Recently, a compound derived from recent scientific advances named 34 has emerged as the focus of this research, the aim of which is to explore its potential impact on solid tumor cell lines. Using a combination of bioinformatics and biological assays, this study conducted an in-depth investigation of the effects of 34. The results of this study have substantial implications for cancer research and treatment. 34 has shown remarkable efficacy in inhibiting the growth of several cancer cell lines, including those representing prostate carcinoma (PC3) and cervical carcinoma (HeLa). The high sensitivity of these cells, indicated by low IC50 values, underscores its potential as a promising chemotherapeutic agent. In addition, 34 has revealed the ability to induce cell cycle arrest, particularly in the G2/M phase, a phenomenon with critical implications for tumor initiation and growth. By interfering with DNA replication in cancer cells, 34 has shown the capacity to trigger cell death, offering a new avenue for cancer treatment. In addition, computational analyses have identified key genes affected by 34 treatment, suggesting potential therapeutic targets. These genes are involved in critical biological processes, including cell cycle regulation, DNA replication and microtubule dynamics, all of which are central to cancer development and progression. In conclusion, this study highlights the different mechanisms of 34 that inhibit cancer cell growth and alter the cell cycle. These promising results suggest the potential for more effective and less toxic anticancer therapies. Further in vivo validation and exploration of combination therapies are critical to improve cancer treatment outcomes. Full article

Soil water and soil temperature are important ecological factors and driving forces for ecosystem restoration and sustainable development, possessing great significance for climate modeling and prediction. The Hengduan Mountains in southeastern Tibet, China, are located in a climate-change-sensitive area, and the study of soil hydrothermal dynamics in this area is of great significance for local and global climatic change and water resource utilization. This study, based on the soil hydrothermal and meteorological data of the Hengduan Mountain area in Southeast Tibet, analyzes the dynamic change patterns of soil hydrothermal and meteorological factors and explores their influencing relationships. It was found that the dynamic change in soil water content affected by precipitation was “bimodal” type. Among the meteorological factors, soil water content has the strongest correlation with relative humidity. The intra-annual variation curve of soil temperature is similar to that of the atmospheric temperature, showing a “unimodal” type, and has the highest correlation with atmospheric temperature. Specifically, it takes 70 mm and 170 mm of precipitation to change the soil water content and soil temperature at the 150 cm depth. For every 20 °C change in atmospheric temperature, soil temperature above 150 cm changes by an average of 7.2 °C. Full article

This study examined the second-order schema domains of Early Maladaptive and Adaptive Schemas based on recent trends and compared them with the five theoretical second-order schema domains commonly used in schema therapy. Using six international Eastern and Western community samples—Singapore (n = 628), Malaysia (n = 229), USA (n = 396), South Africa (n = 390), Nigeria (n = 364), India (n = 306)—confirmatory factor analysis showed that the four second-order domains of EMSs and EASs, which ran almost parallel with each other, were the most robust models calling into question the validity of the five domain model. Given the hypothesized links between schemas and needs, these four categories of EMSs and EASs represent four categories of toxic experiences and core emotional needs, respectively. These categories were supported empirically and are useful to parents as well as to clinicians as they approach child rearing and the treatment of clients in schema therapy from the vantage point of needs. These four categories of psychological core emotional needs, as well as toxic experiences, were found, as expected, to be linked with various measures of well-being and ill-being. Full article

The expression and function of podoplanin (PDPN) in the normal human placenta has been debated in placental evaluation. This study emphasizes the importance of a multimodal approach of PDPN expression in normal human placentas. A complete examination is performed using immunohistochemistry, RNAscope and automated Digital Image examination (DIA) interpretation. QuPath DIA-based analysis automatically generated the stromal and histological scores of PDPN expression for immunohistochemistry and RNAscope stains. The umbilical cord’s isolated fibroblasts and luminal structures expressed PDPN protein and PDPN_mRNA. RNAscope detected PDPN_mRNA upregulation in syncytial placental knots trophoblastic cells, but immunohistochemistry did not certify this at the protein level. The study found a significant correlation between the IHC and RNAscope H-Score (p = 0.033) and Allred Score (p = 0.05). A successful multimodal strategy for PDPN assessment in human placentas confirmed PDPN expression heterogeneity in the full-term human normal placenta and umbilical cord at the protein and mRNA level. In placental syncytial knots trophoblastic cells, PDPN showed mRNA overexpression, suggesting a potential role in placenta maturation. Full article

