Abstract

The heptapeptide core common to melanocyte-stimulating hormone (MSH) and adrenocorticotropin (ACTH) was administered as a single subcutaneous dose of 30 mg to 13 elderly human subjects (9 men, 4 women) in a double-blind, cross-over design. Significant improvement was found in the Benton Visual Retention Test after MSH/ACTH 4–10 as compared with administration of saline. This appeared to be greater in men than women. No side effects or laboratory abnormalities were observed. The behavioral results are consistent with our earlier findings in men and rats of improved visual attention after administration of MSH and extend them to the elderly population.

Effects of peripherally injected pituitary peptides on recognition memory in pigeons

The effects of peripheral injection of arginine vasopressin (AVP), oxytocin (OT), arginine vasotocin (AVT), adrenocorticotrophic hormone (ACTH_4–10) and α-melanocyte-stimulating hormone (α-MSH) were studied, using pigeon subjects and a pair comparison task, with and without intervening delays between stimuli presentation. The highest dose (20 μg/kg) of AVT impaired performance by disrupting input, but not storage. General cognitive ability was unimpaired, as were perceptual mechanisms. None of the other peptides affected recognition memory, in that forgetting curves were unchanged when compared with control. The results are discussed in terms of species-specific roles for these peptides.

The effect of MSH/ACTH 4-10 on delayed response performance and post-test locomotor activity in rats

Delayed response performance was measured in male, Long-Evans rats 1 hr after IP administration of various doses of MSH/ACTH 4–10 or control in a Hunter delayed reaction apparatus. Additional treatments consisting of naloxone 500 μg/kg (IP) and naloxone 500 μg/kg in conjunction with MSH/ACTH 4–10 95 μg/kg were also administered. Directly after delayed response performance was assessed, gross locomotor activity was determined. MSH/ACTH 4–10, at a dose of 95 μg/kg, significantly enhanced retention of a visual stimulus, while MSH/ACTH 4–10, at doses of 195 and 285 μg/kg, significantly impaired delayed response performance. Naloxone treatment resulted in significantly impaired delayed response performance when compared to control. However, naloxone plus MSH/ACTH 4–10 treatment failed to produce a significant difference from control in the delayed response performance paradigm. In post-test locomotor activity determination, an apparent dose-response existed for MSH/ACTH 4–10 with the two highest doses (190 and 285 μg/kg) resulting in significantly increased locomotor activity. The observed delayed response performance data support theories implicating MSH/ACTH peptides in attentional processes involving visual stimuli. The fact that large doses of MSH/ACTH 4–10 disrupt delayed response performance while increasing post-test activity suggest that an optimum level of effect caused by the MSH/ACTH peptide exists in this paradigm.
Disregulation of hypothalamic-pituitary-adrenal axis in the mentally retarded

Previous reports of cognitive and social improvement in the mentally retarded after administration of MSH/ACTH fragments suggested disregulation of the hypothalamic-pituitary-adrenal (HPA) axis. The current study examined the integrity of this system with the Dexamethasone Suppression Test (DST). The DST is a biological index of HPA integrity and recently has been used as a diagnostic aid for endogenous depression. Thirty-five mentally retarded patients were administered 1 mg of dexamethasone just after a sample of blood was taken. Blood samples were analyzed for cortisol by RIA at 11:00 p.m. (basal), 8:00 a.m., 4:00 p.m., and 10:00 p.m. Between 40% and 48% (depending on sampling) of the patients failed to suppress cortisol (>4 μg/dl), after the DST challenge. The results suggested that a significant proportion of mentally retarded patients have a DST index reflecting a disordered HPA axis and complements earlier studies of cognitive enhancement observed after treatment with MSH/ACTH fragments. The possibility that the stress of hospitalization was related to a disordered HPA was suggested. The possible co-existence of depression in the mentally retarded invites further study.
Memory and Learning

Although difficult to find traces, drugs are believed to have effect of memory and learning. Drug-induced changes in acquisition or extinction are interpreted to indicate that the drug has affected memory processes. However, the effectiveness of drugs that impair (retrograde amnesia) or facilitate (delay extinction) retention is sensitive to dose, type and degree of training, and time of drug administration. The recent trend has been to combine the behavioral results with biochemical measurements in an attempt to identify brain structures and systems that may be involved in the memory process. Many studies that have focused on measuring the consequences of electrical stimulation or microinjection of drugs into specific brain loci have shown that behaviorally-active drugs affect neurotransmitter systems, particularly noradrenergic and dopaminergic pathways. Studies in humans tend to corroborate the data in rodents. For example, administration of scopolamine to young students impaired memory storage and retrieval, as well as other cognitive functions. The impairment of cognitive function in both humans and primates was similar to that found in normal aged subjects. It has been shown that rats that are deficient in vasopressin have deficits in acquiring and maintaining avoidance behavior that can be corrected by treatment with this peptide. Interactions between opiate peptide systems and vasopressin pathways may also be of functional significance in memory processes. Experiments performed using subhuman species suggest that cholinomirnetic drugs should improve memory and cognition in humans who do not perform well, and this view is supported by many favorable clinical reports.
