I. Mitteilung Die systematische Stellung und das Farbstoffbildungsvermögen des Pilzes

Zusammenfassung

Der als Allescheria boydii Shear, 1922, Stamm 1699 bekannte biotinheterotrophe Pilz wird morphologisch und physiologisch von der Gattung Allescheria abgegrenzt und als Cephalosporium gordoni nov. spec. beschrieben.

Die Fähigkeit des Pilzes zur Farbstoffsynthese erweist sich von verschiedenen Faktoren abhängig, insbesondere von der Kohlenstoffquelle, demPh-Wert und dem Cu++- Gehalt des Nährsubstrates. Als optimal für die Farbstoffbildung gelten Monosaccharide, ein Anfangs-ph von 4,5 bis 5,0 und 0,2 mg-% Cu++, d.i. die 100 fache Menge des in der normalen synthetischen Nährlösung enthaltenen Kupfers.

Einen Einblick in das fermentative Geschehen geben Versuche zur kompetitiven Hemmung der Pigmentsynthese durch Mononitrophenole. Unter bestimmten Bedingungen blockieren diese nicht nur die Farbstoffsynthese, sondern führen gleichzeitig zu einem nicht-autolytischen Abbau des Mycels. Die dadurch bedingte negative Stoffbilanz wird als Folge einer entkoppelten oxydativen Phosphorylierung gedeutet.

Die Hemmung der Pigmentsynthese ist nicht an die para-Stellung der Substituenten gebunden; dies läßt auf das Vorhandensein einer Polyphenoloxydase vom Laccase-Typ schließen. Es wird die Möglichkeit diskutiert, daß zwischen den die beiden Reaktionen — Farbstoffsynthese bei Allescheria boydii 1699 und oxydativer Phosphorylierung —katalysierenden Fermenten strukturchemische Ähnlichkeiten bestehen.
