    Description: It is now well appreciated that the immune system, in addition to its traditional role in defending the organism against pathogens, communicate in a well-organized fashion with the brain to maintain homeostasis and regulate a set of neural functions. Perturbation in this brain-immune interactions due to inflammatory responses may lead to psychiatric and neurological disorders. Microglia are one of the essential cells involved in the brain-immune interactions. Microglial cells are now not simply regarded as resident tissue macrophages in the brain. These cells are derived from myeloid progenitor cells in the yolk sac in early gestation, travel to the brain parenchyma and interact actively with neurons during the critical period of neurogenesis. Microglia provide a trophic support to developing neurons and take part in the neural wiring through the activity-dependent synapse elimination via direct neuron-microglia interactions. Altered microglial functions including changes in the gene expression due to early life inflammatory events or psychological and environmental stressors can be causally related to neurodevelopmental diseases and mental health disorders. This type of alterations in the neural functions can occur in the absence of infiltration of inflammatory cells in the brain parenchyma or leptomeninges. In this sense, the pathogenetic state underlying a significant part of psychiatric and neurological diseases may be similar to “para-inflammation”, an intermediate state between homeostatic and classical inflammatory states as defined by Ruslan Medzhitov (Nature 454:428-35, 2008). Therefore, it is important to study how systemic inflammation affects brain health and how local peripheral inflammation induces changes in the brain microenvironment. Chronic pain is also induced by disturbance in otherwise well-organized multisystem interplay comprising of reciprocal neural, endocrine and immune interactions. Especially, early-life insults including exposure to immune challenges can alter the neuroanatomical components of nociception, which induces altered pain response later in life. Recently the discrete roles of microglia and blood monocyte-derived macrophages are being defined. The distinction may be further highlighted by disorders in which the brain parenchymal tissue is damaged. Therefore, studies investigating the dynamics of immune cells in traumatic brain injury and neurotropic viral infections including human immunodeficiency virus, etc. as well as neurodegenerative diseases such as amyotrophic lateral sclerosis are promising to clarify the interplay between the central nervous and immune systems. The understanding of the histological architecture providing the infrastructure of such neuro-immune interplay is also essential. This Frontiers research topic brings together fourteen articles and aims to create a platform for researchers in the field of psychoneuroimmunology to share the recent theories, hypotheses and future perspectives regarding open questions on the mechanisms of cell-cell interactions with chemical mediators among the nervous, immune and endocrine systems. We hope that this platform would reveal the relevance of the studies on multisystem interactions to enhance the understanding of the mechanisms underlying a wide variety of neurological and psychiatric disorders.

    Description: Microglia are essential for the development and function of the adult brain. Their ontogeny, together with the absence of turnover from the periphery and the singular environment of the central nervous system (CNS), make microglia a unique cell population compared to other tissue-macrophages. The unique properties and functions of microglial cells, such as their role in synaptic pruning or the exceptional capacity to scan the brain parenchyma and rapidly react to its perturbations, have emerged in recent years. In the coming years, understanding how microglia acquire and maintain their unique profiles in order to fulfil distinct tasks in the healthy CNS and how these are altered in disease, will be essential to develop strategies to diagnose or treat CNS disorders with an immunological component. This Research Topic covers several aspects of microglial biology, ranging from their origin and the functional role of microglia during development and lifespan, their molecular properties compared with other brain and peripheral immune cells to microglial phenotypes and functional states in neurodegenerative diseases and brain tumours. In conclusion, the present Research Topic provides a comprehensive overview of our current understanding of several cellular and molecular mechanisms that make microglia a unique immune cell population within the healthy CNS as well as under inflammatory, neurodegenerative and tumorigenic processes.

    Description: The non-classical HLA class I molecule HLA-G is different from classical HLA class I molecules because of the low polymorphism in the coding region, the fact that HLA-G primary transcript is alternatively spliced in seven isoforms, and the inhibitory action on immune cells. Although HLA-G is low polymorphic, variants in both promoter and 3’ un-translated region (UTR) of HLA-G locus regulate its expression. In healthy conditions, a basal level of HLA-G gene transcription is observed in most cells and tissues; however, translation into HLA-G protein is restricted to trophoblasts in the placenta, where it participates in promoting tolerance at the fetal-maternal interface. HLA-G is also expressed by thymic epitelial, cornea, mesenchymal stem cells, nail matrix, pancreatic beta cells, erythroid, and endothelial precursors. HLA-G can be neo-expressed in adult tissues in pathological conditions, and its expression has been documented autoimmune disorders, viral infections, and cancer. In the latter setting de novo HLA-G expression is associated with the capability of tumor cells to evade the immune control. In the last decade it has become evident that HLA-G expression on T cells and antigenpresenting cells confers to these cells tolerogenic properties. This Research Topic focused on i) summarizing updated clinical and immunological evidences that HLA-G expression is associate with beneficial or detrimental tolerance, ii) gathering new insights into the mechanisms governing the expression of HLA-G in healthy and pathological conditions, such as pre-eclampsia, and iii) examining the mechanisms underlying HLA-G mediated tolerance.

    Description: In the year 2013, ‘selfie’ was named word of the year by Oxford Dictionaries in recognition of dramatic changes in frequency, prominence, and register of the term. This drastic increase in selfie-taking was spurred by two factors. The first was the advent of smartphones equipped with front cameras and preview screens that made it easy to compose a photographic self-portrait by a process of deliberately exploring one’s image, choosing a pose, and finally taking the picture. The second key change contributing to the rise of the selfie age was the increasing availability of internet connections. It is estimated that about 50% of the world population has access to the internet today (2018; https://www.internetworldstats.com). At the end of the past century, this percentage was a mere 1%. The growth of the internet infrastructure simultaneously spurred the development of social network applications such as Facebook, Twitter, Snapchat, and Instagram, providing accessible media for sharing photographs including photographic self-portraits. However, despite their tremendous reach and popularity, selfies have so far received relatively little attention by the scientific community, especially within psychology. Thus, we proposed a Frontiers in Psychology Research Topic to expand empirical and theoretical work on the massively popular, yet scientifically unexplored, phenomenon of the selfie. The articles published in this eBook offer a multifaceted insight into current scholarly work on this topic.

    Description: Lymphocytes constantly survey the lymph nodes in search for potential infection by a pathogen. They enter the afferent lymphatic vessel that serves as a conduit to transport the motile lymphocytes to the draining lymph node. Lymphatic vessels (LVs) are present in most vascularized tissues. They are traditionally regarded as passive conduits for soluble antigens and leukocytes. Afferent LVs begin as blind ended capillaries, which give rise to collecting vessels that merge and connect with draining lymph nodes (dLNs). Initial lymphatic capillaries are composed of Lymphatic Endothelial Cells (LECs) connected by discontinuous cell junctions, which join to form larger collecting lymphatic vessels, and ultimately feed into the LN subcapsular sinus. Within the LN, LECs are localized to the subcapsular, cortical, and medullary sinuses, where they interact with incoming and exiting leukocytes. LECs, and in general LN stromal cells, have emerged in the recent years as active players in the immune response. In support to this,studies have shown that the immune response generated during inflammation and under pathologic conditions is accompanied by modeling of the LVs and generation of new lymphatics, a process known as lymphangiogenesis. These facts strongly suggest that LECs and stromal LN cells in general, are not inert players but rather are part of the immune response by organizing immune cells movement, exchanging information and supplying survival factors. The purpose of this research topic is to review the role of the LECs during immune homeostasis and cancer. Considering the critical role of lymphangiogenesis in many pathologies like chronic and acute inflammation, autoimmunity, wound healing, graft rejection, and tumor metastasis, it is important to understand the molecular mechanisms that govern the cross talks between the LECs and immune cells during homeostasis and inflammation.

    Description: Allogeneic haematopoietic stem cell transplantation (allo-HSCT) is widely used in the treatment of haematological malignancies as a form of immunotherapy acting through a graft-versus-leukemia (GvL) reaction. This curative allogeneic response can be associated with severe drawbacks, such as frequent and severe graft-versus-host disease (GvHD) and a long-lasting immunodeficiency, especially now with the development of innovative strategies such as umbilical cord blood transplantation or transplants from haplo-identical family donors (Haplo-HSCT). In the long-term follow-up of these patients, severe post-transplant infections, relapse or secondary malignancies may be directly related to persistent immune defects.Reconstitution of the different lymphocyte populations (B, T, NK, NKT) and antigen presenting cells of myeloid origin (monocytes, macrophages and dendritic cells) should be considered not only quantitatively but especially qualitatively, in terms of functional subsets. Immune deficiency leading to an increased susceptibility to infections lasts for more than a year. Although infections that occur in the first month mostly result from a deficiency in both granulocytes and mononuclear cells (MNC), later post-engraftment infections are due to a deficiency in MNC subsets, primarily CD4 T-cells and B-cells. T-cell reconstitution has been extensively studied because of the central role of T-cells in mediating both GvHD, evidenced by the reduced incidence of this complication following T-Cell depletion, and a GvL effect as shown by DLI. In the recent years there has been renewed interest in the role of NK-cells, especially in the context of Haplo-HSCT, and in B-cell reconstitution.This Frontiers Research Topic will provide state of the art knowledge of the mechanisms of immune reconstitution in an allogeneic environment, in order to improve monitoring and therapeutic intervention in allo-HSCT patients.

    Description: The integration of multisensory information is an essential mechanism in perception and in controlling actions. Research in multisensory integration is concerned with how the information from the different sensory modalities, such as the senses of vision, hearing, smell, taste, touch, and proprioception, are integrated to a coherent representation of objects. Multisensory integration is central for action control. For instance, when you grasp for a rubber duck, you can see its size and hear the sound it produces. Moreover, identical physical properties of an object can be provided by different senses. You can both see and feel the size of the rubber duck. Even when you grasp for the rubber duck with a tool (e.g. with tongs), the information from the hand, from the effect points of the tool and from the eyes are integrated in a manner to act successfully. Over the recent decade a surge of interest in multisensory integration and action control has been witnessed, especially in connection with the idea that multiple sensory sources are integrated in an optimized way. For this perspective to mature, it will be helpful to delve deeper into the information processing mechanisms and their neural correlates, asking about the range and constraints of this mechanisms, about its localization and involved networks.

    Description: Gamma/delta (γδ) T-cells are a small subset of T-lymphocytes in the peripheral circulation but constitute a major T-cell population at other anatomical localizations such as the epithelial tissues. In contrast to conventional a/ß T-cells, the available number of germline genes coding for T-cell receptor (TCR) variable elements of γδ T-cells is very small. Moreover, there is a prefential localization of γδ T-cells expressing given Vgamma and Vdelta genes in certain tissues. In humans, γδ T-cells expressing the Vg9Vd2-encoded TCR account for anywhere between 50 and 〉95% of peripheral blood γδ T-cells, whereas cells expressing non-Vd2 genes dominate in mucosal tissues. In mice, there is an ordered appearance of γδ T-cell „waves“ during embryonic development, resulting in preferential localization of γδ T-cells expressing distinct VgammaVdelta genes in the skin, the reproductive organs, or gut epithelia. The major function of γδ T-cells resides in local immunosurveillance and immune defense against infection and malignancy. This is supported by the identification of ligands that are selectively recognized by the γδ TCR. As an example, human Vgamma9Vdelta2 T-cells recognize phosphorylated metabolites („phosphoantigens“) that are secreted by many pathogens but can also be overproduced by tumor cells, providing a basis for a role of these γδ T-cells in both anti-infective and anti-tumor immunity. Similarly, the recognition of endothelial protein C receptor by human non-Vdelta2 γδ T-cells has recently been identified to provide a link for the role for such γδ T-cells in immunity against epithelial tumor cells and cytomegalovirus-infected endothelial cells. In addition to „classical“ functions such as cytokine production and cytotoxicity, recent studies suggest that subsets of γδ T-cells can exert additional functions such as regulatory activity and – quite surpisingly – „professional“ antigen-presenting capacity. It is currently not well known how this tremendous extent of functional plasticity is regulated and what is the extent of γδ TCR ligand diversity. Due to their non-MHC-restricted recognition of unusual stress-associated ligands, γδ T-cells have raised great interest as to their potential translational application in cell-based immunotherapy. Topics of this Research Focus include: Molecular insights into the activation and differentiation requirements of γδ T-cells, role of pyrophosphates and butyrophilin molecules for the activation of human γδ T-cells, role of γδ T-cells in tumor immunity and in other infectious and non-infectious diseases, and many others. We are most grateful to all colleagues who agreed to write a manuscript. Thanks to their contributions, this E-book presents an up-to-date overview on many facets of the still exciting γδ T-cells.

    Description: A conversation between two people can only take place if the words intended by each speaker are successfully recognized. Spoken word recognition is at the heart of language comprehension. This automatic and smooth process remains a challenge for models of spoken word recognition. Both the process of mapping the speech signal onto stored representations for words, and the format of the representation themselves are subject to debate. So far, existing research on the nature of spoken word representations has focused mainly on native speakers. The picture becomes even more complex when looking at spoken word recognition in a second language. Given that most of the world’s speakers know and use more than one language, it is crucial to reach a more precise understanding of how bilingual and multilingual individuals encode spoken words in the mental lexicon, and why spoken word recognition is more difficult in a second language than in the native language. Current models of native spoken word recognition operate under two assumptions: (i) that listeners’ perception of the incoming speech signal is optimal; and (ii) that listeners’ lexical representations are accurate. As a result, lexical representations are easily activated, and intended words are successfully recognized. However, these assumptions are compromised when applied to a later-learned second language. For a variety of reasons (e.g., phonetic/phonological, orthographic), second language users may not perceive the speech signal optimally, and they may still be refining the motor routines needed for articulation. Accordingly, their lexical representations may differ from those of native speakers, which may in turn inhibit their selection of the intended word forms. Second language users also have to solve a larger selection challenge—having words in more than one language to choose from. Thus, for second language users, the links between perception, lexical representations, orthography, and production are all but clear. Even for simultaneous bilinguals, important questions remain about the specificity and interdependence of their lexical representations and the factors influencing cross-language word activation. This Frontiers Research Topic seeks to further our understanding of the factors that determine how multilinguals recognize and encode spoken words in the mental lexicon, with a focus on the mapping between the input and lexical representations, and on the quality of lexical representations.

