    Notes: Two-dimensional gel electrophoresis was used to follow low changes in gene expression associated with antibiotic (bialaphos) biosynthesis in Streptomyces hygroscopicus. Cultures were pulse-labelled with [35S]-methionine before, during, and after the switch from primary to secondary metabolism in order to compare kinetic profiles of bialaphos (antibiotic) production (bap) genes during this metabolic transition. Separation of gene products on two-dimensional gels revealed that 27 were dependent on brpA for optimal expression and were activated as the culture approached stationary phase. Genes which encoded 10 brp4-dependent proteins were mapped to a 10 kb Sstl fragment of the 35 kb bap gene cluster by expressing them in Streptomyces lividans using the thiostrepton-inducible tipA promoter. N-terminal amino acid sequences of two brpA-dependent proteins, obtained by direct microsequencing of protein spots excised from two-dimensional gels, identified them as gene products mapping to the same region and involved in secondary metabolic conversions of the bap pathway. The kinetics of synthesis of 16 brpA-dependent gene products were characterized using QUEST computer software. Cluster analysis performed on the kinetics of synthesis of 346 of the most highly expressed gene products of HP5-29, including 16 brpA-dependent ones, identified 75 families having distinct patterns of expression. Many brpA-dependent proteins were clustered together; 10 were found in one kinetic family. These kinetic families also included brpA-independent gene products perhaps subject to similar regulatory mechanisms and thus possibly involved in bialaphos biosynthesis. The activation/derepression of bap expression took place as cultures approached stationary phase and was temporally related to synthesis of ppGpp.

    Notes: Abstract Ras is a low-molecular-weight guanine nucleotide (GDP/GTP)-binding protein that transduces signals for growth and differentiation in eukaryotes. In mammals, the importance of Ras in regulating growth is underscored by the observation that activating mutations in ras genes are found in many animal tumors. Colletotrichum trifolii is a filamentous fungal pathogen of alfalfa which causes anthracnose disease. To investigate signaling pathways that regulate growth and development in this fungus, a gene encoding a Ras homolog (CT-Ras) was cloned from C. trifolii. CT-Ras exhibited extensive amino acid similarity to Ras proteins from higher and lower eukaryotes. A single amino acid change resulting in mutationally activated CT-Ras induced cellular transformation of mouse (NIH 3T3) fibroblasts and tumor formation in nu/nu mice. In Colletotrichum, mutationally activated CT-Ras induced abnormal hyphal proliferation and defects in polarized growth, and significantly reduced differentiation in a nutrient-dependent manner. These results show that C. trifolii Ras is a functional growth regulator in both mammals and fungi, and demonstrate that proper regulation of Ras is required for normal fungal growth and development.

    Notes: Abstract Analysis of the mean wind, equivalent potential temperature and virtual potential temperature profiles observed by the National Center for Atmospheric Research (NCAR) Electra aircraft and obtained from dropwindsondes and ship-launched radiosondes were made in conjunction with synoptic observations to study the structure of the monsoon boundary layer over the Arabian Sea during MONEX 79. Comparison of mean profiles indicates the monsoon boundary layer to be much different from the trade wind boundary layer. Results confirm the existence of a boundary-layer jet known as East African or Somali Jet. Regions of multiple cloud layers at roughly the height of the capping inversion layer were associated with the jet. Regions in which a more well-mixed layer was observed showed a jet structure depressed in height. A free-jet surface-layer model appears to describe the mean wind structure of this jet observed during the present study and by others. An approximate balance of forces was found in the monsoon boundary layer between friction, advective acceleration, Coriolis and pressure gradient forces. Friction and advective acceleration terms were significant in the lower levels of the boundary layer. Forces in a typical trade wind boundary layer were found to be approximately one order of magnitude smaller than those observed in the monsoon boundary layer.