(1) Background: Breast cancer is the leading malignancy worldwide, and in Ghana, it has a poor overall survival rate. However, approximately 50% of cases are cases of early-stage disease, and with advances in breast cancer treatment and improvements in survival, quality of life (QOL) is becoming as important as the treatment of the disease. (2) Methodology: This was a cross-sectional study of survivors who had breast-conserving surgery (BCS), mastectomy only (M) and mastectomy with breast reconstruction (BRS) from 2016 to 2020 at a tertiary hospital in Ghana, comparatively assessing their QOL using EORTC QLQ C-30 and EORTC QLQ BR-23. (3) Results: The study participants had an overall global health status (GHS) median score of 83.3 [IQR: 66.7–91.7] with no significant differences between the surgery types. The BRS group had statistically significant lower median scores for the functional scale (82.8 and 51.0) and the highest scores for the symptomatic scale (15.7 and 16.5). Body image was significantly lowest for the BRS group (83.3) [68.8–91.7] and highest (100) [91.7–100] for the BCS group (p < 0.001). (4) Conclusion: There is a need to develop support systems tailored at improving the QOL of breast cancer survivors taking into consideration the type of surgery performed. Full article

Due to its high complexity and the varied assembly processes, hybrid assembly systems characterized by human–robot collaboration (HRC) are meaningful. Suitable use cases must be identified efficiently to ensure cost-effectiveness and successful deployment in the respective assembly systems. This paper presents a method for evaluating the potential of HRC to derive automation suitability based on existing or to-be-collected time data. This should enable a quick and favorable statement to be made about processes, for efficient application in potential analyses. The method is based on the Methods–Time–Measurement Basic System (MTM-1) procedure, widely used in the industry. This ensures good adaptability in an industrial context. It extends existing models and examines how much assembly activities and processes can be optimized by efficiently allocating between humans and robots. In the process model, the assembly processes are subdivided and analyzed with the help of the specified MTM motion time system. The suitability of the individual activities and sub-processes for automation are evaluated based on criteria derived from existing methods. Two four-field matrices were used to interpret and classify the analysis results. The process is assessed using an example product from electrolyzer production, which is currently mainly assembled by hand. To achieve high statement reliability, further work is required to classify the results comprehensively. Full article