    Description: Epigenetics is the study of changes in gene activity that are heritable but not caused by changes in the DNA sequence. By modulating gene activities, epigenetic changes regulate cell functions. They include DNA methylation, histone posttranslational modifications and gene silencing by the action of non-coding RNAs, particularly microRNAs. It is now clear that epigenetic changes regulate B cell development. By acting in concert with networks of transcription factors, they modulate the activation of B cell lineage specific gene programs and repress inappropriate gene transcription in particular B cell differentiation states. A hallmark of B cell development in the bone marrow is the assembly of the B cell receptor (BCR) for antigen through rearrangement of immunoglobulin heavy (IgH) and light (IgL) chain V(D)J genes, as mediated by RAG1/RAG2 recombinases. Ig V(D)J rearrangement critically times the progression from pro-B cell to pre-B cell and, finally, mature B cell. Such progression is modulated by epigenetic marks, such as DNA methylation and histone posttranslational modifications, that increase chromatin accessibility and target RAG/RAG2 to V, D and J DNA. It is also dependent on the expression of multiple microRNAs. Mice deficient in Ago2, which is essential for microRNA biogenesis and function, have B cell development blocked at the pro-B cell stage. In agreement with this, B cell specific ablation of microRNA by B cell-specific knockout of Dicer virtually blocks B cell differentiation at the pro-B to pre-B cell transition. After mature B cells encounter antigen, changes of the epigenetic landscape are induced by the same stimuli that drive the antibody response; such epigenetic changes underpin the maturation of the antibody response itself. They instruct those B cell differentiation processes, somatic hypermutation (SHM), class switch DNA recombination (CSR) and plasma cell differentiation, that are central to the maturation of the antibody response as well as differentiation of memory B cells. Inducible histone modifications, together with DNA methylation and microRNAs modulate the transcriptome, particularly the expression of activation-induced cytidine deaminase (AID), central to SHM and CSR, and B lymphocyte-induced maturation protein-1 (Blimp-1), which is central to plasma cell differentiation. Combinatorial histone modifications also function as histone codes in the targeting of the CSR and, possibly, the SHM machinery to the Ig locus by recruiting specific adaptors (histone code readers) that can in turn target and/or stabilize CSR/SHM factors. Epigenetic alterations in memory B cells contribute to their functionally distinction from their naive counterparts. Memory B cells inherit epigenetic information from their precursors and acquire new epigenetic marks, which make these resting B cells poised to promptly respond to antigen. The cross/feedback regulation of different epigenetic modifications/elements further increases the complexity of the B cell epigenome, which interacts with the genetic information for precise modulation of gene expression. It is increasingly evident that epigenetic dysregulation in B cells, including aberrant expression of microRNAs, can result in aberrant antibody responses to microbial pathogens, emergence of pathogenic autoantibodies or B cell neoplastic transformation. Epigenetic marks are potential targets for new therapeutics in autoimmunity and B cell malignancy.

    Description: The visual system consists of hierarchically organized distinct anatomical areas functionally specialized for processing different aspects of a visual object (Felleman & Van Essen, 1991). These visual areas are interconnected through ascending feedforward projections, descending feedback projections, and projections from neural structures at the same hierarchical level (Lamme et al., 1998). Accumulating evidence from anatomical, functional and theoretical studies suggests that these three projections play fundamentally different roles in perception. However, their distinct functional roles in visual processing are still subject to debate (Lamme & Roelfsema, 2000). The focus of this Research Topic is the roles of feedforward and feedback projections in vision. Even though the notions of feedforward, feedback, and reentrant processing are widely accepted, it has been found difficult to distinguish their individual roles on the basis of a single criterion. We welcome empirical contributions, theoretical contributions and reviews that fit into any one (or a combination) of the following domains: 1) their functional roles for perception of specific features of a visual object 2) their contributions to the distinct modes of visual processing (e.g., pre-attentive vs. attentive, conscious vs. unconscious) 3) recent techniques/methodologies to identify distinct functional roles of feedforward and feedback projections and corresponding neural signatures. We believe that the current Research Topic will not only provide recent information about feedforward/feedback processes in vision but also contribute to the understanding fundamental principles of cortical processing in general.

    Description: The recent wave of clinical studies demonstrating long-term therapeutic efficacy highlights the enormous potential of gene therapy as an approach to the treatment of inherited disorders and cancer. While in recent years lentiviral vectors have dominated the field of ex vivo gene therapy in man, adeno-associated virus (AAV) vectors have become the platform of choice for the in vivo gene delivery, both local and systemic.Despite the achievements in the clinic however, a number of hurdles remain to be overcome in gene therapy, these include availability of scalable vector production systems, potential issues associated with insertional mutagenesis, and concerns related to immunogenicity of gene therapeutics. For AAV vectors, clinical trials showed that immunity directed against the vector could either prevent transduction of a target tissue or limit the duration of therapeutic efficacy. Initial observations in the context of a gene therapy trial for hemophilia spurred over a decade efforts by gene therapists and immunologists to understand the mechanism and identify factors that contribute to AAV’s immunogenicity, including the prevalence of B cell and T cell immunity to wild type AAV in humans and the interaction of AAV vectors with the innate and adaptive immune system. Despite a number of important contributions in particular in the more recent past, our knowledge on the immunology of gene transfer is still rudimental; this is partly due to the fact that the basic understanding of the complex balance between tolerance and immunity to an antigen, key aspect of gene transfer with AAV, keeps evolving rapidly. However, continuing work towards a better definition of the interaction of viral vectors with the immune system has led to significant advances in the knowledge of the factors influencing the outcome of gene transfer, such as the vector dose, the immune privilege of certain tissues, and the induction of tolerance to an antigen. A better understanding of the structure-function relationship of the viral capsid has boosted the development of novel immune-escape vector variants. In addition, novel immunomodulatory strategies were established to prevent or reduce anti-capsid immunity have been developed and are being tested in preclinical models and in clinical trials. Together, these advances are bringing us closer to the goal of achieving safe and sustained therapeutic gene transfer in humans. In this research topic, a collection of Original Research and Review Articles highlights critical aspects of the interaction between gene AAV vectors and the immune system, discussing how these interactions can be either detrimental or constitute an advantage, depending on the context of gene transfer, and providing tools and resources to better understand the issue of immunogenicity of AAV vectors in gene transfer.

The recent finding that the human nose can discriminate something like a trillion different smells serves as yet another reminder that we have again underestimated the capacity of our sense of smell (Bushdid, Magnasco, Vosshall & Keller, 2014). This volume serves as a further corrective for anyone who should hold the view that olfaction is unimportant in human affairs. The papers presented in this ebook, carefully collated and overseen by Aldo Zucco, Benoist Schaal, Mats Olsson and Ilona Croy, showcase a large number of quite different reasons for studying the applied side of olfaction, and indeed human olfaction in general. The 23 contributions presented here cover a broad range of topics, which illustrate contemporary interests in our field. Although with a strong applied focus, a noteworthy feature of this ebook is the richness of the theoretical perspectives that are developed. These range from considerations of olfactory perception, memory, expertise, and priming right the way through to receptor genetics. These contributions, from many leading experts in the field, will surely shape much of the applied work linking olfaction to disease, which is a further focus of this ebook. In respect to health and disease, the chapters on aging, pregnancy, depression, alcohol dependency and environmental odours, present overviews and rich new data on many contemporary problems, to which the study of olfaction is now contributing. A particularly notable aspect of olfactory experience is the affective impact that odours can have on people and their lives. The ebook covers some particularly intriguing aspects of work in this area, with empirical studies investigating dissociations between wanting and liking, stress reduction in the elderly, mother-infant bonding, and the emotions that different odourants can evoke. This affective line of work is nicely complemented by empirical studies on expertise, the effect of odours on visual attention, and the relationship between particular personality traits and interest in olfaction. The gradual appropriation of methods from cognitive neuroscience into olfaction is also nicely represented in this ebook, with at least three of the chapters reporting data using neuroimaging, including a particular intriguing study looking at recognition of odours in mixtures. Finally, the close links between olfactory perception and sensory evaluation are also reflected in a chapter on wine. I hope that readers of this e-book will be struck, as I have been in reading its various chapters, how much olfaction affects our lives, and how the study of this sense can enrich it.

    Description: CD4+ T lymphocytes play an essential role in host defense against bacterial, parasitic and viral infections. During infection, under the influence of intrinsic signals received through peptide-MHC/TCR interactions and extrinsic signals provided by pathogen-conditioned dendritic and other accessory cells, CD4+ T cells proliferate and differentiate into specialized T helper (Th) effectors, which produce distinct sets of cytokines tailored to combat a specific class of microbes. The concept of CD4+ T cell multi-functionality was developed after the seminal discovery of Th1 and Th2 cells nearly 30 years ago. Although the Th1/Th2 paradigm has successfully withstood the test of time, in the past decade additional Th subsets (Th17, Tfh, Th22, Th9) have been identified. Similarly, single cell analyses of cytokines and master transcriptional factors have revealed that, at the population level, CD4+ T cell responses are far more heterogeneous than initially anticipated. While some of the checkpoints in Th cell specification have been identified, recent studies of transcriptional and epigenetic regulation have uncovered a significant flexibility during the course CD4+ T lymphocyte polarization. In addition, Th cells expressing cytokines with counteracting functions, as a measure of self-regulation, display yet another level of diversity. Understanding the mechanisms that control the balance between stability vs. plasticity of Th effectors both at the time of initiation of immune response and during development of CD4 T cell memory is critical for the rational design of better vaccines and new immunotherapeutic strategies. This research topic will cover current views on Th cell development, with a focus on the mechanisms that govern differentiation, function and regulation of effector Th cells in the context of microbial infections.

    Description: Lipids are best known as energy storing molecules and core-components of cellular membranes, but can also act as mediators of cellular signaling. This is most prominently illustrated by the paramount importance of the phospholipase C (PLC) and phosphoinositide 3-kinase (PI3K) signaling pathways in many cells, including T cells and cancer cells. Both of these enzymes use the lipid phosphatidylinositol(4,5)bisphosphate (PIP2) as their substrate. PLCs produce the lipid product diacylglycerol (DAG) and soluble inositol(1,4,5)trisphosphate (IP3). DAG acts as a membrane tether for protein kinase C and RasGRP proteins. IP3 is released into the cytosol and controls calcium release from internal stores. The PI3K lipid product phosphatidylinositol(3,4,5)trisphosphate (PIP3) controls signaling by binding and recruiting effector proteins such as Akt and Itk to cellular membranes. Recent research has unveiled important signaling roles for many additional phosphoinositides and other lipids. The articles in this volume highlight how multiple different lipids govern T cell development and function through diverse mechanisms and effectors. In T cells, lipids can orchestrate signaling by organizing membrane topology in rafts or microdomains, direct protein function through covalent lipid-modification or non-covalent lipid binding, act as intracellular or extracellular messenger molecules, or govern T cell function at the level of metabolic regulation. The cellular activity of certain lipid messengers is moreover controlled by soluble counterparts, exemplified by symmetric PIP3/inositol(1,3,4,5)tetrakisphosphate (IP4) signaling in developing T cells. Not surprisingly, lipid producing and metabolizing enzymes have gained attention as potential therapeutic targets for immune disorders, leukemias and lymphomas.

    Description: Schistosomiasis is a severe parasitic disease, endemic in 74 developing countries with up to 600 million people, including many children, infected and 800 million at risk of contracting the disease following infection with Schistosoma mansoni, S. haematobium or S. japonicum. Disease burden is estimated to exceed 70 million disability-adjusted life-years, and leads to remarkably high YLD (years lived with disability) rates. Even more importantly, people with schistosomiasis are highly susceptible to malaria, tuberculosis and hepatic and acquired immunodeficiency viruses. There is only one drug, praziquantel, currently available for treatment and it has high efficacy, low cost, and limited side effects. However, only 13% of the target population has received the drug, and those treated are at continuous risk of reinfection necessitating repeated drug administration and the emergence of drug resistant parasites is a constant threat. There currently is no vaccine. While the target of 〉40% protection has been achieved with some molecules such as excretory-secretory proteins including calpain, glyceraldehyde 3-phosphate dehydrogenase, and cysteine peptidases, very recent articles reiterate the findings published during the last 2 decades of the last century, contradicting the established data of the pioneers of schistosome biology. A consensus should be reached without delay, in order to propose collaborative independent experiments and proceed ahead to pre- and clinical trials with efficacious candidate vaccine molecules. The proposed plan aims to finally reach an objective and fruitful agreement , via inviting established and young researchers from the United States, Brazil, China, Australia, and Europe who are working with different vaccine antigens, adjuvants, and approaches for immunization against S. mansoni, S. haematobium, and S. japonicum. It is hoped that the forum will end with a very few candidate antigens and a consensus approach regarding target immune responses, thus leading to encouraging the World Health Organization and other international foundations to sponsor the development and implementation of the urgently required, yet still elusive, vaccine for preventing and eliminating the transmission of schistosomiasis.

This behavior is functional: the reduction of distractions in the environment can improve performance on cognitive tasks, including memory retrieval. Reduction of visual distractions can be operationalized through eye-closure, gaze aversion, or by comparing exposure to simple and complex visual displays, respectively. Reduction of auditory distractions is typically examined by comparing performance under quiet and noisy conditions. Theoretical reasoning regarding this phenomenon draws on various psychological principles, including embodied cognition, cognitive load, and modality-specific interference. Practical applications of the research topic are diverse. For example, the findings could be used to improve performance in forensic settings (e.g., eyewitness testimony), educational settings (e.g., exam performance), occupational settings (e.g., employee productivity), or medical settings (e.g., medical history reporting).     Description: Natural antibodies (NAbs) are found in normal individuals in the absence of exogenous antigenic stimulation. Natural antibodies rapidly recognize and protect against pathogens that have not been previously encountered. NAbs also cross-react with several self-antigens, which, besides their role as a first line of defense against pathogens, affords them the ability to perform important housekeeping functions in healthy organisms. Such housekeeping functions include the clearance of oxidized damaged structures and/or apoptotic cells, which prevents the induction of pro-inflammatory effects. In addition, NAbs play a role in preventing the expansion of specific auto-reactive clones, thereby behaving as regulatory elements in acute or chronic inflammation. To maintain the non-pathogenic balance between the dual pathogen/self-antigen cross-reactivities of NAbs, a strict regulation in NAb secretion and function is necessary to avoid autoimmune disease. Actually, some of the NAbs related auto-reactivities, such as anti-DNA and anti-MOG, have been associated with autoimmunity. Furthermore, NAbs have been shown to bind to ‘neo-self’ carbohydrate antigens on glycolipids and glycoproteins found on malignant but not normal cells, which suggests NAbs may take part in tumor immunosurveillance. Many aspects regarding NAbs have yet to be studied in more detail: the reactivity and function of NAbs in health and disease, the behavior of the NAb repertoire with increasing age, the regulation of natural antibody production and auto-reactivity, the ways to specifically activate NAbs producing cells with desired specificities, the characteristics of human NAbs, among others. This special topics eBook consists of a number of articles exploring the cells that produce NAbs as well as the characteristics, function, specificity, and/or the role of natural antibodies in health and disease.

