial studies in the septin field was restricted to their evolutionarily conserved role in cell division, strong expression of septins in the non-dividing cells of the brain generated great interest in understanding their role in neuronal morphogenesis and other aspects of cellular function. On one hand, recent developments indicate complex non-canonical roles for septins in diverse processes ranging from neuronal development to immune response and calcium signaling. On the other hand several lines of data including those from knockout models question the universal role for septins in animal cell cytokinesis. Mammalian hematopoietic cells seem to proliferate and efficiently undergo cytokinesis in the absence of pivotal septin proteins in a context-dependent manner. The lack of septin-dependence of hematopoiesis also opens the possibility of safely targeting septin-dependent cytokinesis for solid-tumor therapy. Thus the septin field is perfectly poised with novel roles for septins being discovered and the basic understanding on septin assembly and its canonical functions constantly revisited. The objective of this research topic was to provide an exclusive platform for discussing these rapid advances in the septin field. With a mixture of reviews and research articles encompassing diverse areas of septin research, ranging from the humble yeast model to human cancer, this ebook will be an interesting reading material for both experts as well as new comers to the septin field.

    Beschreibung: Eukaryotic cells contain distinct membrane-bound organelles, which compartmentalise cellular proteins to fulfil a variety of vital functions. Many organelles have long been regarded as isolated and static entities (e.g., peroxisomes, mitochondria, lipid droplets), but it is now evident that they display dynamic changes, interact with each other, share certain proteins and show metabolic cooperation and cross-talk. Despite great advances in the identification and characterisation of essential components and molecular mechanisms associated with the biogenesis and function of organelles, information on how organelles interact and are incorporated into metabolic pathways and signaling networks is just beginning to emerge. Organelle cooperation requires sophisticated targeting systems which regulate the proper distribution of shared proteins to more than one organelle. Organelle motility and membrane remodeling support organelle interaction and contact. This contact can be mediated by membrane proteins residing on different organelles which can serve as molecular tethers to physically link different organelles together. They can also contribute to the exchange of metabolites and ions, or act in the assembly of signaling platforms. In this regard organelle communication events have been associated with important cellular functions such as apoptosis, antiviral defense, organelle division/biogenesis, ROS metabolism and signaling, and various metabolic pathways such as breakdown of fatty acids or cholesterol biosynthesis. In this research topic we will focus on recent novel findings on the underlying molecular mechanisms and physiological significance of organelle interaction and cooperation with a particular focus on mitochondria, peroxisomes, endoplasmic reticulum, lysosomes and lipid droplets and their impact on the regulation of cellular homeostasis. Our understanding of how organelles physically interact and use cellular signaling systems to coordinate functional networks between each other is still in its infancy. Nevertheless recent discoveries of defined membrane structures such as the mitochondria-ER associated membranes (MAM) are revealing how membrane domains enriched in specific proteins transmit signals across organelle boundaries, allowing one organelle to influence the function of another. In addition to its role as a mediator between mitochondria and the ER, contacts between the MAM and peroxisomes contribute to antiviral signaling, and specialised regions of the ER are supposed to initiate peroxisome biogenesis, whereas intimate contacts between peroxisomes, lipid droplets and the ER mediate lipid metabolism. In line with these observations it is tempting to speculate that further physical contact sites between other organelles exist. Alternatively, novel regulated vesicle trafficking pathways between organelles (e.g., mitochondria to peroxisomes or lysosomes) have been discovered implying another mode of organelle communication. Identifying the key molecular players of such specialised membrane structures will be a prerequisite to understand how organelle communication is physically accomplished and will lead to the identification of new regulatory networks. In addition to the direct transmission of interorganellar information, cytosolic messenger systems (e.g., kinase/phosphatase systems or redox signaling) may contribute to the coordination of organelle functions. This research topic will integrate new findings from both modes of communication and will provide new perspectives for the functional significance of cross-talk among organelles.     Beschreibung: Biological membranes protect cells and organelles from the surrounding environment, but serve also as organising platforms for physiological processes such as cell signalling. The hydrophobic core of membranes is composed of lipids and proteins influencing each other. Local membrane composition and properties define its molecular organisation and, in this way, regulate the function of all associated molecules. Therefore, studying interactions of components, biophysical properties and overall membrane dynamics provides essential information on its function in the context of cell activities. Such knowledge can contribute to biomedical fields such as pharmacology, immunology, neurobiology and many others. The goal of the Research Topic entitled ‘Molecular organisation of membranes: where biology meets biophysics’ was to provide a comprehensive platform for publishing articles, reviews and opinions focused on membrane organisation and the forces behind its heterogeneous and dynamic structure. We collected 11 works which cover topics as diverse as general membrane organisation models, membrane trafficking and signalling regulation, biogenesis of caveolae, protein-lipid interactions and the importance of membrane-associated tetraspanins networks. The prevalent theme was the existence of membrane nanodomains. To this point, new emerging technologies are presented which own the power to bring a novel insight on how membrane nanodomains are formed and maintained and what is their function. We believe that the collection of works in this Research Topic brings forward some important questions which will stimulate further research in this difficult but exciting field.

    Beschreibung: Mathematical and computational models play an essential role in understanding the cellular metabolism. They are used as platforms to integrate current knowledge on a biological system and to systematically test and predict the effect of manipulations to such systems. The recent advances in genome sequencing techniques have facilitated the reconstruction of genome-scale metabolic networks for a wide variety of organisms from microbes to human cells. These models have been successfully used in multiple biotechnological applications. Despite these advancements, modeling cellular metabolism still presents many challenges. The aim of this Research Topic is not only to expose and consolidate the state-of-the-art in metabolic modeling approaches, but also to push this frontier beyond the current edge through the introduction of innovative solutions. The articles presented in this e-book address some of the main challenges in the field, including the integration of different modeling formalisms, the integration of heterogeneous data sources into metabolic models, explicit representation of other biological processes during phenotype simulation, and standardization efforts in the representation of metabolic models and simulation results.

    Beschreibung: Mitogen-activated protein kinase (MAPK) pathways are evolutionarily conserved in all eukaryotes and allow cells to respond to changes in the physical and chemical properties of the environment and to produce an appropriate response by altering many cellular functions. MAPKs are among the most intensively studied signal transduction systems. MAPK research is a very dynamic field in which new perspectives are continuously opening to the scientific community. Importantly, many MAPK inhibitors have been developed during the last years and are currently being tested in preclinical and clinical assays for inflammatory diseases and cancer treatment. In this research topic, we have gathered 14 papers covering recent advances in different aspects of the MAPK research area that have provided valuable insight into the spatiotemporal dynamics, the regulation and functions of MAPK pathways, as well as their therapeutic potential. We hope that this Research Topic helps readers to have a better understanding of the progresses that have been made recently in the field of MAPK signalling.