Soccer is a sport with a high incidence of injuries. The most common injury occurs when the anterior cruciate ligament of the knee has undergone a sprain, strain, or partial or total rupture. Besides fitness factors (e.g., proprioception, balance and strength capabilities), a principal cause of ACL injuries is sport shoes and playing surfaces. Especially with the emergence of artificial surfaces (rubbers, turfs, concrete, asphalt, red turf), the ACL injuries dramatically increase. The cost of ACL injuries is high both in terms of career termination and in social and economic costs. Thus, it is necessary to understand the impact of sport shoes and playing surfaces on the ACL and how it can be mitigated. The present literature review followed the PRISMA methodology to identify the major biomechanical factors influencing the behavior of surfaces and shoes in relation to the ACL damage. Fifty-eight papers were identified. After reviewing the mechanism of injuries, we identified several distinct factors: type of outsole and mechanical arrangement of the outsoles; shear forces; characteristics of artificial turf surfaces; effect of weather on artificial surface aging and change in mechanical characteristics; outsole/ground interface; and the frictional, tractional and rotational forces in the pathogenesis of ACL injuries in soccer. Youth work is not unitary, has a diverse history, and draws upon differing traditions; however, the focus of all major youth work traditions has been anthropocentric. This approach is now challenged by young people themselves through the climate justice movement, and institutionally through the United Nations Agenda 2030 resolution, which has developed integrated Sustainable Development Goals, which aim to “balance” social, environmental, and economic imperatives. This article examines what insights can be gained from ecopedagogy that would enable youth work to fully integrate an ecological perspective alongside traditional anthropological concerns. The discussion focuses on the nexus and tensions between ecopedagogy and youth work from a theoretical perspective to draw out insights into how changes need to be made to the aims and purposes of youth work, how this might be achieved, and barriers in the current environment. This article explores operational challenges in mining, with a focus on energy management amid depleting ore grades and rising costs. The urgent need for innovative energy management systems and strategies is highlighted by analyzing the unexplored landscape of mine energy budgeting and forecasting and identifying gaps in current practices. Drawing from the literature, this paper offers new insights into energy budgeting and the evaluation of energy efficiency initiatives by integrating traditional and advanced measurement and verification (M&V) techniques. M&V practices are crucial for energy management, particularly in deep-level mines, with a focus on practical knowledge and advanced methodologies. Key findings from this study show that integrating advanced M&V techniques with unplanned events is crucial to improve the financial management of mines. By leveraging these key findings, this article proposes a roadmap of the next seven milestones needed in advanced M&V research to aid effective energy management in a mining environment. If executed successfully, a practical method for applying advanced M&V processes to deep-level mining operations can be constructed. Such a generic method will enhance mining companies’ energy efficiency initiatives and improve financial management practices on a global scale. Full article

Autoclaving Achieves pH-Neutralization, Hydrogelation, and Sterilization of Chitosan Hydrogels in One Step

Conventionally, chitosan hydrogels are acidic and contain toxic chemicals because chitosan is soluble only in acidic solvents and requires toxic additives such as chemical crosslinkers and polymerization agents to fabricate chitosan hydrogels. These properties prevent chitosan hydrogels from being used for medical applications. In this study, chitosan hydrogels were prepared by a simple and versatile process using urea hydrolysis by autoclaving (steam sterilization, 121 °C, 20 min). When autoclaved, urea hydrolyzes in an acidic chitosan aqueous solution, and ammonia is produced, which increases the pH of the solution, and chitosan becomes insoluble, leading to the formation of a chitosan hydrogel. The pH and osmotic concentration of chitosan hydrogels could be adjusted to be suitable for physiological conditions (pH: 7.0–7.5, and osmotic concentration: 276–329 mOsm/L) by changing the amount of urea added to chitosan solutions (chitosan: 2.5% (w/v), urea: 0.75–1.0% (w/v), pH: 5.5). The hydrogels had extremely low cytotoxicity without the washing process. In addition, not only pure chitosan hydrogels, but also chitosan derivative hydrogels were prepared using this method. The autoclaving technique for preparing low-toxic and wash-free sterilized chitosan hydrogels in a single step is practical for medical applications. Full article

Pearl millet [Pennisetum glaucum (L.) R. Br.] is a subtropical grain and forage crop. It is privileged with several desirable forage attributes. Nevertheless, research on pearl millet is limited, especially as a forage crop, in developing countries. Therefore, the objectives of this study were to investigate the field performance and stability of pearl millet genotypes for forage yield across seven environments. The study was conducted in seven environments (combination of locations and seasons) during the 2016/2017–2018/2019 seasons. Twenty-five pearl millet genotypes, selected based on forage yield from a core collection of 200 accessions, were arranged in an alpha lattice design with three replications. The parameters measured were fresh forage yield, days to flowering, plant height, number of culms m−2, leaf-to-stem ratio, and stem girth. The combined analysis revealed that environments, genotypes, and their interaction had significant effects on all traits studied except the genotypic effect on stem girth. Across the seven environments, four genotypes (G14, G01, G12, and G22) outyielded the check genotype in fresh matter yield by 20.7, 16.5, 11.0 and 9.8%, respectively. The additive main effects and multiplicative interaction (AMMI) analysis showed that the genotype, environment, and their interaction were highly significant (p ≤ 0.001) for fresh matter yield. The results of AMMI stability values (ASVs) and the genotype selection index (GSI) combined with the AMMI estimate-based selection showed that genotypes G14, G22 and G01 were the most stable and adapted genotypes and were superior to the check genotype. These results indicate that forage pearl millet varieties could be developed directly through evaluating the wealth of available collections or indirectly through hybridization in crop breeding programs. Non-Fasting Plasma Triglycerides Are Positively Associated with Diabetes Mortality in a Representative US Adult Population

