Charge isomers of urinary bikunin (trypsin inhibitor)

Abstract

It was observed that the purified urinary bikunin (trypsin inhibitor) consisted of four major isomers with different electric charges which could be separated by HPLC using a Mono Q column. These isomers revealed the same antitrypsin activity and did not show any differences in the apparent molecular weight by SDS-PAGE, amino-acid composition, N-terminal amino-acid sequence (1–40) and C-terminal amino acid (Leu). The contents of sialic acid and uronic acid were also identical among these isomers. However, analysis of chondroitin sulfate revealed all the glycosaminoglycan chains of these isomers were undersulfated, comprising nonsulfated and 4-sulfated disaccharide units, and 4-sulfated disaccharide unit ratio varied among these isomers. After the chondroitin ABC lyase digestion, all the isomers were eluted at the same position on a Mono Q column chromatography. These results indicated that charge isomers of urinary bikunin was attributed to the difference on sulfation ratio in a glycosaminoglycan chain.

      The core protein of UTI consists of 143 amino acids and has a single low-sulfated chondroitin 4-sulfate (Ch4S) chain covalently linked to the residue Ser-10 as a glycosaminoglycan (GAG) moiety through a GlcUAβ1-3Galβ1-3Galβ1-4Xylβ1-Ser (GlcUA-Gal-Gal-Xyl-Ser) or GlcUA-Gal(4-sulfate)-Gal-Xyl-Ser linkage region.5–7 Clustered sulfated units (Ch4S disaccharides) are located at the reducing terminus of the GAG chain linked to the linkage region, while clustered non-sulfated units (chondroitin disaccharides, Ch) are located at the non-reducing terminus, and the number of both units can vary in UTI.8–10 UTI has been used as an anti-inflammatory medicine for pancreatitis, threatened premature delivery, and other conditions based on the protease inhibitor activity of its core protein.11

      These data agree with fraction 2 analyzed in this study that eluted at salt concentrations higher than 0.5 m NaCl. Approximately 30 isomers of urinary bikunin have been isolated with differences in the degree of CS chain sulfation and length (23, 47). Separation of urinary bikunin in this study using anion exchange chromatography and subsequent analysis of the disaccharide composition of the CS chains supported the presence of isomers.
