Production of steroid-ring-B-seco acid from β-sitosterol and influence of surfactants on sterol uptake in Nocardia sp. M 29–40

Summary

The effect of several parameters (temperature; pH; main carbon source; time and amount of β-sitosterol addition; Tween 20, 40, 60, 80; Span 20; pluronic F 68, L 64) on the conversion of β-sitosterol to 3-(5α-hydroxy-7aβ-methyl-1-oxo-3aα-H-hexahydroindan-4α-yl) propionic acid (I) by Nocardia sp. M. 29–40 was investigated. A maximal theoretical yield of 65 mol% I (with respect to substrate added) could be achieved during cultivation at pH 8.0 in presence of 6 g/l Tween 40 or Tween 60. Tween 40 and Tween 60 stimulate β-sitosterol cooxidation not by improving the substrate suspension but by providing a fatty acid component as precursor for biosynthesis of surface active cell wall lipids.