This study aimed to investigate whether non-fasting plasma triglycerides were associated with diabetes mortality. It included 7312 US adult participants. Diabetes mortality data were obtained via the linkage to National Death Index (NDI) records. Hazard ratios of non-fasting plasma triglycerides for diabetes mortality were assessed using Cox proportional hazards models, adjusting for age, gender, ethnicity, obesity, poverty–income ratio, education levels, physical activity, alcohol consumption, cigarette smoking status, survey period, hypercholesterolemia, hypertension, diabetes, and family history of diabetes. Among these participants, 1180 had diabetes. A total of 420 diabetes-caused deaths were recorded during a mean follow-up of 16.8 years. A 1-natural-log-unit increase in non-fasting plasma triglycerides was associated with a 41% higher diabetes mortality risk (hazard ratio, 1.41; 95% confidence interval, 1.19–1.67). Participants with non-fasting plasma triglycerides in the highest quintile, versus those in the lowest quintile, had a 141% higher diabetes mortality risk (hazard ratio, 2.41; 95% confidence interval, 1.46–3.97). The positive association of non-fasting plasma triglycerides with diabetes mortality was independent of diabetes status at the baseline. In conclusion, this study demonstrated that non-fasting plasma triglycerides were positively associated with diabetes mortality, independent of diabetes status at baseline. Non-fasting triglycerides may be a therapeutic target for diabetes-related complications. Full article

