The total amount of nucleic acids as well as DNA and RNA were analysed biochemically in embryos of the troutsS. irideus andS. trutta fario from cleavage to early differentiation stages. In the same developmental stages ribonucleoprotein (RNA and basic protein) was investigated cytochemically at different embryonic regions. The phase of gastrulation was of main interest.

The results of both methods show that the quantity of RNA and ribonucleoprotein increases beginning with themorula stage. The maximum is reached in theblastula.

When during the blastoderm stage RNA and basic protein are stained histochemically a characteristic gradient of intensity can be determined. From the increase of RNA and basic protein in periblast and blastoderm and from the gradual staining effect itself one can conclude that RNA and basic protein is transported from the yolk through the periblast into the embryo. There should exist atransfer of substances possibly as ribonucleoprotein particles. The limited time of this transfer during early embryogenesis shows that this phenomenon is not the result of energetic yolk utilization but it is connected with the following differentiation processes.

After injection of actinomycin D into the blastocoel development is stopped at the beginning ofgastrulation. Therefore a new synthesis of messenger RNA should be necessary for further development.

The ribosomal RNA is constant duringgastrulation but the amount of basic protein decreases in this time. During neurulation and the beginning differentiation phase a new increase of RNA takes place but now the quantity of basic protein does not change.

In contrast to investigations of the Urodele development in the trout embryo no change of RNA and basic protein is found during gastrulation in the invaginating material and the covering neuroectoderm. Therefore no transfer of substances takes place between the two blastemas. There are many differences in the embryonic development of Urodela and Teleosts. Nevertheless in both classesa relation between the yolk system and the embryonic blastema results in the beginning of thedifferentiation processes. This results point to the possible morphogenetic importance of the yolk system.

Bei den Keimen der ForellenSalmo irideus undSalmo trutta fario wurden in den Furchungsstadien bis zum Ende der frühen Differenzierungsphase (vollendete Dotterumwachsung) die Gesamtnucleinsäuren sowie DNS und RNS separat biochemisch bestimmt. Mit quantitativen cytochemischen Methoden wurde im gleichen Zeitraum der Ribonucleoproteid-Status (RNS und basisches Protein) in den verschiedenen Keimbereichen untersucht. Besondere Berücksichtigung fand dabei die Phase der Gastrulation.

Mit beiden Methoden lä\t sich vomMorulastadium an eine Vermehrung von RNS bzw. Ribonucleoproteid nachweisen, wobei das Maximum in derBlastula erreicht wird.

Aus der Existenz eines charakteristischen histochemischen Färbegradienten im Blastoderm wird geschlossen, da\ dieser Anstieg auf einemStofftransfer beruht, indem RNS und basisches Protein (möglicherweise in Form von Ribonucleoproteid-Partikeln) aus dem Dotter über das Dottersyncytium in den Keim transportiert werden. Der Zeitraum dieses Stofftransfers sowie dessen zeitlich relativ enge Begrenzung deuten an, da\ dieses Phänomen nicht Ausdruck der energetischen Dotternutzung ist, sondern in Beziehung zu den bevorstehenden Differenzierungsprozessen steht.

Nach Injektion von Actinomycin D in die Subgerminalhöhle kommt die Entwicklung bei Beginn derGastrulation zum Stillstand. Zu diesem Zeitpunkt wäre demnach eine Neusynthese von messenger-RNS erforderlich.

Die ribosomale RNS bleibt während der Gastrulation konstant. Dagegen nimmt das basische Protein im gleichen Zeitraum ab. Erst während der Neurulation und der beginnenden Differenzierungsphase erfolgt eine weitere Zunahme der RNS, während das basische Protein keiner weiteren quantitativen Veränderung unterliegt.

Während der Gastrulation lä\t sich (im Gegensatz zu den bei Urodelen nachgewiesenen Vorgängen) weder im invaginierenden Material noch im darüberliegenden Neuroektoderm irgendeine quantitative Veränderung im Ribonucleoproteid-Status und damit auchkein Stofftransfer zwischen diesen beiden Gewebesystemen nachweisen. Gewisse Parallelen bei diesen entwicklungsgeschichtlich so verschiedenen Wirbeltierklassen scheinen jedoch durch die in beiden Fällen angedeutetenBeziehungen des Dottersystems zu denDifferenzierungs-prozessen gegeben zu sein. Die Vermutung, da\ dem Dotter auch eine morphogenetische Bedeutung zukommt, erfährt damit eine weitere Stütze.
