In this study, we aimed to clarify the roles of insulin and glucose in leptin secretion and mRNA production using inhibitors of insulin signal transduction in adipocytes cultured under glucose-free or normal conditions. Methods Differentiated 3T3-L1 adipocytes were stimulated with insulin in combination with inhibitors for phosphoinositide 3-kinase (PI3K), Akt, and phosphodiesterase 3B (PDE3B), as well as epinephrine and a cyclic AMP (cAMP) analog under glucose-free or normal conditions. After 8 h of stimulation, leptin protein levels in the media and leptin mRNA expression levels in the adipocytes were measured. Results Insulin significantly increased the secretion and mRNA levels of leptin under the depletion of glucose. Glucose augmented basal leptin secretion without insulin, while glucose nullified insulin-induced leptin mRNA upregulation. The PI3K inhibitor BEZ-235, the Akt inhibitor MK-2206, and the PDE3B inhibitor cilostazol attenuated the insulin stimulation of leptin secretion, but did not suppress the insulin-induced leptin mRNA upregulation with glucose depletion. In contrast to the glucose-free condition, insulin failed to upregulate leptin mRNA in the presence of glucose. The cAMP analog dibutyryl cAMP and epinephrine decreased both leptin secretion and mRNA regardless of glucose supplementation. Conclusion Insulin alone stimulates leptin secretion and elevates leptin mRNA levels via cAMP under the lack of glucose metabolism, while glucose is a significant and ambivalent effector on the insulin effects of leptin.

    Description: Publication date: November 2016 Source: Heliyon, Volume 2, Issue 11 Author(s): Akram El Kateb, Claudio Stalder, Christoph Neururer, Chiara Pisapia, Silvia Spezzaferri During an expedition in 2014 in the Gulf of Gabes that aimed to evaluate the impact of the pollution of the phosphate industry on the marine environment, numerous dead coral fragments were retrieved from several stations along a 18 km long transect in front of the industry complex of Gabes. Detailed taxonomy of these coral fragments shows clearly that all fragments belong to the species Cladocora caespitosa (Linnaeus, 1758). Quantitative analysis of the coral fragments indicates a positive correlation with stations characterized by positive bathymetric anomalies. We suggest the presence of probable small-scaled (up to 4 m high) biogenic (palaeo-) build-ups composed mainly of coral colonies and bryozoans. Radiocarbon dating of three coral fragments show ages as old as 1897, 1985 and 1986 AD and suggests the presence of living C. caespitosa as close as 6 km to the phosphate treatment industry of Gabes at least until 1986 AD. This latter age coincides with the construction of the ammonium phosphate production plant, in 1979, in the Gulf of Gabes with an increase of the natural phosphate production. The higher impact of pollution on the marine environment in the inner part of the Gulf of Gabes likely induced the decline of C. caespitosa . This is well in agreement with enhanced siltation processes suggested by the sedimentary facies and grain-size analyses presently characterizing the Gulf of Gabes nowadays.

For these purposes, Italian products belonging to different categories (cooked ham, “selected” cooked ham and “high quality” cooked ham) were evaluated by sensory descriptive analysis and by the application of rapid tools such as image analysis by an “electronic eye” and texture analyzer. The panel of trained assessors identified and evaluated 10 sensory descriptors able to define the quality of the products. Statistical analysis highlighted that sensory characteristics related to appearance and texture were the most significant in discriminating samples belonged to the highest (high quality cooked hams) and the lowest (cooked hams) quality of the product whereas the selected cooked hams, showed intermediate characteristics. In particular, high quality samples were characterized, above all, by the highest intensity of pink intensity, typical appearance and cohesiveness, and, at the same time, by the lowest intensity of juiciness; standard cooked ham samples showed the lowest intensity of all visual attributes and the highest value of juiciness, whereas the intermediate category (selected cooked ham) was not discriminated from the other. Also physical-rheological parameters measured by electronic eye and texture analyzer were effective in classifying samples. In particular, the PLS model built with data obtained from the electronic eye showed a satisfactory performance in terms of prediction of the pink intensity and presence of fat attributes evaluated during the sensory visual phase. This study can be considered a first application of this combined approach that could represent a suitable and fast method to verify if the meat product purchased by consumer match its description in terms of compliance with the claimed quality.

Bode Biological diversity arises among genetically equal subpopulations in the same environment, a phenomenon called phenotypic heterogeneity. The life cycle of the enteric bacterium Photorhabdus luminescens involves a symbiotic interaction with nematodes as well as a pathogenic association with insect larvae. P. luminescens exists in two distinct phenotypic forms designated as primary (1°) and secondary (2°). In contrast to 1° cells, 2° cells are non-pigmented due to the absence of natural compounds, especially anthraquinones (AQs). We identified a novel type of transcriptional regulator, AntJ, which activates expression of the antA - I operon responsible for AQ production. AntJ heterogeneously activates the AQ production in single P. luminescens 1° cells, and blocks AQ production in 2° cells. AntJ contains a proposed ligand-binding WYL-domain, which is widespread among bacteria. AntJ is one of the rare examples of regulators that mediate heterogeneous gene expression by altering activity rather than copy number in single cells.

The criteria for collocations in these languages, however, have not been fully established in the literature, and it is not obvious whether all statistically significant combinations of words could be regarded as collocations. In this article, we point out empirical problems in extracting collocations in Japanese and Korean, and provide a standard for identifying collocations (to be separated from “free combination” and “idiom”) in these languages. We concentrate on the discussion of empirical aspects of collocation research, rather than the statistical analyses of collocational patterns. As a preliminary to developing a database for Japanese-Korean contrastive work on collocations, the present study focuses on ten “Chinese-words” (漢語). We show that (i) the co-occurring verbs for eight Chinese-words in Korean all carry over to Japanese (but not vice versa); (ii) in the other two cases, Korean exhibits co-occurring verbs not found in Japanese; (iii) language-specific patterns of verb co-occurring are also observed in some instances. Overall, it is indicated that a much wider variety of co-occurring verbs are found in Japanese than in Korean.

Kondo The flowing lithium target of a Li(d,xn) fusion relevant neutron source must evacuate the deuteron beam power and generate in a stable manner a flux of neutrons with a broad peak at 14 MeV capable to cause similar phenomena as would undergo the structural materials of plasma facing components of a DEMO like reactors. Whereas the physics of the beam-target interaction are understood and the stability of the lithium screen flowing at the nominal conditions of IFMIF (25 mm thick screen with +/–1 mm surface amplitudes flowing at 15 m/s and 523 K) has been demonstrated, a conclusive assessment of the evaporation and condensation of lithium during operation was missing. First attempts to determine evaporation rates started by Hertz in 1882 and have since been subject of continuous efforts driven by its practical importance; however intense surface evaporation is essentially a non-equilibrium process with its inherent theoretical difficulties. Hertz-Knudsen-Langmuir (HKL) equation with Schrage’s ‘accommodation factor’ η = 1.66 provide excellent agreement with experiments for weak evaporation under certain conditions, which are present during a Li(d,xn) facility operation. An assessment of the impact under the known operational conditions for IFMIF (574 K and 10 −3 Pa on the free surface), with the sticking probability of 1 inherent to a hot lithium gas contained in room temperature steel walls, is carried out. An explanation of the main physical concepts to adequately place needed assumptions is included.

In this study we evaluated the correlation of leptin and CRP in patients with RA and normal controls. Main methods A total of 75 patients with RA and 40 healthy adults were recruited in this case-control study. RA patients were categorized into high (DAS–28 > 3.2) and low activity (DAS ≤ 3.2) group according to their DAS-28 score. Key findings Leptin level was significantly correlated with CRP in healthy controls (r = 0.365; p 〈 0.05), but this correlation was lost in RA patients (r = 0.095, p = 0.41). Patients with RA had higher serum leptin levels compared to healthy controls (P 〈 0.01). No difference in serum leptin level was observed between patients with high and low activity disease. Also leptin was correlated with BMI in healthy controls (r = 0.326, p = 0.037). This correlation was not present in RA patients (r = 0.039, p = 0.756). Significance We observed that the physiologic correlation between leptin and CRP and BMI and CRP was not present RA patients. This is a new study reporting the lost correlation between leptin and CRP in RA patients.

Gaudio An open issue still under investigation by several international entities working on the safety and security field for the foreseen nuclear fusion reactors is the estimation of source terms that are a hazard for the operators and public, and for the machine itself in terms of efficiency and integrity in case of severe accident scenarios. Source term estimation is a crucial key safety issue to be addressed in the future reactors safety assessments, and the estimates available at the time are not sufficiently satisfactory. The lack of neutronic data along with the insufficiently accurate methodologies used until now, calls for an integrated methodology for source term estimation that can provide predictions with an adequate accuracy. This work proposes a complete methodology to estimate dust source terms starting from a broad information gathering. The wide number of parameters that can influence dust source term production is reduced with statistical tools using a combination of screening, sensitivity analysis, and uncertainty analysis. Finally, a preliminary and simplified methodology for dust source term production prediction for future devices is presented.

The latter one requires a paracrine interaction between type II alveolar cells and lipofibroblasts in which leptin triggers PTHrP-induced effects. Whether increased plasma leptin levels, as they occur in high fat diet, modify the expression of PTHrP remains unclear. Furthermore, the effect of high fat diet under conditions of forced pulmonary remodelling such as response to post myocardial infarction remains to be defined. Materials and methods C57 bl/6 mice were randomized to either normal diet or high fat diet at an age of 6 weeks. Seven months later, the mice were euthanized and the lung was removed and frozen in fluid nitrogen until use. Samples were analyzed by real-time RT-PCR and western blot. Leptin deficient mice were used to investigate the effect of leptin on pulmonary expression of PTHrP more directly. A subgroup of mice with and without high fat diet underwent in vivo ischemia (45 min) and reperfusion (4 weeks). Finally, experiments were repeated with prolonged high-fat diet. Key findings High fat diet increased plasma leptin levels by 30.4% and the pulmonary mRNA expression of PTHrP (1,447-fold), PTH-1 receptor (4.21-fold), and PTHrP-downstream targets ADRP (7.54-fold) and PPARγ (5.27-fold). Pulmonary PTHrP expression was reduced in leptin deficient mice by 88% indicating leptin dependent regulation. High fat diet further improved changes in pulmonary adaptation caused by ischemia/reperfusion (1.48-fold increased PTH-1 receptor protein expression). These effects were lost during prolonged high fat diet. Significance This study established that physiological regulation of leptin plasma levels by high fat diet affects the pulmonary PTHrP expression and of PTHrP downstream targets. Modification of pulmonary expression of PTH-1 receptors by high fat diet after myocardial infarction suggests that the identified interaction may participate in the obesity paradox.

However, the use of ZnO NPs as food additive is limited by the perceived high toxicity of ZnO NPs in many earlier toxicity studies. In this study, surface modification by silica coating was used to reduce the toxicity of ZnO NPs by significantly reducing the dissolution of the core ZnO NPs. To more accurately recapitulate the scenario of ingested ZnO NPs, we tested our as synthesized ZnO NPs in ingestion fluids (synthetic saliva and synthetic gastric juice) to determine the possible forms of ZnO NPs in digestive system before exposing the products to colorectal cell lines. The results showed that silica coating is highly effective in reducing toxicity of ZnO NPs through prevention of the dissociation of ZnO NPs to zinc ions in both neutral and acidic condition. The silica coating however did not alter the desired antimicrobial activity of ZnO NPs to E. coli and S. aureus . Thus, silica coating offered a potential solution to improve the biocompatibility of ZnO NPs for applications such as antimicrobial agent in foods or food related products like food packaging. Nevertheless, caution remains that high concentration of silica coated ZnO NPs can still induce undesirable cytotoxicity to mammalian gut cells. This study indicated that upstream safer-by-design philosophy in nanotechnology can be very helpful in a product development.

Cymbidium mosaic virus (CymMV) is one of the common orchid viruses found in Cymbidium species but it infects different orchid genera. In this study Cymbidium aloifolium was propagated in vitro using MS medium at different strength (1.0, ½, and ¼) with or without 0.5 mg/l BAP (6-benzylaminopurine) and 0.5 mg/l NAA (Naphthalene acetic acid). To provide disease-free planting material, source plant for in vitro propagation needs to be screened for pathogenic viruses. In the present study, in vivo -grown source (mother) plants and tissue culture-derived plants of C. aloifolium were tested for CymMV virus using Double antibody sandwich enzyme linked immunosorbent assay (DAS-ELISA). All the tissue cultured plants were found to be 100% virus-free whereas the in vivo grown source plants were highly affected by CymMV virus (83.33%). The virus-free in vitro plantlets were multiplied in large scale and then acclimatized on earthen pot containing a mixture of cocopeat, litter and clay in the ratio of 3:2:1. Eighty five percent of acclimatized plantlets survived making this method an efficient mass production system for high quality virus-free C. aloifolium for commercial floriculture and germplasm preservation.