Accordingly, this volume combats "revisionist science", by those who want to alter history by telling the stories a different way than actually happened. In this regard, one of the good things about science vs. other disciplines is that we have the written record of what was done, when it was done and by whom. Even so, we do not have the complete story or narrative of how and why experiments were done, and what made the differences that led to success.     Description: Character can be defined as self-aware knowledge that helps the individual to set goals, values and ethical principles (Cloninger, 2004). This meta-cognitive dimension of human personality involves ‘Theory of Mind’, and is positively related to measures of well-being, mental health, and constructive behavior patterns. Research from at least three different fields, cultural (Shweder, Much, Mahapatra & Park, 1997), personality (Cloninger, 2004), and social psychology (Abele & Wojcizke, 2007) suggest that character can be organized along three broad principles: agency, which is related to the autonomy and the fulfillment and enhancement of the self; communion, which is related to engagement in the protection and relations to others such as families, companies or nations; and spirituality, which is related to the human ability to transcend the self and find and interconnection with all life and appreciation of the whole world around us (Haidt, 2006; Cloninger, 2013). Using the Temperament and Character Inventory (Cloninger, Svrakic & Przybeck, 1993) researchers have found that agentic (i.e., Self-directedness) and communal (i.e., Cooperativeness) values are associated to high levels of happiness, psychological well-being, and less violent behavior. Moreover, low Self-directedness and Cooperativeness is recurrent among individuals with all types of mental health problems, such as, depression, schizophrenia, anxiety disorder, autism spectrum disorders, attention deficit/hyperactivity disorder and et cetera. Spirituality, in coherence with agency and communion, guides the individual to seek self-realization in harmony with others and nature in the changing world (Cloninger, 2013). Seeing character as self-awareness of the self in three dimensions has also been associated to human responsibility and empowerment.     Description: It is acknowledged that practice could induce rapid change or reorganization of the brain’s cellular or neural networks as well as behaviors. Notably, practice relevant to mental or physical approach attracted great attention in this decade. It highlights profound significance both for human evolvement and individual development. Specifically, acquiring fine motor skills is a crucial premise for human being to evolve to modern human by using tools in one side. In the other side, numerous evidences indicated that motor learning involved in limb and trunks promotes the development of individual brain in anatomy and functions. Hence, motor learning is also tightly associated with developmental plasticity. These studies on brain-mind-body practice illuminate a promising way in promoting human brain health. This editorial covers wide range of brain-mind-body practice forms to summarize recent new findings and development from behavioral, physiological, neurobiological and psychological science approaches. In this research topic, we addressed recent findings from theoretical as well as experimental perspective including contributions under the following three headings: 1) intervention studies to investigate the positive effect of brain-mind-body practice on cognition and relevant brain mechanism. The intervention pattern consisted of short-term practice ranging from few hours to several weeks; 2) cross-sectional studies using expert-novice paradigm to explore the behavioral and neural system change induced by extensive brain-mind-body practice; 3) the mediators influence the relationship between practice and health outcomes and 4) new viewpoints on brain-mind-body practice from theoretical perspectives. Here we briefly highlight these articles aiming to provide a deep understanding for the association between practice, plasticity and health for readers. Additionally, it offers new insights for developing possible practice interventions for clinical treatment of neurological dysfunction or disorders.

    Description: Immune molecules have evolved to distinguish “self “molecules from “non-self”, “altered self” and “danger” molecules. Recognition is mediated via interactions between pattern recognition receptor molecules (PPRs) and their ligands, which include hydrophobic and electrostatic interactions between amino acid residues on the PPRs and uncharged or charged groups on amino acid residues, sugar rings or DNA/RNA molecules. Recognition in innate immunity range from cases (C1q, mannin-binding protein etc) where recognition is orchestrated by interaction between many ligands with one receptor molecule, and density of interaction is necessary for strong specific recognition, distinct from weak non-specific binding, and cases such as TLRs and NLRs where recognition involves complexation of single receptor and ligand, followed by oligomerisation of the receptor molecule. The majority of PPR molecules bind and recognise a wide variety of ligands, e.g TLR4 recognises LPS (gram negative bacteria), Lipotechoic acid (gram positive bacteria), heat shock protein hsp60, respiratory syncytial virus fusion protein etc, molecules that are structurally dissimilar to each other. This indicates considerable flexibility in their binding domains (amino acid residue variations) and modes (hydrophobic and charged, direct or mediated via an adaptor molecule). However, in many cases there is a dearth of structural and molecular data available, required to delineate the mechanism of ligand binding underlining recognition in pathogen receptors in innate immunity. Insights into requirements of conformation, charge, surface etc in the recognition and function of innate immunity receptors and their activation pathways, based on current data can suggest valuable avenues for future work.

    Description: One significant area of research in the multifaceted field of bilingualism over the past two decades has been the demonstration, validation, and account of the so-called ‘bilingual advantage’. This refers to the hypothesis that bilingual speakers have advanced abilities in executive functions and other domains of human cognition. Such cognitive benefits of bilingualism have an impact on the processing mechanisms active during language acquisition in a way that results in language variation. Within bilingual populations, the notion of language proximity (or linguistic distance) is also of key importance for deriving variation. In addition, sociolinguistic factors can invest the process of language development and its outcome with an additional layer of complexity, such as schooling, language, dominance, competing motivations, or the emergence of mesolectal varieties, which blur the boundaries of grammatical variants. This is particularly relevant for diglossic speech communities—bilectal, bidialectal, or bivarietal speakers. The defined goal of the present Research Topic is to address whether the bilingual advantage extends to such speakers as well. Thus, ‘Linguistic and Cognitive Profiles for Speakers of Linguistically Proximal Languages and Varieties’ become an important matter within ‘Developmental, Modal, and Pathological Variation’.

    Description: Attachment theory, assessment and research offers a broad, far-reaching view of human functioning, and it can enrich a psychologist's understanding of subjects and their relational adjustment, both in clinical and non-clinical settings. Ongoing research in attachment has led to a number of individual treatments and prevention and intervention programs. The assessment of an individual's attachment organization, can play a crucial role in explaining and previewing the unfolding treatment, the relational adjustments or concerns, and the psychological well-being. We hope to receive empirical papers that give evidence for the usefulness of attachment assessment in both clinical (e.g., patients with Eating Disorder; or Axis-II; psychotherapy patients…) or not clinical population (e.g. Adoptive and/or foster families or couples, Mother-infant assessment in prevention field…). These papers should include methodological issues and information about the participants, the methods used to assess attachment, the process of scorer training and the availability of the manual used to obtain inter-scorer reliability. Case studies may be of interest to the extent that they demonstrate the value of a systematic approach to attachment material. A range of theoretical perspectives is welcome as well presentation of new emergent tools on attachment. Because Frontiers in Psychology is an international journal, each empirical paper should comment on the international implications of the findings and discuss its cross-cultural use. Such comments may include, for example, its linguistic specificity, its robustness in translation, and the cross-cultural generalizability of the constructs and behaviors of the measure and its usual correlates. Cross-cultural generalizability is not, however, a requirement.

    Description: Music impinges upon the body and the brain. As such, it has significant inductive power which relies both on innate dispositions and acquired mechanisms and competencies. The processes are partly autonomous and partly deliberate, and interrelations between several levels of processing are becoming clearer with accumulating new evidence. For instance, recent developments in neuroimaging techniques, have broadened the field by encompassing the study of cortical and subcortical processing of the music. The domain of musical emotions is a typical example with a major focus on the pleasure that can be derived from listening to music. Pleasure, however, is not the only emotion to be induced and the mechanisms behind its elicitation are far from understood. There are also mechanisms related to arousal and activation that are both less differentiated and at the same time more complex than the assumed mechanisms that trigger basic emotions. It is imperative, therefore, to investigate what pleasurable and mood-modifying effects music can have on human beings in real-time listening situations. This e-book is an attempt to answer these questions. Revolving around the specificity of music experience in terms of perception, emotional reactions, and aesthetic assessment, it presents new hypotheses, theoretical claims as well as new empirical data which contribute to a better understanding of the functions of the brain as related to musical experience.

    Description: Mathematical anxiety is a feeling of tension, apprehension or fear which arises when a person is faced with mathematical content. The negative consequences of mathematical anxiety are well-documented. Students with high levels of mathematical anxiety might underperform in important test situations, they tend to hold negative attitudes towards mathematics, and they are likely to opt out of elective mathematics courses, which also affects their career opportunities. Although at the university level many students do not continue to study mathematics, social science students are confronted with the fact that their disciplines involve learning about statistics - another potential source of anxiety for students who are uncomfortable with dealing with numerical content. Research on mathematical anxiety is a truly interdisciplinary field with contributions from educational, developmental, cognitive, social and neuroscience researchers. The current collection of papers demonstrates the diversity of the field, offering both new empirical contributions and reviews of existing studies. The contributors also outline future directions for this line of research.

    Description: Multitasking refers to performance of multiple tasks. The most prominent types of multitasking are situations including either temporal overlap of the execution of multiple tasks (i.e., dual tasking) or executing multiple tasks in varying sequences (i.e., task switching). In the literature, numerous attempts have aimed at theorizing about the specific characteristics of executive functions that control interference between simultaneously and/or sequentially active component of task-sets in these situations. However, these approaches have been rather vague regarding explanatory concepts (e.g., task-set inhibition, preparation, shielding, capacity limitation), widely lacking theories on detailed mechanisms and/ or empirical evidence for specific subcomponents. The present research topic aims at providing a selection of contributions on the details of executive functioning in dual-task and task switching situations. The contributions specify these executive functions by focusing on (1) fractionating assumed mechanisms into constituent subcomponents, (2) their variations by age or in clinical subpopulations, and/ or (3) their plasticity as a response to practice and training.

    Description: A recent wave of brain research has advanced our understanding of the neural mechanisms of conscious states, contents and functions. A host of questions remain to be explored, as shown by lively debates between models of higher vs. lower-order aspects of consciousness, as well as global vs. local models. (Baars 2007; Block, 2009; Dennett and Cohen, 2011; Lau and Rosenthal, 2011). Over some twenty-five centuries the contemplative traditions have also developed explicit descriptions and taxonomies of the mind, to interpret experiences that are often reported in contemplative practices (Radhakrishnan & Moore, 1967; Rinbochay & Naper, 1981). These traditional descriptions sometimes converge on current scientific debates, such as the question of conceptual vs. non-conceptual consciousness; reflexivity or “self-knowing” associated with consciousness; the sense of self and consciousness; and aspects of consciousness that are said to continue during sleep. These real or claimed aspects of consciousness have not been fully integrated into scientific models so far. This Research Topic in Consciousness Research aims to provide a forum for theoretical proposals, new empirical findings, integrative literature reviews, and methodological improvements inspired by meditation-based models. We include a broad array of topics, including but not limited to: replicable findings from a variety of systematic mental practices; changes in brain functioning and organization that can be attributed to such practices; their effects on adaptation and neural plasticity; measurable effects on perception, cognition, affect and self-referential processes. We include contributions that address the question of causal attribution. Many published studies are correlational in nature, because of the inherent difficulty of conducting longitudinal experiments based on a major lifestyle decision, such as the decision to commit to a mental practice over a period of years. We also feature clinical and case studies, integrative syntheses and significant opinion articles.

    Description: Signaling through the cell surface antigen receptor is a hallmark of various stages of lymphocyte development and adaptive immunity. Besides the adaptive immune system, the innate immunity is equally important for protection. However, the mechanistic connection between signaling, chromatin changes and downstream transcriptional pathways in both innate and adaptive immune system remains incompletely understood in hematopoiesis. A related issue is how the enhancers communicate to the promoters in a stage specific fashion and in the context of chromatin. Because the factors that regulate chromatin are generally present and active in most cell types, how could cell type and/or stage specific chromatin architecture is achieved in response to a particular immune signal?The genetic loci that encode lymphocyte cell surface receptors are in an "unrearranged” or “germline” configuration during the early stages of development. Thus, in addition to expressing lineage and/or stage specific transcription factors during each developmental stage, lymphocytes also need to rearrange their cognate receptor loci in a strictly ordered fashion. Hence, there must be a tightly coordinated communication between the recombination machinery and the transcriptional machinery (including chromatin regulators) at every developmental step. Mature B cells also undergo classswitch recombination and somatic hypermutation. Importantly, along the way, these cells must avoid autoimmune responses and only those cells capable of recognizing foreignantigens are preserved to reach peripheral organs where they must function. The exquisite regulation that govern chromatin accessibility, recombination and transcription regulation in response to the environmental signals in the immune system is discussed here is a series of articles.

    Description: Nowadays, not only psychologists are interested in the study of Emotional Intelligence (EI). Teachers, educator, managers, employers, and people, in general, pay attention to EI. For example, teachers would like to know how EI could affect student’s academic results, and managers are concerned about how EI influences their employees’ performance. The concept of EI has been widely used in recent years to the extent that people start to applying it in daily life. EI is broadly defined as the capacity to process and use emotional information. More specifically, according to Mayer and Salovey, EI is the ability to: “1) accurate perception, appraise, and expression of emotion; 2) access and/or generation of feelings when they facilitate thought; 3) understand emotions and emotional knowledge; and 4) regulate emotions to promote emotional and intellectual growth” (Mayer and Salovey 1997, p. 10). When new information arises into one specific area of knowledge, the work of the scientists is to investigate the relation between this new information and other established concepts. In this sense, EI could be considered as a new framework to explain human behaviour. As a young concept in Psychology, EI could be used to elucidate the performance in the activities of everyday life. Over the past two decades, studies of EI have tried to delimitate how EI is linked to other competences. A vast number of studies have reported a relation between EI and a large list of competences such as academic and work success, life satisfaction, attendee to emotions, assertiveness, emotional expression, emotional-based decision making, impulsive control, stress management, among others. Moreover, recent researches have shown that EI plays an important role in the prediction of behaviour besides personality and cognitive factors. However, it is not until quite recently, that studies on EI have considered the importance of individual differences in EI and their interaction with cognitive abilities. The general issue of this Research Topic was to expose the role of individual differences on EI in the development of a large number of competencies that support a more efficient performance in people’s everyday life. The present Research Topic provide an extensive review that may give light to the better understanding of how individual differences in EI affect human behaviour. We have considered studies that analyse: 1) how EI contributes to emotional, cognitive and social process beyond the well-known contribution of IQ and personality traits, as well as the brain system that supports the EI; 2) how EI contributes to relationships among emotions and health and well-being, 3) the roles of EI during early development and the evaluation in different populations, 4) how implicit beliefs about emotions and EI influence emotional abilities.

