Pupae of the greater wax moth,Galleria mellonella L., were exposed to x-rays with different doses, to a certain wounding, or to both treatments in different combinations. The extent of damage was measured as the decrease of the frequency of ecdysis.

The factors of damage after the different treatments were analyzed by partial irradiation and histological methods. The results of the histological investigation were compared with normal development of important organs of theGalleria pupa, particularly of the ecdysial glands and the corpora allata.

The resistance to x-rays of theGalleria pupa increases about twentyfold during the seven days of the pupal instar. In a given pupal age and after a given dose the radiation damage depends on a direct effect on differentiable tissues and the radiation-induced inability of the hormonal system to repair such a damage.

Wounding evokes a systemic response of the pupa. First this results — as indicated by histological examination of the hormonal glands — from an increased amount of juvenile hormone in relation to ecdyson. Secondly the wound possibly causes an excessive release of neurosecretion in the sense of a stress reaction. The response to wounding becomes maximal on the third and sixth day of the pupal instar. The first maximum of sensitivity on the third pupal day is thought to be correlated-with a critical event on the fourth pupal day, namely the withdrawing of the epidermis from the pupal cuticle. The second maximum of sensitivity on the sixth pupal day coincides with the second maximum of activity of the corpora allata at the end of the fifth pupal day.

A combination damage is always followed by a relatively large decrease of the ecdysial frequency, independent of the effects of the single injuries. The intervals between and the sequence of the treatments have no influence. The combination damage is particularly an amplification of the effect of wounding caused by radiation.

The activity of the corpora allata on the first two days of the pupal instar simultaneously with the ecdysial glands leads to renewed discussion of the “classical scheme”, in which the relation of ecdyson and juvenile hormone determines the following moulting to a larval, pupal, or imaginal moult.

Puppen der großen Wachsmotte,Galleria mellonella L., sind einer Röntgenbestrahlung mit verschiedener Dosis, einer bestimmten Verwundung oder beiden Belastungen in verschiedener Kombination ausgesetzt worden. Das Ausmaß einer Schädigung ist als Herabsetzung der Schlüpfrate registriert worden.

Durch Teilbestrahlungsversuche und mit histologischen Methoden ist versucht worden, die Faktoren des Schadens nach den verschiedenen Eingriffen zu analysieren. Normale Entwicklungsvorgänge an wichtigen Organen derGalleria-Puppe, insbesondere an der Prothoraxdrüse und an den Corpora allata, sind als Vergleichsbasis zur histologischen Analyse behandelter Puppen untersucht worden.

Die Resistenz derGalleria-Puppe gegenüber Röntgenstrahlen steigt während des 7 Tage dauernden Puppenstadiums bis auf etwa das Zwanzigfache des Ausgangswertes an. Der Strahlenschaden in einem bestimmten Puppenalter und nach einer bestimmten Dosis hängt ab von einer direkten Schädigung differenzierungsfähiger Gewebe und von einer strahlenbedingten Unfähigkeit des Hormonsystems, eine solche Schädigung zu reparieren.

Eine Wundsetzung ruft eine systemische Antwort der Puppe hervor. Diese kommt erstens — nach dem histologischen Befund an den Hormondrüsen — daher, daß in einem bestimmten Alter das Verhältnis zwischen Ekdyson und Juvenilhormon zugunsten des letzteren verschoben wird. Zweitens bewirkt die Wunde möglicherweise eine exzessive Ausschüttung von Neurosekret im Sinne einer Stressreaktion. Die Empfindlichkeit der Puppe gegenüber einer Wunde ist am III. Puppentag und am VI. Puppentag am größten. Das erste Empfindlichkeitsmaximum am III. Puppentag wird mit einer kritischen Periode am IV. Puppentag, nämlich dem Abheben der Epidermis von der Puppenkutikula, in Zusammenhang gebracht, und das zweite Empfindlichkeitsmaximum am VI. Puppentag mit einem zweiten Aktivitätsmaximum der Corpora allata am Ende des V. Puppentags.

Ein Kombinationsschaden äußert sich unabhängig von der Summe der Einzelbelastungen immer in einer relativ starken Verminderung der Schlüpfrate. Der zeitliche Abstand und die Reihenfolge der Belastungen sind dabei weitgehend ohne Einfluß. Der Kombinationsschaden stellt vor allem eine strahlenbedingte Verstärkung des Wundschadens dar.

Der Befund, daß an den ersten beiden Puppentagen die Corpora allata gleichzeitig mit der Prothoraxdrüse aktiv sind, gibt Anlaß, das „klassische Schema“, wonach durch das Verhältnis von Ekdyson zu Juvenilhormon bestimmt wird, ob die folgende Häutung zu einer Larve, einer Puppe oder einer Imago führt, erneut zu diskutieren.
