An in situ fluorimetric method for the determination of Sevin and α-naphthol on thin-layer chromatograms

Zusammenfassung

Die Substanzen werden extrahiert, auf Silicagel-Dünnschichten getrennt und mit Natronlauge besprüht, wodurch sie fluorescierend werden. Sevin wird dabei in α-Naphthol umgewandelt. Die Fluorescenz der beiden Flecke wird bei 488 nm gemessen (Anregung: 338 nm). Die Nachweisgrenze liegt visuell bei 0,006 μg je Fleck, bei instrumenteller Messung bei 0,001 μg. Reproduzierbarkeit und Genauigkeit des Verfahrens sind der Gas-Chromatographie und anderen Methoden vergleichbar.

Summary

A fluorimetric in situ method is described for the determination of Sevin and α-naphthol. The compounds are extracted, separated on silicagel-gel thin layers and sprayed with NaOH solution to render them fluorescent. Sevin is converted to α-naphthol by this treatment. The fluorescence of both spots is measured at 488 nm (excitation: 338 nm). Visual detection limit is 0.006 μg/spot, by instrumental measurement it is 0.001 μg/spot. Reproducibility and accuracy are comparable to gas chromatography and other methods.
