Preparation of R-type lipopolysaccharides ofAcinetobacter calcoaceticus by EDTA-salt extraction

Abstract

An EDTA-salt extraction method is presented for the preparation of R-type LPS fromAcinetobacter calcoaceticus. Since the cell extract is free of nucleic acids and contains only few protein contaminations, it is easily purified by Sephadex G-200 chromatography and CaCl2 precipitation. Yield and chemical composition are similar to the LPS prepared by the phenol-chloroform-light petroleum method of Brade and Galanos [Eur J Biochem 122:233–237]. Significant differences were found in the fatty acid composition between both preparations, mainly with respect to the proportions of unsaturated fatty acids and chain length. This finding may be attributed to heterogeneities demonstrated within both R-type LPS isolated.
