    Abstract: Rabies is one of the oldest known pathogens, is incurable, and has the highest fatality rate of all infectious diseases. The Americas is the only region with bat rabies virus, including vampire bat rabies. The region is rich in cultural references and notable for many discoveries in the field, including the current vaccine potency test, diagnostic assay, conception of oral vaccines for wildlife, the first human survivor and the first successful canine rabies program executed at a broad level. Rabies remains the most important viral zoonosis, with tens of thousands of human fatalities and tens of millions of exposures annually, which can be used to model for other pathogens, such as COVID-19. There is an international effort to eliminate human rabies caused by dogs over the next decade, and the Americas represent the primary region with the greatest proof-of-concept evidence to accomplish this goal. This two-volume set addresses the medical history and modern results of rabies in countries throughout the Americas, including the implications of and on cultural, economic, sociological, and research developments in the region. Volume I presents an overview of concepts critical to the study of rabies in the region, including evolutionary aspects, reservoir ecology and control, elimination efforts, vaccine development, and disease hallmarks and progression.     Abstract: Nutrition is an important environmental factor for the maturation of the human immune system and essential for maintaining immunological homeostasis. Based on this, a variety of food applications with medical claims are being generated by food manufacturers worldwide in order to expand the market potential of products creating interesting linkages with other market segments, such as cosmetics and pharmaceuticals. However, in addition to the health benefits, active principles of such components often remain unexplored. This book focusses on the specific interactions between food ingredients and the immune system along the entire immune defense response. Starting from the immune barrier, through the innate and adaptive immune response, to active limitation and termination, all major mechanisms of the immune response are addressed and different biochemical, cellular and genetic interactions of components of our diet are discussed.     Abstract: This volume provides a comprehensive review of programmed cell death pathways and their fundamental role in antiviral host defense. The book deep-dives into the molecular functions and regulation of necroptosis and discusses how viruses induce and manipulate this potent innate cellular sensing system. Initially, understanding of necroptosis emerged from studies on tumor necrosis factor (TNF) signaling that showed the key role of receptor interacting protein kinase 1 (RIPK1) in the activation of receptor interacting protein kinase 3 (RIPK3) which then phosphorylates mixed lineage kinase domain like pseudokinase (MLKL) to execute cells via plasma membrane leakage of cytosolic contents. Since its discovery, multiple facets of the RIPK3-dependent necroptotic machinery have evolved where the requirements for execution of death varies depending on the stimulus. Virus-induced necroptosis was discovered over 10 years ago in studies on murine cytomegalovirus (MCMV) where a virus-encoded inhibitor was shown to prevent the recruitment of RIPK3 (RIP3). This transformative evidence identified a novel pathway acting independent of TNF, interferon or RIPK1 that can stop virus from infecting its natural mouse host by killing off infected cells to halt replication. Over the past decade influenza A virus (IAV), herpes simplex virus (HSV) and poxvirus vaccinia (VACV) have all been shown to trigger the pathway. Herpesviruses and poxviruses also encode inhibitors of caspase-8 whose elaboration unleashes the necroptosis pathway. IAV and other RNA viruses do not encode programmed cell death inhibitors. RIPK3 is also known to induce apoptosis by recruiting RIPK1 as shown nearly a decade ago and this dual apoptosis/necroptosis induction occurs naturally during influenza A virus infection. RIPK3 is also able to induce an inflammatory response independently of programmed cell death that can predominate to drive inflammatory disease outcomes. This volume is a must-read for researchers and advanced students in immunology and virology.

Chikungunya virus (CHIKV) is a mosquito-borne pathogen that has re-emerged to become a significant global disease threat. Although CHIKV has historically been associated with localized outbreaks of debilitating polyarthralgia in Africa and Asia, factors such as increased global travel and viral adaptation to new mosquito vectors have led to the introduction and spread of CHIKV in new regions of the world, including the Americas. The re-emergence of CHIKV has stimulated significant interest both in understanding the biology of CHIKV and in the development of medical countermeasures against the virus.

