Gehaltebepaling van hydrocortisonacetaat in zalf met behulp van transmissiedensitometrie

Quantitative TLC analysis of hydrocortison acetate

Abstract

Spraying of a chromatogram in order to colour TLC-spots is no impediment for precise quantitative‘in situ’ analysis. However, therefore it is necessary that the spots are thoroughly coloured, as can be realised by repeated spraying with a dilute reagens. A residual standard deviation of 0.2μg is obtained for 0.4μg spots of hydrocortison acetate charred withp-toluene sulfonic acid.

Samenvatting

De kleuring van een chromatogram door middel van sproeien hoeft geen beletsel te zijn voor nauwkeurige kwantitatieve ‘in situ’-bepalingen op de dunnelaag-plaat. Van belang is echter dat de vlekken volledig zijn gekleurd. Dit kan worden gerealiseerd door enige malen te sproeien met verdund reagens. Voor vlekken van 0,4μg hydrocortisonacetaat, verkoold metp-tolueensulfonzuur, is een residuele standaardafwijking van 0,02μg gevonden.