The initial Swedish discourse of transnational adoption as a win-win situation has changed over its more than 60-year-long history. This article aims to trace and present some themes in this history, with a particular focus on the public debate and the different narratives that representatives of the adoption triangle—the adoptees, the adoptive parents, and the biological parents—tell when dealing with transnational and transracial adoption as a personal and political phenomenon. The article draws from an ongoing study of discourses and narratives of transnational adoption based, above all, on journalism, memoirs, governmental documents, and previous research. It attempts to present the contradictory perspectives on transnational adoption to create a diffractive pattern. The diffractive analysis makes it possible to show how the investigated narratives and discourses on transnational adoption change in encounters with experiences made, and even more so, made visible and knowledgeable, in social practices. However, challenges such as treatment resistance, medication side effects, and long-term tolerability persist, prompting the exploration of novel therapeutic approaches. We aimed to highlight the characteristics and related comorbidities of HIV/AIDS cases in which the antiretroviral therapy was modified. Material and Methods: A cross-sectional clinical investigation was conducted on adults diagnosed with HIV/AIDS who were hospitalized at the “St. Parascheva” Clinical Hospital of Infectious Diseases in Iasi in the Northeastern region of Romania. The timeframe under investigation was 1 January 2023 to 30 June 2023. Results: In the Northeastern part of Romania, from a total of 1692 patients in the active records, there were a total of 148 recorded cases of antiretroviral therapy switch in HIV-infected patients. The main reason for the ART switch was the simplification of the ART regimen (82 cases, 55.40%), viro-immunological failure (16 cases, 10.66%), other disturbances correlated to the ART regimen, dyslipidemia (34 cases 22.97%), depression (3 cases, 2.02%), suicide attempt (1 case, 0.67%), new situations, including the appearance of pregnancy (3 cases 2.02%), and tuberculosis (9 cases, 6.08%). ART before the switch was represented by protease inhibitors that accounted for 84 cases (56.75%) of the ART switch. Following the therapy switch, integrase inhibitor-based ART single-tablet regimens accounted for 43.91% (65 cases) of all changeovers, with non-nucleoside reverse transcriptase inhibitor regimens coming in second, in 63 cases, 42.66%. Conclusions: ART switch as an experimental therapy offers a promising approach to optimizing HIV treatment outcomes. By focusing on viral suppression and immune reconstitution, addressing treatment challenges, and exploring novel ARV agents, ART switch strategies aim to improve the overall health and well-being of individuals living with HIV. Background and Objectives: This study aimed to investigate the relationship between the systemic immune inflammation (SII) index and the development of micro and macro complications and mortality within the first year and the following three years in type 2 diabetic retinopathy patients. Materials and Methods: The retrospective study included 523 type 2 diabetic retinopathy patients seen in the endocrinology outpatient clinic of our hospital between January and December 2019. Their demographic and clinical characteristics were analyzed using descriptive statistics. The normal distribution of quantitative data was assessed by the Shapiro–Wilk test. Mann–Whitney U, McNemar–Chi-square, and Cochran’s Q tests were used to analyze the SII values and complication rates over time. An ROC analysis determined the sensitivity and specificity of SII. A multiple linear regression analysis examined the relationship between variables and SII, while Spearman’s test assessed the correlation between CRP and SII. p < 0.05 was accepted as significant. Results: The mean age of patients was 63.5 ± 9.3 years, with mean SII values of 821.4 ± 1010.8. Higher SII values were significantly associated with acute–chronic renal failure, peripheral arterial disease, and hospitalization rates in both the first year and the following three years (p < 0.05 for all). Significant cut-off values for SII were found for micro- and macrovascular complications and death within the first year (p < 0.05 for all). The ROC curve analysis identified an optimal SII cut-off value of >594.0 for predicting near-term (1-year) complications and mortality, with a sensitivity of 73.8% and specificity of 49.4% (area under the ROC curve: 0.629, p = 0.001). Multiple linear regression indicated that smoking of at least 20 pack-years had a significant positive effect on SII. The Spearman test showed a weak positive correlation between SII and CRP. Conclusions: High SII values predict both early and late acute–chronic renal failure, peripheral arterial disease, and hospitalizations in patients with type 2 diabetic retinopathy. The study also shows that high SII values may predict microvascular and macrovascular complications of type 2 DM and mortality risk in the early period in patients with type 2 diabetic retinopathy. In addition, comorbidities and inflammatory habits, such as long-term smoking, should be considered in the clinical use of SII. Full article

Photosynthetic pigments are fundamental for plant photosynthesis and play an important role in plant growth. Currently, the frequently used method for measuring photosynthetic pigments is spectrophotometry. Additionally, the SPAD-502 chlorophyll meter, with its advantages of easy operation and non-destructive testing, has been widely applied in land agriculture. However, the application prospects of its test results in horticultural plants have not yet been proven. This study examines the reliability of SPAD values for predicting chlorophyll concentrations. Using fresh and senescent leaves from four common horticultural plants, we measured SPAD values, photosynthetic pigment concentrations, and leaf color parameters. A generalized linear mixed model demonstrated that SPAD values are a reliable indicator for predicting chlorophyll concentrations, yet interspecific variations exist. Based on the predictive power of SPAD values for chlorophyll, we first propose an Enrichment Index (CEI) and a Normal Chlorophyll Concentration Threshold (NCCT). The CEI can be used to compare SPAD values among different species, and the NCCT value can serve as a more accurate indicator for assessing the growth potential of old trees. However, due to a limited sample size, further research with larger samples is needed to refine the diagnosis of plant growth potential and enhance the management of ornamental plant cultivation. Background and Objectives: The risk of developing glycemic dysregulation up to overt diabetes mellitus (DM) after an episode of acute pancreatitis (AP) is increasingly being analyzed. We aimed to assess the changes in serum glucose levels associated with the first episode of AP, as well as the impact of dysglycemia on outcomes such as the severity of inflammation, the length of hospitalization, mortality, and the persistence of hyperglycemia at follow-up. Materials and Methods: All patients experiencing their first episode of AP, who presented to the Emergency Room (ER) between 1 January 2020 and 31 December 2023, were retrospectively included. On-admission serum glucose and peak serum glucose during hospitalization were the biological markers used to assess glucose metabolism impairment, and they were correlated with outcomes of AP. Results: Our study included 240 patients, 46.67% (112 patients) having a biliary etiology for an AP flare. Patients with COVID-19-associated AP exhibited the highest on-admission and peak serum glucose levels (244.25 mg/dL and 305.5 mg/dL, respectively). A longer hospital stay was noted in patients with peak serum glucose levels of ≥100 mg/dL (9.49 days) compared to normoglycemic patients (6.53 days). Both on-admission and peak glucose levels were associated with elevated CRP levels during hospitalization. A total of 83.78% of patients who received antibiotics exhibited on-admission hyperglycemia, and 72.07% had peak serum glucose levels of ≥100 mg/dL. The presence of hyperglycemia at follow-up was associated with both on-admission and peak serum glucose levels of ≥100 mg/dL, as well as with a longer stay, higher CRP levels, and antibiotic use during index admission. Conclusions: On-admission hyperglycemia predicts a higher inflammatory response in patients at the first episode of AP, while the presence of hyperglycemia during hospitalization is associated with imaging and biological severity and longer hospitalizations, indicating a more severe disease course. Both on-admission and peak in-hospital hyperglycemia were identified as risk factors for sustained hyperglycemia at follow-up. Full article

