    Abstract: The CTBN volume Therapeutic Applications of Dopamine D3 Receptor Function reviews the state of the knowledge on the dopamine D3 receptor and its role in human behavior and disease (i.e.: neuropsychiatric illnesses including schizophrenia, mood disorders, Parkinson’s disease, restless legs syndrome, addictions and substance use disorders). The volume is written by leading experts across multidisciplinary areas (imaging, biobehavioral testing and clinical trials, preclinical models / molecular pharmacology) converging on the therapeutic implications / potential of the D3 receptor. The D3 dopamine receptor is a member of the D2-like family of G protein-coupled receptors. It was cloned and characterized almost 25 years ago. A key feature of the D3 dopamine receptor system, which has attracted considerable attention, is its anatomical localization remarkably restricted to the limbic circuitry. This has spurred the hypothesis that D3 involvement could contribute to the pathophysiology of neuropsychiatric disorders (or to some features of neuropsychiatric disorders), including but not limited to psychosis, addictions and substance abuse, mood and movement disorders.

    Abstract: This book provides an overview of Data Monitoring Committees - what was done in the past, what is currently being done, and thoughts on improvements for the future. Previous works focused primarily on large cardiovascular studies (where DMCs originated more than 30 years ago) but updated references are needed that discuss smaller, more flexible studies in areas such as oncology. The authors have attended ~800 DMC meetings from ~200 distinct studies across all areas of clinical studies (oncology, rheumatology, rare diseases, cardiology, immunology, etc.) This wide range of expertise will be used, as well as the expertise that comes from working with virtually every large biotech/pharma and CRO for DMC work. The reader of the book will know when DMCs are needed or helpful, how to form the DMC, how to work with external CROs and with sponsor teams and the DMC to create needed DMC outputs, how the DMC meetings are conducted, and - especially for DMC members - what are considerations within the Closed Session to review safety/efficacy outputs to assess risk/benefit to make appropriate recommendations that protect the patient safety and trial integrity. This is a practical hands-on book on how to decide if a DMC is necessary, how to form the DMC, how to smoothly create the necessary materials for the DMC and have smooth running DMC meetings. There is no specialized training in school about how DMCs work - frequently people may have been in industry for many years without ever needing to work with a DMC. The concept of life cycle and regulatory perspectives are discussed. Three chapters are entirely dedicated to DOE theory from screening to optimization designs. Moreover, a comprehensive discussion on modelling and data treatment is presented. Practical aspects of QbD and DOE for pharmaceutical product and process of different dosage forms is included, as well as a practical guide of the input process variables, material attributes, intermediate, and final quality attributes for the most representative pharmaceutical processes.     Abstract: The current book is focussed on the Toll-like receptors (TLRs), which are the first pattern recognition receptors (PRRs) discovered in humans. For example, TLR4 was first recognized in humans in 1997 as a PRR recognizing the Gram-negative bacterial lipopolysaccharide (LPS). This discovery revolutionized the field of innate immunity and filled the long-standing gap in the pathogen recognition by the immune system. Now, it is well established that humans have 10 (TLR1-TLR10) and mice have 12 (TLR1-TLR13) functional TLRs, excluding TLR10 that is present as a defective pseudogene. TLRs are present as both membrane-bound extracellular (TLR1, TLR2, TLR4, TLR5, TLR6, and TLR10) and intracellular (TLR3, TLR7, TLR8, and TLR9) PRRs in humans, which identify different pathogen or microbe-associated molecular patterns (PAMPs or MAMPs) and death or damage-associated molecular patterns (DAMPs) released by the host cells. A lot of development in the TLR biology has occurred in last 24 years since there first discovery in humans. The book is intended to describe their role in the host defence, human reproduction, non-infectious sterile inflammatory conditions, including brain immunity and cerebrovascular diseases, signaling mechanisms, adaptive immunity, and their targeting for drug development.