Inness, Avril Mansfield Performance decrements in balance tasks are often observed when a secondary cognitive task is performed simultaneously. This study aimed to determine whether increased cognitive load resulted in altered reactive stepping in individuals with sub-acute stroke, compared to a reactive stepping trial with no secondary task. The secondary purpose was to determine whether differences existed between the first usual-response trial, subsequent usual-response trials, and the dual-task condition. Individuals with sub-acute stroke were exposed to external perturbations to elicit reactive steps. Perturbations were performed under a usual-response (single-task) and dual-task condition. Measures of step timing and number of steps were based on force plate and video data, respectively; these measures were compared between the usual-response and dual-task trials, and between the first usual-response trial, later usual-response trials (trials 2–5) and a dual-task trial. A longer time of unloading onset and greater number of steps were identified for the first usual-response trial compared to later usual-response trials. No significant differences were identified between usual-response and dual-task trials. Although improvements were observed from the first to subsequent usual-response lean-and-release trials, performance then tended to decrease with the introduction of the dual-task condition. These findings suggest that when introduced after usual-response trials, the dual-task trial may represent the first trial of a new condition, which may be beneficial in reducing the potential for adaptation that may occur after multiple repetitions of a reactive stepping task. Therefore, these findings may lend support to the introduction of a new condition (i.e. a dual-task trial) in addition to usual-response trials when assessing reactive balance in individuals with stroke.

Paul The present study employed mass spectrometry (ICP-MS) to measure the internal cadmium concentrations (Cd int ) in Caenorhabditis elegans to determine Cd uptake from a Cd-containing environment as well as Cd release under Cd-free conditions. To analyze the functional role of several ATP binding cassette (ABC) transporters (e.g., HMT-1 and MRP-1) and phytochelatin synthase (PCS), we compared wild-type (WT) and different mutant strains of C. elegans . As a pre-test on selected mutant strains, several time-resolved experiments were performed to determine the survival rate and avoidance behavior of C. elegans under Cd stress, which confirmed the already known Cd sensitivity of the deletion mutants mrp-1 Δ, pcs-1 Δ, and hmt-1 Δ. In addition, these experiments revealed flight reactions under Cd stress to be almost completely absent in mrp-1 Δ mutants. The ICP-MS studies showed Cd uptake to be significantly higher in mrp-1 Δ and WT than in hmt-1 Δ. As Cd is ingested with food, food refusal due to very early Cd stress and its perception was likely the reason for the reduced Cd uptake of hmt-1 Δ. Cd release (detoxification) was found to be maximal in mrp-1 Δ, minimal in hmt-1 Δ, and intermediate in WT. High mortality under Cd stress, food refusal, and minimal Cd release in the case of hmt-1 Δ suggest a vital importance of the HMT-1/PCS-1 detoxification system for the survival of C. elegans under Cd stress. High mortality under Cd stress, absence of an avoidance behavior, missing food refusal, and maximal Cd release in the case of mrp-1 Δ indicate that MRP-1 is less important for Cd detoxification under severe stress, but is probably important for Cd perception. Accordingly, our results suggest that the survival of WT under Cd stress (or possibly other forms of metal stress) primarily depends on the function of the HMT-1/PCS-1 detoxification system and the presence of a sensing mechanism to control the uptake of Cd (or other metals), which keeps internal Cd (or metal) concentrations under control, to some extent, for the timely mobilization of protection and repair systems.

Nevertheless, intraocular pressure (IOP) reduction alone was not enough to inhibit the progression of disease, which suggests the contribution of other factors to the glaucoma pathogenesis. In this study, we investigated the cytoprotective effect of geranylgeranylacetone (GGA) on RGCs degeneration using a normal tension glaucoma (NTG) mouse model, which lacks glutamate/aspartate transporter (GLAST) and demonstrates spontaneous RGC and optic nerve degeneration without elevated intraocular pressure (IOP). Three-week-old GLAST +/− mice were given oral administration of GGA at 100, 300, or 600 mg/kg/day or vehicle alone, and littermate control mice were given vehicle alone for 14 days, respectively. At 5 weeks after birth, the number of RGCs was counted in paraffin sections of retinal tissues stained with hematoxylin and eosin. In addition, retrograde labeling technique was also used to quantify the number of RGC. Expression and localization of heat shock protein 70 (HSP70) in retinas were evaluated by reverse transcription polymerase chain reaction and immunohistochemistry, respectively. Activities of caspase-9 and -3 in retinas were also assessed. The number of RGCs of GLAST +/− mice significantly decreased, as compared to that of control mice. RGC loss was significantly suppressed by administration of GGA at 600 mg/kg/day, compared with vehicle alone. Following GGA administration, HSP70 was significantly upregulated together with reduction in the activities of caspase-9 and -3. Our studies highlight HSP70 induction in the retina is available to suppress RGC degeneration, and thus GGA may be applicable for NTG as a promising therapy.

The structural, topological, electronic and vibrational properties were investigated in both media by using the hybrid B3LYP/6-31G* method and the integral equation formalism variant polarised continuum model (IEFPCM). Here, the characteristics of the S═O, S−O, N⋯H, O⋯H, Na⋯O bonds were completely revealed by using atomic charges, natural bond orbital (NBO) and atoms in molecules (AIM) studies. The infrared, 1 H NMR, 13 C NMR and UV-visible spectra are in reasonable concordance with those experimental available in the literature. The vibrational analysis of sodium picosulphate was performed considering C 3V symmetries for both SO 4 2− groups and the complete assignments of the 126 vibration modes were reported in gas phase and aqueous solution together with their corresponding force fields. In addition, the reactivities of sodium picosulfate increase in solution due to their ionic characteristic which probably justifies their behaviour as a stimulant cathartic and their easy metabolic conversion, as reported in the literature.

The damage resulting from collisions could be reduced through several techniques such as designing appropriate hull structures, ensuring tightness of cargo tanks as well as observation and review on structural behaviors, whilst accounting for all involved parameters. The position during a collision can be influenced by the collisions’ location and angle as these parts are included in the external dynamics of ship collisions. In this paper, the results of several collision analyses using the finite element method were used and reviewed regarding the effect of location and angle on energy characteristic. Firstly, the capabilities of the structure and its ability to resist destruction in a collision process were presented and comparisons were made to other collision cases. Three types of collisions were identified based on the relative location of contact points to each other. From the results, it was found that the estimation of internal energy by the damaged ships differed in range from 12%–24%. In the second stage, the results showed that a collision between 30 to 60 degrees produced higher level energy than a collision in the perpendicular position. Furthermore, it was concluded that striking and struck objects in collision contributed to energy and damage shape.

We postulate that such exploratory behavior and emotional experience both are regulated based on the interdependent perception of one’s body and stimuli that collectively define a spatial context such as a cliff. Here we examined this by testing if the interaction of the sensory input on one’s gait and the sensory input on the spatial context is modulating both the emotional experience of the environment and its exploration through head motion. To this end, we asked healthy humans to explore a life-sized Virtual Reality simulation of a forest glade by physically walking around in this environment on two narrow rectangular platforms connected by a plank. The platforms and the plank were presented such that they were either placed on ground or on the top of two high bridge piers. Hence, the forest glade was presented either as a “ground” or as a “height” context. Within these two spatial contexts the virtual plank was projected either on the rigid physical floor or onto a bouncy physical plank. Accordingly, the gait of our participants while they crossed the virtual plank was either “smooth” or “bouncy.” We found that in the height context bouncy gait compared to smooth gait increased the orientation of the head below the horizon and intensified the experience of the environment as negative. Whereas, within the ground context bouncy gait increased the orientation of the head towards and above the horizon and made that the environment was experienced as positive. Our findings suggest that the brain of healthy humans is using the interaction of the sensory input on their gait and the sensory input on the spatial context to regulate both the emotional experience of the environment and its exploration through head motion.

The model is based on a 2-port transmission line representation of each conductor, and the further assembling of a network representing the complete structure. Horizontal and vertical conductors are modeled using formulas based on the complex images method, in order to take into account frequency dependence. Variation of electrical parameters with height is also considered for vertical conductors. This is accomplished by means of a non-uniform modeling approach based on conductor subdivision and cascaded connection of chain matrices computed for each segment. The results from the model are validated by means of comparisons with measurements reported elsewhere, as well as simulations using PSCAD/EMTDC.

However, the differences in dynamic verbal processing under different cognitive conditions are unclear. To clarify this, we conducted an electrophysiological experiment involving a semantic priming paradigm in which semantically congruent or incongruent word sequences (prime nouns–target verbs) were randomly presented. We examined the event-related brain potentials that occurred in response to congruent and incongruent target words that were preceded by primes with or without grammatical case markers. The two participant groups performed either the shallow (lexical judgment) or deep (direct semantic judgment) semantic tasks. We hypothesized that, irrespective of the case markers, the congruent targets would reduce centro-posterior N400 activities under the deep semantic condition, which induces selective attention to the semantic relatedness of content words. However, the same congruent targets with correct case markers would reduce lateralized negativity under the shallow semantic condition because grammatical case markers are related to automatic structural integration under semantically unattended conditions. We observed that congruent targets (e.g., ‘open') that were preceded by primes with congruent case markers (e.g., ‘shutter-object case') reduced lateralized negativity under the shallow semantic condition. In contrast, congruent targets, irrespective of case markers, consistently yielded N400 reductions under the deep semantic condition. To summarize, human neural verbal processing differed in response to the same grammatical markers in the same verbal expressions under semantically attended or unattended conditions.

Sutherland Small cell lung cancer (SCLC) is the most aggressive type of lung cancer, with almost 95% of patients succumbing to the disease. Although RBM5 , a tumor suppressor gene, is downregulated in the majority of lung cancers, its role in SCLC is unknown. Using the GLC20 SCLC cell line, which has a homozygous deletion encompassing the RBM5 gene locus, we established stable RBM5 expressing sublines and investigated the effects of RBM5 re-expression. Transcriptome and target identification studies determined that RBM5 directly regulates the cell cycle and apoptosis in SCLC cells, as well as significantly downregulates other important transformation-associated pathways such as angiogenesis and cell adhesion. RNA sequencing of paired non-tumor and tumor SCLC patient specimens showed decreased RBM5 expression in the tumors, and expression alterations in the majority of the same pathways that were altered in the GLC20 cells and sublines. Functional studies confirmed RBM5 expression slows SCLC cell line growth, and increases sensitivity to the chemotherapy drug cisplatin. Overall, our work demonstrates the importance of RBM5 expression to the non-transformed state of lung cells and the consequences of its deletion to SCLC development and progression.

This individual difference is conceptualized in different aspects, such as wake up times, bed times, times of peak performance, as well as in morning affect. A total of 14,987 visitors of an exhibition in the LWL State Museum of Natural History, Münster (Germany), did the survey on chronotype and gave their consent that these data can be used for a scientific study. Age groups were coded into 5-year bins. Mean age (mean ± SD) was 28.2 ± 17.5 years. There were 8075 females (54%) and 6912 males in the sample. The German version of the rMEQ (reduced Morningness-Eveningness-Questionnaire) was used for data collection. The data showed clear age effects. Younger children are more morning oriented and become rapidly evening oriented during puberty, while the more attenuated turn towards morningness occurs from the age of 20 years. Then between the ages 25 to 30 morningness-eveningness remained rather stable. Significant gender differences existed in the reproductive age, i.e., the age groups 20 to 50 (corresponding to the age 16–50 years). In other age groups, no gender differences could be detected. Seasonal effects were also found. Chronotype score was lowest during the summer months (and more evening oriented). Based on the single item analysis of the five questions of the rMEQ, we found age group differences in all items. Gender differences occurred in all items except item 1, which deals with the preferred wake-up time. Men always scored significantly lower (i.e. more evening oriented) than women except in item 2 (tiredness after awakening). Seasonal effects were only significant in item 3, which is related to preferred bed times. People showed a later bed time preference during summer. The classification of chronotypes according to the cut-off scores provided by Adan and Almirall (1991) and by using the 20/80 percentile provided identical cut-off scores (values of 11 and below for evening types and 18 and above for morning types).

This is likely to increase the financial crisis due to limited resources for funding procurement of ready-made products from the pharmaceuticals manufacturing leading countries. Therefore, production of local oral tablets containing Lamivudine and TDF FDC is inevitable. Lamivudine 300 mg/TDF 300 mg tablets were developed and optimized by D-optimal mixture design and produced by direct compression technique. Twenty trial formulations with independent variables, including PVP-CL 1–12.00%, PVP-K30 1–10.00%, starch-1500 2.5–12.5% and Avicel-PH102 2–19.25% were prepared by direct compression technique. The formulations were assessed on assay, dissolution, friability, weight variation and disintegration time. It was found that assay ranged from 98.13–101.95% for Lamivudine, 98.25–102.84 for TDF, both were within the in-house assay specification of 95 to 105%. Dissolution at single point was above 80% for Lamivudine 93.96–100.55% and 95.85–103.15% for TDF, disintegration time was between 1.92–66.33 min and friability 0.06–12.56%. Out of twenty formulation trials, eight formulations had all parameters in proven acceptable range. On optimization, one formulation with independent variables, PVP-CL 5.67%, PVP-K30 1.00%, Starch-1500 5.76% was selected. The optimized formulation was comparable to the reference product on the market with similarity factor (f2) and difference factor (f1) within the acceptable range for both Lamivudine and TDF.

