    Beschreibung: Street trees have aesthetic, environmental, human health, and economic benefits in urban ecosystems. Street tree populations are constructed by cycles of planting, growth, death, removal and replacement. The goals of this study were to understand how tree mortality and planting rates affect net population growth, evaluate the shape of the mortality curve, and assess selected risk factors for survival. We monitored a street tree population in West Oakland, CA for 5 years after an initial inventory (2006). We adapted the classic demographic balancing equation to quantify annual inputs and outputs to the system, tracking pools of live and standing dead trees. There was a 17.2 % net increase in live tree counts during the study period (995 in 2006, 1166 in 2011), with population growth observed each year. Of the live trees in 2006, 822 survived to 2011, for an annual mortality rate of 3.7 %. However, population growth was constrained by high mortality of young/small trees. Annual mortality was highest for small trees, and lower for mid-size and large trees; this represents a Type III mortality curve. We used multivariate logistic regression to evaluate the relationship between 2011 survival outcomes and inventory data from 2006. In the final model, significant associations were found for size class, foliage condition, planting location, and a multiplicative interaction term for size and foliage condition. Street tree populations are complex cultivated systems whose dynamics can be understood by a combination of longitudinal data and demographic analysis. Urban forest monitoring is important to understand the impact of tree planting programs.

    Beschreibung: Hong Kong’s rugged topography demands creation of developable land by reclaiming the harbor and terracing hillslopes. The vertical faces between adjoining hillslope-platforms are often stabilized by stone retaining walls. Such artificial cliffs embedded in the urban matrix, as a habitat analogue of rocky cliffs, have been colonized by spontaneous-ruderal vegetation. Detailed field assessment of 289 walls with 793 trees permitted in-depth analysis of this unique tropical urban ecology to inform conservation. The 28 stonewall tree species, mainly native, are dominated by six keystone and companion Ficus (Moraceae) species. They can develop into large cliff hangers 〉 20 m height on the apparently precarious and inhospitable walls. The many joints between masonry blocks provide opportunities for lodging and germination of seeds brought by frugivorous birds and bats. The aft-soil behind walls provides catchment for normal root functions. Beginning as pioneers and persisting as climax members, they short-circuit the seral processes. Wall-gripping capability as large epilithic woody hemi-epiphytes is due to the strangler-fig habit, an evolutionary trait that originated in the tropical forest. The polymorphic roots with morphological plasticity and self-grafting ability, and their multiple modes of interactions with wall niches, account for firm attachment on the vertical habitat and exploration of proximal soils. A model of strangler-fig growth on stone walls serving as surrogate host tree is developed. The diverse and versatile rooting modes are the basis to sustain strangler-fig wall conditions to permit continuation of strangler-fig habit. Such unique nature-in-city and natural-cum-cultural gems deserve conservation as urban ecological heritage.

    Beschreibung: Norway rats ( Rattus norvegicus ) and black rats ( Rattus rattus ) are among the most prolific and widespread urban pest species in the world. Yet despite their ubiquity, a unified understanding of the ecology of these species in urban habitats eludes us. A comprehensive understanding of urban rat ecology is important for managing rat populations and mitigating the harmful effects that they may have on urban ecosystems (e.g., structural damage, food contamination, and disease spread). The objective of this systematic review and narrative synthesis is to collate, compare, and contrast data from the published literature regarding the ecology of Norway and black rats in urban centers. Themes emerging from the synthesis process, and discussed in detail, include population dynamics, behavior, movement, and environmental influences on rat populations.

    Beschreibung: Estimating the relative importance of vegetation on residential land (gardens, yards, and street-trees) and vegetation on non-residential land (parks and other large green spaces) is important so that competing options for urban conservation planning can be prioritized. We used data from an urban breeding-bird monitoring program to compare the relative effects vegetation on residential land and vegetation on non-residential land (both the amount and type of vegetation at local and landscape scales) on bird species richness and an index of conservation value for the bird community. We then estimated the realised relative benefit of managing the amount of vegetation on these two types of land (i.e., as alternative management options for promoting biodiversity), which might be achieved within the practical limits imposed by human population density. The local effects of increasing residential and non-residential vegetation amount were similar and positive on all measures of bird species richness and conservation value. Non-residential vegetation had an additional landscape-scale influence on bird diversity that residential vegetation did not. Options for managing the amount of non-residential vegetation appear to be more limited by high human population density than for managing the amount of residential vegetation. This suggests that there may be greater realised benefits to bird diversity from managing the amount of vegetation on residential land than from the more common focus of urban planning of managing vegetation on non-residential land.

    Beschreibung: Many regions of the globe are experiencing rapid urban growth, the location and intensity of which can have negative effects on ecological and social systems. In some locales, planners and policy makers have used urban growth boundaries to direct the location and intensity of development; however the empirical evidence for the efficacy of such policies is mixed. Monitoring the location of urban growth is an essential first step in understanding how the system has changed over time. In addition, if regulations purporting to direct urban growth to specific locales are present, it is important to evaluate if the desired pattern (or change in pattern) has been observed. In this paper, we document land cover and change across six dates (1986, 1991, 1995, 1999, 2002, and 2007) for six counties in the Central Puget Sound, Washington State, USA. We explore patterns of change by three different spatial partitions (the region, each county, 2000 U.S. Census Tracks), and with respect to urban growth boundaries implemented in the late 1990’s as part of the state’s Growth Management Act. Urban land cover increased from 8 to 19% of the study area between 1986 and 2007, while lowland deciduous and mixed forests decreased from 21 to 13% and grass and agriculture decreased from 11 to 8%. Land in urban classes outside of the urban growth boundaries increased more rapidly (by area and percentage of new urban land cover) than land within the urban growth boundaries, suggesting that the intended effect of the Growth Management Act to direct growth to within the urban growth boundaries may not have been accomplished by 2007. Urban sprawl, as estimated by the area of land per capita, increased overall within the region, with the more rural counties within commuting distance to cities having the highest rate of increase observed. Land cover data is increasingly available and can be used to rapidly evaluate urban development patterns over large areas. Such data are important inputs for policy makers, urban planners, and modelers alike to manage and plan for future population, land use, and land cover changes.

    Beschreibung: Due to economic decline and the recent rise in home foreclosure, many U.S. cities are faced with managing large acreages of vacant land. Interest in local food production on this land has the potential to dramatically reshape the composition of greenspace found within urban landscapes. This study examined how the conversion of vacant land to urban gardens and farms influenced arthropod generalist predator populations and their ability to support biocontrol services. We found that the abundance of Coccinellidae and Syrphidae, and the activity density of Carabidae, Formicidae, and Lycosidae were equivalent among vacant lot and urban garden sites. Dolichopodidae abundance and the activity density of Linyphiidae and Opiliones were reduced in urban gardens whereas Anthocoridae abundance and the activity density of Staphylinidae were greater within urban gardens relative to vacant lots. The biocontrol service supplied by generalist predators was measured using sentinel eggs ( Helicoverpa zea (Noctuiidae)) and pupae ( Sarcophaga bullata (Sarcophagidae) and Musca domestica (Muscidae)). We found no difference in the biocontrol of H . zea eggs or M . domestica pupae among the focal greenspaces. We found no difference in the removal of S . bullata pupae by predators in June 2010, whereas in August 2010 a greater proportion of pupae were removed in urban garden sites relative to vacant lots. Based on these findings, we discuss the potential of the urban landscape to support generalist predators and sustainable urban gardening and farming.