    Description: More than 90% of diseases possess immunological abnormalities. Disorders such as inflammation, hypersensitivity, autoimmunity and immunodeficiency are simple examples of how the immune system misinterprets its surroundings and goes awry. Multiple sclerosis, rheumatoid arthritis, systemic lupus erythematosus, inflammatory bowel diseases, among many others are manifestations of immune cells attacking normal tissues. On the other hand, damping the immune system leads to diseases such as cancer, AIDS, and severe combined immunodeficiency. The last ten years witnessed an explosion in developing drugs that target the immune system. Several novel monoclonal antibodies have been approved for treatment of various diseases confirming that personalized medicine approach is robust in combating diseases. Hence, the future holds great promise for using personalized and targeted medicine rather than generalized medications that, in most circumstances, proven to be ineffective and characteristically exert side effects. Approaches such as generating novel adjuvants that can stimulate the immune system without harmful side effects, targeting inflammatory cytokines and chemokines, harnessing and activating innate immune cells such as natural killer cells or dendritic cells, are examples of future approaches to treat autoimmune diseases, AIDS, and various forms of cancer resulting from chronic inflammation. More recently, targeting immune checkpoint molecules have shown therapeutic response against lung cancer and melanoma. Identifying molecules involved in autophagy is another example of how personalized medicine might help treat patients with refractory asthma and autoimmune diseases. This topic introduces the reader to these novel approaches of manipulating the immune system and developing targeted therapeutic strategies for treatment of various diseases.

    Description: The PI3Ks control many key functions in immune cells. PI3Ks phosphorylate PtdIns(4,5)P2 to yield PtdIns(3,4,5)P3. Initially, PI3K inhibitors such as Wortmannin, LY294002 and Rapamycin were used to establish a central role for Pi3K pathway in immune cells. Considerable progress in understanding the role of this pathway in cells of the immune system has been made in recent years, starting with analysis of various PI3K and Pten knockout mice and subsequently mTOR and Foxo knockout mice. Together, these experiments have revealed how PI3Ks control B cell and T cell development, T helper cell differentiation, regulatory T cell development and function, B cell and T cell trafficking, immunoglobulin class switching and much, much more. The PI3Kd inhibitor idelalisib has recently been approved for the treatment of B cell lymphoma. Clinical trials of other PI3K inhibitors in autoimmune and inflammatory diseases are also in progress. This is an opportune time to consider a Research Topic considering when what we have learned about the PI3K signalling module in lymphocyte biology and how this is making an impact on clinical immunology and haematology.

    Description: The immunological synapse (IS) is a specialised cell-cell adhesion that mediates antigen acquisition and regulates the activation of lymphocytes. Initial studies of the IS showed a structure composed of stable supra-molecular activation clusters (SMAC) organised during the interaction of helper T lymphocytes with B lymphocytes, working as antigen presenting cells. A central SMAC of coalesced T cell receptors (TCRs) and a peripheral SMAC for cell-cell adhesion were observed. IS with similar structure was later described during antigen acquisition by B cells and during the interaction of NK cells with target and healthy cells. More recent research developed with microscopy systems that improve the spatial and temporal resolution has showed the complex molecular dynamics at the IS that governs lymphocyte activation. Currently, the IS is seen as a three-dimensional structure where signalling networks for lymphocyte activation and endosomal and cytoskeleton machinery are polarised. A view has emerged in which dynamic microclusters of signalling complexes are composed of molecular components attached to the plasma membrane and other components conveyed on sub-synaptic vesicles transported to the membrane by cytoskeletal fibers and motor proteins. Much information is nonetheless missing about how the dynamics of the endosomal compartment, the cytoskeleton, and signalling complexes are reciprocally regulated to achieve the function of lymphocytes. Experimental evidence also suggests that the environment surrounding lymphocytes exposed to different antigenic challenge regulates IS assembly and functional output, making an even more complex scenario still far from being completely understood. Also, although some signalling molecular components for lymphocyte activation have been identified and thoroughly studied, the function of other molecules has not been yet uncovered or deeply characterised. This research topic aims to provide the reader with the latest information about the molecular dynamics governing lymphocyte activation. These molecular dynamics dictate cell decisions. Thus, we expect that understanding them will provide new avenues for cell manipulation in therapies to treat different immune-related pathologies.

    Description: The pathogenic mechanisms underlying primary T-cell disorders are mainly related to molecular alterations of genes whose expression is intrinsic to hematopoietic cells. However, since the differentiation process requires a crosstalk among thymocytes and the thymic microenvironment, molecular alterations of genes, involved in the differentiation and functionality of the stromal component of the thymus, may lead to a severe T-cell defect or failure of central tolerance, as well. The first example of severe combined immunodeficiency (SCID) not related to an intrinsic alteration of the hematopoietic cell but rather of the thymic epithelial component is the Nude/SCID phenotype, inherited as an autosomal recessive disorder, whose hallmarks are the T-cell defect and the absence of the thymus. The clinical and immunological phenotype is the human equivalent of the murine Nude/SCID syndrome, which represents the first spontaneous SCID identified in nude mice in 1966. For over 3 decades studies of immune system in these mice enormously contributed to the overall knowledge of cell mediated immunity, in the assumption that the athymia of these mice was solely responsible for the T-cell immunological defect. This syndrome is due to mutations of the transcription factor FOXN1, belonging to the forkhead-box gene family, which is mainly expressed in the thymus and skin epithelial cells, where it plays a critical role in differentiation and survival. An alteration of the thymic structure is also a feature of the DiGeorge syndrome (DGS), which has been long considered the human counterpart of the nude mice phenotype. This syndrome is frequently associated to a deletion of the 22q11 region, which contains approximately 30 genes, including the TBX1 gene, which is responsible for most of the clinical features of DGS in humans and mice. In this syndrome common manifestations are cardiac malformations, speech delay, hypoparathyrodism and immunodeficiency, even though the immunological hallmarks of the T-cell defect in DiGeorge syndrome are profoundly different from those reported in human Nude/SCID. The divergence of the phenotype among these 2 entities raised the possibility that the FOXN1 transcription factor represents the real key stromal molecule implicated in directing the hematopoietic stem cell toward a proper T-cell fate. Thymic stromal component of the primary lymphoid organ is also required to negatively select the autoreactive clones, a process driven by the expression of tissue specific antigens (TSA) by medullary thymic epithelial cells (mTECs). The expression of genes encoding TSA antigens is mediated by autoimmune regulator (AIRE) gene, encoding a transcription factor expressed in mTECs. Molecular alterations of this gene are associated to autoimmune polyendocrinopathy candidiasis ectodermal dystrophy (APECED), a rare autosomal disorder, which may be considered the prototype of an autoimmune disease due to the failure of central tolerance homeostasis. All these "experiments of nature" led to unravel novel pathogenic mechanisms underlying inherited disorders of immune system and, of note, to clarify the pivotal role of epithelial cells in the maturation and education process of T-cell precursors.

    Description: In multicellular organisms, states with a high degree of tissue turnover like embryogenesis, development, and adult tissue homeostasis need an instantaneous, tightly regulated and immunologically silent clearance of these dying cells to ensure appropriate development of the embryo and adult tissue remodelling. The proper and swift clearance of apoptotic cells is essential to prevent cellular leakage of damage associated molecular patterns (DAMPs) which would lead to the stimulation of inflammatory cytokine responses. In addition to the clearance of apoptotic cells (efferocytosis), backup mechanisms are required to cope with DAMPs (HMGB-1, DNA, RNA, S100 molecules, ATP and adenosine) and other intracellular material (uric acid, intracellular proteins and their aggregates) released from cells, that were not properly cleared and have entered the stage of secondary necrosis. Furthermore, under certain pathologic conditions (e.g. gout, cancer, diabetes) non-apoptotic cell death may transiently occur (NETosis, necroptosis, pyroptosis) which generates material that also has to be cleared to avoid overloading tissues with non-functional cellular waste. Efficient efferocytosis is therefore indispensable for normal tissue turnover and homeostasis. The characterization of various signalling pathways that regulate this complex and evolutionary conserved process has shed light on new pathogenetic mechanisms of many diseases. Impaired clearance promotes initiation of autoimmunity as well as the perpetuation of chronic inflammation, but may also foster anti-tumor immunity under certain microenvironmental conditions. Immunological tolerance is continuously being challenged by the presence of post-apoptotic remnants in peripheral lymphoid tissues. Besides the autoimmune phenotype of chronic inflammatory rheumatoid disorders a plethora of pathologies have been associated with defects in genes involved in clearance, e.g. atherosclerosis, cancer, gout, diabetes, some forms of blindness, neuropathy, schizophrenia and Alzheimer’s disease. The main goal of this research topic is to collect contributions from various disciplines committed to studying pathogenetic mechanisms of the aforementioned disorders and dealing with alterations in the clearance of dying and dead cells, their remnants, and their constituents that leak out after membrane rupture. Integrating the combined collection of knowledge on efferocytosis and clearance of dead cells and their derived waste from different fields of research in physiology and pathophysiology could improve the molecular understanding of these increasingly prevalent diseases and may ultimately result in new therapeutic strategies.

    Description: Recognition and killing of aberrant, infected or tumor targets by Natural Killer (NK) cells is mediated by positive signals transduced by activating receptors upon engagement of ligands on target surface. These stimulatory pathways are counterbalanced by inhibitory receptors that raise NK cell activation threshold through negative antagonist signals. While regulatory effects are necessary for physiologic control of autoimmune aggression, they may restrain the ability of NK cells to activate against disease. Overcoming this barrier to immune surveillance, multiple approaches to enhance NK-mediated responses are being investigated since two decades. Propelled by considerable advances in the understanding of NK cell biology, these studies are critical for effective translation of NK-based immunotherapy principles into the clinic. In humans, dominant inhibitory signals are transduced by Killer Immunoglobulin Like Receptors (KIR) recognizing cognate HLA class I on target cells. Conversely, KIR recognition of “missing self-HLA” - due to HLA loss or HLA/ KIR mismatch - triggers NK-mediated tumor rejection. Initially observed in murine transplant models, these antitumor effects were later found to have important implications for the clinical outcome of haplotype-mismatched stemcell transplantation. Here, donor NK subsets protect against acute myeloid leukemia (AML) relapse through missing self recognition of donor HLA-C allele groups (C1 or C2) and/or Bw4 epitope. These studies were subsequently extended by trials investigating the antileukemia effects of adoptively transferred haplotype-mismatched NK cells in non-transplant settings. Other mechanisms have been found to induce clinically relevant NK cell alloreactivity in transplantation, e.g., post-reconstitution functional reversal of anergic NK cells. More recently, activating KIR came into the spotlight for their potential ability to directly activate donor NK cells through in vivo recognition of HLA or other ligands. Novel therapeutic monoclonal antibodies (mAb) may optimize NK-mediated effects. Examples include obinutuzumab (GA101), a glyco-engineered anti-CD20 mAb with increased affinity for the FcγRIIIA receptor, enhancing antibody-dependent cellular cytotoxicity; lirilumab (IPH2102), a first-in-class NK-specific checkpoint inhibitor, blocking the interaction between the major KIR and cognate HLA-C antigens; and elotuzumab (HuLuc63), a humanized monoclonal antibody specific for SLAMF7, whose anti-myeloma therapeutic effects are partly due to direct activation of SLAMF7-expressing NK cells. In addition to conventional antibodies, NK cell-targeted bispecific (BiKEs) and trispecific (TriKEs) killer engagers have also been developed. These proteins elicit potent effector functions by binding target ligands (e.g., CD19, CD22, CD30, CD133, HLA class II, EGFR) on one arm and NK receptors on the other. An additional innovative approach to direct NK cell activity is genetic reprogramming with chimeric antigen receptors (CAR). To date, primary NK cells and the NK92 cell line have been engineered with CAR specific for antigens expressed on multiple tumors. Encouraging preclinical results warrant further development of this approach. This Research Topic welcomes contributions addressing mechanisms of NK-mediated activation in response to disease as well as past and contemporary strategies to enhance NK mediated reactivity through control of the interactions between NK receptors and their ligands.

    Description: Evaluations of intervention programs seek to present high-quality design, measures and data to assess their merit and worth. While evaluations differ in their purpose, theoretical framework and methodology, their collective aim is to obtain relevant and meaningful information to inform practice, research, and policy. As such, evaluation findings serve to build a body of knowledge on effective approaches to promote designated psychological outcomes, critical to an individual's overall health and well-being. However, as examined in this e-book, methodological weaknesses directly limit the potential of evaluations of intervention programs. As discussed by Chacón-Moscoso and Sanduvete-Chaves, methodological weaknesses can be attributed to how to define and measure methodological quality and the contextual dependency of instruments designed to measure this quality. In response, this e-book provides a collection of studies on methodological approaches to promote the quality of psychological interventions. Specifically, 10 original works published in the Research Topic Methodological Quality of Interventions in Psychology are included. The papers are organized into two chapters. Concretely, Chapter 1 includes studies pertaining to methodological approaches to enhance the quality of psychological intervention, being context independent solutions. Furthermore, Chapter 2 presents original work in different areas (health, education, sport and social welfare) where methodological quality has been better assessed. Collectively, the papers in this e-book serve to expand the awareness of practitioners and researchers interested in psychological interventions of the critical role of methodological quality in this work.     Description: Successful containment of an infection is dependent on both innate and adaptive immune response. Cytokines are essential effectors of both of these systems. In particular, type I interferons (IFN-I) are important components of early innate immunity against an infection. However, the production of IFN-I could serve as a double edge sword, either containing an infection or enhancing susceptibility. For example, IFN-I, which is essential for early containment of viral infections, has been shown to be detrimental to the host during bacterial infections. In fact, recent significant reports have shown that influenza virus induced IFN-I responses can enhance the host susceptibility to secondary bacterial infections. These recent reports highlight the expanding immunoregulatory role of IFN-I in the host immunity. With these recent findings in mind, the aim of this research topic is to welcome novel data, opinion and literature reviews on the newly identified dual functions of IFN-I. This research topic wills focus on the following areas of IFN-I: 1) a detrimental role of IFN-I during primary bacterial infection; 2) a detrimental role of viral infection induced IFN-I during secondary bacterial infections; 3) evolutionary pressure that drove detrimental IFN-I response during primary bacterial infection; and 4) does benefit of IFN-I responses during primary viral infections outweigh the adverse consequences of IFN-I mediated enhanced susceptibility to secondary bacterial infections.

    Description: Model building is typically based on the identification of a set of established facts in any given field of research, insofar as the model is then evaluated on how well it accounts for these facts. Psychology – and specifically visual word identification and reading – is no exception in this sense (e.g., Amenta & Crepaldi, 2012; Coltheart et al., 2001; Grainger & Jacobs, 1996). What counts as an established fact, however, was never discussed in great detail. It was typically considered, for example, that experimental effects need to replicate across, e.g., individuals, experimental settings, and languages if they are to be believed. The emphasis was on consistency, perhaps under a tacit assumption that the universal principles lying behind our cognitive structures determine our behaviour for the most part (or at least for that part that is relevant for model building). There are signs that a different approach is growing up in reading research. On a theoretical ground, Dennis Norris’ Bayesian reader (2006, 2009) has advanced the idea that models can dispense of static forms of representation (i.e., fixed architectures), and process information in a way that is dynamically constrained by context-specific requirements. Ram Frost (2012) has focused on language-specific constraints in the development of general theories of reading. On an empirical ground, the most notable recent advance in visual word identification concern the demonstration that some previously established (in the classic sense) effects depend heavily on language (Velan and Frost, 2011), task (e.g., Duñabeitia et al., 2011; Marelli et al., 2013; Kinoshita and Norris, 2009), or even individual differences (Andrews & Lo, 2012, 2013). Variability has become an intrinsic and informative aspect of cognitive processing, rather than a sign of experimental weakness. This Research Topic aims at moving forward in this new direction by providing an outlet for experimental and theoretical papers that: (i) explore more in depth the theoretical basis for considering variability as an intrinsic property of the human cognitive system; (ii) highlight new context-dependent experimental effects, in a way that is informative on the dynamics of the underlying cognitive processing; (iii) shed new light on known context-dependent experimental effects, again in a way that enhances their theoretical informativeness.