    Abstract: This edited collection of 12 chapters by research workers from a wide range of disciplines resolves the confusion that currently surrounds the “hygiene hypothesis” by considering the human need for exposure to microorganisms from an evolutionary point of view.     Abstract: Lung diseases are leading causes of death and disability globally, with about 65 million people suffering from COPD, and 334 million from asthma. Each year, tens of millions of people develop and can die from lung infections such as pneumonia and TB. Systemic inflammation may induce and exacerbate local inflammatory diseases in the lungs, and local inflammation can in turn cause systemic inflammation. There is increasing evidence of the coexistence of systemic and local inflammation in patients suffering from asthma, COPD, and other lung diseases, and the co-morbidity of two or more local inflammatory diseases often occurs. For example, rheumatoid arthritis frequently occurs together with, and promotes the development of, pulmonary hypertension. This co-morbidity significantly impacts quality of life, and can result in death for some patients. Current treatment options for lung disease are neither always effective, nor condition-specific; there is a desperate need for novel therapeutics in the field. Additionally, the molecular and physiological significance of most major lung diseases is not well understood, which further impedes development of new treatments, especially in the case of coexistent lung diseases with other inflammatory diseases. Great progress has been made in recent years in many areas of the field, particularly in understanding the molecular geneses, regulatory mechanisms, signalling pathways, and cellular processes within lung disease, as well as basic and clinical technology, drug discovery, diagnoses, treatment options, and predictive prognoses. This is the first text to aggregate these developments. In two comprehensive volumes, experts from all over the world present state-of-the-art advances in the study of lung inflammation in health and disease.     Abstract: This book summarizes recent advances in antibody glycosylation research. Covering major topics relevant for immunoglobulin glycosylation - analytical methods, biosynthesis and regulation, modulation of effector functions - it provides new perspectives for research and development in the field of therapeutic antibodies, biomarkers, vaccinations, and immunotherapy. Glycans attached to both variable and constant regions of antibodies are known to affect the antibody conformation, stability, and effector functions. Although it focuses on immunoglobulin G (IgG), the most explored antibody in this context, and unravels the natural phenomena resulting from the mixture of IgG glycovariants present in the human body, the book also discusses other classes of human immunoglobulins, as well as immunoglobulins produced in other species and production systems.     Abstract: This book explains about amino acids (AAs) which are not only building blocks of protein, but are also signaling molecules as well as regulators of gene expression and the protein phosphorylation cascade. Additionally, AAs are key precursors for syntheses of hormones and low-molecular-weight nitrogenous substances with each having enormous biological importance. For example, physiological concentrations of AA metabolites (e.g., nitric oxide, polyamines, glutathione, taurine, thyroid hormones, and serotonin) are required for cell functions. Growing evidence shows that humans and animals have dietary requirements for all proteinogenic AAs. Mammals, birds, and fish also have species- and age-dependent needs for some AA-related substances. However, elevated levels of other products (e.g., ammonia, homocysteine, H2S, and asymmetric dimethylarginine) are pathogenic factors for neurological disorders, oxidative stress, and cardiovascular disease. Thus, optimal amounts of AAs and their ratios in diets and circulation are crucial for whole-body homeostasis and health. Adequate provision of one or a mixture of functional AAs or metabolites may be beneficial for ameliorating health problems at various stages of the life cycle (e.g., fetal growth restriction, neonatal morbidity and mortality, weaning-associated intestinal dysfunction and wasting syndrome, obesity, diabetes, cardiovascular disease, the metabolic syndrome, and infertility). Dietary supplementation of these nutrients can also optimize the efficiency of metabolic transformations to enhance muscle growth, milk production, and athletic performance, while preventing excess fat deposition and reducing adiposity. Therefore, functional AAs hold great promise in improving the growth, health, and well-being of individuals.