Sweet Objectives Microfluidic perfusion systems are used for assessing cell and tissue function while assuring cellular viability. Low perfusate flow rates, desired both for conserving reagents and for extending the number of channels and duration of experiments, conventionally depend on peristaltic pumps to maintain flow yet such pumps are unwieldy and scale poorly for high-throughput applications requiring 16 or more channels. The goal of the study was to develop a scalable multichannel microfluidics system capable of maintaining and assessing kinetic responses of small amounts of tissue to drugs or changes in test conditions. Methods Here we describe the BaroFuse, a novel, multichannel microfluidics device fabricated using 3D-printing technology that uses gas pressure to drive large numbers of parallel perfusion experiments. The system is versatile with respect to endpoints due to the translucence of the walls of the perifusion chambers, enabling optical methods for interrogating the tissue status. The system was validated by the incorporation of an oxygen detection system that enabled continuous measurement of oxygen consumption rate (OCR). Results Stable and low flow rates (1–20 μL/min/channel) were finely controlled by a single pressure regulator (0.5–2 psi). Control of flow in 0.2 μL/min increments was achieved. Low flow rates allowed for changes in OCR in response to glucose to be well resolved with very small numbers of islets (1–10 islets/channel). Effects of acetaminophen on OCR by precision-cut liver slices of were dose dependent and similar to previously published values that used more tissue and peristaltic-pump driven flow. Conclusions The very low flow rates and simplicity of design and operation of the BaroFuse device allow for the efficient generation of large number of kinetic profiles in OCR and other endpoints lasting from hours to days. The use of flow enhances the ability to make measurements on primary tissue where some elements of native three-dimensional structure are preserved. We offer the BaroFuse as a powerful tool for physiological studies and for pharmaceutical assessment of drug effects as well as personalized medicine.

Walker Wind erosion not only affects agricultural productivity but also soil, air, and water quality. Dust and specifically particulate matter ≤10 μm (PM-10) has adverse effects on respiratory health and also reduces visibility along roadways, resulting in auto accidents. The Wind Erosion Prediction System (WEPS) was developed by the USDA-Agricultural Research Service to simulate wind erosion and provide for conservation planning on cultivated agricultural lands. A companion product, known as the Single-Event Wind Erosion Evaluation Program (SWEEP), has also been developed which consists of the stand-alone WEPS erosion submodel combined with a graphical interface to simulate soil loss from single (i.e., daily) wind storm events. In addition to agricultural lands, wind driven dust emissions also occur from other anthropogenic sources such as construction sites, mined and reclaimed areas, landfills, and other disturbed lands. Although developed for agricultural fields, WEPS and SWEEP are useful tools for simulating erosion by wind for non-agricultural lands where typical agricultural practices are not employed. On disturbed lands, WEPS can be applied for simulating long-term (i.e., multi-year) erosion control strategies. SWEEP on the other hand was developed specifically for disturbed lands and can simulate potential soil loss for site- and date-specific planned surface conditions and control practices. This paper presents novel applications of WEPS and SWEEP for developing erosion control strategies on non-agricultural disturbed lands. Erosion control planning with WEPS and SWEEP using water and other dust suppressants, wind barriers, straw mulch, re-vegetation, and other management practices is demonstrated herein through the use of comparative simulation scenarios. The scenarios confirm the efficacy of the WEPS and SWEEP models as valuable tools for supporting the design of erosion control plans for disturbed lands that are not only cost-effective but also incorporate a science-based approach to risk assessment.

UV-Vis spectra, electrical conductivity and pH method were also used to characterize the final product. The stoichiometry of the reaction complex formation was determined via different referential methods. It was observed that, the reaction complex was formed when the concentrations were smaller than a certain limit (10 −5 M). Adjusting the pH of the reaction (typically from 3.7 to 6.0) also resulted in the formation of the complex. The formed complex was highly stable in dark conditions (absence of sunlight) and at ambient temperature. Without the use of additives and by employing the investigated optimal conditions (i.e. pH: 5.0, i: 6 mA/cm 2 , t: 5 min, C: 0.1 M, d: 1.273 × 10 −4 cm), electrodeposition of tin (II) was demonstrated to be successful.

We studied the biological alterations in the medial prefrontal cortex (mPFC), which is a region that is highly implicated in the neurobiology of MDD, and the blood cells (BCs) of ovariectomized (OVX) mice subjected to chronic mild stress (CMS), which represents a mouse model of depression during menopause. Main methods The mPFC and the BCs were obtained from the same individuals. Gene expression levels were analyzed by microarray. The data were used for the Ingenuity Pathway Analysis and the Gene Ontology analysis. Key findings The gene expression alterations (GEAs) induced by OVX were mainly associated with ribosomal and mitochondrial functions in both the mPFC and the BCs. Rapamycin-insensitive companion of mTOR (RICTOR) was identified as a possible upstream regulator of the OVX-induced GEAs in both tissues. The CMS-induced GEAs were associated with retinoic acid receptor signaling, inflammatory cytokines and post-synaptic density in the mPFC, but not in the BCs. Significance OVX and CMS independently affect biological pathways in the mPFC, which is involved in the development of the depression-like phenotype. Because a subset of the OVX-induced GEAs in the mPFC also occurred in the BCs, the GEAs in the BCs might be a useful probe to predict biological pathways in the corresponding brain tissue under specific conditions such as OVX in females.

It has been suggested that S1P gradient with high concentrations in the blood and lymphatic fluid and low concentrations in the peripheral tissue plays important roles in immune cell trafficking and potentially cancer progression. However, only a few reports have assessed S1P levels in the lymphatic fluid due to lack of an established easy-to-use method. Here, we report a simple technique for collection of lymphatic fluid to determine S1P. Materials and methods Lymphatic fluid was collected directly with a catheter needle (classical method) or was absorbed onto filter paper after incision of cisterna chyli (new method) in murine models. Blood, lymphatic fluid and mesenteric lymph nodes were corrected from wild type and sphingosine kinase 2 (SphK2) knockout mice to determine S1P levels by mass spectrometry. Results The volume of lymphatic fluid collected by the new method was at least three times greater than those collected by the classical method. S1P concentrations in lymphatic fluid are lower than in blood and higher than in lymph nodes. Interestingly, S1P levels in lymphatic fluid from SphK2 knockout mice were significantly higher than those in wild type, suggesting an important role of SphK2 and/or SphK1 to regulate S1P levels in lymphatic fluid. Conclusions In agreement with the previous theory, our results confirm “S1P gradient” among blood, lymphatic fluid and peripheral lymphatic tissues. Convenient methods for collection and measurement of sphingolipids in lymphatic fluid are expected to provide new insights on functions of sphingolipids.

The pressure-volume-temperature ( P-V-T ) data were collected at up to 111 GPa and room temperature for the diamond-anvil cell experiments and at up to 15 GPa and 1300 K for the multianvil experiments. No phase transition was observed through the entire range of experimental conditions. The pressure-volume data at room temperature were fitted using a Vinet EOS to obtain the isothermal bulk modulus, B T0 = 256.7 GPa, and its pressure derivative, B ’ T0 = 3.83. When fitting a thermal EOS using the P-V-T data for the multianvil experiments, we find that ( ∂ B T / ∂ T ) V = 0.095 (GPa/K) and α 0 = 2.49 × 10 −5 K −1 .

The current work investigates the protein extractability and techno-functionality of insect flour fractions recovered from Tenebrio molitor and Hermetia illucens . T. molitor and H. illucens flours contained about 20% crude fat and 60% and 36 % crude protein, respectively. Defatting reduced the crude fat content to 2.8% ( T. molitor ) and 8.8% ( H. illucens ) and increased the crude protein content to 68% and 47%, respectively. To isolate proteins from the flours, protein solubility was optimized by varying the pH, the ionic strength, and the extraction temperature of the solvent. All products and by-products accumulated in the protein production process were characterized by composition, selected techno-functional properties, protein solubility, composition and structure as well as their microbial load.

Chiu Sensory evaluation of quinoa gluten-free whole grain low fat and salt snacks was conducted. The snacks were Quinoa, Quinoa-Cayenne Pepper, Quinoa-Ginger and Quinoa-Turmeric. Cayenne pepper, ginger and turmeric are common spices that contain health promoting nutrients. Cayenne pepper has been associated with enhancing heat production. Ginger has been reported to improve blood flow and prevent joint pains. Turmeric has been observed to have wound healing potential. All the snacks contained 6% corn oil and 2% salt. Snack dough was prepared using 120 mL water for 100 g dry ingredients. About 20 g of snack dough was placed on center of preheated KrumKake Express Baker and cooked for 2 min. Seventy in-house volunteers judged Color/Appearance of Quinoa, Quinoa-Cayenne Pepper and Quinoa-Ginger snacks significantly ( p ≤ 0.05) higher than Quinoa-Turmeric snacks. Odor/Aroma of Quinoa-Ginger snacks was significantly higher than other snacks tested. Texture/Mouth-feel of Quinoa-Cayenne Pepper, Quinoa-Ginger and Quinoa-Turmeric snacks was similar and significantly higher than Quinoa snacks. Taste/Flavor and Acceptance was similar in four kinds of snacks tested. Water activity of all the snacks tested ranged from 0.41–0.55 suggesting that these snacks were crispy with good antimicrobial stability. These snacks would be quite filling due to their expansion of 2.6–3.1 times due to high porosity. Acceptance of snacks tested was Quinoa 79%, Quinoa-Cayenne Pepper 77%, Quinoa-Ginger 73% and Quinoa-Turmeric 70%. These snacks contained only 3–4 ingredients and could be made in any house kitchen or commercial production. Acceptance of 70–79% is very desirable. These healthy nutritious gluten-free quinoa snacks offer choice for all including vegetarians and individuals hypersensitive to gluten.

Construction of the IMFET biosensor started with adsorption of 1.5 mg/mL of protein A (PA) onto the insulated gate surface of ISFET for 90 min. Next, the immobilized 1/500 dilution of anti-biotin antibody was adsorbed onto the PA layer for 60 min. The IMFET biosensor was subsequently ready for detection of the biotinylated amplicon. The IMFET biosensor showed highly specific binding to the biotinylated PCR product of the phaE gene of Haloquadratum walsbyi DSM 16854. The phaE gene is a biomarker of polyhydroxyalkanoate (PHA) producers that contain PHA synthase class III. The lowest amount of DNA template of H. walsbyi DSM 16854 that the IMFET biosensor could detect was 125 fg. The IMFET biosensor has a lower amount of detection compared with a DNA lateral flow biosensor from our previous study. The degree of linearity of the biosensor signal was influenced by the concentration of the biotinylated amplicon. The IMFET biosensor also has a short response time (approximately 30 times) to detect the phaE amplicon compared to an agarose gel electrophoresis. The IMFET biosensor is a promising tool for the detection of the biotinylated PCR product, and it can be integrated into a micro total analysis system (μTAS).

Montagna A microfluidic system consisting of generic single use cartridges which interface with a workstation allows the automatic performance of all necessary sample preparation, PCR analysis and interpretation of multiplex PCR assays. The cartridges contain a DNA array with 20 different 16mer DNA “universal” probes immobilized at defined locations. PCR amplicons can be detected via hybridization of user-defined “reporter” probes that are complementary at their 3′ termini to one or more of the universal probes and complementary to the target amplicons at their 5′ termini. The system was able to detect single-plex and multiplex PCR amplicons from various infectious agents as well as wild type and mutant alleles of single nucleotide polymorphisms. The system's ease of use was further demonstrated by converting a published PCR assay for the detection of Mycobacterium genitalium in a fully automated manner. Excellent correlation between traditional manual methods and the automated analysis performed by the workstation suggests that the system can provide a means to easily design and implement a variety of customized PCR-based assays. The system will be useful to researchers or clinical investigators seeking to develop their own user defined assays. As the U.S. FDA continues to pursue regulatory oversight of LDTs, the system would also allow labs to continue to develop compliant assays.

We explored the experiences and concerns of key actors in the informed consent process in two HIV clinical trials. Method Semi-structured interviews were conducted with 46 respondents including trial participants, research study team and research ethics committee members about their experiences during the informed consent process. Three focus group discussions were conducted with 14 Community Advisory Board (CAB) members and 17 trial participants. Data were analysed to identify key themes. Findings The consent process was highlighted as an important procedure by all the key actors however each group had a particular area of emphasis. Signing a consent form was given importance by research team and ethics committee members, because it provided documented evidence of a participant’s willingness to join a clinical trial. Participants did not welcome the presence of a witness for a non-literate participant because understanding study information was not closely related to an ability to read and write. Conclusion This study’s findings indicated that obtaining a volunteer’s signature or thumbprint on a consent form did not necessarily mean that the participant was fully-informed about the information relevant to their taking part nor that they understood all the information shared with them. Informed consent requires sufficient time in the research process to have staff trained well enough before research begins. Ensuring and gaining informed consent should be understood and treated as a relation-centred, dynamic supportive process throughout the duration of a research study.

The survey associated the geotechnical engineering and the sedimentary geology of the late Pleistocene–Holocene deposits at five sites and also undifferentiated Pleistocene sediments. A cross-section which was rebuilt in the delta progradation-direction and between the Mekong and Bassac rivers represents the stratigraphy. Each sedimentary unit was formed under a different delta formation mechanism and revealed a typical geotechnical property sequence. The mechanical behaviors of the sediment succession in the tide-dominated delta with significant fluvial-activity and material source tend to be more cohesionless soils and strengths than those in the tide- and wave-dominated delta and even the coast. The particular tendency of the mechanical behavior of the deposit succession can be reasonably estimated from the delta formation mechanism. The characteristics of the clay minerals from the Mekong River produced the argillaceous soil which does not have extremely high plasticity. The microstructure development-levels are low to very high indicating how to choose hydraulic conductivity value, k, for estimating overconsolidation ratio, OCR, by the piezocone penetration tests (CPTU). The OCR of sediments in the delta types strangely change with depth but none less than 1. The post-depositional processes significantly influenced the microstructure development, particularly the dehydrating and oxidizing processes.