    Beschreibung: Urbanization processes challenge ecosystem health in many metropolitan areas. New policy and program approaches are needed to restore and sustain natural systems as public agencies and organizations face greater demands and declining budgets. Environmental stewardship is an often overlooked intervention strategy, and the full potential of civic engagement by citizens on behalf of ecosystem health is little understood. Using a coupled systems approach, integrated analysis of social and ecological footprints can lead to greater theoretical understanding and more effective programs at the landscape scale. Here we outline two pilot studies as part of an emergent research program that is investigating the extent and impact of environmental stewardship. Qualitative interviews of stewardship managers revealed multiple dimensions of motivations and purposes for stewardship, ranging from the practical to the conceptual. A regional organization census yielded a surprisingly large number of organizations that conduct stewardship, with social and ecological values being of comparable emphasis. The initial research is based in the Puget Sound area of Washington State, U.S., but results have relevance to other urban areas. Pilot study findings now guide additional research effort about motivations, organizational networks, and theory of integrated socio-ecological systems to be derived from comprehensive footprint analysis of stewardship activity.

    Beschreibung: Urban land area is expanding worldwide and may contribute to long-term carbon (C) storage; however, little is known about potential drivers of soil C in urban areas. Residential areas are one of the largest urban land use zones and lawns can provide stable chronosequences for studying soil C dynamics. In residential lawns containing no trees ( n  = 23), the relationships between soil C and four potential drivers [home age (1–51 years), yard maintenance practices (fertilization, irrigation, and bagging or mulching lawn clippings), soil nitrogen (N) and soil texture] were investigated. Soil C increased with home age at 0–15 cm depth by 0.026 kg C m −2  yr −1 , declined by −0.011 kg C m −2  yr −1 at 15–30 cm depth, and was stable at 30–50 cm depth. Soil C had a positive relationship with soil N ( R 2  = 0.55) at the 0–15 cm depth. Soil C and N were not related to yard maintenance practices or soil texture. The low soil C sequestration rate and limited relationships between soil C and home age, yard maintenance, soil N and soil texture may have resulted from the positive influence of Auburn’s humid, subtropical climate on residue decomposition.

    Beschreibung: High total organic carbon (TOC) concentrations in Converse Reservoir, a water source for Mobile, Alabama, have concerned water treatment officials due to the potential for disinfection byproduct (DBP) formation. TOC reacts with chlorine during drinking water treatment to form DBPs. This study evaluated how increased urbanization can alter watershed-derived total nitrogen (TN), total phosphorus (TP) and TOC inputs to the Converse Reservoir. Converse Watershed, on the urban fringe of Mobile, is projected to undergo urbanization increasing watershed urban land from 3% in 1992 to 22% urban land by 2020. A pre-urbanization scenario using 1992 land cover was coupled with 2020 projections of land use. The Loading Simulation Program C++ watershed model was used to evaluate changes in nutrient concentrations (mg L −1 ) and loads (kg) to Converse Reservoir. Urban and suburban growth of 52 km 2 simulated from 1991 to 2005 (15 year) caused overall TN and TP loads to increase by 109 and 62%, respectively. Simulated urban growth generally increased monthly flows by 15%, but resulted in lower streamflows (2.9%) during drought months. Results indicate that post-urbanization median TN and TP concentrations were 59 and 66% higher than corresponding pre-urbanization concentrations, whereas TOC concentrations were 16% lower. An increase in urban flow caused TOC loads to increase by 26%, despite lower post-urbanization TOC concentrations.

    Beschreibung: Due to increasing exurban settlement around Austin, Texas and subsequent land use changes, the City of Austin Wildland Division faces competing pressures from residential communities regarding land and wildlife management actions. Current environmental education and outreach efforts in Austin focus on developing environmental literacy about the city’s wildland areas adjacent to exurban settlement as well as their management program. The desired outcome is to promote more positive perceptions and motivate pro-environmental behaviors that are consistent with the city’s wildland program. We conducted a mail survey of 1,000 residents living near the urban wildlands to better understand factors that influence residents’ proficiency in four knowledge domains: karst aquifers, endangered species, rangeland ecology, and the city’s management program. Regression analyses identified positive associations between local newspaper readership and residents’ environmental knowledge of karst aquifer geology, rangelands, and city’s wildland program. Previous engagement in environmentally oriented education programs or organization was positively associated with all four knowledge domains. Some strategies for information dissemination about the wildlands include the use of local newspapers and homeowners association newsletters. Newer, younger residents are the suggested target audience for initiating a proposed environmental education and outreach program. Such programs should incorporate both local environmental organizations and homeowners associations.

    Beschreibung: The environment is both a setting for and a product of human interactions. Understanding the dynamic nature of human-environment interactions is critical for mitigating the impacts of human induced environmental change and understanding how the environment shapes social systems. Current research has focused on the reduced ability of many natural systems to provide ecosystem services and the subsequent impact on human well-being. Furthermore, there has been a proliferation of cases analyzing the impacts of payment programs designed to enhance ecosystem services. However, analyses that link environmental policies through to their ecological results are not common and methods to do so are not thoroughly developed. To better analyze these interactions, a theory or framework is necessary. This article presents a framework of social ecological complex adaptive systems (SECAS). The framework links structuration theory from social science with the theories of complex adaptive systems from ecology to provide an enhanced understanding of the human drivers and responses to environmental change. The framework is presented as a recursive process where social and ecological systems are both the medium for and product of social action and ecological disturbance. A case study of Costa Rica’s ecosystem service payment program is presented as a demonstration of empirical applicability. This framework is proposed as a method to evaluate payments for ecosystem services, conservation policies, urban ecosystems, and for land use change in general.

    Beschreibung: Changes to the ground layer in urban areas are extensive, but the effects on arthropod fauna are poorly understood. We undertook a manipulative experiment to examine the response of arthropods to small-scale variation in ground covers commonly found in urban parks and gardens in Australia. The ground covers tested were bare ground, leaf litter, woodchips and grass, with plot sizes of 3.6 m 2 . Epigeic arthropods were sampled with pitfall traps and Tullgren funnels over 12 months following establishment of the treatments. All epigeic arthropods were sorted to order and the ants (Hymenoptera: Formicidae), beetles (Coleoptera), millipedes (Diplopoda) and slaters (Isopoda: Oniscidea) were examined at lower taxonomic levels. Diverse arthropods rapidly colonised previously cleared plots in all four treatments and were most abundant in grass plots. The diversity of ants and beetles was significantly different in different ground covers and tended to be most diverse in grass plots. Despite the treatments providing very different microclimates, the fauna studied did not show strong selection for a particular cover type overall. The abundance of grass cover in the surrounding area may have led to the grass plots having the greatest abundance of arthropods. These results have important implications for developing effective small-scale conservation efforts for arthropods in anthropogenically modified landscapes, especially for species with poor dispersal abilities.

    Beschreibung: Green infrastructure (GI) is gaining wide popularity as a means to reduce urban stormwater runoff. The challenges to long-term success of many GI policies, however, depend on citizen understanding and willingness to be engaged in the process of GI implementation. In this study, we developed a conceptual model that incorporated human preferences and biophysical processes in a coupled manner to estimate stormwater runoff variation at the sewershed scale under different acceptable GI implementation scenarios. To investigate resident receptivity toward GI implementation, we conducted surveys in selected Syracuse sewersheds and developed the scenario of residents implementing rain barrels and rain gardens in their private properties based on the survey results. We simulated this scenario at the lot level using the model we developed within the framework of the EPA SWMM 5 model to estimate reductions in peak flow and total runoff volume for major storm events. With our model, we also tested the effect of planned government tree planting scenarios. The scenario simulations were applied in three urban sewersheds of varying socio-economic and biophysical structures in the City of Syracuse, New York. The simulation results indicated that both the “government participation” and “household participation” scenarios, compared to pre-GI-development conditions, would contribute to a modest reduction in stormwater peak flow (〉4 %) and total runoff volume (〉5 %) across the simulated sewersheds. This study provides decision makers with a scientific methodology to quantify how human decisions can shape ecosystem function, and thus support sustainable stormwater management planning while addressing citizen preferences and needs.