    Notes: The complete characterization of a branched polymer involves many factors, the most difficult to assess being the average number of elementary linear portions, or branches, of which the polymer is composed, the degree of polymerization of the branches and the extent of cyclic structure formation. A method of analysis of branched polymers is proposed which should be applicable to polymers of any polyfunctional monomer the unsaturated groups of which are separated by saponifiable links. The method is based on the measurement of degree of polymerization and residual unsaturation of the polymer followed by saponification and determination of degree of polymerization of the resulting polymer, and involves the assumptions that branching in the polymer occurs only by the sharing of a monomer unit by two or more linear portions or branches depending on the number of double bonds in the monomer and also that the average degree of polymerization of the branches remains unchanged by the saponification treatment. Evidence from experiments on the saponification and re-esterification of poly(allyl acetate) and poly(allyl benzoate) indicates, that, in the case of diallyl esters, the above conditions are satisfied. The technique was therefore applied to three polymers of diallyl phthalate, and in each case the average number of chains and degree of polymerization of the chains were estimated. Evidence of considerable extents of cyclic structure formation was found, each individual branch in the polymers consisting of approximately fourteen diallyl phthalate units, six of which are doubly reacted. It is suggested that the existence of extensive formation of cyclic structures may be partly responsible for the poor correlation of predicted with actual degrees of conversion at gelation in polymerizations involving multifunctional monomers.

    Notes: Recent investigations have shown that cyclization or intramolecular crosslinking plays a major part in the polymerization of diallyl phthalate, and an extension of the network theory of gelation to allow for this led to satisfactory agreement between actual and predicted degrees of conversion at gelation for this monomer. It became important, therefore, to find whether satisfactory agreement could be obtained for other systems, and a detailed examination of the polymerization of the diallyl esters of isophthalic, terephthalic, oxalic, and sebacic acids has now been made. In all of these cases gelation occurs at a conversion higher than the predicted value, the discrepancy being considerably greater than can be accounted for by experimental error. Complicating factors in the interpretation of the experimental results are shown to be absent and it is concluded that cyclization in the poly(diallyl esters) does not affect gelation in the manner expected from theory, and that the satisfactory correlation in the case of diallyl phthalate is fortuitous. It is shown, in the case of the isomeric phthalates which are considered strictly comparable, that all three systems gel at the same degree of conversion, although the ortho ester polymer has considerably fewer pendent allyl groups available for crosslinking and is also of a lower chain length than the other two polymers. This fact seems irreconcilable with the network theory of the gel point as it exists at present. The results are discussed in relation to Walling's diffusion control theory, and although the constancy of conversion at gelation is consistent with the predictions of this theory, supporting evidence regarding molecular complexity is lacking.

    Description: We use the Shallow Radar (SHARAD) on the Mars Reconnaissance Orbiter to search for subsurface interfaces and characterize surface roughness at the landing sites of Viking Landers 1 and 2, Mars Pathfinder, the Mars Exploration Rovers Spirit and Opportunity, the Phoenix Mars lander, the Mars Science Laboratory Curiosity rover, and three other sites proposed for Curiosity. Only at the Phoenix site do we find clear evidence of subsurface radar returns, mapping out an interface that may be the base of ground ice at depths of ~15–66 m across 2900 km 2 in the depression where the lander resides. At the Opportunity , Spirit , and candidate Curiosity sites, images and altimetry show layered materials 10s to 100s of m thick extending 10s to 100s of km laterally. These scales are well within SHARAD's resolution limits, so the lack of detections is attributable either to low density contrasts in layers of similar composition and internal structure or to signal attenuation within the shallowest layers. At each site, we use the radar return power to estimate surface roughness at scales of 10–100 m, a measure that is important for assessing physical properties, landing safety, and site trafficability. The strongest returns are found at the Opportunity site, indicating that Meridiani Planum is exceptionally smooth. Returns of moderate strength at the Spirit site reflect roughness more typical of Mars. Gale crater, Curiosity's ultimate destination, is the smoothest of the four proposed sites we examined, with Holden crater, Eberswalde crater, and Mawrth Vallis exhibiting progressively greater roughness.
