    Description: The isotope (dO18 or dD) variability in ice cores from non-tempered Alpine glaciers might provide long term temperature proxy records at mid-latitudes supplementing respective Polar records. Concerning the last millennium, this exclusively applies to the low accumulation Colle Gnifetti (4450 m asl, Swiss-Italian Alps), though depositional noise challenges reliable temperature reconstruction there. For two ice cores from this drill site a substantial covariation in trends between atmospheric temperature and water isotopes has already been found for the last 250 years- including important exceptions, however. Thus, the decadal isotope variations remain still to be proved as a faithful temperature signal. For Colle Gnifetti, this task is adressed on the basis of the extensive instrumental temperature records provided by the HISTALP network. Here we restrict ourselves as a first step on the last century, where shortcommings in both archives are not yet obstructive- such as ice core dating uncertainties and ambiguities associated with the early instrumental period. Aimed at assessing the temperature related signal embedded in the isotope variability we include isotope time series from four down-to-bedrock ice cores subject to different glaciological regimes. Dedicated time series analysis yields a common signal in the decadal components. For a sound comparsion, a special Colle Gnifetti relevant temperature time series is constructed from the high elevation stations of the HISTALP network. This attempt includes weighting for precipitation and net accumulation, resulting in an ice core relevant temperature record mostly associated with growing seasons. The intercomparison of trends yields a common decadal variability between temperature and ice core isotope data. Thereby the potential of the Colle Gnifetti ice cores in Alpine paleoclimate research is assessed, which may include the early instrumental correction debate.

    Description: The variabilty of water isotopes (delta-O18 or delta-D) preserved in Alpine glaciers may provide mid-latitude temperature records supplementing respective proxy information from other archives. However, concerning long term records (i.e. exceeding 100 years) the limited glacier depth of suitable Alpine drill sites requires a relatively low accumulation rate. As a result, depositional noise is found to strongly challenge the interpretation of these isotope records in terms of net temperature change. Here we present results from our multi core array at the low accumulation Colle Gnifetti (CG) site (4450m asl, Swiss-Italian Alps) dedicated to assess the potential of long term ice core isotope records for paleoclimate reconstruction. To do so, a specific CG relevant temperature time series is synthesized from the high elevation stations of the instrumental HISTALP network considering among others the accumulation bias towards growing seasons. Within the last century (where dating and homogenity problems do not matter) dedicated time series analysis reveals a common signal in the decadal components of the temperature record and isotope records of four ice cores. Throughout the entire 250 year instrumental period, exceptions occur within the early instrumental period, as well as an unexpectedly high isotope-temperature sensitivity. An inter-core correspondence in the temperature related decadal trends can be revealed for two cores throughout the last 500 years. On a tentative basis, we discuss the delta O18-record of a new CG ice core dedicated to last 1000 years in view of constraints arising from varying time resolution, dating uncertainties and systematic glaciological up-stream effects.

    Notes: Summary The total and free steady-state plasma levels of nifedipine in patients with renal failure have been compared with those in subjects with normal renal function. Studies were done after administration of nifedipine 10 mg t.d.s. p.o. for 5 days, after i.v. infusion of 4·4 mg, and after a single 10 mg oral dose. The systemic clearance of nifedipine after a single i.v.-dose was higher in subjects with renal insufficiency (854 ml/min) than in those with normal renal function (468 ml/min). After the single oral dose the AUC (6100 ng·min·ml−1) and maximum plasma concentration (75.0 ng·ml−1) were lower than in subjects with normal renal function (19300 ng·ml−1; 122 ng·ml−1). The plasma protein binding of nifedipine averaged 95.5% in normal subjects and 94.8% in patients with renal failure. Although free and total steady-state plasma levels of nifedipine tended to be somewhat lower than normal in renal failure, the changes in pharmacokinetics and decreased protein binding of nifedipine did not result in a significantly different steady-state plasma level of the drug. The blood pressure response to a given plasma nifedipine level appeared to be enhanced in renal failure.