    Beschreibung: Bogotá, the capital of the Republic of Colombia, is a tropical highland city located 2,650 m above sea level. It is the 25th largest city in the world and, among large cities, is also one of the highest. In common with other cities in Latin America, a large part of its urban growth during recent centuries has been unplanned and informal. The introduction of green spaces into urban planning in Bogotá began in the mid-20th century, but was first included in official legislation during the 1990s through the concept of Ecological Main Structure (EMS). Initially developed by Dutch scholars, EMS was brought to Colombia via biological conservation practitioners as a means of enhancing biological connectivity in rural and natural landscapes, extended in this case to urban landscapes as a top-level planning instrument. EMS originally included a variety of components, from protected areas and biological conservation tools to environmental urban elements - the emphasis being on biodiversity conservation, without sufficient recognition of specific urban structures and functions. This process led to conceptual disciplinary-based divergence and conflicting political interpretations. The current emergence of EMS as a planning tool for urban regions represents an opportunity for integration, although the risk of divergent interpretations remains, as no integrative conceptual framework has yet been developed. In this paper we review the concepts underlying EMS that have been incorporated within urban and regional planning, especially those of ecological networks and green infrastructure, and also diagnose conceptual and institutional barriers to its current integration, challenges and opportunities which are set in the context of an emerging urban region. We propose a trans-disciplinary framework for multi-level integration of EMS, along a gradient from wild environments to built structures that incorporates emerging concepts such as urban biodiversity, ecosystem services and design in the urban landscape, with the aim of contributing to the creation of an urban landscape that is resilient to environmental change and suitable for human well-being and adaptation.

    Beschreibung: Coyotes ( Canis latrans ) have inhabited Florida (USA) since the 1960s and are currently found throughout the state. Our objective was to obtain information on enteric parasites of coyotes from two different habitats. Fresh coyote fecal samples were collected from protected and suburban habitats in Pinellas County, Florida, USA (27 o 54′ N, 82 o 41′W) from May 2005 to March 2007. A standard fecal flotation examination and formalin-ethyl acetate sedimentation protocol were utilized on fecal samples from both habitats. Five newly documented coyote parasites were documented: one cestode ( Hymenolepis spp.), one nematode ( Ascaris spp.), and three protozoa ( Balantidium coli , Blastocystis spp., and Entamoeba histolytica ). Nine hitherto unreported parasites for FL coyotes were also discovered: two cestodes ( Diphyllobothrium latum and Dipylidium caninum ), two nematodes ( Toxocara canis and Uncinaria stenocephala ), one trematode ( Paragonimus spp.), and four protozoa ( Cryptosporidium spp., Giardia canis , Cystoisospora spp., and Sarcocystis cruzi ). The protected area supported significantly more undocumented (i.e., newly identified) parasites for FL coyotes, and Protozoa as compared to the suburban area. Florida coyotes are likely more susceptible to infection by novel parasites because of their rapid range expansion and lack of acquired immunity. In addition, rapid habitat loss and urbanization in Florida may increase the probability of disease transmission between wild and domestic canids. We suggest preventative measures that may lower the risk of parasitic infection and promote co-existence with coyotes in urban landscapes.

    Beschreibung: Assessing in situ microbial abilities of soils to degrade pesticides is of great interest giving insight in soil filtering capability, which is a key ecosystem function limiting pollution of groundwater. Quantification of pesticide-degrading gene expression by reverse transcription quantitative PCR (RT-qPCR) was tested as a suitable indicator to monitor pesticide biodegradation performances in soil. RNA extraction protocol was optimized to enhance the yield and quality of RNA recovered from soil samples to perform RT-qPCR assays. As a model, the activity of atrazine-degrading communities was monitored using RT-qPCRs to estimate the level of expression of atzD in five agricultural soils showing different atrazine mineralization abilities. Interestingly, the relative abundance of atzD mRNA copy numbers was positively correlated to the maximum rate and to the maximal amount of atrazine mineralized. Our findings indicate that the quantification of pesticide-degrading gene expression may be suitable to assess biodegradation performance in soil and monitor natural attenuation of pesticide.

    Beschreibung: This unique study describes how Aspergillus japonicus , Penicillium brocae and Purpureocillium lilacinum , three novel isolates of our laboratory from heavily plastics-contaminated soil completely utilized the plasticizer di(2-ethylhexyl)phthalate (DEHP) bound to PVC blood storage bags (BB) in simple basal salt medium (BSM) by static submerged growth (28 °C). Initial quantification as well as percentage utilization of DEHP blended to BB were estimated periodically by extracting it into n -hexane. A two-stage cultivation strategy was employed for the complete mycoremediation of DEHP from BB in situ. During the first growth stage, about two-third parts of total (33.5 % w/w) DEHP bound to BB were utilized in two weeks, accompanied by increased fungal biomass (~0.15–0.32 g per g BB) and sharp declining (to ~3) of initial pH (7.2). At this stagnant growth state (low pH), spent medium was replaced by fresh BSM (pH, 7.2), and thus in the second stage the remaining DEHP (one-third) in BB was utilized completely. The ditches and furrows seen from the topology of the BB as seen by the 3D AFM image further confirmed the bioremediation of DEHP physically bound to BB in situ. Of the three mycelial fungi employed, P. lilacinum independently showed highest efficiency for the complete utilization of DEHP bound to BB, whose activity was comparable to that of the consortium comprising all the three fungi described herein. To sum up, the two-stage cultivation strategy demonstrated in this study shows that a batch process would efficiently remediate the phthalic acid esters blended in plastics on a large scale, and thus it offers potentials for the management of plastics wastes.

    Beschreibung: A comprehensive study on the effects of different carbon sources during the bacterial enrichment on the removal performances of benzene, toluene, ethylbenzene, and xylenes (BTEX) compounds when present as a mixture was conducted. Batch BTEX removal kinetic experiments were performed using cultures enriched with individual BTEX compounds or BTEX as a mixture or benzoate alone or benzoate–BTEX mixture. An integrated Monod-type non-linear model was developed and a ratio between maximum growth rate ( μ max ) and half saturation constant (K s ) was used to fit the non-linear model. A higher μ max /K s indicates a higher affinity to degrade BTEX compounds. Complete removal of BTEX mixture was observed by all the enriched cultures; however, the removal rates for individual compounds varied. Degradation rate and the type of removal kinetics were found to be dependent on the type of carbon source during the enrichment. Cultures enriched on toluene and those enriched on BTEX mixture were found to have the greatest μ max /K s and cultures enriched on benzoate had the least μ max /K s . Removal performances of the cultures enriched on all different carbon sources, including the ones enriched on benzoate or benzoate–BTEX mixture were also improved during a second exposure to BTEX. A molecular analysis showed that after each exposure to the BTEX mixture, the cultures enriched on benzoate and those enriched on benzoate–BTEX mixture had increased similarities to the culture enriched on BTEX mixture.

