    Description: The RNA binding protein HuD plays essential roles in neuronal development and plasticity. We have previously shown that HuD stimulates translation. Key for this enhancer function is the linker region and the poly(A) binding domain of HuD that are also critical for its function in neurite outgrowth. Here, we further explored the underlying molecular interactions and found that HuD but not the ubiquitously expressed HuR interacts directly with active Akt1. We identify that the linker region of HuD is required for this interaction. We also show by using chimeric mutants of HuD and HuR, which contain the reciprocal linker between RNA-binding domain 2 (RBD2) and RBD3, respectively, and by overexpressing a dominant negative mutant of Akt1 that the HuD–Akt1 interaction is functionally important, as it is required for the induction of neurite outgrowth in PC12 cells. These results suggest the model whereby RNA-bound HuD functions as an adapter to recruit Akt1 to trigger neurite outgrowth. These data might also help to explain how HuD enhances translation of mRNAs that encode proteins involved in neuronal development.

    Description: Stem cells are maintained in the niche by intercellular interactions and signaling networks. In this work, we study extracellular signals required for maintenance of the root stem cell niche in higher plants. We identify a family of functionally redundant homologous peptides that are secreted, tyrosine-sulfated, and expressed mainly in the stem cell area and the innermost layer of central columella cells. We name these peptides root meristem growth factors (RGFs). RGFs are required for maintenance of the root stem cell niche and transit amplifying cell proliferation in Arabidopsis. RGF1 defines expression levels and patterns of the stem cell transcription factor PLETHORA, mainly at the posttranscriptional level. The RGFs function independently of the auxin pathway.     Description: The transcriptional profiles of mouse embryonic, neural, and hematopoietic stem cells were compared to define a genetic program for stem cells. A total of 216 genes are enriched in all three types of stem cells, and several of these genes are clustered in the genome. When compared to differentiated cell types, stem cells express a significantly higher number of genes (represented by expressed sequence tags) whose functions are unknown. Embryonic and neural stem cells have many similarities at the transcriptional level.     Description: Vasoactive intestinal peptide, a smooth-muscle relaxant neuropeptide with neurotransmitter properties, was relaxed during electrical field stimulation of guinea pig trachea. The amount released correlated with the degree of relaxation, and the release was blocked by tetrodotoxin. Prior incubation of the trachea with antiserum to vasoactive intestinal peptide reduced the relaxation.     Description: Geographical variability in daily precipitation was investigated in Izu-Oshima, Miyakejima, and Hachijojima Islands in the Izu Islands, located in the southeastern ocean of Japan, utilizing data observed by surface rain gauges in the dense network constructed by the Japan Meteorological Agency and the Tokyo Metropolitan Government Bureau of Construction. These islands have sizes on a scale of less than 10 km. Izu-Oshima and Miyakejima have larger geographical variability than Hachijojima in the mean precipitation of the individual rain gauges within each island. Some rain gauges have differences from the island-mean precipitation by up to 40%. The variability is relatively large accompanied by inconsistency in temporal patterns of daily precipitation within each island in the summer months. This result implies that scattered rainfall systems with a smaller size than the islands dominated in these months, characterized by less frequent passages of synoptic disturbances in the southern part of Japan. A season-independent analysis reveals that the variability is large on days with relatively light precipitation or weak wind speed and independent of wind direction.

    Notes: Reactions of a hydrogen molecule with small platinum clusters Ptn (n=1,2,3) are studied theoretically by ab initio methods. This provides a cluster model study for hydrogen chemisorption on a Pt surface. The results suggest that the Pt atom and the linear Pt3 cluster will react with H2 and dissociatively adsorb it, making two Pt–H bonds, whereas the Pt2 cluster will not react with H2 because of a high energy barrier. The dissociative adsorption of a hydrogen molecule occurs at a side-on, on-top site of a surface Pt atom and molecular adsorption does not seem to occur. Essentially only one Pt atom is involved in the initial adsorption process. These findings are in contrast to the palladium case previously reported. Almost no energy barrier exists for the hydrogen migration from one Pt atom to an adjacent one, with a preference being shown for one H atom on each Pt atom rather than two H atoms on one Pt atom. The heat of adsorption, the stability of the catalytic surface, etc., are best represented by the Pt3–H2 system. Spin–orbit coupling effect is examined for the Pt–H2 system and found to be small for the singlet A1 state which is the most important state for the dissociative adsorption. This effect is important, however, to obtain natural potential curve in the large Pt–H2 separation.