    Description: Plasticity is the hallmark of stem cells. At the same time, stem cells, like any other cell type, are influenced by their microenvironment and respond to it accordingly. A specific microenvironment is defined by a variety of factors, including biological and chemical factors, cell-cell interactions, but also metabolic and mechanical cues. Such dynamic and specialized microenvironment where the stem cells reside is considered a stem cell niche. Tissue injury as well as malignant tissue alterations lead to changes in the niche influencing the plasticity and biology of residing stem cells. Similarly, the niche changes upon tissue damage, which eventually induces differentiation of stem cells and ultimately regeneration of the tissue.

    Description: NETosis, a form of cell death that manifests by the release of decondensed chromatin to the extracellular space, provides valuable insights into mechanisms and consequences of cellular demise. Because extracellular chromatin can immobilize microbes, the extended nucleohistone network was called a neutrophil extracellular trap (NET), and the process of chromatin release was proposed to serve an innate immune defense function. Extracellular chromatin NETs were initially observed in studies of neutrophils and are most prominent in these types of granulocytes. Subsequent studies showed that other granulocytes and, in a limited way, other cells of the innate immune response may also release nuclear chromatin following certain kinds of stimulation. Variations of NETosis were noted with cells that remain temporarily motile after the release of chromatin. Numerous stimuli for NETosis were discovered, including bacterial breakdown products, inflammatory stimuli, particulate matter, certain crystals, immune complexes and activated thrombocytes. Fundamental explorations into the mechanisms of NETosis observed that neutrophil enzyme activity (PAD4, neutrophil elastase, proteinase 3 and myeloperoxidase) and signal transduction pathways contribute to the regulation of NETosis. Histones in NET chromatin become modified by peptidylarginine deiminase 4 (PAD4) and cleaved at specific sites by proteases, leading to extensive chromatin externalization. In addition, NETs serve for attachment of bactericidal enzymes including myeloperoxidase, leukocyte proteases, and the cathelicidin LL-37. NETs are decorated with proteases and may thus contribute to tissue destruction. However, the attachment of these enzymes to NET-associated supramolecular structures restricts systemic spread of the proteolytic activity. While the benefit of NETs in an infection appears obvious, NETs also participate as key protagonists in various pathologic states. Therefore, it is essential for NETs to be efficiently cleared; otherwise digestive enzymes may gain access to tissues where inflammation takes place. Persistent NET exposure at sites of inflammation may lead to a further complication: NET antigens may provoke acquired immune responses and, over time, could initiate autoimmune reactions, serve as antigen for nuclear autoantibodies and foster DNA immune complex-related inflammation. Neutrophil products and deiminated proteins comprise an important group of autoantigens in musculoskeletal disorders. Aberrant NET synthesis and/or clearance are often associated with inflammatory and autoimmune conditions. Recent evidence also implicates aberrant NET formation in the development of endothelial damage, atherosclerosis and thrombosis. Intravital microscopy provides evidence for conditions that induce NETosis in vivo. Furthermore, NETs can easily be detected in synovial fluid and tissue sections of patients with arthritis and gout. NETosis is thus of interest to researchers who investigate innate immune responses, host-pathogen interactions, chronic inflammatory disorders, cell and vascular biology, biochemistry, and autoimmunity. As we enter the second decade of research on NETosis, it is useful and timely to review the mechanisms and pathways of NET formation, their role in bacterial and fungal defense and their importance as inducers of autoimmune responses.

    Description: Early studies recognized the unique phenotype and attributes of T cells found in mucosal tissues, such as the intestines, skin, lung and female reproductive tract. This special topic issue will cover many aspects of mucosal-resident T cell biology during infection and disease and is dedicated to Leo Lefrancois, a pioneer in this field who recently passed away. A major proportion of these mucosal T cells are memory T cells, now recognized as a major constituent of memory T cells referred to as tissue-resident memory T cells. Unlike central and effector memory T cell subsets, tissue-resident memory T cells exhibit tissue specificity with minimal systemic migration. Nonetheless, tissue-resident memory T cells share a similar origin and display some overlapping phenotypes with their other memory T cell counterparts. Articles in this issue will describe the different types of memory T cells residing in mucosal tissues, their origins and functions as well as how they vary among discrete mucosal sites. Manuscripts will consider the unique physiological environments and cellular constituents which facilitate tissue residency while preserving tissue function. Additionally, there will be descriptions of the various mechanisms responsible for the migration and segregation of tissue resident memory CD8 T cells from the peripheral T cell pool. Although the mechanisms facilitating the sequestration of tissue-resident memory T cells within a respective tissue has not well characterized, various theories will also be discussed. Lastly, how these T cells contribute to immunity to pathogens, cancer, and autoimmunity and could be modified through vaccination or therapeutic intervention will be described. As mucosal tissues are the major portals of pathogen entry and frequent transformation, the activities and persistence of tissue resident memory T cells is crucial for mediating protection at these sites.

    Description: Powerful and economic sensors such as high definition cameras and corresponding recognition software have become readily available, e.g. for face and motion recognition. However, designing user interfaces for robots, phones and computers that facilitate a seamless, intuitive, and apparently effortless communication as between humans is still highly challenging. This has shifted the focus from developing ever faster and higher resolution sensors to interpreting available sensor data for understanding social signals and recognising users' intentions. Psychologists, Ethnologists, Linguists and Sociologists have investigated social behaviour in human-human interaction. But their findings are rarely applied in the human-robot interaction domain. Instead, robot designers tend to rely on either proof-of-concept or machine learning based methods. In proving the concept, developers effectively demonstrate that users are able to adapt to robots deployed in the public space. Typically, an initial period of collecting human-robot interaction data is used for identifying frequently occurring problems. These are then addressed by adjusting the interaction policies on the basis of the collected data. However, the updated policies are strongly biased by the initial design of the robot and might not reflect natural, spontaneous user behaviour. In the machine learning approach, learning algorithms are used for finding a mapping between the sensor data space and a hypothesised or estimated set of intentions. However, this brute-force approach ignores the possibility that some signals or modalities are superfluous or even disruptive in intention recognition. Furthermore, this method is very sensitive to peculiarities of the training data. In sum, both methods cannot reliably support natural interaction as they crucially depend on an accurate model of human intention recognition. Therefore, approaches to social robotics from engineers and computer scientists urgently have to be informed by studies of intention recognition in natural human-human communication. Combining the investigation of natural human behaviour and the design of computer and robot interfaces can significantly improve the usability of modern technology. For example, robots will be easier to use by a broad public if they can interpret the social signals that users spontaneously produce for conveying their intentions anyway. By correctly identifying and even anticipating the user's intention, the user will perceive that the system truly understands her/his needs. Vice versa, if a robot produces socially appropriate signals, it will be easier for its users to understand the robot's intentions. Furthermore, studying natural behaviour as a basis for controlling robots and other devices results in greater robustness, responsiveness and approachability. Thus, we welcome submissions that (a) investigate how relevant social signals can be identified in human behaviour, (b) investigate the meaning of social signals in a specific context or task, (c) identify the minimal set of intentions for describing a context or task, (d) demonstrate how insights from the analysis of social behaviour can improve a robot's capabilities, or (e) demonstrate how a robot can make itself more understandable to the user by producing more human-like social signals.

An important factor that has exacerbated the epidemiological picture in Brazil, Colombia and Venezuela is infection after the oral intake of contaminated foods such as sugar cane, açai and bacaba juices. Trypanosoma cruzi is an intracellular protozoan that exhibits a complex life cycle, involving multiple developmental stages found in both vertebrate and invertebrate hosts. In vertebrate hosts, the trypomastigote form invades a large variety of nucleated cells using multiple mechanisms. The invasion process involves several steps: (a) attraction of the protozoan to interact with the host cell surface; (b) parasite-host cell recognition; (c) adhesion of the parasite to the host cell surface; (d) cell signalling events that culminate in the internalization of the parasite through endocytic processes; (e) biogenesis of a large vacuole where the parasite is initially located, and is also known as parasitophorous vacuole (PV); (f) participation of endocytic pathway components in the internalization process; (g) participation of cytoskeleton components in the internalization process; (h) transformation of the trypomastigote into the amastigote form within the PV; (i) lysis of the membrane of the PV; (j) multiplication of amastigotes within the host cell in direct contact with cell structures and organelles; (k) transformation of amastigotes into trypomastigotes, and (l) rupture of the host cell releasing trypomastigotes into the extracellular space. The kinetics of the interaction process and even the fate of the parasite within the cell vary according to the nature of the host cell and its state of immunological activation.

    Description: Over the last years it has become evident that many neurological diseases of the central nervous system (CNS) are induced by a specific adaptive immune response directed against molecules expressed on CNS-resident cells. Well-recognized examples are anti-N-Methyl-D-Aspartate Receptor (NMDAR) encephalitis which is characterized by the presence of antibodies against neuron-expressed NMDAR, or neuromyelitis optica (NMO), induced by antibodies to astrocyte-expressed aquaporin-4. Many more examples exist, and antibodies, and T or/and B cells have increasingly been associated with CNS disease. Often the symptoms of these diseases have not been typically reported to have an immune aetiology. Beside classical neurological symptoms like ataxia, vision disturbance, and motor or sensory symptoms, these can include cognitive disturbances, behavioral abnormalities, or/and epileptic seizures. Although much has been learned regarding the pathophysiology of prototypic examples of these disorders, there are still major gaps in our understanding of their biology. This may be due to the fact that they are rare diseases, and their therapies are still very limited. This research topic includes contributions addressing the analysis of the adaptive immune response driving disease including target antigens, molecular epitope mapping, and factors involved in the disease pathogenesis such as complement activation cascades, genetic and genomic regulation, as well as environmental triggers. Diagnostic criteria and methods, and treatment are also discussed. The overall aim of the volume is to review progress in our pathophysiological understanding of immune-mediated CNS disorders in order to advance diagnostic and therapeutic approaches, and ultimately improve outcomes for patients.

    Description: The mononuclear phagocyte system (MPS) comprises dendritic cells (DCs), monocytes and macrophages (MØs) that together play crucial roles in tissue immunity and homeostasis, but also contribute to a broad spectrum of pathologies. They are thus attractive therapeutic targets for immune therapy. However, the distinction between DCs, monocytes and MØ subpopulations has been a matter of controversy and the current nomenclature has been a confounding factor. DCs are remarkably heterogeneous and consist of multiple subsets traditionally defined by their expression of various surface markers. While markers are important to define various populations of the MPS, they do not specifically define the intrinsic nature of a cell population and do not always segregate a bona fide cell type of relative homogeneity. Markers are redundant, or simply define distinct activation states within one subset rather than independent subpopulations. One example are the steady-state CD11b+ DCs which are often not distinguished from monocytes, monocyte-derived cells, and macrophages due to their overlapping phenotype. Lastly, monocyte fate during inflammation results in cells bearing the phenotypic and functional features of both DCs and MØs significantly adding to the confusion. In fact, depending on the context of the study and the focus of the laboratory, a monocyte-derived cell will be either be called "monocyte-derived DCs" or "macrophages". Because the names we give to cells are often associated with a functional connotation, this is much more than simple semantics. The "name" we give to a population fundamentally changes the perception of its biology and can impact on research design and interpretation. Recent evidence in the ontogeny and transcriptional regulation of DCs and MØs, combined with the identification of DC- and MØ-specific markers has dramatically changed our understanding of their interrelationship in the steady state and inflammation. In steady state, DCs are constantly replaced by circulating blood precursors that arise from committed progenitors in the bone marrow. Similarly, some MØ populations are also constantly replaced by circulating blood monocytes. However, others tissue MØs are derived from embryonic precursors, are seeded before birth and maintain themselves in adults by self-renewal. In inflammation, such differentiation pathways are fundamentally changed and unique monocyte-derived inflammatory cells are generated. Current DC, monocyte and MØ nomenclature does not take into account these new developments and as a consequence is quite confusing. We believe that the field is in need of a fresh view on this topic as well as an upfront debate on DC and MØ nomenclature. Our aim is to bring expert junior and senior scientists to revisit this topic in light of these recent developments. This Research Topic will cover all aspects of DC, monocyte and MØ biology including development, transcriptional regulation, functional specializations, in lymphoid and non-lymphoid tissues, and in both human and mouse models. Given the central position of DCs, monocytes and MØs in tissue homeostasis, immunity and disease, this topic should be of interest to a large spectrum of the biomedical community.

    Description: Early experience plays a crucial role in determining the trajectory of cognitive development. For example, early sensory deprivation is known to induce neural reorganization by way of adaptation to the altered sensory experience. Neville and Bavelier’s “compensatory theory’’ hypothesizes that loss of one sense may bring about a sensory enhancement in the remaining modalities. Sensory deprivation will, however, also impact the age of emergence, or the speed of acquisition of cognitive abilities that depend upon sensory inputs. Understanding how a child’s early environment shapes their cognition is not only of theoretical interest. It is essential for the development of early intervention programs that address not just the early deprivation itself, but also the cognitive sequelae of such deprivation. The articles in this e-book all address different aspects of deprivation - sensory, linguistic, and social - and explore the impacts of such deprivation on a wide range of cognitive outcomes. In reading these contributions, it is important to note that sensory, linguistic, and social deprivation are not independent factors in human experience. For example, a child born deaf into a hearing family is likely to experience delays in exposure to natural language, with subsequent limits on their linguistic competence having an effect on social interactions and inclusion: a child raised in environments where social interaction is highly limited is also likely to experience reductions in the quantity and quality of linguistic inputs. Future work will need to carefully examine the complex interactions between the sensory, linguistic and social environments of children raised in atypical or impoverished environments.