    Description: Publication date: November 2016 Source: Heliyon, Volume 2, Issue 11 Author(s): Nkongho Ayuketang Arreyndip, Ebobenow Joseph, Afungchui David For the future installation of a wind farm in Cameroon, the wind energy potentials of three of Cameroon’s coastal cities (Kribi, Douala and Limbe) are assessed using NASA average monthly wind data for 31 years (1983–2013) and compared through Weibull statistics. The Weibull parameters are estimated by the method of maximum likelihood, the mean power densities, the maximum energy carrying wind speeds and the most probable wind speeds are also calculated and compared over these three cities. Finally, the cumulative wind speed distributions over the wet and dry seasons are also analyzed. The results show that the shape and scale parameters for Kribi, Douala and Limbe are 2.9 and 2.8, 3.9 and 1.8 and 3.08 and 2.58, respectively. The mean power densities through Weibull analysis for Kribi, Douala and Limbe are 33.7 W/m2, 8.0 W/m2 and 25.42 W/m2, respectively. Kribi’s most probable wind speed and maximum energy carrying wind speed was found to be 2.42 m/s and 3.35 m/s, 2.27 m/s and 3.03 m/s for Limbe and 1.67 m/s and 2.0 m/s for Douala, respectively. Analysis of the wind speed and hence power distribution over the wet and dry seasons shows that in the wet season, August is the windiest month for Douala and Limbe while September is the windiest month for Kribi while in the dry season, March is the windiest month for Douala and Limbe while February is the windiest month for Kribi. In terms of mean power density, most probable wind speed and wind speed carrying maximum energy, Kribi shows to be the best site for the installation of a wind farm. Generally, the wind speeds at all three locations seem quite low, average wind speeds of all the three studied locations fall below 4.0m/s which is far below the cut-in wind speed of many modern wind turbines. However we recommend the use of low cut-in speed wind turbines like the Savonius for stand alone low energy needs

Livrea Thirty samples from thirteen Sicilian monofloral honeys by the local black honeybee, and two honeydew honeys, were studied to assess phenol content, reducing power and antioxidant capacity as well as correlations among these parameters. Honeys from Apiaceae showed the highest phenol amount and capacity to reduce ferric ion and stable chemical radicals, whereas honeys from Leguminosae the lowest. All honeys were active against myoglobin-derived radicals usually formed in red meat after storage and/or heating and significant correlation ( p = 0.023) was found between flavonoid content and deactivation rate of this radical. Dill > almond > tangerine > thistle > sulla honeys inhibited formation of lipoperoxides in either iron/ascorbate or azoinitiator -induced membrane lipid oxidation, whereas eucalyptus honey was mostly effective in the metal-dependent model. Honeys by black honeybee possess remarkable reducing power and antioxidant potential against radicals of interest in dietary foodstuffs.

However, serotype 3 remains as an important cause of invasive pneumococcal disease (IPD) in Denmark. IPD surveillance data (1999–2016) was used to calculate the incidence and age-distribution of serotype 3 IPD, and the effect of PCV-13 on serotype 3 IPD incidence was examined. The incidence of serotype 3 IPD in the age group below 65 years was 0.51/100,000 pre PCV-13, and 0.45/100,000 post PCV-13. In the group 0–4 years, serotype 3 IPD incidence was 0.28/100,000 pre PCV-13, and 0.16/100,000 post PCV-13. Serotype 3 IPD incidence in the elderly showed a mean of 4.27/100,000 pre PCV-13, and 4.32/100,000 post PCV-13. PCV-13 childhood immunization in Denmark has not lead to a reduction of the incidence of IPD caused by serotype 3. The reason behind this missing effect needs to be investigated further.

Scully, Nick Birbilis Atomic hydrogen (H) was introduced into steel (AISI 1018 mild steel) by controlled cathodic pre-charging. The resultant steel sample, comprising about 1 ppmw diffusible H, and a reference uncharged sample, were studied using atomic emission spectroelectrochemistry (AESEC). AESEC involved potentiodynamic polarisation in a flowing non-passivating electrolyte (0.6 M NaCl, pH 1.95) with real time reconciliation of metal dissolution using on-line inductively coupled plasma-atomic emission spectroscopy (ICP-OES). The presence of absorbed H was shown to significantly increase anodic Fe dissolution, as evidenced by the enhanced detection of Fe 2+ ions by ICP-OES. We discuss this important finding in the context of previously proposed mechanisms for H-effects on the corrosion of steels.

In India, oral squamous cell carcinoma (OSCC) is the most common cancer with half a million new cases diagnosed every year. More than 50% of patients eventually develop local recurrence or metastasis usually within the first 2-years following completion of treatment. It is beneficial to analyze the prognostic significance of Cyclin D1, p53 and EGFR which are critical mediators in the pathogenesis of OSCC. The objective of this study was to assess the association of expression of these markers with recurrence and pattern of recurrence in OSCC patients undergoing chemoradiation. Materials and Methods A Total 290 OSCC cases of locally advanced stage (III, IV) oral cancer with World Health Organization (W.H.O.) performance status of grade 0/1 in the year 2009–2012 were enrolled in the study. Treatment response was assessed according to W.H.O. criteria. Cyclin D1, EGFR and p53 expression in tumor tissue was estimated by immunohistochemical (IHC) method and quantified as percentage positive nuclei. Results During the 2-years follow up, 56 (19.3%) patients recurred, out of which, 47 (83.9%) were locoregional and 9 (16.1%) distant sites. On correlating, χ 2 test showed significant ( P 〈 0.05 or P 〈 0.01 or P 〈 0.001) association of marker expressions (Cyclin D1, EGFR and p53) with recurrence. The strong positive expressions of all three markers showed significant association with early time of recurrence. The multivariate logistic regression analysis showed significant ( P 〈 0.05 or P 〈 0.01 or P 〈 0.001) association of recurrence with primary site, differentiation, Cyclin D1 and p53 expressions indicating these as an independent predictors of recurrence in OSCC. The Cyclin D1, EGFR and p53 expressions also showed significant ( P 〈 0.001) poor survivals (OS, DFS and RFS) in patients with positive/strong positive expressions than negative expression suggesting their prognosis in OSCC. Conclusion Our results signifies that tumors over expressing Cyclin D1, EGFR and p53 are resistant to chemoradiation and are associated with increased risk of locoregional recurrence and metastasis in OSCC patients undergoing chemoradiation.

Patient outcomes could potentially be improved by targeting genes and pathways that drive neoplastic cell motility and invasiveness, including hypoxia-inducible factor-1 (HIF-1α), NOTCH, and aspartate-β-hydroxylase (ASPH). Methods Human astrocytoma biopsy specimens (n = 37), WHO Grades II–IV, were analyzed for levels and distributions of ASPH and HIF-1α immunoreactivity by immunohistochemical staining, and ASPH, Notch, JAG, HES1, HEY1 and HIF1α mRNA expression by quantigene multiplex analysis. The effects of small molecule inhibitors on ASPH’s catalytic activity, cell viability and directional motility were examined in vitro in established GBM cell lines and primary tumor cells from an invasive mouse model of GBM. Results The highest grade astrocytoma, i.e. GBM was associated with the highest levels of ASPH and HIF1α, and both proteins were more abundantly distributed in hypoxic compared with normoxic regions of tumor. Furthermore, mining of the TCGA database revealed higher levels of ASPH expression in the mesenchymal subtype of GBM, which is associated with more aggressive and invasive behavior. In contrast, lower grade astrocytomas had low expression levels of ASPH and HIF1α. In vitro experiments demonstrated that small molecule inhibitors targeting ASPH’s catalytic activity significantly reduced GBM viability and directional motility. Similar effects occurred in GBM cells that were transduced with a lentiviral sh-ASPH construct. Conclusion This study demonstrates that increased ASPH expression could serve as a prognostic biomarker of gliomas and may assist in assigning tumor grade when biopsy specimens are scant. In addition, the findings suggest that GBM treatment strategies could be made more effective by including small molecule inhibitors of ASPH.

However, the detailed dynamic relations among these changes and EF-G sampling are not clear. Here, based on our proposed pathway of ribosomal translocation, we study theoretically the dynamic relations among these changes exhibited in the single molecule data and those exhibited in the ensemble kinetic data. It is shown that the timing of these changes in the single molecule data and that in the ensemble kinetic data show very different. The theoretical results are in agreement with both the available ensemble kinetic experimental data and the single molecule experimental data.

Roberts While northern and southern Chinese are genetically correlated, there exists notable environmental differences in their living conditions. This study aimed to evaluate validity of the southern Chinese reference dataset for dental age estimation applied to northern Chinese. Dental panoramic tomographs of 437 northern Chinese aged 3 to 21 years were analysed. All the left maxillary and mandibular permanent teeth plus the 2 third molars on the right side were scored based on Demirjian’s classification of tooth development stages. Mean and standard error of dental age were obtained for each tooth development stage, followed by random effect meta-analysis for mean dental age estimation. Validity of the method was examined through measures of agreement (95% limits of agreement, standard error of measurement, and Lin’s concordance correlation coefficient) and measure of reliability (Intraclass correlation coefficient). On average, the estimated dental age overestimated chronological age by only around 1 month in both females and males. The Intraclass correlation coefficient values were 0.99 for both sexes, suggesting excellent reliability of the method. Reference dataset for dental age estimation developed on the basis of southern Chinese was applicable for use among the northern Chinese.

The less negative slope of the fractal scaling of heart rate variability (HRV) of astronauts exposed long-term to microgravity reflects cardiovascular deconditioning. We here focus on specific frequency regions of HRV. Methods Ten healthy astronauts (8 men, 49.1 ± 4.2 years) provided five 24-hour electrocardiographic (ECG) records: before launch, 20.8 ± 2.9 (ISS01), 72.5 ± 3.9 (ISS02) and 152.8 ± 16.1 (ISS03) days after launch, and after return to Earth. HRV endpoints, determined from normal-to-normal (NN) intervals in 180-min intervals progressively displaced by 5 min, were compared in space versus Earth. They were fitted with a model including 4 major anticipated components with periods of 24 (circadian), 12 (circasemidian), 8 (circaoctohoran), and 1.5 (Basic Rest-Activity Cycle; BRAC) hours. Findings The 24-, 12-, and 8-hour components of HRV persisted during long-term spaceflight. The 90-min amplitude became about three times larger in space (ISS03) than on Earth, notably in a subgroup of 7 astronauts who presented with a different HRV profile before flight. The total spectral power (TF; p 〈 0.05) and that in the ultra-low frequency range (ULF, 0.0001–0.003 Hz; p 〈 0.01) increased from 154.9 ± 105.0 and 117.9 ± 57.5 msec 2 (before flight) to 532.7 ± 301.3 and 442.4 ± 202.9 msec 2 (ISS03), respectively. The power-law fractal scaling β was altered in space, changing from -1.087 ± 0.130 (before flight) to -0.977 ± 0.098 (ISS01), -0.910 ± 0.130 (ISS02), and -0.924 ± 0.095 (ISS03) (invariably p 〈 0.05). Interpretation Most HRV changes observed in space relate to a frequency window centered around one cycle in about 90 min. Since the BRAC component is amplified in space for only specific HRV endpoints, it is likely to represent a physiologic response rather than an artifact from the International Space Station (ISS) orbit. If so, it may offer a way to help adaptation to microgravity during long-duration spaceflight.

The melodies so generated are derived from the sequence, and by inference, of the fold, in ways that give each fold a sound representation that may facilitate analysis, recognition, or comparison with other sequences.

Persinger Structural imaging tools can be used to identify neuropathology in post-mortem tissue whereas functional imaging tools including quantitative electroencephalography (QEEG) are thought to be restricted for use in living subjects. We are not aware of any study which has used electrophysiological methods decades after death to infer pathology. We therefore attempted to discriminate between chemically preserved brains which had incurred electrical seizures and those that did not using functional imaging. Our data indicate that modified QEEG technology involving needle electrodes embedded within chemically fixed neural tissue can be used to discriminate pathology. Forty (n = 40) rat brains preserved in ethanol-formalin-acetic acid (EFA) were probed by needle electrodes inserted into the dorsal and ventral components of the left and right cerebral hemispheres. Raw microvolt potentials were converted to spectral power densities within classical electroencephalographic frequency bands (1.5 Hz to 40 Hz). Brain mass differences were shown to scale with left hemispheric ventral theta-band spectral power densities in lithium-pilocarpine seized rats. This relationship was not observed in non-seized rats. A conspicuous absence of pathological indicators within dorsal regions as inferred by microvolt fluctuations was expected given the known localization of post-ictal damage in lithium-pilocarpine seized rats. Together, the data demonstrate that post-mortem neuroimaging is both possible and potentially useful as a means to identify neuropathology without structural imaging techniques or dissection.