    Beschreibung: An investigation was done to determine the occurrence and composition of avian fauna community in the urbanizing city of Nairobi, Kenya. We conducted bird counts in sample sites randomly distributed over the Nairobi landscape within a two-year period. The proportion of seven different land cover types derived from within a 500 m radius of classified satellite image described the habitat condition of each sample site. Multivariate analysis of the site and species data indicated that savannah vegetation, forest and agriculture land cover types were the main environmental gradients that differentiated the sample sites. Four clusters of sample sites occurred on the ordination plane according to canonical correspondence analysis (CCA). From 50 families of birds observed, species related to bush and scrub habitats occurred at a rate of about 31 %, followed by grassland species at 20 % and forest species at about 16 % rate. Out of CCA, five functional groups of birds were distinguishable. The proportion of sites occupied by birds per functional group and mean count of individual birds declined significantly, p  〈 0.0001, as groups of birds changed from being urban-related to savannah and then to forest-related species. One cluster comprised birds marked as having conservation concern status and related mainly to savannah vegetation of scrub and grasslands. In this landscape context, features such as native savannah vegetation and woodland accompanied by a process of controlled land use, could greatly mitigate negative impacts of ecosystem degradation on the sensitive tropical urban bird biodiversity.

    Beschreibung: The residential yard is an integral part of the urban ecosystem. Individual preferences and social expectations influence homeowners’ yard care choices, which in turn affect urban ecology. However, little is known about residents’ conceptions of their yards as part of the urban ecosystem. We asked how homeowners conceive of their yard as part of the urban ecosystem by examining urban ecosystem concepts embedded within homeowners’ descriptions and stories of their yards. Our study sites included an urban and suburban area in the Saint Paul-Minneapolis metropolitan area of Minnesota, USA. We found that people’s understandings of their yards as urban ecosystems are complex but have prominent gaps. Salient concepts included biotic and abiotic interactions within the yard, linkages of human inputs and weeds across yards and watersheds, and yards as social space. Stories described managing dynamic ecological processes within yards to maintain a steady state and limiting linkages of human inputs beyond the yard. Prominent gaps included ecological cycles, biodiversity, and ecosystem services within yards and ecological linkages across yards. In general, people conceived of their yards in terms of inputs rather than cycles and in terms of creating barriers between their yards and surrounding areas rather than fostering ecological interconnections across them. We provide recommendations for resident outreach programs based on our findings. Finally, our study presents a challenge to urban ecosystem research to unravel where there are gaps in understandings of urban ecosystems versus where there is resistance to incorporating certain ecological interactions within the residential yard.

    Beschreibung: In this paper, we report a study on how property taxes influence land use change decisions of Georgia private landowners. We analyze National Resources Inventory plot-level land use data and county-level socio-economic data using a panel random parameter logit model. Our results indicate that property taxes have significantly negative impacts on private landowners’ land-use change decisions, but these impacts are relatively small.

    Beschreibung: A genetically engineered microorganism (GEM) capable of simultaneously degrading organophosphate and organochlorine pesticides was constructed for the first time by display of organophosphorus hydrolase (OPH) on the cell surface of a hexachlorocyclohexane (HCH)-degrading Sphingobium japonicum UT26. The GEM could potentially be used for removing the two classes of pesticides that may be present in mixtures at contaminated sites. A surface anchor system derived from the truncated ice nucleation protein (INPNC) from Pseudomonas syringae was used to target OPH onto the cell surface of UT26, reducing the potential substrate uptake limitation. The surface localization of INPNC–OPH fusion was verified by cell fractionation, western blot, proteinase accessibility, and immunofluorescence microscopy. Furthermore, the functionality of the surface-exposed OPH was demonstrated by OPH activity assays. Surface display of INPNC–OPH fusion (82 kDa) neither inhibited cell growth nor affected cell viability. The engineered UT26 could degrade parathion as well as γ-HCH rapidly in minimal salt medium. The removal of parathion and γ-HCH by engineered UT26 in sterile and non-sterile soil was also studied. In both soil samples, a mixture of parathion (100 mg kg −1 ) and γ-HCH (10 mg kg −1 ) could be degraded completely within 15 days. Soil treatment results indicated that the engineered UT26 is a promising multifunctional bacterium that could be used for the bioremediation of multiple pesticide-contaminated environments.

    Beschreibung: Stimulation of native microbial populations in soil by the addition of small amounts of secondary carbon sources (cosubstrates) and its effect on the degradation and theoretical mineralization of DDT [l,l,l-trichloro-2,2-bis( p -chlorophenyl)ethane] and its main metabolites, DDD and DDE, were evaluated. Microbial activity in soil polluted with DDT, DDE and DDD was increased by the presence of phenol, hexane and toluene as cosubstrates. The consumption of DDT was increased from 23 % in a control (without cosubstrate) to 67, 59 and 56 % in the presence of phenol, hexane and toluene, respectively. DDE was completely removed in all cases, and DDD removal was enhanced from 67 % in the control to ~86 % with all substrates tested, except for acetic acid and glucose substrates. In the latter cases, DDD removal was either inhibited or unchanged from the control. The optimal amount of added cosubstrate was observed to be between 0.64 and 2.6 mg C $ {\text{g}}^{ - 1}_{\text{dry soil}} $ . The CO 2 produced was higher than the theoretical amount for complete cosubstrate mineralization indicating possible mineralization of DDT and its metabolites. Bacterial communities were evaluated by denaturing gradient gel electrophoresis, which indicated that native soil and the untreated control presented a low bacterial diversity. The detected bacteria were related to soil microorganisms and microorganisms with known biodegradative potential. In the presence of toluene a bacterium related to Azoarcus , a genus that includes species capable of growing at the expense of aromatic compounds such as toluene and halobenzoates under denitrifying conditions, was detected.

    Beschreibung: Because benzene, toluene, ethylbenzene, and xylenes (BTEX) and ethanol are important contaminants present in Brazilian gasoline, it is essential to develop technology that can be used in the bioremediation of gasoline-contaminated aquifers. This paper evaluates the performance of a horizontal-flow anaerobic immobilized biomass (HAIB) reactor fed with water containing gasoline constituents under denitrifying conditions. Two HAIB reactors filled with polyurethane foam matrices (5 mm cubes, 23 kg/m 3 density and 95 % porosity) for biomass attachment were assayed. The reactor fed with synthetic substrate containing protein, carbohydrates, sodium bicarbonate and BTEX solution in ethanol, at an Hydraulic retention time (HRT) of 13.5 h, presented hydrocarbon removal efficiencies of 99 % at the following initial concentrations: benzene 6.7 mg/L, toluene 4.9 mg/L, m -xylene and p -xylene 7.2 mg/L, ethylbenzene 3.7 mg/L, and nitrate 60 mg N/L. The HAIB reactor fed with gasoline-contaminated water at an HRT of 20 h showed hydrocarbon removal efficiencies of 96 % at the following initial concentrations: benzene, 4.9 mg/L; toluene, 7.2 mg/L; m -xylene, 3.7 mg/L; and nitrate 400 mg N/L. Microbiological observations along the length of the HAIB reactor fed with gasoline-contaminated water confirmed that in the first segment of the reactor, denitrifying metabolism predominated, whereas from the first sampling port on, the metabolism observed was predominantly methanogenic.