    Description: In the ancient past, cocoa has been appreciated as a high-calorie food to boost energy in soldiers and for its undefined medicinal and mystical properties. During other times, chocolate has been considered as the forbidden “food of God”: a treasure of pleasure for the mind and the soul. The overall perception of the consumer for chocolate was of a “charming” and appealing food with lots of negative aspects related to high sugar content leading to consider chocolate as “junk food” for its “obesigen” calories. Recently, in association with the renewed interest of nutrition science in alternative source of health-promoting foods and ingredients, a large body of research has been conducted to unravel the pro and cons of cocoa in relation to human health. Epidemiological evidences indicate that cocoa consumption helps preventing cardiovascular disease for its high content in bioactive flavonoids. Clinical trials show that chocolate consumption might improve vascular function, decreasing platelet aggregation and display an antioxidant and anti-inflammatory effect. The putative protective action of cocoa seems to be multi-factorial and involving different aspects of vascular, antioxidant and endothelial function. However, the mechanism(s) that account for the benefits of cocoa it is still unclear. The aim of this Research Topic is therefore to provide the reader with an objective picture of the state of art on the association between cocoa and health, mainly through the evidences of human trials; overwhelmingly considered the golden standard for nutritional science. The Research Topic will cover the analysis of the manufacturing processes of the chocolate and the antioxidant effects in humans as well as the majority of the putative health effects of chocolate and cocoa, such as anti-inflammatory properties, effect on immunity, platelet aggregation, blood pressure, endothelial function and cognitive behavior. Unraveling the functional properties of cocoa will help to understand if the 'food of God' is a primordial gift for the health of mankind.

    Description: This Research Topic aims to showcase the state of the art in language research while celebrating the 25th anniversary of the tremendously influential work of the PDP group, and the 50th anniversary of the perceptron. Although PDP models are often the gold standard to which new models are compared, the scope of this Research Topic is not constrained to connectionist models. Instead, we aimed to create a landmark forum in which experts in the field define the state of the art and future directions of the psychological processes underlying language learning and use, broadly defined.     Description: In the Research Topic "History of Chemoattractant Research" we will portray some of the key discoveries that helped to transform cell migration research into a global playing field within immunology (and beyond). Early progress had a profound effect on both, academia and industry. Today, numerous academic laboratories are fully engaged in compiling a detailed road map describing the highly complex network of immune and tissue cells that respond to chemoattractants. Industrial research, on the other hand, centers on drugs that interfere with immune cell traffic in inflammatory diseases and cancer. The following series of “short stories” provide personal accounts on key discoveries. The individual molecular discoveries enabled numerous research laboratories worldwide to unravel their significance in steady-state or pathological immune processes. Although ground-breaking in their own right, it is therefore worth emphasizing that rapid progress in chemoattractant research was made possible by many other laboratories who were not directly involved in the original discovery process. Therefore, the authors of this mini-series are discussing their findings in the context of time, place and subsequent progress enabled by their discoveries.     Description: Embodied cognition represents one of most important research programs in contemporary cognitive science. Although there is a diversity of opinion concerning the nature of embodiment, the core idea is that cognitive processes are influenced by body morphology, emotions, and sensorimotor systems. This idea is supported by an ever increasing collection of empirical studies that fall into two broad classes: one consisting of experiments that implicate action, emotion, and perception systems in seemingly abstract cognitive tasks and the other consisting of experiments that demonstrate the contribution of bodily interaction with the external environment to the performance of such tasks. Now that the research program of embodied cognition is well established, the time seems right for assessing its further promise and potential limitations. This research topic aims to create an interdisciplinary forum for discussing where we go from here. Given that we have good reason to think that the body influences cognition in surprisingly robust ways, the central question is no longer whether or not any cognitive processes are embodied. Instead, other questions have come to the fore: To what extent are cognitive processes in general embodied? Are there disembodied processes? Among those that are embodied, how are they embodied? Is there more than one kind of embodiment? Is embodiment a matter of degree? There are a number of specific issues that could be addressed by submissions to this research topic. Some supporters of embodied cognition eschew representations. Should anti-representationalism be a core part of an embodied approach? What role should dynamical models play? Research in embodied cognition has tended to focus on the importance of sensorimotor areas for cognition. What are the functions of multimodal or amodal brain areas? Abstract concepts have proved to be a challenge for embodied cognition. How should they be handled? Should researchers allow for some form of weak embodiment? Currently, there is a split between those who offer a simulation-based approach to embodiment and those who offer an enactive approach. Who is right? Should there be a rapprochement between these two groups? Some experimental and robotics researchers have recently shown a great deal of interest in the idea that external resources such as language can serve as form of cognitive scaffolding. What are the implications of this idea for embodied cognition? This research topic aims to bring together empirical and theoretical work from a diversity of perspectives. Submissions are sought from any of the major disciplines associated with cognitive science, including but not necessarily limited to anthropology, cognitive psychology, computational modeling, linguistics, neuroscience, philosophy, robotics, and social psychology.     Description: The pervasive idea that madness and creativity are intricately linked is one that holds tremendous fascination for both scientists and the general public alike. Although this view was at first largely driven by anecdotal evidence showcasing the manifestation of mental illness in individuals who exhibited extraordinary levels of creativity in various spheres of life, it initiated a strong impetus to empirically investigate the association between mental health and creativity. A variety of approaches (and combinations of approaches) have been adopted to address this association including clinical, personality, psychometric, behavioral, cognitive, historiometric and neuroscientific. Despite the ever accumulating body of evidence over the past six decades investigating this link, what is lacking is a comprehensive overview of the disparate findings from these different approaches that will enable us to address the question of whether there is an empirically founded relationship between creativity and mental illness. And if such a link does exist, what is the nature of this association? The purpose of this Research Topic was to motivate theorists and researchers to answer this question (or at least attempt to do so) given the available evidence thus far. The themes of interest that were open to exploration in view of this topic included: (a) Which mental disorders are positively associated with creativity? (b) Which mental disorders are negatively associated with creativity? (c) The dynamics of information processing biases (positive versus negative) associated with psychiatric and high-risk populations (d) Theories regarding the madness-creativity link (e) Personality-based studies on creativity (f) Creativity, mental illness and the brain (g) Genes and creativity (h) How can studies on neurological populations inform this debate? (i) What are the areas of impact with regard to real world applications and practice? (j) Historical timeline of this question (k) Evolutionary perspectives on the madness-creativity link (l) Methodological problems associated with this field (m) Philosophical issues to bear in mind when investigating this domain (n) The usefulness of the “troubled genius” concept The invitation to contribute was open to all interested academics regardless of whether they were seasoned explorers within this field of study or just beginning to get their feet wet in its murky waters. As a result of adopting this inclusive approach, the contributions showcase a wide variety of perspectives from academic departments and institutions the world over. What is most encouraging is that so many were willing to openly take on the challenge of tackling this difficult question head on. We hope future discussions that follow through as a result of this collective effort will prove to be just as fruitful.

The cohering theme of this publication revolves around the question of how people can establish meaningful lives and meaningful work experiences in light of the many challenges that are reducing access to decent work. Another essential contextual factor that is explored in this volume is the Decent Work Agenda (International Labour Organization, 2008), which represents an initiative by the International Labour Organization. In this book, we hope to enrich the Decent Work Agenda by infusing the knowledge and perspectives of psychology into contemporary discourses about work, and well-being. Another inspiration for this project emerged from the UNESCO Chair in Lifelong guidance and counseling, recently established in Poland in 2013 under the leadership of Jean Guichard, which has focused on advancing research and policy advocacy about decent work. This new era calls for an innovative perspective in constructing decent work and decent lives: the passage from the paradigm of motivation to the paradigm of meaning, where the sustainability of the decent life project is anchored to a meaningful construction. During this period when work is changing so rapidly, leaving people yearning for a sense of connection and meaning, it’s fundamental to create a framework for an explicitly psychological analysis of decent work.

    Description: There is an extensive literature conducted from a range of theoretical perspectives and methodologies on the role of groups and student learning in higher education. However here the concept of the ‘group’ is heavily contested at a theoretical level but within higher education practice, characterizing the group has tended to be clear cut. Groups of students are often formed within the parameters of specific educational programs to address explicitly defined learning objectives. These groups are often small scale and achieve tasks through cooperative or collaborative learning. Cooperative learning involves students dividing roles and responsibilities between group members, so learning becomes an independent process and outcome. On the other hand, collaborative learning involves students working together by developing shared meanings and knowledge to solve a task or problem. From this perspective, learning is conceptualized as both a social process and individual outcome. That is, collaborative learning may facilitate individual student conceptual understanding and hence lead to higher academic achievement. The empirical evidence is encouraging as has been shown that students working collaboratively tend to achieve higher grades than students working independently. However the above perspectives on student engagement assume that groups are formed within the confines of formal learning environments (e.g. lecture theaters), involve students on the same degree program, have the explicit function of achieving a learning task and disband once this has been achieved. However, students may also use existing social networks such as friendship groups as a mechanism for learning, which may occur outside of formal learning environments. There is an extensive literature on the role and benefits of friendship groups on student learning within primary and secondary education but there is a distinct lack of research within higher education. This ebook is innovative and ambitious and will highlight and consolidate, the current understanding of the role that student based engagement behaviors may serve in effective pedagogy. A unique aspect of this research topic will be the fact that scholars will also be welcome to submit articles that describe the efficacy of the full range of approaches that have been employed to facilitate student engagement across the sector.

    Description: Learning to read, and to spell are two of the most important cultural skills that must be acquired by children, and for that matter, anyone learning a second language. We are not born with an innate ability to read. A reading system of mental representations that enables us to read must be formed in the brain. Learning to read in alphabetic orthographies is the acquisition of such a system, which links mental representations of visual symbols (letters) in print words, with pre-existing phonological (sound) and semantic (comprehension) cognitive systems for language. Although spelling draws on the same representational knowledge base and is usually correlated with reading, the acquisition processes involved are not quite the same. Spelling requires the sequential production of letters in words, and at beginning levels there may not be a full degree of integration of phonology with its representation by the orthography. Reading, on the other hand, requires only the recognition of a word for pronunciation. Hence, spelling is more difficult than reading, and learning to spell may necessitate more complete representations, or more conscious access to them. The learning processes that children use to acquire such cognitive systems in the brain, and whether these same processes are universal across different languages and orthographies are central theoretical questions. Most children learn to read and spell their language at the same time, thus the co-ordination of these two facets of literacy acquisition needs explication, as well as the effect of different teaching approaches on acquisition. Lack of progress in either reading and/or spelling is also a major issue of concern for parents and teachers necessitating a cross-disciplinary approach to the problem, encompassing major efforts from researchers in neuroscience, cognitive science, experimental psychology, and education. The purpose of this Research Topic is to summarize and review what has been accomplished so far, and to further explore these general issues. Contributions from different perspectives are welcomed and could include theoretical, computational, and empirical works that focus on the acquisition of literacy, including cross-orthographic research.

    Description: It is impossible to perceive the innumerable stimuli impinging on our senses, all at once. Out of the myriad stimuli, external and internal, a few are selected for further processing; and even among these, we try to put each in some sort of relation with the others, to be able to make some sense about them all. Time, of course, is an elementary dimension we use to organize our experiences. Thus, the perception of sequences is basic to human cognition. Nevertheless, research addressing sequences is rather sparse. Partly, this is due to difficulty in designing experiments in this area due to huge individual differences. Then, there is the assumption that temporal order has more to do with memory than perception. Another problem is that sequences seem endemic to the auditory world. So much so that some researchers have suggested that sound provides the ‘auditory scaffolding’ for sequencing behavior. Little wonder that research studies addressing sequences in modalities other than audition are extremely rare.This research topic aimed to gather a holistic picture of sequencing behaviour among humans by collecting snapshots of the current research on the topic of sequencing. We particularly sought contributions which addressed sequences beyond the auditory modality. The single unifying criteria for these diverse contributions was that they shed new light on previously unexplored empirical relationships and/or provoked new lines of research with incisive ideas regarding sequencing behavior. Seasoned researchers contributed their views on perception, memory, and production of sequences.

In this context, this Research Topic is aimed to offer a snapshot of the present knowledge and questions raised in this field.     Description: In an attempt to cease from reducing the world and its phenomena to linear modeling and analytic dissection, Dynamic Systems Theories (DST) and Embodiment theories and methods aim to account for the complex, dynamic, and non-linear phenomena that we constantly deal with in psychology. For instance, a DST and Embodiment perspective can enrich psychology’s understanding of communicative processes both in clinical and non-clinical settings. In psychotherapy, research has shown that there are a number of common factors contributing to psychotherapy outcome, of which the therapeutic relationship is the most important one. These findings give communication a central role in the psychotherapy process. In the traditional view, the underlying model of understanding psychotherapy processes is that of a number of components summatively coming together enabling us to make a linear causal prediction. Yet, communication is inherently dynamic. A shift to viewing the communication process in psychotherapy as a field dynamic phenomenon helps us to take into account nonlinear phenomena, such as feedback processes within and between persons. We thus propose an embodied enactive dynamic systems view as a new theoretical and methodological perspective that can more realistically capture what happens among and between two persons in psychotherapy. This view is broader than that of most current models in psychotherapy research. DST and Embodied Enactive Approaches can offer solutions to the prevailing neglect of non-linear phenomena in Western science, to better account for the complex dynamics of reality, and to move to a more holistic level of analysis. DST and Embodied Enactive Approaches have developed not in a single discipline but in a joined movement based on various fields such as physics, biology, robotics, anthropology, philosophy, linguistics, neuroscience, and psychology, and have only recently entered clinical theorizing. The two new paradigms are presently triggering a rethinking of the therapeutic process by recognizing the embodied nature of psychological and communicative phenomena. Their integration opens up a promising scenario in the field of psychotherapy research, developing new, profoundly transdisciplinary, theoretical concepts, methodologies, and standards of knowledge. The notion of field dynamics enables us to account for the role of the communicational context in the regulation of intra-psychological processes, while at the same time avoiding the pitfalls of an ontologization of the hierarchy of systemic organization. Moreover, the new approach implements methodological strategies that can transcend the conventional opposition between idiographic and nomothetic sciences.

    Description: Mammalian pregnancy represents a unique immunological riddle in that the mother does not reject her allogeneic fetus. In part this is largely due to a general sequestration or diminution of T cell activity, and an increased involvement of the innate immune system. The field of immunology is concerned primarily with how innate and adaptive mechanisms collaborate to protect vertebrates from infection. Although many cellular and molecular actors have evidently important roles, antibodies and lymphocytes are considered to be the principal players. Yet despite their importance, it would be definitely simplistic to conclude that they are solely essential for immunity overall. A major distinction between adaptive and innate immunity is the spontaneity of the innate immune response, which utilizes an already pre-existing but limited repertoire of responding modules. The slower onset of adaptive immunity compensates by its ability to recognize a much broader repertory of foreign substances, and also by its power to constantly improve during a response, whereas innate immunity remains relatively unaffected. The interactions between the reproductive system and the immune system are of particular interest, since the reproductive system is unique in that its primary role is to assure the continuity of the species, while the immune system provides internal protection and thus facilitates continued health and survival. The modus operandi of these two morphologically diffuse systems involves widely distributed chemical signals in response to environmental input, and both systems must interact for the normal functioning of each. Furthermore, dysregulation of normal physiological interactions between the reproductive and immune systems can lead to severe pregnancy-related disorders or complications. On the other hand, by ameliorating auto-inflammatory conditions such as MS and RA, pregnancy may provide a unique insight into novel immune modulatory strategies. The scientific focus on reproductive–immune research has historically provided substantial insight into the interface between these two physiological systems. A translational research approach would involve a tight interaction between diverse scientific and clinical disciplines including immunology, obstetrics, haematology, haemostasis and endocrinology. With so much recent progress in the field, we believe that it is valuable and well-timed to review the broad variety of the relevant physiologic and pathologic aspects – from menstruation to fertilization and implantation, and from placentation and pregnancy per se to the post partum condition - in which the immune system takes part. We are looking forward to a wide and vivid discussion of these and related issues, and we sincerely expect that our readers profoundly benefit from new exciting insights and fruitful collaborations.