The thermogravimetric data for the cake decomposition at six different heating rates (5, 10, 15, 20, 30 and 40 °C/min) were used to evaluate the kinetic decomposition of the cake using Friedman (FD), Kissinger-Akahira-Sunose (KAS) and Flynn-Wall-Ozawa (FWO) models. Thermal decomposition of the cake showed thermograms indicating dehydration and devolitilization stages (200–400 °C). The maximum temperature for the decomposition of the cake (T max ) increases from 289.42–335.96 °C with increase in heating rates. The average apparent activation energy (Ea) values of 153.15, 145.14 and 147.15 kJ/mol were calculated using Friedman, Kissinger-Akahira-Sunose, and Flynn-Wall-Ozawa models respectively. The extent of mass conversion (α) shows dependence on apparent activation Ea values which is an evidence of multi-step decomposition kinetic. The thermal profile and kinetic data obtained could be helpful in evaluating the thermal stability of the cake as well as modeling, designing and developing a thermo-chemical system for the conversion of the cake to fuel.

Hawbaker The wildland-urban interface (WUI), the area where human development encroaches on undeveloped land, is expanding throughout the western United States resulting in increased wildfire risk to homes and communities. Although census based mapping efforts have provided insights into the pattern of development and expansion of the WUI at regional and national scales, these approaches do not provide sufficient detail for fine-scale fire and emergency management planning, which requires maps of individual building locations. Although fine-scale maps of the WUI have been developed, they are often limited in their spatial extent, have unknown accuracies and biases, and are costly to update over time. In this paper we assess a semi-automated Object Based Image Analysis (OBIA) approach that utilizes 4-band multispectral National Aerial Image Program (NAIP) imagery for the detection of individual buildings within the WUI. We evaluate this approach by comparing the accuracy and overall quality of extracted buildings to a building footprint control dataset. In addition, we assessed the effects of buffer distance, topographic conditions, and building characteristics on the accuracy and quality of building extraction. The overall accuracy and quality of our approach was positively related to buffer distance, with accuracies ranging from 50 to 95% for buffer distances from 0 to 100 m. Our results also indicate that building detection was sensitive to building size, with smaller outbuildings (footprints less than 75 m 2 ) having detection rates below 80% and larger residential buildings having detection rates above 90%. These findings demonstrate that this approach can successfully identify buildings in the WUI in diverse landscapes while achieving high accuracies at buffer distances appropriate for most fire management applications while overcoming cost and time constraints associated with traditional approaches. This study is unique in that it evaluates the ability of an OBIA approach to extract highly detailed data on building locations in a WUI setting.

Here, we synthesise magnetic alloy-filling carbon nanoparticles (MA@C NPs) via the following two-step procedure; (1) Irradiation of a laser beam of 266 nm wavelength into super-critical benzene, in which both ferrocene and cobaltocene are dissolved, at 290 °C; and (2) annealing of the particles at 600 and 800 °C. We find that the core particles are composed of cobalt (Co), iron (Fe) and oxygen (O) and covered with carbon layers. The structure of the core particles as-synthesised, and annealed at 600 and 800 °C, is, respectively, amorphous, CoFe 2 O 4 and FeCo. We also investigate the viability of L929 cells in the presence of MA@C NPs and find that there is no serious advert effect of the MA@C NPs on the cell viability thanks to the carbon layers covering the core particles. The magnetic properties are well characterised. The saturation and remnant magnetisation and coercivity increase and as a result, the hyperthermic efficiency becomes higher with an increase in the annealing temperature. The further modification of the surface of the present particles with several functional molecules becomes easier due to the carbon layers, which makes the present particles more valuable. It is therefore supposed that the presently synthesised MA@C NPs may well be utilised for nanotechnology-based biomedical engineering; e.g., nano bioimaging, nano hyperthermia and nano surgery.

Muller, Arianna Muller The nature of emerging patterns concerning water quality stressors and the evolution of hypoxia within sub-estuaries of the Chesapeake Bay has been an important unresolved question among the Chesapeake Bay community. Elucidation of the nature of hypoxia in the tributaries of the Chesapeake Bay has important ramifications to the successful restoration of the Bay, since much of Bay states population lives within the watersheds of the tributaries. Very little to date, is known about the small sub-estuaries of the Chesapeake Bay due to limited resources and the difficulties in resolving both space and time dimensions on scales that are adequate to resolve this question. We resolve the spatio-temporal domain dilemma by setting up an intense monitoring program of water quality stressors in the Severn and South Rivers, MD. Volume rendered models were constructed to allow for a visual dissection of the water quality times series which illustrates the life cycle of hypoxia and anoxia at the mid to upper portions of the tidal tributaries. The model also shows that unlike their larger Virginian tributary counterparts, there is little to no evidence of severe hypoxic water intrusions from the main-stem of the Chesapeake Bay into these sub-estuaries.

This framework encompasses linear reversible and irreversible processes with thermal, electrical, magnetic, and/or mechanical effects coupled. The comprehensive framework combines the principles of classical equilibrium and non-equilibrium thermodynamics with electrodynamics of continua in the infinitesimal strain regime. In the first part of this paper, linear Thermo-Electro-Magneto-Mechanical (TEMM) quasistatic processes are characterized. Thermodynamic stability conditions are further imposed on the linear constitutive model and restrictions on the corresponding material constants are derived. The framework is then extended to irreversible transport phenomena including thermoelectric, thermomagnetic and the state-of-the-art spintronic and spin caloritronic effects. Using Onsager's reciprocity relationships and the dissipation inequality, restrictions on the kinetic coefficients corresponding to charge, heat and spin transport processes are derived. All the constitutive models are accompanied by multiphysics interaction diagrams that highlight the various processes that can be characterized using this framework.

G2P[4] is one of the most common genotypes among human rotavirus (HRV) strains, and has been persistently prevalent in South Asia including Bangladesh. In the present study, whole genome sequences of a total of 16 G2P[4] HRV strains (8 strains each in 2010 and 2013) detected in Mymensingh, north-central Bangladesh were determined. These strains had typical DS-1-like genotype constellation. Most of gene segments from DS-1 genogroup exhibited high level sequence identities to each other (>98%), while slight diversity was observed for VP1, VP3, and NSP4 genes. By phylogenetic analysis, individual RNA segments were classified into one (V) or two-three lineages (V–VI or V–VII). In terms of lineages (sublineages) of 11 gene segments, the 16 Bangladeshi strains could be further classified into four clades (A-D) containing 8 lineage constellations, revealing the presence of three clades (A-C) with three lineage constellations in 2010, and a single clade (D) with four constellations in 2013. Therefore, co-existence of multiple G2P[4] HRV strains with different lineage constellations, and change in clades for the study period were demonstrated. Although amino acids in the antigenic regions on VP7 and VP4 were mostly identical to those of global G2P[4] strains after 2000, VP4 of clade D RVAs in 2013 had alanine and proline at positions 88 and 114, respectively, which are novel substitutions compared with recent global G2P[4] strains. Replacement of lineage constellations associated with unique amino acid changes in the antigenic region in VP4 suggested continuous genetic evolutionary state for emerging new G2P[4] rotavirus strains in Bangladesh.

Adams, Nils Welsh Objectives Insulin expression is highly controlled on the posttranscriptional level. The RNA binding proteins (RBPs) responsible for this result are still largely unknown. Methods and results To identify RBPs that bind to insulin mRNA we performed mass spectrometry analysis on proteins that bound synthetic oligonucloetides mimicing the 5′- and the 3′-untranslated regions (UTRs) of rat and human insulin mRNA in vitro . We observed that the RBPs heterogeneous nuclear ribonucleoprotein (hnRNP) U, polypyrimidine tract binding protein (PTB), hnRNP L and T-cell restricted intracellular antigen 1-related protein (TIA-1-related protein; TIAR) bind to insulin mRNA sequences, and that the in vitro binding affinity of these RBPs changed when INS-1 cells were exposed to glucose, 3-isobutyl-1-methylxanthine (IBMX) or nitric oxide. High glucose exposure resulted in a modest increase in PTB and TIAR binding to an insulin mRNA sequence. The inducer of nitrosative stress DETAnonoate increased markedly hnRNP U and TIAR mRNA binding. An increased PTB to TIAR binding ratio in vitro correlated with higher insulin mRNA levels and insulin biosynthesis rates in INS-1 cells. To further investigate the importance of RNA-binding proteins for insulin mRNA stability, we decreased INS-1 and EndoC-βH1 cell levels of PTB and TIAR by RNAi. In both cell lines, decreased levels of PTB resulted in lowered insulin mRNA levels while decreased levels of TIAR resulted in increased insulin mRNA levels. Thapsigargin-induced stress granule formation was associated with a redistribution of TIAR from the cytosol to stress granules. Conclusions These experiments indicate that alterations in insulin mRNA stability and translation correlate with differential RBP binding. We propose that the balance between PTB on one hand and TIAR on the other participates in the control of insulin mRNA stability and utilization for insulin biosynthesis.

This article reports improved B-factor prediction achieved by sampling the atomic positional fluctuations in multiple picosecond molecular dynamics simulations that use uniformly increased atomic masses by 100-fold to increase time resolution. Using the third immunoglobulin-binding domain of protein G, bovine pancreatic trypsin inhibitor, ubiquitin, and lysozyme as model systems, the B-factor root mean square deviations (mean ± standard error) of these proteins were 3.1 ± 0.2–9 ± 1 Å 2 for Cα and 7.3 ± 0.9–9.6 ± 0.2 Å 2 for Cγ, when the sampling was done for each of these proteins over 20 distinct, independent, and 50-picosecond high-mass molecular dynamics simulations with AMBER forcefield FF12MC or FF14SB. These results suggest that sampling the atomic positional fluctuations in multiple picosecond high-mass molecular dynamics simulations may be conducive to a priori prediction of crystallographic B-factors of a folded globular protein.

Concerns regarding the toxic fly ashes can be solved by transforming them into resource as recycled materials. The commitment to recycle is driven by the need to conserve natural resources, reduce imports of raw materials, save landfill space and reduce pollution. A new method to stabilize fly ash from Municipal Solid Waste Incinerator (MSWI) at room temperature has been developed thanks to COSMOS-RICE LIFE+ project ( www.cosmos-rice.csmt.eu ). This process is based on a chemical reaction that occurs properly mixing three waste fly ashes with rice husk ash, an agricultural by-product. COSMOS inert can replace critical raw materials (i.e. silica, fluorspar, clays, bentonite, antimony and alumina) as filler. Moreover the materials employed in the stabilization procedure may be not available in all areas. This paper investigates the possibility of substituting silica fume with corresponding condensed silica fume and to substitute flue-gas desulfurization (FGD) residues with low-cost calcium hydroxide powder. The removal of coal fly ash was also considered. The results will be presented and a possible substitution of the materials to stabilize fly ash will be discussed.

However, it is unknown whether variation in mood affects glucose metabolism. The aim of this study was to assess whether changes in affect were related to fasting plasma glucose and glycated haemoglobin. Methods 379 men and 441 women who took part in the 2003 Health Survey for England and had valid data for GHQ12 and fasting blood glucose were included. Mood variability was assessed by the General Health Questionnaire 12 (GHQ12). Fasting plasma glucose and glycated haemoglobin (HbA 1c ) were measured by standard laboratory methodology and their relationship to variability assessed using linear regression. Results There was a significant inverse relationship between fasting blood glucose, but not HbA 1c , and variability score (R 2 = 0.327, p = 0.02) after adjusting for sociodemographic factors, anthropometric measurements, lifestyle, and use of medication. Conclusion This study has shown an inverse association between changes in affect and fasting plasma glucose. This unexpected finding suggests that the association between affect and glucose is more complex than previously thought. Fasting blood glucose may reflect the operation of homeostatic mechanisms that are disturbed in certain mental states and are associated, therefore, with altered risk of diabetes and related metabolic conditions. This may have implications for the management of those with such conditions and with mental disorders.

These residues were obtained from a pilot plant in Spain that produces biofuel, the biochar represented 10–15% in weight. It was observed that the highest activation was achieved at a temperature of 800 °C, the specific surface increased with time but, on the contrary, high loss of matter was observed. At 600 °C, although there was an important increase of the specific surface and the volume of micropores, at this temperature it was observed that the activation time was not an influential parameter. Finally, at 400 °C it was observed that the activation process was not very significant. Assessing the average pore diameter it was found that the lowest value corresponded to the activation temperature of 600 °C, which indicated the development of microporosity. When the activation temperature increases up to 800 °C the pore diameter increased developing mesoporosity.

Al-Faraj This study aims to assess the potential of trees for integration in urban development by evaluating the damage caused by trees in relation to various tree characteristics. Tree damage to permeable pavement systems and other urban structures such as impermeable pavements, kerbs, roads, retaining walls, footpaths, walls and buildings were assessed to identify the most suitable trees for the urban environment. One hundred square sites of 100 m × 100 m were randomly selected in Greater Manchester for this representative example case study to demonstrate the assessment methodology. Among tree species in this study, Acer platanoides L. (Norway maple) occurred most frequently (17%); others were Tilia spp. L. (Lime; 16%), Fraxinus excelsior L. (common ash; 12%), Acer pseudoplatanus L. (sycamore; 10%) and Prunus avium L. (wild cherry; 8%). The study concludes that 44% of the damage was to impermeable pavements and 22% to permeable pavements. Other damage to structures included kerbs (19%), retaining walls (5%), footpaths (4%), roads (3%) and walls (3%). Concerning the severity of damage, 66% were moderate, 21% light and 19% severe. Aesculus hippocastanum L. (horse chestnut) caused the greatest damage (59%) expressed in percentage as a ratio of the tree number related to damage over the corresponding tree number that was found close to structures.