The improvement of battery management systems (BMSs) requires the incorporation of advanced battery status detection technologies to facilitate early warnings of abnormal conditions. In this study, acoustic data from batteries under two discharge rates, 0.5 C and 3 C, were collected using a specially designed battery acoustic test system. By analyzing selected acoustic parameters in the time domain, the acoustic signals exhibited noticeable differences with the change in discharge current, highlighting the potential of acoustic signals for current anomaly detection. In the frequency domain analysis, distinct variations in the frequency domain parameters of the acoustic response signal were observed at different discharge currents. The identification of acoustic characteristic parameters demonstrates a robust capability to detect short-term high-current discharges, which reflects the sensitivity of the battery’s internal structure to varying operational stresses. Acoustic emission (AE) technology, coupled with electrode measurements, effectively tracks unusually high discharge currents. The acoustic signals show a clear correlation with discharge currents, indicating that selecting key acoustic parameters can reveal the battery structure’s response to high currents. This approach could serve as a crucial diagnostic tool for identifying battery abnormalities. Full article

The milk fat globule membrane (MFGM) contains bioactive proteins, carbohydrates, and lipids. Polar lipids found in the MFGM play a critical role in maintaining cell membrane integrity and neuronal signalling capacity, thereby supporting brain health. This review summarises the literature on the MFGM and its phospholipid constituents for improvement of mental health across three key stages of the human lifespan, i.e., infancy, adulthood, and older age. MFGM supplementation may improve mental health by reducing neuroinflammation and supporting neurotransmitter synthesis through the gut–brain axis. Fortification of infant formula with MFGMs is designed to mimic the composition of breastmilk and optimise early gut and central nervous system development. Early behavioural and emotional development sets the stage for future mental health. In adults, promising results suggest that MFGMs can reduce the negative consequences of situational stress. Preclinical models of age-related cognitive decline suggest a role for the MFGM in supporting brain health in older age and reducing depressive symptoms. While there is preclinical and clinical evidence to support the use of MFGM supplementation for improved mental health, human studies with mental health as the primary target outcome are sparce. Further high-quality clinical trials examining the potential of the MFGM for psychological health improvement are important. Full article

This case explores the subjective nature of professional judgment related to audit risk through the lens of a fictitious company, Sparked Intuition Power (SIP). SIP has just been accepted as a new audit client by an international accounting firm. Students play the role of associates on the audit engagement team. As part of their work on the audit, the students identify potential risks present at SIP as part of the initial risk assessment process. Once these risks have been identified, the students decide how to assess the potential severity of each risk and determine the resulting effect on the overall auditor workload. Through interaction and discussion with their instructors, the students should be better able to navigate the decision-making process related to risk evaluation in an audit engagement. Full article