    Description: Parenthood represents a fundamental construct that identifies the quality of early adult-infant interactions. In both short and long period, relationships, as primary interactional experiences, have an essential role in influencing individual’s adjustment and psychopathology development during in lifetime. In this scenario, the most important areas of interest and innovation are (a) parents’ representation of themselves, both in relation to the child and to their care-giving role; (b) the quality of couple relationship, in terms of both conjugal and co-parental bounds, and its influence on the quality of early mother-father-child interactions; (c) the early models of caregiver-baby interaction; (d) the recent approach to the “maternal brain”, that represents the contribution coming from neurosciences, linked to the adult’s activation and cerebral functioning processes, in association with the parental role. These data are a starting point for the individuation of functioning mechanisms and developmental trajectories of parenting in groups of adults and babies, belonging to normative populations. At the same time, those studies may provide an important introduction to the detection of critical and/or dysfunctional aspects in populations of babies and adults at risk (e.g. preterm babies, adopted children, etc.) or in adults’ clinical group (e.g. a depressed parents, addicted parents, etc.) or in children with one or more impaired characteristic (e.g. children with organic diseases, with autistic disorders, etc.).. The present Research Topic puts the attention on these themes, particularly, considering the possible aftermaths that empirical research may have on planning and realizing interventional models to support the parental functioning. We will consider papers focused on the individuation of methods of observation and assessment, which allow to plan and realize prevention programs and/or interventions, primarily focused on parental support, both in the early stadium of the child’s development and in the long-term period. Thus, the focus is both on the prevention and sustenance sphere and, also, on specific manners of taking on the therapeutic responsibility of the adult-infant relationship.

    Description: The ability of pathogens, such as parasites, bacteria, fungi and viruses to invade, persist and adapt in both invertebrate and vertebrate hosts is multifactorial and depends on both pathogen and host fitness. Communication between a pathogen and its host relies on a wide and dynamic array of molecular interactions. Through this constant communication most pathogens evolved to be relatively benign, whereas killing of its host by a pathogen represents a failure to adapt. Pathogens are lethal to their host when their interaction has not been long enough for adaptation. Evolution has selected conserved immune receptors that recognize signature patterns of pathogens as non-self elements and initiate host innate responses aimed at eradicating infection. Conversely, pathogens evolved mechanisms to evade immune recognition and subvert cytokine secretion in order to survive, replicate and cause disease. The cell signaling machinery is a critical component of the immune system that relays information from the receptors to the nucleus where transcription of key immune genes is activated. Host cells have developed signal transduction systems to maintain homeostasis with pathogens. Most cellular processes and cell signaling pathways are tightly regulated by protein phosphorylation in which protein kinases are key protagonists. Pathogens have developed multiple mechanisms to subvert important signal transduction pathways such as the mitogen activated protein kinase (MAPK) and the nuclear factor kB (NF-kB) pathways. Pathogens also secrete effectors that manipulate actin cytoskeleton and its regulators, hijack cell cycle machinery and alter vesicular trafficking. This research topic focuses on the cellular signaling mechanisms that are essential for host immunity and their subversion by pathogens.

    Description: Clearance of apoptotic cells is essential for proper development, homeostasis and termination of immune responses in multicellular organisms. Thus, cellular and molecular players taking part in the sequential events of this process are of great interest. Research in the last 20 years has indicated that specific ligands and receptors take part in the attraction of immune cells toward apoptotic targets and in the interactions between apoptotic cells and professional as well as non-professional phagocytes that engulf them. Moreover, phagocytosis of apoptotic cells (efferocytosis) leads to significant phenotypic changes in the engulfing cells suggesting that it is a major fate-determining event for phagocytes. Particularly, efferocytosis has an important impact on the inflammation-resolution axis as well as embryonic development and tissue morphogenesis. Deficiencies in these processes can result in health threats, such as autoimmunity, atherosclerosis, bone loss, obesity, infertility, neurodegeneration, fibrosis and cancer.     Description: Attentional biases (ABs) play a prominent role in the development and maintenance of clinically relevant symptoms of, for example, anxiety and depression. In particular, increased attentional orienting and preoccupation with biologically relevant and mood-congruent stimuli has been observed, suggesting that the visual-attentional system is overly sensitive towards threat cues and avoidant of cues of reward in these disorders. First, several experimental paradigms have been used to assess ABs, e.g., the dot probe task, the emotional stroop task, and the spatial cueing task amongst others. Yet, these paradigms are based on different theoretical backgrounds and target different stages of the attentional process. Thus, different paradigms provided converging as well as diverging evidence with regard to ABs. However, it is often not entirely clear to what extent this reflects real differences and commonalities, or is caused by differences in methodology. For example, behavioral reaction time data can only provide a snapshot of selective attention. Measuring event-related potentials, eye movements, or functional brain imaging data enables exploring the exact temporal and spatial dynamics of attentional processes. Moreover, neuroimaging data reveal specific cortical networks involved in directing attention toward a stimulus or disengaging from it. Second, ABs have been mainly discussed as symptoms of psychopathology, while results in healthy participants are still scarce; previous studies mostly compared extreme groups. However, a comprehensive theoretical and empirical account of ABs in psychopathology also requires a thorough account of ABs in the general healthy population. Moreover, the effect of gender, as an important contributing factor in processing of emotional stimuli, has also not been considered systematically in previous research. Third, a variety of stimuli has been used in the assessment of ABs. So far, mostly facial or word stimuli have been applied. However, in everyday life not only facial emotion recognition but also a fast evaluation of complex social situations is important to be effective in social interactions. Recent research started using more complex stimuli to raise ecological validity. However, the use of ecologically valid stimuli poses some methodological challenges and needs to be applied more systematically. The aim of this research topic is to integrate different paradigms and stimuli, addressing individuals from the whole range of the population continuum, and to apply different methodological approaches. It is intended to bring together expertise in stimulus selection, timing and implementing issues, advancing and broadening the overall understanding of ABs.

    Description: Dendritic cells (DC) are among the first cells to encounter pathogens and damage in peripheral tissues and, upon activation, DC migrate to lymph nodes where they activate and educate T cells to initiate and shape the immune response. DC present pathogen-derived antigen to T cells and drive T cell differentiation into particular effector cells through the expression and secretion of co-stimulatory molecules and cytokines respectively. The study of DC biology has included the identification of multiple DC subsets in tissues and lymphoid organs, the differentiation and plasticity of DC subsets, the functional consequences of DC interaction with pathogen, control of DC migratory properties and the impact of DC on T cell activation and differentiation. In recent years sophisticated systems biology approaches have been developed to deepen our understanding of DC function. These studies have identified differences between DC subsets located in various tissues and critical factors that drive the outcome of the interaction between DC and T cells. DC are currently being used in in various clinical therapeutic settings, including as vaccines for cancer and autoimmune disease. A clear understanding of DC factors that contribute to specific immune responses is vital to the success of DC based therapies. This research topic will give a comprehensive overview of current issues in DC biology and provides an update on the clinical uses of DC in the therapy of autoimmunity and cancer.

    Description: Neuroenhancement (NE) is a behavior conceptualized as the use of a potentially psychoactive substance to enhance ones’ already proficient cognitive capacities. Depending on the specific definitions used, prevalence estimates vary greatly between very low 0.3% (for illicit substances) to astonishingly high 89% (for freely available lifestyle substances). These variations indicate that further research and more conceptual and theoretical clarification of the NE construct is dearly needed. The contributions of this research topic aim to do just that. Specific questions addressed are: How prevalent is NE behavior? How can NE research profit from the already more evolved field of social science research on doping in sports? How is NE perceived by the public? What psychological processes and variables play a role in the decision to neuroenhance? A wide array of methodological approaches is used to investigate these questions. The topics contributions range from theoretical to experimental accounts on NE, and they utilize a diverse set of methods ranging from qualitative to neuroscientific approaches. The research presented here represents a first step towards what we have labeled a psychological approach to NE. By addressing the questions above this research topic hopefully advances our understanding of NE behavior. As with every new field of research, new answers always prompt new questions. In light of what we know now about NE, we hope that the findings presented here will be pursued by other researchers in the future. Clearly, the endeavor to understand NE behavior has only just begun.

    Description: Recognition and killing of aberrant, infected or tumor targets by Natural Killer (NK) cells is mediated by positive signals transduced by activating receptors upon engagement of ligands on target surface. These stimulatory pathways are counterbalanced by inhibitory receptors that raise NK cell activation threshold through negative antagonist signals. While regulatory effects are necessary for physiologic control of autoimmune aggression, they may restrain the ability of NK cells to activate against disease. Overcoming this barrier to immune surveillance, multiple approaches to enhance NK-mediated responses are being investigated since two decades. Propelled by considerable advances in the understanding of NK cell biology, these studies are critical for effective translation of NK-based immunotherapy principles into the clinic. In humans, dominant inhibitory signals are transduced by Killer Immunoglobulin Like Receptors (KIR) recognizing cognate HLA class I on target cells. Conversely, KIR recognition of “missing self-HLA” - due to HLA loss or HLA/ KIR mismatch - triggers NK-mediated tumor rejection. Initially observed in murine transplant models, these antitumor effects were later found to have important implications for the clinical outcome of haplotype-mismatched stemcell transplantation. Here, donor NK subsets protect against acute myeloid leukemia (AML) relapse through missing self recognition of donor HLA-C allele groups (C1 or C2) and/or Bw4 epitope. These studies were subsequently extended by trials investigating the antileukemia effects of adoptively transferred haplotype-mismatched NK cells in non-transplant settings. Other mechanisms have been found to induce clinically relevant NK cell alloreactivity in transplantation, e.g., post-reconstitution functional reversal of anergic NK cells. More recently, activating KIR came into the spotlight for their potential ability to directly activate donor NK cells through in vivo recognition of HLA or other ligands. Novel therapeutic monoclonal antibodies (mAb) may optimize NK-mediated effects. Examples include obinutuzumab (GA101), a glyco-engineered anti-CD20 mAb with increased affinity for the FcγRIIIA receptor, enhancing antibody-dependent cellular cytotoxicity; lirilumab (IPH2102), a first-in-class NK-specific checkpoint inhibitor, blocking the interaction between the major KIR and cognate HLA-C antigens; and elotuzumab (HuLuc63), a humanized monoclonal antibody specific for SLAMF7, whose anti-myeloma therapeutic effects are partly due to direct activation of SLAMF7-expressing NK cells. In addition to conventional antibodies, NK cell-targeted bispecific (BiKEs) and trispecific (TriKEs) killer engagers have also been developed. These proteins elicit potent effector functions by binding target ligands (e.g., CD19, CD22, CD30, CD133, HLA class II, EGFR) on one arm and NK receptors on the other. An additional innovative approach to direct NK cell activity is genetic reprogramming with chimeric antigen receptors (CAR). To date, primary NK cells and the NK92 cell line have been engineered with CAR specific for antigens expressed on multiple tumors. Encouraging preclinical results warrant further development of this approach. This Research Topic welcomes contributions addressing mechanisms of NK-mediated activation in response to disease as well as past and contemporary strategies to enhance NK mediated reactivity through control of the interactions between NK receptors and their ligands.

    Description: Plasma cells (PCs) are terminally differentiated B-cells producing large amounts of immunoglobulins (Ig). In humans, most of circulating Ig are produced by bone marrow plasma cells. PCs differentiate from activated naïve or memory B-cells usually activated by specific antigens. It is still controversial whether the regulation of PCs numbers and the “active” in vivo Ig diversity depend or not on non-specific reactivation of B-cells during infections. Depending on the stimulus (T-independent/T-dependent antigen, cytokines, partner cells) and B-cell types (naïve or memory, circulating or germinal center, lymph nodes or spleen, B1 or B2...), both the phenotype and isotype of PCs differ suggesting that PC diversity is either linked to B-cell diversity or to the type of stimulus or to both. Knowledge of the mechanisms supporting PC diversity has important consequences for the management of i) plasma cell neoplasia such as Multiple Myeloma and Waldenström's Macroglobulinemia, ii) vaccine protection against pathogens and iii) auto-immune diseases.

    Description: Despite continuous progress in the development of anti-viral and anti-bacterial/parasite drugs, the high cost of medicines and the potential for re-infection, especially in high risk groups, suggest that protective vaccines to some of the most dangerous persistent infections are still highly desirable. There are no vaccines available for HIV, HCV and Malaria, and all attempts to make a broadly effective vaccine have failed so far. In this Research Topic we look into why vaccines have failed over the years, and what we have learn from these attempts. Rather than only showing positive results, this issue aims to reflect on failed efforts in vaccine development. Coming to understand our limitations will have theoretical and practical implications for the future development of vaccines to these major global disease burdens.

    Description: Humans are remarkably adept at identifying individuals on the basis of their facial features, or other traits such as gait or vocal timbre. Besides voice, another auditory medium capable of carrying identity information is music. Indeed, certain famous musicians, such as John Coltrane or Sonny Rollins, need only to play a few notes to be unequivocally recognized. Along with emotion and structural cues, artistic individuality seems to be a key element communicated in music performance. Yet, the means by which individuality is expressed in performance, as well as the cognitive processes employed by listeners to perceive identity cues, remain poorly elucidated. Other pertinent issues, including the connection between a performer’s technical competence and ability to convey a specific musical identity, as well as potential links between individuality and career-defining outcomes such as critical recognition and aesthetic appraisal, warrant further exploration. Quantitative approaches to the study of music performance have benefited greatly from MIDI technology and the application of computational methods, leading to the flourishing of empirical music performance research over the last few decades. More recently, neuroimaging techniques have provided valuable insights into the neural mechanisms involved in the cognitive processes of performing music. Nevertheless, this field continues to benefit greatly from qualitative approaches, given that the communication of affect and identity cues in music performance leads to a rich subjectivity of impressions that must be accounted for in order to lead to a greater understanding of this multifaceted phenomenon. The aim of this Research Topic is to provide a forum for interdisciplinary research broadly related to the expression and perception of individuality in music performance. Research methodology includes behavioral, psychophysiological, and neuroimaging techniques. Both quantitative and qualitative approaches are presented The scope of this Research Topic includes laboratory studies as well as studies in real-life performance settings and longitudinal studies on performers.