    Beschreibung: Burkholderia sp. C3, an efficient polycyclic aromatic hydrocarbon degrader, can utilize nine of the ten N -methylcarbamate insecticides including carbaryl as a sole source of carbon. Rapid hydrolysis of carbaryl in C3 is followed by slow catabolism of the resulting 1-naphthol. This study focused on metabolomes and proteomes in C3 cells utilizing carbaryl in comparison to those using glucose or nutrient broth. Sixty of the 867 detected proteins were involved in primary metabolism, adaptive sensing and regulation, transport, stress response, and detoxification. Among the 41 proteins expressed in response to carbaryl were formate dehydrogenase, aldehyde-alcohol dehydrogenase and ethanolamine utilization protein involved in one carbon metabolism. Acetate kinase and phasin were 2 of the 19 proteins that were not detected in carbaryl-supported C3 cells, but detected in glucose-supported C3 cells. Down-production of phasin and polyhydroxyalkanoates in carbaryl-supported C3 cells suggests insufficient carbon sources and lower levels of primary metabolites to maintain an ordinary level of metabolism. Differential metabolomes (~196 identified polar metabolites) showed up-production of metabolites in pentose phosphate pathways and metabolisms of cysteine, cystine and some other amino acids, disaccharides and nicotinate, in contract to down-production of most of the other amino acids and hexoses. The proteomic and metabolomic analyses showed that carbaryl-supported C3 cells experienced strong toxic effects, oxidative stresses, DNA/RNA damages and carbon nutrient deficiency.

    Beschreibung: Species of the genus Variovorax are often isolated from nitrile or amide-containing organic compound-contaminated soil. However, there have been few biological characterizations of Variovorax and their contaminant-degrading enzymes. Previously, we reported a new soil isolate, Variovorax boronicumulans CGMCC 4969, and its nitrile hydratase that transforms the neonicotinoid insecticide thiacloprid into an amide metabolite. In this study, we showed that CGMCC 4969 is able to degrade acrylamide, a neurotoxicant and carcinogen in animals, during cell growth in a mineral salt medium as well as in its resting state. Resting cells rapidly hydrolyzed 600 mg/L acrylamide to acrylic acid with a half-life of 2.5 min. In in vitro tests, CGMCC 4969 showed plant growth-promoting properties; it produced a siderophore, ammonia, hydrogen cyanide, and the phytohormone salicylic acid. Interestingly, in soil inoculated with this strain, 200 mg/L acrylamide was completely degraded in 4 days. Gene cloning and overexpression in the Escherichia coli strain Rosetta (DE3) pLysS resulted in the production of an aliphatic amidase of 345 amino acids that hydrolyzed acrylamide into acrylic acid. The amidase contained a conserved catalytic triad, Glu59, Lys 134, and Cys166, and an “MRHGDISSS” amino acid sequence at the N-terminal region. Variovorax boronicumulans CGMCC 4969, which is able to use acrylamide for cell growth and rapidly degrade acrylamide in soil, shows promising plant growth-promoting properties. As such, it has the potential to be developed into an effective Bioaugmentation strategy to promote growth of field crops in acrylamide-contaminated soil.

    Beschreibung: Organic and metallic pollutants are ubiquitous in the environment. Many metals are reported to be toxic to microorganisms and to inhibit biodegradation. The effect of the metals iron, copper and silver on the metabolism of Labrys portucalensis F11 and on fluorobenzene (FB) biodegradation was examined. The results indicate that the addition of 1 mM of Fe 2+ to the culture medium has a positive effect on bacterial growth and has no impact in the biodegradation of 1 and 2 mM of FB. The presence of 1 mM of Cu 2+ was found to strongly inhibit the growth of F11 cultures and to reduce the biodegradation of 1 and 2 mM of FB to ca. 50 %, with 80 % of stoichiometrically expected fluoride released. In the experiments with resting cells, the FB degraded (from 2 mM supplied) was reduced ca. 20 % whereas the fluoride released was reduced to 45 % of that stoichiometrically expected. Ag + was the most potent inhibitor of FB degradation. In experiments with growing cells, the addition of 1 mM of Ag + to the culture medium containing 1 and 2 mM of FB resulted in no fluoride release, whereas FB degradation was only one third of that observed in control cultures. In the experiments with resting cells, the addition of Ag + resulted in 25 % reduction in substrate degradation and fluoride release was only 20 % of that stoichiometrically expected. The accumulation of catechol and 4-fluorocatechol in cultures supplemented with Cu 2+ or Ag + suggest inhibition of the key enzyme of FB metabolism—catechol 1,2-dioxygenase.

    Beschreibung: In the present study, the influence of kaolinite and goethite on microbial degradation of methyl parathion was investigated. We observed that the biodegradation process was improved by kaolinite and depressed by goethite. Calorimetric data further showed that the metabolic activities of degrading cells ( Pseudomonas putida ) were enhanced by the presence of kaolinite and depressed by the presence of goethite. A semipermeable membrane experiment was performed and results supported the above observations: the promotive effect of kaolinite and the inhibition of goethite for microbial degradation was not found when the bacteria was enclosed by semipermeable membrane and had no direct contact with these minerals, suggesting the important function of the contact of cellular surfaces with mineral particles. The relative larger particles of kaolinite were loosely attached to the bacteria. This attachment made the cells easy to use the sorbed substrate and then stimulated biodegradation. For goethite, small particles were tightly bound to bacterial cells and limited the acquisition of substrate and nutrients, thereby inhibiting biodegradation. These results indicated that interfacial interaction between bacterial cells and minerals significantly affected the biodegradation of pesticides.

    Beschreibung: Although 4- tert -butylphenol (4- t -BP) is a serious aquatic pollutant, its biodegradation in aquatic environments has not been well documented. In this study, 4- t -BP was obviously and repeatedly removed from water from four different environments in the presence of Spirodela polyrrhiza , giant duckweed, but 4- t -BP persisted in the environmental waters in the absence of S. polyrrhiza . Also, 4- t -BP was not removed from autoclaved pond water with sterilized S. polyrrhiza . These results suggest that the 4- t -BP removal from the environmental waters was caused by biodegradation stimulated by the presence of S. polyrrhiza rather than by uptake by the plant. Moreover, Sphingobium fuliginis OMI capable of utilizing 4- t -BP as a sole carbon and energy source was isolated from the S. polyrrhiza rhizosphere. Strain OMI degraded 4- t -BP via a meta -cleavage pathway, and also degraded a broad range of alkylphenols with linear or branched alkyl side chains containing two to nine carbon atoms. Root exudates of S. polyrrhiza stimulated 4- t -BP degradation and cell growth of strain OMI. Thus, the stimulating effects of S. polyrrhiza root exudates on 4- t -BP-degrading bacteria might have contributed to 4- t -BP removal in the environmental waters with S. polyrrhiza . These results demonstrate that the S. polyrrhiza –bacteria association may be applicable to the removal of highly persistent 4- t -BP from wastewaters or polluted aquatic environments.

    Beschreibung: The biodegradation of heptadecane in five sand columns was modeled using a multiplicative Monod approach. Each column contained 1.0 kg of sand and 2 g of heptadecane, and was supplied with an artificial seawater solution containing nutrients at a flow rate that resulted in unsaturated flow through the column. All nutrients were provided in excess with the exception of nitrate whose influent concentration was 0.1, 0.5, 1.0, 2.5, or 5.0 mg N/L. The experiment was run around 912 h until no measurable oxygen consumption or CO 2 production was observed. The residual mass of heptadecane was measured at the end of the experiments and the biodegradation was monitored based on oxygen consumption and CO 2 production. Biodegradation kinetic parameters were estimated by fitting the model to experimental data of oxygen, CO 2 , and residual mass of heptadecane obtained from the two columns having influent nitrate–N concentration of 0.5 and 2.5 mg/L. Noting that the oxygen and CO 2 measurements leveled off at around 450 h, we fitted the model to these data for that range. The estimated parameters fell in within the range reported in the literature. In particular, the half-saturation constant for nitrate utilization,  $ K_{\text{N}} $ , was estimated to be 0.45 mg N/L, and the yield coefficient was found to be 0.15 mg biomass/mg heptadecane. Using these values, the rest of experimental data from the five columns was predicted, and the model agreed with the observations. There were some consistent discrepancies at large times between the model simulation and observed data in the cases with higher nitrate concentration. One plausible explanation for these differences could be limitation of biodegradation by reduction of the heptadecane–water interfacial area in these columns while the model uses a constant interfacial area.

