2013, Small Ruminant ResearchCitation Excerpt :The final pellet was resuspended in the second preparation buffer and stored at −80 °C. Whole membrane fractions were prepared as already described (Schröder et al., 1990) with slight modifications as described elsewhere (Muscher et al., 2008) for caprine kidney cortex. Whole cell lysates were prepared by homogenising the tissue in preparation buffer containing 50 mmol/L HEPES, 4 mmol/L EGTA, 10 mmol/L EDTA, 0.1% Triton X-100, 100 mmol/L β-glycerophosphate, 15 mmol/L pyrophosphate, 15 mmol/L sodium orthovanadate, 5 mmol/L sodium fluoride, protease and phosphatase inhibitors (Complete mini, Roche, Mannheim, Germany), pH 7.4.