    Description: Elite sport typically provides obvious rewards in terms of recognition, finance and acclaim for athletic performance. Increasingly, we are becoming aware of the risks that elite athletes, their entourage, including families, sport-science support team and coaches are exposed to. Twelve original articles, seven commentaries and a corrigendum, are structured in a five chapter format. Chapter 1, comprising the Editorial is titled “An Overview of Mental Health in Elite Sport: Changing the Play Book” to reflect the advocacy role of this article. Chapter 2 (“Finding the Sweet Spot”) amplifies the voice of key stakeholders across three qualitative studies with three additional commentaries. Quantitative evidence is presented in Chapter 3 which has the sub-title the “State of Play.” Chapter 4, entitled the “Field of Play” includes three original publications which present contrasting conceptual approaches to guide researchers in hypothesis generation, formulation and implementation science. Finally, in Chapter 5, “Seeing the Ball Early” prospective perspectives are provided in three publications reinforced by two commentaries. The future thinking ideas includes the use of virtual reality training, a broadening of the concept of mental health literacy, tackling stigma and focusing on the potential positive effect of the natural environment on well-being and recovery.To date the research topic has generated widespread in the field. For example, several articles have generated an Altmetric score above 40 with one publication meriting an Altmetric score of 102. We envisage that the impact of this e-book will not simply be measured in citations, views, downloads nor social media impact, but in the discourse that emerges from this collection of contributions from a combined total of 53 authors from across three continents. It is our hope that this e-book, providing a snapshot of global challenges for elite athletes mental health and well-being, becomes a touchstone for researchers and practitioners in the field.

    Description: Motor skills are a vital part of healthy development and are featured prominently both in physical examinations and in parents’ baby diaries. It has been known for a long time that motor development is critical for children’s understanding of the physical and social world. Learning occurs through dynamic interactions and exchanges with the physical and the social world, and consequently movements of eyes and head, arms and legs, and the entire body are a critical during learning. At birth, we start with relatively poorly developed motor skills but soon gain eye and head control, learn to reach, grasp, sit, and eventually to crawl and walk on our own. The opportunities arising from each of these motor milestones are profound and open new and exciting possibilities for exploration and interactions, and learning. Consequently, several theoretical accounts of child development suggest that growth in cognitive, social, and perceptual domains are influences by infants’ own motor experiences. Recently, empirical studies have started to unravel the direct impact that motor skills may have other domains of development. This volume is part of this renewed interest and includes reviews of previous findings and recent empirical evidence for associations between the motor domain and other domains from leading researchers in the field of child development. We hope that these articles will stimulate further research on this interesting question.

    Description: Two of the most important social skills in humans are the ability to determine the moods of those around us, and to use this to guide our behavior. To accomplish this, we make use of numerous cues. Among the most important are vocal cues from both speech and non-speech sounds. Music is also a reliable method for communicating emotion. It is often present in social situations and can serve to unify a group's mood for ceremonial purposes (funerals, weddings) or general social interactions. Scientists and philosophers have speculated on the origins of music and language, and the possible common bases of emotional expression through music, speech and other vocalizations. They have found increasing evidence of commonalities among them. However, the domains in which researchers investigate these topics do not always overlap or share a common language, so communication between disciplines has been limited. The aim of this Research Topic is to bring together research across multiple disciplines related to the production and perception of emotional cues in music, speech, and non-verbal vocalizations. This includes natural sounds produced by human and non-human primates as well as synthesized sounds. Research methodology includes survey, behavioral, and neuroimaging techniques investigating adults as well as developmental populations, including those with atypical development. Studies using laboratory tasks as well as studies in more naturalistic settings are included.

    Description: f we want to understand people’s responses to threats in social interactions we can distinguish between three levels of analysis:On a social level of analysis we can describe people’s interpersonal behavior, on a cognitive level we can identify corresponding information processing mechanisms, and on a neural level we can specify neural systems, which underlie these processes.In this Research Topic we want to present research connecting these three levels of analysis and propose their functional interconnection in social interaction.We propose that threats in social interactions activate basic motivational processes, which manifest in neural processes related to behavioral inhibition vs. activation in a social situation. This shapes our attention to new information, and affects our cognitions about social identities, belief systems and worldviews. These changes in social cognition in turn affect people’s behavior in social interactions and lead to corresponding reactions on behalf of the interaction partner. Thus, we assume that people’s reactions to threat in interactions can be described as sequences of broader attentional processes resulting from basic motivational tendencies leading to specific social cognitions and subsequent behavior within social interactions. We can analyze this sequence in order to contribute to a better understanding of social interactions.The three levels of analyses (social, cognitive, neural) shed light on social interactions from different angles:On the social level we can analyze how the behaviors of the interaction partners mutually affect each other and how this is accompanied by specific cognitive, emotional and motivational processes. On the cognitive level we can analyze people’s perception of a social situation leading to attentional and reasoning processes with regard to their interaction partner/s, which may be accompanied by certain emotional and motivational processes and determines the behavior towards the partner/s. Finally, we can focus on the neural mechanisms underlying cognitive, emotional, and motivational processes in social interactions.

    Description: According to management and psychology courses, as well as legions of consultants in organizational psychology, shared vision in dyads, teams and organizations can fill us with hope and inspire new possibilities, or delude us into following false prophets. However, few research studies have empirically examined the impact of shared vision on key organizational outcomes such as leadership effectiveness, employee engagement, organizational citizenship, coaching and organizational change. As a result, the field of organizational psychology has not yet established a causal pattern of whether, if, and how shared vision helps dyads, teams and organizations function more effectively. The lack of empirical work around shared vision is surprising given its long-standing history in the literature. Bennis and Nanus (1982) showed that distinctive leaders managed attention through vision. The practitioner literature has long proclaimed that vision is a key to change, while Conger and Kanungo (1998) discussed its link to charismatic leadership. Around the same time, positive psychology appeared in the forms of Appreciative Inquiry (Cooperrider, Sorensen, Whitney, & Yaeger, 2000) and Positive Organizational Scholarship (Cameron, Dutton, & Quinn, 2003). In this context, a shared vision or dream became a legitimate antecedent to sustainable change. But again, empirical measurement has been elusive. More recently, shared vision has been the focus of a number of dissertations and quantitative studies building on Intentional Change Theory (ICT) (Boyatzis, 2008) at dyad, team and organization levels of social systems. These studies are beginning to lay the foundations for a systematic body of empirical knowledge about the role of shared vision in an organizational context. For example, we now know that shared vision can activate neural networks that arouse endocrine systems and allow a person to consider the possibilities of a better future (Jack, Boyatzis, Leckie, Passarelli & Khawaja, 2013). Additionally, Boyatzis & Akrivou (2006) have discussed the role of a shared vision as the result of a well-developed set of factors that produce a desired image of the future. Outside of the organizational context, positive visioning has been known to help guide future behavior in sports psychology (Loehr & Schwartz, 2003), medical treatment (Roffe, Schmidt, & Ernst, 2005), musical performance (Meister, Krings, Foltys, Boroojerdi, Muller, Topper, & Thron, 2004), and academic performance (Curry, Snyder, Cook, Ruby, & Rehm, 1997). This Research Topic for Frontiers in Psychology is a collection of 14 original papers examining the role of vision and shared vision on a wide variety of desired dependent variables from leadership effectiveness and executive performance to organizational engagement, citizenship and corporate social responsibility, and how to develop it through coaching.

    Description: A line of research in cognitive science over several decades has been dedicated to finding an innate, language-specific cognitive system, a faculty which allows human infants to acquire languages natively without formal instruction and within short periods of time. In recent years, this search has attracted significant controversy in cognitive science generally, and in the language sciences specifically. Some maintain that the search has had meaningful results, though there are different views as to what the findings are: ranging from the view that there is a rich and rather specific set of principles, to the idea that the contents of the language faculty are - while specifiable - in fact extremely minimal. But other researchers rigorously oppose the continuation of this search, arguing that decades of effort have turned up nothing. The fact remains that the proposal of a language-specific faculty was made for a good reason, namely as an attempt to solve the vexing puzzle of language in our species. Much work has been developing to address this, and specifically, to look for ways to characterize the language faculty as an emergent phenomenon; i.e., not as a dedicated, language-specific system, but as the emergent outcome of a set of uniquely human but not specifically linguistic factors, in combination. A number of theoretical and empirical approaches are being developed in order to account for the great puzzles of language - language processing, language usage, language acquisition, the nature of grammar, and language change and diversification. This research topic aims at reviewing and exploring these recent developments and establishing bridges between these young frameworks, as well as with the traditions that have come before. The goal of this Research Topic is to focus on current developments in what many regard as a paradigm shift in the language sciences. In this Research Topic, we want to ask: If current explicit proposals for an innate, dedicated faculty for language are not supported by data or arguments, how can we solve the problems that UG was proposed to solve? Is it possible to solve the puzzles of language in our species with an appeal to causes that are not specifically linguistic?

    Description: Coordinating one’s behavior with the behavior of other individuals is a fundamental feature of everyday social interaction. A defining feature of such behavior is that it is dynamic, that is, it evolves over time. This is true whether one considers the linguistic, gestural and non-verbal coordination that occurs between two or more individuals engaged in a conversation or the physical movement coordination that occurs when two or more people clear a dinner table or load a dishwasher together. Such behavior is also emergent and robust to sudden changes in task context or unexpected environmental or social perturbations. Accordingly, robust social action and multi-agent coordination is synergistic, with co-acting individuals adapting to each other and the environment around them in a mutual and reciprocal manner. Research investigating the behavioral dynamics of joint-action and multi-agent coordination has steadily increased over the last several decades. Spurred by several factors, including (i) the increased accessibility of technologies for recording and extracting the time-evolution of multi-agent behavior (e.g., motion tracking, eye-tracking, EEG), (ii) the development of new nonlinear techniques for analyzing behavioral and linguistic time-series data, and (iii) a growing appreciation that social cognition, perception, and action are interdependent, embodied and embedded processes, this research has not only been directed towards measuring and identifying the stable patterns of coordinated social and multi-agent activity that emerge over time, but also how these stable pattern are activated, dissolved, transformed, and exchanged over time. Not surprisingly, researchers have begun to investigate the implications of this behavioral dynamics perspective for understanding social cognitive processes as well as clinical disorders with social deficits such as autism and schizophrenia. Attempts at modeling the dynamics of social action and multi-agent behavior using various nonlinear and complex systems methods has also increased over the last several years, with many researchers demonstrating how simple low-dimensional dynamical or computational models can be employed to capture and explain the dynamics of ongoing joint-action and multi-agent behavior. A characteristic feature of these dynamical models is that they reveal how stable social action and multi-agent coordination arises naturally from the interaction of the physical, biomechanical, neural, informational, and social cognitive properties of a joint-action task context and goal, and cannot be ascribed to any one singular processes, agent, or level of analysis.     Description: Deception is a ubiquitous phenomenon in social interactions and has attracted a significant amount of research during the last decades. The majority of studies in this field focused on how deception modulates behavioral, autonomic, and brain responses and whether these changes can be used to validly identify lies. Especially the latter question, which historically gave rise to the development of psychophysiological "lie detection" techniques, has been driving research on deception and its detection until today. The detection of deception and concealed information in forensic examinations currently constitutes one of the most frequent applications of psychophysiological methods in the field. With the increasing use of such methods, the techniques for detecting deception have been controversially discussed in the scientific community. It has been proposed to shift from the original idea of detecting deception per se to a more indirect approach that allows for determining whether a suspect has specific knowledge of crime-related details. This so-called Concealed Information Test is strongly linked to basic psychological concepts concerning memory, attention, orienting, and response monitoring. Although research in this field has intensified with the advancement of neuroimaging techniques such as PET and fMRI in the last decade, basic questions on the psychological mechanisms underlying modulatory effects of deception and information concealment on behavioral, autonomic, and brain responses are still poorly understood. This Research Topic brings together contributions from researchers in experimental psychology, psychophysiology, and neuroscience focusing on the understanding of the broad concept of deception including the detection of concealed information, with respect to basic research questions as well as applied issues. This Research Topic is mainly composed of originalresearch articles but reviews and papers elaborating on novel methodological approaches have also been included. Experimental methods include, but are not limited to, behavioral, autonomic, electroencephalographic or brain imaging techniques that allow for revealing relevant facets of deception on a multimodal level. While this Research Topic primarily includes laboratory work, relevant issues for the field use of such methods are also discussed.

    Description: 80% of the bodies’ immune cells are harbored within the intestine. They are only separated from 1014 microorganisms by a single layer of intestinal epithelial cells and a secreted superficial mucus layer. Therefore, the intestinal epithelial surface represents a main frontier in host defense. Providing an intact mucosal barrier is vital for the host to limit bacterial entry and spread to the circulation. This specialized localization requires dynamic responses of intestinal epithelial cells to both pathogen- and immune-derived signals. Moreover, emergency barriers are needed in the setting of epithelial damage, which allow provisional microbial control and a timely restitution of mucosal integrity. Epithelial cells constantly interact with subjacent immune cells and fibroblasts, actively directing the immune response and also shaping the luminal microbiota. Epithelial dysfunction has been appreciated in recent years as a driving element in the pathogenesis of Inflammatory Bowel Diseases (IBD). Additionally, primary immune deficiencies may manifest in the form of chronic intestinal inflammation mimicking features of IBD. Recent advances in the techniques of epithelial cell culture and the discovery of new immune cell types and cellular properties have tremendously advanced the understanding in this interesting field of research. In this research topic, we want to focus on the complex interaction of intestinal epithelial cells, luminal flora and adjacent immune cells and invite manuscripts which highlight the dynamic responses of both epithelium and immune cells under steady-state or inflammatory conditions, and envision how this may be translated to the benefit of patient-care.

    Description: Behaviour, language, and reasoning are expressions of neural functions par excellence, as the brain must draw on sensory modalities to gather information on the rest of the body and on the outer world. Cortical areas processing the identity and location of sensory inputs were once thought to be organised hierarchically, with some branches dedicated to basic features and other branches dedicated to complex features. Yet current studies have uncovered synergistic effects at early sensory cortices as well as at higher-level association areas. A less hierarchical functional architecture of the brain has emerged such that, irrespective of sensory modality, inputs would be allocated to the best suited cortical substrate. It is our hope that the articles included in this special issue will offer novel insights into recent developments relating to multisensory integration and brain functioning.

    Description: New therapies are currently being developed in the field of skin cancer. In particular, advances in melanoma now represent the frontline of cancer immunotherapy, as immunological findings in the disease have led to the development of highly effective immune-checkpoint inhibitors. However, these immune-checkpoint inhibitors are only effective in a subset of patients, and may not work in other skin cancer types, thus highlighting the need for further innovation in the field of skin cancer treatment. The purpose of this Research Topic is therefore to provide an up-to-date overview of immune processes and new therapies for melanoma and other skin cancers in order to further stimulate the development of new and even more successful treatments.