    Beschreibung: Leaf litter may be an important source of nutrients to stormwater and ultimately contribute to eutrophication of surface waters associated with urbanization. Thus, understanding decomposition and nutrient release from leaf litter that falls on impervious surfaces is important for stormwater management. However, few studies have examined leaf litter decomposition in the unique urban environment of the street gutter. We compared decomposition of leaf litter of five street tree species in a parking lot gutter in St. Paul, Minnesota, USA. In contrast to our expectations, comparisons with past studies revealed that litter decomposed more rapidly in the gutter than in nearby natural areas. And decomposition rates were as rapid as those measured in other urban settings (forests and streams), with most species losing 80 % of their initial mass after 1 year. Litter of most species had retained more than half of its initial N and P after 1 year. However, in contrast to N, litter P dynamics largely were uncoupled from litter mass dynamics, with litter P increasing and decreasing unpredictably over the year. Short-term (24 h) laboratory studies revealed that litter had the potential to lose a high fraction of its initial P, with high variation among species (from 27 to 88 %), and a smaller fraction of its initial N (〈10 %) via leaching. Thus, street tree species may differ in their potential contributions to nutrients that are released during decomposition. Our results suggest that careful selection of street tree species and timely removal of litterfall have significant potential to reduce nutrient fluxes from streets to storm drains, particularly for P.