    Abstract: This book collects information about the most popular ethnomedicinal plants, which are common in Turkey and around the world. It presents the ethnopharmacological records, in vivo and in vitro studies, side effects, chemical compositions and clinical studies of these medicinal plants. Chapters of the second part focus on translational challenges from the bench to the bedside in CNS drug development, shed light on the importance to understand the brain distribution of drugs related to their efficacy, elaborate on general pharmacokinetic considerations for CNS drugs and introduce current and novel drug delivery strategies to overcome the blood-brain barrier. The experimental part of the book covers mathematical and in vitro models as well as animal and human methods in blood-brain barrier research.     Abstract: Reactive oxygen species (ROS) have been implicated in almost every human disease phenotype, without much, if any, therapeutic consequence foremost exemplified by the failure of the so-called anti-oxidants. The term ‘oxidative stress’ is abandoned and replaced with a systems medicine and network pharmacology-based mechanistic approach to disease.     Abstract: This book reflects the state of the art of our understanding of the WNT signaling system, which comprises a network of signaling pathways initiated by the secreted WNT lipoglycoproteins, which are crucial for embryonal development, stem cell regulation, tissue homeostasis and repair. Dysfunction of this evolutionarily conserved signaling system leads to many diseases including developmental disorders, diverse forms of cancer, fibrosis, neurodegenerative disease and many more. The WNT signaling system is built upon 19 mammalian, secreted WNT lipoglycoproteins that interact with a plethora of distinct receptors, such as the G protein-coupled receptors called Frizzleds (FZD1-10), LDL receptor-like proteins (LRP5/6), receptor tyrosine kinases (ROR1, 2, RYK and PTK7). In addition, WNT pathways are tightly regulated by many secreted and cell-intrinsic negative regulators, such as soluble FZD-related proteins (SFRPs), Dickkopfs (DKKs), WNT-inhibitory proteins, TIKI, RNF43 and more. Understanding the basic mechanism in terms of receptor-ligand interaction, receptor selectivity, signal initiation and desensitization remain poorly understood, even though substantial advances have been made the recent years. Due to the involvement of the WNT signaling system in human disease, it appears obvious to target diverse branches pharmacologically and therapeutically. However, given the complexity of the system and its importance for stem cell regulation and tissue maintenance, therapy comes with obvious risks for severe side effects. The field is addressing the challenge to identify suitable targets and selective compounds for therapy allowing disease-selective therapeutic effects and balancing unwanted side effects. This book summarizes the current understanding of the basic and applied pharmacology in the WNT signaling system and bridges disciplines such as pharmacology, physiology, neurosciences, oncology and drug development.

    Abstract: Medicinal plants and their derived products remain as an indispensable source of bioactive molecules that serve as either drug candidates or lead compounds for drug design and discovery. There are several advantages for plant-derived therapeutics including wide availability, diverse pharmacological actions and a generally good profile of safety and tolerability. Over the recent years, there have been numerous reports from clinical studies testifying to the efficacy and safety of medicinal plants and phytochemicals in ameliorating several human diseases. A plethora of basic studies has also unravelled molecular mechanisms underlying the health benefits of herbal medicines. Nevertheless, issues such as identification of bioactive ingredients, standardization of the products and drug interactions remain to be further studied. In this book, we aim to put together several chapters on the medicinal properties and pharmacological action of medicinal plants, plant species and phytochemicals. The goal of this contributed volume is to present a comprehensive collection on most important therapeutic aspects of plant-derived natural products and their molecular mechanisms of action.

    Abstract: Proteins are exposed to various interfacial stresses during drug product development. They are subjected to air-liquid, liquid-solid, and, sometimes, liquid-liquid interfaces throughout the development cycle-from manufacturing of drug substances to storage and drug delivery. Unlike small molecule drugs, proteins are typically unstable at interfaces where, on adsorption, they often denature and form aggregates, resulting in loss of efficacy and potential immunogenicity. This book covers both the fundamental aspects of proteins at interfaces and the quantification of interfacial behaviors of proteins.     Abstract: This book is designed to provide pharmacologists and researchers of natural products a comprehensive review of 200 medicinal plants, their vernacular names in various languages and their medicinal uses around the world, and in some cases, a historical perspective. Chemical constituents of each plant with the putative active constituent, and available up to date pharmacological studies (until 2017 on PubMed) with each medical activity explored and its relationship with traditional uses, are described for each plant. Any variations in chemical constituents and their effects on pharmacological studies outcome have been highlighted. All clinical trials conducted, with sufficient details, have been included. Nationalities and racial identities of participants of clinical trials are identified to impress upon the social, cultural and dietary influences on the clinical outcomes. Toxicity studies and potential interactions with prescribed drugs, and full spectrum of references are included.

