In contrast to humans, prolactin is luteotropic in rats, i.e. it promotes progesterone production in the corpus luteum after ovulation and maintains gestation (Neumann, 1991; Ben-Jonathan et al., 2008). Progesterone antagonizes the estrogenic stimulation of uterine growth by e.g. inhibiting estrogen receptor alpha (ERα) chromatin binding and non-transcriptional activity (Kim et al., 2013).

The causal relationship between sustained reduction in Prl, altered progesterone production and eventual promotion of uterine adenocarcinomas late in the life is a well-studied process in rats (Neumann, 1991). Additional confidence that this causal relationship is present at high doses of Afidopyropen is provided by the characteristic lesion signature induced by hypoprolactinemia (i.e., increased incidence of uterine adenocarcinomas and reduced incidence of mammary neoplasia) (Harleman et al., 2012).

2017, Toxicology LettersCitation Excerpt :Administration of a dopamine agonist reverses this situation, lowers circulating prolactin levels promoting luteolysis and a return to cyclicity, which increases exposure to periods of estrogen dominance (proestrus/estrus). An increased exposure to periods of endogenous estrogen dominance associated with chronic administration of a dopamine agonist promotes an increased incidence of uterine tumors in the rat (Neumann, 1991; Gopinath, 1999). This manuscript details our investigation of the potential for chronic exposure to 700 ppm D4 and 160 ppm D5 to affect estrous cyclicity and other markers of reproductive senescence in aging female F344 rats.

Moreover, Martin et al. (1998) found that persistently high LH and T concentrations might increase testicular seminoma risk.