Deep learning (DL)-oriented document processing is widely used in different fields for extraction, recognition, and classification processes from raw corpus of data. The article examines the application of deep learning approaches, based on different neural network methods, including Gated Recurrent Unit (GRU), long short-term memory (LSTM), and convolutional neural networks (CNNs). The compared models were combined with two different word embedding techniques, namely: Bidirectional Encoder Representations from Transformers (BERT) and Gensim Word2Vec. The models are designed to evaluate the performance of architectures based on neural network techniques for the classification of CVs of Moroccan engineering students at ENSAK (National School of Applied Sciences of Kenitra, Ibn Tofail University). The used dataset included CVs collected from engineering students at ENSAK in 2023 for a project on the employability of Moroccan engineers in which new approaches were applied, especially machine learning, deep learning, and big data. Accordingly, 867 resumes were collected from five specialties of study (Electrical Engineering (ELE), Networks and Systems Telecommunications (NST), Computer Engineering (CE), Automotive Mechatronics Engineering (AutoMec), Industrial Engineering (Indus)). The results showed that the proposed models based on the BERT embedding approach had more accuracy compared to models based on the Gensim Word2Vec embedding approach. Accordingly, the CNN-GRU/BERT model achieved slightly better accuracy with 0.9351 compared to other hybrid models. On the other hand, single learning models also have good metrics, especially based on BERT embedding architectures, where CNN has the best accuracy with 0.9188. Full article

Monascus pigments (MPs), a class of secondary metabolites produced by Monascus spp., can be classified into yellow, orange, and red MPs according to their differences in the wavelength of the maximum absorption. However, the biosynthetic sequence and cellular biosynthesis mechanism of different MPs components are still not yet completely clear in Monascus spp. In this study, the subcellular localization of five MPs synthases was investigated using fluorescent protein fusion expression. The results revealed that the proteins encoded by the MPs biosynthetic gene cluster were compartmentalized in various subcellular locations, including the mitochondrial polyketide synthase MrPigA, cytosolic enzymes consisting of the ketoreductase MrPigC, the oxidoreductase MrPigE, and the monooxygenase MrPigN, and the cell-wall-bound oxidoreductase MrPigF. Moreover, the correct localization of MrPigF to the cell wall was crucial for the synthesis of orange MPs. Lastly, we discussed the compartmentalized biosynthetic pathway of MPs. This study will not only be helpful in clarifying the biosynthetic sequence and biosynthesis mechanism of different MPs but also provides new insights into the cellular biosynthesis of secondary metabolites in filamentous fungi. Efficient thawing can preserve the quality of frozen hairtail (Trichiurus lepturus) close to that of fresh hairtail. In contrast to air thawing (AT) and radio-frequency thawing (RT), this study looked at how graphene oxide (GO) and graphene magnetic (GM) nanoparticles paired with RT affect the microstructure and protein conformation of hairtails after thawing. The results suggested that GM-RT can reduce the myofibrillar protein (MP) damage and be more effective than other thawing treatments, like AT, RT, and GO-RT, in maintaining the microstructure of hairtail. The particle size and zeta potential showed that GM-RT could reduce the aggregation of MP during the thawing process compared to other thawing methods. Moreover, the texture of the hairtail after GM-RT exhibited higher hardness (1185.25 g), elasticity (2.25 mm), and chewiness (5.75 mJ) values compared to other thawing treatments. Especially compared with RT, the GM-RT treatment displayed significant improvements in hardness (27.24%), a considerable increase in springiness (92.23%), and an increase in chewiness (57.96%). GO-RT and GM-RT significantly reduced the centrifugal loss. The scanning electron microscopy results demonstrated that the effect of GM-RT was more akin to that of a fresh sample (FS) and characterized by a well-organized microstructure. In conclusion, GM-RT effectively diminished the MP aggregation and improved the texture of thawed fish. It can be regarded as a viable alternative thawing technique to enhance MP stability, which is vital for preserving meat quality. Full article