    Notes: Summary The influence of antituberculosis drugs on the plasma level of verapamil was studied after its oral and intravenous administration. Six patients who had been treated for at least 6 months with a combination of rifampicin, ethambutol and isoniazid received a single oral dose of 40 mg verapamil. As compared to untreated subjects, the antituberculosis drugs greatly reduced the bioavailability of the calcium antagonist. Studies in patients in whom treatment with ethambutol and isoniazid had been discontinued revealed that the effect was due to rifampicin. The drugs for tuberculosis had no influence on the plasma level of verapamil when it was given intravenously. The findings can be explained by the induction of verapamil metabolizing liver enzymes in patients treated with rifampicin.

    Notes: Summary The time course of the blood pressure lowering effect and the dose-response relationship of the new angiotensin converting enzyme inhibitor ramipril (HOE 498) were studied in 8 patients with essential hypertension. As compared with placebo, a single oral dose of 2.5 mg ramipril lowered systolic and diastolic blood pressure. The antihypertensive action of single oral doses of 5, 7.5 and 10 mg ramipril was more pronounced. No change in heart rate occurred. Angiotensin converting enzyme activity was suppressed after all doses of ramipril studied. Plasma renin activity increased after 2.5 mg and 5 mg ramipril. Plasma aldosterone was not affected by 2.5 mg, but it fell after 5 mg ramipril. Thus, ramipril produced prolonged inhibition (more than 12 hours) of angiotensin converting enzyme activity and lowered blood pressure in patients with essential hypertension.

    Notes: Summary The pharmacokinetics of propranolol after oral and intravenous administration was studied at rest and on an exercise day in 8 healthy subjects. On the exercise day the subjects performed physical exercise for 7 h, consisting of bicycle ergometer exercise at 50% of maximal work capacity and outdoor walking. Propranolol (80 mg p.o., or 0.2 mg/kg body weight i.v.) was administered 30 min before the start of the exercise. After oral administration the terminal phase halflife, (t1/2β) and area under the curve (AUC) were both significantly reduced on the exercise day compared to the rest day. The bioavailability of propranolol was reduced by prolonged physical exercise and plasma levels of propranolol were about 30% lower at the end of the exercise day than at the end of the rest day. After intravenous administration, t1/2β was also reduced on the exercise day as compared to the rest day. AUC, clearance and volume of distribution did not differ on the two days. On the other hand, indocyanine green (ICG) clearance was significantly reduced during the bicycle ergometry periods on the exercise day. The combination of reduced ICG clearance, suggesting a reduction in hepatic blood flow, and a decreased t1/2β and unchanged clearance of propranolol on the exercise day was unexpected.

    Notes: Summary The pharmacokinetics and plasma protein binding of nitrendipine in patients with terminal renal failure have been compared with those in subjects with normal renal function. Kinetic parameters were calculated after a single 40 mg oral dose, an i.v. injection of 3 mg and after a 15 mg i.v. infusion of nitrendipine. Steady-state plasma levels were determined after 5 days of oral treatment with 20 mg b.d. Pharmacokinetic parameters and steady-state plasma levels in patients with renal failure did not differ from those in subjects with normal renal function. Nitrendipine was as highly bound to plasma proteins in patients with renal failure, as in subjects with normal renal function. The plasma protein did not differ between the two. The dosage of nitrendipine need not be modified for kinetic reasons in patients with renal failure.

    Notes: Summary In a double-blind, placebo-controlled crossover study, the plasma atrial natriuretic peptide (ANP) levels of nine young and ten elderly hypertensive patients were compared after placebo and after treatment with 120 mg verapamil given three times daily over 4 weeks. During placebo, plasma ANP levels proved to be higher in elderly patients than in young subjects. Chronic treatment with verapamil induced a rise in ANP levels in both young and elderly patients with hypertension.
