Histochemical investigations on sialic acid containing compounds in the CNS of teleosts

Summary

By means of histochemical experiments the qualitative (microphotographs) and quantitative (cytophotometry) distribution of sialic acid containing compounds was investigated in the mesencephalon and cerebellum of Carassius carassius.

  1. 1.

    The perikarya of nerve cells are especially stained by means of Azur A.

  2. 2.

    Nerve fibres, on the other hand, showed positive reactions with colloidal ironhydroxyd (CIH).

  3. 3.

    In different structures of the CNS the intensity of CIH-staining is decreased up to about 55% following treatment with neuraminidase.

  4. 4.

    There are no differences in the reactions of 7°C as compared to 20°C adapted animals.

Zusammenfassung

Am Mesencephalon und Cerebellum von Karauschen (Carassius carassius) wurden qualitative (mikroskopische Aufnahmen) und quantitative (Extinktionsmessungen mit Hilfe des Cytophotometers) Untersuchungen zur histochemischen Lokalisation von sialinsäurehaltigen Verbindungen durchgeführt:

  1. 1.

    Mit Hilfe von Azur A lassen sich NANA-haltige Verbindungen besonders im Bereich von Nervenzell-Perikaryen darstellen.

  2. 2.

    Die Eisenbindungsreaktion (EBR) wirkt dagegen insbesondere auf das Nervenfasermaterial positiv.

  3. 3.

    In verschiedenen Hirnstrukturen wurde nach Neuraminidase-Behandlung und anschließender EBR ein Extinktionsverlust von 55% festgestellt und so die Spezifität der Reaktion zur Darstellung von Sialinsäure aufgezeigt.

  4. 4.

    Zwischen den Hirnen von Fischen, die an Wassertemperaturen von 7°C gegenüber 20°C adaptiert waren, ergaben sich keine histochemisch erfaßbaren NANA-Unterschiede.