To study this suppression activity, anacardic acids, cardanols and cardols having different numbers of double bonds in alk(en)yl chains were prepared. The O 2 - scavenging activity and H 2 O 2 formation from O 2 - using PMS-NADH were examined. Anacardic acids and cardols indicated sigmoidal O 2 - scavenging activity but cardanols did not. The O 2 - scavenging activity of anacardic acid C 15:3 was weaker than the suppression activity using xanthine oxidase, but the scavenging activity of cardol C 15:3 was quite similar to the suppression using xanthine oxidase. The H 2 O 2 formation from O 2 - decreased by the addition of anacardic acids, cardanols and cardols but increased by the addition of gallic and caffeic acids. From these results, we deduced that the O 2 - suppression activity of xanthine oxidase reaction with cardols is the O 2 - scavenging activity and that anacardic acids and cardols are O 2 - scavengers having low prooxidant property.

A total of 440,738 sequences from bacteria with complete taxonomic classifications were downloaded from the high quality ribosomal RNA database SILVA. Amplicons produced (86,371 sequences; 1450 bp) by virtual PCR with primers covering the V1–V9 region of the 16S-rRNA gene were used as reference. Virtual PCŔs of internal fragments V3–V4, V4–V5 and V3–V5 were performed. A total of 81,523, 82,334 and 82,998 amplicons were obtained for regions V3–V4, V4–V5 and V3–V5 respectively. Differences in depth of taxonomic classification were detected among the internal fragments. For instance, sensitivity and specificity of sequences classified up to subspecies level were higher when the largest internal fraction (V3–V5) was used (54.0 and 74.6% respectively), compared to V3–V4 (45.1 and 66.7%) and V4–V5 (41.8 and 64.6%) fragments. Similar pattern was detected for sequences classified up to more superficial taxonomic categories (i.e. family, order, class…). Results also demonstrate that internal fragments lost specificity and some could be misclassified at the deepest taxonomic levels (i.e. species or subspecies). It is concluded that the larger V3–V5 fragment could be considered for massive high throughput sequencing reducing the loss of sensitivity and sensibility.

Here, we show that γ-tubulin is located throughout the chromatin of demembranated Xenopus laevis sperm and, as the nucleus is formed, γ-tubulin recruits lamin B3 and nuclear membranes. Immunodepletion of γ-tubulin impairs X. laevis assembly of both the lamina and the nuclear membrane. During nuclear formation in mammalian cell lines, γ-tubulin establishes a cellular protein boundary around chromatin that coordinates nuclear assembly of the daughter nuclei. Furthermore, expression of a γ-tubulin mutant that lacks the DNA-binding domain forms chromatin-empty nuclear like structures and demonstrate that a constant interplay between the chromatin-associated and the cytosolic pools of γ-tubulin is required and, when the balance between pools is impaired, aberrant nuclei are formed. We therefore propose that the nuclear protein meshwork formed by γ-tubulin around chromatin coordinates nuclear formation in eukaryotic cells.

Karl An ordinary least squares (OLS) analysis of the relationship between chlorophyll a (chl a ) concentrations and photosynthetic rates at depths of 5 and 25 m at Station ALOHA produced a slope that was only 28% of the mean productivity index at those depths and an intercept at zero chl a that equaled 70% of the mean photosynthetic rate. OLS regression lines are known to produce a slope and intercept that are biased estimates of the true slope and intercept when the explanatory variable, X , is uncontrolled, but in this case the measurement errors and natural variability of the chl a concentrations were much too small to explain the apparent bias. The bias was traceable to the fact that the photosynthetic rates were determined by more than one explanatory variable, a source of variability that is typically overlooked in discussions of OLS bias. Modeling the photosynthetic rates as a function of the product of chl a and surface irradiance produced a much more accurate and realistic description of the data, but the OLS continued to be biased, presumably because the photosynthetic rates were functions of factors in addition to chl a and surface irradiance (e.g., temperature, macronutrients, trace metals, and vitamins). The results underscore the need to recognize that the absence of bias in an OLS when X is not controlled implies that all scatter in the data about the OLS is due to errors in the dependent variable, an unlikely scenario. In most cases, resolution of the bias problem will require identification of the explanatory variables in addition to X that determine the dependent variable.

Its etiology is multifactorial, arising from predisposing environmental stress conditions as well as the action of several different respiratory pathogens. This situation has hindered the development of effective control strategies. Although different type of vaccines are available, many currently marketed vaccines are based on inactivated cultures of the main viral and bacterial agents involved in this pathology. The molecular composition of commercial veterinary vaccines is a critical issue. The present work aims to define at the proteomic level the most relevant valence of a line of commercial respiratory vaccines widely used in Central and South America. Since Mannheimia haemolytica is responsible for most of the disease associated morbid-mortality, we focused on the main proteins secreted by this pathogen, in particular Leukotoxin A, its main virulence factor. By Western blot analysis and mass spectrometry, Leukotoxin A was identified as a major component of M. haemolytica culture supernatants. We also identified other ten M. haemolytica proteins, including outer membrane proteins, periplasmic transmembrane solute transporters and iron binding proteins, which are relevant to achieve protective immunity against the pathogen. This work allowed a detailed molecular characterization of this vaccine component, providing evidence of its quality and efficacy. Furthermore, our results contributed to the identification of several proteins of interest as subunit vaccine candidates.

Patel Background and purpose Clinical history is known to influence interpretation of a wide range of radiologic examinations. We sought to evaluate the influence of the clinical history on MRI interpretation of optic neuropathy. Materials and methods 107 consecutive orbital MRI scans were retrospectively reviewed by three neuroradiologists. The readers independently evaluated the coronal STIR sequence for optic nerve hyperintensity and/or atrophy (yes/no) and the coronal post-contrast T1WI for optic nerve enhancement (yes/no). Readers initially evaluated the cases blinded to the clinical history. Following a two week washout period, readers again evaluated the cases with the clinical history provided. Inter-reader and reader-clinical radiologist agreement was assessed using Cohen's simple kappa coefficient. Results Intra-reader agreement, without and with provision of clinical history, was 0.564–0.716 on STIR and 0.270–0.495 on post-contrast T1WI. Inter-reader agreement was overall fair-moderate. On post-contrast T1WI, inter-reader agreement was significantly higher when the clinical history was provided (p = 0.001). Reader-clinical radiologist agreement improved with provision of the clinical history to the readers on both the STIR and post-contrast T1WI sequences. Conclusions In the MRI assessment of optic neuropathy, only modest levels of inter-reader agreement were achieved, even after provision of clinical history. Provision of clinical history improved inter-reader agreement, especially when assessing for optic nerve enhancement. These findings confirm the subjective nature of orbital MRI interpretation in cases of optic neuropathy, and point to the importance of an accurate clinical history. Of note, the accuracy of orbital MRI in the context of optic neuropathy was not assessed, and would require further investigation.

Martínez-Avila Stability of the total phenolic content, ascorbic acid, total carotenoids and antioxidant activity in eight fruit beverages was analyzed. The influence of storage temperature (4, 8 and 11 °C) during the product shelf-life (20 days) was evaluated. Pomegranate Juice presented the highest values for antioxidant activity by DPPH assay (552.93 ± 6.00 GAE μg mL −1 ), total carotenoids (3.18 ± 0.11 βCE μg mL −1 ), and total phenolic content (3967.07 ± 2.47 GAE μg mL −1 ); while Splash Blend recorded the highest levels of ascorbic acid (607.39 ± 2.13 AAE μg mL −1 ). The antioxidant capacity was stable at 4 and 8 °C for the first 8 days of storage; while carotenoids and ascorbic acid were slightly degraded through the storage time, possibly due to oxidation and/or reactions with other compounds. The results suggest that the observed variation during testing could be related to storage conditions of the final product.

This pathway is called antigen cross-presentation and plays an important role in the stimulation of naïve CD8 + T cells for infectious and tumor immunity. Our previous studies in DC2.4 cells and bone marrow-derived DCs revealed that exogenously added ovalbumin (OVA) is processed through endoplasmic reticulum (ER)-associated degradation (ERAD) for cross-presentation. In this study, we aimed to further confirm these results by purification of the subcellular compartment in which exogenous antigens undergo ERAD from homogenates of DC2.4 cells pretreated with biotinylated OVA (bOVA). bOVA-containing vesicles were purified using streptavidin (SA)-magnetic beads from cell homogenates and were found to contain ER chaperones and ERAD components together with proteins for antigen presentation. In purified microsomes, bOVA was retained in membranous fractions and degraded by the ubiquitin proteasome system in presence reticulocyte lysates and ATP. These results strongly suggested that DCs processed and degraded exogenous antigens through ERAD for cross-presentation in this purified subcellular compartment.

Wildtype (wt) RON has been shown to play a role in metastasis of epithelial malignancies. It presents an important potential therapeutic target for colorectal, breast, gastric and pancreatic cancer. Little is known about functional differences amongst RON isoforms RON155, RON160 and RON165. The purpose of this study was to determine the effect of various RON kinase isoforms on cell motility. Methods Cell lines with stable expression of wtRON were generated by inserting the coding region of RON in pTagRFP (tagged red fluorescence protein plasmid). The expression constructs of RON variants (RON155, RON160 and RON165) were generated by creating a mutagenesis-based wtRON-pTag RFP plasmid and stably transfected into HEK 293 cells. The wound closure scratch assay was used to investigate the effect on cell migratory capacity of wild type RON and its variants. Results RON transfected cells demonstrated increased cell motility compared to HEK293 control cells. RON165 cell motility was significantly increased compared to RON160 (mean percentage of wound covered 37.37% vs. 32.40%; p = 0.03). Conclusions RON tyrosine kinase isoforms have variable cell motility. This may reflect a difference in the behavior of malignant epithelial cells and their capacity for metastasis.

Despite a variety of anti-smoking products available to aid with tobacco cessation, the majority of people who attempt to quit smoking relapse within 6 months due to the addictive nature of nicotine. An immunotherapy approach could offer a promising treatment option by inducing a potent selective antibody response against nicotine in order to block its distribution to the brain and its addictive effects in the central nervous system. Our nicotine vaccine candidate was administered intranasally using the Neisseria meningitidis serogroup B Adjuvant Finlay Proteoliposome 1 (AFPL1) as a part of the delivery system. This system was designed to generate a robust immune response by stimulating IL-1β production through Toll-like receptor 4 (TLR4), a potent mechanism for mucosal immunity. The vaccine induced high antibody titers in mice sera in addition to inducing mucosal antibodies. The efficacy of our vaccine was demonstrated using in vivo challenge experiments with radioactive [ 3 H]-nicotine, followed by an analysis of nicotine distribution in the lung, liver, blood and brain. Our results were encouraging as the nicotine concentration in the brain tissue of mice vaccinated with our candidate vaccine was four times lower than in non-vaccinated controls; suggesting that the anti-nicotine antibodies were able to block nicotine from crossing the blood brain barrier. In summary, we have developed a novel nicotine vaccine for the treatment of tobacco addiction by intranasal administration and also demonstrated that the AFPL1 can be used as a potential adjuvant for this vaccine design.

The present study was carried out to quantify N transfer from legumes ( Lupinus albus L., Trifolium incarnatum L. or Vicia sativa L.) to rapeseed ( Brassica napus L.) using the split-root 15 N-labelling method. After three months we observed that legumes did not alter the growth of rapeseed. Vetch showed the lowest growth and demonstrated low 15 N shoot to root translocation and no significant N transfer to rapeseed. In contrast, significant 15 N enrichment was found in lupine and clover and 15 N was transferred to the associated rapeseed plants (around 6 and 4 mg N plant −1 , respectively), which contributed 2 to 3% of the rapeseed total N. Additionally, the data revealed that N 2 fixation dominated the N nutrition in lupine despite the high N level provided in the donor compartment, suggesting a greater niche segregation between companion plants. Based on the results of this study we suggest that intercropping can be a relevant contributor to rapeseed N nutrition. Among the three legumes tested, clover and lupine seemed to be the best intercropping candidates.

The bottom wall of the cavity is heated isothermally, the left inclined wall is non-isothermal and the right inclined wall is considered to be cold. The concentration is higher at bottom wall, lower at right inclined wall and non-isoconcentration at left inclined wall of the cavity. The governing equations are transformed to the dimensionless form and solved numerically using Galerkin weighted residual technique of finite element method. The results are obtained in terms of streamlines, isotherms, isoconcentrations, average Nueeslt number ( Nu ) and average Sherwood number ( Sh ) for the parameters thermal Rayleigh number ( Ra T ), dimensionless heat generation parameter ( λ ), solid volume fraction ( ϕ ) and Lewis number ( Le ) while Prandtl number ( Pr ), Buoyancy ratio ( N ) and Darcy number ( Da ) are considered to be fixed. It is observed that flow pattern, temperature fields and concentration fields are affected by the variation of above considered parameters.

Dimitrijević-Branković This study has explored the feasibility of using spent coffee grounds as a good supporting material for the Paenibacillus chitinolyticus CKS1 cellulase immobilization. An optimal operational conditions in a batch-adsorption system were found to be: carrier mass of 12 g/L, under the temperature of 45 °C and no pH adjustments. The immobilization yield reached about 71%. An equilibrium establishment between the cellulase and the carrier surface occurred within 45 min, whereas the process kinetics may be predicted by the pseudo-second-order model. An immobilized cellulase preparation expressed very good avicelase activity, this reached up to 2.67 U/g, and revealed an improved storage stability property, compared to free enzyme sample counterpart. The addition of metal ions, such as K + and Mg 2+ did not affect positively immobilization yield results, but on the contrary, contributed to an improved bio-activities of the immobilized cellulase, thus may be employed before each enzyme application. The method developed in this study offers a cheap and effective alternative for immediate enzyme isolation from the production medium and its stabilization, compared to other carriers used for the immobilization.

