Next, the glutamate levels in these samples were analysed using high-performance liquid chromatography (HPLC) coupled to a spectrofluorometric detector (Beckman, USA) with a fixed excitation wavelength set to 340 nm and a fixed emission wavelength set to 460 nm [23]. Sodium-binding benzofuran isophthalate acetoxymethyl ester (SBFI-AM, Life Technologies) was employed as a sodium indicator to measure [Na+]i. Briefly, the cells were loaded with 2 μM of SBFI/AM and 0.02% Pluronic F-127 (Life Technologies) for 40 min at room temperature in the dark.

At 12 h post-TBI, the glutamate level in CSF was assayed by high-performance liquid chromatography (HPLC, Beckman Company, Berkeley, CA, USA) as previously reported [Begley et al., 1994]. CSF separation was achieved on a C18 column (Supelcosil, USA) with the mobile phase composed of a mixture of methanol, sodium acetate buffer (pH 6.5; 0.1 M), and tetrahydrofuran (95:900: 5, v/v/v), at a flow rate of 1.0 mL/min and detected by a 157 spectrofluorometric detector (Beckman Company) with the excitation wavelength set at 330 nm and the emission wavelength was fixed at 450 nm.