    Abstract: Respiratory diseases are leading causes of death and disability globally, with about 65 million people suffering from COPD, and 334 million from asthma, the most common chronic disease. Each year, tens of millions of people develop and can die from respiratory infections such as pneumonia and TB. Systemic inflammation may induce and exacerbate local inflammatory diseases in the lungs, and local inflammation can in turn cause systemic inflammation. There is increasing evidence of the coexistence of systemic and local inflammation in patients suffering from asthma, COPD, and other lung diseases, and the co-morbidity of two or more local inflammatory diseases often occurs. For example, rheumatoid arthritis frequently occurs together with, and promotes the development of, pulmonary hypertension. This co-morbidity significantly impacts quality of life, and can result in death for those affected. Current treatment options for lung diseases are neither always effective, nor often condition-specific; there is a desperate need for novel therapeutics in the field. Additionally, the molecular and physiological significance of most major lung diseases are not well understood, which further impedes development of new treatments, especially in the case of coexistent lung diseases with other inflammatory diseases. Great progress has been made in recent years in many areas of the field, particularly in understanding the molecular geneses, regulatory mechanisms, signalling pathways, and cellular processes within lung diseases, as well as basic and clinical technology, drug discovery, diagnoses, treatment options, and predictive prognoses.     Abstract: The scope of this contributed volume is to provide an overview of the latest translational research in the field of lupus pathogenesis, with particular emphasis on how these discoveries progress in parallel with therapeutic drug development. Systemic lupus erythematosus (SLE) is a multifaceted disease with a number of well-defined immune pathways that are dysregulated, resulting in an immune-mediated chronic inflammatory injury at target organs. As knowledge of these pathways evolves to provide opportunities for targeted drug therapy and lays the foundation for personalized medicine, clinicians and researchers need to keep up with the ever-expanding medical literature. This book will critically appraise the current understanding of important immunological pathways that contribute to the pathogenesis of lupus. We will review the role of interferons as part of the innate immune defects that perpetuate the loss of self-tolerance in SLE. B cell hyperactivity, as a defining hallmark of SLE, and different strategies of B cell targeted therapy will be discussed. The role of co-stimulation or immune checkpoint molecules in activating B and T cells will be reviewed, as well as other cytokines that serve in the amplification loop promoting a more proinflammatory Th1 or Th17 responses. Intracellular targets, such as signaling molecules in the JAK/STAT pathway, or a variety of kinases and proteasomes, can cause a cascading downstream effect of transcriptional responses that are important in SLE. Immune homeostasis can also be restored by bolstering the naturally occurring anti-inflammatory mechanisms. Glucocorticoid, as a potent natural anti-inflammatory hormone, can mediate its effects by recruiting histone deacetylase that serve to repress gene transcription. Glucocorticoid-induced leucine zipper is a gene upregulated by glucocorticoid that can be a potential target for development of anti-inflammatory strategy. Finally, T regulatory cells can be utilized to help restore to immune tolerance and are amongst the latest focus of therapeutic development in SLE.

    Abstract: The topic of this book, Collectins, is a family of proteins whose major function is in innate immunity, where Collectins act as pattern recognition receptors (PRRs). In general they recognize targets such as microbial surfaces and apoptotic cells, and once bound to a target, Collectins promote the clearance of microorganisms and damaged host tissue. New cell-surface proteins and glycoproteins, which act as Collectin receptors, are currently being identified. Some Collectins, particularly MBL, activate the complement system, which enhances the ability of antibodies to fight pathogens, via three MBL-associated proteases, the MASPs. Additionally, recent research has begun to show wider-ranging activities of Collectins, such as: · Their role in metabolism, and therefore their involvement in lifestyle diseases such as obesity and cardiovascular disease. · Their ability to modulate the adaptive immune response, as well as to recognize and trigger apoptosis of cancer cells, which makes them effective in the annihilation of cancer cells with multiple mutations. · The regulation of their expression by gonadal steroid hormones implicates them with critical roles in both male and female fertility. · Altered levels of Collectins have been associated with various autoimmune diseases.     Abstract: Macrophages are core components of the innate immune system. Once activated, they may have either pro- or anti-inflammatory effects that include pathogen killing, safe disposal of apoptotic cells or tissue renewal. The activation state of macrophages is conceptualized by the so-called M1/M2 model of polarization. M2 macrophages are not simply antagonists of M1 macrophages; rather, they represent a network of tissue resident macrophages with roles in tissue development and organ homeostasis. M2 macrophages govern functions at the interfaces of immunity, tissue development and turnover, metabolism, and endocrine signaling.     Abstract: Phagocytosis is the engulfment of particulate matter by cells. It is a fundamental (and probably “primitive”) cell biological process which is important in single celled organisms such as amoeba; multicellular animals including coelenterates; and in higher animals. In humans and other mammals, specialised immune cells (phagocytes) utilise phagocytosis in their crucial role of engulfing and destroying infecting microbes. Yet, surprisingly, the biophysics and biochemistry underlying the process has only become clear recently with the advent of genetic manipulation and advances in single cell imaging. In this volume, the aim is to bring together recent fundamental advances that give a clear picture of the underlying mechanism involved in phagocytosis. Not only is this an important topic in its own right, but a full understanding of the process will have a potential impact on human medicine, since as antibiotics become less effective in fight infection, researchers are looking at alternative approaches, including enhancing the “natural” immunity brought about by immune phagocytes. The aim is to provide a comprehensive volume on the topic, with separate chapters on identified recent advances, each written by the major contributors in each area.