Manjunath, Haoquan Wu 5′-isomiRs expand the repertoire of miRNA targets. However, how they are generated is not well understood. Previously, we showed that for some miRNAs in mammalian cells, Drosha cleaves at multiple sites to generate multiple pre-miRNAs that give rise to multiple 5′-isomiRs. Here, we showed that for some other miRNAs, 5′-isomiRs are generated by alternative Dicer processing. In addition, we showed that in miR-203, alternative Dicer processing is regulated by a conserved sliding-bulge structure at the Dicer processing site, which allows the pre-miRNA molecule to fold into two different structures that are processed differently by Dicer. So far no RNA motif that slides to change conformation and alter a protein–RNA interaction has been reported. Thus, our study revealed a novel RNA motif that regulates 5′-isomiR generation in some miRNAs. It might also contribute to regulating protein–RNA interactions in other biological processes, since it takes only one point mutation to generate the sliding bulge, and there are a large number of different RNAs in the cell.

Microscopic fluorescence lifetime measurement can provide information about localized biochemical conditions such as pH and the concentrations of oxygen and calcium. We hypothesized that combining these techniques could assist accurate cancer discrimination by providing both biochemical and morphological information. We designed a dual-mode experimental setup for confocal endomicroscopic imaging and fluorescence lifetime measurement and applied it to a mouse xenograft model of activated human pancreatic cancer generated by subcutaneous injection of AsPC-1 tumor cells. Using this method with pH-sensitive sodium fluorescein injection, we demonstrated discrimination between normal and cancerous tissues in a living mouse. With further development, this method may be useful for clinical cancer detection.

The resultant concentrations of nitrate and nitrite for both current and future land-use scenarios are all below the daily limit suggested by the WHO, (World Health Organization). For the phosphate concentration, an increase of 4.21% was depicted under cassava land-use scenario. The results show that SWAT is a reliable water quality model, capable of simulating accurate information for developing environmental management plans.

In the absence of topology changes, both types of models self-organize to a critical state. Species extinctions in the replicator system degenerates the self-organization to a random walk, as does vanishing of species interaction for the BS-model. A replicator model with speciation is introduced, experiencing dramatic topology changes. It produces a variety of features, but self-organizes to a possibly critical state only in a few special cases. Speciation-extinction dynamics interfering with self-organization, biological macroevolution probably is not a self-organized critical system.

Environmental variables used in modeling consisted of 42 environmental and climate variables for Nayband wildlife refuge and 36 environmental and climate variables for Petregan protected area. Also, seasonal overlap area were obtained using the ENM TOOLS software. The results showed that the most important factors affecting habitat suitability of the Asian Houbara in all seasons included the ratio of distance to hill, the type of Artemisia−Gymnocarpus, distance to the slope (8–12%) in the Nayband wildlife refuge, distance to the type of Artemisia aucheri, distance to the Land Passion, and distance to the dry land farming in the Petregan region. In summer, the most suitable habitat is Nayband but is Petergan during fall-winter. there is maximum overlap in summer, and the least overlap in the spring these areas. The results of this study can be used as a valuable tool in implementing conservation and management strategies, in order to increase desirable habitats in the eastern part of Iran.

However, exploring the structure of the multiplex network time series is still an open problem. Here we propose a two-step strategy to tackle this problem based on the concept of distance (metric) between networks. Given a multiplex graph, first a network of networks is built for each time step, and then a real valued time series is obtained by the sequence of (simple) networks by evaluating the distance from the first element of the series. The effectiveness of this approach in detecting the occurring changes along the original time series is shown on a synthetic example first, and then on the Gulf dataset of political events.

Here we report that through a chemical screen, we identified two novel and structurally similar small molecules BIR1 and BIR2 which robustly induced the self-organization of a balloon-shaped three-dimensional structure when applied to two-dimensional adherent hESC cultures in the absence of growth factors. Gene expression analyses and functional assays demonstrated an endothelial identity of this balloon-like structure, while cell surface marker analyses revealed a VE-cadherin + CD31 + CD34 + KDR + CD43 − putative endothelial progenitor population. Furthermore, molecular marker labeling and morphological examinations characterized several other distinct DiI-Ac-LDL + multi-cellular modules and a VEGFR3 + sprouting structure in the balloon cultures that likely represented intermediate structures of balloon-formation.

The aim of this study was to investigate whether CS/DS rich in GalNAc(4,6SO 4 ) residues participate in the progression or resolution of liver fibrosis. Methods Wild type (WT) and GalNAc4S-6ST KO mice were treated with CCl 4 for 5 weeks. After discontinuation of CCl 4 administration, histochemical and biochemical changes and expression of genes related to matrix components were compared between WT and GalNAc4S-6ST KO mice. Results and conclusion On 2 days after cessation of CCl 4 administration, higher fibrosis was observed in KO mice than in WT mice by Sirius Red staining. Serum alanine aminotransferase activity was higher in KO mice than in WT mice. Hydroxyproline contents and Sirius Red staining showed that repair of liver fibrosis in the recovery stages appeared to be delayed in KO mice. Expression of mRNA of matrix metalloproteinase (MMP)-2, MMP-13 and versican peaked at 2 days after cessation of CCl 4 administration and was higher in KO mice than in WT mice. Expression of MMP-9 in the recovery stage was lower in KO mice than in WT mice. Our findings demonstrate that defect in GalNAc4S-6ST, which resulted in disappearance of CS/DS containing GalNAc(4,6SO 4 ), appear to contribute to progression of liver fibrosis, delayed recovery from fibrosis, and various changes in the expression of proteoglycans and MMPs in carbon tetrachloride–treated mice.

    Description: Publication date: August 2016 Source: Heliyon, Volume 2, Issue 8 Author(s): Ichiro Fujihara, Mitsuru Furusawa It has been shown that the disparity model, in which mutations are introduced exclusively in the lagging strand, has a great advantage in the promotion of evolution, compared with the conventional parity mutagenesis model. To understand much more about the characteristic of the disparity model, a novel 2-D genetic algorithm (GA) which reflected gene interactions was developed. The GA consisting of the lattice model showed powerful abilities to evolve. Even under conditions of high mutation rates with an extra genetic load, the GA could quickly evolve and finally attain a long stable state with high fitness scores. Arbitrary interruption of the stable state of evolution by various lengths of environmental stress could produce new individuals with different genotypes within relatively short periods and the mutants finally occupy the population; that brought back the picture of “punctuated equilibrium”. Interestingly, this phenomenon could be reproduced with certainty without changing mutation rates at all. As long as the fidelity difference between the lagging and leading strand was kept high enough, the robustness of the disparity model was very high. The acceleration or slowdown of evolution can be unambiguously introduced only by environmental changes, and the seesawing mutation rate is not the necessary condition for changing the speed of evolution.

IgA1 was observed in the cytoplasm and the plasma membrane. A correlation was found between intra-tumour IgA1 and poor overall survival in a large cohort of bladder cancer patients (n = 99, p = 0.011, log-rank test). The number of IgA1-positive tumour cells was also found to be higher in female than male bladder cancer patients. The presence of IgA1 was confirmed in formalin-fixed paraffin-embedded ovarian carcinoma samples using LC-MS/MS analysis. Uptake of IgA1 was also observed in breast cancer and melanoma cell lines when cultivated in the presence of serum from healthy individuals, indicating a possible origin of the IgA1 antibodies in cancer cells.

We previously reported that LATS1/2 localize to the mitotic apparatus, where they are involved in the phosphorylation and activation of the mitotic kinase Aurora-B; however, the detailed mechanism of LATS1/2 action remains obscure. The activity of Aurora-B is stringently regulated by formation of the chromosomal passenger complex containing the inner centromere protein (INCENP), which leads to appropriate activation of Aurora-B during mitosis and cytokinesis. In this study, we found that LATS1/2 phosphorylated INCENP at S894 in the Thr-Ser-Ser motif. Moreover, the LATS-mediated phosphorylation of S894 was necessary and sufficient for the activation of Aurora-B, which is required for completion of cytokinesis in cells engaged in multipolar division. We propose a novel mechanism for regulation of Aurora-B via INCENP phosphorylation by LATS1/2 during cytokinesis.

Main methods An MTT assay and FACS flow cytometry were performed to assess the cellular viability and apoptosis and cellular ROS levels, respectively. To examine the expression of pro-inflammatory cytokines and cellular signaling pathways, semi-quantitative RT-PCR and Western blotting were also used in this study. Key findings Among the six newly synthesized DMNQ derivatives, 2-cyclohexylamino-5,8-dimethoxy-1,4-naphthoquinone (R6) significantly inhibited the NO production, cellular ROS levels and the cytokines expression in BV-2 microglial cells, which stimulated by LPS. Signaling study showed that compound R6 treatment also significantly down-regulated the LPS-induced phosphorylation of MAPKs (ERK, JNK and p38) and decreased the degradation of IκB-α in BV2 microglial cells. Significance Our findings demonstrate that our newly synthesized compound derived from DMNQ, 2-cyclohexylamino-5,8-dimethoxy-1,4-naphthoquinone (R6), might be a therapeutic agent for the treatment of glia-mediated neuroinflammatory diseases.

Nara, Noriyoshi Tsuchiya, Zhong-Qiang Chen The largest mass extinction of biota in the Earth’s history occurred during the Permian–Triassic transition and included two extinctions, one each at the latest Permian (first phase) and earliest Triassic (second phase). High seawater temperature in the surface water accompanied by euxinic deep-intermediate water, intrusion of the euxinic water to the surface water, a decrease in pH, and hypercapnia have been proposed as direct causes of the marine crisis. For the first-phase extinction, we here add a causal mechanism beginning from massive soil and rock erosion and leading to algal blooms, release of toxic components, asphyxiation, and oxygen-depleted nearshore bottom water that created environmental stress for nearshore marine animals. For the second-phase extinction, we show that a soil and rock erosion/algal bloom event did not occur, but culmination of anoxia–euxinia in intermediate waters did occur, spanning the second-phase extinction. We investigated sedimentary organic molecules, and the results indicated a peak of a massive soil erosion proxy followed by peaks of marine productivity proxy. Anoxic proxies of surface sediments and water occurred in the shallow nearshore sea at the eastern and western margins of the Paleotethys at the first-phase extinction horizon, but not at the second-phase extinction horizon. Our reconstruction of ocean redox structure at low latitudes indicates that a gradual increase in temperature spanning the two extinctions could have induced a gradual change from a well-mixed oxic to a stratified euxinic ocean beginning immediately prior to the first-phase extinction, followed by culmination of anoxia in nearshore surface waters and of anoxia and euxinia in the shallow-intermediate waters at the second-phase extinction over a period of approximately one million years or more. Enhanced global warming, ocean acidification, and hypercapnia could have caused the second-phase extinction approximately 60 kyr after the first-phase extinction. The causes of the first-phase extinction were not only those environmental stresses but also environmental stresses caused by the soil and rock erosion/algal bloom event.

Purushotham, J.Siva Kumar Ion conducting polymer electrolytes composed of poly(vinylidene fluoride- co -hexafluoropropylene) (PVdF-HFP), poly(methyl methacrylate) (PMMA) and lithium triflate (LiTf) were prepared using the solution casting method. Structural change and complex formation in the blend electrolyte systems were confirmed from the X-ray diffraction (XRD), Fourier transform infrared (FTIR) spectroscopy and scanning electron microscopy (SEM) studies. Thermal properties of the samples were investigated by the differential scanning calorimetry (DSC) technique. The ionic conductivity of these polymer electrolytes was studied by impedance spectroscopy at various temperatures ranging from 303–393 K. The results reveal that the ionic conductivity of the polymer blend electrolytes depends on the PVdF-HFP:PMMA composition as well as the temperature. Maximum room temperature conductivity of 7.4 × 10 − 5 S cm −1 was achieved with 22.5 wt.% PMMA. The blending of PVdF-HFP with PMMA improved the thermal stability and ionic conductivity of the polymer electrolyte. Estimated transference numbers suggest the charge transport is predominantly ionic.

Abdalla The aim of the study was to provide values for morphometric parameters of histological components of normally delivered full term placentas of Sudanese women and compare them with reported parameters for other ethnic groups. A total of 200 histological sections, stained with hematoxylin and eosin and trichrome stains were used to give a final sample of 1000 fields saved as PowerPoint images for histomorphometry. A systematic random sampling procedure was adopted to ensure the optimum sample size that keeps the percentage error below 5% for the volume estimates. Standard stereological methods of point-counting and intersection-counting were applied to the microscopic fields to determine the volumes of placental components and surface area of fetal-maternal interface. The morphometric parameters showed no variations either between the placentas or between central and peripheral regions. The placental villi and the intervillus space occupied 65% and 35% of placental volume respectively with mean absolute values of 318 cm 3 and 169 cm 3 . The mean absolute volume of the intervillus space was less than that of other ethnic groups by 8.67% but was significantly larger than that of the fetal capillaries which measured 41.2 cm 3 . The ratio of the absolute volume of the intervillus space to the volume of the fetal blood capillaries was 4:1 in both Sudanese and other ethnic groups. In the placental villi the fetal connective tissue together with the contained blood vessels larger than capillaries occupied 88% of the villus volume. The mean surface area of the fetal-maternal interface of the placental villi (syncytiotrophoblast) was 12.59 M 2 .