    Abstract: This volume – for pharmacologists, systems biologists, philosophers and historians of medicine – points to investigate new avenues in pharmacology research, by providing a full assessment of the premises underlying a radical shift in the pharmacology paradigm. The pharmaceutical industry is currently facing unparalleled challenges in developing innovative drugs. While drug-developing scientists in the 1990s mostly welcomed the transformation into a target-based approach, two decades of experience shows that this model is failing to boost both drug discovery and efficiency. Selected targets were often not druggable and with poor disease linkage, leading to either high toxicity or poor efficacy. Therefore, a profound rethinking of the current paradigm is needed. Advances in systems biology are revealing a phenotypic robustness and a network structure that strongly suggest that exquisitely selective compounds, compared with multitarget drugs, may exhibit lower than desired clinical efficacy. This appreciation of the role of polypharmacology has significant implications for tackling the two major sources of attrition in drug development, efficacy and toxicity. Integrating network biology and polypharmacology holds the promise of expanding the current opportunity space for druggable targets.

    Abstract: Medicine has entered a golden age in which therapeutic agents are becoming widely available due to advances in basic science and technology. As such, many drugs have been developed that target inflammatory processes and/or the immune system. This book is intended for health professionals examining the modulation of inflammation by immunotherapeutic drugs. The immune system fills the primordial role of host defense and resistance to infections with pathogenic microorganisms. Several hematopoietic-derived cells constituting the innate and adaptive immune systems cooperate to provide barriers for microbial colonization and/or promote pathogen destruction within the host. Conversely, many immune cells are also involved in the pathogenesis and propagation of chronic inflammatory diseases. The beginning of this book details various components of the immune system including the cell types, lymphoid tissues, soluble cytokines and surface molecules that are essential for host defense. Breakdowns in immune tolerance, or dysregulated immune responses to antigens derived from self tissues or innocuous sources, can lead to the development of autoimmunity or chronic inflammatory diseases. Pathophysiologic roles for the immune system are detailed in corresponding chapters on autoimmunity, epithelial surfaces (lungs, skin, intestine), and transplantation, with special emphasis placed on immunotherapeutic drug targets. The last section of the book focuses on treatments that stimulate our immune system to specifically target and fight infectious diseases and cancer. In each chapter, the medications used to treat various diseases/conditions in terms of their mechanism of action and other pharmacologic properties are discussed in detail. Chapters begin with a table showing drug names and classifications. The importance of basic science and clinical trials cannot be understated in the context of drug development. As such, the discovery of certain medications that had a lasting impact in medicine and pharmacy are highlighted in chapter subsections named “Bench to Bedside.” Several clinical applications of immunotherapeutic drugs are described within end-of -chapter case studies including practice questions. The Pharmacology of Immunotherapeutic Drugs is a reference for immunologists and clinicians (medical doctors, pharmacists, nurses) examining the modulation of inflammatory processes by a variety of medications targeting the cells and mediators of our immune system.

    Abstract: The main goal of the present book is to deal with the role of nanobiotechnology in skin, soft tissue and bone infections since it is difficult to treat the infections due to the development of resistance in them against existing antibiotics. The present interdisciplinary book is very useful for a diverse group of readers including nanotechnologists, medical microbiologists, dermatologists, osteologists, biotechnologists, bioengineers. Nanotechnology in Skin, Soft-Tissue, and Bone Infections is divided into four sections: Section I- includes role of nanotechnology in skin infections such as atopic dermatitis, and nanomaterials for combating infections caused by bacteria and fungi. Section II- incorporates how nanotechnology can be used for soft-tissue infections such as diabetic foot ulcer and other wound infections; Section III- discusses about the nanomaterials in artificial scaffolds bone engineering and bone infections caused by bacteria and fungi; and also about the toxicity issues generated by the nanomaterials in general and nanoparticles in particular. The readers will be immensely enriched by the knowledge of new and emerging nanobiotechnologies in a variety of platforms.

    Abstract: Lipids are an integral part of cell membrane architecture, are intermediaries in cell metabolism, and are involved in transmitting cell signals from hormones, growth factors and nutrients. A number of lipases and phospholipases, lipid kinases, lipid phosphatases, sphingosine kinases, and their reaction products have been implicated in fundamental cellular processes including cell proliferation, division and migration. These enzymes and their products underlie the molecular mechanisms of numerous human diseases, in particular metabolic disease (diabetes), cancer, neurodegenerative disease and cardiovascular disease. Over the last decade, studies have advanced to the point that a number of inhibitors for these enzymes have been developed to attempt to ameliorate these conditions; some of the inhibitors are currently in human clinical trial. The need for this book is to review the current status of this field and the prospect for the inhibitors to be clinically important. .
