Abstract

Nitrogen-dried smears ofEhrlich ascites tumor cells are stained with the tetrazolium saltMTT for succinodehydrogenase activity.

The intensity of the staining is measured cytospectrometrically by scanning the single cells.

The statistic distribution of the total extinctions of the single cells is analysed and shown graphically. Thus it is possible to obtain basic values for quantitative comparisons.

As an example for application, the strong inactivation of SDH by 4-Hydroxypentenal is presented.

Zusammenfassung

Im Stickstoffstrom getrocknete Ausstriche vonEhrlich-Ascites-Tumorzellen werden mit dem TetrazoliumsalzMTT auf SDH-Aktivität gefärbt und die Intensität der Färbung cytospektrometrisch durch mäanderförmiges Absuchen der Einzelzellen gemessen.

Die statistische Verteilung der erhaltenen Gesamtextinktionen wird analysiert und graphisch dargestellt.

Dadurch ist es möglich, Bezugswerte für quantitative Vergleiche zu erhalten.

Als Anwendungsbeispiel dient die Inaktivierung der SDH durch 4-Hydroxypentenal.