Sotelo Anchovies have been traditionally captured and processed for human consumption for millennia. In the case of Spain, ripened and salted anchovies are a delicacy, which, in some cases, can reach high commercial values. Although there have been a number of studies presenting DNA methodologies for the identification of anchovies, this is one of the first studies investigating the level of mislabelling in this kind of products in Europe. Sixty-three commercial semipreserved anchovy products were collected in different types of food markets in four Spanish cities to check labelling accuracy. Species determination in these commercial products was performed by sequencing two different cyt-b mitochondrial DNA fragments. Results revealed mislabelling levels higher than 15%, what authors consider relatively high considering the importance of the product. The most frequent substitute species was the Argentine anchovy, Engraulis anchoita , which can be interpreted as an economic fraud.

Native small islets are known to have a higher insulin secretion ability in vitro and to provide better transplantation outcomes when compared with large islets. In this study, we prepared microengineered pseudo-islets from dispersed rat islet cells using precisely-fabricated agarose gel-based microwells with different diameters (100, 300, or 500 μm) to investigate the function and survival of islet cell aggregates with well-controlled sizes. We observed that dead cells were rarely present in the small pseudo-islets with an average diameter of ∼60 μm prepared using 100 μm microwells. In contrast, we observed more dead cells in the larger pseudo-islets prepared using 300 and 500 μm microwells. The relative amount of hypoxic cells was significantly low in the small pseudo-islets whereas a hypoxic condition was present in the core region of the larger pseudo-islets. In addition, we found that the small-sized pseudo-islets reconstituted the in vivo -tissue like arrangement of the α and β cells, and restored the high insulin secretory capacity in response to high glucose. These results clearly suggest that precise size control of pseudo-islets is essential for maintaining islet cell function and survival in vitro . The small-sized pseudo-islets may be advantageous for providing a better therapeutic approach for treating type 1 diabetes mellitus via islet reorganization and transplantation.

Elmarakby Background/Aim Hepatic injury is a hallmark adverse reaction to Valproate (VPA), a common used drug in the management of numerous CNS disorders, including epilepsy. DHA has a myriad of health benefits, including renal- and hepato-protective effects. Unfortunately, however, the underpinnings of such liver-pertinent VPA- and DHA-actions remain largely undefined. Accordingly, this study attempted to unveil the cellular and molecular triggers whereby VPA evokes, while DHA abates, hepatotoxicity. Methods We evaluated activity and/or expression of cellular markers of oxidative stress, inflammation, and apoptosis in rat liver, following treatment with VPA (500 mg/kg/day) with and without concurrent treatment with DHA (250 mg/kg/day) for two weeks. Results and conclusion VPA promoted hepatic oxidative stress as evidenced by enhancing activity/expression of NADPH-oxidase and its subunits, a ROS-generator, and by accumulation of lipid-peroxides. Moreover, VPA enhanced hepatic phosphorylation/activation of mitogen-activated protein kinase (MAPK), and expression of cyclooxygenase-2(COX-2), as proinflammatory signals. Besides, VPA promoted hepatocellular apoptosis, as attested by enhanced expression of cleaved caspase-9 and increased number of TUNEL-positive hepatocytes. Lastly, VPA upregulated levels of hypoxia-inducible factor-1-alpha (HIF-1α), a multifaceted modulator of hepatocytic biology, and activity of its downstream antioxidant enzyme heme-oxygenase-1(HO-1). These changes were significantly blunted by co-administration of DHA. Our findings demonstrate that VPA activated NADPH-oxidase and HIF-1α to induce oxidative-stress and hypoxia as initiators of hepatic injury. These changes were further aggravated by up-regulation of inflammatory (MAPK and COX-2) and apoptotic cascades, but could be partly lessened by HO-1 activation. Concurrent administration of DHA mitigated all VPA-induced anomalies.

    Description: Publication date: June 2016 Source: Heliyon, Volume 2, Issue 6 Author(s): Zia Mahmood Siddiqi, Mohammad Saleem, Chanbasha Basheer Water quality in a run-off canal system in an industrial area was evaluated for a range of physical and chemical properties comprising trace metals (including mercury (Hg), chromium (Cr), iron (Fe), manganese (Mn), salinity, pH, turbidity, total dissolved solids, total suspended solids, chemical oxygen demand (COD), and dissolved oxygen). High concentrations of potassium (K) (1.260–2.345 mg/l) and calcium (Ca) (19.170–35510 mg/l) demonstrated that the salinity in the water was high, which indicates that industrial effluents from fertilizer manufacturing and Chlor-alkali units are being discharged into the canal system. Almost all the metal concentrations in water and sediment were within the thresholds established by the local regulatory body. Concentrations of Cr (0.0154–0.0184 mg/l), Mn (0.0608–0.199 mg/l), Fe (0.023–0.035 mg/l), COD (807–916 mg/l), and turbidity (633 ± 15–783 ± 22 NTU) were high where the canal discharges into the Persian Gulf; these discharges may compromise the health of the aquatic ecosystem. There is concern about the levels of Hg in water (0.00135–0.0084 mg/l), suspended sediment (0.00308–0.0096 mg/l), and bed sediment (0.00172–0.00442 mg/l) because of the bio-accumulative nature of Hg. We also compared the total Hg concentrations in fish from Jubail, and two nearby cities. Hg contents were highest in fish tissues from Jubail. This is the first time that heavy metal pollution has been assessed in this water run-off canal system; information about Hg is of particular interest and will form the basis of an Hg database for the area that will be useful for future investigations.

In this article, we illustrated the physical behaviors were obviously influenced by synthetic routes. And their chemical and physical structures were investigated by gel permeation chromatograph (GPC), differential scanning calorimeter (DSC), fourier transform infrared spectrography (FT-IR) and mechanical properties tests. The surface wettability were studied by contact angle measurement (CA). As a kind of short-term implant biomaterial, the results of the hemolysis and platelet adhesive tests were recorded by spectrophotometer and scanning electron microscopy (SEM), indicating the materials have a great potential for developments and applications in biomedical field.

This article presents the outcomes of a recent structural investigation concerning a supply network in the central area of Japan. We investigated the effectiveness of statistical learning theory to express the individual differences of a supply chain of enterprises within a certain business community using social network analysis. In the experiments, we employ support vector machine to train a customer–supplier relationship model on one of the main communities extracted from a supply network in the central area of Japan. The prediction results reveal an F-value of approximately 70% when the model is built by using network-based features, and an F-value of approximately 77% when the model is built by using attribute-based features. When we build the model based on both, F-values are improved to approximately 82%. The results of this research can help to dispel the implicit design space concerning customer–supplier relationships, which can be explored and refined from detailed topological information provided by network structures rather than from traditional and attribute-related enterprise profiles. We also investigate and discuss differences in the predictive accuracy of the model for different sizes of enterprises and types of business communities.

Nicolau Introduction We aimed to describe the in vivo efficacy of meropenem, in addition to cefepime and levofloxacin as comparators against VIM-producing Pseudomonas aeruginosa and compare the findings to our previous observations with Enterobacteriaceae . Methods Eight clinical P. aeruginosa isolates with meropenem MICs from 4 to 512 mg/L were studied in a murine neutropenic thigh infection model. Animals were treated with doses of the antibiotics to simulate the human exposure of meropenem 2 g q8 h 30-min infusion, cefepime 2 g q8 h 30-min infusion and levofloxacin 500 mg q24 h. After 24 hours, the animals were euthanized and efficacy was calculated as the change in thigh bacterial density (log 10 CFU) relative to the starting inoculum (0 h). Results As expected, levofloxacin was ineffective against all isolates due to their resistant phenotype (8 to>64 mg/L). Cefepime also showed minimal activity against all isolates consistent with its failure to achieve pharmacodynamic target exposures due to high MICs of 32 to>512 mg/L. In the presence of low MICs (4 to 16 mg/L), the f T> MIC of meropenem was sufficiently high to result in CFU reductions. However, conflicting activity was noted for isolates with MICs = 128 mg/L that possessed the same enzymatic profile, suggesting that other mechanisms of resistance are responsible for driving CFU outcomes. No activity was noted for organisms with a meropenem MIC = 512 mg/L. Conclusion Unlike previous observations with MBL-producing Enterobacteriaceae that showed discordance between in vitro resistance and in vivo efficacy in the murine infection model, we found that the efficacy of humanized cefepime and meropenem was generally concordant with the phenotypic profile of VIM-producing P. aeruginosa .

Tallamy This paper reports data from a residential landscape preference study conducted in Delaware, USA. The researchers constructed an ecologically designed exurban residential landscape, which delivered 20 new environmental and human-related impacts, including 7 that delivered ecosystem services. Ecosystem services included impacts such as improved flood control and enhanced plant diversity. Using pictures before and after the intervention, an intercept survey of 105 non-neighboring residents estimated whether the 20 impacts positively, negatively, or did not affect the respondents’ household wellbeing. The public found that most landscape-intervention impacts had a positive effect on their quality of life, especially those impacts involving ecosystem services. All but one ecosystem service were found to be strong amenities and the other (moving indoor activities outside) was an amenity. However, the landscape intervention delivered one clear disamenity: increased undesirable wildlife. Respondents also identified what impacts were the most important in affecting their welfare: undesirable wildlife (negative); flood control (positive); and water quality (positive). Ecosystem services accounted for 41.6% of the public’s importance rating, while undesirable wildlife was 12.9%. A planning process seeking more ecosystem services from residential landscapes should focus on all the most important drivers of preference, if it is to be accepted by residents.

Four-week-old male ddY mice were fed a Ca-deficient diet for 4 weeks as a part of the experimental protocol. While hypocalcemia was observed, there was no significant change in body weight. The CCl 4 -exposed hypocalcemic mice exhibited a significant decrease in alanine aminotransferase and aspartate aminotransferase activities at both 6 h and 24 h even though markers of renal function remained unchanged. Moreover, lipid peroxidation was impaired and total antioxidant power was partially recovered in the liver. Studies conducted in parallel with the biochemical analysis revealed that hepatic histopathological damage was attenuated 24 h post CCl 4 injection in hypocalcemic mice fed the Ca-deficient diet. Finally, this diet impaired CCl 4 -induced inflammatory responses. Although upregulation of Ca concentration is a known indicator of terminal progression to cell death in the liver, these results suggest that Ca is also involved in other phases of CCl 4 -induced hepatotoxicity, via regulation of oxidative stress and inflammatory responses.

Smirnova This article highlights certain inconsistencies that exist in modern lexicography with respect to word meaning representation and proposes an alternative outlook on the nature of word meaning based on the principles of the functional linguo-anthropological theory. Through the case study of the lexeme bank , it has been demonstrated that word meaning can be presented as a more coherent structure if only the functional evaluative nature of language be taken into account more accurately while compiling a dictionary entry. The article defends the idea that the evaluative nature of human experience as a source of meaning in language determines the way lexical items behave in real texts and, therefore, demonstrates the necessity of abandoning basic principles of referential semantics (with its stress on objective features of referents as physical objects) in lexicographical descriptions.

Patterson Despite improvements made in stroke rehabilitation, motor impairment and gait deficits persist at discharge. New interventions are needed. Mirror therapy has promise as one element of a rehabilitation program. The primary objectives were to 1) describe a bilateral, lower extremity mirror therapy (LE-MT) device and training protocol and 2) investigate the feasibility of LE-MT. A LE-MT device was constructed to train bilateral LE movements for 30 min, 3 times/week for 4 weeks, as an adjunct to physiotherapy in three individuals post-stroke. Sessions were digitally recorded and reviewed to extract feasibility measures; repetitions, rests and session duration. Pre and post measures of gait and motor impairment were taken. Two participants completed 100% of the sessions and a third completed 83% due to a recurrence of pre-existing back pain. Repetitions increased and session duration was maintained. Number of rests decreased for two participants and increased for one participant. Participants reported fatigue and mild muscle soreness but also that the intervention was tolerable. Positive gait changes included increased velocity and decreased variability. LE motor impairment also improved. A bilateral LE-MT adjunct intervention for stroke is feasible and may have positive effects. A history of low back pain should be a precaution.

Animal experiments assessed the effect of SPP on body weight, abdominal adipose tissue mass, serum lipids and adipocytokines. Body and liver weight and epididymal and mesenteric fat of mice fed a high-fat diet containing 0.5% or 5% SPP for 28 days were significantly lower than control mice. Triglyceride and cholesterol in VLDL and LDL and leptin levels were significantly lower in the serum of SPP-administered mice compared to control mice. Biomarker arrays showed that adiponectin, melanocyte-stimulating-hormone-alpha and neuromedin U were more than 1.5 times higher, while TNF-alpha was about 1.5 times lower in the livers of SPP-administered mice compared to control mice. These results suggest SPP mitigated leptin resistance in mice administered a high-fat diet, and maintained anorexigenic peptide levels. SPP administration may suppress lipogenesis by increasing adiponectin levels and decreasing TNF-alpha levels in adipocytes.