    Beschreibung:    White-rot fungi are a group of microorganisms capable of degrading xenobiotic compounds, such as polycyclic aromatic hydrocarbons or synthetic dyes, by means of the action of extracellular oxidative enzymes secreted during secondary metabolism. In this study, the transformation of three anti-inflammatory drugs: diclofenac, ibuprofen and naproxen were carried out by pellets of Phanerochaete chrysosporium in fed-batch bioreactors operating under continuous air supply or periodic pulsation of oxygen. The performance of the fungal reactors was steady over a 30-day treatment and the effect of oxygen pulses on the pellet morphology was evidenced. Complete elimination of diclofenac was achieved in the aerated and the oxygenated reactors, even with a fast oxidation rate in the presence of oxygen (77% after 2 h), reaching a total removal after 23 h. In the case of ibuprofen, this compound was completely oxidized under air and oxygen supply. Finally, naproxen was oxidized in the range of 77 up to 99% under both aeration conditions. These findings demonstrate that the oxidative capability of this microorganism for the anti-inflammatory drugs is not restricted to an oxygen environment, as generally accepted, since the fungal reactor was able to remove these compounds under aerated and oxygenated conditions. This result is very interesting in terms of developing viable reactors for the oxidation of target compounds as the cost of aeration can be significantly reduced.     Beschreibung:    In this work, two novel iron oxidizing bacteria (IOB), namely Gordonia sp. MZ-89 and Enterobacter sp . M01101, were isolated from sewage treatment plants and identified by biochemical and molecular methods. Then, microbially influenced corrosion (MIC) of carbon steel in the presence of these bacteria was investigated. The electrochemical techniques such as potentiodynamic polarization measurements and electrochemical impedance spectroscopy (EIS) were used to measure the corrosion rate and observe the corrosion mechanism. The results showed that the existence of these microorganisms decreased the corrosion potential and enhanced the corrosion rate. Scanning electron microscopy (SEM) images revealed the ground boundary attacks and pitting on carbon steel samples in the presence of these bacteria after polarization. Corrosion scales were identified with X-ray diffraction (XRD). It was demonstrated that these bacteria can greatly affect the crystalline phase of corrosion products that also confirmed by SEM results. It was inferred that these bacteria were responsible for the corrosion of carbon steel, especially in the form of localized corrosion.     Beschreibung:    The common grass Calamagrostis epigeions produces a large amount of dead biomass, which remain above the soil surface for many months. In this study, we determined how exposure of dead biomass above the soil affects its subsequent decomposition in soil. Collected dead standing biomass was divided in two parts, the first one (initial litter) was stored in a dark, dry place. The other part was placed in litterbags in the field. The litterbags were located in soil, on the soil surface, or hanging in the air without contact with soil but exposed to the sun and rain. After 1 year of field exposure, litter mass loss and C and N content were measured, and changes in litter chemistry were explored using NMR and thermochemolysis-GC–MS. The potential decomposability of the litter was quantified by burying the litter from the litterbags and the initial litter in soil microcosms and measuring soil respiration. Soil respiration was greater with litter that had been hanging in air than with all other kinds of litter. These finding could not be explained by changes in litter mass or C:N ratio. NMR indicated a decrease in polysaccharides relative to lignin in litter that was buried in soil but not in litter that was placed on soil surface or that was hanging in the air. Thermochemolysis indicated that the syringyl units of the litter lignin were decomposed when the litter was exposed to light. We postulate that photochemical decay of lignin increase decomposability of dead standing biomass.     Beschreibung:    Arsenic is a carcinogenic compound widely distributed in the groundwater around the world. The fate of arsenic in groundwater depends on the activity of microorganisms either by oxidizing arsenite (As III ), or by reducing arsenate (As V ). Because of the higher toxicity and mobility of As III compared to As V , microbial-catalyzed oxidation of As III to As V can lower the environmental impact of arsenic. Although aerobic As III -oxidizing bacteria are well known, anoxic oxidation of As III with nitrate as electron acceptor has also been shown to occur. In this study, three As III -oxidizing bacterial strains, Azoarcus sp. strain EC1-pb1, Azoarcus sp. strain EC3-pb1 and Diaphorobacter sp. strain MC-pb1, have been characterized. Each strain was tested for its ability to oxidize As III with four different electron acceptors, nitrate, nitrite, chlorate and oxygen. Complete As III oxidation was achieved with both nitrate and oxygen, demonstrating the novel ability of these bacterial strains to oxidize As III in either anoxic or aerobic conditions. Nitrate was only reduced to nitrite. Different electron donors were used to study their suitability in supporting nitrate reduction. Hydrogen and acetate were readily utilized by all the cultures. The flexibility of these As III -oxidizing bacteria to use oxygen and nitrate to oxidize As III as well as organic and inorganic substrates as alternative electron donors explains their presence in non-arsenic-contaminated environments. The findings suggest that at least some As III -oxidizing bacteria are flexible with respect to electron-acceptors and electron-donors and that they are potentially widespread in low arsenic concentration environments.     Beschreibung:    To reduce the volume of seaweed wastes and extract polysaccharides, seaweed-degrading bacteria were isolated from drifting macroalgae harvested along the coast of Toyama Bay, Japan. Sixty-four bacterial isolates were capable of degrading “Wakame” ( Undaria pinnatifida ) thallus fragments into single cell detritus (SCD) particles. Amongst these, strain 6532A was the most active degrader of thallus fragments, and was capable of degrading thallus fragments to SCD particles within a day. Although the sequence similarity of the 16S rRNA gene of strain 6532A was 100% similar to that of Microbulbifer elongatus JAMB-A7, several distinct differences were observed between strains, including motility, morphology, and utilization of d -arabinose and gelatin. Consequently, strain 6532A was classified as a new Microbulbifer strain, and was designated Microbulbifer sp. 6532A. Strain 6532A was capable of degrading both alginate and cellulose in the culture medium, zymogram analysis of which revealed the presence of multiple alginate lyases and cellulases. To the best of our knowledge, this is the first study to directly demonstrate the existence of these enzymes in Microbulbifer species. Shotgun cloning and sequencing of the alginate lyase gene in 6532A revealed a 1,074-bp open reading frame, which was designated algMsp . The reading frame encoded a PL family seven enzyme composed of 358 amino acids (38,181 Da). With a similarity of 74.2%, the deduced amino acid sequence was most similar to a Saccharophagus enzyme ( alg 7C ). These findings suggest that algMsp in strain 6532A is a novel alginate lyase gene.     Beschreibung:    The performance of an Arthrobacter viscosus culture to remove diethylketone from aqueous solutions was evaluated. The effect of initial concentration of diethylketone on the growth of the bacteria was evaluated for the range of concentration between 0 and 4.8 g/l, aiming to evaluate a possible toxicological effect. The maximum specific growth rate achieved is 0.221 h −1 at 1.6 g/l of initial diethylketone concentration, suggesting that for higher concentrations an inhibitory effect on the growth occurs. The removal percentages obtained were approximately 88%, for all the initial concentrations tested. The kinetic parameters were estimated using four growth kinetic models for biodegradation of organic compounds available in the literature. The experimental data found is well fitted by the Haldane model ( R 2  = 1) as compared to Monod model ( R 2  = 0.99), Powell ( R 2  = 0.82) and Loung model ( R 2  = 0.95). The biodegradation of diethylketone using concentrated biomass was studied for an initial diethylketone concentration ranging from 0.8–3.9 g/l in a batch with recirculation mode of operation. The biodegradation rate found followed the pseudo-second order kinetics and the resulting kinetic parameters are reported. The removal percentages obtained were approximately 100%, for all the initial concentrations tested, suggesting that the increment on the biomass concentration allows better results in terms of removal of diethylketone. This study showed that these bacteria are very effective for the removal of diethylketone from aqueous solutions.     Beschreibung:    Trichloroethylene (TCE) is extensively used in commercial applications, despite its risk to human health via soil and groundwater contamination. The stability of TCE, which is a useful characteristic for commercial application, makes it difficult to remove it from the environment. Numerous studies have demonstrated that TCE can be effectively removed from the environment using bioremediation. Pseudomonas putida F1 is capable of degrading TCE into less hazardous byproducts via the toluene dioxygenase pathway (TOD). Unfortunately, these bioremediation systems are not self-sustaining, as the degradation capacity declines over time. Fortunately, the replacement of metabolic co-factors is sufficient in many cases to maintain effective TCE degradation. Thus, monitoring systems must be developed to predict when TCE degradation rates are likely to decline. Herein, we show evidence that tod expression levels correlate with the ability of P. putida F1 to metabolize TCE in the presence of toluene. Furthermore, the presence of toluene improves the replication of P . putida F1, even when TCE is present at high concentration. These findings may be applied to real world applications to decide when the bioremediation system requires supplementation with aromatic substrates, in order to maintain maximum TCE removal capacity.     Beschreibung:    Molasses melanoidin (MM) is a major pollutant in biomethanated distillery spent wash (BMDS) due to its recalcitrant properties. The 75% colour and 71% COD of MM (1,000 ppm) were reduced with developed bacterial consortium comprising Proteus mirabilis (IITRM5; FJ581028), Bacillus sp. (IITRM7; FJ581030), Raoultella planticola (IITRM15; GU329705) and Enterobacter sakazakii (IITRM16, FJ581031) in the ratio of 4:3:2:1 within 10 days at optimized nutrient. Bacterial consortium showed manganese peroxidase and laccase activity during MM decolourisation. The dominant growth of R . planticola and E . sakazakii was noted in consortium during MM decolourisation. The comparative GC–MS analysis of extracted compounds of control and degraded samples showed that most of the compounds present in control were completely utilized by bacterial consortium along with production of some metabolites. The developed bacterial consortium could be a tool for the decolourisation and degradation of melanoidin containing BMDS.     Beschreibung:    The capacity of an anaerobic sediment to achieve the simultaneous biodegradation of phenol and carbon tetrachloride (CT) was evaluated, using humic acids (HA) as redox mediator. The presence of HA in sediment incubations increased the rate of biodegradation of phenol and the rate of dehalogenation (2.5-fold) of CT compared to controls lacking HA. Further experiments revealed that the electron-accepting capacity of HA derived from different organic-rich environments was not associated with their reducing capacity to achieve CT dechlorination. The collected kinetic data suggest that the reduction of CT by reduced HA was the rate-limiting step during the simultaneous biodegradation of phenol and CT. To our knowledge, the present study constitutes the first demonstration of the simultaneous biodegradation of two priority pollutants mediated by HA.     Beschreibung:    In order to build competence for sustainability analysis and assessment of urban systems, it is seen as essential to build on models representing urban form, landuse and transportation, urban metabolism, as well as ecological processes. This type of analysis of interacting sub-systems requires an advanced model integration platform, yet open for learning and for further development. Moreover, since the aim is to increase urban experience with ecosystem management in the wide sense, the platform needs to be open and easily available, with high visualisation capacity. For this purpose, the LEAM model was applied to the Stockholm Region and two potential future scenarios were developed, resulting from alternative policies. The scenarios differed widely and the dense urban development of Scenario Compact could be visualised, destroying much of the Greenstructure of Stockholm, while Scenario Urban Nature steered the development more to outer suburbs and some sprawl. For demonstration of the need for further development of biodiversity assessment models, a network model tied to a prioritised ecological profile was applied and altered by the scenarios. It could be shown that the Greenstructure did not support this profile very well. Thus, there is a need for dynamic models for negotiations, finding alternative solutions and interacting with other models. The LEAM Stockholm case study is planned to be further developed, to interact with more advanced transport and land use models, as well as analysing energy systems and urban water issues. This will enable integrated sustainability analysis and assessment of complex urban systems, for integration in the planning process in Stockholm as well as for comparative sustainability studies between different cities, with the goal to build more sustainable urban systems and to increase urban experiences in ecosystem management.     Beschreibung:    Urban marine habitats are often utilized by wildlife for foraging and other activities despite surrounding anthropogenic impact or disturbance. However little is known of the ecological factors that determine habitat value of these and other remnant natural habitats. We examined the preferential use of urban marine habitats in a northeast US estuary to try to elucidate the factors driving enhanced foraging activity at these sites. Using a bioenergetic model, we compared energy intake to energy expenditure and examined differences in behavior and foraging success of great egrets Ardea alba at three urban and three rural salt marshes in Narragansett Bay, Rhode Island USA. Mean per site available nekton energy averaged 4.44 ± 0.97 GJ site −1 and was significantly higher at urban than at rural sites. While energy expenditure by birds was similar across all sites, mean strike and prey capture rate were significantly greater at urban sites, and 70.1 ± 12.2 % of strikes by egrets at urban sites were successful. Egrets foraging at urban sites consumed significantly more energy (23.2 ± 6.62 W bird −1 ) than those at rural sites. Model results indicated a net energy gain by egrets foraging at urban sites, versus a net energy loss at rural sites. Our results may help explain previously observed increases in the numbers of egrets foraging at urban marine habitats, and help provide input into decisions about the extent to which these areas should be considered for restoration or protection.     Beschreibung:    Assessing in situ microbial abilities of soils to degrade pesticides is of great interest giving insight in soil filtering capability, which is a key ecosystem function limiting pollution of groundwater. Quantification of pesticide-degrading gene expression by reverse transcription quantitative PCR (RT-qPCR) was tested as a suitable indicator to monitor pesticide biodegradation performances in soil. RNA extraction protocol was optimized to enhance the yield and quality of RNA recovered from soil samples to perform RT-qPCR assays. As a model, the activity of atrazine-degrading communities was monitored using RT-qPCRs to estimate the level of expression of atzD in five agricultural soils showing different atrazine mineralization abilities. Interestingly, the relative abundance of atzD mRNA copy numbers was positively correlated to the maximum rate and to the maximal amount of atrazine mineralized. Our findings indicate that the quantification of pesticide-degrading gene expression may be suitable to assess biodegradation performance in soil and monitor natural attenuation of pesticide.     Beschreibung:    Urban lakes are often monitored and managed with limited consideration of adjacent communities. Generally, when communities are considered in relation to urban lakes, they are viewed simply as sources of pollutants. Given the inevitable interactions between an urban lake and the surrounding inhabitants, the community must be considered explicitly when assessing the ecosystem health of urban lakes, as the two entities intrinsically comprise interrelated parts of a single ecosystem. In this study, the reciprocal links between a residential community and a series of urban lakes in South East Queensland have been examined to facilitate a dynamically linked, fully integrated ecosystem health assessment of constructed urban lakes. Residents’ attitudes towards, and values derived from, a series of urban lakes were surveyed, as well as residents’ behaviours which may impact upon urban lake health. The results indicate that residents derive both tangible and intangible benefits from the urban lakes, but feel little responsibility for lake health or custodianship over the lakes. Greater recognition within urban lake management frameworks of the links between urban lake systems and their surrounding communities may help to foster and enhance both community well-being, a greater sense of custodianship for such systems and improved management.     Beschreibung:    The concept of ecosystem goods and services is increasingly used to describe how biodiversity and ecosystems are linked to human well-being and that it should be placed at the core of sustainable urban development. Predictions of a tremendous future increase of urbanization in Africa necessitate an investigation into the research on ecosystem goods and services in the urban green infrastructure of Africa. Ecosystem goods and services (ES) are described as the benefits humans derive directly or indirectly from ecosystem functions and are classified as supporting, provisioning, regulating and cultural services. A literature study focusing on journal papers was conducted. Additionally a case study based on two masters studies was further refined. ES studies in African cities are biased towards South Africa and include assessments and economic valuations in which several different methods were used to determine direct consumptive and non-consumptive and indirect use values. Emphasis was placed on the multifunctional nature of ES. The main objectives of these studies were to sensitize policy makers, planners and the general public about the importance of biodiversity and ES. Ecosystem-based adaptation is discussed as the most appropriate approach in terms of applying knowledge about ES and their values in African cities as many residents still rely on ES from natural areas, but the major ecological, economic and political challenges are acknowledged. A case study focusing on domestic gardens (private green spaces) have indicated that the demand and supply of certain ES differ along a socio-economic gradient due to poor service delivery and smaller plots in the poorer areas mainly due to the legacy of separate development of the past. Where provisioning services are mainly outsourced in cities, it was found that plant species useful as food, medicine, etc. were more frequent in the gardens of poorer residents than in those of more affluent areas. The tendency to pay more for residential properties close to public open spaces, as in Europe, could not be statistically proven in the more affluent areas of a South African city, although the property values in proximity of public open spaces in some of the areas studied were lower than further away.     Beschreibung:    One of the most effective ways to describe the successional gradient of an environment is by using measures of biodiversity. We investigated the biodiversity of ant communities (known tramp/structure invaders) in an environment undergoing urban succession to characterize how ant communities changed along a disturbance gradient. Ant community dynamics were examined within three Puerto Rican housing developments of different ages. Within each development, we assessed the ant community using biodiversity measures of richness, abundance, diversity and evenness. We also studied changes in biodiversity within each development over time as well as how biodiversity related to food preferences within the ant communities. In general, the biodiversity measures increased as the age of the housing development increased. Monthly changes in biodiversity indicated that month was a better predictor of biodiversity in the most recently disturbed development, while the biodiversity in the older developments was less predictable from month to month. The biodiversity measures also suggested that the ant community in the youngest development was comprised mostly of generalist feeders. However, the older sites’ ant species appeared to have more specific feeding preferences. Based on the biodiversity measures of these ant communities, our findings suggest that the housing developments of different ages represented advancing stages of secondary succession. Furthermore, this study indicated that the succession of ant species occurs rapidly (〈10 years) in urban environments, resulting in a relatively diverse ant community.     Beschreibung:    While former salt marsh sites filled with human transported material (HTM) have altered the surface marsh ecosystem, if artificial drainage is absent, subsurface conditions may continue favorable for denitrification, a microbial process reducing nitrogen (N) export to estuaries. We used piezometer networks to evaluate the in situ groundwater denitrification capacity and nitrous oxide (N 2 O) flux (with 15 N-enriched nitrate-N via the push-pull method) in four former fringing salt marshes topped by HTM along the Rhode Island coast, U.S.A. Groundwater at these sites commonly interacted with the buried marsh horizon and the HTM. In situ groundwater denitrification capacity site means ranged from 15.2 to 71.7 μg N kg −1 d −1 with no significant differences between sites due to high intrasite variability. The site with the highest and most consistent denitrification capacity also had HTM of the finest texture and highest soluble organic carbon. Three of four sites had minimal N 2 O flux [mean N 2 O:(N 2 O + N 2 ) = 0.082] while the final site had N 2 O generation rates up to 52.5 μg N kg −1  d −1 . The site with the highest N 2 O contributions also had the lowest ambient groundwater nitrate-N indicating lack of priming for N 2 O reduction to N 2 . Former salt marshes with HTM deposits may still have the capacity for substantial groundwater denitrification capacity, similar to that observed in undisturbed salt marshes, but may also contribute substantially to global N 2 O emissions. For both salt marsh restoration and greenhouse gas mitigation efforts, attention should be given to ensuring that a tidally-driven, fluctuating water table regularly intercepts the buried organic horizons of the filled salt marsh.     Beschreibung:    Plasmid-mediated bioaugmentation was demonstrated using sequencing batch reactors (SBRs) for enhancing 2,4-dichlorophenoxyacetic acid (2,4-D) removal by introducing Cupriavidus necator JMP134 and Escherichia coli HB101 harboring 2,4-D-degrading plasmid pJP4. C. necator JMP134(pJP4) can mineralize and grow on 2,4-D, while E. coli HB101(pJP4) cannot assimilate 2,4-D because it lacks the chromosomal genes to degrade the intermediates. The SBR with C. necator JMP134(pJP4) showed 100 % removal against 200 mg/l of 2,4-D just after its introduction, after which 2,4-D removal dropped to 0 % on day 7 with the decline in viability of the introduced strain. The SBR with E. coli HB101(pJP4) showed low 2,4-D removal, i.e., below 10 %, until day 7. Transconjugant strains of Pseudomonas and Achromobacter isolated on day 7 could not grow on 2,4-D. Both SBRs started removing 2,4-D at 100 % after day 16 with the appearance of 2,4-D-degrading transconjugants belonging to Achromobacter , Burkholderia , Cupriavidus , and Pandoraea . After the influent 2,4-D concentration was increased to 500 mg/l on day 65, the SBR with E. coli HB101(pJP4) maintained stable 2,4-D removal of more than 95 %. Although the SBR with C. necator JMP134(pJP4) showed a temporal depression of 2,4-D removal of 65 % on day 76, almost 100 % removal was achieved thereafter. During this period, transconjugants isolated from both SBRs were mainly Achromobacter with high 2,4-D-degrading capability. In conclusion, plasmid-mediated bioaugmentation can enhance the degradation capability of activated sludge regardless of the survival of introduced strains and their 2,4-D degradation capacity.
